Abstract In the field of information processing, all-optical routers are significant for achieving high-speed, high-capacity signal processing and transmission. In this study, we developed three types of structurally simple and flexible routers using the deep diffractive neural network (D2NN), capable of routing incident light based on wavelength and polarization. First, we implemented a polarization router for routing two orthogonally polarized light beams. The second type is the wavelength router that can route light with wavelengths of 1550, 1300, and 1100 nm, demonstrating outstanding performance with insertion loss as low as 0.013 dB and an extinction ratio of up to 18.96 dB, while also maintaining excellent polarization preservation. The final router is the polarization-wavelength composite router, capable of routing six types of input light formed by pairwise combinations of three wavelengths (1550, 1300, and 1100 nm) and two orthogonal linearly polarized lights, thereby enhancing the information processing capability of the device. These devices feature compact structures, maintaining high contrast while exhibiting low loss and passive characteristics, making them suitable for integration into future optical components. This study introduces new avenues and methodologies to enhance performance and broaden the applications of future optical information processing systems. Graphical Abstract

Abstract Cancer immunotherapy has demonstrated significant efficacy in various tumors, but its effectiveness in treating Hepatocellular Carcinoma (HCC) remains limited. Therefore, there is an urgent need to identify a new immunotherapy target and develop corresponding intervention strategies. Bioinformatics analysis has revealed that growth differentiation factor 15 (GDF15) is highly expressed in HCC and is closely related to poor prognosis of HCC patients. The previous study revealed that GDF15 can promote immunosuppression in the tumor microenvironment. Therefore, knocking out GDF15 through gene editing could potentially reverse the suppressive tumor immune microenvironment permanently. To deliver the CRISPR/Cas9 system specifically to HCC, nanocapsules (SNC) coated with HCC targeting peptides (SP94) on their surface is utilized. These nanocapsules incorporate disulfide bonds (SNCSS) that release their contents in the tumor microenvironment characterized by high levels of glutathione (GSH). In vivo, the SNCSS target HCC cells, exert a marked inhibitory effect on HCC progression, and promote HCC immunotherapy. Mechanistically, CyTOF analysis showed favorable changes in the immune microenvironment of HCC, immunocytes with killer function increased and immunocytes with inhibitive function decreased. These findings highlight the potential of the CRISPR‐Cas9 gene editing system in modulating the immune microenvironment and improving the effectiveness of existing immunotherapy approaches for HCC.

Ischemic heart disease (IHD) is a condition where the heart muscle does not receive enough blood flow, leading to cardiac dysfunction. Restoring blood flow to the coronary artery is an effective clinical therapy for myocardial ischemia. This strategy helps lower the size of the myocardial infarction and improves the prognosis of patients. Nevertheless, if the disrupted blood flow to the heart muscle is restored within a specific timeframe, it leads to more severe harm to the previously deprived heart tissue. This condition is referred to as myocardial ischemia/reperfusion injury (MIRI). Until now, there is a dearth of efficacious strategies to prevent and manage MIRI. Hormones are specialized substances that are produced directly into the circulation by endocrine organs or tissues in humans and animals, and they have particular effects on the body. Hormonal medications utilize human or animal hormones as their active components, encompassing sex hormones, adrenaline medications, thyroid hormone medications, and others. While several studies have examined the preventive properties of different endocrine hormones, such as estrogen and hormone analogs, on myocardial injury caused by ischemia-reperfusion, there are other hormone analogs whose mechanisms of action remain unexplained and whose safety cannot be assured. The current study is on hormones and hormone medications, elucidating the mechanism of hormone pharmaceuticals and emphasizing the cardioprotective effects of different endocrine hormones. It aims to provide guidance for the therapeutic use of drugs and offer direction for the examination of MIRI in clinical therapy.

Abstract Cyclic dimeric guanosine monophosphate (c-di-GMP) serves as a bacterial second messenger that modulates various processes including biofilm formation, motility, and host-microbe symbiosis. Numerous studies have conducted comprehensive analysis of c-di-GMP. However, the mechanisms by which certain environmental signals such as iron control intracellular c-di-GMP levels are unclear. Here, we show that iron regulates c-di-GMP levels in Pseudomonas aeruginosa by modulating the interaction between an iron-sensing protein, IsmP, and a diguanylate cyclase, ImcA. Binding of iron to the CHASE4 domain of IsmP inhibits the IsmP-ImcA interaction, which leads to increased c-di-GMP synthesis by ImcA, thus promoting biofilm formation and reducing bacterial motility. Structural characterization of the apo-CHASE4 domain and its binding to iron allows us to pinpoint residues defining its specificity. In addition, the cryo-electron microscopy structure of ImcA in complex with a c-di-GMP analog (GMPCPP) suggests a unique conformation in which the compound binds to the catalytic pockets and to the membrane-proximal side located at the cytoplasm. Thus, our results indicate that a CHASE4 domain directly senses iron and modulates the crosstalk between c-di-GMP metabolic enzymes.

Adolescent binge drinking leads to long-lasting disorders of the adult central nervous system, particularly aberrant hippocampal neurogenesis. In this study, we applied in vivo fluorescent tracing using NestinCreERT2::Rosa26-tdTomato mice and analyzed the endogenous neurogenesis lineage progression of neural stem cells (NSCs) and dendritic spine formation of newborn neurons in the subgranular zone of the dentate gyrus. We found abnormal orientation of tamoxifen-induced tdTomato+ (tdTom+) NSCs in adult mice 2 months after treatment with EtOH (5.0 g/kg, i.p.) for 7 consecutive days. EtOH markedly inhibited tdTom+ NSCs activation and hippocampal neurogenesis in mouse dentate gyrus from adolescence to adulthood. EtOH (100 mM) also significantly inhibited the proliferation to 39.2% and differentiation of primary NSCs in vitro. Adult mice exposed to EtOH also exhibited marked inhibitions in dendritic spine growth and newborn neuron maturation in the dentate gyrus, which was partially reversed by voluntary running or inhibition of the mammalian target of rapamycin-enhancer of zeste homolog 2 pathway. In vivo tracing revealed that EtOH induced abnormal orientation of tdTom+ NSCs and spatial misposition defects of newborn neurons, thus causing the disturbance of hippocampal neurogenesis and dendritic spine remodeling in mice.

Summary: This study systematically reviewed the application of large language models (LLMs) in medicine, analyzing 550 selected studies from a vast literature search. LLMs like ChatGPT transformed healthcare by enhancing diagnostics, medical writing, education, and project management. They assisted in drafting medical documents, creating training simulations, and streamlining research processes. Despite their growing utility in assisted diagnosis and improving doctor-patient communication, challenges persisted, including limitations in contextual understanding and the risk of over-reliance. The surge in LLM-related research indicated a focus on medical writing, diagnostics, and patient communication, but highlighted the need for careful integration, considering validation, ethical concerns, and the balance with traditional medical practice. Future research directions suggested a focus on multimodal LLMs, deeper algorithmic understanding, and ensuring responsible, effective use in healthcare.

