A view that has been gaining prevalence over the past decade is that the human conceptual system is malleable, dynamic, context-dependent, and task-dependent, that is, flexible. Within the flexible conceptual representation framework, conceptual representations are constructed ad hoc, forming a different, idiosyncratic instantiation upon each occurrence. In this review, we scrutinize the neurocognitive literature to better understand the nature of this flexibility. First, we identify some key characteristics of these representations. Next, we consider how these flexible representations are constructed by addressing some of the open questions in this framework: We review the age-old question of how to reconcile flexibility with the apparent need for shareable stable definitions to anchor meaning and come to mutual understanding, as well as some newer questions we find critical, namely, the nature of relations among flexible representations, the role of feature saliency in activation, and the viability of all-or-none feature activations. We suggest replacing the debate about the existence of a definitional stable core that is obligatorily activated with a question of the degree and probability of activation of the information constituting a conceptual representation. We rely on published works to suggest that (1) prior featural salience matters, (2) feature activation may be graded, and (3) Bayesian updating of prior information according to current demands offers a viable account of how flexible representations are constructed. This proposal provides a theoretical mechanism for incorporating a changing momentary context into a constructed representation, while still preserving some of the concept's constituent meaning.

Sentence-length event-related potential (ERP) waveforms were obtained from 23 scalp sites as 24 subjects listened to normally spoken sentences of various syntactic structures. The critical materials consisted of 36 sentences each containing one of 2 types of relative clauses that differ in processing difficulty, namely Subject Object (SO) and Subject Subject (SS) relative clauses. Sentence-length ERPs showed several differences in the slow scalp potentials elicited by SO and SS sentences that were similar in their temporal dynamics to those elicited by the same stimuli in a word-by-word reading experiment, although the effects in the two modalities have non identical distributions. Just as for written sentences, there was a large, fronto-central negativity beginning at the linguistically defined "gap" in the SO sentences; this effect was largest for listeners with above-median comprehension rates, and is hypothesized to index changes in on-line processing demands during comprehension.

To investigate the neural processing of different word categories, we recorded event-related potentials (ERPs) from 32 individuals listening to sentences, beginning either with a proper name (first name), the subject's own name, or a common noun. Names and nouns both elicited ERP waveforms with the same early componentry, but the N1 and P2 components were larger for proper names than common nouns. The ERPs to the subject's own name also had a large N1/P2 plus a prominent negativity at parieto-central site peaking around 400 ms and a late positivity between 500-800 ms over left lateral-frontal sites. These findings are consistent with differential processing of people's first names within the category of nouns.

Reliable biomarkers of memory decline are critical for the early detection of Alzheimer's disease. Previous work has found three EEG measures, namely the event-related brain potential P600, suppression of oscillatory activity in the alpha frequency range (∼10 Hz) and cross-frequency coupling between low theta/high delta and alpha/beta activity, each of which correlates strongly with verbal learning and memory abilities in healthy elderly and patients with mild cognitive impairment or prodromal Alzheimer's disease. In the present study, we address the question of whether event-related or oscillatory measures, or a combination thereof, best predict the decline of verbal memory in mild cognitive impairment and Alzheimer's disease. Single-trial correlation analyses show that despite a similarity in their time courses and sensitivities to word repetition, the P600 and the alpha suppression components are minimally correlated with each other on a trial-by-trial basis (generally |r s| < 0.10). This suggests that they are unlikely to stem from the same neural mechanism. Furthermore, event-related brain potentials constructed from bandpass filtered (delta, theta, alpha, beta or gamma bands) single-trial data indicate that only delta band activity (1-4 Hz) is strongly correlated (r = 0.94, P < 0.001) with the canonical P600 repetition effect; event-related potentials in higher frequency bands are not. Importantly, stepwise multiple regression analyses reveal that the three event-related brain potential/oscillatory measures are complementary in predicting California Verbal Learning Test scores (overall R 2 ' s in 0.45-0.63 range). The present study highlights the importance of combining EEG event-related potential and oscillatory measures to better characterize the multiple mechanisms of memory failure in individuals with mild cognitive impairment or prodromal Alzheimer's disease.

