Multiple sclerosis (MS) is an incurable autoimmune disease and is currently treated by systemic immunosuppressants with off-target side effects. Although aberrant myeloid function is often observed in MS plaques in the central nervous system (CNS), the role of myeloid cells in therapeutic intervention is currently overlooked. Here, we developed a myeloid cell-based strategy to reduce the disease burden in experimental autoimmune encephalomyelitis (EAE), a mouse model of progressive MS. We developed monocyte-adhered microparticles (“backpacks”) for activating myeloid cell phenotype to an anti-inflammatory state through localized interleukin-4 and dexamethasone signals. We demonstrate that backpack-laden monocytes infiltrated into the inflamed CNS and modulated both the local and systemic immune responses. Within the CNS, backpack-carrying monocytes regulated both the infiltrating and tissue-resident myeloid cell compartments in the spinal cord for functions related to antigen presentation and reactive species production. Treatment with backpack-monocytes also decreased the level of systemic pro-inflammatory cytokines. Additionally, backpack-laden monocytes induced modulatory effects on T H 1 and T H 17 populations in the spinal cord and blood, demonstrating cross talk between the myeloid and lymphoid arms of disease. Backpack-carrying monocytes conferred therapeutic benefit in EAE mice, as quantified by improved motor function. The use of backpack-laden monocytes offers an antigen-free, biomaterial-based approach to precisely tune cell phenotype in vivo, demonstrating the utility of myeloid cells as a therapeutic modality and target.

Nitric oxide (NO·), an important mediator of inflammation, and β-catenin, a component of the Wnt–adenomatous polyposis coli signaling pathway, contribute to the development of cancer. We have identified two T-cell factor 4 (Tcf-4)-binding elements (TBE1 and TBE2) in the promoter of human inducible NO synthase 2 (NOS2). We tested the hypothesis that β-catenin regulates human NOS2 gene. Mutation in either of the two TBE sites decreased the basal and cytokine-induced NOS2 promoter activity in different cell lines. The promoter activity was significantly reduced when both TBE1 and TBE2 sites were mutated (P < 0.01). Nuclear extract from HCT116, HepG2, or DLD1 cells bound to NOS2 TBE1 or TBE2 oligonucleotides in electrophoretic mobility shift assays and the specific protein-DNA complexes were supershifted with anti-β-catenin or anti-Tcf-4 antibody. Overexpression of β-catenin and Tcf-4 significantly increased both basal and cytokine-induced NOS2 promoter activity (P < 0.01), and the induction was dependent on intact TBE sites. Overexpression of β-catenin or Tcf-4 increased NOS2 mRNA and protein expression in HCT116 cells. Lithium chloride (LiCl), an inhibitor of glycogen synthase kinase-3β, increased cytosolic and nuclear β-catenin level, NOS2 expression, and NO· production in primary human and rat hepatocytes and cancer cell lines. Treatment with Wnt-3A-conditioned medium increased β-catenin and NOS2 expression in fetal human hepatocytes. When administered in vivo, LiCl increased hepatic β-catenin level in a dose-dependent manner with simultaneous increase in NOS2 expression. These data are consistent with the hypothesis that β-catenin up-regulates NOS2 and suggest a novel mechanism by which the Wnt/β-catenin signaling pathway may contribute to cancer by increasing NO· production. Simple strategy to stably preserve the smaller Ni nanoparticles (NPs) having homogeneous crystallite size of ∼5 nm was proposed using ordered mesoporous supports under dry reforming of methane with CO2 (DRM). The NPs-impregnated ordered mesoporous SBA-15 was effective to preserve the smaller Ni nanoparticles with their lower thermal aggregations and less coke depositions by their spatial confinement effects inside of the ordered mesopore structures compared to the irregular conventional SiO2 support. Although the highly ordered mesoporous NPs-impregnated Al2O3 was also found to be effective, the acidic natures of the Al2O3 surfaces accelerated coke depositions by preferentially forming inactive phases.