Abstract Background The vein of Marshall (VOM) ethanol infusion is increasingly performed in combination with catheter ablation in atrial fibrillation (AF). The cannulation of the VOM can sometimes be challenging. This study aimed to evaluate the double-wire technique in cases of difficult cannulation of the VOM. Case presentation Patients with AF scheduled for combined catheter ablation and VOM ethanol infusion were consecutively enrolled. The procedure was performed via the femoral vein. If the regular cannulation technique with one angioplasty wire failed or took more than 20 min, the double-wire technique using a stabilizing wire and a cannulation wire was performed. The unique technique was used mainly in two scenarios, when the Eustachian ridge was too prominent as a barrier for catheter manipulation or when the VOM ostium was close to the coronary sinus ostium. Of 162 patients scheduled for VOM ethanol infusion, the double-wire technique was applied in 6 (3.7%) patients and led to a 100% successful cannulation rate of the VOM. Of the six patients, two had a prominent Eustachian ridge, and four had a VOM ostium close to the coronary sinus ostium. The mean cannulation time was 33.3 ± 7.3 min. The ethanol infusion was successfully performed in 5 patients. One patient had a collateral circulation in the distal VOM, and ethanol infusion was not performed. Conclusions The double-wire technique can facilitate VOM cannulation and ethanol infusion in challenging cases. Word count : 231.

In this paper, the effects of laser heat input on the microstructures, tensile strength, and fatigue properties of Ti60 laser welded joints were investigated. The results show that with the increase in laser heat input, the macro morphology of the weld zone (WZ) changes from the Y-type to X-type. In the Y-type WZ, the porosity defects are almost eliminated. In contrast, there are a lot of porosity defects in the lower part of the X-type WZ. The microstructure of the base metal (BM) comprises equiaxed α phases, and β phases are mainly distributed at the boundaries of α phases. The heat-affected zone (HAZ) is comprised of α phases and acicular α′ phases, while the WZ mainly contains acicular α′ phases. With the increase in laser heat input, the quantity of the α phase gradually decreases and the acicular α′ phase gradually increases in the HAZ, and the size of the acicular α′ phase in the WZ gradually decreases. Due to the different microstructures, the hardness of BM is lower than the HAZ and WZ under different laser heat input conditions. In the tensile tests and low-cycle fatigue tests, the welded joints are fractured in BM. The porosity defects do not have decisive effects on the tensile and low-cycle fatigue properties of Ti60 laser welded joints.

Hexagonal boron nitride (h-BN) nanoparticles have attracted increasing attention due to their unique structure and properties. However, it is difficult to synthesize h-BN nanoparticles with uniform spherical morphology due to their crystal characteristic. The morphology control by tuning their precursor synthesis is a promising and effective strategy to solve this problem. Especially, the treatment temperature of precursors plays an important role in the morphology and surface area of h-BN nanoparticles. Herein, h-BN nanoparticles with different morphologies were synthesized via regulating the treatment temperature of precursors. The result shows that treatment temperature will affect the microstructure and state of precursor and further influence the morphology of h-BN products. Benefiting from the unique structure, the h-BN obtained using 250 °C precursors shows higher specific surface area (61.1 m2 g−1) than that of 85 °C (36.5 m2 g−1) and 145 °C (27.9 m2 g−1). h-BN products obtained using 250 °C precursors show higher specific surface area than that of 85 °C and 145 °C. The optimal condition for obtaining high-quality spherical h-BN is the pretreatment temperature of 250 °C and sintering temperature of 1300 °C. Importantly, compared with commercial h-BN nanoparticles, the synthesized h-BN nanoparticles show more uniform structure and larger specific surface area, indicating that sintering activity will be greatly improved. Furthermore, the reaction pathway and formation mechanism of h-BN was revealed by DFT calculations. The result shows that the five stationary states and five transition states exist in the reaction pathway, and the energy barrier can be overcome at high temperatures to form a ring h-BN. In view of its simplicity and efficiency, this work is promising for designing and guiding the synthesis of h-BN nanoparticles with uniform morphology.

The migration, proliferation, and apoptosis of trophoblastic cells play a crucial role in ensuring the effective preservation of pregnancy at the maternal–fetal interface. Any deviations in the structure and function of these cells might potentially result in the development of numerous pregnancy-related disorders, including missed abortion (MA). This study involved the examination of semaphorin 4A (SEMA4A) expression in missed abortion (n = 18) and normal early pregnancy (n = 18) villus. The findings of this study indicate a statistically significant decrease in the expression of SEMA4A in the villi of individuals diagnosed with missed abortion, as compared to the control group. The results of our vitro study showed that SEMA4A promoted the migration and proliferation of trophoblast cells and inhibited their apoptosis. Subsequent studies have shown that SEMA4A may be involved in regulating p-STAT3/STAT3, MMP9, bcl-2, and BAX levels. In summary, the findings of this study indicate a correlation between the decreased level of SEMA4A in chorionic villi and missed abortion. These results offer novel theoretical insights into the proper implantation and development of SEMA4A embryos at the maternal–fetal interface.

Abstract Background The impact of insulin resistance on the prognosis of heart failure with preserved ejection fraction (HFpEF) remains unknown. This study aimed to investigate the association between the triglyceride-glucose (TyG) index, an easily calculated marker of insulin resistance, and the long-term prognosis of HFpEF. Methods A total of 823 patients with HFpEF were enrolled in the study. The TyG index was determined using the formula ln(fasting triglycerides [mg/dL] × fasting glucose [mg/dL]/2). The primary endpoint was all-cause death. The secondary endpoints were cardiovascular (CV) death and heart failure (HF) rehospitalization. Restricted cubic spline, multivariate Cox proportional hazard models, and competing risk models were used for analyses. Results During a median follow-up period of 3.16 years, 147 (17.8%) all-cause deaths, 139 (16.8%) CV deaths, and 222 (27.0%) HF rehospitalizations occurred. Restricted cubic spline analysis revealed a J-shaped association between the TyG index and the mortality and rehospitalization rates. In the multivariate Cox proportional hazard models, compared with those in the lowest TyG index tertile, patients in the highest tertile exhibited the greatest susceptibility to all-cause death (HR 1.53, 95% CI 1.19–1.98) and CV death (HR 1.52, 95% CI 1.19–1.96). In the competing risk model, a significant association between the TyG index and HF rehospitalization was observed (HR 1.31, 95% CI, 1.07–1.61). Conclusion A high TyG index is associated with an increased risk of mortality and rehospitalization in patients with HFpEF. The TyG index may serve as a promising prognostic marker for patients with HFpEF.

