Zorunlu hücre içi parazitler olan mikrosporidia türleri, özellikle immün sistemi baskılanmış hastalarda ciddi enfeksiyonlara yol açan en önemli fırsatçı patojenlerdendir. İnsanı enfekte eden 14 mikrosporidia türü arasında Enterocytozoon bieneusi ve Encephalitozoon intestinalis en yaygın olanlarıdır. Bu çalışmada, kemoterapi alan kanserli hastalarda E.intestinalis ve E.bieneusi prevalansının, immünofloresan antikor ve konvansiyonel boyama yöntemleri ile araştırılması amaçlanmıştır. Çalışmaya, hastanemizin onkoloji ve hematoloji kliniklerinde takip edilen 93 kanserli hasta (58 erkek, 35 kadın) ile kontrol olarak 30 (13 erkek, 17 kadın) sağlıklı gönüllüden alınan toplam 123 dışkı örneği dahil edilmiştir. Hastaların 51 (%55)’inde ishal şikayeti mevcuttur. Tüm örneklerde E.intestinalis ve E.bieneusi varlığı, monoklonal antikorların kullanıldığı ticari bir immünofloresan antikor yöntemi (IFA-MAb; Bordier Affinity Products, İsviçre) ile araştırılmış; ayrıca 50 örnek modifiye trikrom, aside dirençli trikrom ve kalkoflor boyama yöntemleri ile de incelenmiştir. IFA-MAb yöntemi ile hastaların 43 (%46.2)’ünde E.intestinalis, 9 (%9.7)’unda E.bieneusi ve 13 (%14)’ünde karışık enfeksiyon olmak üzere toplam 65 (%69.9) olguda pozitiflik saptanmış; kon- trol grubunda ise 2 (%6.7) E.intestinalis, 1 (%3.3) E.bieneusi ve 2 (%6.7) karışık enfeksiyon olmak üzere toplam 5 (%16.7) pozitif sonuç alınmıştır. Hasta ve kontrol grubunun pozitiflik oranları arasındaki fark istatistiksel olarak anlamlı bulunmuştur (p< 0.05). İshalli hastalardan %68.6 (35/51)’sının mikrosporidia ile enfekte olduğu izlenmiş; microsporidia pozitiflik oranı ishali olan ve olmayan (%48.6) olgular arasında anlamlı fark göstermiştir (p< 0.05). Tüm yöntemlerin birlikte uygulandığı 50 örnek değerlendirildiğinde; microsporidia pozitiflik oranları IFA-MAb yöntemi ile %66 (n= 33), modifiye trikrom boyama ile %34 (n= 17), aside dirençli trikrom boyama ile %24 (n= 12) ve kalkoflor boyama ile %42 (n= 21) olarak belirlenmiştir. Bu veriler, mikrosporidia tanısında konvansiyonel boyama yöntemleri ile birlikte IFA-MAb yönteminin kullanılmasının duyarlılığı artıracağını göstermektedir. Sonuç olarak çalışmamızda, kemoterapi alan kanserli hastalarda E.intestinalis ve E.bieneusi prevalansının oldukça yüksek (%69.9) olduğu saptanmış; bu hastaların düzenli olarak mikrosporidian patojenler açısından taranmasının uygun olacağı düşünülmüştür., Microsporidia species are obligate intracellular parasites and constitute one of the most important opportunistic pathogens that can cause severe infections especially in immunocompromised patients. Enterocytozoon bieneusi and Encephalitozoon intestinalis are the most common species among 14 microsporidia species identified as human pathogens. The aim of this study was to investigate the prevalence of E.intestinalis and E.bieneusi in cancer patients under chemotherapy by immunofluorescent antibody and conventional staining methods. A total of 123 stool samples obtained from 93 patients (58 male, 35 female) with cancer who were followed in oncology and hematology clinics of our hospital and 30 healthy volunteers (13 male, 17 female) were included in the study. Fifty-one (55%) of the patients had complain of diarrhea. The presence of E.intestinalis and E.bieneusi were investigated by a commercial immunofluorescence antibody test using monoclonal antibodies (IFA-MAbs; Bordier Affinity Products, Switzerland) in all of the samples, and 50 of the samples were also investigated by modified trichrome, acid-fast trichrome and calcofluor staining methods. A total of 65 (69.9%) patients were found positive with IFA-MAbs method, including 43 (46.2%) E.intestinalis, 9 (9.7%) E.bieneusi and 13 (14%) mixed infections. In the control group, 5 (16.7%) subjects were positive with IFA-MAbs method, including 2 (6.7%) E.intestinalis, 1 (3.3%) E.bieneusi and 2 (6.7%) mixed infections. The difference between the positivity rate of the patient and control groups was statistically significant (p< 0.05). Of the patients with diarrhea, 68.6% (35/51) were infected with microsporidia, and the difference between cases with and without (48.6%) diarrhea was statistically significant (p< 0.05). When 50 samples in which all of the methods could be performed were evaluated, the frequency of microsporidia were detected as follows; 66% (n= 33) with IFA-MAbs, 34% (n= 17) with modified trichrome staining, 24% (n= 12) with acid-fast trichrome staining and 42% (n= 21) with calcofluor staining methods. Our data indicated that the use of IFA-MAbs method along with the conventional staining methods in diagnosis of microsporidia will increase the sensitivity. As a conclusion, the prevalence of E.intestinalis and E.bieneusi in cancer patients under chemotherapy was detected quite high (69.9%) in our study, it would be appropriate to screen these patients regularly in terms of microsporidian pathogens.