Background Nanoparticles have been used for biomedical applications, including drug delivery, diagnosis, and imaging based on their unique properties derived from small size and large surface-to-volume ratio. However, concerns regarding unexpected toxicity due to the localization of nanoparticles in the cells are growing. Herein, we quantified the number of cell-internalized nanoparticles and monitored their cellular localization, which are critical factors for biomedical applications of nanoparticles. Methods This study investigates the intracellular trafficking of silica-coated magnetic nanoparticles containing rhodamine B isothiocyanate dye [MNPs@SiO2(RITC)] in various live single cells, such as HEK293, NIH3T3, and RAW 264.7 cells, using site-specific direct stochastic optical reconstruction microscopy (dSTORM). The time-dependent subdiffraction-limit spatial resolution of the dSTORM method allowed intracellular site-specific quantification and tracking of MNPs@SiO2(RITC). Results The MNPs@SiO2(RITC) were observed to be highly internalized in RAW 264.7 cells, compared to the HEK293 and NIH3T3 cells undergoing single-particle analysis. In addition, MNPs@SiO2(RITC) were internalized within the nuclei of RAW 264.7 and HEK293 cells but were not detected in the nuclei of NIH3T3 cells. Moreover, because of the treatment of the MNPs@SiO2(RITC), more micronuclei were detected in RAW 264.7 cells than in other cells. Conclusion The sensitive and quantitative evaluations of MNPs@SiO2(RITC) at specific sites in three different cells using a combination of dSTORM, transcriptomics, and molecular biology were performed. These findings highlight the quantitative differences in the uptake efficiency of MNPs@SiO2(RITC) and ultra-sensitivity, varying according to the cell types as ascertained by subdiffraction-limit super-resolution microscopy. Nutritional metal ions play critical roles in many important immune processes. Hence, the effective modulation of metal ions may open up new forms of immunotherapy, termed as metalloimmunotherapy. Here, we demonstrate a prototype of cancer metalloimmunotherapy using cyclic dinucleotide (CDN) stimulator of interferon genes (STING) agonists and Mn2+. We screened various metal ions and discovered specific metal ions augmented STING agonist activity, wherein Mn2+ promoted a 12- to 77-fold potentiation effect across the prevalent human STING haplotypes. Notably, Mn2+ coordinated with CDN STING agonists to self-assemble into a nanoparticle (CDN–Mn2+ particle, CMP) that effectively delivered STING agonists to immune cells. The CMP, administered either by local intratumoural or systemic intravenous injection, initiated robust anti-tumour immunity, achieving remarkable therapeutic efficacy with minute doses of STING agonists in multiple murine tumour models. Overall, the CMP offers a new platform for local and systemic cancer treatments, and this work underscores the great potential of coordination nanomedicine for metalloimmunotherapy. The stimulation of interferon genes (STING) pathway with STING agonists such as cyclic dinucleotides (CDNs) has emerged as a promising immunotherapeutic approach. Here, the authors show that Mn2+ can amplify the STING-promoted anti-tumour immune response in challenging murine tumour models by coordinating with CDNs and self-assembling into nanoparticles that can be delivered locally and systemically.

Various anti-tumor nanomedicines have been developed based on the enhanced permeability and retention effect. However, the dense extracellular matrix (ECM) in tumors remains a major barrier for the delivery and accumulation of nanoparticles into tumors. While ECM-degrading enzymes, such as collagenase, hyaluronidase, and bromelain, have been used to facilitate the accumulation of nanoparticles, serious side effects arising from the current non-tumor-specific delivery methods limit their clinical applications. Here, we report targeted delivery of bromelain into tumor tissues through its covalent attachment to a hyaluronic acid (HA)-peptide conjugate with tumor ECM targeting ability. The ECM targeting peptide, collagen type IV-binding peptide (C4BP), was chosen from six candidate-peptides based on their ability to bind to frozen sections of triple-negative breast cancer, 4T1 tumor ex vivo. The HA- C4BP conjugate showed a significant increase in tumor accumulation in 4T1-bearing mice after intravenous administration compared to unmodified HA. We further demonstrated that the systemic administration of bromelain conjugated C4BP-HA (C4BP-HA-Bro) potentiates the anti-tumor efficacy of liposomal doxorubicin. C4BP-HA-Bro decreased the number and length of collagen fibers and improved the distribution of doxorubicin within the tumor. No infusion reaction was noted after delivery of C4BP-HA-Bro. C4BP-HA thus offers a potential for effective and safe delivery of bromelain for improved intratumoral delivery of therapeutics.

