Abstract Background Postoperative pneumonia (POP) is the most prevalent of all nosocomial infections in patients who underwent cardiac surgery. The aim of this study was to identify independent risk factors for pneumonia after cardiac surgery, from which we constructed a nomogram for prediction. Methods The clinical data of patients admitted to the Department of Cardiothoracic Surgery of Nanjing Drum Tower Hospital from October 2020 to September 2021 who underwent cardiac surgery were retrospectively analyzed, and the patients were divided into two groups according to whether they had POP: POP group (n=105) and non-POP group (n=1083). Preoperative, intraoperative, and postoperative indicators were collected and analyzed. Logistic regression was used to identify independent risk factors for POP in patients who underwent cardiac surgery. We constructed a nomogram based on these independent risk factors. Model discrimination was assessed via area under the receiver operating characteristic curve (AUC), and calibration was assessed via calibration plot. Abstract Background Postoperative hyper-inflammation is a frequent event in patients with acute Stanford type A aortic dissection (ATAAD) after surgical repair. This study's objective was to determine which inflammatory biomarkers could be used to make a better formula for identifying postoperative hyper-inflammation, and which risk factors were associated with hyper-inflammation. Methods A total of 405 patients were enrolled in this study from October 1, 2020 to April 1, 2023. Of these patients, 124 exhibited poor outcomes. In order to investigate the optimal cut-off values for poor outcomes, logistic and receiver operating characteristic analyses were performed on the following parameters on the first postoperative day: procalcitonin (PCT), C-reactive protein (CRP), interleukin-6 (IL-6), and systemic immune-inflammation index (SII). These cut-off points were used to separate the patients into hyper-inflammatory (n = 52) and control (n = 353) groups. Finally, the logistic were used to find the risk factors of hyper-inflammatory. Results PCT, CRP, IL-6, and SII were independent risk factors of poor outcomes in the multivariate logistic model. Cut-off points of these biomarkers were 2.18 ng/ml, 49.76 mg/L, 301.88 pg/ml, 2509.96 × 109/L respectively. These points were used to define postoperative hyper-inflammation (OR 2.97, 95% CI 1.35–6.53, P 180 min, and deep hypothermia circulatory arrest (DHCA) > 40 min were the independent risk factors for hyper-inflammation. Conclusions PCT > 2.18, CRP > 49.76, IL-6 > 301.88, and SII 180 min and DHCA > 40 min were separate risk factors for postoperative hyper-inflammation.

Regulatory T cells (Tregs) are crucial immune cells that initiate a tolerable immune response. Transforming growth factor-beta (TGFβ) is a key cytokine produced by Tregs and plays a significant role in stimulating tissue fibrosis. Systemic sclerosis, an autoimmune disease characterized by organ fibrosis, is associated with an overrepresentation of regulatory T cells. This review aims to identify Treg-dominant tolerable host immune reactions and discuss their association with scleroderma and end-stage organ failure. End-stage organ failures, including heart failure, liver cirrhosis, uremia, and pulmonary fibrosis, are frequently linked to tissue fibrosis. This suggests that TGFβ-producing Tregs are involved in the pathogenesis of these conditions. However, the exact significance of TGFβ and the mechanisms through which it induces tolerable immune reactions during end-stage organ failure remain unclear. A deeper understanding of these mechanisms could lead to improved preventive and therapeutic strategies for these severe diseases.

Summary: Fish cells, such as grass carp (Ctenopharyngodon idella) kidney (CIK) cells, are harder to transfect than mammalian cells. There is a need for an efficient gene delivery system for fish cells. Here, we used CIK cell line as a model to develop a strategy to enhance RNA and plasmid DNA transfection efficiency using a nanocarrier generated from α-lactalbumin (α-NC). α-NC absorbed nucleic acid cargo efficiently and exhibited low cytotoxicity. Plasmid transfection was more efficient with α-NC than with liposomal transfection reagents. We used α-NC to co-transfect Tol2 transposase mRNA and a plasmid containing Cas9 and GFP, generating a stable transgenic CIK cell line. Genome and RNA sequencing revealed that the Cas9 and GFP fragments were successfully inserted into the genome of CIK cells and efficiently transcribed. In this study, we established an efficient transfection system for fish cells using α-NC, simplifying the process of generating stable transgenic fish cell lines.