Mikrosporidian patojenler her yerde bulunabilen, omurgalı ve omurgasız konaklarda enfeksiyona neden olabilen, 144 cins altında 1200'den fazla türü tanımlanmış zorunlu hücre içi ökaryotik parazitlerdir. İnsanlarda patojen olarak tanımlanan 14 microsporidia türü arasında en sık saptananlar Enterocytozoon bieneusi ve Encephalitozoon intestinalis olup gastrointestinal sistemde enfeksiyona yol açmaktadırlar. Bu etkenler, özellikle immün sistemi baskılanmış hastalarda kronik ishallerin yanı sıra, yaygın enfeksiyonlara neden olarak hayatı tehdit eden ağır klinik tablolar oluşturmaktadır. Microsporidia sporları çok küçük olduklarından, rutin dışkı incelemelerinde genellikle gözden kaçmaktadır. Bu nedenle tanıda moleküler yöntemler ve mümkünse transmisyon elektron mikroskopi altın standart olarak kullanılmaktadır. Bu yöntemlerin uygulanamadığı laboratuvarlarda ise, monoklonal antikorların kullanıldığı immünofl oresan antikor (IFA-MAbs) yöntemi, konvansiyonel yöntemlere üstünlüğü nedeniyle tercih edilebilir. Bu çalışmada, kemik iliği transplant (KİT) hastalarında E.intestinalis ve E.bieneusi varlığının IFA-MAbs yöntemi ile araştırılması amaçlanmıştır. Çalışmaya, 147'sinde ishal şikayeti olan 200 KİT hastası (134'ü erkek, 66'sı kadın; yaş ortalaması: 43.2 ± 15.01 yıl) ile kontrol olarak 80 sağlıklı birey (43'ü erkek, 37'si kadın; yaş ortalaması: 31.9 ± 11.76 yıl) dahil edilmiştir. Tüm olguların dışkı örnekleri, konvansiyonel yöntemlerin (nativ-lugol ve modifi ye aside dirençli boyama) yanı sıra, ticari bir IFA-MAbs (Bordier Affi nity Products, İsviçre) yöntemiyle de değerlendirilmiştir. Hastaların %25.5 (51/200)'inde E.intestinalis, %4 (8/200)'ünde E.bieneusi ve %9.5 (19/200)'inde her ikisi birden olmak üzere, toplam %39 (78/200)'unda pozitifl ik saptanmıştır. Bu oranlar kontrol grubu için sırasıyla, %5 (4/80), %2.5 (2/80), %3.8 (3/80) ve %11.3 (9/80) olarak tespit edilmiştir. Hasta ve kontrol gruplarında saptanan pozitifl ik oranları arasındaki fark istatistiksel olarak anlamlı bulunmuştur (%39'a karşı %11.3, p< 0.05). Hasta grubunda pozitif bulunan 78 hastanın 67 (%85.9)'sinin ishalli olduğu izlenmiş; ishal varlığı ile parazit pozitifl iği arasında istatistiksel olarak anlamlı bir ilişkinin olduğu görülmüştür (p< 0.05). Sonuç olarak, KİT hastalarının yaşam kalitesini yükseltmek ve tedavi sürecindeki olumsuzlukları azaltmak için, bu hastaların, özellikle de gastrointestinal yakınması olanların düzenli aralıklarla E.intestinalis ve E.bieneusi açısından değerlendirilmesi gerektiği düşünülmüştür, Microsporidian pathogens are obligatory intracellular eukaryotic parasites which can be found worldwide. They have been represented in 144 genera and more than 1200 species that may cause infections in both vertebrate and invertebrate hosts. Enterocytozoon bieneusi and Encephalitozoon intestinalis are the most common species among 14 species of microsporidia identifi ed as human pathogens and they cause infections in the gastrointestinal tract. These species may also cause chronic diarrhea particularly in immunocompromised patients, as well as disseminated infections with severe clinical conditions which can be life-threatening. Since the spores of microsporidia are quite small-sized structures, they frequently may be overlooked in routine stool examinations. Therefore, molecular methods and transmission electron microscopy, if possible, are used as the gold standard methods in laboratory diagnosis. In laboratories in which those methods could not be applied, immunofl