1. Development of a Continuous Biofilm-Based Process for the Bioconversion of Cheese Whey into Volatile Fatty Acids

Abstract: The main goal of the present work was to study the continuous production of volatile fatty acids from a cheese whey powder in anaerobic packed bed biofilm reactors (PPBRs). Vukopor S10 ceramic cubes and granular activated carbon were employed as cell immobilization materials. Experiments were done at 37°C, pH 6. Reactors were frequently sampled for metabolites and biogas analyses. Preliminary batch tests, which were carried out in 100-mL anaerobic Pyrex bottles, demonstrated that the process consisted of two sequential phases: (a) the conversion of lactose into lactic acid and (b) the conversion of lactic acid into a mixture of VFAs. The continuous process was settled up in 1-L glass column and two different hydraulic retention times (9 and 6 days) were applied during the continuous process. Almost 80% of lactose bioconversion was achieved in Vukopor reactors, this corresponding to a 10 g/L VFAs concentration.

Abstract: Predicting the ocean state in a reliable and interoperable way, while ensuring high-quality products, requires forecasting systems that synergistically combine science-based methodologies with advanced technologies for timely, user-oriented solutions. Achieving this objective necessitates the adoption of best practices when implementing ocean forecasting services, resulting in the proper design of system components and the capacity to evolve through different levels of complexity. The vision of OceanPrediction Decade Collaborative Center, endorsed by the UN Decade of Ocean Science for Sustainable Development 2021-2030, is to support this challenge by developing a “predicted ocean based on a shared and coordinated global effort” and by working within a collaborative framework that encompasses worldwide expertise in ocean science and technology. To measure the capacity of ocean forecasting systems, the OceanPrediction Decade Collaborative Center proposes a novel approach based on the definition of an Operational Readiness Level (ORL). This approach is designed to guide and promote the adoption of best practices by qualifying and quantifying the overall operational status. Considering three identified operational categories - production, validation, and data dissemination - the proposed ORL is computed through a cumulative scoring system. This method is determined by fulfilling specific criteria, starting from a given base level and progressively advancing to higher levels. The goal of ORL and the computed scores per operational category is to support ocean forecasters in using and producing ocean data, information, and knowledge. This is achieved through systems that attain progressively higher levels of readiness, accessibility, and interoperability by adopting best practices that will be linked to the future design of standards and tools. This paper discusses examples of the application of this methodology, concluding on the advantages of its adoption as a reference tool to encourage and endorse services in joining common frameworks.

Abstract: Urban waste can be valorized within a biorefinery approach, producing platform chemicals, biopolymers and energy. In this framework, hydrodynamic cavitation (HC) is a promising pre-treatment for improving biodegradability due to its high effectiveness and low cost. This paper deals with the effect of HC pre-treatment on the acidogenic co-fermentation process of thickened sewage sludge from a WWTP and seasonal vegetable waste from a wholesale market. Specifically, HC was assessed by testing two sets of parameters (i.e., treatment time of 30 and 50 min; vacuum pressure 1.4 and 2.0 bar; applied power 8 and 17 kW) to determine its effectiveness as a pre-treatment of the mixture. 4. Multivariate state and parameter estimation with data assimilation applied to sea-ice models using a Maxwell elasto-brittle rheology

Abstract: In this study, we investigate the fully multivariate state and parameter estimation through idealised simulations of a dynamics-only model that uses the novel Maxwell elasto-brittle (MEB) sea-ice rheology and in which we estimate not only the sea-ice concentration, thickness and velocity, but also its level of damage, internal stress and cohesion. Specifically, we estimate the air drag coefficient and the so-called damage parameter of the MEB model. Mimicking the realistic observation network with different combinations of observations, we demonstrate that various issues can potentially arise in a complex sea-ice model, especially in instances for which the external forcing dominates the model forecast error growth. Even though further investigation will be needed using an operational (a coupled dynamics–thermodynamics) sea-ice model, we show that, with the current observation network, it is possible to improve both the observed and the unobserved model state forecast and parameter accuracy.

5. Improving short-term sea ice concentration forecasts using deep learning

Abstract: Reliable short-term sea ice forecasts are needed to support maritime operations in polar regions. While sea ice forecasts produced by physically based models still have limited accuracy, statistical post-processing techniques can be applied to reduce forecast errors. In this study, post-processing methods based on supervised machine learning have been developed for improving the skill of sea ice concentration forecasts from the TOPAZ4 prediction system for lead times from 1 to 10 d. The deep learning models use predictors from TOPAZ4 sea ice forecasts, weather forecasts, and sea ice concentration observations. Predicting the sea ice concentration for the next 10 d takes about 4 min (including data preparation), which is reasonable in an operational context. On average, the forecasts from the deep learning models have a root mean square error 41 % lower than TOPAZ4 forecasts and 29 % lower than forecasts based on persistence of sea ice concentration observations. They also significantly improve the forecasts for the location of the ice edges, with similar improvements as for the root mean square error. Furthermore, the impact of different types of predictors (observations, sea ice, and weather forecasts) on the predictions has been evaluated. Sea ice observations are the most important type of predictors, and the weather forecasts have a much stronger impact on the predictions than sea ice forecasts.

