Recent years have seen an increase in the use of wood for energy production of over 30 %, and this trend is expected to continue due to the current energy crisis and geopolitical instability. At present, residential wood burning (RWB) is one of the most important sources of organic aerosols (OAs) and black carbon (BC), posing a significant risk to air quality and health. Simultaneously, as a substantial aerosol source, RWB also holds relevance in the context of aerosol radiative effects and climate. While BC is recognized for its large light absorption cross-section, the role of OAs in light absorption is still under evaluation due to their heterogeneous composition and source-dependent optical properties. Existing studies that characterize wood-burning aerosol emissions in Europe primarily concentrate on urban and background sites and focus on BC properties. Despite the significant RWB emissions in rural areas, these locations have received comparatively less attention. The present scenario underscores the imperative for an improved understanding of RWB pollution, aerosol optical properties, and their subsequent connection to climate impacts, particularly in rural areas. We have characterized atmospheric aerosol particles from a central European rural site during wintertime in the village of Retje in Loški Potok, Slovenia, from 1 December 2017 to 7 March 2018. The village experienced extremely high aerosol concentrations produced by RWB and near-ground temperature inversion. The isolated location of the site and the substantial local emissions made it an ideal laboratory-like place for characterizing RWB aerosols with low influence from non-RWB sources under ambient conditions. The mean mass concentrations of OA and BC were 35 µg m−3 (max⁡=270 µg m−3) and 3.1 µg m−3 (max⁡=24 µg m−3), respectively. The mean total particle number concentration (10–600 nm) was 9.9×103 particles cm−3 (max⁡=59×103 particles cm−3). The mean total light absorption coefficients at 370 and 880 nm measured by an AE33 Aethalometer were 120 and 22 Mm−1 and had maximum values of 1100 and 180 Mm−1, respectively. The aerosol concentrations and absorption coefficients measured during the campaign in Loški Potok were significantly larger than reported values for several urban areas in the region with larger populations and a larger extent of aerosol sources. Here, considerable contributions from brown carbon (BrC) to the total light absorption were identified, reaching up to 60 % and 48 % in the near-UV (370 nm) and blue (470 nm) wavelengths. These contributions are up to 3 times higher than values reported for other sites impacted by wood-burning emissions. The calculated mass absorption cross-section and the absorption Ångström exponent for RWB OA were MACOA,370nm=2.4 m2 g−1, and AAEBrC,370-590nm=3.9, respectively. Simple-forcing-efficiency (SFE) calculations were performed as a sensitivity analysis to evaluate the climate impact of the RWB aerosols produced at the study site by integrating the optical properties measured during the campaign. The SFE results show a considerable forcing capacity from the local RWB aerosols, with a high sensitivity to OA absorption properties and a more substantial impact over bright surfaces like snow, typical during the coldest season with higher OA emissions from RWB. Our study's results are highly significant regarding air pollution, optical properties, and climate impact. The findings suggest that there may be an underestimation of RWB emissions in rural Europe and that further investigation is necessary.

Secondary organic aerosols (SOAs) formed by oxidation of typical precursors largely emitted by biomass burning, such as polycyclic aromatic hydrocarbons (PAHs) and furans, are still poorly characterized. We evaluated and compared the formation yields, effective density (ρeff), absorption Ångström exponent (α), and mass absorption coefficient (MAC) of laboratory-generated SOAs from three furan compounds and four PAHs. SOAs were generated in an oxidation flow reactor under day- (OH radicals) or nighttime (NO3 radicals) conditions. The ρeff, formation yields, α, and MAC of the generated SOAs varied depending on the precursor and oxidant considered. The ρeff of SOAs formed with OH and NO3 tended to increase with particle size before reaching a “plateau”, highlighting potential differences in SOA chemical composition and/or morphology, according to the particle size. Three times lower SOA formation yields were obtained with NO3 compared with OH. The yields of PAH SOAs (18 %–76 %) were five to six times higher than those obtained for furans (3 %–12 %). While furan SOAs showed low or negligible light absorption properties, PAH SOAs had a significant impact in the UV–visible region, implying a significant contribution to atmospheric brown carbon. No increase in the MAC values was observed from OH to NO3 oxidation processes, probably due to a low formation of nitrogen-containing chromophores with NO3 only (without NOx). The results obtained demonstrated that PAHs are significant SOA precursors emitted by biomass burning, through both, day- and nighttime processes, and have a substantial impact on the aerosol light absorption properties.