The catalytic activity for dehydrogenation of C2H6 and successive CO2 activation was studied by using hierarchically-structured bimetallic FeM/ZSM-5 (M=Ce, Ga, and Sn metal) to verify the redox property of the Fe nanoparticles and metal promoters on the acidic ZSM-5. Based on the surface characteristics, the reducibility and oxygen vacant sites of metal oxides on the ZSM-5 largely altered the reduction-oxidation nature and catalytic cracking behavior. The metal-promoted Fe/ZSM-5, especially with CeO2 promoter on the FeCe/ZSM-5, revealed excellent redox cycles and higher steady-state dehydrogenation activity such as a comparable C2H6 conversion of 6.1% as well as C2H4 selectivity of 89.8% at 600 °C with a larger CO production with 9.7 mmol/g by CO2 activation at 700 °C. This observation was attributed to the incorporated partially reducible CeO2 species by enhancing their interaction with ZSM-5 as well as by easily stabilizing the oxidation states of Ce and Fe metal oxides with its higher thermal stability during C2H6 dehydrogenation through an initial oxidative dehydrogenation followed by a steady-state catalytic cracking and subsequent CO2 activation to CO.

The current health crisis of corona virus disease 2019 (COVID-19) highlights the urgent need for vaccine systems that can generate potent and protective immune responses. Protein vaccines are safe, but conventional approaches for protein-based vaccines often fail to elicit potent and long-lasting immune responses. Nanoparticle vaccines designed to co-deliver protein antigens and adjuvants can promote their delivery to antigen-presenting cells and improve immunogenicity. However, it remains challenging to develop vaccine nanoparticles that can preserve and present conformational epitopes of protein antigens for induction of neutralizing antibody responses. Here, we have designed a new lipid-based nanoparticle vaccine platform (NVP) that presents viral proteins (HIV-1 and SARS-CoV-2 antigens) in a conformational manner for induction of antigen-specific antibody responses. We show that NVP was readily taken up by dendritic cells (DCs) and promoted DC maturation and antigen presentation. NVP loaded with BG505.SOSIP.664 (SOSIP) or SARS-CoV-2 receptor-binding domain (RBD) was readily recognized by neutralizing antibodies, indicating the conformational display of antigens on the surfaces of NVP. Rabbits immunized with SOSIP-NVP elicited strong neutralizing antibody responses against HIV-1. Furthermore, mice immunized with RBD-NVP induced robust and long-lasting antibody responses against RBD from SARS-CoV-2. Facile, economic methods of preparing tungsten (W) nanopowder are critically needed to meet industrial demand. Herein, we report a method of preparing single-phase alpha-W (α-W) nanopowders using ammonium paratungstate (APT) as a starting material and the optimum synthesis conditions. The process involves two stages: i) the radio-frequency (RF) induction thermal plasma treatment of APT, followed by ii) thermochemical reduction at 600-900 oC. The crystallographic phase and morphological evolution of all products were systematically investigated using X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), and transmission electron microscopy (TEM), and the effects of the annealing temperature on the phase and particle size of the obtained powders were also evaluated. When the RF induction thermal plasma treatment was conducted with and without H2, the XRD and FESEM results showed the formation of mixed-phase α- and beta-W (β-W) nanopowder and WO3 nanopowder, respectively. Single-phase α-W nanopowder was achieved by annealing the WO3 nanopowder in an H2 reductive atmosphere at 700 oC for 10 min, resulting in homogenous nanoparticles with a small particle size (d50) of 21.16 nm without any aggregation.