6. Assimilation of satellite swaths versus daily means of sea ice concentration in a regional coupled ocean–sea ice model

Abstract: Operational forecasting systems routinely assimilate daily means of sea ice concentration (SIC) from microwave radiometers in order to improve the accuracy of the forecasts. However, the temporal and spatial averaging of the individual satellite swaths into daily means of SIC entails two main drawbacks: (i) the spatial resolution of the original product is blurred (especially critical in periods with strong sub-daily sea ice movement), and (ii) the sub-daily frequency of passive microwave observations in the Arctic are not used, providing less temporal resolution in the data assimilation (DA) analysis and, therefore, in the forecast. Within the SIRANO (Sea Ice Retrievals and data Assimilation in NOrway) project, we investigate how challenges (i) and (ii) can be avoided by assimilating individual satellite swaths (level 3 uncollated) instead of daily means (level 3) of SIC. To do so, we use a regional configuration of the Barents Sea (2.5 km grid) based on the Regional Ocean Modeling System (ROMS) and the Los Alamos Sea Ice Model (CICE) together with the ensemble Kalman filter (EnKF) as the DA system. The assimilation of individual swaths significantly improves the EnKF analysis of SIC compared to the assimilation of daily means; the mean absolute difference (MAD) shows a 10 % improvement at the end of the assimilation period and a 7 % improvement at the end of the 7 d forecast period. This improvement is caused by better exploitation of the information provided by the SIC swath data, in terms of both spatial and temporal variance, compared to the case when the swaths are combined to form a daily mean before assimilation.

Abstract: Plain abdominal radiography (PAR) is routinely performed in emergency departments (EDs). This study aimed to (1) identify the indications for PAR in EDs and compare them against international guidelines, (2) uncover predictors of non-compliance with guidelines, and (3) describe the use of additional radiological examinations in EDs. Retrospective cohort study in the EDs of two hospitals in Geneva, Switzerland, including all adult patients who underwent PAR in the EDs. Indications were considered “appropriate” if complying with guidelines. Predictors of non-compliance were identified by univariate and multivariate analyses. Over 1 year, PAR was performed in 1997 patients (2.2% of all admissions). Their mean age was 59.7 years, with 53.1% of female patients. The most common indications were constipation (30.8%), suspected ileus (28.9%), and abdominal pain (15.3%). According to the French and American guidelines, only 11.8% of the PARs were indicated, while 46.2% of them complied with the Australian and British guidelines. On multivariate analysis, admission to the private hospital ED (odds ratio [OR] 3.88, 95% CI 1.78–8.45), female gender (OR 1.95, 95% CI 1.46–2.59), and an age > 65 years (OR 2.41, 95%CI 1.74–3.32) were associated with a higher risk of inappropriate PAR. Additional radiological examinations were performed in 73.7% of patients. Most indications for PAR did not comply with guidelines and elderly women appeared particularly at risk of being exposed to inappropriate examination. PAR did not prevent the need for additional examinations. Local guidelines should be developed, and initiatives should be implemented to reduce unnecessary PARs. Abstract: This paper intends to provide an overview of test results regarding the behavior of electric vehicle (EV) embedded chargers at various operating points. These results were obtained thanks to the development of a laboratory test bench that automatizes EV charging scenarios. This bench allows to check if each individual EV is suitable for smart charging, to assess its impact in terms of power quality on the network, and to assess its sensitivity to deteriorated grid conditions. Several successful tests are described as well as particular phenomena that appeared when testing some EVs.

11. Arctic sea ice data assimilation combining an ensemble Kalman filter with a novel Lagrangian sea ice model for the winter 2019–2020

Abstract: Advanced data assimilation (DA) methods, widely used in geophysical and climate studies to merge observations with numerical models, can improve state estimates and consequent forecasts. We interface the deterministic ensemble Kalman filter (DEnKF) to the Lagrangian neXt generation Sea Ice Model, neXtSIM. The ensemble is generated by perturbing the atmospheric and oceanic forcing throughout the simulations and randomly initialized ice cohesion. Our ensemble–DA system assimilates sea ice concentration (SIC) from the Ocean and Sea Ice Satellite Application Facility (OSI-SAF) and sea ice thickness (SIT) from the merged CryoSat-2 and SMOS datasets (CS2SMOS). Because neXtSIM is computationally solved on a time-dependent evolving mesh, it is a challenging application for ensemble–DA. As a solution, we perform the DEnKF analysis on a fixed and regular reference mesh, on which model variables are interpolated before the DA and then back to each member's mesh after the DA. We evaluate the impact of assimilating different types of sea ice observations on the model's forecast skills of the Arctic sea ice by comparing satellite observations and a free-run ensemble in an Arctic winter period, 2019–2020. Significant improvements in modeled SIT indicate the importance of assimilating weekly CS2SMOS SIT, while the improvements of SIC and ice extent are moderate but benefit from daily ingestion of the OSI-SAF SIC. For most of the winter, the correlation between SIT and SIC is weaker, which results in little cross-inference between the two variables in the assimilation step. Overall, the ensemble–DA system based on the stand-alone sea ice model demonstrates the feasibility of winter Arctic sea ice prediction with good computational efficiency. These results open the path toward operational implementation and the extension to multi-year assimilation.

