We explored links between competitive authoritarianism and populism in Serbia under Vučić via mixed methods. We conducted a quantitative content analysis of 228 political cartoons (2013--2017) by Dušan Petričić, a leading cartoonist and government critic, and qualitative visual and contextual analysis of four cartoons that reflected key themes--media freedom violations, unfair political competition, and populist and abusive rule. We found that populism appeared as the 'soft' face of competitive authoritarianism, tolerable to domestic audiences and acceptable to influential international actors because of its reliance on more informal and sophisticated and less repressive forms of authoritarian manipulation. Serbia was highly polarized ahead of the 2010 Pride parade in Belgrade, an event marred by violence. It is often assumed that the media played a negative role by uncritically giving voice to anti-LGBT activists. To investigate this claim, we conducted a quantitative and qualitative analysis of media reporting as well as 25 in-depth interviews with journalists. We conclude that the media denied extremists direct access to the media. However, due to the high polarization, the media did empower nonviolent opponents of the parade, and hence indirectly contributed to the stereotypical and sometimes derogatory representation of the LGBT population. The 2010 Belgrade Pride Parade represents a critical moment in the story of Serbia's democratisation process and highlights the threat that right-wing extremism poses to democratic rights and personal freedoms. Through a focus on patterns of visibility and visuality in the coverage of different protagonists in the streets of Belgrade, we explore the ways in which distinct communities perform their affinities, their right to be seen in public spaces, and rejection of 'the other'. We conduct a visual framing analysis across four news programmes (RTS, Prva TV, TV B92 and Pink TV), emphasising the stylistic-semiotic choices which work to construct the contested spaces of the city. In shifting attention to how the news images work to create the spaces of political 'appearance' and the potentials for political agency through mediated visibility, the article explores the uneasy ambivalence of the democratisation process for authorities and the resulting marginalisation of the lesbian, gay, bisexual and transgender community in news coverage. In this article, we use the hierarchy-of-influences model as a framework for examining the ways in which media owners, managers and journalists perceive the influence exerted on their work during 12-year democratic transition in Serbia. We aim to explain how factors perceived as influential at the highest system level gradually transfer and relate to the factors on the subsumed levels. Using the concepts such as corruption and the culture of corruption to interpret hierarchy between different levels of influence on transitional journalism, we argue that coupling extra-media actors at the system level can be considered corruption – understood as abuse of power for personal gain or benefit of the aligned group – which translates to all other levels of influence. The aim of this paper is to examine in which way the following factors influence TV owners‟ strategies for the development of television as a business in the turbulent media market of Serbia: 1) the size of the news market and local media environment, 2) the response to technological changes, 3) economic status of the television outlet, and 4) clientelism. For the purpose of this case study, in-depth interviews were conducted with the owners of six local commercial TV stations located in different regions in Serbia. The results show that instead of developing professional resources for a highly competitive media environment, TV owners are fighting for bare survival. However, their strategies for the development of television as a business can be divided into two camps: one group expects the help of the state to overcome financial constraints, while the other seeks to develop e-business and web advertising as models of sustainable growth on the market. The mouse is a suitable experimental model to study the biology of mesenchymal stem cells (MSCs), as well as to be used in biocompatibility studies and tissue engineering models. However, the isolation and purification of murine MSCs is far more challenging than their counterparts from other species. In this study, we isolated, expanded and characterized mouse MSCs from bone marrow (BM-MSCs). Additionally, we analyzed the effects of two regulatory molecules, interleukin 17 (IL-17) and basic fibroblast growth factor (bFGF), on BM-MSCs growth and elucidated the signaling pathways involved. The results revealed that IL-17 increased the frequency of colony-forming units fibroblast (CFU-F) as well as the BM-MSCs proliferation in a dose-dependent manner, while bFGF supplementation had no significant effect on CFU-F frequency but induced an increase in cell proliferation. Their combined usage did not produce additive effects