Neuroinflammation is a common pathological hallmark of various neurological disorders affecting the central nervous system (CNS) including Parkinson’s disease (PD), multiple system atrophy (MSA) and multiple sclerosis (MS). MS and the animal model experimental autoimmune encephalomyelitis (EAE) are prototypical neuroinflammatory demyelinating diseases of the CNS, which can cause severe neurological dysfunction. Despite intense research, the underlying pathomechanisms still remain incompletely understood. Traditionally, MS and EAE are considered as autoimmune disorders characterized by the activation of CNS-reactive immune cells in the periphery. Infiltration of these cells into the CNS facilitates the damage of myelin and oligodendrocytes, ultimately resulting in axonal loss and extensive tissue injury. However, in MS, this view is challenged by another model proposing primary damage of oligodendroglia and neurons as initial event in the disease pathogenesis. In this thesis, the immunopathological as well as the primary degenerative component in neuroinflammatory demyelination is addressed by investigating the contribution of two molecules associated with immune system homeostasis and neurodegeneration, respectively. Lymphocyte migration is critical for functional immune responses and its dysregulation contributes to the development of various inflammatory diseases. Mucosal addressin cell adhesion molecule (MAdCAM) 1 is mainly expressed in the intestinal tract where it facilitates the entry of lymphocytes. Recently, the gut has been identified as an important modulator of the immune system and is supposed to be involved in the development of autoimmune diseases such as MS. Therefore, the first hypothesis of this thesis is that MAdCAM-1-mediated lymphocyte homing into the intestinal tract plays a role in immune-mediated neuroinflammatory processes. To test this, the murine EAE model was employed and interaction of MAdCAM-1 with lymphocytes was inhibited using a knock out approach (MAdCAM-1-KO). Genetic ablation of MAdCAM-1 rendered mice less susceptible to the development of EAE and resulted in an ameliorated disease course. This was accompanied by reduced immune cell infiltration into the spinal cord as well as into the intestinal tract, while peripheral immune responses were not affected. Cell tracking experiments indicated that MAdCAM-1 is also required for trafficking of myelin-activated lymphocytes to the gut. However, MAdCAM-1-KO mice were equally susceptible to adoptive transfer-EAE. In neuroinflammatory demyelinating diseases, persistent demyelination triggers axonal loss, which accounts for irreversible neurological disability. Additionally, chronic demyelination is often accompanied by oligodendrocyte injury and wide-spread CNS degeneration. Alpha-Synuclein (aSyn) is a protein, which has been associated with neuroinflammatory processes in neurodegenerative diseases, in particular PD and MSA. Considering that MSA shares some features with autoimmune-mediated neuroinflammatory demyelinating diseases, the second part of this work aims at investigating the role of aSyn in de- and regenerative processes during (immune-mediated) neuroinflammation. For this purpose, the effect of genetic aSyn deletion was studied in different animal models of demyelination. During acute EAE, aSyn-deficient mice showed an earlier disease onset accompanied by pronounced activation and interleukin-2-mediated proliferation of T helper (Th) 1 cells. In chronic EAE, aSyn-deficient mice exhibited an attenuated disease course with a concomitant reduction in demyelination, axonal loss and immune cell infiltration in spinal cord lesions. Finally, the impact of aSyn on de- and remyelination in the absence of peripheral inflammatory processes was assessed using the cuprizone model. De- as well as regenerative responses in the CNS following cuprizone treatment remained majorly unaffected by aSyn deficiency. In conclusion, the results obtained within the scope of this work demonstrate a MAdCAM-1-associated modulation of CNS autoimmunity within the intestinal tract and give new insights into the functional role of the gut in neuroinflammation. Importantly, these data reinforce the concept that the intestine is critically involved in the development of autoimmune reactions and constitutes an important site of immune cell regulation. Furthermore, aSyn was identified as a modulator of immunological processes during early neuroinflammation, particularly by acting as regulator of Th1 cell responses. Additionally, findings presented here implicate aSyn in the modulation of degenerative processes during chronic inflammatory demyelination, thus establishing aSyn as potential bridging element between classical neurodegenerative disorders and neuroinflammatory demyelinating diseases. Die Neuroinflammation ist ein gemeinsames Kennzeichen vieler neurologischer Erkrankungen, die das zentrale Nervensystem (ZNS) betreffen. Hierzu zählen unter anderem die Parkinson-Erkrankung, die Multisystematrophie (MSA) und die Multiple Sklerose (MS). Letztere, sowie das Tiermodell der experimentellen autoimmunen Encephalomyelitis, sind prototypische neuroinflammatorische Entmarkungserkrankungen, welche schwere neurologische Ausfälle zur Folge haben können. Trotz