Oxidative stress plays a key role in chronic kidney disease (CKD) development and progression, inducing kidney cell damage, inflammation, and fibrosis. However, effective therapeutic interventions to slow down CKD advancement are currently lacking. The multifaceted pharmacological effects of molecular hydrogen (H2) have made it a promising therapeutic avenue. H2 is capable of capturing harmful •OH and ONOO- while maintaining the crucial reactive oxygen species (ROS) involved in cellular signaling. The NRF2-KEAP1 system, which manages cell redox balance, could be used to treat CKD. H2 activates this pathway, fortifying antioxidant defenses and scavenging ROS to counteract oxidative stress. H2 can improve NRF2 signaling by using the Wnt/β-catenin pathway and indirectly activate NRF2-KEAP1 in mitochondria. Additionally, H2 modulates NF-κB activity by regulating cellular redox status, inhibiting MAPK pathways, and maintaining Trx levels. Treatment with H2 also attenuates HIF signaling by neutralizing ROS while indirectly bolstering HIF-1α function. Furthermore, H2 affects FOXO factors and enhances the activity of antioxidant enzymes. Despite the encouraging results of bench studies, clinical trials are still limited and require further investigation. The focus of this review is on hydrogen's role in treating renal diseases, with a specific focus on oxidative stress and redox signaling regulation, and it discusses its potential clinical applications.

The preparation parameters of PM6:Y6 non-fullerene organic solar cells (OSCs) have significant influence on the power conversion efficiency (PCE). Herein, machine learning (ML) models are applied for analyzing the quantitative effects of the PCE on PM6:Y6 non-fullerene OSCs from the perspective of fabrication parameters. Random Forest (RF) model has the best evaluation performance and is considered as the best model among the six different algorithms. The Pearson correlation coefficient, coefficient of determination, root mean square error, mean absolute error, and mean absolute percentage error of the test set in the RF model are 0.836, 0.668, 0.695, 0.538, and 0.035, respectively. In addition, the most important preparation parameters for PCE are the effective area of OSCs, type of solvent and solvent additives, spin-coating speed, total concentration, annealing temperature, annealing time from SHAP analysis. The optimized PCE can be obtained in PM6:Y6 non-fullerene OSCs when the effective area is between 4 and 6 mm2, the spin-coating speed is 3000 rpm, and the total concentration is 18 mg ml−1, respectively. The contour map further reflects that the optimal annealing temperature are 80 or 105 °C, and the corresponding annealing time are 5 or 9 min, respectively. Based on the above preparation conditions, we verified through the experimental design that the optimal PCE is 16.91%, and the relative error with the ML prediction is only 1.24%. Our research provides low time and cost guidance on preparation parameter matching for the development of high-performance the PM6:Y6 non-fullerene OSCs.

The composition and content of fatty acids (FAs) and amino acids (AAs) are important nutritional and sensory traits in aquaculture fish. To meet increasing demand while retaining quality, genetic selection in breeding programs is urgently needed to improve these traits. In this study, we performed the genome-wide association study (GWAS) to identify genetic variants and candidate genes potentially linked to FA and AA compositions in grass carp muscle. We focused on key dietary components, including unsaturated FAs and essential AAs. Through profiling of total FA and AA contents, we identified strong positive correlations among the compositions of most FAs and AAs. From our GWAS panel, 4,913,199 high-quality single nucleotide polymorphisms (SNPs) were identified across the genome, of which 32 SNPs linked to 18 candidate genes were significantly associated with FA levels, and 202 SNPs linked to 115 candidate genes associated with AA contents. In addition, we categorized our sequencing population by region and detected allele frequencies of SNPs across four geographic locations. Notably, four FA-associated SNPs and 28 AA-associated SNPs exhibited geographic specificity. These results offered valuable insights into the complex genetic basis of FA and AA metabolism and deposition. Furthermore, this work provides potential genetic markers for selective breeding aimed at enhancing flesh quality in the economically important fish species, grass carp.