on BM-MSCs proliferation and even induced reduction in the number of CFU-F. Also, the involvement of both p38 and extracellular signal-regulated kinase (ERK) mitogen-activated protein kinases (MAPKs) signaling in proliferative activity of IL-17 and bFGF on murine BM-MSCs and, moreover, the increased co-activation of a common signaling molecule, p38 MAPK, were demonstrated. Together, the data presented highlighted the role of IL-17 and bFGF in murine BM-MSCs proliferation and pointed to the complexity and specificity of the signaling networks leading to MSCs proliferation in response to different regulatory molecules. Abstract: Syphacia obvelata is a rodent nematode parasite with high prevalence in laboratory mice. In our previous work we have demonstrated that this gut-dwelling helminth induces significant hematopoietic changes, characterized by increased myelopoiesis and erythropoiesis in infected animals, and accompanied with altered reactivity of bone marrow hematopoietic progenitors to interleukin (IL)-17. In this study we extended these investigations by demonstrating that naturally acquired S. obvelata infection induces significant alterations in murine bone marrow cells manifested at the molecular level. Namely, S. obvelata infection induced sustained phosphorylation of the members of three major groups of distinctly regulated mitogen-activated protein kinases (MAPKs), the p38, the c-Jun amino-terminal kinase (JNK) and the extracellular signal-regulated kinase (ERK), as well as enhanced expression of mRNA for the inducible nitric oxide synthase (iNOS) in the bone marrow cells of infected animals. Furthermore, the infection interfered with the IL-17-mediated effects in bone marrow cells, since in normal mice IL-17 significantly enhanced phosphorylation of p38 MAPK and upregulated the expression of iNOS and the constitutive, endothelial (e)NOS mRNA, while in S. obvelata-infected animals IL-17 did not influence the MAPKs activation, but markedly down-regulated the expression of both NOS isoforms. The data obtained demonstrating that S. obvelata is able to manipulate signal transduction pathways in the hosts'' bone marrow cells, pointed to the multiple layers of immunomodulatory ability of this pinworm parasite and highlighted the importance of working under pinworm-free conditions when using experimental murine models for immunohematopoietic investigations. The effects of interleukin (IL)-17 on nitric oxide (NO) synthase (NOS) expression, as well as the participation of mitogen-activated protein kinases (MAPKs) in IL-17-mediated effects were examined in murine bone marrow cells. The results demonstrated the ability of IL-17 to upregulate the expression of mRNA for both inducible NOS and constitutive, endothelial NOS isoforms, as well as to enhance the phosphorylation of p38 MAPK. Moreover, both the NOS-inducing effect of IL-17 and the in vitro IL-17-mediated inhibition colony forming unit-erythroid (CFU-E) growth were dependent on p38 MAPK activity. The data demonstrating that the in vivo reducing effect of IL-17 on bone marrow CFU-E was prevented by co-treatment with the NOS inhibitor Nw-nitro-l-arginine methyl ester hydrochloride (L-NAME), implied that this effect is mediated through NOS activation. Besides revealing a link between the IL-17, NO, and haematopoiesis, data presented gave an insight into the mechanisms by which IL-17 exerts its modulatory effects on bone marrow cells. Through the use of effectively unlimited energy from the environment, contemporary heat pump installations have proven to be a highly efficient means for satisfying both heating and cooling needs. As such, they are often regarded as a key driver in the electrification process of the thermal domain. Keeping in mind the significant share of the thermal demand in building energy requirements, this aspect in particular is projected to have a great impact on the overall energy efficiency improvement of buildings and increase of demand-side flexibility. The latter point is considered crucial for improving the precarious balance against the contemporary intermittent power supply obtained from a variety of renewable sources. However, in the process of increasing their efficiency, heat pump systems are becoming significantly more complex with combinations of various different sources of energy from the ground, air and the Sun. By extension, the complexity of managing their operation has also increased and proves to be a challenge even for experienced plant operators. In order to aid in this regard, relevant literature suggests that optimization algorithms can be considered as a viable tool. This paper explores the limited extent of existing state-of-the-art approaches for hybrid heat pump control via metaheuristic optimization and broadens the subject matter with a comparative study that analyzes and benchmarks several metaheuristic approaches found in the literature on similar problems. Abstract: Interleukin-17 belongs to a family of pro-inflammatory cytokines with pleiotropic effects, which can be associated