13. Assimilation of sea surface salinities from SMOS in an Arctic coupled ocean and sea ice reanalysis

Abstract: In the Arctic, the sea surface salinity (SSS) plays a key role in processes related to water mixing and sea ice. However, the lack of salinity observations causes large uncertainties in Arctic Ocean forecasts and reanalysis. Recently the Soil Moisture and Ocean Salinity (SMOS) satellite mission was used by the Barcelona Expert Centre to develop an Arctic SSS product. In this study, we evaluate the impact of assimilating this data in a coupled ocean–ice data assimilation system. Using the deterministic ensemble Kalman filter from July to December 2016, two assimilation runs respectively assimilated two successive versions of the SMOS SSS product on top of a pre-existing reanalysis run. The runs were validated against independent in situ salinity profiles in the Arctic. The results show that the biases and the root-mean-squared differences (RMSD) of SSS are reduced by 10 % to 50 % depending on the area and highlight the importance of assimilating satellite salinity data. The time series of freshwater content (FWC) further shows that its seasonal cycle can be adjusted by assimilation of the SSS products, which is encouraging of the assimilation of SSS in a long-time reanalysis to better reproduce the Arctic water cycle.

14. Couverture vaccinale antigrippale des internes : une analyse régionale

Abstract: Introduction Le virus de la grippe est responsable d’epidemies hivernales touchant 2 a 7 millions de personnes en France. Souvent benigne, la grippe est a l’origine de complications et de deces chez les personnes fragiles. La surmortalite due a la grippe dans la region etudiee est estimee a 1623 deces pour la saison 2017–18 (+ 8,4 %). Les professionnels de sante sont en contact frequent avec des personnes a risque. La grippe pouvant avoir des consequences personnelles et professionnelles (grippe nosocomiale, absenteisme), il est necessaire de prevenir la diffusion du virus. Pour cela, l’European Center for Disease Control recommande une couverture vaccinale de 75 % dans cette population. Des etudes sur la couverture vaccinale du personnel medical existent, mais tres peu sur les internes, pourtant professionnels de sante essentiels a l’hopital. Cette etude a cible les internes de cette region et les objectifs ont ete : (i) determiner la couverture vaccinale antigrippale des internes (ii) connaitre les determinants a la vaccination. Materiels et methodes Un questionnaire en ligne anonyme a ete diffuse via les listes de diffusion les associations representant des internes en medecine, pharmacie et odontologie et les reseaux sociaux professionnels, du 25/01/2019 au 10/02/2019. Le critere d’inclusion etait d’etre interne dans la region etudiee. Resultats Parmi les 282 reponses, 254 correspondaient au critere d’inclusion. Les internes en medecine, pharmacie, biologie medicale et odontologie ont represente respectivement 47 %, 35 %, 17 % et 0,3 % des reponses. Pour la saison 2018–2019, 75 % des internes etaient vaccines contre la grippe. La couverture vaccinale a ete differente en fonction de la specialite : 84 % en medecine, 70 % en pharmacie et 50 % en biologie medicale (p Conclusion La couverture vaccinale globale des internes de la region etudiee est satisfaisante au regard des recommandations dans cette population. Les internes des specialites pharmacie et biologie medicale ont cependant des taux de couverture inferieurs a 75 %. Bien que moins en contact avec les patients, ils font partie du corps medical hospitalier. Le manque de temps et l’oubli etant les deux freins majeurs a la vaccination, il serait interessant de proposer une vaccination dans les services. Une enquete d’ampleur nationale est prevue pour augmenter la puissance de nos resultats.

16. Sea-level variability and change along the Norwegian coast between 2003 and 2018 from satellite altimetry, tide gauges, and hydrography

Abstract: Sea-level variations in coastal areas can differ significantly from those in the nearby open ocean. Monitoring coastal sea-level variations is therefore crucial to understand how climate variability can affect the densely populated coastal regions of the globe. In this paper, we study the sea-level variability along the coast of Norway by means of in situ records, satellite altimetry data, and a network of eight hydrographic stations over a period spanning 16 years (from 2003 to 2018). At first, we evaluate the performance of the ALES-reprocessed coastal altimetry dataset (1 Hz posting rate) by comparing it with the sea-level anomaly from tide gauges over a range of timescales, which include the long-term trend, the annual cycle, and the detrended and deseasoned sea-level anomaly. We find that coastal altimetry and conventional altimetry products perform similarly along the Norwegian coast. However, the agreement with tide gauges in terms of trends is on average 6 % better when we use the ALES coastal altimetry data. We later assess the steric contribution to the sea level along the Norwegian coast. While longer time series are necessary to evaluate the steric contribution to the sea-level trends, we find that the sea-level annual cycle is more affected by variations in temperature than in salinity and that both temperature and salinity give a comparable contribution to the detrended and deseasoned sea-level variability along the entire Norwegian coast. A conclusion from our study is that coastal regions poorly covered by tide gauges can benefit from our satellite-based approach to study and monitor sea-level change and variability.