Abstract Objective This study aimed to investigate the relationship between the TyG (Triglyceride-glucose index) and the prognosis of patients with HOCM (hypertrophic obstructive cardiomyopathy) without diabetes. Research design and methods A total of 713 eligible patients with HOCM were enrolled in this study and divided into two groups based on treatment: an invasive treatment group (n = 461) and a non-invasive treatment group (n = 252). The patients in both two groups were then divided into three groups based on their TyG index levels. The primary endpoints of this study were Cardiogenic death during long-term follow-up. Kaplan–Meier analysis was used to study the cumulative survival of different groups. Restricted cubic spline was used to model nonlinear relationships between the TyG index and primary endpoints. Myocardial perfusion imaging/Myocardial metabolic imaging examinations were performed to assess glucose metabolism in the ventricular septum of the HOCM patients. Results The follow-up time of this study was 41.47 ± 17.63 months. The results showed that patients with higher TyG index levels had better clinical outcomes (HR, 0.215; 95% CI 0.051,0.902; P = 0.036, invasive treatment group; HR, 0.179; 95% CI 0.063,0.508; P = 0.001, non-invasive treatment group). Further analysis showed that glucose metabolism in the ventricular septum was enhanced in HOCM patients. Conclusions The findings of this study suggest that the TyG index may serve as a potential protective factor for patients with HOCM without diabetes. The enhanced glucose metabolism in the ventricular septum of HOCM patients may provide a potential explanation for the relationship between the TyG index and HOCM prognosis.

Abstract Background Cardiometabolic disease is a clinical syndrome characterized by multiple metabolic disorders, with atherosclerosis as the core and cardiovascular and cerebrovascular events as the outcome. Drug research and development (R&D) in cardiometabolic diseases has grown rapidly worldwide. However, the development of cardiometabolic drug clinical trials in China remains unclear. This study aims to depict the changing landscape of drug clinical trials for cardiometabolic diseases in China during 2009–2021. Methods The detailed information of drug trials on cardiometabolic diseases registered in the National Medical Products Administration (NMPA) Registration and Information Disclosure Platform was collected between January 1, 2009, and July 1, 2021. The landscape of cardiometabolic drug clinical trials was analyzed by the characteristics, time trends, indications, pharmacological mechanisms, and geographical distribution. Results A total of 2466 drug clinical trials on cardiometabolic diseases were extracted and analyzed. The annual number of drug trials increased rapidly in the past twelve years. Among all the trials, the bioequivalence trials (1428; 58.3%) accounted for the largest proportion, followed by phase I (555; 22.5%), phase III (278; 11.3%), phase II (169; 6.9%), and phase IV (26; 1.1%). Of 2466 trials, 2133 (86.5%) trials were monomer drugs, only 236 (9.6%) trials were polypills and 97 (3.9%) were traditional Chinese medicine (TCM) compounds. In terms of pharmacological mechanisms, the number of trials in dihydropyridine (DHP) calcium antagonists 321 (11.9%) ranked first, while trials in angiotensin receptor blocker (ARB) 289 (10.7%) and dipeptidyl peptidase-4 (DPP-4) inhibitor 205 (7.6%) ranked second and third place respectively. Of 236 chemical polypills trials, 23 (9.7%) polypills were the combination of DHP calcium antagonists and statins, while others were the combination of two same pharmacological effect agents. As for the geographical distribution of leading units, 36 trials were led by principal investigators (PI) units from Beijing, followed by Jiangsu (n = 29), Shanghai (n = 19), Guangdong (n = 19), and Hunan (n = 19), showing an uneven regional distribution. Conclusions Great progress has been made in drug clinical trials on cardiometabolic diseases, especially in antihypertensive agents, hypoglycemic agents, and hypolipidemic agents. However, the insufficient innovation of first-in-class drugs and polypills should be carefully considered by all stakeholders in drug trials.

Heart failure has become a public health problem that we cannot avoid choosing to face in today's context. In the case of heart failure, pathological cardiac hypertrophy plays a major role because of its condition of absolute increase in ventricular mass under various stresses. Ferroptosis, it could be defined as regulatory mechanisms that regulate cell death in the absence of apoptosis in iron-dependent cells. This paper introduces various new research findings on the use of different regulatory mechanisms of cellular ferroptosis for the treatment of heart failure and cardiac hypertrophy, providing new therapeutic targets and research directions for clinical treatment. The role and mechanism of ferroptosis in the field of heart failure has been increasingly demonstrated, and the relationship between cardiac hypertrophy, which is one of the causes of heart failure, is also an area of research that we should focus on. In addition, the latest applications and progress of inducers and inhibitors of ferroptosis are reported in this paper, updating the breakthroughs in their fields.

The biomimetic strategy has been widely used in multiple fields, and provides huge convenience to human life. For biomedical application in vivo, the importance of biomimetic materials lies on mimicking the natural process/behavior as well as mimicking the structure/ingredient. Herein, recent advances of materials processing biomimetic recognition and assembly behavior for in vivo applications, including the natural source materials and artificial materials, are summarized. The biological processes are divided as recognition and assembly as well as recognition and self‐amplifying assembly. Finally, to conclude, the application of recognition and assembly process‐biomimetic materials in vivo in the future is discussed.

Background: The vein of Marshall (VOM) ethanol infusion improves rhythm control in atrial fibrillation (AF). The identification and cannulation of the VOM can be technically challenging. This study aimed to assess the angiographic morphology of the VOM and investigate its value in the VOM ethanol infusion. Methods: Patients with AF (n = 162) scheduled for combined catheter ablation and VOM ethanol infusion were enrolled. The VOM morphologic features in the right anterior oblique (RAO), the left anterior oblique (LAO), and the LAO cranial views were analyzed. The impact of morphology on the identification and cannulation of the VOM was investigated. Results: The VOM was identified in 159 (98.1 %) and cannulated in 150 (92.6 %) patients. The VOM identification rate in the RAO and LAO/LAO cranial view was 97.3 % and 89.3 %, respectively. Of 134 patients with VOM identification in the LAO/LAO cranial view, 104 (77.6 %) had a VOM ostium clock location (VOMoClock) of ≤3 and 3–4 o'clock. The VOM cannulation success rate in the ≤3, 3–4, 4–5, and 5–6 o'clock groups was 100 %, 92.6 %, 88.5 %, and 77.8 %, respectively (p = 0.032). Glioma is the most common tumor of the nervous system. The diffuse growth and proliferation of glioma poses great challenges for its treatment. Here, Transcriptomic analysis revealed that Rac GTPase activating protein 1 (RACGAP1) is highly expressed in glioma. RACGAP1 has been shown to play an important role in the malignant biological progression of a variety of tumors. However, the underlying role and mechanism in glioma remain poorly understood. By using quantitative real-time polymerase chain reaction (qRT-PCR), western blot, immunohistochemistry and Orthotopic mouse xenografts, we confirmed that knockdown of RACGAP1 impeded cell proliferation in glioma and prolonged the survival of orthotopic mice. Interestingly, we also found that inhibiting the expression of RACGAP1 reduced the expression of minichromosome maintenance 3 (MCM3) through RNA-seq and rescue assay, while Yin Yang 1 (YY1) transcriptionally regulated RACGAP1 expression. Furthermore, T7 peptide-decorated exosome (T7-exo) is regard as a promising delivery modality for targeted therapy of glioma, and the T7-siRACGAP1-exo significantly improved the survival time of glioma bearing mice. These results suggested that targeting RACGAP1 may be a potential strategy for glioma therapy.