intensiver Forschung sind die zugrundeliegenden Pathomechanismen noch nicht vollständig aufgeklärt. Traditionell werden die MS und die EAE als Autoimmunerkrankungen angesehen, welche durch die Aktivierung von ZNS-reaktiven Immunzellen in der Peripherie verursacht wird. Die Infiltration dieser Zellen in das ZNS führt zu einer Schädigung von Myelin und Oligodendrozyten, was letztendlich in Neurodegeneration und beträchtlichen Gewebeschäden resultiert. Bei der Pathogenese der MS wird diese Ansicht allerdings von einem anderen Modell in Frage gestellt, welches die Schädigung von oligodendroglialen Zellen und Neuronen als initiales Ereignis in der Krankheitsentstehung auffasst. In dieser Arbeit wird sowohl die immunpathologische, als auch die primär degenerative Komponente der neuroinflammatorischen Demyelinisierung aufgegriffen und der Beitrag zweier Moleküle, die mit dem Gleichgewicht des intestinalen Immunsystems beziehungsweise mit Neurodegeneration in Zusammenhang stehen, erforscht. Die Wanderung von Lymphozyten durch den Organismus ist für die Ausbildung einer funktionellen Immunantwort von großer Bedeutung und eine Fehlregulation wird mit der Entstehung einer Reihe von entzündlichen Erkrankungen in Verbindung gebracht. Das Mukosale Addressin Zell-Adhäsionsmolekül (MAdCAM) 1 wird hauptsächlich im Darm exprimiert, wo es den Eintritt von Lymphozyten ermöglicht. Kürzlich ist der Verdauungstrakt als wichtiger Modulator des Immunsystems erkannt worden und es gibt vermehrt Hinweise darauf, dass dieser an der Entstehung von Autoimmunerkrankungen wie der MS beteiligt ist. Daraus ergibt sich die erste Hypothese dieser Arbeit, dass die von MAdCAM-1 vermittelte Wanderung von Immunzellen in den Gastrointestinaltrakt bei neuroinflammatorischen Prozessen eine Rolle spielt. Um dies zu testen, wurde das EAE-Maus-Modell genutzt und die Interaktion von MAdCAM-1 mit Lymphozyten durch eine Deletion des MAdCAM-1-Gens (MAdCAM-1-KO) verhindert. Diese Gendeletion führte bei MAdCAM-1-KO-Mäusen zu einer verringerten Anfälligkeit für die Entstehung von EAE und resultierte in einem milderen Krankheitsverlauf. Diese Beobachtungen gingen mit einer reduzierten Infiltration von Immunzellen in das Rückenmark und den Darm einher. Die Immunantwort in der Peripherie dagegen war im Vergleich zu Kontrollmäusen nicht verändert. Des Weiteren konnte hier gezeigt werden, dass MAdCAM-1 auch für die Migration von Myelin-aktivierten Lymphozyten in den Gastrointestinaltrakt benötigt wird. Jedoch waren Mäuse ohne funktionelles MAdCAM-1-Gen genauso empfänglich für eine passiv induzierte EAE wie entsprechende Wildtyp-Mäuse. Bei neuroinflammatorisch bedingten Entmarkungserkrankungen führt eine dauerhafte Demyelinisierung zu einem Verlust von Axonen, welcher für irreversible neurologische Beeinträchtigungen verantwortlich gemacht wird. Zusätzlich sind im Zuge chronischer Entmarkung häufig ein Verlust von Oligodendrozyten und eine weitreichende Degeneration des ZNS zu beobachten. Das Protein alpha-Synuklein (aSyn) wird mit neuroinflammatorischen Prozessen bei neurodegenerativen Erkrankungen, insbesondere der Parkinson-Erkrankung und der MSA, in Verbindung gebracht. Unter Berücksichtigung, dass die MSA einige Gemeinsamkeiten mit autoimmun-vermittelten neuroinflammatorischen Entmarkungserkrankungen aufweist, befasst sich der zweite Teil dieser Arbeit mit der Rolle von aSyn bei de- und regenerativen Vorgängen während solcher (inflammatorischer) Entmarkungsprozesse. Hierzu wurde der Einfluss der Deletion des aSyn-Gens in verschiedenen Tiermodellen untersucht. Während der akuten EAE wiesen Mäuse ohne aSyn-Gen einen früheren Krankheitsbeginn auf, welcher mit einer verstärkten Aktivierung und einem Interleukin-2-vermittelten Anstieg der Proliferation von T-Helfer (TH) 1-Zellen verknüpft war. Im Verlauf der chronischen EAE zeigten aSyn-defiziente Mäuse einen milderen Krankheitsverlauf mit gleichzeitig verringerter Demyelinisierung, reduziertem Axonverlust und geringerer Immunzellansammlung in Rückenmarksläsionen. Schließlich wurde der Einfluss von aSyn auf die De- und Remyelinisierung im Cuprizone-Modell, in Abwesenheit von peripheren Entzündungsprozessen, bestimmt. Hier wurden in aSyn-defizienten Mäusen keine wesentlichen Veränderungen bei de- und regenerativen Vorgängen im ZNS festgestellt. Zusammenfassend kann gesagt werden, dass die im Rahmen dieser Arbeit erhobenen Daten eine mit MAdCAM-1 verknüpfte Modulation der ZNS-Autoimmunität innerhalb des Gastrointestinaltrakts aufzeigen und neue Einsichten in die funktionelle Rolle dieses Organs bei der Neuroinflammation gewährleisten. Insgesamt bestärken die hier gewonnenen Ergebnisse das Konzept, dass der Darm wesentlich an der Entstehung von Autoimmunreaktionen beteiligt ist und einen wichtigen Ort der Immunzellregulation darstellt. Des Weiteren wurde aSyn als Modulator von immunologischen Vorgängen während früh auftretender Neuroinflammation, speziell als ein Regulator von TH1-Zellen, identifiziert. Die vorliegenden Resultate deuten darüber hinaus eine Rolle bei der Steuerung von degenerativen Prozessen während der chronisch-entzündlichen Entmarkung an. Dies lässt vermuten, dass aSyn eine Verknüpfung zwischen klassischen neurodegenerativen Erkrankungen und neuroinflammatorischen Entmarkungserkrankungen darstellen könnte.