with several inflammatory diseases of the muscle tissue. Although elevated levels of interleukin-17 have been described in inflammatory myopathies, its role in muscle homeostasis remains to be elucidated. The requirement of the urokinase type plasminogen activator in skeletal myogenesis was recently demonstrated in vivo and in vitro, suggesting its involvement in the regulation of extracellular matrix remodeling, cell migration and myoblast fusion. Our previous results have demonstrated that interleukin-17 inhibits myogenic differentiation of C2C12 myoblasts in vitro concomitantly with the inhibition of cell migration. However, the involvement of urokinase type plasminogen activator in interleukin-17-inhibited myogenesis and migration remained to be analyzed. Therefore, the effect of interleukin-17 on the production of urokinase type plasminogen activator by C2C12 myoblasts was determined in the present study. Our results demonstrated that interleukin-17 strongly inhibits urokinase type plasminogen activator expression during myogenic differentiation. This reduction of urokinase type plasminogen activator production corresponded with the inhibition of cell migration by interleukin-17. Activation of p38 signaling pathway elicited by interleukin-17 mediated the inhibition of both urokinase type plasminogen activator expression and cell migration. Additionally, IL-17 inhibited C2C12 cells migration by causing the cells to reorganize their cytoskeleton and lose polarity. Therefore, our results suggest a novel mechanism by which interleukin-17 regulates myogenic differentiation through the inhibition of urokinase type plasminogen activator expression and cell migration. Abstract: The present study evaluated the role of interleukin (IL) 17 in multilineage commitment of C2C12 myoblastic cells and investigated associated signaling pathways. The results concerning the effects on cell function showed that IL-17 inhibits the migration of C2C12 cells, while not affecting their proliferation. The data regarding the influence on differentiation demonstrated that IL-17 inhibits myogenic differentiation of C2C12 cells by down-regulating the myogenin mRNA level, myosin heavy chain expression and myotube formation, but promotes their osteogenic differentiation by up-regulating the Runt-related transcription factor 2 mRNA level, cyclooxygenase-2 expression and alkaline phosphatase activity. IL-17 exerted these effects by activating ERK1,2 mitogen activated protein kinase signaling pathway, which in turn regulated the expression of relevant genes and proteins to inhibit myogenic differentiation and induce osteogenic differentiation. Additional analysis showed that the induction of osteogenic differentiation by IL-17 is independent of BMP signaling. The results obtained demonstrate the potential of IL-17 not only to inhibit the myogenic differentiation of C2C12 myoblasts but also to convert their differentiation pathway into that of osteoblast lineage providing new insight into the capacities of IL-17 to modulate the differentiation commitment. Transforming growth factor-beta (TGF-β1) is a potent inductor of matrix metalloproteinase-9 (MMP-9) in transformed cells. Recently, Ski-interacting protein (SKIP) has been described as a regulator of TGF-β1 signal transduction, but its role in the induction of cell malignance by TGF-β1 has not been fully elucidated so far. In the present study, we analyzed the role of SKIP on TGF-β1-induced MMP-9 production. Mouse transformed keratinocytes (PDV) were stably transfected with SKIP antisense construct. We observed that SKIP depletion provoked an enhancement in the expression of MMP-9 in response to TGF-β1 treatment. The downregulation of SKIP produced an enhancement in TGF-β1-activated ERK1,2 MAP kinase as well as increased transactivation of downstream Elk1 transcription factor. ?e increased MMP-9 production in response to TGF-β1 was dependent of MAPK activation as PD98059, an MEK inhibitor, reduced MMP-9 expression in SKIP antisense transfected cells. Thus, we propose SKIP as a regulatory protein in TGF-β1-induced MMP-9 expression acting by controlling ERK1,2 signaling in transformed cells. [ABSTRACT FROM AUTHOR]

The essence of this study is synthesis of a newly developed and efficient heterogeneous catalyst based on aluminum oxide support promoted with potassium iodide (KI) as active dopant responsible for pronounced catalyst activity. Modified sol–gel, reflux and hydrothermal method were used for synthesis of aluminum-oxide support using aluminum isopropoxide as a precursor. The γ-Al 2 O 3 support and KI/γ-Al 2 O 3 catalyst samples were examined by various experimental techniques. Effects of several operational parameters were considered and those are: methanol to sunflower oil molar ratio, catalyst loadings, reaction time and reaction stirring rate. The obtained results indicated that modified sol–gel method for synthesis of γ-Al 2 O 3 catalyst support is the most favorable for obtaining the catalyst with superior catalytic performances in transesterification of sunflower