Abstract Background Familial hypercholesterolemia (FH) is an inherited metabolic disorder with a high level of low-density lipoprotein cholesterol and the worse prognosis. The triglyceride-glucose (TyG) index, an emerging tool to reflect insulin resistance (IR), is positively associated with a higher risk of atherosclerotic cardiovascular disease (ASCVD) in healthy individuals, but the value of TyG index has never been evaluated in FH patients. This study aimed to determine the association between the TyG index and glucose metabolic indicators, insulin resistance (IR) status, the risk of ASCVD and mortality among FH patients. Methods Data from National Health and Nutrition Examination Survey (NHANES) 1999–2018 were utilized. 941 FH individuals with TyG index information were included and categorized into three groups: 9.0. Spearman correlation analysis was used to test the association of TyG index and various established glucose metabolism-related indicators. Logistic and Cox regression analysis were used to assess the association of TyG index with ASCVD and mortality. The possible nonlinear relationships between TyG index and the all-cause or cardiovascular death were further evaluated on a continuous scale with restricted cubic spline (RCS) curves. Abstract Although temozolomide (TMZ) provides significant clinical benefit for glioblastoma (GBM), responses are limited by the emergence of acquired resistance. Here, we demonstrate that exosomal circCABIN1 secreted from TMZ-resistant cells was packaged into exosomes and then disseminated TMZ resistance of receipt cells. CircCABIN1 could be cyclized by eukaryotic translation initiation factor 4A3 (EIF4A3) and is highly expressed in GBM tissues and glioma stem cells (GSCs). CircCABIN1 is required for the self-renewal maintenance of GSCs to initiate acquired resistance. Mechanistically, circCABIN1 regulated the expression of olfactomedin-like 3 (OLFML3) by sponging miR-637. Moreover, upregulation of OLFML3 activating the ErbB signaling pathway and ultimately contributing to stemness reprogramming and TMZ resistance. Treatment of GBM orthotopic mice xenografts with engineered exosomes targeting circCABIN1 and OLFML3 provided prominent targetability and had significantly improved antitumor activity of TMZ. In summary, our work proposed a novel mechanism for drug resistance transmission in GBM and provided evidence that engineered exosomes are a promising clinical tool for cancer prevention and therapy. Oxygen post annealing is a promising method for improving the quality of the SiC metal oxide semiconductor (MOS) interface without the introduction of foreign atoms. In addition, a low oxygen partial pressure annealing atmosphere would prevent the additional oxidation of SiC, inhibiting the generation of new defects. This work focuses on the effect and mechanism of low oxygen partial pressure annealing at different temperatures (900–1250 °C) in the SiO2/SiC stack. N2 was used as a protective gas to achieve the low oxygen partial pressure annealing atmosphere. X-ray photoelectron spectroscopy (XPS) characterization was carried out to confirm that there are no N atoms at or near the interface. Based on the reduction in interface trap density (Dit) and border trap density (Nbt), low oxygen partial pressure annealing is proven to be an effective method in improving the interface quality. Vacuum annealing results and time of flight secondary ion mass spectrometry (ToF-SIMS) results reveal that the oxygen vacancy (V[O]) filling near the interface is the dominant annealing mechanism. The V[O] near the interface is filled more by O2 in the annealing atmosphere with the increase in temperature.

Abstract Background The prognostic implication of liver fibrosis in acute coronary syndrome (ACS) patients are scarce. We sought to evaluate whether liver fibrosis scores (LFS) were associated with thrombotic or bleeding events in patients with acute coronary syndrome. Methods We included 6386 ACS patients who underwent percutaneous coronary intervention (PCI). This study determined liver fibrosis with aspartate aminotransferase to platelet ratio index (APRI), aspartate aminotransferase to alanine aminotransferase ratio (AST/ALT ratio), Forns score, and nonalcoholic fatty liver disease fibrosis score (NFS). The primary endpoint was major adverse cardiac and cerebrovascular events (MACCE), a composite of all-cause mortality (ACM), myocardial infarction (MI), and ischemic stroke (IS). Abstract Aims Recent studies suggested that both left ventricular ejection fraction (LVEF) lower than 60% or higher than 65% were associated with an increased mortality in the general population. Uncertainty remains regarding adverse outcomes across LVEF in coronary artery disease (CAD). The common understanding was that LVEF 40%. We aimed to evaluate the relationship between LVEF and mortality or adverse events in CAD patients undergoing percutaneous coronary intervention (PCI). Methods and results A total of 10 252 CAD patients undergoing PCI from an observational cohort were studied. All‐cause mortality and major adverse cardiovascular and cerebrovascular events (MACCE) were set as outcomes. Kaplan–Meier curves, adjusted Cox regression models, and restricted cubic spline analyses were used for evaluation. A total of 137 (1.3%) patients had all‐cause mortality, and 816 (8.0%) patients had MACCE during a median of 2.4 years of follow‐up. The median LVEF was 64%. All‐cause mortality and MACCE rates changed substantially across LVEF categories, and a linear inverse relationship of LVEF with all‐cause mortality and MACCE risk was observed. In this work, the response surface methodology (RSM) was used to model and optimize the hydroxylamine (HA) enhanced Fe(II)/peroxymonosulfate (PMS) process. The enhanced effect of HA on the degradation efficiency of Orange II (AO7) in the Fe(II)/PMS system was quantitatively analyzed. Pareto analysis showed that the individual and interactive effects of HA, Fe(II) and PMS were of the following order: HA > Fe(II) > PMS and Fe(II)/PMS > HA/PMS > Fe(II)/HA. The optimal conditions of HA/Fe(II)/PMS system were as follows: Fe(II) concentration was 34.0 μM, HA concentration was 0.4 mM, and PMS concentration was 0.9 mM. When the initial pH was 4.0–6.0, the degradation efficiency of AO7 in the HA/Fe(II)/PMS system was significantly higher than that in the Fe(II)/PMS system (P < 0.05). Hydroxylamine enhances the degradation of AO7 in the Fe(II)/PMS system by reducing Fe(III) to Fe(II). The results of quenching experiment showed that SO4•− was the dominating reactive oxygen species (ROS) in the HA/Fe(II)/PMS system. In the HA/Fe(II)/PMS system, CO32− and humic acid inhibited the degradation efficiency of AO7. This work provides a novel mathematical model for the degradation of AO7 in the HA/Fe(II)/PMS system, which can be popularized and applied in similar experiments. Previous research has revealed that graded pre-activation rather than specific lexical prediction is more likely to be the mechanism for the word predictability effect in English. However, whether graded pre-activation underlies the predictability effect in Chinese reading is unknown. Accordingly, the present study tested the generality of the graded pre-activation account in Chinese reading. We manipulated the contextual constraint of sentences and the predictability of target words as independent variables. Readers’ eye movement behaviors were recorded via an eye tracker. We examined whether processing an unpredictable word in a solid constraining context incurs a prediction error cost when this unpredictable word has a predictable alternative. The results showed no cues of prediction error cost on the early eye movement measures, supported by the Bayes Factor analyses. The current research indicates that graded predictive pre-activation underlies the predictability effect in Chinese reading.