17. Improved BEC SMOS Arctic Sea Surface Salinity product v3.1

Abstract: Measuring salinity from space is challenging since the sensitivity of the brightness temperature (TB) to sea surface salinity (SSS) is low (about 0.5 K psu−1), while the SSS range in the open ocean is narrow (about 5 psu, if river discharge areas are not considered). This translates into a high accuracy requirement of the radiometer (about 2–3 K). Moreover, the sensitivity of the TB to SSS at cold waters is even lower (0.3 K psu−1), making the retrieval of the SSS in the cold waters even more challenging. Due to this limitation, the ESA launched a specific initiative in 2019, the Arctic+Salinity project (AO/1-9158/18/I-BG), to produce an enhanced Arctic SSS product with better quality and resolution than the available products. This paper presents the methodologies used to produce the new enhanced Arctic SMOS SSS product (Martínez et al., 2019) . The product consists of 9 d averaged maps in an EASE 2.0 grid of 25 km. Abstract: We compared the efficacy and safety of subcutaneous (SC) sumatriptan (6 mg) with that of dihydroergotamine (DHE) nasal spray (1 mg plus optional 1 mg) in the acute treatment of migraine.Two hundred sixty-six adult migraineurs (International Headache Society criteria) completed a multicenter, double-blind, double-dummy, cross-over study. Patients took SC sumatriptan for one attack and DHE nasal spray for the other in random order. Data from both treatment periods show that at all time points from 15 minutes, SC sumatriptan was significantly better than DHE nasal spray at providing both headache relief (moderate/severe headache improving to mild/none) and resolution of headache. Similarly, SC sumatriptan was superior to DHE nasal spray for the other efficacy end points assessed in the study. Patients reported that both treatments were well tolerated. Adverse events were reported by 43% of patients taking SC sumatriptan and 22% of patients taking DHE nasal spray. These were usually mild and transient. We conclude that subcutaneous sumatriptan has a faster onset of action than DHE nasal spray and provides greater relief of acute migraine symptoms.NEUROLOGY 1996;47: 361-365

Abstract: A recent empirical literature has shaken economists' confidence in the value of aggregate (industry-level) data to illuminate production relationships. But the statistical finding 'you can't aggregate,' however well documented, is not an economic explanation. Plant-level relationships do aggregate in Depression-era blast furnace operations despite the presence of very substantial interplant heterogeneity, the most common economic cause of nonaggregability. The economic explanation of this lies in poor short-run substitutability of one plant's output for another's. Substitutability determines the importance of composition effects in understanding aggregate time series, constrains the potential cleansing effects of recessions, and therefore influences industry evolution quite broadly. Abstract: The Arctic climate system is rapidly transitioning into a new regime with a reduction in the extent of sea ice, enhanced mixing in the ocean and atmosphere, and thus enhanced coupling within the ocean–ice–atmosphere system; these physical changes are leading to ecosystem changes in the Arctic Ocean. In this review paper, we assess one of the critically important aspects of this new regime, the variability of Arctic freshwater, which plays a fundamental role in the Arctic climate system by impacting ocean stratification and sea ice formation or melt. Liquid and solid freshwater exports also affect the global climate system, notably by impacting the global ocean overturning circulation. We assess how freshwater budgets have changed relative to the 2000–2010 period. We include discussions of processes such as poleward atmospheric moisture transport, runoff from the Greenland Ice Sheet and Arctic glaciers, the role of snow on sea ice, and vertical redistribution. Notably, sea ice cover has become more seasonal and more mobile; the mass loss of the Greenland Ice Sheet increased in the 2010s (particularly in the western, northern, and southern regions) and imported warm, salty Atlantic waters have shoaled. During 2000–2010, the Arctic Oscillation and moisture transport into the Arctic are in-phase and have a positive trend. This cyclonic atmospheric circulation pattern forces reduced freshwater content on the Atlantic–Eurasian side of the Arctic Ocean and freshwater gains in the Beaufort Gyre. We show that the trend in Arctic freshwater content in the 2010s has stabilized relative to the 2000s, potentially due to an increased compensation between a freshening of the Beaufort Gyre and a reduction in freshwater in the rest of the Arctic Ocean. However, large inter-model spread across the ocean reanalyses and uncertainty in the observations used in this study prevent a definitive conclusion about the degree of this compensation.

Abstract: To associate anthropometric measures with arterial hypertension and develop a mathematical model to estimate this probability.A cross-sectional study was carried out with 3,445 adults of both sexes, between 18 and 60 years of age. Body mass index (BMI), waist-to-height ratio (WHR), waist circumference (HC), in addition age and sex were assessed. Blood pressure (BP) was measured and classified according to World Health Organization (WHO) and Brazilian Society of Hypertension (SBH) recommendations, using frequency analyses, correlation and logistic regression between variables, with p0.05.Males were twice as likely as woman to have hypertension, and for each year of life, the chance of hypertension increased 1.04 times. Odds ratio of hypertension in overweight individual increased two times, and in obese subjects, four times.Changes in body composition with advancing age, especially in men, were associated with hypertension, and assessment of these changes was relevant in the identification of this disease.