There exists a lack of aerosol absorption measurement techniques with low uncertainties and without artefacts. We have developed the two-wavelength Photothermal Aerosol Absorption Monitor (PTAAM-2λ), which measures the aerosol absorption coefficient at 532 and 1064 nm. Here we describe its design, calibration and mode of operation and evaluate its applicability, limits and uncertainties. The 532 nm channel was calibrated with ∼ 1 µmol mol−1 NO2, whereas the 1064 nm channel was calibrated using measured size distribution spectra of nigrosin particles and a Mie calculation. Since the aerosolized nigrosin used for calibration was dry, we determined the imaginary part of the refractive index of nigrosin from the absorbance measurements on solid thin film samples. The obtained refractive index differed considerably from the one determined using aqueous nigrosin solution. PTAAM-2λ has no scattering artefact and features very low uncertainties: 4 % and 6 % for the absorption coefficient at 532 and 1064 nm, respectively, and 9 % for the absorption Ångström exponent. The artefact-free nature of the measurement method allowed us to investigate the artefacts of filter photometers. Both the Aethalometer AE33 and CLAP suffer from cross-sensitivity to scattering – this scattering artefact is most pronounced for particles smaller than 70 nm. We observed a strong dependence of the filter multiple scattering parameter on the particle size in the 100–500 nm range. The results from the winter ambient campaign in Ljubljana showed similar multiple scattering parameter values for ambient aerosols and laboratory experiments. The spectral dependence of this parameter resulted in AE33 reporting the absorption Ångström exponent for different soot samples with values biased 0.23–0.35 higher than the PTAAM-2λ measurement. Photothermal interferometry is a promising method for reference aerosol absorption measurements.

Residential wood combustion is a widespread practice in Europe with a serious impact on air quality, especially in mountainous areas. While there is a significant number of studies conducted in deep urbanized valleys and basins, little is known about the air pollution processes in rural shallow hollows, where around 30 % of the people in mountainous areas across Europe live. We aim to determine the influence of ground temperature inversions on wood combustion aerosol pollution in hilly, rural areas. The study uses Retje karst hollow (Loški Potok, Slovenia) as a representative site for mountainous and hilly rural areas in central and south-eastern Europe with residential wood combustion. Sampling with a mobile monitoring platform along the hollow was performed in December 2017 and January 2018. The backpack mobile monitoring platform was used for the determination of equivalent black carbon (eBC) and particulate matter (PM) mass concentrations along the hollow. To ensure high quality of mobile measurement data, intercomparisons of mobile instruments with reference instruments were performed at two air quality stations during every run. Our study showed that aerosol pollution events in the relief depression were associated with high local emission intensities originating almost entirely from residential wood burning and shallow temperature inversions (58 m on average). The eBC and PM mass concentrations showed stronger associations with the potential temperature gradient (R2=0.8) than with any other meteorological parameters taken into account (ambient temperature, relative humidity, wind speed, wind direction, and precipitation). The strong association between the potential temperature gradient and pollutant concentrations suggests that even a small number of emission sources (total 243 households in the studied hollow) in similar hilly and mountainous rural areas with frequent temperature inversions can significantly increase the levels of eBC and PM and deteriorate local air quality. During temperature inversions the measured mean eBC and PM2.5 mass concentrations in the whole hollow were as high as 4.5±2.6 and 48.0 ± 27.7 µg m−3, respectively, which is comparable to larger European urban centres.

