IntroductionAneurysmal subarachnoid hemorrhage (aSAH) often necessitates prolonged sedation to manage elevated intracranial pressure (ICP) and to prevent secondary brain injury. Optimal timing and biomarkers for predicting adverse events (AEs) during interruption of sedation (IS) after prolonged sedation are not well established. To guide sedation management in aSAH, we aimed to explore the frequency, risk factors, and outcomes of IS in aSAH.MethodsIn a retrospective cohort study, a total of 148 patients with aSAH from January 2015 to April 2020 were screened. In total, 30 patients accounting for 42 IS were included in the analysis. Adverse events (AEs) during IS were used as core outcome measures and were categorized into neurological and non-neurological AEs. Baseline characteristics, clinical parameters before IS, AEs, and functional outcomes were collected using health records. Statistical analysis used generalized linear mixed-effects models with regularization to identify candidate predictors with subsequent bootstrapping to test model stability. As an exploratory analysis, multivariate linear and logistic regression was used to analyze the association between IS and intensive care unit length of stay, duration of mechanical ventilation, and functional outcomes.ResultsThe mean age was 56.9 (SD 14.8) years, and a majority of the patients presented with poor-grade SAH (16/30, 53.3%). Neurological and non-neurological AEs occurred in 60.0% (18/30) of the patients. Timing, number of IS attempts, ICP burden, craniectomy status, level of consciousness, heart rate, cerebral perfusion pressure, oxygen saturation, fraction of inspired oxygen, and temperature were selected as candidate predictors. Through bootstrapping, elapsed time since disease onset (OR 0.85, 95% confidence interval (95% CI) 0.75–0.97), ICP burden (OR 1.24, 95% CI 1.02–1.52), craniectomy (OR 0.68, 95% CI 0.48–0.69), and oxygen saturation (OR, 0.80 0.72–0.89) were revealed as relevant biomarkers for neurological AEs, while none of the pre-selected predictors was robustly associated with non-neurological AEs.ConclusionIn aSAH, complications during the definite withdrawal of sedation are frequent but can potentially be predicted using clinical parameters available at the bedside. Prospective multicenter studies are essential to validate these results and further investigate the impact of IS complications.

The Ekström Ice Shelf is one of numerous small ice shelves that fringe the coastline of western Dronning Maud Land, East Antarctica. Reconstructions of past ice-sheet extent in this area are poorly constrained, due to a lack of geomorphological evidence. Here, we present a compilation of geophysical surveys in front of and beneath the Ekström Ice Shelf, to identify and interpret evidence of past ice-sheet flow, extent and retreat. The sea floor beneath the Ekström Ice Shelf is dominated by an incised trough, which extends from the modern-day grounding line onto the continental shelf. Our surveys show that mega-scale glacial lineations cover most of the mouth of this trough, terminating 11 km away from the continental shelf break, indicating the most recent minimal extent of grounded ice in this region. Beneath the front ∼30 km of the ice shelf measured from the ice shelf edge towards the inland direction, the sea floor is characterised by an acoustically transparent sedimentary unit, up to 45 m thick. This is likely composed of subglacial till, further corroborating the presence of past grounded ice cover. Further inland, the sea floor becomes rougher, interpreted as a transition from subglacial tills to a crystalline bedrock, corresponding to the outcrop of the volcanic Explora Wedge at the sea floor. Ice retreat in this region appears to have happened rapidly in the centre of the incised trough, evidenced by a lack of overprinting of the lineations at the trough mouth. At the margins of the trough uniformly spaced recessional moraines suggest ice retreated more gradually. We estimate the palaeo-ice thickness at the calving front around the Last Glacial Maximum to have been at least 305 to 320 m, based on the depth of iceberg ploughmarks within the trough and sea level reconstructions. Given the similarity of the numerous small ice shelves along the Dronning Maud Land coast, these findings are likely representative for other ice shelves in this region and provide essential boundary conditions for palaeo ice-sheet models in this severely understudied region.

The West Antarctic Ice Sheet (WAIS) presently holds enough ice to raise global sea level by 4.3 m if completely melted. The unknown response of the WAIS to future warming remains a significant challenge for numerical models in quantifying predictions of future sea level rise. Sea level rise is one of the clearest planet-wide signals of human-induced climate change. The Sensitivity of the West Antarctic Ice Sheet to a Warming of 2 ∘C (SWAIS 2C) Project aims to understand past and current drivers and thresholds of WAIS dynamics to improve projections of the rate and size of ice sheet changes under a range of elevated greenhouse gas levels in the atmosphere as well as the associated average global temperature scenarios to and beyond the +2 ∘C target of the Paris Climate Agreement. Despite efforts through previous land and ship-based drilling on and along the Antarctic margin, unequivocal evidence of major WAIS retreat or collapse and its causes has remained elusive. To evaluate and plan for the interdisciplinary scientific opportunities and engineering challenges that an International Continental Drilling Program (ICDP) project along the Siple coast near the grounding zone of the WAIS could offer (Fig. 1), researchers, engineers, and logistics providers representing 10 countries held a virtual workshop in October 2020. This international partnership comprised of geologists, glaciologists, oceanographers, geophysicists, microbiologists, climate and ice sheet modelers, and engineers outlined specific research objectives and logistical challenges associated with the recovery of Neogene and Quaternary geological records from the West Antarctic interior adjacent to the Kamb Ice Stream and at Crary Ice Rise. New geophysical surveys at these locations have identified drilling targets in which new drilling technologies will allow for the recovery of up to 200 m of sediments beneath the ice sheet. Sub-ice-shelf records have so far proven difficult to obtain but are critical to better constrain marine ice sheet sensitivity to past and future increases in global mean surface temperature up to 2 ∘C above pre-industrial levels. Thus, the scientific and technological advances developed through this program will enable us to test whether WAIS collapsed during past intervals of warmth and determine its sensitivity to a +2 ∘C global warming threshold (UNFCCC, 2015).