Therapeutic cancer vaccines require robust cellular immunity for the efficient killing of tumor cells, and recent advances in neoantigen discovery may provide safe and promising targets for cancer vaccines. However, elicitation of T cells with strong antitumor efficacy requires intricate multistep processes that have been difficult to attain with traditional vaccination approaches. Here, a multifunctional nanovaccine platform has been developed for direct delivery of neoantigens and adjuvants to lymph nodes (LNs) and highly efficient induction of neoantigen-specific T cell responses. A PEGylated reduced graphene oxide nanosheet (RGO-PEG, 20–30 nm in diameter) is a highly modular and biodegradable platform for facile preparation of neoantigen vaccines within 2 h. RGO-PEG exhibits rapid, efficient (15–20% ID/g), and sustained (up to 72 h) accumulation in LNs, achieving >100-fold improvement in LN-targeted delivery, compared with soluble vaccines. Moreover, RGO-PEG induces intracellular reactive oxygen species in dendritic cells, guiding antigen processing and presentation to T cells. Importantly, a single injection of RGO-PEG vaccine elicits potent neoantigen-specific T cell responses lasting up to 30 days and eradicates established MC-38 colon carcinoma. Further combination with anti-PD-1 therapy achieved great therapeutic improvements against B16F10 melanoma. RGO-PEG may serve a powerful delivery platform for personalized cancer vaccination.

Light-emitting diodes (LEDs) are energy-efficient and mercury-free, conventionally it is considered environment-friendly, which is not true depending upon kind of LED material constituents. Those LEDs provides yellow, red light and used for infrared light resources are mainly made from Ga, As and P. End of life LED bearing waste is an environmental challenge considering hazardous As content. Also, Ga is a vital metal for the electronics industry, is classified as significant from the industrial application and critical from a supply chain scarcity perspective. The Republic of Korea depends upon import of Ga, under the national policy of securing a stable supply, much attention has been paid to the notion of “urban mining”. Hence, Ga values recovery and detoxification of As from EOL LED can address two different issues. Through high-temperature sublimation followed As recovery by condensation for safe disposal or reuse purpose could be a potential technique. For the purpose, reactor design and thermal parameters are two important aspects needed to be addressed. Hence, in our current study focusses on understanding of thermochemistry and thermodynamics of GaAs bearing LED. It can provide potential information for the recovery of metal values and detoxification of hazardous substances from the LED.

CD4/CD8 T-cell lineage differentiation is a key process in immune system development; however, a defined regulator(s) that converts the signal from T-cell receptor and co-receptor complexes into lineage differentiation remains unclear. Here, we show that Twist2 is a critical factor in CD4/CD8 thymocyte differentiation. Twist2 expression is differentially regulated by T-cell receptor signaling, leading to differentiation into the CD4 or CD8 lineage. Forced Twist2 expression perturbed CD4+ thymocyte differentiation while enhancing CD8+ thymocyte differentiation. Furthermore, Twist2 expression produced mature CD8+ thymocytes in B2m-/- mice, while its deficiency significantly impaired CD8+ cells in MHC class-II-/- and TCR transgenic mice, favoring CD8 T-cell differentiation. During CD8 lineage differentiation, Twist2 interacted with Runx3 to bind to the silencer region of the ThPOK locus, thereby blocking ThPOK expression. These findings indicate that Twist2 is a part of the transcription factor network controlling CD8 lineage differentiation.

The selective production of aromatic chemicals such as benzene, toluene and xylenes from biomass-derived furans are quite challenging since zeolites and other supported heterogeneous catalysts are easily deactivated by trapping oligomeric species over their pores. Herein, a series of zirconium phosphates (ZrP) grafted on ordered mesoporous architectures of silicas was prepared by nonhydrolytic sol-gel method, and evaluated for the selective production of valuable para-xylene (pX) by catalytic reaction of 2,5-dimethylfuran (DMF) with gaseous ethylene through tandem Diels-Alder cycloaddition and dehydration reactions. The surface properties such as P/Zr ratio, Bronsted (B) to Lewis (L) acid sites and grafting layer thickness of the ZrP grafted silica catalysts were largely altered by changing P/Zr ratios and parent mesoporous silica surfaces. The amounts and strengths of acidic sites including B and L sites from the active (Zr–O)2–PO(OH) and (Zr–O)3–PO species were crucial factors for the differences in pX production rate on the ZrP grafted KIT-6, SBA-15 and fumed SiO2.