Summary: The abnormal reproduction of algae in water worldwide is prominent in the context of human interference and global climate change. This study first thoroughly analyzed the effects of physical factors, such as light, temperature, hydrodynamics, and operational strategies, on algal growth and their mechanisms. Physical control techniques are safe and have great potential for preventing abnormal algal blooms in the absence of chemical reagents. The focus was on the principles and possible engineering applications of physical shading, ultrasound, micro-current, and ultraviolet (UV) technologies, in controlling abnormal algal reproduction. Physical shading can inhibit or weaken photosynthesis in algae, thereby inhibiting their growth. Ultrasound mainly affects the physiological and biochemical activities of cells by destroying the cell walls, air cells, and active enzymes. Micro-currents destroy the algal cell structure through direct and indirect oxidation, leading to algal cell death. UV irradiation can damage DNA, causing organisms to be unable to reproduce or algal cells to die directly. This article comprehensively summarizes and analyzes the advantages of physical prevention and control technologies for the abnormal reproduction of algae, providing a scientific basis for future research. In the future, attempts will be made toward appropriately and comprehensively utilizing various physical technologies to control algal blooms. The establishment of an intelligent, comprehensive physical prevention and control system to achieve environmentally friendly, economical, and effective physical prevention and control of algae, such as the South-to-North Water Diversion Project in China, is of great importance for specific waters.

Abstract Evidence indicates that patients with chronic low back pain (CLBP) have lumbar multifidus muscle (LM) activation deficit which might be caused by changes in cortical excitability. Anodal transcranial direct current stimulation (a-tDCS) can be used to restore cortical excitability. This study aimed to (1) determine the immediate effects of a-tDCS on the cortical excitability and LM activation and (2) explore the relationship between cortical excitability and LM activation. Thirteen participants with CLBP during remission and 11 healthy participants were recruited. Cortical excitability (peak-to-peak motor evoked potential amplitude; P2P and cortical silent period; CSP) and LM activation were measured at pre- and post-intervention. We found significant difference (P 0.05) in P2P, CSP and LM activation were found between pre- and post-intervention in CLBP. The CLBP group demonstrated significant correlation (P = 0.05) between P2P and LM activation. Although our finding demonstrates change in P2P in the CLBP group, one-session of a-tDCS cannot induce changes in cortical excitability and LM activation. However, moderate to strong correlation between P2P and LM activation suggests the involvement of cortical level in LM activation deficit. Therefore, non-significant changes could have been due to inadequate dose of a-tDCS.

Catalyzed by Rh2(esp)2 (10 mol%) and (±)-BINAP (20 mol%) in DCE at 80 °C, the cascade assembly between diazobarbiturates and alkylidene pyrazolones proceeded readily and produced spiro-furopyrimidines in 38–96% chemical yields. The chemical structure of the prepared spirofuro-pyrimidines was firmly confirmed by X-ray diffraction analysis.

Cladophora is commonly found in marine and freshwater around the globe and provides productivity for littoral zone microorganisms and invertebrates. The eutrophication of the water body has led to the abnormal proliferation of Cladophora in some water, often in river coast channel outbreaks. Even under the nutritional deficiency systems, such as the central trunk canal of the South-to-North Water Diversion in China, Cladophora’s blooms affect water quality and seriously jeopardize human health. Thus, the structural characteristics of Cladophora cells and spores, the factors affecting the growth of Cladophora cells, and the mechanisms of damage and destruction of Cladophora cells and spores were investigated. Cladophora cells are cylindrical with very thick cell walls. The inner layer of the spore wall is a thin membrane which contains the nucleus of the spore. The growth and spreading of Cladophora cells are affected by various environmental factors such as light, temperature, water depth, water level, nutrient salts, pH, etc. Some physical treatment measures, such as ultrasounds, would destroy the cell walls and membranes of Cladophora by its high-intensity mechanical action. Chemicals and aquatic plant measures can destroy Cladophora cells’ photosynthesis system, antioxidant enzyme systems, proteins, and ultrastructure. Based on the mechanisms for these cell damage and destruction, a combination of measures that are likely to inhibit the growth of Cladophora cells effectively was suggested. Furthermore, the damaged cells of Cladophora could provide some environmental benefits. Aggregated results could provide a scientific basis for further research on the control of blooms of Cladophora or the reuse of Cladophora cells as a natural resource.