In order to elucidate the petrogenesis of the Hughes Bluff granitic pluton, the petrological and geochemical studies were conducted, and the results show that the Hughes Bluff granitic pluton is composed of monzogranite, intruded by fine-grained monzogranite dikes in the later period. They both are characterized by high abundance of silicon, alkali and potassium, enriched in Rb, Th, U and K and depleted in Nb, Ta, Nd and Ti relative to those of the primitive mantle, with the Rittmann Indexes less than 3 and the A/CNK values less than 1. They both also have a low total amount of rare earth elements and an abundance of light rare earth, showing weakly negative Eu anomaly and slightly positive Eu anomaly in the chondrite-normalized REE pattern for the monzogranite and granitic monzogranite dike respectively. All the data show that the rocks from the Hughes Bluff granitic pluton belong to the I-type granites, and the source region is probably the lower continental crust, but the contribution of mantle material cannot be ruled out. The magma in the source region underwent varying degrees of fractional crystallization of plagioclase, ilmenite, rutile and apatite, and was derived from a volcanic island arc environment related to subduction.

Bedrock geology from Antarctica remains largely unknown since it is hidden beneath thick ice sheets. Geophysical methods such as gravity and magnetic inverse modelling provide a framework to infer crustal rock properties indirectly in Antarctica. However, due to limited availability of rock samples, validation against direct geological information is challenging. We present a new rock property catalogue containing density and susceptibility measurements on 320 rock samples from northern Victoria Land. This catalogue is used to assess the reliability of local and regional scale inverse results, including a new local high resolution magnetic inversion in the Mesa Range region and a previously published regional scale joint inversion of gravity and magnetic data in northern Victoria Land and the Wilkes Subglacial Basin. We compare our density and susceptibility measurements to global and local measurements from the literature to access the correlation to rock types and geological units. Furthermore, the measured values are compared against inverted values. The close correspondence between inverted and measured rock properties allows us to predict locations of rock types where currently such information is missing. The utility of measured susceptibility and density relationships for interpreting inversion output provides a strong incentive to incorporate local rock samples into geophysical studies of subglacial geology across Antarctica. Caldicellulosiruptor is a genus of thermophilic to hyper-thermophilic microorganisms that express and secrete an arsenal of enzymes degrading lignocellulosic biomasses into fermentable sugars. Because of this distinguished feature, strains of Caldicellulosiruptor have been considered as promising candidates for consolidated bioprocessing. Although a few Caldicellulosiruptor strains with industrially relevant characteristics have been isolated to date, it is apparent that further improvement of the strains is essential for industrial application. The earlier identification of the HaeIII-like restriction-modification system in C. bescii strain DSM 6725 has formed the basis for genetic methods with the aim to improve the strain’s lignocellulolytic activity and ethanol production. In this study, a novel SfaNI-like restriction-modification system was identified in Caldicellulosiruptor sp. strain BluCon085, consisting of an endonuclease and two methyltransferases that recognize the reverse-complement sequences 5’-GATGC-3’ and 5‘-GCATC-3’. Methylation of the adenine in both sequences leads to an asymmetric methylation pattern in the genomic DNA of strain BluCon085. Proteins with high percentage of identity to the endonuclease and two methyltransferases were identified in the genomes of C. saccharolyticus strain DSM 8903, C. naganoensis strain DSM 8991, C. changbaiensis strain DSM 26941 and Caldicellulosiruptor sp. strain F32, suggesting that a similar restriction-modification system may be active also in these strains and respective species. We show that methylation of plasmid and linear DNA by the identified methyltransferases, obtained by heterologous expression in Escherichia coli, is sufficient for successful transformation of Caldicellulosiruptor sp. strain DIB 104C. The genetic engineering toolbox developed in this study forms the basis for rational strain improvement of strain BluCon085, a derivative from strain DIB 104C with exceptionally high L-lactic acid production. The toolbox may also work for other species of the genus Caldicellulosiruptor that have so far not been genetically tractable.
