Nous avons étudié les performances de survie et de croissance, au cours de la résorption vitelline, de descendants issus d'une souche domestique, d'une souche sauvage et des deux croisements réciproques entre celles-ci. Les différents génotypes ont été élevés sur substrats lisses et rugueux. Nous avons pu mettre en évidence un effet positif du substrat rugueux sur la croissance et la survie de tous les lots ainsi qu'un effet souche en faveur de la souche domestique. Par contre, aucun effet génétique significatif n'a pu être décelé.

Deux modes de transfert automnal en mer (direct ou progressif) ont été testés chez la truite commune (Salmo truttaj sur des poissons âgés de 10 mois et deux catégories de poids différentes. (I : Pm = 40 g, S : Pm = 50 g). Au cours des trois premières semaines qui suivent le transfert, une mortalité relativement élevée (22 %) est observée sur le lot " I " transféré directement, les trois autres lots (I et S transférés progressivement, S transféré directement) présentant une bonne survie (97 %). Cette mortalité affecte essentiellement les plus petits individus (inférieurs à 40 grammes). Au cours de la période d'observation ultérieure (5 mois), les survies sont élevées et comparables dans les quatre lots. A l'issue de cette période, les poids moyens des lots ne diffèrent pas selon le mode de transfert et sont en outre supérieurs à ceux des témoins de même taille initiale, conservés en eau douce. Il apparaît donc possible d'effectuer un transfert automnal direct en mer pour des animaux supérieurs à 40 grammes sans effets néfastes sur leur survie et leur croissance ultérieure.

La comparaison d'échantillons de truite fario (Salmo trutta L.) prélevés dans différents bassins hydrographiques de Corse avec la souche de pisciculture utilisée pour le repeuplement dans cette zone a permis de définir des variants électrophorétiques caractéristiques de la souche domestique., L'absence de ces allèles dans les échantillons prélevés dans les rivières non sujettes au repeuplement et leur présence simultanée dans les échantillons provenant de cours d'eau, où les repeuplements sont régulièrement réalisés, mettent clairement en évidence une modification de la structure génétique des stocks locaux par les sujets de pisciculture déversés. L'analyse de la structure génotypique des échantillons génétiquement "contaminés" indique, en outre, qu'il n'y a pas d'isolement reproducteur total, en milieu naturel, entre souche de pisciculture et populations naturelles de Corse.

Strains of two salmonid species, brown trout, Salmo trutta L., and Atlantic salmon, Salmo salar L., were compared for the purpose of marine aquaculture. Abstract Growth rate is the main breeding goal of fish breeders, but individual selection has often shown poor responses in fish species. The PROSPER method was developed to overcome possible factors that may contribute to this low success, using (1) a variable base population and high number of breeders (Ne > 100), (2) selection within groups with low non-genetic effects and (3) repeated growth challenges. Using calculations, we show that individual selection within groups, with appropriate management of maternal effects, can be superior to mass selection as soon as the maternal effect ratio exceeds 0.15, when heritability is 0.25. Practically, brown trout were selected on length at the age of one year with the PROSPER method. The genetic gain was evaluated against an unselected control line. After four generations, the mean response per generation in length at one year was 6.2% of the control mean, while the mean correlated response in weight was 21.5% of the control mean per generation. At the 4th generation, selected fish also appeared to be leaner than control fish when compared at the same size, and the response on weight was maximal (≈130% of the control mean) between 386 and 470 days post fertilisation. This high response is promising, however, the key points of the method have to be investigated in more detail.