Bone has a robust regenerative potential, but its capacity to repair critical-sized bone defects is limited. In recent years, stem cells have attracted significant interest for their potential in tissue engineering. Applying mesenchymal stem cells (MSCs) for enhancing bone regeneration is a promising therapeutic strategy. However, maintaining optimal cell efficacy or viability of MSCs is limited by several factors. Epigenetic modification can cause changes in gene expression levels without changing its sequence, mainly including nucleic acids methylation, histone modification, and non-coding RNAs. This modification is believed to be one of the determinants of MSCs fate and differentiation. Understanding the epigenetic modification of MSCs can improve the activity and function of stem cells. This review summarizes recent advances in the epigenetic mechanisms of MSCs differentiation into osteoblast lineages. We expound that epigenetic modification of MSCs can be harnessed to treat bone defects and promote bone regeneration, providing potential therapeutic targets for bone-related diseases.

The waste pomelo peel was pyrolyzed at 400 °C to prepare biochar and used as adsorbent to remove norfloxacin (NOR) from simulated wastewater. The adsorption conditions of norfloxacin by biochar were optimized by response surface methodology (RSM). On the basis of single-factor experiment, the adsorption conditions of biochar dosage, solution pH and reaction temperature were optimized by Box-Behnken Design (BBD), and the quadratic polynomial regression model of response value Y1 (NOR removal efficiency) and Y2 (NOR adsorption capacity) were obtained respectively. The results show that the two models are reasonable and reliable. The influence of single factor was as follows: solution pH > biochar dosage > reaction temperature. The interaction between biochar dosage and solution pH was very significant. The optimal adsorption conditions after optimization were as follows: biochar dosage = 0.5 g/L, solution pH = 3, and reaction temperature = 45 °C. The Y1 and Y2 obtained in the verification experiment were 75.68% and 3.0272 mg/g, respectively, which were only 2.38% and 0.0242 mg/g different from the theoretical predicted values of the model. Therefore, the theoretical model constructed by response surface methodology can be used to optimize the adsorption conditions of norfloxacin in water. In this study, a new type of iron/copper bimetallic combined with hydroxylamine (HA) activated peroxymonosulfate (PMS) was constructed to treat organic pollutants. Selecting the azo dye acid orange 7 (AO7) as the representative of organic pollutants, the new system can achieve nearly 100% degradation of AO7 within 5 min. The Fe(III)/Cu(II)/HA/PMS system mainly generates SO4·− to achieve the degradation of AO7 in acidic environment, while neutral and alkaline environments rely on ·OH. The presence of hydroxylamine accelerates the cycle of Fe(III)/Fe(II) and Cu(II)/Cu(I) in the system, and enhances the degradation ability of the system for organic pollutants. The addition of trace Cu(II) (1 μM) enhances the ability of a single Fe(III)/HA/PMS system to degrade AO7 in neutral and alkaline environments without causing secondary copper pollution. The common inorganic anions Cl− and NO3− in water have almost no effect on the degradation of AO7 in the system. The constructed Fe(III)/Cu(II)/HA/PMS system is an efficient and clean organic pollutant wastewater treatment process, which has very promising application prospects. Abstract Background Non-diabetic coronary artery disease (CAD) patients are thought to encounter metabolic dysfunction and while these changes may be imperceptible to the patient they probably influence outcomes. At present, there is no system to support patients sensing these subtle changes, nor is there an established model for prognoses. The Atherogenic Index of Plasma (AIP) index has already proven useful for atherosclerosis although further research is needed, especially for those without hyperglycemia. Methods This is a prospective study of 5538 non-diabetic CAD patients who had received percutaneous coronary intervention (PCI). Participants were assigned to one of three groups according to their AIP index. High AIP index cases were then compared to low index patients according to major adverse cardiac events (MACE). Restricted cubic spline (RCS) analysis was also conducted to investigate interrelations between AIP index levels and hazard ratios (HR) for MACEs. Results Patients with a high AIP index encountered metabolic dysfunction compared to those with a low AIP index i.e., higher Body Mass Index (BMI), Total Cholesterol (TC), Triglycerides (TG), and uric acid as well as lower HDL-C. Each of the aforementioned interrelations were significant with p values of less than 0.001. There was also a significant increase in the number of MACEs in the high AIP index group compared to the low AIP index group (HR: 1.37, 95% CI 1.04–1.81; p = 0.025). A J-shaped RCS curve highlighted a change in the HR after the 0.18 juncture (HR per SD: 1.20, 95% CI 0.96–1.50). Further subgroup analysis supported the main findings, all with HRs greater than one. Conclusion The AIP index could be used in prognostics for non-diabetic CAD patients 2 years after PCI. The relationship between hazard ratio and the AIP index appears to be J-shaped. Although, further multi-center studies designed for non-diabetic patients with potential metabolic dysfunction should be conducted to determine the value of the AIP index.

