The goal of this study was to evaluate the influence of various supplementary feeds on the chemical composition and production of bioactive substances in Protaetia brevitarsis larvae. The primary feed-oak-fermented sawdust-was supplemented with a variety of substances, including aloe, apple, banana, sweet persimmon (S. persimmon) and sweet pumpkin (S. pumpkin). Crude protein and fat content were the highest in the control and S. pumpkin group, respectively. Supplementary feeds increased the content of unsaturated fatty acids, except in the group receiving S. pumpkin, in which oleic acid was the most abundant (58.2%-64.5%). Free essential amino acids in larvae receiving supplementary aloe were higher compared with the control group except for Lys and His. Polyphenol and flavonoid contents and the antioxidant activities of ABTS and DPPH were higher in all treated groups compared with the control group. Although supplementary feeds led to a decreased crude protein content in the treated larvae when compared with the control group, these treatments generally improved the levels of unsaturated fatty acids and antioxidative activity. Therefore, we suggest that among the supplementary foods tested, aloe is a better resource for P. brevitarsis based on crude protein content, free amino acids and other bioactive compounds such as unsaturated fatty acids and antioxidants.

This study characterized the antimicrobial substances produced by the radiation-resistant bacterium Nibribacter radioresistens. The antimicrobial substances showed activity against Salmonella Gallinarum, pathogenic Escherichia coli, Bacillus cereus, Streptococcus iniae, and Saccharomyces cerevisiae. The substances showed higher activity against Gram-positive bacteria than against Gram-negative bacteria and yeast. N. radioresistens showed the best growth rate in LB liquid medium at 37oC; however, production of the antimicrobial substances was not associated with growth. Since the activity of the antimicrobial substances was affected by proteinase K and EDTA, the substances were presumed to be antimicrobial peptides (AMPs). The antimicrobial substances produced by N. radioresistens were unstable at higher temperatures and in acidic and basic pH ranges, and most of the activity was attributed to either low ( 30 kDa) molecules. When S. Gallinarum was treated with the antimicrobial substances, the cell destruction was acted on the cell envelope. Therefore, we concluded that N. radioresistens produces broad-spectrum and very unstable antimicrobial substances that mostly consist of low- and high-molecular weight peptides.

This study was done to analyze the antimicrobial activity of gut-derived microbes of Protaetiabrevitarsisseulensis larvae. Among the lactic acid bacterial strains isolated from the intestinal microbes of P. brevitarsisseulensis larvae, 10 strains were firstly selected from results of antimicrobial activity against Salmonella Typhimurium χ3339 strain as a susceptible strain. As a result, since the Minimum Inhibitory Concentration (MIC) was the best result in the K9 strain, further tests were performed on the K9 strain in subsequent studies. The K9 strain was identified as Lactobacillus plantarum from 16 SrDNA sequencing, named as L. plantarum K9. The antibacterial activity according to the amount of susceptible strain showed no or lower activity at 108 and 107 CFU/ml, whereas stronger antibacterial activity below 106 CFU/ml. The antibacterial activity test according to the cultural temperature showed the strongest antibacterial activity at 37℃. In the thermal stability test, it was confirmed that the antimicrobial activity was maintained up to 50℃ but significantly decreased from 60℃ or higher. However, some of the AMPs exhibited thermal stability because of remaining the activity above 60℃ or higher. Since proteinase K treatmentdecreases the antibacterial activity, we confirm that antibacterial activity is done by peptide-based bioactive substances. In summary, we suggest that AMPs in L. plantarum K9 strain are a potential of application as a natural antibacterial agent owing to thermal stability.

The aim of the study was to formulate the stable amlodipine maleate tablet by selecting and combining of suitable ingredients. Amlodipine tablets were designed by using different manufacturing methods or formulations. Dissolution rate at 30 min of newly formulated tablets was over 98% in 0.1 M HCl medium. After 4 months storage under accelerated condition, the changes of appearance, loss on drying, content and total impurity were investigated. For long-term stability tests, two formulations of K017 (direct compressed tablets consisting of microcrystalline cellulose and pregelatinized starch) and K018 (wet granulated tablets by OpadryAMB) were stored under , 60% RH for 24 months. Under the accelerated condition, moisture content in K017 formulation was increased as 5.96% for 4 months, while other formulations with anhydrous monobasic phosphoric potassium or by wet granulation showed higher increase in moisture content compared to K017. In addition, K017 formulation showed a low decrease in contents and total relative substance as 0.8% and 0.7%, respectively. Similar stability of amlodipine in K017 was obtained under the long-term stability test. These results indicate that the K017 combined with microcrystalline cellulose and pregelatinized starch as ingredients is very stable formulation to protect active substance from moisture contact and sustain stability. Therefore, suitable combination of ingredients such as microcrystalline cellulose and pregelatinized starch could attribute to enhance the stability of moisture-labile drug such as amlodipine maleate.