Abstract Background Rainbow trout is an economically important fish and a suitable experimental organism in many fields of biology including genome evolution, owing to the occurrence of a salmonid specific whole-genome duplication (4th WGD). Rainbow trout is among some of the most studied teleosts and has benefited from substantial efforts to develop genomic resources (e.g., linkage maps. Here, we first generated a synthetic map by merging segregation data files derived from three independent linkage maps. Then, we used it to evaluate genome conservation between rainbow trout and three teleost models, medaka, stickleback and zebrafish and to further investigate the extent of the 4th WGD in trout genome. Results The INRA linkage map was updated by adding 211 new markers. After standardization of marker names, consistency of marker assignment to linkage groups and marker orders was checked across the three different data sets and only loci showing consistent location over all or almost all of the data sets were kept. This resulted in a synthetic map consisting of 2226 markers and 29 linkage groups spanning over 3600 cM. Blastn searches against medaka, stickleback, and zebrafish genomic databases resulted in 778, 824 and 730 significant hits respectively while blastx searches yielded 505, 513 and 510 significant hits. Homology search results revealed that, for most rainbow trout chromosomes, large syntenic regions encompassing nearly whole chromosome arms have been conserved between rainbow trout and its closest models, medaka and stickleback. Large conserved syntenies were also found between the genomes of rainbow trout and the reconstructed teleost ancestor. These syntenies consolidated the known homeologous affinities between rainbow trout chromosomes due to the 4th WGD and suggested new ones. Conclusions The synthetic map constructed herein further highlights the stability of the teleost genome over long evolutionary time scales. This map can be easily extended by incorporating new data sets and should help future rainbow trout whole genome sequence assembly. Finally, the persistence of large conserved syntenies across teleosts should facilitate the identification of candidate genes through comparative mapping, even if the occurrence of intra-chromosomal micro-rearrangement may hinder the accurate prediction their genomic location.

Abstract Background There is increasing evidence that the ability to adapt to seawater in teleost fish is modulated by genetic factors. Most studies have involved the comparison of species or strains and little is known about the genetic architecture of the trait. To address this question, we searched for QTL affecting osmoregulation capacities after transfer to saline water in a nonmigratory captive-bred population of rainbow trout. Results A QTL design (5 full-sib families, about 200 F2 progeny each) was produced from a cross between F0 grand-parents previously selected during two generations for a high or a low cortisol response after a standardized confinement stress. When fish were about 18 months old (near 204 g body weight), individual progeny were submitted to two successive hyper-osmotic challenges (30 ppt salinity) 14 days apart. Plasma chloride and sodium concentrations were recorded 24 h after each transfer. After the second challenge, fish were sacrificed and a gill index (weight of total gill arches corrected for body weight) was recorded. The genome scan was performed with 196 microsatellites and 85 SNP markers. Unitrait and multiple-trait QTL analyses were carried out on the whole dataset (5 families) through interval mapping methods with the QTLMap software. For post-challenge plasma ion concentrations, significant QTL (P < 0.05) were found on six different linkage groups and highly suggestive ones (P < 0.10) on two additional linkage groups. Most QTL affected concentrations of both chloride and sodium during both challenges, but some were specific to either chloride (2 QTL) or sodium (1 QTL) concentrations. Six QTL (4 significant, 2 suggestive) affecting gill index were discovered. Two were specific to the trait, while the others were also identified as QTL for post-challenge ion concentrations. Altogether, allelic effects were consistent for QTL affecting chloride and sodium concentrations but inconsistent for QTL affecting ion concentrations and gill morphology. There was no systematic lineage effect (grand-parental origin of QTL alleles) on the recorded traits. Conclusions For the first time, genomic loci associated with effects on major physiological components of osmotic adaptation to seawater in a nonmigratory fish were revealed. The results pave the way for further deciphering of the complex regulatory mechanisms underlying seawater adaptation and genes involved in osmoregulatory physiology in rainbow trout and other euryhaline fishes.

Abstract Background Rainbow trout (Oncorhynchus mykiss) are the most-widely cultivated cold freshwater fish in the world and an important model species for many research areas. Coupling great interest in this species as a research model with the need for genetic improvement of aquaculture production efficiency traits justifies the continued development of genomics research resources. Many quantitative trait loci (QTL) have been identified for production and life-history traits in rainbow trout. An integrated physical and genetic map is needed to facilitate fine mapping of QTL and the selection of positional candidate genes for incorporation in marker-assisted selection (MAS) programs for improving rainbow trout aquaculture production. Results The first generation integrated map of the rainbow trout genome is composed of 238 BAC contigs anchored to chromosomes of the genetic map. It covers more than 10% of the genome across segments from all 29 chromosomes. Anchoring of 203 contigs to chromosomes of the National Center for Cool and Cold Water Aquaculture (NCCCWA) genetic map was achieved through mapping of 288 genetic markers derived from BAC end sequences (BES), screening of the BAC library with previously mapped markers and matching of SNPs with BES reads. In addition, 35 contigs were anchored to linkage groups of the INRA (French National Institute of Agricultural Research) genetic map through markers that were not informative for linkage analysis in the NCCCWA mapping panel. The ratio of physical to genetic linkage distances varied substantially among chromosomes and BAC contigs with an average of 3,033 Kb/cM. Conclusions The integrated map described here provides a framework for a robust composite genome map for rainbow trout. This resource is needed for genomic analyses in this research model and economically important species and will facilitate comparative genome mapping with other salmonids and with model fish species. This resource will also facilitate efforts to assemble a whole-genome reference sequence for rainbow trout.

