1. Manipulation of microfluidic flow pattern by optically controlled electroosmosis

Abstract

Light is used to dynamically control the pattern of electroosmotic flow in a microfluidic channel. One wall of the channel is formed by a photoconductor film in a specific geometry. Illumination of this surface results in an increase of the conductivity that modifies the structure of the electric field inside the channel. A dramatic change of the electroosmotic flow pattern can be achieved. This approach provides useful capabilities for the manipulation of fluids in microfluidic systems like directing flow and mixing.

Abstract

We present a microfluidic cell sorter that is able to count, characterize and sort micrometer sized particles and cells. In addition to optical counting and characterization, also sorting is performed by optical forces. The device is optimized for simplicity. The microfluidic channels and optical waveguides that carry the illuminating, detecting and sorting light form a single integrated structure, all built from the same material in a single photopolymerization step.

Abstract

Background: Light therapy is thought to be the first choice treatment of winter depression. However, its way of action is poorly understood. In order to find a solid effect of bright artificial light, we studied its possible alerting action through the spontaneous fluctuations of the pupil, considered to be an objective measurement of vigilance. Methods: Pupillary fluctuations of 10 healthy subjects (mean age: 22±1 S.D. years) were measured for 60 s before and 15 min after 0.5 h, 10 000-lux light exposure. The cumulative change in pupil size, characterised by the pupillary unrest index (PUI) decreased at each subject, and this decrease was in average 35±4.4% S.E.M. The average pupillary diameters were unchanged (101±2.2% S.E.M.). This analysis revealed that the slow components of the pupillary fluctuations also decreased considerably. Limitations: There was no dim light or other placebo control of the light treatment. Conclusions: Bright light exposure significantly influenced the pupillary fluctuations. We suppose that bright light exposure increases the level of alertness, and this could be a possible way by which bright artificial light exerts a beneficial effect also in affective disorders.

4. Interpretation of the spatial charge displacements in bacteriorhodopsin in terms of structural changes during the photocycle

Abstract

We have recently introduced a method, made possible by an improved orienting technique using a combination of electric and magnetic fields, that allows the three-dimensional detection of the intramolecular charge displacements during the photocycle of bacteriorhodopsin. This method generates electric asymmetry, a prerequisite for the detection of electric signal on the macroscopic sample, in all three spatial dimensions. Purple membrane fragments containing bacteriorhodopsin were oriented so that their permanent electric dipole moment vectors were perpendicular to the membrane plane and pointed in the same direction. The resulting cylindrical symmetry was broken by photoselection, i. e. , by flash excitation with low intensity linearly polarized light. From the measured electric signals, the three-dimensional motion of the electric charge center in the bacteriorhodopsin molecules was calculated for the first 400 μs. Simultaneous absorption kinetic recording provided the time-dependent concentrations of the intermediates. Combining the two sets of data, we determined the discrete dipole moments of intermediates up to M. When compared with the results of current molecular dynamics calculations, the data provided a decisive experimental test for selecting the optimal theoretical model for the proton transport and should eventually lead to a full description of the mechanism of the bacteriorhodopsin proton pump.

Abstract

Self propelled robots are interesting both for technology (primarily biotechnology) and for basic science in the investigation of collective motion. Numerous experimental model systems have been developed and used for different applications. The solutions range from simple specially shaped particles [1] to complex robots [2].Here we introduce moving microscopic swimmers where the objects move on a flat horizontal surface and the energy neeeded for the motion is provided by light. In contrast to the usual optical micromanipulation techniques, the direction of motion is not determined by the illuminating light. The whole area where the particles move is illuminated vertically from above and the objects move in an autonomous manner, in a direction determined by their position and orientation.The specially shaped objects are built by two photon induced photopolymerization. Two types of swimmers are discussed: a simple vedge shape [3] and a roller. Both are reflective and they are driven by the momentum change of light reflected from their surface.References:1. H-R. Abstract

We introduce a system of light driven microscopic autonomous moving particles that move on a flat surface. The design is simple, yet effective: Micrometer sized objects with wedge shape are produced by photopolymerization, and they are covered with a reflective surface. When the area of motion is illuminated perpendicularly from above, the light is deflected to the side by the wedge shaped objects, in the direction determined by the position and orientation of the particles. The momentum change during reflection provides the driving force for an effectively autonomous motion. The system is an efficient tool to study self propelled microscopic robots. © 2012 American Institute of Physics.