A facile one-pot wet chemical process to prepare pure anatase TiO2 hollow structures using ammonium hexafluorotitanate as a precursor is developed. By defining the formic acid ratio, we fabricate TiO2 hollow structures containing fluorine on the surface . The TiO2 hollow sphere is composed of an anatase phase containing fluorine by various analytical techniques . A possible formation mechanism for the obtained hollow samples by self -transformation and Ostwald ripening is proposed. The TiO2 hollow structures containing fluorine exhibits 1.2 - 2.7 times higher performance than their counterparts in photocatalytic activity . The enhanced photocatalytic activity of the TiO2 hollow structures is attributed to the combined effects of high crystallinity, specific surface area (62 ㎡g-1), and the advantage of surface fluorine ions (at 8%) having strong electron -withdrawing ability of the surface ≡ Ti-F groups reduces the recombination of photogenerated electrons and holes.

TiO2 is an attractive catalyst for the photocatalytic degradation of organic pollutants. However, owing to its large band gap, it can only be activated by ultraviolet (UV) light, which constitutes a small portion of solar energy. Therefore, there has been significant interest in extending its light absorption range from UV to visible light. In this study, fluorinated TiO2 hollow spheres (FTHSs) were prepared via a rapid and simple wet chemical process using ammonium hexafluorotitanate, and then FTHS/WO3 heterostructures with different weight ratios of the FTHS and WO3 nanoparticles were synthesized via a simple wet impregnation method. The formation of the hybrid structure was confirmed by various characterization techniques. The photocatalytic activity of the synthesized photocatalysts in the photodegradation of rhodamine B, a model pollutant, was evaluated under visible light irradiation. The FTHS/WO3 heterostructures exhibited significantly improved photocatalytic activity compared to the bare FTHS or WO3 nanoparticles. The photodegradation efficiency of the FTHS/WO3 heterostructure in the present study was up to 0.0581 min–1. Detailed mechanisms that lead to the enhanced photocatalytic activity of the heterostructures are discussed. In addition, comparative experiments reveal that the photodegradation efficiency of the FTHS/WO3 heterostructure under visible light irradiation is superior to that of the P25/WO3 heterostructure prepared from the commercially available TiO2 catalyst (P25) via the same impregnation method.

The on-board voltage regulator in the DDR5 memory module is required to resiliently supply current at large load transient events and alleviate output noise at the same time. We present an adaptive on-time (AOT) buck regulator with a turbo dual-phase interleaving logic for stable regulation and on-time control with dithered pseudo-constant switching frequency to suppress output harmonics by 6dB. The voltage regulator delivers up to 10A with a peak efficiency of 92.5% and covers 10A/μs steep load transients.

Cancer immunotherapy is revolutionizing oncology. However, dose-limiting toxicities and low patient response rates remain major challenges in the clinic. Cancer nanomedicine in combination with immunotherapies offers the possibility to amplify antitumour immune responses and to sensitize tumours to immunotherapies in a safe and effective manner. In this Review, we discuss opportunities for combination immunotherapy based on nanoparticle platforms designed for chemotherapy, photothermal therapy, photodynamic therapy, radiotherapy and gene therapy. We highlight how nanoparticles can be used to reprogramme the immunosuppressive tumour microenvironment and to trigger systemic antitumour immunity, synergizing with immunotherapies against advanced cancer. Finally, we discuss strategies to improve tumour and immune cell targeting while minimizing toxicity and immune-related adverse events, and we explore the potential of theranostic nanoparticles for combination immunotherapy. Cancer nanomedicine in combination with immunotherapies offers the possibility to amplify antitumour immune responses and to sensitize tumours to immunotherapies. In this Review, the authors discuss combination immunotherapy based on nanoparticle platforms designed for chemotherapy, photothermal therapy, photodynamic therapy, radiotherapy and gene therapy.