We report on an inter-comparison of black-carbon- and aerosol-absorption-measuring instruments with laboratory-generated soot particles coated with controlled amounts of secondary organic matter (SOM). The aerosol generation setup consisted of a miniCAST 5201 Type BC burner for the generation of soot particles and a new automated oxidation flow reactor based on the micro smog chamber (MSC) for the generation of SOM from the ozonolysis of α-pinene. A series of test aerosols was generated with elemental to total carbon (EC / TC) mass fraction ranging from about 90 % down to 10 % and single-scattering albedo (SSA at 637 nm) from almost 0 to about 0.7. A dual-spot Aethalometer AE33, a photoacoustic extinctiometer (PAX, 870 nm), a multi-angle absorption photometer (MAAP), a prototype photoacoustic instrument, and two prototype photo-thermal interferometers (PTAAM-2λ and MSPTI) were exposed to the test aerosols in parallel. Significant deviations in the response of the instruments were observed depending on the amount of secondary organic coating. We believe that the setup and methodology described in this study can easily be standardised and provide a straightforward and reproducible procedure for the inter-comparison and characterisation of both filter-based and in situ black-carbon-measuring (BC-measuring) instruments based on realistic test aerosols.

Aerosolized black carbon is monitored worldwide to quantify its impact on air quality and climate. Given its importance, measurements of black carbon mass concentrations must be conducted with instruments operating in quality-checked and ensured conditions to generate data which are reliable and comparable temporally and geographically. In this study, we report the results from the largest characterization and intercomparison of filter-based absorption photometers, the Aethalometer model AE33, belonging to several European monitoring networks. Under controlled laboratory conditions, a total of 23 instruments measured mass concentrations of black carbon from three well-characterized aerosol sources: synthetic soot, nigrosin particles, and ambient air from the urban background of Leipzig, Germany. The objective was to investigate the individual performance of the instruments and their comparability; we analyzed the response of the instruments to the different aerosol sources and the impact caused by the use of obsolete filter materials and the application of maintenance activities. Differences in the instrument-to-instrument variabilities from equivalent black carbon (eBC) concentrations reported at 880 nm were determined before maintenance activities (for soot measurements, average deviation from total least square regression was −2.0 % and the range −16 % to 7 %; for nigrosin measurements, average deviation was 0.4 % and the range −15 % to 17 %), and after they were carried out (for soot measurements, average deviation was −1.0 % and the range −14 % to 8 %; for nigrosin measurements, the average deviation was 0.5 % and the range −12 % to 15 %). The deviations are in most of the cases explained by the type of filter material employed by the instruments, the total particle load on the filter, and the flow calibration. The results of this intercomparison activity show that relatively small unit-to-unit variability of AE33-based particle light absorbing measurements is possible with well-maintained instruments. It is crucial to follow the guidelines for maintenance activities and the use of the proper filter tape in the AE33 to ensure high quality and comparable black carbon (BC) measurements among international observational networks.

We have developed a novel single-beam photothermal interferometer and present here its application for the measurement of aerosol light absorption. The use of only a single laser beam allows for a compact optical set-up and significantly easier alignment compared to standard dual-beam photothermal interferometers, making it ideal for field measurements. Due to a unique configuration of the reference interferometer arm, light absorption by aerosols can be determined directly – even in the presence of light-absorbing gases. The instrument can be calibrated directly with light-absorbing gases, such as NO2, and can be used to calibrate other light absorption instruments. The detection limits (1σ) for absorption for 10 and 60 s averaging times were determined to be 14.6 and 7.4 Mm−1, respectively, which for a mass absorption cross section of 10 m2 g−1 leads to equivalent black carbon concentration detection limits of 1460 and 740 ng m−3, respectively. The detection limit could be reduced further by improvements to the isolation of the instrument and the signal detection and processing schemes employed.