Abstract: We wished to assess the efficacy of inhaled salmeterol (SML; 50 micrograms b.i.d.) compared to a combination of slow-release theophylline and ketotifen p.o. (TK; T 300 mg+K 1 mg b.i.d.) for the treatment of nocturnal asthma. Ninety six patients with nocturnal asthma, (forced expiratory volume in one second (FEV1) 60-90% of predicted value, reversibility > or = 15%, at least two nocturnal awakenings per week) were eligible for a multicentre, double-blind, double-dummy cross-over study (14-day run-in, two successive 28-day treatment periods). Efficacy was assessed as success/failure, success being defined as the complete disappearance of nocturnal symptoms/awakening during the last week of each treatment period. There was a statistically significant difference between SML and TK for this criterion: 46% and 39% success with SML during periods I (first 28-day period) and II (following the cross-over), compared to only 15% and 26% with TK, respectively (p < 0.01). SML was also significantly better for the other criteria (lung function, rescue salbutamol intake during day and night). Side-effects were five times less frequent in SML-treated patients (p < 0.004). Efficacy and tolerance of SML were obviously far better than those of TK in patients with nocturnal asthma.

Abstract: Theophylline determination in saliva was proposed several years ago as a convenient and non-invasive alternative to monitoring plasma in children and adults. Published data demonstrated that theophylline saliva concentration linearly correlates plasma concentration. However, the variability found in interindividual serum/saliva ratios and the wide scattering among the data points precluded the clinical use of saliva for theophylline monitoring. The purpose of this study was to compare different standardized methods for obtaining stimulated saliva intending to reduce the variability in plasma/saliva ratios and to determine the most reliable one. A group of 150 ambulatory chronic asthmatic 4.5 to 20.83 (10 +/- 3.7; M +/- SD) year-old patients receiving theophylline 6.85 +/- 1.88 mg/kg every 12 h as slow release preparations for 4 to 100 days was studied. One ml venous blood and salivary specimens were simultaneously collected 5.15 +/- 0.36 h after the morning maintenance dose. In a subgroup of 75 patients, saliva was collected using first a new device called salivette, immediately followed by the collection of an expectorated sample 30 s after citric acid crystals stimulation. In the other patients saliva was collected using citric acid containing salivette. Theophylline concentration was determined using HPLC. For all types of saliva collection, salivary and plasma theophylline concentrations correlated significantly. However whichever method was used, based on the -2 to +2 SD interval, a large range of plasma theophylline was predicted from a single salivary theophylline concentration. Despite a further standardization of the sampling of saliva, saliva theophylline could not accurately predict plasma concentration.

34. Combined influence of oceanic and atmospheric circulations on Greenland sea ice concentration

Abstract: The amount and spatial extent of Greenland Sea (GS) ice are primarily controlled by the sea ice export across the Fram Strait (FS) and by local seasonal sea ice formation, melting, and sea ice dynamics. In this study, using satellite passive microwave sea ice observations, atmospheric and a coupled ocean-sea ice reanalysis system, TOPAZ4, we show that both the atmospheric and oceanic circulation in the Nordic Seas (NS) act in tandem to explain the SIC variability in the south-western GS. Northerly wind anomalies associated with anomalously low sea level pressure (SLP) over the NS reduce the sea ice export in the south-western GS due to westward Ekman drift of sea ice. On the other hand, the positive wind stress curl strengthens the cyclonic Greenland Sea Gyre (GSG) circulation in the central GS. An intensified GSG circulation may result in stronger Ekman divergence of surface cold and fresh waters away from the south-western GS. Both of these processes can reduce the freshwater content and weaken the upper-ocean stratification in the south-western GS. At the same time, warm and saline Atlantic Water (AW) anomalies are recirculated from the FS region to the south-western GS by a stronger GSG circulation. Under weakly stratified conditions, enhanced vertical mixing of these subsurface AW anomalies can warm the surface waters and inhibit new sea ice formation, further reducing the SIC in the south-western GS.

Abstract: A Known Good Die (KGD) is defined as “a package type fully supported by suppliers to meet or exceed quality, reliability, and functional data sheet specifications, with non-standardized (die specific) but completely and electronically transferable mechanical specifications”. In other words, the category of challenges associated with KGD are no different than those faced by any customer using a new or different package type. Electrical performance and power dissipation values and variations, mechanical specifications, handling issues, cooling, attachment to the next level of assembly, etc., all must be reevaluated and solutions found whenever the package changes, even though the die remains identical. Hence, KGD is no more than a different package type for a known die that incorporates mechanical specifications that cannot be standardized (unless or until die designers agree on standard die sizes and pad layouts, etc.). The challenges associated with KGD usage may be greater than with standardized packaged dice because of less standardization of mechanical specifications, handling of bare dice, etc. The ben-efits, challenges, and approaches to KGD usage are discussed in this chapter.