TiO2 nanoparticles as an active sunscreen ingredient generate reactive oxygen species (ROS) upon UVA irradiation which is cytotoxic, genotoxic and potential to damage the DNA. The health concern and potential risks from TiO2 can be mitigated by shielding the particles through the suitable coating. Considering the advantages of SiO2, SiO2 coated TiO2 nanoparticles can be a potential material which can replace TiO2 for thickening, whitening, lubricating, and sunscreen ingredient in cosmetics. This article reports the synthesis of cosmetic grade TiO2-SiO2 core-shell nanopowder from mechanically milled TiO2 nanopowder for commercial mass production. From commercial TiO2 nanopowder was fabricated through size reduction by nanoset milling. Followed by the fabricated TiO2 nanopowder coated with SiO2 through sol-gel technique. A suitable optimum condition was explored for cosmetic grade TiO2-SiO2 core-shell nanopowder. Various physical properties and optical properties were analyzed. Synthesized of cosmetic grade TiO2-SiO2 core-shell nanopowder found to be at 100 nm size, with a homogeneous SiO2 coating having UVA protection factor 39 and sun protection factor (SPF) is 42. From the size, safety, and SPF perspective it can be an excellent cosmetic grade powder and from process simplicity perspective it can be commercially viable.

Two types of TiO2 nanoparticles: i) fluorine-containing TiO2 (F-TiO2) and ii) fluorine-free TiO2 (H-TiO2) nanoparticles, were prepared through a simple, scalable wet-chemical synthesis process, and a comparative study of their photocatalytic properties was conducted. This facile process begins with a one-pot precipitation process at 90 °C, followed by heat treatment at 600 °C for 1 h in air, resulting in hierarchical, sphere-like, mesoporous structures composed of primary nanoparticles. The microstructural features and crystallographic structures of both types of nanoparticles were systematically investigated by X-ray diffraction, thermogravimetric/differential thermal analysis, X-ray photoelectron spectroscopy, N2 physical adsorption-desorption, field emission scanning electron microscopy, and transmission electron microscopy analyses. The photocatalytic activities were also evaluated by measuring the degradation of MB. The F-TiO2 nanoparticles showed enhanced photocatalytic activity compared to H-TiO2, which can be attributed to adsorbed fluorine on the surface which leads to various positive effects on the photocatalytic degradation reactions.

Nanoparticles (NPs) can serve as a promising vaccine delivery platform for improving pharmacological property and codelivery of antigens and adjuvants. However, NP‐based vaccines are generally associated with complex synthesis and postmodification procedures, which pose technical and manufacturing challenges for tailor‐made vaccine production. Here, modularly programmed, polyethyleneimine (PEI)‐based NP vaccines are reported for simple production of personalized cancer vaccines. Briefly, PEI is conjugated with neoantigens by facile coupling chemistry, followed by electrostatic assembly with CpG adjuvants, leading to the self‐assembly of nontoxic, sub‐50 nm PEI NPs. Importantly, PEI NPs promote activation and antigen cross‐presentation of antigen‐presenting cells and cross‐priming of neoantigen‐specific CD8+ T cells. Surprisingly, after only a single intratumoral injection, PEI NPs with optimal PEGylation elicit as high as ≈30% neoantigen‐specific CD8+ T cell response in the systemic circulation and sustain elevated CD8+ T cell response over 3 weeks. PEI‐based nanovaccines exert potent antitumor efficacy against pre‐established local tumors as well as highly aggressive metastatic tumors. PEI NPs elicit strong, long‐lasting neoantigen‐specific CD8+ T cell response with potent antitumor efficacy against local as well as metastatic tumors. PEI NPs offer a promising strategy for personalized cancer vaccination.