Body-protective compound (BPC) 157 demonstrates protective effects against damage to various organs and tissues. For future clinical applications, we had previously established a solid-phase synthesis process for BPC157, verified its biological activity in different wound models, and completed preclinical safety evaluations. This study aimed to investigate the pharmacokinetics, excretion, metabolism, and distribution profiles of BPC157. After a single intravenous (IV) administration, single intramuscular (IM) administrations at three doses in successive increments along with repeated IM administrations, the elimination half-life (t1/2) of prototype BPC157 was less than 30 min, and BPC157 showed linear pharmacokinetic characteristics in rats and beagle dogs at all doses. The mean absolute bioavailability of BPC157 following IM injection was approximately 14%–19% in rats and 45%–51% in beagle dogs. Using [3H]-labeled BPC157 and radioactivity examination, we proved that the main excretory pathways of BPC157 involved urine and bile. [3H]BPC157 was rapidly metabolized into a variety of small peptide fragments in vivo, thus forming single amino acids that entered normal amino acid metabolism and excretion pathways. In conclusion, this study provides the first analysis of the pharmacokinetics of BPC157, which will be helpful for its translation in the clinic.

ObjectivesThis study aimed to find novel oxidative stress (OS)-related biomarkers of osteoporosis (OP), together with targeting the macromolecule Mitogen-activated protein kinase-activated protein kinase 2 (MAPKAPK2) protein to further discover potential novel materials based on an advanced structural biology approach.MethodsGene expression profiles of GSE35958 were obtained from the Gene Expression Omnibus (GEO) database, which were included for weighted gene co-expression network analysis (WGCNA) and differential analysis to identify the most correlated module, to identify OS-related hub genes in the progression of OP. Functional annotations were also analyzed on the interested module to get a comprehensive understanding of these genes. Then, a series of advanced structural biology methods, including high-throughput screening, pharmacological characteristic prediction, precise molecular docking, molecular dynamics simulation, etc., was implemented to discover novel natural inhibitor materials against the MAPKAPK2 protein.ResultsThe brown module containing 720 genes was identified as the interested module, and a group set of genes was determined as the hub OS-related genes, including PPP1R15A, CYB5R3, BCL2L1, ABCD1, MAPKAPK2, HSP90AB1, CSF1, RELA, P4HB, AKT1, HSP90B1, and CTNNB1. Functional analysis demonstrated that these genes were primarily enriched in response to chemical stress and several OS-related functions. Then, Novel Materials Discovery demonstrated that two compounds, ZINC000014951634 and ZINC000040976869, were found binding to MAPKAPK2 with a favorable interaction energy together with a high binding affinity, relatively low hepatoxicity and carcinogenicity, high aqueous solubility and intestinal absorption levels, etc., indicating that the two compounds were ideal potential inhibitor materials targeting MAPKAPK2.ConclusionThis study found a group set of OS-related biomarkers of OP, providing further insights for OS functions in the development of OP. This study then focused on one of the macromolecules, MAPKAPK2, to further discover potential novel materials, which was of great significance in guiding the screening of MAPKAPK2 potential materials.

Summary: Many viruses encode RNA-modifying enzymes to edit the 5′ end of viral RNA to mimic the cellular mRNA for effective protein translation, genome replication, and evasion of the host defense mechanisms. Alphavirus nsP1 synthesizes the 5′ end Cap-0 structure of viral RNAs. However, the molecular basis of the capping process remains unclear. We determine high-resolution cryoelectron microscopy (cryo-EM) structures of Chikungunya virus nsP1 in complex with m7GTP/SAH, covalently attached m7GMP, and Cap-0 viral RNA. These structures reveal details of viral-RNA-capping reactions and uncover a sequence-specific virus RNA-recognition pattern that, in turn, regulates viral-RNA-capping efficiency to ensure optimal genome replication and subgenomic RNA transcription. This sequence-specific enzyme-RNA pairing is conserved across all alphaviruses.

A back-propagation neural network (BPNN) was used to model and optimize the process of hydroxylamine (HA)-enhanced Fe2+ activating peroxymonosulfate (PMS). Using HA-enhanced Fe2+ to activate PMS is a cost-effective method to degrade orange II (AO7). We investigated the individual and interactive effects of the concentrations of Fe2+, HA, and PMS on the degradation of AO7. The R2 of the BPNN model was 0.99852, and the data were distributed around y = x. Sensitivity analysis showed that the relative importance of each factor was as follows: HA > Fe2+ > PMS. The optimized results obtained by the genetic algorithm were as follows: the concentration of Fe2+ was 35.33 μmol·L−1, HA was 0.46 mmol·L−1, and PMS was 0.93 mmol·L−1. Experiments verified that the AO7 degradation effect within 5 min was 95.7%, whereas the predicted value by the BPNN was 96.2%. The difference between predicted and experimental values is 0.5%. This study provides a new tool (machine learning) to accurately predict the concentrations of HA, Fe2+, and PMS to degrade AO7 under various conditions.

Abstract. Background:. Thyroid dysfunction is associated with cardiovascular diseases. However, the role of thyroid function in lipid metabolism remains partly unknown. The present study aimed to investigate the causal association between thyroid function and serum lipid metabolism via a genetic analysis termed Mendelian randomization (MR). Methods:. The MR approach uses a genetic variant as the instrumental variable in epidemiological studies to mimic a randomized controlled trial. A two-sample MR was performed to assess the causal association, using summary statistics from the Atrial Fibrillation Genetics Consortium (n = 537,409) and the Global Lipids Genetics Consortium (n = 188,577). The clinical measures of thyroid function include thyrotropin (TSH), free triiodothyronine (FT3) and free thyroxine (FT4) levels, FT3:FT4 ratio and concentration of thyroid peroxidase antibodies (TPOAb). The serum lipid metabolism traits include total cholesterol (TC) and triglycerides, high-density lipoprotein, and low-density lipoprotein (LDL) levels. The MR estimate and MR inverse variance-weighted method were used to assess the association between thyroid function and serum lipid metabolism. Results:. The results demonstrated that increased TSH levels were significantly associated with higher TC (β = 0.052, P = 0.002) and LDL (β = 0.041, P = 0.018) levels. In addition, the FT3:FT4 ratio was significantly associated with TC (β = 0.240, P = 0.033) and LDL (β = 0.025, P = 0.027) levels. However, no significant differences were observed between genetically predicted FT4 and TPOAb and serum lipids. Conclusion:. Taken together, the results of the present study suggest an association between thyroid function and serum lipid metabolism, highlighting the importance of the pituitary-thyroid-cardiac axis in dyslipidemia susceptibility.