We present a new method for the determination of the source-specific black carbon emission rates. The methodology was applied in two different environments: an urban location in Ljubljana and a rural one in the Vipava valley (Slovenia, Europe), which differ in pollution sources and topography. The atmospheric dynamics was quantified using the atmospheric radon (222Rn) concentration to determine the mixing layer height for periods of thermally driven planetary boundary layer evolution. The black carbon emission rate was determined using an improved box model taking into account boundary layer depth and a horizontal advection term, describing the temporal and spatial exponential decay of black carbon concentration. The rural Vipava valley is impacted by a significantly higher contribution to black carbon concentration from biomass burning during winter (60 %) in comparison to Ljubljana (27 %). Daily averaged black carbon emission rates in Ljubljana were 210 ± 110 and 260 ± 110 µgm-2h-1 in spring and winter, respectively. Overall black carbon emission rates in Vipava valley were only slightly lower compared to Ljubljana: 150 ± 60 and 250 ± 160 µgm-2h-1 in spring and winter, respectively. Different daily dynamics of biomass burning and traffic emissions was responsible for slightly higher contribution of biomass burning to measured black carbon concentration, compared to the fraction of its emission rate. Coupling the high-time-resolution measurements of black carbon concentration with atmospheric radon concentration measurements can provide a useful tool for direct, highly time-resolved measurements of the intensity of emission sources. Source-specific emission rates can be used to assess the efficiency of pollution mitigation measures over longer time periods, thereby avoiding the influence of variable meteorology.

We present a newly developed total carbon analyzer (TCA08) and a method for online speciation of carbonaceous aerosol with a high time resolution. The total carbon content is determined by flash heating of a sample collected on a quartz-fiber filter with a time base between 20 min and 24 h. The limit of detection is approximately 0.3 µg C, which corresponds to a concentration of 0.3 µg C m−3 at a sample flow rate of 16.7 L min−1 and a 1 h sampling time base. The concentration of particulate equivalent organic carbon (OC) is determined by subtracting black carbon concentration, concurrently measured optically by an Aethalometer®, from the total carbon concentration measured by the TCA08. The combination of the TCA08 and Aethalometer (AE33) is an easy-to-deploy and low-maintenance continuous measurement technique for the high-time-resolution determination of equivalent organic and elemental carbon (EC) in different particulate matter size fractions, which avoids pyrolytic correction and the need for high-purity compressed gases. The performance of this online method relative to the standardized off-line thermo-optical OC–EC method and respective instruments was evaluated during a winter field campaign at an urban background location in Ljubljana, Slovenia. The organic-matter-to-organic-carbon ratio obtained from the comparison with an aerosol chemical speciation monitor (ACSM) was OM/OC=1.8, in the expected range.

Atmospheric mineral dust influences Earth's radiative budget, cloud formation, and lifetime; has adverse health effects; and affects air quality through the increase of regulatory PM10 concentrations, making its real-time quantification in the atmosphere of strategic importance. Only few near-real-time techniques can discriminate dust aerosol in PM10 samples and they are based on the dust chemical composition. The online determination of mineral dust using aerosol absorption photometers offers an interesting and competitive alternative but remains a difficult task to achieve. This is particularly challenging when dust is mixed with black carbon, which features a much higher mass absorption cross section. We build on previous work using filter photometers and present here for the first time a highly time-resolved online technique for quantification of mineral dust concentration by coupling a high-flow virtual impactor (VI) sampler that concentrates coarse particles with an aerosol absorption photometer (Aethalometer, model AE33). The absorption of concentrated dust particles is obtained by subtracting the absorption of the submicron (PM1) aerosol fraction from the absorption of the virtual impactor sample (VI-PM1 method). This real-time method for detecting desert dust was tested in the field for a period of 2 months (April and May 2016) at a regional background site of Cyprus, in the Eastern Mediterranean. Several intense desert mineral dust events were observed during the field campaign with dust concentration in PM10 up to 45 µg m−3. Mineral dust was present most of the time during the campaign with an average PM10 of about 8 µg m−3. Mineral dust absorption was most prominent at short wavelengths, yielding an average mass absorption cross section (MAC) of 0.24±0.01 m2 g−1 at 370 nm and an absorption Ångström exponent of 1.41±0.29. This MAC value can be used as a site-specific parameter for online determination of mineral dust concentration. The uncertainty of the proposed method is discussed by comparing and validating it with different methods.