Abstract

Hydrodynamic synchronization is a fundamental physical phenomenon by which self-sustained oscillators communicate through perturbations in the surrounding fluid and converge to a stable synchronized state. This is an important factor for the emergence of regular and coordinated patterns in the motions of cilia and flagella. When dealing with biological systems, however, it is always hard to disentangle internal signaling mechanisms from external purely physical couplings. We have used the combination of two-photon polymerization and holographic optical trapping to build a mesoscale model composed of chiral propellers rotated by radiation pressure. The two microrotors can be synchronized by hydrodynamic interactions alone although the relative torques have to be finely tuned. Dealing with a micron sized system we treat synchronization as a stochastic phenomenon and show that the phase lag between the two microrotors is distributed according to a stationary Fokker-Planck equation for an overdamped particle over a tilted periodic potential. Synchronized states correspond to minima in this potential whose locations are shown to depend critically on the detailed geometry of the propellers.

Abstract

The static and dynamic response of optical waveguides coated with a thin protein film of bacteriorhodopsin was investigated. The size and kinetics of the light-induced refractive index changes of the adlayer were determined under different conditions of illumination. The results demonstrate the applicability of this protein as an active, programmable nonlinear optical material in all-optical integrated circuits.

Abstract

We have studied the conditions of rotation induced by collimated light carrying no angular momentum. Objects of different shapes and optical properties were examined in the nontrivial case where the rotation axis is perpendicular to the direction of light propagation. This geometry offers important advantages for application as it fundamentally broadens the possible practical arrangements to be realised. We found that collimated light cannot drive permanent rotation of 2D or prism-like 3D objects (i.e. fixed cross-sectional profile along the rotation axis) in the case of fully reflective or fully transparent materials. Based on both geometrical optics simulations and theoretical analysis, we derived a general condition for rotation induced by collimated light carrying no angular momentum valid for any arrangement: Permanent rotation is not possible if the scattering interaction is two-dimensional and lossless. In contrast, light induced rotation can be sustained if partial absorption is present or the object has specific true 3D geometry. Abstract

Electro-osmosis is an efficient means to move fluid in microfluidic channels. The flow is driven by the interaction of the electrical double layer at the channel wall with an electric field along the channel. The flow can be controlled by modifying the electrical &muparameters, either the charge of the channel wall or the electric field. If the surface chagre or the surface rsistance of the channel wall is sensitive to light, the flow can be modulated by light. We have demonstrated this effect by using photoconductive surfaces. The resistance change due to the illumination changes the electric field above the photoconductive layer and consequently changing the rate of fluid flow. By using channels where upon a photoresistive CdS surface a linear PDMS channel was placed, flow rate changes of an order of magnitude were achieved. This gives serious possibilities for optical control of flow. We further developed the method by building channel structures of more complicated patterns, e.g. Y-junctions. By appropriate illumination of the arms the flow direction could be selected between the arms optically. This unit is the basis of more complex flow patterns, it demonstrates the feasibility of optical control of such devices.

Abstract

We present a method that offers the possibility to directly apply and measure torque on particles in an optical trap. It can be used to rotationally manipulate biopolymers attached to appropriate particles. A flat object is trapped and oriented in the focus of a linearly polarized laser light. The direction and power of the orientational trap are controlled by the polarization state of the light. As a demonstration of the capabilities of the method, we examined the torsional stiffness of dsDNA (lambda-DNA) in its linear torsional regime by directly measuring the torque generated by the molecule.

Abstract

Electro-osmotic pumping is an efficient way to move fluids in microfluidic systems. It is driven by the interaction of the Debye layer formed in the vicinity of the charged channel wall with a tangential electric field. The key parameters that determine the flow properties are the zeta potential of the surface and the electric field that drives the flow. Consequently, the flow can be controlled by appropriately modifying these parameters. Controlling the charge on the channel wall makes it possible to modify fluid flow. Likewise, the electric field close to the surface can be modified by changing the conductivity of the surface. The surface charge of appropriate materials can be changed by light illumination: the application of this phenomenon offers the possibility to optically control flow parameters. We have tested this possibility with several light sensitive surfaces. In the class of materials that change their charge upon illumination TiO 2 , a well known photoactive material was investigated. Experiments were also performed with the protein bacteriorhodopsin, known to change its surface charge following the release of protons into the solvent upon illumination. CdS was tested as the photoconductive material to modify the electric field by light. Linear microfluidic channels were prepared by soft lithography: a PDMS mold was placed upon a planar glass surface so that a rectangular cross section channel was formed upon the glass. The photosensitive materials covered the bottom glass surface. The experiments show that the flow can be readily modulated by illumination. The results demonstrate that it is possible to dynamically control microfluidic flow, opening up the prospect to create optically controlled complex microfluidic networks.