38. A dose-defining study of sumatriptan suppositories in the acute treatment of migraine

Abstract: In this dose-ranging, randomised, multinational, multicentre, double-blind, placebo-controlled, parallel group study, 431 patients treated a single migraine attack with study medication: sumatriptan suppository 6 mg, 12.5 mg, 25 mg, 50 mg, 100 mg, or placebo. Patients were treated in the clinic with a single dose in suppository form. All doses of sumatriptan, except 6 mg, were significantly better than placebo (p0.004) and achieved similar rates of headache relief within two hours of dosing. The highest response rate was in the 25 mg group (72%) compared with placebo (37%) (p0.001). Fewer patients required rescue medication in the active groups (1% 100 mg to 13% 6 mg) compared with placebo (17%), and more patients were able to work and function normally two hours after dosing (41%, 100 mg; 20%, placebo). The overall incidence of adverse events was similar in the placebo, 6 mg and 12.5 mg groups (14-17%) but higher in the 25 mg, 50 mg and 100 mg groups (25%, 32% and 29% respectively). Analysis of plasma sumatriptan levels indicated rapid rectal absorption for all doses (median tmax = 1.0 hr). It is concluded that sumatriptan, in doses above 6 mg, is an effective and well tolerated treatment for acute migraine. From this study doses of 12.5 mg and 25 mg sumatriptan were identified as having the best efficacy/safety profile and were evaluated further.

Abstract: Twelve healthy children in three age groups anaesthetized for minor surgery were given a single oral dose of tiaprofenic acid (3 mg.kg-1) (TA). Seven blood samples and zero to 8 and 8 to 24 h urines were collected. TA concentrations in plasma and urine were measured by HPLC. No significant difference was found between the age groups in the kinetic parameters of TA and no correlation was found between these parameters and age: tmax = 2.12 h, Cmax = 8.78 mg.l-1, AUC(0----8 h) 33.9 mg.h.l-1, AUC = 39.3 mg.h.l-1, t1/2 = 2.35 h, Vt = 0.319 l.kg-1, CL = 0.094 l.h-1.kg-1. Renal clearance was 14 ml.h-1.kg-1.33% of the TA dose was recovered in the 24 h urine, 48% of which was conjugated, whereas in adults, TA is only found in urine as conjugates. The apparent plasma clearance was significantly higher (56%) than in 12 healthy adults given 1.5 mg.kg-1 TA. Volume of distribution and t1/2 did not significantly differ between children and adults. Since no relationship has been established between plasma TA and either efficacy or toxicity, a different dose regimen cannot be recommended in 3-11 year-old children from that in adults.

Abstract: Intra-arterial chemotherapy combined with haemofiltration enables the administration of drugs to confined neoplastic tissue while limiting the systemic drug exposure. During the procedure, the cytotoxic drugs are injected into the arterial supply of the tumour and the venous blood coming from the tumour bed is pumped out and filtered trough haemofiltration unit in order to extract the cytotoxic drug not fixed on the tumour. The patients selected for such treatment failed previous intravenous chemotherapy. Thirteen patients underwent intra-arterial chemotherapy injections combined with haemofiltration procedures: 8 unresectable liver metastases from colorectal cancer and 5 pelvic recurrences from rectal cancer. Fluorouracil, mitomycin C and doxorubicin were infused. One out of thirteen patients presented a complete regression of his liver metastases, 5/13 presented a partial regression, 2/13 patients did not show any significant modification of the size of their lesions and were classified as stable disease, 5/13 patients showed a progression of their neoplastic disease. No nephrotoxicity or major gastrointestinal side effects were observed. Intra-arterial chemotherapy combined with haemofiltration procedure is a therapeutic modality that could be considered in patients with locally advanced cancer who failed previous first line intravenous chemotherapy.

Abstract: Two hundred and nineteen children (boys: 56%, girls: 44%) were included in a randomized, double-blind, multicentre (4 centres) controlled trial designed to assess the efficacy and safety of ibuprofen (IBU) in the treatment of 1 to 6 year-old children with otoscopically proven acute otitis media (AOM), either unilateral or bilateral. They randomly received 10 mg/kg IBU (n = 71), or acetaminophen (PARA) (n = 73) or placebo (PLA) (n = 75), orally, tid, for 48 hours. All received oral cefaclor (Alfatil, Lilly, France) for seven days. They were evaluated before (D0) and at the end of treatment (D2). The main criterion of response was the aspect (landmarks and color) of the tympanic membrane assessed on a semi-quantitative scale from 0 to 6. Other criteria, assessed on semi-quantitative scales, included relief of pain (0 or 1), rectal temperature (0 to 2), and overall evaluation by parents of the improvement of quality of life on three items: appetite (0 to 2), sleep (0 to 2), and playing activity (0 to 2). The results at D2 were as follows: there was no significant difference between treatment groups as to the main criterion, but only a trend for IBU and PARA to do better than PLA but not for IBU to do better than PARA. From these data there is no argument to emphasize the utility of non-steroidal anti-inflammatory drugs (NSAIDs) in treating the inflammatory signs of the tympanic membrane in otitis. There was a statistically significant difference between treatment groups at D2 for pain, IBU being superior to PLA (P < 0.01): 7%, 10% and 25% of the children were still suffering at D2 in the IBU, PARA and PLA treatment groups, respectively. The difference between PARA and PLA for pain was not statistically significant. There was no significant difference between treatment groups for the other criteria. All treatments were well and equally tolerated. Although no significant difference was found between the treatment groups on the aspect of the tympanic membrane, the efficacy of IBU was evidenced on the relief of pain, the symptom that most disturbs the child.