Infant formula, an important food for babies, is convenient and nutritious, and hydrolyzed formulas have attracted much attention due to their non-allergicity. However, it is uncertain whether hydrolyzed formulars cause obesity and other side effects in infants. Herein, three infant formulas, standard (sIF), partially hydrolyzed (pHIF), and extensively hydrolyzed (eHIF), were analyzed in an in vitro gastrointestinal digestion model. With increasing degree of hydrolysis, the protein moleculars, and allergenicity of the proteins decreased and the long-chain polyunsaturated fatty acid content increased. Moreover, the digestion model solutions quickly digested the small fat globules and proteins in the hydrolyzed formula, allowing it to become electrostatically stable sooner. The eHIF-fed mice presented larger body sizes, and exhibited excellent exploratory and spatial memory abilities in the maze test. Based on villus height and crypt depth histological characterizations and amplicon sequencing, eHIF promoted mouse small intestine development and changed the gut microbiota composition, eventually favoring weight gain. The mouse spleen index showed that long-term infant formula consumption might be detrimental to immune system development, and the weight-bearing swimming test showed that eHIF could cause severe physical strength decline. Therefore, long-term consumption of infant formula, especially eHIF, may have both positive and negative effects on mouse growth and development, and our results might shed light on feeding formula to infants.

Sorafenib is one of the few effective first-line drugs approved for the treatment of advanced hepatocellular carcinoma (HCC). However, the development of drug resistance is common among individuals with HCC. Recent evidence indicated that the anticancer activity of sorafenib mainly relies on the induction of ferroptosis. Furthermore, in our study, genes that suppress ferroptosis, especially GPX4 and DHODH, were enriched in sorafenib-resistant cells and primary tissues and were associated with poor prognosis of HCC patients who received sorafenib treatment. Therefore, a new ferroptosis inducer comprising a multiplex small interfering RNA (multi-siRNA) capable of simultaneously silencing GPX4 and DHODH was created. Then, exosomes with high multi-siRNA loading and HCC-specific targeting were established by fusing the SP94 peptide and the N-terminal RNA recognition motif (RRM) of U1-A with the exosomal membrane protein Lamp2b. The results from the in vitro and in vivo experiments indicate that this tumor-targeting nano-delivery system (ExoSP94-lamp2b-RRM-multi-siRNA) could enhance sorafenib-induced ferroptosis and overcome sorafenib resistance. Taken together, HCC-targeted exosomes (ExoSP94-Lamp2b-RRM) could specifically deliver multi-siRNA to HCC tissues, enhance sorafenib-induced ferroptosis by silencing GPX4 and DHODH expression and consequently increase HCC sensitivity to sorafenib, which opens a new avenue for clinically overcoming sorafenib resistance from the perspective of ferroptosis.

Aluminum grating has wide applications in laser systems and photoelectric equipment. Research on the laser damage characteristics of aluminum grating has guiding significance and application value for improving the laser damage resistance. The aim of this study is to investigate the characteristics of damage induced by nanosecond pulsed lasers on the aluminum grating. To better understand the laser damage characteristics of aluminum grating, herein, Maxwell’s equations were numerically solved according to the finite difference time-domain method, and the electric field model of 1,064 nm Gaussian laser damage aluminum grating was established. The simulation results showed that the light field is modulated by the grating; furthermore, the maximum value of the electric field occurred at the ridge of the grating when the laser is irradiated vertically. Analysis suggested that the electric field distribution is in accordance with the laser energy distribution, and the distribution region of the maximum electric field is a vulnerable location. To further verify the local electric field enhancement effect, based on the 1-on-1 laser damage measurement method, an experimental study of the nanosecond laser (@1,064 nm, 6.5 ns) damage to the aluminum grating was carried out. Moreover, the damage morphology was analyzed using a scanning electron microscope (SEM), and the simulation results showed good agreement with the experimental results.

A lot of previous research has highlighted the negative consequences of Internet addiction. However, relatively few is known about the underlying mechanism for Internet addiction among college students in relation to family function. The present study explored the relationship between family function and Internet addiction among college students, as well as the mediating effects of alexithymia and loneliness. A sample of 783 Chinese college students were administered a number of psychometric scales including the “General Function” subscale of the Chinese version of the Family Assessment Device, Toronto Alexithymia Scale, UCLA Loneliness Scale, and Revised Chinese Internet Addiction Scale. The results showed that family function was negatively associated with Internet addiction; the association was significantly mediated by alexithymia; the association was significantly mediated by loneliness; and alexithymia and loneliness sequentially mediated the association. The total mediating effect was 63.96%. The results of the present study are of great significance to the prevention and intervention of Internet addiction among college students.

BackgroundObservational studies have shown an association between early age at menarche (AAM) and myocardial infarction (MI) with recorded cases. In this Mendelian randomization (MR) study, we used large amounts of summary data from genome-wide association studies (GWASs) to further estimate the association of genetically predicted AAM with genetically predicated risk of MI and investigate to what extent this association is mediated by genetically determined lifestyles, cardiometabolic factors, and estrogen exposure.MethodsA two-step, two-sample MR study was performed by mediation analysis. Genetic variants identified by GWAS meta-analysis of reproductive genetics consortium (n = 182,416) were selected for genetically predicted AAM. Genetic variants identified by the Coronary ARtery DIsease Genome-wide Replication and Meta-analysis plus The Coronary Artery Disease Genetics Consortium (n = 184,305) were selected for genetically predicted risk of MI. Genetic variants from other international GWAS summary data were selected for genetically determined mediators.ResultsThis MR study showed that increase in genetically predicted AAM was associated with lower risk of genetically predicted MI (odds ratio 0.91, 95% confidence interval 0.84–0.98). Inverse variance weighted (IVW) MR analysis also showed that decrease in genetically predicted AAM was associated with higher genetically predicted alcohol intake frequency, current smoking behavior, higher waist-to-hip ratio, and higher levels of systolic blood pressure (SBP), fasting blood glucose, hemoglobin A1c (HbA1c), and triglycerides (TGs). Furthermore, increase in genetically predicted AAM was associated with genetically predicted longer sleep duration, higher levels of high-density lipoproteins, and older age at which hormone replacement therapy was started. The most essential mediators identified were genetically predicted current smoking behavior and levels of HbA1c, SBP, and TGs, which were estimated to genetically mediate 13.9, 12.2, 10.5, and 9.2%, respectively, with a combined mediation proportion of 37.5% in the association of genetically predicted AAM with genetically predicted increased risk of MI in an MR framework.ConclusionOur MR analysis showed that increase in genetically predicted AAM was associated with lower genetically predicted risk of MI, which was substantially mediated by genetically determined current smoking behavior and levels of HbA1c, SBP, and TGs. Intervening on the above mediators may reduce the risk of MI.