Abstract Background Myocardial injury-related cardiogenic shock (MICS) is significantly associated with poor outcomes in patients after cardiac surgery. Herein, we aimed to investigate the risk factor for postoperative MICS. Methods We performed a case-control study on 792 patients undergoing cardiac surgery from 2016 to 2019, including 172 patients with postoperative MICS and 620 age- and sex-matched controls. MICS was defined as composite criteria: a cardiac index of 5 mmol/L at the end of the surgery, a vasoactive-inotropic score of > 40 at the end of the surgery, and a cardiac troponin T (cTnT) level of > 0.8 µg/L on postoperative day 1 (POD1) with an increase of > 10% on POD 2. Results A total of 4671 patients who underwent cardiac surgery in our hospital between 2016 and 2019 were included; of these, 172 (3.68%) had MICS and the remaining 4499 did not. Psychological stress increases risk of gastrointestinal tract diseases. However, the mechanism behind stress-induced gastrointestinal injury is not well understood. The objective of our study is to elucidate the putative mechanism of stress-induced gastrointestinal injury and develop an intervention strategy. To achieve this, we employed the restraint stress mouse model, a well-established method to study the pathophysiological changes associated with psychological stress in mice. By orally administering gut-nonabsorbable Evans blue dye and monitoring its plasma levels, we were able to track the progression of gastrointestinal injury in live mice. Additionally, flow cytometry was utilized to assess the viability, death, and inflammatory status of splenic leukocytes, providing insights into the stress-induced impact on the innate immune system associated with stress-induced gastrointestinal injury. Our findings reveal that neutrophils represent the primary innate immune leukocyte lineage responsible for stress-induced inflammation. Splenic neutrophils exhibited elevated expression levels of the pro-inflammatory cytokine IL-1, cellular reactive oxygen species, mitochondrial burden, and cell death following stress challenge compared to other innate immune cells such as macrophages, monocytes, and dendritic cells. Regulated cell death analysis indicated that NETosis is the predominant stress-induced cell death response among other analyzed regulated cell death pathways. NETosis culminates in the formation and release of neutrophil extracellular traps, which play a crucial role in modulating inflammation by binding to pathogens. Treatment with the NETosis inhibitor GSK484 rescued stress-induced neutrophil extracellular trap release and gastrointestinal injury, highlighting the involvement of neutrophil extracellular traps in stress-induced gastrointestinal inflammation. Our results suggest that neutrophil NETosis could serve as a promising drug target for managing psychological stress-induced gastrointestinal injuries.

In recent years, the outbreak of infectious diseases has highlighted the need for improved planning of hospital buildings. Traditional planning for infectious disease hospitals only considers the impact of wind and pollutant diffusion, without analysing pollutant diffusion under different thermal conditions. To reveal the distribution of pollutants in infectious disease hospitals under different thermal conditions, this study conducted wind tunnel tests and numerical analyses of pollutant diffusion in the environment surrounding an infectious disease hospital in Changsha, China. The results show that the pollutant concentration mainly depends on the local wind speed. In the range of Rb = −1.25 to 1.25, the concentration of pollutants was mainly affected by the disturbance of the flow field in areas with rough surfaces, where the effect of the thermal stability of the atmosphere on pollutant diffusion was relatively small. However, in relatively flat regions, the thermal stability of the atmosphere played a significant role in pollutant diffusion around the buildings.

The characterization of dissolved organic matter (DOM) is important for better understanding of the migration and transformation mechanisms of DOM in water bodies and its interaction with other contaminants. In this work, fluorescence characteristics and molecular compositions of the DOM samples collected from the mainstream, tributary, and sewage outfall of the Inner Mongolia section of the Yellow River (IMYR) were determined by using fluorescence spectroscopy and Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS). In addition, concentrations of potentially toxic elements (PTEs) in the relevant surface water and their potential relationships with DOM were investigated. The results showed that the abundance of tyrosine-like components increased significantly in downstream waters impacted by outfall effluents and was negatively correlated with the humification index (HIX). Compared to the mainstream, outfall and tributaries have a high number of molecular formulas and a higher proportion of CHOS molecular formulas. In particular, the O5S class has a relative intensity of 41.6% and the O5-7S class has more than 70%. Thirty-eight PTEs were measured in the surface water samples, and 12 found above their detective levels at all sampling sites. Protein-like components are positively correlated with Cu, which is likely indicating the source of Cu in the aquatic environment of the IMYR. Our results demonstrated that urban wastewater discharges significantly alter characteristics and compositions of DOM in the mainstream of IMYR with strongly anthropogenic features. These results and conclusions are important for understanding the role and sources of DOM in the Yellow River aquatic environment.