43. The Arctic Front and its variability in the Norwegian Sea

Abstract: The Arctic Front (AF) in the Norwegian Sea is an important biologically productive region which is well-known for its large feeding schools of pelagic fish. A suite of satellite data, a regional coupled ocean–sea ice data assimilation system (the TOPAZ reanalysis) and atmospheric reanalysis data are used to investigate the variability in the lateral and vertical structure of the AF. A method, known as “singularity analysis”, is applied on the satellite and reanalysis data for 2-D spatial analysis of the front, whereas for the vertical structure, a horizontal gradient method is used. We present new evidence of active air–sea interaction along the AF due to enhanced momentum mixing near the frontal region. The frontal structure of the AF is found to be most distinct near the Faroe Current in the south-west Norwegian Sea and along the Mohn Ridge. Coincidentally, these are the two locations along the AF where the air–sea interactions are most intense. This study investigates in particular the frontal structure and its variability along the Mohn Ridge. The seasonal variability in the strength of the AF is found to be limited to the surface. The study also provides new insights into the influence of the three dominant modes of the Norwegian Sea atmospheric circulation on the AF along the Mohn Ridge. The analyses show a weakened AF during the negative phase of the North Atlantic Oscillation (NAO−), even though the geographical location of the front does not vary. The weakening of AF during NAO− is attributed to the variability in the strength of the Norwegian Atlantic Front Current over the Mohn Ridge associated with the changes in the wind field.

Abstract: Recently two gridded sea surface salinity (SSS) products that cover the Arctic Ocean have been derived from the European Space Agency (ESA)'s Soil Moisture and Ocean Salinity (SMOS) mission: one developed by the Barcelona Expert Centre (BEC) and the other developed by the Ocean Salinity Expertise Center of the Centre Aval de Traitement des Données SMOS at IFREMER (The French Research Institute for Exploitation of the Sea) (CEC). The uncertainties of these two SSS products are quantified during the period of 2011–2013 against other SSS products: one data assimilative regional reanalysis; one data-driven reprocessing in the framework of the Copernicus Marine Environment Monitoring Services (CMEMS); two climatologies – the 2013 World Ocean Atlas (WOA) and the Polar science center Hydrographic Climatology (PHC); and in situ datasets, both assimilated and independent. The CMEMS reanalysis comes from the TOPAZ4 system, which assimilates a large set of ocean and sea-ice observations using an ensemble Kalman filter (EnKF). Another CMEMS product is the Multi-OBservations reprocessing (MOB), a multivariate objective analysis combining in situ data with satellite SSS. The monthly root mean squared deviations (RMSD) of both SMOS products, compared to the TOPAZ4 reanalysis, reach 1.5 psu in the Arctic summer, while in the winter months the BEC SSS is closer to TOPAZ4 with a deviation of 0.5 psu. The comparison of CEC satellite SSS against in situ data shows Atlantic Water that is too fresh in the Barents Sea, the Nordic Seas, and in the northern North Atlantic Ocean, consistent with the abnormally fresh deviations from TOPAZ4. Abstract: Recent progress in machine learning has shown how to forecast and, to some extent, learn the dynamics of a model from its output, resorting in particular to neural networks and deep learning techniques. We will show how the same goal can be directly achieved using data assimilation techniques without leveraging on machine learning software libraries, with a view to high-dimensional models. The dynamics of a model are learned from its observation and an ordinary differential equation (ODE) representation of this model is inferred using a recursive nonlinear regression. Because the method is embedded in a Bayesian data assimilation framework, it can learn from partial and noisy observations of a state trajectory of the physical model. Moreover, a space-wise local representation of the ODE system is introduced and is key to coping with high-dimensional models. It has recently been suggested that neural network architectures could be interpreted as dynamical systems. Reciprocally, we show that our ODE representations are reminiscent of deep learning architectures. Furthermore, numerical analysis considerations of stability shed light on the assets and limitations of the method. The method is illustrated on several chaotic discrete and continuous models of various dimensions, with or without noisy observations, with the goal of identifying or improving the model dynamics, building a surrogate or reduced model, or producing forecasts solely from observations of the physical model.

Abstract: We wished to assess the efficacy of inhaled salmeterol (SML; 50 micrograms b.i.d.) compared to a combination of slow-release theophylline and ketotifen p.o. (TK; T 300 mg+K 1 mg b.i.d.) for the treatment of nocturnal asthma. Ninety six patients with nocturnal asthma, (forced expiratory volume in one second (FEV1) 60-90% of predicted value, reversibilityor = 15%, at least two nocturnal awakenings per week) were eligible for a multicentre, double-blind, double-dummy cross-over study (14-day run-in, two successive 28-day treatment periods). Efficacy was assessed as success/failure, success being defined as the complete disappearance of nocturnal symptoms/awakening during the last week of each treatment period. There was a statistically significant difference between SML and TK for this criterion: 46% and 39% success with SML during periods I (first 28-day period) and II (following the cross-over), compared to only 15% and 26% with TK, respectively (p0.01). SML was also significantly better for the other criteria (lung function, rescue salbutamol intake during day and night). Side-effects were five times less frequent in SML-treated patients (p0.004). Efficacy and tolerance of SML were obviously far better than those of TK in patients with nocturnal asthma.