uorescence assay using monoclonal antibodies (IFA-MAbs) may be advantageous compared to conventional methods. The aim of this study was to investigate the presence of E.intestinalis and E.bieneusi in bone marrow transplant (BMT) patients by using IFA-MAbs method. A total of 200 BMT patients (134 male, 66 female; mean age: 43.2 ± 15.01 years), of them 147 with diarrhea and 80 healthy subjects (43 male, 37 female; mean age: 31.9 ± 11.76 years) as control group were included in the study. All of the stool samples were examined by a commercial IFA-MAbs (Bordier Affi nity Products, Switzerland) method as well as conventional (native-lugol and modifi ed acid-fast staining) methods. Of the patients 25.5% (51/200) were positive for E.intestinalis, 4% (8/200) for E.bieneusi and 9.5% (19/200) for both of them, giving a total positivity rate of 39% (78/200). Those rates were 5% (4/80), 2.5% (2/80), 3.8% (3/80) and 11.3% (9/80), respectively for control group. The difference between the patient and control groups in terms of positivity was found statistically signifi cant (39% vs 11.3%, p< 0.05). Among 78 positive BMT patients, 67 (85.9%) were suffering from diarrhea. The correlation between the presence of diarrhea and the presence of microsporidia was statistically signifi cant (p< 0.05). It was concluded that, BMT patients particularly those with gastrointestinal complaints, have to be evaluated for microsporidian pathogens regularly to improve quality of life and to decrease the problems during the treatment period

Objective: Enterobius vermicularis is very common parasite in children especially of early school age both world and Turkey. The aim of this study was to investigate the screening of Enterobius vermicularis among the students of the primary schools in our city. Materials and Methods: This investigation was made on a total of 164 students including 84 (51.22%) girls and 80 (48.78%) boys. Results: We found two students to be infected with E. vermicularis and one student to be infected with Taenia spp. using the cellophane tape method. In addition, three cellophane tape were applied to 22 students who were not determined E. vermicularis in one classroom and E. vermicularis was found in two students. Conclusion: We consider that further research with a wide series is warranted and preventive strategies for both E. vermicularis and Taenia spp. are needed. Application of one cellophane tape for the diagnosis of E. vermicularis may not to be adequate., Amaç: Bağırsak parazitleri paraziter hastalıklar içinde günümüzde de önemini korumaktadır. Enterobius vermicularis ise dünyada olduğu kadar ülkemizde de yaygın görülen ve önemli rahatsızlıklara neden olan bir parazittir. Bu çalışmada, ilimizde bir ilköğretim okulunun birinci sınıfında okuyan öğrencilerde E. vermicularis taraması ve yaygınlığı etkileyen bazı faktörlerin araştırılması amaçlanmıştır. Gereç ve Yöntem: Çalışmaya 84 (% 51.22)'u kız, 80 (% 48.78)'i erkek 164 birinci sınıf öğrencisi katıldı. Bulgular: Tek bir selefon bant uygulaması ile 2 öğrencide E. vermicularis bir öğrencide ise Taenia spp. saptandı. E. vermicularis saptanmayan bir sınıftaki 22 öğrenciye ardışık üç adet selefon bant uygulaması yapıldı ve 2 öğrencide E. vermicularis bulundu. Sonuç: Bu çalışmada E. vermicularis'in olduğu kadar Taenia spp.'nın da çocuklar arasında görüldüğü, bu nedenle gerekli önlemlerin alınması gerekliliği ve E. vermicularis tanısında tek bir selefon bant uygulamasının yetersiz olabileceği vurgulanmıştır.