Abstract

We describe a novel method by which it is possible to apply and measure torque directly on particles grabbed in optical tweezers. It can be used to orient particles of micron size or even on single molecules, biopolymers by the use of test particles. The procedure is based on the observation that flat objects are oriented in an optical trap formed by linearly polarized light. The orienting torque originates from the anisotropic scattering of polarized light by the trapped particle. The phenomenon is characterized in detail, the physics is analysed. A tool is developed that exploits this effect to manuipulate biological macromolecules. Microscopic particles are produced by photopolymerisation that exploit this orientation effect and by which the torque is applied upon the biological object. In our system the applied torque can be turned on and off, it is controlled independently of the grabbing force of the tweezers during the manipulation process. The capabilities of the method are demonstrated. The method has great promise for application on DNA, DNA-protein complexes, actin filaments and other biopolymers.

Abstract

A simplified Brownian dynamics model and the corresponding software implementation have been developed for the simulation of electrolyte dynamics on the mesoscopic scale. In addition to direct control simulations, the model system has been verified by a quantitative comparison with the Debye-Huckel theory. As a first application, the model was used to simulate ionic relaxation processes following abrupt intramembrane charge rearrangements in the case of a disk shaped membrane. In addition to its general implications, the obtained properties of the relaxation kinetics confirm the assumptions of the theory of the so-called suspension method, a technique capable of tracing molecular charge motions of membrane proteins in three dimensions.

Abstract

A concept of a new application based on the outstanding nonlinear optical properties of the chromoprotein bacteriorhodopsin was investigated. Using the Optical Waveguide Lightmode Spectroscopy technique on dried bacteriorhodopsin films gave us the possibility to exploit the large refractive index changes corresponding to the absorption changes during the photocycle. The results demonstrate the applicability of this protein as an active nonlinear optical material in all-optical integrated circuits.

Abstract

Transmembrane ion pumps are often investigated experimentally by photoelectric measurements in model systems. In addition to the most widely used systems based on model membranes, a fundamentally different class is represented by the so-called suspension methods. In this technique the electric signal is measured on a bulk suspension of oriented ion pumps in the form of a displacement current. On this system, electric and spectroscopic experiments can be performed simultaneously. Using the information from both types of measurements, and utilizing the three-dimensional nature of the system, it is possible to follow the intramolecular charge motions in all three spatial directions. The derivable dipole moment changes associated with conformational transitions allow the verification of molecular dynamic models. In this work a theory is presented to describe the suspension method; samples with different symmetry properties and the possibilities of photoselection to obtain the desired three-dimensional information are analyzed.

Abstract

State-of-the-art photonic integration technology is ready to provide the passive elements of optical integrated circuits, based either on silicon, glass or plastic materials. The bottleneck is to find the proper nonlinear optical (NLO) materials in waveguide-based integrated optical circuits for light-controlled active functions. Recently, we proposed an approach where the active role is performed by the chromoprotein bacteriorhodopsin as an NLO material, that can be combined with appropriate integrated optical devices. Here we present data supporting the possibility of switching based on a fast photoreaction of bacteriorhodopsin. The results are expected to have important implications for photonic switching technology.

Abstract

We present a method to build an optical tip at the end of a single-mode optical fiber. The tip is grown by a self-writing process: photopolymerization by the light coming from the optical fiber. We developed a technique to produce a flat end surface on the tip. The good optical quality of the tip and the output laser beam was demonstrated by the fact that a counterpropagating optical trap could be constructed by using the tips with parameters comparable to regular fiber traps. Because of the small size of the tips, the tweezers require a much smaller space than regular fiber traps.

Abstract

The author introduce a technique for the optical control of electro-osmosis. One wall of the microfluidic channel is formed by a photoconductive material. Illumination of the photoconductor results in a local drop of the electric field in the microchannel, reducing the electro-osmotic driving force. Fluid flow can be effectively modulated by light utilizing this effect. They verified the phenomenon in linear channels and further developed the method by building a Y junction, an optically controlled microfluidic switch. This unit is the basis of more complex flow patterns: it demonstrates the feasibility of dynamic optical control of microfluidic devices.