How does individual-level variation in experience andknowledge influence neural mechanisms recruited during real-time language comprehension? We used event-related brainpotentials (ERPs) combined with lateralized visualpresentations of critical sentence-final words to examineasymmetries in hemispheric processing as individuals whovaried in their knowledge of the fictional world of Harry Potter(HP) read sentences about general topics / HP. HP sentenceendings were either contextually supported, unrelatedanomalies, or semantically related anomalies. Amongst HP“experts,” both hemispheres were sensitive to contextualsupport, but only the right hemisphere (RH) was sensitive tothe related anomaly manipulation. The exact pattern of resultsdepended on the relationship (categorical vs event). Ourfindings are in line with accounts on which the left hemisphere(LH) activates narrow/specific semantic contents and the RHactivates a broader range. We tentatively hypothesize thatcontent experts may exploit these hemispheric differences inscope of activation.

In Troyer and Kutas (2018), individual differences in knowledge of the world of Harry Potter (HP) rapidly modulated individuals' average electrical brain potentials to contextually supported words in sentence endings. Using advances in single-trial electroencephalogram analysis, we examined whether this relationship is strictly a result of domain knowledge mediating the proportion of facts each participant knew; we find it is not. Participants read sentences ending in a contextually supported word, reporting online whether they had known each fact. Participants' reports correlated with HP domain knowledge and reliably modulated event-related brain potentials to sentence-final words within 250 ms. Critically, domain knowledge had a dissociable influence in the same time window for endings that participants reported not having known and/or were less likely to be known/remembered across participants. We hypothesize that knowledge impacts written word processing primarily by affecting the neural processes of (implicit) retrieval from long-term memory (LTM): Greater knowledge eases otherwise difficult retrieval processes.

Uncanny valley refers to humans' negative reaction to almost-but-not-quite-human agents. Theoretical work proposes prediction violation as an explanation for uncanny valley but no empirical work has directly tested it. Here, we provide evidence that supports this theory using event-related brain potential recordings from the human scalp. Human subjects were presented images and videos of three agents as EEG was recorded: a real human, a mechanical robot, and a realistic robot in between. The real human and the mechanical robot had congruent appearance and motion whereas the realistic robot had incongruent appearance and motion. We hypothesize that the appearance of the agent would provide a context to predict her movement, and accordingly the perception of the realistic robot would elicit an N400 effect indicating the violation of predictions, whereas the human and the mechanical robot would not. Our data confirmed this hypothesis suggesting that uncanny valley could be explained by violation of one's predictions about human norms when encountered with realistic but artificial human forms. Importantly, our results implicate that the mechanisms underlying perception of other individuals in our environment are predictive in nature.

Visual narratives sometimes depict successive images with different characters in the same physical space; corpus analysis has revealed that this occurs more often in Japanese manga than American comics. We used event-related brain potentials to determine whether comprehension of "visual narrative conjunctions" invokes not only incremental mental updating as traditionally assumed, but also, as we propose, "grammatical" combinatoric processing. We thus crossed (non)/conjunction sequences with character (in)/congruity. Conjunctions elicited a larger anterior negativity (300-500 ms) than nonconjunctions, regardless of congruity, implicating "grammatical" processes. Conjunction and incongruity both elicited larger P600s (500-700 ms), indexing updating. Both conjunction effects were modulated by participants' frequency of reading manga while growing up. Greater anterior negativity in frequent manga readers suggests more reliance on combinatoric processing; larger P600 effects in infrequent manga readers suggest more resources devoted to mental updating. As in language comprehension, it seems that processing conjunctions in visual narratives is not just mental updating but also partly grammatical, conditioned by comic readers' experience with specific visual narrative structures.

In grapheme-color synesthesia, seeing particular letters or numbers evokes the experience of specific colors. We investigate the brain's real-time processing of words in this population by recording event-related brain potentials (ERPs) from 15 grapheme-color synesthetes and 15 controls as they judged the validity of word pairs ('yellow banana' vs. 'blue banana') presented under high and low visual contrast. Low contrast words elicited delayed P1/N170 visual ERP components in both groups, relative to high contrast. When color concepts were conveyed to synesthetes by individually tailored achromatic grapheme strings ('55555 banana'), visual contrast effects were like those in color words: P1/N170 components were delayed but unchanged in amplitude. When controls saw equivalent colored grapheme strings, visual contrast modulated P1/N170 amplitude but not latency. Color induction in synesthetes thus differs from color perception in controls. Independent from experimental effects, all orthographic stimuli elicited larger N170 and P2 in synesthetes than controls. While P2 (150-250ms) enhancement was similar in all synesthetes, N170 (130-210ms) amplitude varied with individual differences in synesthesia and visual imagery. Results suggest immediate cross-activation in visual areas processing color and shape is most pronounced in so-called projector synesthetes whose concurrent colors are experienced as originating in external space.