The submodule capacitor voltage fluctuation of the modular multilevel converter (MMC) leads to the increase of the size of the submodule capacitor, the increase of system cost, and the decrease of system reliability and stability. In order to solve the problem, the mathematical model of circulating current voltage drop is derived, and the mechanism of circulating current on the submodule capacitor voltage fluctuation is studied through the submodule switching function. A control strategy for suppressing the submodule capacitor voltage fluctuation is proposed, which is based on circulating current voltage drop balance (CCVDB). The simulation model is built to compare three control strategies for suppressing submodule capacitor voltage fluctuation under the step changes of DC bus voltage and active power, namely, the circulating current is suppressed to zero, the second-order component of the submodule capacitor voltage is suppressed to zero, and the CCVDB strategy. The research results show that the CCVDB strategy can effectively suppress the submodule capacitor voltage fluctuation, reduce the capacitance demand, and reduce the total harmonic distortion (THD) of the submodule capacitor voltage, which verifies the feasibility and effectiveness of the CCVDB strategy.

The scatterometer onboard the China-France Oceanography Satellite (CFOSAT) is the first rotating fan beam scatterometer (CSCAT) in operation. It is used to measure the ocean surface wind fields over a large observation swath. Geolocation is an important part of the preprocessing of scatterometer data. The accuracy of geolocation has a large impact on the accuracy of wind field retrieval. It is necessary to estimate and analyze the geolocation processing of CSCAT. The challenge is understanding how to estimate the geolocation errors of CSCAT. In this study, the coastline inflection method is used to estimate the geolocation errors of CSCAT based on the distribution characteristics of the backscatter coefficients at both the ocean and the land. The coastline inflection points are obtained using the gradient changes of backscatter coefficients near the coastline. The locations of these points are in line with the trend of the high-precision coastline data. The geolocation errors are estimated by comparing these points with the high-precision coastline. The results show that the current geolocation errors of CSCAT are about 7.31 km, which meets the requirements of 25 × 25 km wind field retrieval. In addition, because the geolocation of scatterometer data is affected by terrain, the geolocation errors of CSCAT are corrected based on an elevation correction algorithm using high-precision digital elevation model data. The results verify the effectiveness of the algorithm.

The microwave scatterometer onboard the China-France Oceanography Satellite (CFOSAT Scatterometer, CSCAT) is the first rotating fan-beam scatterometer (RFSCAT) in operation. It is used to obtain the ocean surface wind field by measuring the backscattering coefficient over a large observation swath. To estimate the backscattering coefficient, the scatterometer needs to measure the scattered signal accurately. The process of removing noise from the echo signal (scattered signal plus noise) is called noise subtraction. Noise subtraction has a great influence on the measurement of the backscattering coefficient and subsequent wind field retrieval. Therefore, accurate noise subtraction is necessary and important. In this article, an improved noise subtraction method for CSCAT is proposed. According to the distribution characteristics of backscattering coefficients, noise correction factors are introduced to improve the noise subtraction of CSCAT. The ocean surface wind fields are obtained using the improved backscattering coefficient data, and the deviation of wind speed and direction are calculated by comparison with the European Centre for Medium Range Weather Forecasts wind fields. The results show that the improved noise subtraction method can lead to improvement of the quality of wind field effectively. In addition, based on the improved method of noise subtraction, a method of estimating the noise correction factor is proposed at a large incidence angle, furtherly improving the noise subtraction method. The results show that the proposed method is effective.

The purpose of this paper is to put forward a decision model with wide applicability and differentiated decision scheme scores so as to improve the ability of students to learn during a water engineering economics course. The main novelty and contributions of this paper are that the multi-attribute decision-making method proposed is more objective and does not require rich subjective experience from decision-makers in the application process, which is particularly suitable for beginners who are learning in a water engineering economics course. The method involves standardizing each index value of the decision scheme first, constructing the objective function of maximum entropy distribution, calculating the weight of each index by the genetic algorithm, and finally ranking the pros and cons of the scheme according to the score of each scheme. The example results of three water engineering scheme decisions show that the maximum entropy model proposed in this paper can achieve reasonable decision results, and there is a large degree of differentiation between the decision schemes. The proposed scheme, a decision maximum entropy model, has wide applicability, can improve the rationality of the decisions made regarding water engineering schemes, and can be popularized and applied when teaching decision-making in water engineering economics courses.

Alpine lakes, which include glacial and nonglacial lakes, are widely distributed in high mountain areas and are sensitive to climate and environmental changes. Remote sensing is an effective tool for identifying alpine lakes over large regions, but in the case of small lakes, the complex terrain and extreme weather make their accurate identification extremely challenging. This paper presents an automated method for alpine lake identification developed by leveraging deep learning algorithms and multi-source high-resolution satellite data. The method is able to detect the outlines and types of alpine lakes from high-resolution optical and Synthetic Aperture Radar (SAR) satellite data. In this study, a total of 4584 alpine lakes (including 2795 glacial lakes) were identified in the Eastern Himalayas from Sentinel-1 and Sentinel-2 data acquired during 2016–2020. The average area of the lakes was 0.038 km2, and the average elevation was 4974 m. High accuracy was reported for the dataset for both segmentation (mean Intersection Over Union (MIoU) > 72%) and classification (Overall Accuracy, User’s and Producer’s Accuracies, and F1-Score are all higher than 85%). A higher accuracy was found for the combination of optical and SAR data than relying on single-sourced data, for which the MIoU increased by at least 12%, suggesting that the combination of optical and SAR data is critical for improving the identification of alpine lakes. The deep learning-based method demonstrated a significant improvement over traditional spectral extraction methods.

In order to control the discharge of regional total pollutants in the region and improve the ability of water environment management and decision making, a multi-objective decision-making optimization model of water pollution load allocation was constructed, which took into account economy and fairness. The model takes the maximum environmental benefit and the minimum weighted comprehensive Gini coefficient as the objective function and takes into account the uncertainty and multi-objectives of the model, which is conducive to promoting economic development and ensuring the fairness of regional water pollutant discharge. A method based on Monte Carlo simulation coupled with a genetic algorithm was designed to obtain the optimal solution set through multiple simulation optimization. This model is applied to Anhui Province to solve the allocation optimization problem of total pollutant reduction in the 13th Five-Year Energy Conservation and Emission Reduction Plan. After the optimization of water pollution load distribution, the comprehensive Gini coefficients of COD and NH3-N are reduced by different ranges. The comprehensive Gini coefficient after COD optimization decreased by 2.4–4.6%, and the comprehensive Gini coefficient after NH3-N optimization decreased by 25.1–32.5%, which verified the feasibility and rationality of the model in the optimal allocation of the total discharge of regional water pollutants. The model takes into account uncertain subjective and objective factors that have an important impact on water pollutant discharge targets and decision variables, thus optimizing the total emissions of the entire regional control unit in both space and time.