Abstract Background To explore the effects of Ving Tsun (VT) sticking-hand training on knee joint proprioception and leg muscular performance among community-dwelling middle-aged and older adults. Methods Thirty-three middle-aged and older adults were randomly assigned to either the VT or control group. The VT group received sticking-hand training twice/week for 3 months. Data were collected before and after the intervention period. The primary outcome was knee joint repositioning error, which was measured using knee joint active repositioning tests. The secondary outcomes included the maximum muscle strength and time to maximum muscle strength of the major leg muscles. Results No significant group, time, or group-by-time interaction effects were found for the knee joint repositioning error (p > 0.05). The maximum muscle strength of the knee flexors increased over time in the VT group only (p = 0.038). Abstract Background Recent high-quality trials have shown that the anti-inflammatory effects of colchicine reduce the risk of cardiovascular events in patients suffering post-myocardial infarction and chronic coronary disease. The effect of colchicine in patients undergoing non-coronary artery bypass grafting (non-CABG) with cardiopulmonary bypass remains unclear. We aim to evaluate the effect of colchicine on myocardial protection in patients who underwent non-CABG cardiac surgery. Method Patients were randomly assigned to colchicine or placebo groups starting 72 h before scheduled cardiac surgery and for 5 days thereafter (0.5 mg daily).The primary outcome was the level of cardiac troponin T (cTnT) at postoperative 48 h. The secondary outcomes included troponin I (cTnI) and creatine kinase-MB (CK-MB), inflammatory biomarkers (procalcitonin and interleukin-6, etc.), and adverse events (30-day mortality, stroke, ECMO and IABP use, etc.). Results A total of 132 patients underwent non-CAGB cardiac surgery, 11were excluded because of diarrhea (n = 6) and long aortic cross-clamp time > 2 h (n = 5), 59 were assigned to the colchicine group and 62 to the placebo group. 本研究運用行動研究協助研究者進行教學實習課的規劃、實施與省思，研究目的為探討授課研究融入教學實習課中的實踐情形，並瞭解授課研究融入教學實習課中職前教師的學習成效。研究參與者是國民小學職前教師 36 位，資料蒐集方式為訪談、觀察與文件蒐集。在資料分析之後，本研究得到的結論為：首先，授課研究透過理念介紹、微型教學與集中實習等實踐歷程，有助於教學實習課的實施。其次，職前教師在微型教學時逐漸掌握設計與實施教學，對小學生的理解仍有困難。在集中實習時可深化職前教師的教學設計與實施教學的成效，及更加理解小學生的學習。本研究的建議是引導職前教師在集中實習後再一次修訂教學設計，及進一步探討職前教師在教材教法課或教育實習階段參與授課研究的情形。 This study utilized action research to conduct planning, implementation, and reflection on a teaching internship. The objectives of this study were to understand the implementation of integrating a lesson study with a teaching internship course as well as pre-service teachers’ learning effectiveness. The participants were 36 elementary school pre-service teachers. Interviews, observations, and documents collection were conducted for data collection. After data analysis, the following conclusions were obtained. First, the lesson study was conducive to the practice of the teaching internship course through the processes of an introduction to ideas, microteaching and intensive internship. Second, pre-service teachers gradually mastered the design and implementation of teaching during microteaching, but there were still difficulties in understanding elementary school students. During the intensive internship, it was possible to deepen the effectiveness of pre-service teachers’ teaching design and implementation and to better understand elementary school students’ learning. The suggestions of this research are to guide preservice teachers to revise their teaching designs after the intensive internship. This study also advises to further explore the situation of pre-service teachers’ participation in the lesson study in regards to teaching materials, methods course, or educational practicum.