Various e-wastes like waste LCD, LED, and LCD etching industry wastewater are important secondary resources for indium, which is a critical metal. In this research, the industrial-scale indium recovery from e-waste resources like waste LCD, LED, and LCD industry etching wastewater is being emphasized through simulation and integration of the developed processes. A demonstration plant for indium recovery on one ton/day of ITO etching wastewater has been developed with almost complete (99%) recovery of indium. For the indium recovery, integration of the processes can be managed by following two approaches unique to this system, (i) utilization of ITO etching industry wastewaters for the leaching of waste LCD, (ii) integration of leaching processes developed for waste LCD and LED with that of the treatment process for ITO wastewaters. Through the proposed approach, the semiconductor manufacturing industry and ITO industry can address various pressing issues like (i) waste disposal, (ii) indium recovery, (iii) circular economy.

The novel corona virus termed severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has spread throughout the globe at a formidable speed, causing tens of millions of cases and more than one million deaths in less than a year of its report in December 2019. Since then, companies and research institutions have raced to develop SARS-CoV-2 vaccines, ranging from conventional viral and protein-based vaccines to those that are more cutting edge, including DNA- and mRNA-based vaccines. Each vaccine exhibits a different potency and duration of efficacy, as determined by the antigen design, adjuvant molecules, vaccine delivery platforms, and immunization method. In this review, we will introduce a few of the leading non-viral vaccines that are under clinical stage development and discuss delivery strategies to improve vaccine efficacy, duration of protection, safety, and mass vaccination., Graphical abstract Unlabelled Image

The synergetic effects of Co oxides on the Ni/CoAl (NCA) catalysts were observed at an optimal molar ratio of Al/Co = 2 (NCA(2)) due to the partial formations of thermally stable spinel CoAl2O4 phases for the steam reforming of propane (SRP). The optimal content of the spinel CoAl2O4 phases on the NCA(2) was responsible for the formation of the relatively active oxophilic metallic Co nanoparticles with a smaller amount of less active NiAl2O4 on the surfaces by preserving the relative amount of metallic Co of 68% and 52% in the reduced and used catalysts, which enhanced the catalytic activity and stability with the largest specific rate of 1.37 C3H8/(Ni + Co)h&minus, 1 among the tested NCA catalysts. The larger or smaller amounts of Co metal on the less active NCA mainly caused the preferential formation of larger aggregated Ni nanoparticles ~16 nm in size due to their weaker interactions, or induced the smaller formations of active metal phases by selectively forming the spinel NiAl2O4 phases with ~60% in the NCA(4), resulting in a fast deactivation.

For a solid acid-catalyzed dehydration of biomass-derived carbohydrates into useful furan derivatives, a suitable porous solid acid catalyst having an optimum acidic density and its strength is required to avoid cascade reactions in biomass conversion processes. A large-pore mesoporous zirconium phosphate (m-ZrP) was prepared hydrothermally using P123 as a template in water solvent, which resulted in a higher pore diameter (>9 nm) having wormhole-like pore structures with balanced Lewis (L) to Brönsted (B) acid sites. The effects of calcination temperature (500–800 °C) on the textural, acidic/basic, and structural properties of the m-ZrP with its catalytic performance for glucose dehydration to 5-hydroxymethylfurfural (HMF) were investigated in a pure water media as a green and sustainable alternative solvent. The larger number of L and B acid sites and basic sites with their appropriate strengths were clearly related with a better catalytic performance in terms of glucose conversion and HMF yield. The strong L acid and basic sites in the m-ZrP efficiently promoted the glucose isomerization to fructose, which dehydrated exclusively on the weak B acid sites resulting in a maximum conversion of glucose (83.8%) and HMF yield (46.6%). The adjusted acidic and basic sites with large mesopore sizes make the m-ZrP yield a higher reaction rate (2.78 mmol gcat–1 h–1) and turnover frequency (11.68/h) for conversion of glucose to HMF, which showed higher catalytic activity than those of a small-pore m-ZrP and other mesoporous heterogeneous and homogeneous acid catalysts.