Abstract Background The development of large genomic resources has become a prerequisite to elucidate the wide-scale evolution of genomes and the molecular basis of complex traits. Linkage maps represent a first level of integration and utilization of such resources and the primary framework for molecular analyses of quantitative traits. Previously published linkage maps have already outlined the main peculiarities of the rainbow trout meiosis and a correspondance between linkage groups and chromosome arms has been recently established using fluorescent in situ hybridization. The number of chromosome arms which were covered by these maps remained unknown. Results We report an updated linkage map based on segregation analysis of more than nine hundred microsatellite markers in two doubled haploid gynogenetic lines. These markers segregated into 31 linkage groups spanning an approximate total map length of 2750 cM. Centromeres were mapped for all the linkage groups using meiogenetic lines. For each of the 31 linkage groups, the meta or acrocentric structure infered from centromere mapping was identical with those recently found with fluorescent in situ hybridization results. The present map is therefore assumed to cover the 52 chromosome arms which constitute the rainbow trout karyotype. Our data confirm the occurrence of a high interference level in this species. Homeologous regions were identified in eleven linkage groups, reflecting the tetraploid nature of the salmonid genome. The data supported the assumption that gene orders are conserved between duplicated groups and that each group is located on a single chromosome arm. Overall, a high congruence with already published rainbow trout linkage maps was found for both gene syntenies and orders. Conclusion This new map is likely to cover the whole set of chromosome arms and should provide a useful framework to integrate existing or forthcoming rainbow trout linkage maps and other genomic resources. Since very large numbers of EST containing microsatellite sequences are available in databases, it becomes feasible to construct high-density linkage maps localizing known genes. This will facilitate comparative mapping and, eventually, identification of candidate genes in QTL studies.

Inorganic colloidal cesium lead halide perovskite nanocrystals (NCs) encapsulated by surface capping ligands exhibit tremendous potential in optoelectronic applications, with their surface structure playing a pivotal role in enhancing their photophysical properties. Soy lecithin, a tightly binding zwitterionic surface-capping ligand, has recently facilitated the high-yield synthesis of stable ultraconcentrated and ultradilute colloids of CsPbX 3 NCs, unlocking a myriad of potential device applications. However, the atomic-level understanding of the ligand-terminated surface structure remains uncertain. Herein, we use a versatile solid-state nuclear magnetic resonance (NMR) spectroscopic approach, in combination with dynamic nuclear polarization (DNP) and atomistic molecular dynamics (MD) simulations, to explore the effect of lecithin on the core-to-surface structures of CsPbX 3 (X = Cl or Br) perovskites, sized from micron to nanoscale. Surface-selective (cross-polarization, CP) solid-state and DNP NMR ( 133 Cs and 207 Pb) methods were used to differentiate the unique surface and core chemical environments, while the head-groups {trimethylammonium [-N(CH 3 ) 3 + ] and phosphate (-PO 4 - )} of lecithin were assigned via 1 H, 13 C, and 31 P NMR spectroscopy. A direct approach to determining the surface structure by capitalizing on the unique heteronuclear dipolar couplings between the lecithin ligand ( 1 H and 31 P) and the surface of the CsPbCl 3 NCs ( 133 Cs and 207 Pb) is demonstrated. The 1 H- 133 Cs heteronuclear correlation (HETCOR) DNP NMR indicates an abundance of Cs on the NC surface and an intimate proximity of the -N(CH 3 ) 3 + groups to the surface and subsurface 133 Cs atoms, supported by 1 H{ 133 Cs} rotational-echo double-resonance (REDOR) NMR spectroscopy. Moreover, the 1 H- 31 P{ 207 Pb} CP REDOR dephasing curve provides average internuclear distance information that allows assessment of -PO 4 - groups binding to the subsurface Pb atoms. Atomistic MD simulations of ligand-capped CsPbCl 3 surfaces aid in the interpretation of this information and suggest that ligand -N(CH 3 ) 3 + and -PO 4 - head-groups substitute Cs + and Cl - ions, respectively, at the CsCl-terminated surface of the NCs. These detailed atomistic insights into surface structures can further guide the engineering of various relevant surface-capping zwitterionic ligands for diverse metal halide perovskite NCs.