Abstract Background Heme oxygenase one (HO-1) is considered a poor prognostic factor for survival in patients with severe-to-critical coronavirus disease (COVID-19), but the clinical correlation between heme catabolism biomarkers and COVID-19-related sepsis is unknown. The etiopathogenetic hypothesis of HO-1 response during sepsis in patients with poor prognosis should be clarified. This study aimed to investigate sepsis development within 48 h following moderate-to-critical COVID-19 and examined heme/HO-1 catabolism biomarkers associated with sepsis. We also studied the HO-1 and traditional prognostic factors for predicting survival in patients with COVID-19. Methods This retrospective observational study included patients unvaccinated for COVID-19 with moderate-to-critical COVID-19 (n = 156) who had been admitted to Taipei Tzu Chi Hospital in 2021. All COVID-19 patients were diagnosed by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) reverse transcriptase polymerase chain reaction. For analysis of heme catabolism in SARS-CoV-2-induced sepsis, we excluded patients with co-infection and severe anemia. Heme catabolism biomarkers were compared between groups of patients with COVID-19 and sepsis (sepsis) and those with COVID-19 without sepsis (no sepsis), and a control group comprising 100 healthy individuals. All clinical and laboratory data were collected retrospectively and blood specimens were collected from Biobank. Multivariable logistic regression analysis was used to compare all variables between the sepsis and no-sepsis groups. Cox regression analysis was used to determine predictors of survival in patients with COVID-19. Results There were 71 and 85 patients with and without sepsis, respectively. Heme and HO-1 levels differed significantly between the sepsis, no sepsis, and control groups. In the field of reliability engineering, covariate information shared among product units within a specific group (e.g., a manufacturing batch, an operating region), such as operating conditions and design settings, exerts substantial influence on product lifetime prediction. The covariates shared within each group may be missing due to sensing limitations and data privacy issues. The missing covariates shared within the same group commonly encompass a variety of attribute types, such as discrete types, continuous types, or mixed types. Existing studies have mainly considered single-type missing covariates at the individual level, and they have failed to thoroughly investigate the influence of multi-type group-shared missing covariates. Ignoring the multi-type group-shared missing covariates may result in biased estimates and inaccurate predictions of product lifetime, subsequently leading to suboptimal maintenance decisions with increased costs. To account for the influence of the group-shared missing covariates with different structures, a new flexible lifetime model with multi-type group-shared latent heterogeneity is proposed. We further develop a Bayesian estimation algorithm with data augmentation that jointly quantifies the influence of both observed and multi-type group-shared missing covariates on lifetime prediction. A tripartite method is then developed to examine the existence, identify the correct type, and quantify the influence of group-shared missing covariates. To demonstrate the effectiveness of the proposed approach, a comprehensive simulation study is carried out. A real case study involving tensile testing of molding material units is conducted to validate the proposed approach and demonstrate its practical applicability.

The precipitation characteristics in Hangzhou of Zhejiang Province, China under the background of global climate change are analyzed using the meteorological observation data obtained from the Hangzhou base station in this study. We investigate the climate characteristics of precipitation in Hangzhou from several aspects, such as centennial trend, seasonal change, periodicity and the variation of extreme precipitation. Our results show a linear decreasing trend and obvious interdecadal characteristics in the precipitation of Hangzhou on a centennial timescale. Significantly increased amplitude of precipitation fluctuation was observed since the beginning of the 21st century. For the interdecadal variation of seasonal precipitation on a centennial timescale, precipitation in autumn showed a decreasing trend of 8.1 mm/10a, whereas the trends for the other three seasons were statistically insignificant. The precipitation in Hangzhou showed a decreasing trend in spring and an increasing trend in winter over the past 30 years. Our analyses reveal distinct precipitation cycles, including a quasi-30-year cycle since the 1960s and a quasi-10-year cycle since the 1980s. However, the periodicity has weakened in the past 10 years. In addition, the occurrence of torrential rain has increased rapidly in the past 10 years. Furthermore, influenced by global climate change and regional processes, the variation of extreme precipitation in Hangzhou has changed, which shows strong correlations with the overall trend of annual precipitation. The annual maximum daily precipitation in Hangzhou was mainly in the range of 0–40 mm from 1951 to 1980 and in the range of 40–80 mm from 1981 to 2010 with the maximum daily precipitation occurrence rate of 4.7 times/10a and 6.3 times/10a, respectively. This study emphasizes the risk of urban waterlogging caused by short-term heavy rainfall and provides useful reference to the assessment of extreme meteorological and hydrological disaster risk in Hangzhou.