Aim: The present experiment was carried out to determine economic loss caused by cystic echinococcosis in the animals slaughtered in Elazıg ELET corporation slaughterhouse between 1998 and 2000 years. Material and Methods: The records of slaughterhouse were retrospectively investigated to determine the organs with cystic echinococcosis in the slaughtered animals, which include cows, sheep and goats. Results: The percentages of cystic echinococcosis was found to be 2.52%, 2.32% and 0.12%; 2.51%, 2.37% and 0.13%; 1.85%, 1.74% and 0% in the lung, liver and heart from the cows, sheep and goats, respectively. According to the company's wholesale prices, the economical loss due to cystic echinococcosis were 17.031.064.000 TL in the cows, 8.258.000. 000 TL in the sheep and the goats. Total economical loss was calculated as 29.099.064.000 TL. Conclusion: The incidence of location of cystic echinococcosis at liver, lung or heart in cows, sheep and goats is similar but reflection of this involvement to the economic loss is much higher in cows. These results indicate that cystic echinococcosis presents more important consequences in cows in terms of economic loss compared to sheep and goats., Amaç: Bu çalışmada 1 Ocak 1998-31. 12. 2000 tarihlerini kapsayan 3 yıllık döneminde kesilen kasaplık hayvanlarda kistik ekinokokkozise (KE) bağlı oluşan ekonomik kayıpların boyutunun ve hayvan türlerine göre dağılımını araştırmayı amaçladık. Gereç ve Yöntem: Elazığ ELET AŞ. Kesimhane'sinde veteriner kayıt defterleri incelenerek bu dönemde kesimi yapılan hayvanlarda KE'lu organların hayvan türüne ve yıllara göre dağılımları irdelendi. Bulgular: Kesimi yapılan hayvanlarda akciğer, karaciğer ve kalpte KE yerleşme sıklığı sırasıyla sığırlarda %2.52, %2.32 ve %0.12 koyunlarda %2.51, %2.37 ve %0.13 keçilerde %1.85, %1.74 ve %0 olarak bulundu. Ekonomik değer olarak bakıldığında 3 yıllık süre içerisinde toplam ekonomik kaybın 29.099.064.000 TL olduğu bulunmuştur. Bunun 17.031.064.000 TL sığırlardaki, 8.258.000. 000 TL koyunlardaki ve 3.810.000.000 TL ise keçilerdeki ekonomik kayıp olduğu görüldü. Sonuç: Sığır, koyun ve keçilerde KE'nin karaciğer, akciğer ve kalbe benzer oranlarda yerleştiği ancak bu tutulum oranlarının ekonomik kayba yansımasının sığırlarda çok daha yüksek olduğu görülmektedir. Bu sonuçlar ekonomik değer açısından bakıldığında sığırlarda KE'nin koyun ve keçilere göre çok daha önemli olduğu görülmektedir.

Amaç: Bu çalışma nonspesifik karın ağrısı (NKA) tanısı alan hastalarda Toxocara canis'in etyolojideki rolünü belirlemek amacıyla yapıldı. Gereç ve yöntem: 15 Mart-15 Temmuz 2001 tarihleri arasında, Fırat Üniversitesi Tıp Fakültesi Genel Cerrahi Kliniği'ne karın ağrısı nedeniyle yatırılan hastalarda ELISA yöntemi ile anti-T. canis IgG/M seropozitifliği araştırıldı. Bulgular: Çalışmaya alınan 41 olgudan 15'i NKA tanısı alırken, 26'sında karın ağrısını izah edecek etyolojik bir neden saptanarak kontrol grubu olarak ayrıldı. NKA'lı olgulardan 5'inde (%33.33) ve kontrol grubunun 6'sında (%23.07), T. canis seropozitifliği saptandı. Sonuç: Seropozitiflik oranı her iki grupta da normal popülasyona göre yüksek olmasına karşılık iki grup arasında anlamlı bir fark saptanamadı. Bu durum T. canis'in NKA olan hastaların etyolojisinde anlamlı bir yeri olmadığı kanısını destekledi., Aim: To determine the role of Toxocara canis in the etiology of non-specific abdominal pain (NAP), Materials and method: We examined anti-T. canis IgG/M seropositivity by ELISA method in 41 patients with abdominal pain who attended to Fırat University Faculty of Medicine, Department of Surgery between the March 15 and July 15, 2001. Results: The NAP was defined in 15 cases and other 26 cases, in which etiological cause for the abdominal pain has been found, were separated as a control group. T. canis seropositivity was found in 5 cases (33.33%) of NPA group and in 6 cases of control group (%23.07). Despite the high incidence of T. canis seropozitivity in both study groups compared to the normal population, there was no statistically differences among the study groups. Conclusion: The findings of present study support the opinion that T. canis has not important influence in NAP aetiology.