Background Increased α-synuclein immunoreactivity has been associated with inflammatory activity in multiple sclerosis (MS) lesions, but the function of α-synuclein in neuroinflammation remains unknown. The aim of this study was to examine the role of α-synuclein in immunological processes in murine experimental autoimmune encephalomyelitis (EAE) as a model of MS. Findings We studied EAE in wildtype (aSyn+/+) and α-synuclein knockout (aSyn−/−) mice on a C57BL/6N background. In the spleen and spinal cord of aSyn+/+ mice, we observed a gradual reduction of α-synuclein expression during EAE, starting already in the pre-symptomatic disease phase. Compared to aSyn+/+ mice, aSyn−/− mice showed an earlier onset of symptoms but no differences in symptom severity at the peak of disease. Earlier symptom onset was accompanied by increased spinal cord infiltration of CD4+ T cells, predominantly of interferon-γ-producing T helper 1 (Th1) cells, and reduced infiltration of regulatory T cells, whereas antigen-presenting cells were unaltered. Pre-symptomatically, aSyn−/− mice exhibited hyperproliferative CD4+ splenocytes consistent with increased splenic interleukin-2 mRNA expression, resulting in increased numbers of Th1 cells in the spleen at the onset of symptoms. Conclusions Our findings indicate a functional role of α-synuclein in early EAE by increasing Th1 cell-mediated immune response.

Background ApoE4, the most significant genetic risk factor for late-onset Alzheimer's disease (AD), sequesters a pro-synaptogenic Reelin receptor, Apoer2, in the endosomal compartment and prevents its normal recycling. In the adult brain, Reelin potentiates excitatory synapses and thereby protects against amyloid-β toxicity. Recently, a gain-of-function mutation in Reelin that is protective against early-onset AD has been described. Alternative splicing of the Apoer2 intracellular domain (Apoer2-ICD) regulates Apoer2 signaling. Splicing of juxtamembraneous exon 16 alters the g-secretase mediated release of the Apoer2-ICD as well as synapse number and LTP, and inclusion of exon 19 ameliorates behavioral deficits in an AD mouse model. The Apoer2-ICD has also been shown to alter transcription of synaptic genes. However, the role of Apoer2 splicing for transcriptional regulation and its role in AD pathogenesis is unknown. Methods To assess in vivo mRNA-primed ribosomes specifically in hippocampi transduced with Apoer2-ICD splice variants, we crossed wild-type, cKO, and Apoer2 cleavage-resistant mice to a Cre-inducible translating ribosome affinity purification (TRAP) model. This allowed us to perform RNA-Seq on ribosome-loaded mRNA harvested specifically from hippocampal cells transduced with Apoer2-ICDs. Results Across all conditions, we observed ~ 4,700 altered ribosome-associated transcripts, several of which comprise key synaptic components such as extracellular matrix and focal adhesions with concomitant perturbation of critical signaling cascades, energy metabolism, translation, and apoptosis. We further demonstrated the ability of the Apoer2-ICD to rescue many of these altered transcripts, underscoring the importance of Apoer2 splicing in synaptic homeostasis. A variety of these altered genes have been implicated in AD, demonstrating how dysregulated Apoer2 splicing may contribute to neurodegeneration. Conclusions Our findings demonstrate how alternative splicing of the APOE and Reelin receptor Apoer2 and release of the Apoer2-ICD regulates numerous ribosome-associated transcripts in mouse hippocampi in vivo . These transcripts comprise a wide range of functions, and alterations in these transcripts suggest a mechanistic basis for the synaptic deficits seen in Apoer2 mutant mice and AD patients. Our findings, together with the recently reported AD-protective effects of a Reelin gain-of-function mutation in the presence of an early-onset AD mutation in Presenilin-1, implicate the Reelin/Apoer2 pathway as a target for AD therapeutics.