The present study investigates and compares the ground and in-flight performance of three miniaturized aerosol absorption sensors integrated on board small-sized Unmanned Aerial Systems (UASs). These sensors were evaluated during two contrasted field campaigns performed at an urban site, impacted mainly by local traffic and domestic wood burning sources (Athens, Greece), and at a remote regional background site, impacted by long-range transported sources including dust (Cyprus Atmospheric Observatory, Agia Marina Xyliatou, Cyprus). The miniaturized sensors were first intercompared at the ground-level against two commercially available instruments used as a reference. The measured signal of the miniaturized sensors was converted into the absorption coefficient and equivalent black carbon concentration (eBC). When applicable, signal saturation corrections were applied, following the suggestions of the manufacturers. The aerosol absorption sensors exhibited similar behavior against the reference instruments during the two campaigns, despite the diversity of the aerosol origin, chemical composition, sources, and concentration levels. The deviation from the reference during both campaigns concerning (eBC) mass was less than 8 %, while for the absorption coefficient it was at least 15 %. This indicates that those sensors that report black carbon mass are tuned and corrected to measure eBC more accurately than the absorption coefficient. The overall potential use of miniature aerosol absorption sensors on board small UASs is also illustrated. UAS-based absorption measurements were used to investigate the vertical distribution of eBC over Athens up to 1 km above sea level during January 2016, exceeding the top of the planetary boundary layer (PBL). Our results reveal a heterogeneous boundary layer concentration of absorbing aerosol within the PBL intensified in the early morning hours due to the concurrent peak traffic emissions at ground-level and the fast development of the boundary layer. After the full development of the PBL, homogenous concentrations are observed from 100 m a.g.l. to the PBL top.

We investigate the optical properties of light-absorbing organic carbon (brown carbon) from domestic wood combustion as a function of simulated atmospheric aging. At shorter wavelengths (370–470 nm), light absorption by brown carbon from primary organic aerosol (POA) and secondary organic aerosol (SOA) formed during aging was around 10 % and 20 %, respectively, of the total aerosol absorption (brown carbon plus black carbon). The mass absorption cross section (MAC) determined for black carbon (BC, 13.7 m2 g−1 at 370 nm, with geometric standard deviation GSD =1.1) was consistent with that recommended by Bond et al. (2006). The corresponding MAC of POA (5.5 m2 g−1; GSD =1.2) was higher than that of SOA (2.4 m2 g−1; GSD =1.3) at 370 nm. However, SOA presents a substantial mass fraction, with a measured average SOA ∕ POA mass ratio after aging of ∼5 and therefore contributes significantly to the overall light absorption, highlighting the importance of wood-combustion SOA as a source of atmospheric brown carbon. The wavelength dependence of POA and SOA light absorption between 370 and 660 nm is well described with absorption Ångström exponents of 4.6 and 5.6, respectively. UV-visible absorbance measurements of water and methanol-extracted OA were also performed, showing that the majority of the light-absorbing OA is water insoluble even after aging.