Colloidal nuclear magnetic resonance (cNMR) spectroscopy on inorganic cesium lead halide nanocrystals (CsPbX 3 NCs) is found to serve for noninvasive characterization and quantification of disorder within these structurally soft and labile particles. In particular, we show that 133 Cs cNMR is highly responsive to size variations from 3 to 11 nm or to altering the capping ligands on the surfaces of CsPbX 3 NCs. Distinct 133 Cs signals are attributed to the surface and core NC regions. Increased heterogeneous broadening of 133 Cs signals, observed for smaller NCs as well as for long-chain zwitterionic capping ligands (phosphocholines, phosphoethanol(propanol)amine, and sulfobetaines), can be attributed to more significant surface disorder and multifaceted surfaces (truncated cubes). On the contrary, capping with dimethyldidodecylammonium bromide (DDAB) successfully reduces signal broadening owing to better surface passivation and sharper (001)-bound cuboid shape. DFT calculations on various sizes of NCs corroborate the notion that the surface disorder propagates over several octahedral layers. 133 Cs NMR is a sensitive probe for studying halide gradients in mixed Br/Cl NCs, indicating bromide-rich surfaces and chloride-rich cores. In semiconductors, exciton or charge carrier diffusivity is typically described as an inherent material property. Here, we show that the transport of excitons among CsPbBr 3 perovskite nanocrystals (NCs) depends markedly on how recently those NCs were occupied by a previous exciton. Using transient photoluminescence microscopy, we observe a striking dependence of the apparent exciton diffusivity on excitation laser power that does not arise from nonlinear exciton-exciton interactions or thermal heating. We interpret our observations with a model in which excitons cause NCs to transition to a long-lived metastable configuration that markedly increases exciton transport. The exciton diffusivity observed here (>0.15 square centimeters per second) is considerably higher than that observed in other NC systems, revealing unusually strong excitonic coupling between NCs. The finding of a persistent enhancement in excitonic coupling may help explain other photophysical behaviors observed in CsPbBr 3 NCs, such as superfluorescence, and inform the design of optoelectronic devices.

The relaxation of the above-gap ("hot") carriers in lead halide perovskites (LHPs) is important for applications in photovoltaics and offers insights into carrier-carrier and carrier-phonon interactions. However, the role of quantum confinement in the hot carrier dynamics of nanosystems is still disputed. Here, we devise a single approach, ultrafast pump-push-probe spectroscopy, to study carrier cooling in six different size-controlled LHP nanomaterials. In cuboidal nanocrystals, we observe only a weak size effect on the cooling dynamics. In contrast, two-dimensional systems show suppression of the hot phonon bottleneck effect common in bulk perovskites. The proposed kinetic model describes the intrinsic and density-dependent cooling times accurately in all studied perovskite systems using only carrier-carrier, carrier-phonon, and excitonic coupling constants. This highlights the impact of exciton formation on carrier cooling and promotes dimensional confinement as a tool for engineering carrier-phonon and carrier-carrier interactions in LHP optoelectronic materials.

Due to the growing demand for patient-friendly subcutaneous dosage forms, the ability to increasing protein solubility and stability in formulations to deliver on the required high protein concentrations is crucial. A common approach to ensure protein solubility and stability in high concentration protein formulations is the addition of excipients such as sugars, amino acids, surfactants, approved by the Food and Drug Administration. In a best-case scenario, these excipients fulfil multiple demands simultaneously, such as increasing long-term stability of the formulation, reducing protein adsorption on surfaces/interfaces, and stabilizing the protein against thermal or mechanical stress. 2-Hydroxylpropyl-β-cyclodextrin (derivative of β-cyclodextrin) holds this potential, but has not yet been sufficiently investigated for use in protein formulations. Within this work, we have systematically investigated the relevant molecular interactions to identify the potential of Kleptose ® HPB (2-hydroxylpropyl-β-cyclodextrin from Roquette Freres, Lestrem, France) as "multirole" excipient within liquid protein formulations. Based on our results three factors determine the influence of Kleptose ® HPB on protein formulation stability: (1) concentration of Kleptose ® HPB, (2) protein type and protein concentration, and (3) quality of the protein formulation. The soft lattice of lead-halide perovskite nanocrystals (NCs) allows tuning their optoelectronic characteristics via anion exchange by introducing halide salts to a solution of perovskite NCs. Similarly, cross-anion exchange can occur upon mixing NCs of different perovskite halides. This process, though, is detrimental for applications requiring perovskite NCs with different halides in close proximity. We study the effects of various stabilizing surface ligands on the kinetics of the cross-anion exchange reaction, comparing zwitterionic and ionic ligands. The kinetic analysis, inspired by the "cage effect" for solution reactions, showcases a mechanism where the surface capping ligands act as anion carriers that diffuse to the NC surface, forming an encounter pair enclosed by the surrounding ligands that initiates the anion exchange process. The zwitterionic ligands considerably slow down the cross-anion exchange process, and while they do not fully inhibit it, they confer improved stability alongside enhanced solubility relevant for various applications.