In this study, results of parasitological examinations of stools in the Parasitology laboratory of the Fırat University Medical Centre were evaluated retrospectively in order to determine the distribution of intestinal parasites in our region for the last year. The study included the results of the stool samples taken from outpatients and inpatients that were referred to our hospital between 2004 and 2005. All of the samples were investigated by the cellophane tape method and the direct wet mount method. Also, trichrome stained and acid-fast stained preparations were examined in ambiguous cases. During the period intestinal parasitic disease was detected in 210 (17.24%) of all patients. Only one species of parasite was found in 195 patients whereas two species were found in 15 cases. The most parasites were B. Hominis (26.66%), G. İntestinalis (24.76%) ve E. coli (20.95%) respectively. The result of this study is similar to our country and previously our results. It has been shown that intestinal parasites are still important for public health. ©2006, Fırat Üniversitesi, Tıp Fakültesi, Bu çalışma, ilimiz Tıp Fakültesi hastanesine başvuran hastalardaki bağırsak parazitlerinin dağılımının son bir yıllık durumunu incelemek amacıyla retrospektif olarak yapıldı. Çalışma kapsamına polikliniklerden veya kliniklerde yatırılarak izlenen hastalardan gelen, selefon bant ile nativ-Iugol yöntemi ve şüpheli durumlarda trikrom ve asit-fast boyalı preparasyonlar hazırlanarak incelenen dışkı örneklerinin sonuçları değerlendirmeye alındı. İncelenen 1218 örneğin 210(%17.24)'unda parazit saptanmış olup en sık saptanan parazitlerin sırasıyla Blastocystis hominis %26.66, Giardia intestinalis %24.76 ve Entamoeba coli %20.95 olduğu görüldü. Ayrıca tek tür parazit sıklığının %16.01, iki tür parazit sıklığının ise %1.23 olduğu saptandı. Bu sonuçlar bağırsak parazitlerinin günümüzde de önemini koruyan bir halk sağlığı sorunu olduğunu göstermektedir. Bu sebeple konuya dikkat çekici bu tür çalışmalara devam edilmesinin ve koruyucu önlemlere yönelik çalışmalara daha fazla önem verilmesinin gerektiğini düşünmekteyiz.©2006, Fırat Üniversitesi, Tıp Fakültesi

Objectives: Diarrhea may be of nonspecific, unknown etiology or bacterial, viral, parasitic, or toxic etiology it's a problem not only developing countries but also developed country. The aim of this retrospective study was to acces patients who were up to 15 years old. Materials and Methods: This study presented emergency depertmant complaining of diarrhea between January 2005 and December 2005. Results: It was determineted 167 (76.5%) nause, 136 (62.4%) vomiting, 139 (63.8%) abdominal pain as a the most common symptoms.It was found 44 (20.2%) patients with fever. The parasites that were found and their prevalence is G. intestinalis 11 (5%), E. histolytica/E. dispar 5 (1.8%). Conclusion: Although the ratio of the patient that admitted to the emergency department with a complaint of diarrhea is not so much high, but the symptoms to death. So we emphasized on the clinical importance of personal hygiene, providing clean water and food, establishing substructure, increasing the education of public health about prevention of the infections. ©2007, Firat University, Medical Faculty, Amaç: İshal daha önceleri gelişmekte olan ülkelerin sorunu iken globalleşen dünyada bugün bütün ülkelerin sorunu halini almıştır. Nonspesifik nedenler, virusler, bakteriler, parazitler ve