Abstract: The survival patterns of two wild strains of Escherichia coli (clinical isolates from human faeces) were compared during in situ exposure of the organisms to environmental freshwater in dialysis tubing sacs. The sublethal stress was evaluated by both standard enumeration techniques on selective or non-selective solid media and an electrochemical detection procedure based on the potentiometric measurement of lipoic acid reduction. Apparent growth rates were calculated from the potentiometric data, providing accurate information on the ability of injured bacteria to multiply. Significant differences between the survival patterns of the two bacterial strains were highlighted by both the electrochemical and the conventional techniques. The recovery of both strains on coliform-selective, lactose-deoxycholate agar was much less efficient than on non-selective solid medium. From the standard plate counts on non-selective agar, the time required for the cell population to decrease by 90% of its original value varied from 7 to 20 days, according to the strain tested. Using coliform-selective plate counts, this time was reduced to 3 or 10 days, respectively. The loss in viability was correlated with an increase in the potentiometric generation time, which increased from 19 to 33 min in 22 days for one strain, from 20 to 48 min in 8 days for the other strain. This study shows that the potentiometric technique may be suitable to evaluate the sublethal stress of micro-organisms released in natural environments.

Abstract: The binding of 125I-Tyr4 bombesin was investigated on plasma membranes of 8 human breast cancer cell lines and 2 long-term cultures of normal human breast epithelial cells. Scatchard plots were compatible with high-affinity, single-site class of receptors in 3 cell lines (KD of 0.75 x 10(-9) and 10(-9) M, Bmax of 0.75 x 10(-13) and 9.7 x 10(-13) M/mg protein in MDA-MB231 and in T47D cells, respectively) while no binding was observed in 5 other cell lines and normal epithelial cells. The neuropeptide and its structural analogues (natural or synthetic) inhibited the binding of 125I-Tyr4 bombesin in the following order of potency: gastrin-releasing peptide (GRP, EC50 = 1.7 x 10(-10) M) greater than BIM 26159 greater than bombesin, Tyr4 bombesin greater than BIM 26147 greater than litorin greater than neuromedin C. In contrast, 125I-Tyr4 bombesin binding was not displaced by neuromedin B, somatostatin, bradykinin and insulin. In agreement with our binding data, SDS-PAGE of the complex 125I-Tyr4 bombesin-receptor covalently linked by ethylene glycol-bis succinimidyl succinate (EGS) identified after autoradiography a single band with a molecular weight of 75,000, which disappeared in the presence of bombesin in excess. No transcription of either GRP or neuromedin B mRNA could be shown in tumor or normal cells. Exogenous gastrin-releasing peptide had no effect on growth of the cell lines when a serum-free medium was used, implicating that in breast cancer cell lines this receptor does not mediate growth but has a functional role.

49. Post-Irradiation Tensile Behavior and Residual Activity of Several Ferritic/Martensitic and Austenitic Steels Irradiated in Osiris Reactor at 325°C up to 9 dpa

Abstract: An experimental irradiation, named “Alexandre,” has been carried out in the Osiris experimental reactor to perform a generic study on the mechanical behavior after irradiation at 325°C of different kinds of steels suitable for use as irradiated components in a nuclear reactor [1]. The irradiated steels were austenitic stainless, martensitic (conventional and reduced activation), and ferritic-martensitic Oxide Dispersion Strengthened steels in various initial metallurgical conditions. The final dose was 9 dpa, which represents nearly a “saturation” dose for the hardening/embrittlement of both austenitic and martensitic steels. At this dose, the Yield Strength and the Ultimate Tensile Strengths are almost equal, and strong localization of the plastic deformation is often observed. After irradiation, as-quenched martensitic steels exhibit very large tensile strengths, and some of them show ductility parameters comparable with those of the tempered martensitic steels. At 9 dpa, the behavior of cold-worked steels tends to be similar to that of tempered steels with a more pronounced localization of the deformation. This indicates that the recovery of the cold-worked microstructure is not achieved at 9 dpa. Certain martensitic steels (MANET II and HT9) show tendencies to brittle behavior and exhibit a considerable degradation of their ductility parameters after irradiation. The behavior of the most chromium-rich ferritic steel (ODS-MA957) is quite surprising. It exhibits moderate hardening and good post-irradiation ductility parameters. Measurements of the dose rate have been performed after several cooling-times, of up to 53 months after unloading, to study the radioactive decay of the steels. Experimental data show that the reduced activation steels exhibit the lowest residual activity and the highest rate of relative activity decrease (90 %) during the cooling period.

Abstract: The data presented in this article are related to the research article entitled “Effect of collagen packing and moisture content on leather stiffness” (Kelly et al., 2018). This article describes how moisture content affects collagen packing and leather stiffness. Structural changes were experimentally introduced into ovine leather through biaxial strain during tanning (׳stretch tanning׳). Leather samples produced normally without strain (׳non-stretch tanned׳) and those produced by stretch tanning, were conditioned in a range of relative humidity environments and then analysed by small angle X-ray scattering and three point bend testing. The collagen D-spacing, lateral intermolecular spacing and flexural properties were measured under these varying moisture contents.