Semiconductor quantum dots have long been considered artificial atoms, but despite the overarching analogies in the strong energy-level quantization and the single-photon emission capability, their emission spectrum is far broader than typical atomic emission lines. Here, by using ab-initio molecular dynamics for simulating exciton-surface-phonon interactions in structurally dynamic CsPbBr 3 quantum dots, followed by single quantum dot optical spectroscopy, we demonstrate that emission line-broadening in these quantum dots is primarily governed by the coupling of excitons to low-energy surface phonons. Mild adjustments of the surface chemical composition allow for attaining much smaller emission linewidths of 35-65 meV (vs. initial values of 70-120 meV), which are on par with the best values known for structurally rigid, colloidal II-VI quantum dots (20-60 meV). A comprehensive microscopic description of thermally induced distortions in lead halide perovskites is crucial for their realistic applications, yet still unclear. Here, we quantify the effects of thermal activation in CsPbBr 3 nanocrystals across length scales with atomic-level precision, and we provide a framework for the description of phase transitions therein, beyond the simplistic picture of unit-cell symmetry increase upon heating. The temperature increase significantly enhances the short-range structural distortions of the lead halide framework as a consequence of the phonon anharmonicity, which causes the excess free energy surface to change as a function of temperature. As a result, phase transitions can be rationalized via the soft-mode model, which also describes displacive thermal phase transitions in oxide perovskites. Our findings allow to reconcile temperature-dependent modifications of physical properties, such as changes in the optical band gap, that are incompatible with the perovskite time- and space-average structures.

Nonresonant optical driving of confined semiconductors can open up exciting opportunities for experimentally realizing strongly interacting photon-dressed (Floquet) states through the optical Stark effect (OSE) for coherent modulation of the exciton state. Here we report the first room-temperature observation of the Floquet biexciton-mediated anomalous coherent excitonic OSE in CsPbBr 3 quantum dots (QDs). Remarkably, the strong exciton-biexciton interaction leads to a coherent red shift and splitting of the exciton resonance as a function of the drive photon frequency, similar to Autler-Townes splitting in atomic and molecular systems. The large biexciton binding energy of ∼71 meV and exciton-biexciton transition dipole moment of ∼25 D facilitate the hallmark observations, even at large detuning energies of >300 meV. This is accompanied by an unusual crossover from linear to nonlinear fluence dependence of the OSE as a function of the drive photon frequency. Our findings reveal crucial information on the unexplored many-body coherent interacting regime, making perovskite QDs suitable for room temperature quantum devices.

Fundamental photophysical behavior in CsPbBr 3 nanocrystals (NCs), especially at low temperatures, is under active investigation. While many studies have reported temperature-dependent photoluminescence, comparatively few have focused on understanding the temperature-dependent absorption spectrum. Here, we report the temperature-dependent (35-300 K) absorption and photoluminescence spectra of zwitterionic ligand-capped CsPbBr 3 NCs with four different edge lengths ( d = 4.9, 7.2, 8.1, and 13.2 nm). The two lowest-energy excitonic transitions are quantitatively modeled over the full temperature range within the effective mass approximation considering the quasi-cubic NC shape and nonparabolicity of the electronic bands. Significantly, we find that the effective dielectric constant determined from the best fit model parameters is independent of temperature. Moreover, we observe a temperature-dependent Stokes shift that saturates at a finite value of Δ ≈ 10 meV at low temperatures for d = 7.2 nm NCs, which is absent in bulk CsPbBr 3 films. Overall, these observations highlight differences between the temperature-dependent dielectric behavior of NC and bulk perovskites and point to the need for a more unified theoretical understanding of absorption and emission in halide perovskites.