toksinler daha çok su ve yiyeceklerle bulaşarak ishal nedeni olarak karşımıza çıkmaktadır. Bu çalışmada, hastanemiz acil servisine 15 yaş ve üzeri ishal şikayeti ile başvuran hastaların bir yıllık değerlendirilmesi amaçlanmıştır. Gereç ve Yöntem: Çalışma, Tıp Fakültesi acil servisine Ocak-Aralık 2005 tarihleri arasında ishal şikayeti ile başvuran tüm hastaların protokol defterinden dosyalarına ulaşılarak yapılmıştır. Bulgular: İshalle birlikte hastaların 167'sinde (%76.5) bulantı, 136'sında (%62.4) kusma, 139'da (%63.8) karın ağrısı, tespit edilmiştir. Gastrointestinal sistem bulguları dışında en fazla 44'ünde (%20.2) ateş olduğu bulunmuştur. Çalışmamızdan elde edilen sonuçların incelenmesi ile en fazla görülen parazitlerin sırasıyla G. intestinalis, E. histolytica/E. dispar olduğu görülmüştür. Hastaların hastaneye yatışında yaş önemli faktör olarak bulunmuştur ve ishal nedeniyle başvuranların ortalama yaş 38.78 iken yatışlarda ortalama yaş 56.56 olarak karşımıza çıkmaktadır. Sonuç: Acil servise başvuran ishalli hastaların oranı çok yüksek olmamakla birlikte ishalin basit semptomlardan ölüme kadar uzanan bir tabloya sebep olmasından dolayı kişisel hijyen, temiz yiyecek ve suyun temini, alt yapının düzeltilmesi ve infeksiyonun yayılımı konusunda halk eğitiminin artırılması konuları üzerinde önemle durulması gerekliliğini vurgulamaktayız. ©2007, Fırat Üniversitesi, Tıp Fakültesi

Aim: This study was designed in order to find out the year-by-year distribution of fasciolosis prevalence in animals that were slaughtered in slaughterhouse of Elazığ ELET Company between January 1, 1998 and December 31, 2000 and to figure out the dimensions of economic loss because of this condition. Meterial and Methods: Yearly prevalence of fasciolosis in cows, sheep and goats slaughtered during the above stated period has been detected from the veterinary registration books of the ELET Company. The calculated economic loss caused by fasciolosis was based on the wholesale prices of the company. Results: Fasciolosis was present in 1.368 of 6.1157 cows (2.23%), in 1.740 of 82.250 sheep (2.11 %) and in 766 of 54.700 goats (1.40%). 3.874 of a total number of 198.107 animals (1.95%) were determined to have fasciolosis. Total value of livers in animals with fasciolosis was 21.428.000.000 TL. Conclusion: 3874 animals were detected with fasciolosis in only one slaughterhouse in our province during a three year period and yearly average economic losses related to this condition is 7.142.000.000 TL. Preventive measures should be implemented in order to decrease the incidence of fasciolosis in animals and to minimize the economic losses related to this condition., Amaç: Bu çalışma 1 Ocak 1998-31 Aralık 2000 tarihleri arasında Elazığ ELET AŞ. Kesimhane'sinde kesilen kasaplık hayvanlarda görülen fasciolosisin yıllara göre dağılımını ve bu nedenle oluşan ekonomik kayıpların boyutunu araştırmak amacıyla yapıldı. Toplam olarak kesimi yapılan 198.107 hayvanın 3.874'ünde (%1.95) fasciolosis saptandı. Fasciolosis nedeniyle imha edilen karaciğerlerin toplam ekonomik değeri 21.428.000.000 TL olarak hesaplandı. Sonuç: İlimizde tek bir kesimhanede 3 yıllık sürede toplam 3874 fasciolosisli hayvan saptandığı ve buna bağlı yıllık ortalama ekonomik kaybın 7.142.000.000 TL olduğu görülmüştür. Hayvanlardaki fasciolosis yaygınlığının azaltılması ve ekonomik kayıpların asgari düzeye çekilebilmesi için önlemler alınması gerekli görülmektedir.