DNA methylation and chromatin accessibility play important roles in gene expression, but their function in subgenome expression dominance remains largely unknown. We conducted comprehensive analyses of the transcriptome, DNA methylation, and chromatin accessibility in liver and muscle tissues of allotetraploid common carp, aiming to reveal the function of epigenetic modifications in subgenome expression dominance. A noteworthy overlap in differential expressed genes (DEGs) as well as their functions was observed across the two subgenomes. In the promoter and gene body, the DNA methylation level of the B subgenome was significantly different than that of the A subgenome. Nevertheless, differences in DNA methylation did not align with changes in homoeologous biased expression across liver and muscle tissues. Moreover, the B subgenome exhibited a higher prevalence of open chromatin regions and greater chromatin accessibility, in comparison to the A subgenome. The expression levels of genes located proximally to open chromatin regions were significantly higher than others. Genes with higher chromatin accessibility in the B subgenome exhibited significantly elevated expression levels compared to the A subgenome. Contrastingly, genes without accessibility exhibited similar expression levels in both subgenomes. This study contributes to understanding the regulation of subgenome expression dominance in allotetraploid common carp.

Dissolved organic matter (DOM) plays a major role in ecological systems, affecting the fate and transportation of iron (Fe) and phosphorus (P). To better understand the geochemical cycling of these components, soil and sediment samples were collected around a reservoir downstream of a typical temperate forest in Northeast China. The DOM fractions from these soils, river, and reservoir sediments were extracted and then characterized by spectroscopic techniques. Abstract Introduction Autoimmune encephalitis (AE) is caused by autoantibodies attacking neuronal cell surface antigens and/or synaptic antigens. We previously demonstrated that S100A6 was hypomethylated in patients with AE and that it promoted B lymphocyte infiltration through the simulated blood–brain barrier (BBB). In this study, we focused on the epigenetic regulation of S100A6, the process by which S100A6 affects B lymphocyte infiltration, and the therapeutic potential of S100A6 antibodies. Methods We enrolled and collected serum from 10 patients with AE and 10 healthy control (HC) subjects. Promoter methylation and 5‐azacytidine treatment assays were conducted to observe the methylation process of S100A6. The effect of S100A6 on B lymphocytes was analyzed using an adhesion assay and leukocyte transendothelial migration (LTEM) assay. A LTEM assay was also used to compare the effects of the serum of HCs, serum of AE patients, S100A6 recombinant protein, and S100A6 antibodies on B lymphocytes. Result The promoter methylation and 5‐azacytidine treatment assays confirmed that S100A6 was regulated by DNA methylation. The adhesion study demonstrated that the addition of S100A6 enhanced adhesion between B lymphocytes and a BBB endothelial cell line in a concentration‐dependent manner. The LTEM assay showed that the serum of AE patients, as well as S100A6, promoted B lymphocyte infiltration and that this effect could be attenuated by S100A6 antibodies. Conclusion We clarified that S100A6 was under epigenetic regulation in patients with AE and that it helped B lymphocytes to adhere to and infiltrate the BBB endothelial layer, which could be counteracted by S100A6 antibodies. Therefore, the methylation profile of S100A6 could be a marker of the activity of AE, and countering the effect of S100A6 may be a potential treatment target for AE.

Abstract Study objective The purpose of the present study was to evaluate the efficacy of levosimendan in patients with acute myocardial infarction related ventricular septal rupture (AMI-VSR) underwent cardiac surgery. Design Prospective observational cohort study with propensity score analysis. Patients There were 261 patients with AMI-VSR in our study. After 1:1 propensity matching, 106 patients (53 levosimendan and 53 control) were selected in the matched cohort. Interventions None. Measurements Patients who received levosimendan were assigned to the levosimendan group (n = 164). The patients who were not received were levosimendan assigned to the control group (n = 97). The levosimendan was initiated immediately after cardiopulmonary bypass. Then, it has been maintained during the postoperative 3 days.