Black carbon is a primary aerosol tracer for high-temperature combustion emissions and can be used to characterize the time evolution of its sources. It is correlated with a decrease in public health and contributes to atmospheric warming. Black carbon measurements are usually conducted with absorption filter photometers, which are prone to several artifacts, including the filter-loading effect – a saturation of the instrumental response due to the accumulation of the sample in the filter matrix. In this paper, we investigate the hypothesis that this filter-loading effect depends on the optical properties of particles present in the filter matrix, especially on the black carbon particle coating. We conducted field campaigns in contrasting environments to determine the influence of source characteristics, particle age and coating on the magnitude of the filter-loading effect. High-time-resolution measurements of the filter-loading parameter in filter absorption photometers show daily and seasonal variations of the effect. The variation is most pronounced in the near-infrared region, where the black carbon mass concentration is determined. During winter, the filter-loading parameter value increases with the absorption Ångström exponent. It is suggested that this effect is related to the size of the black carbon particle core as the wood burning (with higher values of the absorption Ångström exponent) produces soot particles with larger diameters. A reduction of the filter-loading effect is correlated with the availability of the coating material. As the coating of ambient aerosols is reduced or removed, the filter-loading parameter increases. Coatings composed of ammonium sulfate and secondary organics seem to be responsible for the variation of the loading effect. The potential source contribution function analysis shows that high values of the filter-loading parameter in the infrared are indicative of local pollution, whereas low values of the filter-loading parameter result from ageing and coating during long-range transport. Our results show that the filter-loading parameter can be used as a proxy for determination of the particle coating, thus allowing for differentiation between local/fresh and transported/aged particles.

Aerosol black carbon is a unique primary tracer for combustion emissions. It affects the optical properties of the atmosphere and is recognized as the second most important anthropogenic forcing agent for climate change. It is the primary tracer for adverse health effects caused by air pollution. For the accurate determination of mass equivalent black carbon concentrations in the air and for source apportionment of the concentrations, optical measurements by filter-based absorption photometers must take into account the "filter loading effect". We present a new real-time loading effect compensation algorithm based on a two parallel spot measurement of optical absorption. This algorithm has been incorporated into the new Aethalometer model AE33. Intercomparison studies show excellent reproducibility of the AE33 measurements and very good agreement with post-processed data obtained using earlier Aethalometer models and other filter-based absorption photometers. The real-time loading effect compensation algorithm provides the high-quality data necessary for real-time source apportionment and for determination of the temporal variation of the compensation parameter k.

Residential wood burning contributes to the total atmospheric aerosol burden; however, large uncertainties remain in the magnitude and characteristics of wood burning products. Primary emissions are influenced by a variety of parameters, including appliance type, burner wood load and wood type. In addition to directly emitted particles, previous laboratory studies have shown that oxidation of gas-phase emissions produces compounds with sufficiently low volatility to readily partition to the particles, forming considerable quantities of secondary organic aerosol (SOA). However, relatively little is known about wood burning SOA, and the effects of burn parameters on SOA formation and composition are yet to be determined. There is clearly a need for further study of primary and secondary wood combustion aerosols to advance our knowledge of atmospheric aerosols and their impacts on health, air quality and climate. For the first time, smog chamber experiments were conducted to investigate the effects of wood loading on both primary and secondary wood combustion products. Products were characterized using a range of particle- and gas-phase instrumentation, including an aerosol mass spectrometer (AMS). A novel approach for polycyclic aromatic hydrocarbon (PAH) quantification from AMS data was developed and results were compared to those from GC-MS analysis of filter samples. Similar total particle mass emission factors were observed under high and average wood loadings; however, high fuel loadings were found to generate significantly higher contributions of PAHs to the total organic aerosol (OA) mass compared to average loadings. PAHs contributed 15 ± 4% (mean ±2 sample standard deviations) to the total OA mass in high-load experiments, compared to 4 ± 1% in average-load experiments. With aging, total OA concentrations increased by a factor of 3 ± 1 for high load experiments compared to 1.6 ± 0.4 for average-load experiments. In the AMS, an increase in PAH and aromatic signature ions at lower m / z values, likely fragments from larger functionalized PAHs, was observed with aging. Filter samples also showed an increase in functionalized PAHs in the particles with aging, particularly oxidized naphthalene species. As PAHs and their oxidation products are known to have deleterious effects on health, this is a noteworthy finding to aid in the mitigation of negative wood burning impacts by improving burner operation protocols.