Two neolignan compounds, magnolol (5,5'-diallyl-2,2'-dihydroxybiphenyl, 1) and honokiol (5,5'-diallyl-2,4'-dihydroxybiphenyl, 2), were isolated from the stem bark of Magnolia obovata and evaluated for antifungal activity against various human pathogenic fungi. Compound 1 and 2 showed significant inhibitory activities against Trichophyton mentagrophytes, Microsporium gypseum, Epidermophyton floccosum, Aspergillus niger, Cryptococcus neoformans, and Candida albicans with minimum inhibitory concentrations (MIC) in a range of 25-100 microg/ml. Therefore, compound 1 and 2 could be used as lead compounds for the development of novel antifungal agents.

An efficient process for the preparation of poly(3-hydroxybutyrate) (PHB) microspheres with a narrow size distribution was developed. PHB was produced by a fed-batch culture of Ralstonia eutropha using fructose syrup as the sole carbon source. After autoclaving the bacteria, PHB granules, which accumulated in the cells, were isolated by a detergent/hypochlorite treatment and then spray-dried to obtain the microspheres. The diameters of the PHB microspheres ranged from 0.6 to 1.1 μm and the weight-average molecular weights were approximately 50 000 with polydispersity indexes of 5.0. The microspheres had a porous internal structure with an average porosity value of 72% and efficiently blocked UV light shorter than 220 nm. When isosorbide dinitrate was used as a model drug, the optimal drug loading concentration of the microspheres for controllable retardation was 3% (w/w). Almost 80% of the loaded drug (3%, w/w) was released within 12 h with typical sustained drug release behaviors.

Antifungal activities of the compounds isolated from Kalopanax pictus against representative fungi of dermatomycosis were investigated using paper disc diffusion method. It was found that kalopanaxsaponins A and I were effective in inhibiting the growth of Candida albicans KCTC 1940 and Cryptococcus neoformans KCTC 7224 with minimum inhibitory concentration (MIC) of 25 micrograms/ml. It showed that antifungal activity of both compounds have strong selectivity against the fungi of dermatomycosis.

This study examined the inhibitory effects of trans-cinnamaldehyde (CA), an aromatic aldehyde derived from Cinnamomi Cortex, on Saccharomyces cerevisiae cell wall synthesizing enzymes in vitro. This compound was found to be a noncompetitive inhibitor of beta-(1,3)-glucan synthase and a mixed inhibitor of chitin synthase 1 with 50% inhibitory concentrations (IC50) of 0.84 and 1.44 mM, respectively. Chitin synthases 2 and 3 were less sensitive than chitin synthase 1 to CA. CA can be useful as a model compound of cell wall inhibitors for the development of effective antifungal agents.

NIR spectroscopy has been extensively employed for the in-line monitoring of pharmaceutical processes as one of the key PAT implementation tools. Nevertheless, pharmaceutical processes such as fluid-bed coating have not fully made the most of the NIR in-line monitoring primarily due to a difficulty in handling random in-line spectra. In this study, novel approaches to develop a reasonable dynamic calibration model were proposed; averaging and clustering. Pharmaceutical test tablets were coated with HPMC-based materials using a fluid-bed processor. During the 160 min coating process under tangential spraying mode, 10 tablets were sampled out at every 10 min mark for actual coating thickness measurements. NIR spectra at and near each 10 min mark were treated and processed by the averaging and clustering operations. Averaging of 21 spectra resulted in a reasonably good dynamic calibration model whose determination coefficient was estimated as high as 0.9916. The PCA-based clustering turned out to be substantially helpful especially when a large number of NIR spectra were averaged. A prediction experiment verified that our dynamic calibration model can control the coating thickness in-line as good as 3% deviated from the actual thickness, which can offer a reasonable end-point for the fluid-bed coating process.