Steam reforming of propane (SRP) for the stable production of hydrogen-rich reformates was investigated using the Ni-supported on the cobalt-modified SiC-embedded MgAl2O4 support (denoted as NCMAS). The adjusted interactions of the Ni nanoparticles with the cobalt-modified SiC-embedded MgAl2O4 (MgAl2O4-SiC) and its crystallite size distributions largely altered the catalytic activity and stability of NCMAS. The introductions of SiC on the NCMAS, where SiC has a higher thermal conductivity, also increased the dispersion of smaller MgAl2O4 grains with the less formations of inactive NiAl2O4 species, which resulted in the higher catalytic activity with smaller formations of unreformed light hydrocarbons. The positive roles of cobalt promoter on the MgAl2O4-SiC matrices were mainly attributed to the suppressed aggregation of nickel nanoparticles by their strong and intimate interactions with the cobalt-modified MgAl2O4. The effects of cobalt promoter at an optimal 5wt%Co in the MgAl2O4-SiC (NCMAS(5)) enhanced the oxidation-resistance of the nickel nanoparticles with less formations of inactive metal aluminates by being reversibly re-reduced under the SRP reaction conditions. These phenomena further lessen coke depositions by intimately interacting with highly dispersed oxophilic cobalt or cobalt aluminate species. The optimal NCMAS(5) was applied to derive kinetic parameters using Langmuir-Hinshelwood-Hougen-Watson (LHHW) mechanisms, and the reasonable activation energy of 73 kJ/mol and optimal operating parameters to maximize hydrogen production by SRP reaction was estimated in terms of the reaction conditions such as space velocity, feed ratio and reaction temperature.

Photothermal therapy (PTT) and neoantigen cancer vaccine each offers minimally invasive and highly specific cancer therapy; however, they are not effective against large established tumors due to physical and biological barriers that attenuate thermal ablation and abolish anti-tumor immunity. Here, we designed and performed comparative study using small (~ 50 mm(3)) and large (> 100 mm(3)) tumors to examine how tumor size affects the therapeutic efficiency of PTT and neoantigen cancer vaccine. We show that spiky gold nanoparticle (SGNP)-based PTT and synergistic dual adjuvant-based neoantigen cancer vaccine can efficiently regress small tumors as a single agent, but not large tumors due to limited internal heating and immunosuppressive tumor microenvironment (TME). We report that PTT sensitizes tumors to neoantigen cancer vaccination by destroying and compromising the TME via thermally induced cellular and molecular damage, while neoantigen cancer vaccine reverts local immune suppression induced by PTT and shapes residual TME in favor of anti-tumor immunity. The combination therapy efficiently eradicated large local tumors and also exerted strong abscopal effect against pre-established distant tumors with robust systemic anti-tumor immunity. Thus, PTT combined with neoantigen cancer vaccine is a promising nano-immunotherapy for personalized therapy of advanced cancer.

Identification of tumor-specific mutations, called neoantigens, offers new exciting opportunities for personalized cancer immunotherapy . However, it remains challenging to achieve robust induction of neoantigen-specific T cells and drive their infiltration into the tumor microenvironment (TME). Here, we have developed a novel polyethyleneimine (PEI)-based personalized vaccine platform carrying neoantigen peptides and CpG adjuvants in a compact nanoparticle (NP) for their spatio-temporally concerted delivery. The NP vaccine significantly enhanced activation and antigen cross-presentation of dendritic cells, resulting in strong priming of neoantigen-specific CD8 + T cells with the frequency in the systemic circulation reaching as high as 23 ± 7% after a single subcutaneous administration. However, activated CD8 + T cells in circulation exhibited limited tumor infiltration, leading to poor anti-tumor efficacy. Notably, local administration of stimulator of interferon genes (STING) agonist promoted tumor infiltration of vaccine-primed CD8 + T cells, thereby overcoming one of the major challenges in achieving strong anti-tumor efficacy with cancer vaccination. The NP vaccination combined with STING agonist therapy eliminated tumors in murine models of MC-38 colon carcinoma and B16F10 melanoma and established long-term immunological memory . Our approach provides a novel therapeutic strategy based on combination nano-immunotherapy for personalized cancer immunotherapy.