We have used two methods for measuring emission factors (EFs) in real driving conditions on five cars in a controlled environment: the stationary method, where the investigated vehicle drives by the stationary measurement platform and the composition of the plume is measured, and the chasing method, where a mobile measurement platform drives behind the investigated vehicle. We measured EFs of black carbon and particle number concentration. The stationary method was tested for repeatability at different speeds and on a slope. The chasing method was tested on a test track and compared to the portable emission measurement system. We further developed the data processing algorithm for both methods, trying to improve consistency, determine the plume duration, limit the background influence and facilitate automatic processing of measurements. The comparison of emission factors determined by the two methods showed good agreement. EFs of a single car measured with either method have a specific distribution with a characteristic value and a long tail of super emissions. Measuring EFs at different speeds or slopes did not significantly influence the EFs of different cars; hence, we propose a new description of vehicle emissions that is not related to kinematic or engine parameters, and we rather describe the vehicle EF with a characteristic value and a super emission tail.

We present a new mobile environmental reaction chamber for the simulation of the atmospheric aging of different emission sources without limitation from the instruments or facilities available at any single site. Photochemistry is simulated using a set of 40 UV lights (total power 4 KW). Characterisation of the emission spectrum of these lights shows that atmospheric aging of emissions may be simulated over a range of temperatures (−7 to 25 °C). A photolysis rate of NO2, JNO2, of (8.0 ± 0.7) × 10−3 s−1 was determined at 25 °C. We demonstrate the utility of this new system by presenting results on the aging (OH = 12 × 106 cm−3 h) of emissions from a modern (Euro 5) gasoline car operated during a driving cycle (New European Driving Cycle, NEDC) on a chassis dynamometer in a vehicle test cell. Emissions from the entire NEDC were sampled and aged in the chamber. Total organic aerosol (OA; primary organic aerosol (POA) emission + secondary organic aerosol (SOA) formation) was (369.8–397.5)10−3 g kg−1 fuel, or (13.2–15.4) × 10−3 g km−1, after aging, with aged OA/POA in the range 9–15. A thorough investigation of the composition of the gas phase emissions suggests that the observed SOA is from previously unconsidered precursors and processes. This large enhancement in particulate matter mass from gasoline vehicle aerosol emissions due to SOA formation, if it occurs across a wider range of gasoline vehicles, would have significant implications for our understanding of the contribution of on-road gasoline vehicles to ambient aerosols.

This study aimed to functionally characterize β 2 -adrenergic (β 2 AR) and insulin receptor (IR) heteromers in regard to β-arrestin 2 (βarr2) recruitment and cAMP signaling and to examine the involvement of the cytoplasmic portion of the IR β chain in heteromerization with β 2 AR. Evidence for β 2 AR:IR:βarr2 complex formation and the specificity of the IR:βarr2 interaction was first provided by bioinfomatics analysis. Receptor-heteromer investigation technology (HIT) then provided functional evidence of β 2 AR:IR heterodimerization by showing isoproterenol-induced but not insulin-induced GFP 2 -βarr2 recruitment to the β 2 AR:IR complex; the IR:βarr2 interaction was found to only be constitutive. The constitutive IR:βarr2 BRET signal (BRET const ) was significantly smaller in cells coexpressing IR-RLuc8 and a GFP 2 -βarr2 1-185 mutant lacking the proposed IR binding domain. β 2 AR:IR heteromerization also influenced the pharmacological phenotype of β 2 AR, i.e., its efficacy in recruiting βarr2 and activating cAMP signaling. Evidence suggesting involvement of the cytoplasmic portion of the IR β chain in the interaction with β 2 AR was provided by BRET 2 saturation and HIT assays using an IR 1-1271 stop mutant lacking the IR C-terminal tail region. For the complex consisting of IR 1-1271-RLuc8:β 2 AR-GFP 2 , saturation was not reached, most likely reflecting random collisions between IR 1-1271 and β 2 AR. Furthermore, in the HIT assay, no substantial agonist-induced increase in the BRET 2 signal was detected that would be indicative of βarr2 recruitment to the IR 1-1271:β 2 AR heteromer. Complementary 3D visualization of β 2 AR:IR provided supporting evidence for stability of the heterotetramer complex and identified amino acid residues involved in β 2 AR:IR heteromerization. Residential wood burning is a major source of poorly characterized, deleterious particulate matter, whose composition and toxicity may vary with wood type, burning condition and photochemical age. The causative link between ambient wood particle constituents and observed adverse health effects is currently lacking. Here we investigate the relationship between chemical properties of primary and atmospherically aged wood combustion particles and acute toxicity in human airway epithelial cells. Emissions from a log wood burner were diluted and injected into a smog chamber for photochemical aging. After concentration-enrichment and removal of oxidizing gases, directly emitted and atmospherically aged particles were deposited on cell cultures at the air-liquid interface for 2 hours in an aerosol deposition chamber mimicking physiological conditions in lungs. Cell models were fully differentiated normal and diseased (cystic fibrosis and asthma) human bronchial epithelia (HBE) and the bronchial epithelial cell line BEAS-2B. Cell responses were assessed at 24 hours after aerosol exposure. Atmospherically relevant doses of wood combustion particles significantly increased cell death in all but the asthma cell model. Expression of oxidative stress markers increased in HBE from all donors. Increased cell death and inflammatory responses could not be assigned to a single chemical fraction of the particles. Exposure to primary and aged wood combustion particles caused adverse effects to airway epithelia, apparently induced by several interacting components.