Real-time sentence processing involves connecting linguistic inputwith knowledge. Here, we ask how variability in semantic memory(specific domain knowledge) may influence semantic access in real-time sentence processing. We recorded EEG while participantsmore/less knowledgeable about the narrative world of Harry Potter(HP) read sentences. In Experiment 1, all participants showed N400predictability effects for general-knowledge sentences, but onlythose with high HP knowledge showed predictability effects forsentences about Harry Potter. This effect was driven by graded brainresponses to predictable endings as a function of knowledge.Experiment 2 revealed greater semantic activation (inferred fromN400 effects) for HP items participants reported knowing. High-knowledge participants also showed greater semantic activation foritems they reported not knowing/remembering. These findingssuggest that amount and/or functional organization of knowledgehas real-time consequences on written sentence processing andimplicate implicit/partial access to domain knowledge for expertswhen information is not explicitly recalled.

Language comprehension requires access to stored knowledge and the ability to combine knowledge in new, meaningful ways. Previous work has shown that processing linguistically more complex expressions ('Texas cattle rancher' vs. 'rancher') leads to slow-downs in reading during initial processing, possibly reflecting effort in combining information. Conversely, when this information must subsequently be retrieved (as in filler-gap constructions), processing is facilitated for more complex expressions, possibly because more semantic cues are available during retrieval. To follow up on this hypothesis, we tested whether information distributed across a short discourse can similarly provide effective cues for retrieval. Participants read texts introducing two referents (e.g., two senators), one of whom was described in greater detail than the other (e.g., 'The Democrat had voted for one of the senators, and the Republican had voted for the other, a man from Ohio who was running for president'). The final sentence (e.g., 'The senator who the {Republican/Democrat}had voted for…') contained a relative clause picking out either the Many-Cue referent (with 'Republican') or the One-Cue referent (with 'Democrat'). We predicted facilitated retrieval (faster reading times) for the Many-Cue condition at the verb region ('had voted for'), where readers could understand that 'The senator' is the object of the verb. As predicted, this pattern was observed at the retrieval region and continued throughout the rest of the sentence. Participants also completed the Author/Magazine Recognition Tests (ART/MRT; Stanovich and West, 1989), providing a proxy for world knowledge. Since higher ART/MRT scores may index (a) greater experience accessing relevant knowledge and/or (b) richer/more highly structured representations in semantic memory, we predicted it would be positively associated with effects of elaboration on retrieval. We did not observe the predicted interaction between ART/MRT scores and Cue condition at the retrieval region, though ART/MRT interacted with Cue condition in other locations in the sentence. In sum, we found that providing more elaborative information over the course of a text can facilitate retrieval for referents, consistent with a framework in which referential elaboration over a discourse and not just local linguistic information directly impacts information retrieval during sentence processing.

BackgroundFragile X-associated tremor/ataxia syndrome (FXTAS), a neurodegenerative disorder caused by FMR1 gene premutations, typically associated with frontal-subcortical type cognitive impairments. High prevalence (~50%) of superimposed Alzheimer׳s pathology has been reported in FMR1 premutation carriers, and standardized neuropsychological tests have not yielded any robust discriminators between FXTAS and Alzheimer׳s disease (AD) dementia. The similarities/differences in memory processes between FXTAS and early AD remain underexplored.Methods32-channel event-related potentials (ERPs) were obtained from a semantic judgment task in which semantically congruous (50%) and incongruous pairs repeat pseudorandomly. The N400 and late positive component (LPC) of 25 FXTAS patients (M(age)=71.2, MMSE=26.6) were compared to a matched group of 25 patients with MCI or early AD (1 mild AD dementia, 24 amnestic MCI, of whom 18 later converted to AD; M(age)=73.4, MMSE=26.4), and 25 healthy elderly.ResultsBoth patient groups showed similar reductions in the N400 repetition effect and N400 congruity effect amplitudes, compared to controls, reflecting abnormal semantic priming and repetition priming. The MCI/AD group, however, had significantly smaller LPC word repetition effects and poorer learning and memory on the CVLT than FXTAS. The LPC and N400 repetition effects both correlated with verbal memory across all subjects, but only N400 correlated with memory in FXTAS.ConclusionFXTAS patients show relative sparing of the LPC repetition effect, and less disruption of explicit memory than prodromal/early AD. N400 abnormalities in FXTAS appear to account for much of their mild impairments in verbal learning and memory.