Aim: In this study, it was investigated that bacterial colonies obtained after transformation were tested whether they contained positive recombinant colonies by using PCR screening method. Material and method: For this aim, Fasciola hepatica cathepsin L1 gene and Rinderpest virus matrix gene were cloned into three different vector systems and transformed into E. coli. It was determined some bacterial colonies which had the right size band after PCR screening. Plasmid DNA of those bacterial colonies was extracted by classical miniprep method. The colonies positive with PCR screening were assayed by enzyme digestion assays to verify the PCR screening results. Results: There was 100% correlation between PCR screening and enzyme digestion assays. Conclusion: Using PCR screening method reduce the time requirement for detection the positive colonies and saving the materials used for plasmid DNA extraction., Amaç: Bu çalışmada Polimeraz Zincir Reaksiyonu Tarama (PZR-T) yöntemiyle klonlama sonucu elde edilen bakteriyel kolonilerin pozitifliği araştırılmıştır. Gereç ve yöntem: Fasciola hepatica katepsin L1 geni ve sığır vebası virüs matriks genleri üç farklı vektör sistemine klonlandıktan sonra E.coli kompetan hücrelerine transformasyon yapıldı. Transformasyon sonucu elde edilen kolonilerin bir kısmı PZR-T yöntemi kullanılarak değerlendirmeye tabi tutuldu. PZR-T sonucu istenilen büyüklükte elde edilen bantları içeren koloniler belirlenerek klasik miniprep yöntemiyle plasmid DNA'lar elde edildi. Kesim deneyleri yapılarak istenilen gen bölgelerinin varlığı doğrulandı. Bulgular: PZR-T ile pozitif olduğu saptanan 61 örneğin tamamının miniprep sonrası yapılan kesim deneylerinde de pozitif olduğu belirlendi. Sonuç: PZR-T yöntemiyle hem çok daha kısa zamanda pozitif koloniler belirlenecek hem de materyal açısından büyük ölçüde tasarruf edilmiş olacaktır.

Acanthamoeba, Naegleria, Balamuthia, Sappinia, Vahlkampfia and Hartmannella of the free-living ameba are commonly encountered in soil, air, lens solutions and water sources such as tap water, lakes, rivers and swimming pools. Some of those free-living ameba may cause serious diseases in human. The main objective of this study is to isolate and genotype the free-living potential pathogen amoeba species in the soil samples obtained from different regions of Kayseri. Each of the total 20 samples are passed through cellulose nitrate filter and transported to the non nutritional plates and coated with Escherichia coli agar. The samples which are cultured at 37 0C are observed for two weeks in order to observe the reproduction and isolate the DNA samples that are thought to be positive. Eight of the 20 samples which are undergoing PCR with specific primers were Acanthamoeba positive and three of them were Naegleria positive. Besides, Balamuthia was not detected in any of the samples. In conclusion, in this study the presence of some free-living amoeba species which may threaten human health is shown in the soil samples obtained from Kayseri, Serbest yaşayan amiplerden Acanthamoeba, Naegleria, Balamuthia, Sappinia, Vahlkampfia ve Hartmannella; toprakta, musluk suyu, göl, nehir, yüzme havuzu gibi çeşitli su kaynaklarında, havada, lens solüsyonlarında bulunabilmektedir. Doğada serbest olarak yaşayan bu amiplerin bir kısmı insanlarda ciddi hastalıklara yol açabilmektedir. Bu çalışmada; Kayseri’nin çeşitli bölgelerinden alınan toprak örneklerinde serbest yaşayan potansiyel patojen amip türlerinin izolasyonu ve genotiplendirilmesi amaçlanmıştır. Toplam 20 farklı bölgeden alınan birer örnek selüloz nitrat filtreden geçirilip Escherichia coli ile kaplanmış besleyici değeri olmayan agar plaklarına aktarılmıştır. 37 0C’de kültürü yapılan örneklerin iki hafta boyunca üremeleri gözlenerek, pozitif olduğu düşünülen örneklerden DNA izolasyonu gerçekleştirildi. Acanthamoeba, Balamuthia spesifik primerlerle PCR’ı yapılan toplam 20 toprak örneğinin sekizinin Acanthamoeba, üçünün Naegleria pozitif olduğu belirlenirken, örneklerin hiçbirinde Balamuthia saptanmamıştır. Bu çalışma ile insan sağlığı açısından tehdit oluşturabilecek serbest yaşayan bazı amip türlerinin varlığı Kayseri’deki toprak örneklerinde gösterilmiştir