oil. A maximum conversion (triglycerides to methyl esters) of 99.99% was achieved using the SG-KI/γ-Al 2 O 3 -770 catalyst under following optimal process parameters: 15:1 methanol to sunflower oil molar ratio, 2.5 wt% catalysts loading, 600 rpm stirring rate and 4 h of the reaction run. Abstract: Monophase silver-doped hydroxyapatite (Ag x Ca10−x (PO4)6(OH)2; 0.002≤ x ≤0.04) nanoparticles were prepared using a neutralization method and investigated with respect to potential medical applications. This method consists of dissolving Ag2O in solution of H3PO4, and the slow addition to suspension of Ca(OH)2 was applied for the purpose of homogenous distribution of silver ions. Characterization studies from XRD, TEM and FTIR spectra showed that obtained crystals are monophase hydroxyapatites and that particles of all samples are of nano size, with average length of 70nm and about 15–25nm in diameter. Antimicrobial studies have demonstrated that all silver-doped hydroxyapatite samples exhibit excellent antimicrobial activity in vitro against the following pathogens: Staphylococcus aureus, Escherichia coli and Candida albicans. The hydroxyapatite sample with the highest content of silver has shown the highest antimicrobial activity; killed all cells of E. coli and brought to more than 99% reduction in viable counts of S. aureus and C. albicans. The atomic force microscopic studies illustrate that silver-doped hydroxyapatite sample causes considerable morphological changes of microorganism cells which might be the cause of cells’ death. Hemolysis ratios of the silver-doped hydroxyapatite samples were below 3%, indicating good blood compatibility and that are promising as biomaterials. [ABSTRACT FROM AUTHOR]

Low O2 concentration (1%) favors the self-renewal of hematopoietic stem cells and inhibits committed progenitors (CFC). Since IL-6 influences both stem cells and committed progenitors at 20% O2, we studied its effects in cultures at 1% O2. The pre-CFC activity in Lin- population of mouse bone marrow was analyzed following 10 days of serum-free culture in medium (LC1) supplemented with IL-3 with and without IL-6, at 20 and 1% O2 and phenotypic differentiation and proliferative history monitored. The IL-6 receptor expression and initiation of VEGF-A synthesis were also investigated. At 20% O2, the effects of IL-6 on pre-CFC were negligible but effects on CFC were apparent; conversely, at 1% O2, the IL-6 enhances activity of pre-CFC but not of CFC. Unlike at 20% O2, at 1% O2 a subpopulation of cells remained Lin- in spite of extensive proliferation. However, the absolute number of Lin- cells, did not correlate with pre-CFC activity. A relative increase in VEGF transcripts at 1% O2 in presence of IL-3 alone was enhanced by the addition of IL-6. IL-6 enhanced pre-CFC activity at 1% O2 and this was correlated to the induction of VEGF. These data reinforce the concept that physiologically low oxygenation of bone marrow is a regulator of stem cell maintenance. Since the 20% O2 does not exist in tissues in vivo, further studies in vitro at lower O2 concentrations should revise our knowledge relating to cytokine effects on stem and progenitor cells. J. Cell. Abstract: Pinworm parasites commonly infect laboratory mice with high prevalence even in well-managed animal colonies. Although often considered as irrelevant, these parasites if undetected may significantly interfere with the experimental settings and alter the interpretation of final results. There are a few reports documenting the effects of pinworms on research and the effects of pinworms on the host hematopoiesis have not yet been investigated. In this study we examined the changes within various hematopoietic cell lineages in the bone marrow, spleen, peripheral blood and peritoneal space during naturally acquired Syphacia obvelata infection in inbred CBA mice. The data obtained showed significant hematopoietic alterations, characterized by increased myelopoiesis and erythropoiesis in S. obvelata-infected animals. In order to additionally evaluate if this pinworm infection modifies hematopoietic cells'' reactivity, we examined the effect of murine interleukin-17, T cell-derived cytokine implicated in the regulation of hematopoiesis and inflammation, on the growth of bone marrow progenitor cells and demonstrated that bone marrow myeloid and erythroid progenitors from S. obvelata-infected mice displayed altered sensitivity to IL-17 when compared to non-infected controls. Taken together the alterations presented pointed out that this rodent pinworm is an important environmental agent that might significantly modify the hosts'' hematopoietic response, and therefore interfere with the experimental settings and alter the interpretation of the final results. However, the results obtained also contributed new data concerning the activity of IL-17 on bone marrow hematopoietic cells, supporting our previous reports that depending on physiological/pathological status of the organism IL-17 exerts differential effects on the growth of progenitor cells.