Despite indications in the split-brain and lesion literatures that the right hemisphere is capable of some syntactic analysis, few studies have investigated right hemisphere contributions to syntactic processing in people with intact brains. Here we used the visual half-field paradigm in healthy adults to examine each hemisphere's processing of correct and incorrect grammatical number agreement marked either lexically, e.g., antecedent/reflexive pronoun ("The grateful niece asked herself/*themselves…") or morphologically, e.g., subject/verb ("Industrial scientists develop/*develops…"). For reflexives, response times and accuracy of grammaticality decisions suggested similar processing regardless of visual field of presentation. In the subject/verb condition, we observed similar response times and accuracies for central and right visual field (RVF) presentations. For left visual field (LVF) presentation, response times were longer and accuracy rates were reduced relative to RVF presentation. An event-related brain potential (ERP) study using the same materials revealed similar ERP responses to the reflexive pronouns in the two visual fields, but very different ERP effects to the subject/verb violations. For lexically marked violations on reflexives, P600 was elicited by stimuli in both the LVF and RVF; for morphologically marked violations on verbs, P600 was elicited only by RVF stimuli. These data suggest that both hemispheres can process lexically marked pronoun agreement violations, and do so in a similar fashion. Morphologically marked subject/verb agreement errors, however, showed a distinct LH advantage.

In previous work investigating the relationship between domain knowledge (of the fictional world of Harry Potter) and sentence comprehension, domain knowledge had a greater impact on electrical brain potentials to words which completed sentences about fictional "facts" participants reported they did not know compared to facts they did. This suggests that individuals use domain knowledge continuously to activate relevant/related concepts as they process sentences, even with only partial knowledge. As that study relied on subjective reports, it may have resulted in response bias related to an individual's overall domain knowledge. In the present study, we therefore asked participants with varying degrees of domain knowledge to complete sentences describing fictional "facts" as an objective measure of sentence‐level knowledge. We then recorded EEG as the same individuals (re‐)read the same sentences, including their appropriate final words, and sorted these according to their objective knowledge scores. Replicating and extending Troyer et al., domain knowledge immediately facilitated access to meaning for unknown words; greater domain knowledge was associated with reduced N400 amplitudes for unknown words. These findings constitute novel evidence for graded preactivation of conceptual knowledge (e.g., at the level of semantic features and/or relations) in the absence of lexical prediction. Knowledge also influenced post‐N400 memory/integration processes for these same unknown words; greater domain knowledge was associated with enhanced late positive components (LPCs), suggesting that deeper encoding during language processing may be engendered when knowledgeable individuals encounter an apparent gap in their knowledge. We examined how content/domain expertise influences electrophysiology during language processing. Within a third of a second, neural signatures indexing access to word meaning reveal sensitivity to fine‐grained variation in domain knowledge—especially for words not explicitly recalled during a fact‐completion task. For the same words, domain knowledge also influenced neural signatures thought to index elaborative encoding processes, likely because generally knowledgeable individuals could more readily link "unknown" words with existing knowledge structures. We sought cognitive event-related potential (ERP) biomarkers of "Preclinical Alzheimer's disease" (Pre-AD) using an incidental verbal learning paradigm with high sensitivity to prodromal AD. Seven elderly persons, with normal cognition at the time of ERP recordings, but who showed subsequent cognitive decline or AD pathology at autopsy (n = 5, mean Braak stage = 2.8), were compared to 12 "robust" normal elderly (RNE) persons who remained cognitively normal (Mfollow-up = 9.0 years). EEG was recorded during a word repetition paradigm (semantically congruous (50%) and incongruous target words repeat ~10-140 seconds later). The RNE P600 congruous word repetition ERP effects (New minus Old congruous words) were significantly larger than in Pre-AD (mean amplitudes = 3.28 vs. 0.10 μV, p = .04). High group discrimination (84%) was achieved (by a P600 amplitude cutoff of ~1.5 μV). Abnormal P600 word repetition effects in cognitively normal elderly persons may be an important sign of synaptic dysfunction and Preclinical AD.

The Brain Research through Advancing Innovative Neurotechnologies (BRAIN) Initiative has focused scientific attention on the necessary tools to understand the human brain and mind. Here, we outline our collective vision for what we can achieve within a decade with properly targeted efforts and discuss likely technological deliverables and neuroscience progress.