Glucose metabolism is under the cooperative regulation of both insulin receptor (IR) and β(2)-adrenergic receptor (β(2)AR), which represent the receptor tyrosine kinases (RTKs) and seven transmembrane receptors (7TMRs), respectively. Studies demonstrating cross-talk between these two receptors and their endogenous coexpression have suggested their possible interactions. To evaluate the effect of IR and prospective heteromerization on β(2)AR properties, we showed that IR coexpression had no effect on the ligand binding properties of β(2)AR; however, IR reduced β(2)AR surface expression and accelerated its internalization. Additionally, both receptors displayed a similar distribution pattern with a high degree of colocalization. To test the possible direct interaction between β(2)AR and IR, we employed quantitative BRET(2) saturation and competition assays. Saturation assay data suggested constitutive β(2)AR and IR homo- and heteromerization. Calculated acceptor/donor (AD50) values as a measure of the relative affinity for homo- and heteromer formation differed among the heteromers that could not be explained by a simple dimer model. In heterologous competition assays, a transient increase in the BRET(2) signal with a subsequent hyperbolical decrease was observed, suggesting higher-order heteromer formation. To complement the BRET(2) data, we employed the informational spectrum method (ISM), a virtual spectroscopy method to investigate protein-protein interactions. Computational peptide scanning of β(2)AR and IR identified intracellular domains encompassing residues at the end of the 7th TM domain and C-terminal tail of β(2)AR and a cytoplasmic part of the IR β chain as prospective interaction domains. ISM further suggested a high probability of heteromer formation and homodimers as basic units engaged in heteromerization. In summary, our data suggest direct interaction and higher-order β(2)AR:IR oligomer formation, likely comprising heteromers of homodimers.

The idea that seven transmembrane receptors (7TMRs; also designated G-protein coupled receptors, GPCRs) might form dimers or higher order oligomeric complexes was formulated more than 20 years ago and has been intensively studied since then. In the last decade, bioluminescence resonance energy transfer (BRET) has been one of the most frequently used biophysical methods for studying 7TMRs oligomerization. This technique enables monitoring physical interactions between protein partners in living cells fused to donor and acceptor moieties. It relies on non-radiative transfer of energy between donor and acceptor, depending on their intermolecular distance (1-10 nm) and relative orientation. Results derived from BRET-based techniques are very persuasive; however, they need appropriate controls and critical interpretation. To overcome concerns about the specificity of BRET-derived results, a set of experiments has been proposed, including negative control with a non-interacting receptor or protein, BRET dilution, saturation, and competition assays. This article presents the theoretical background behind BRET assays, then outlines mathematical models for quantitative interpretation of BRET saturation and competition assay results, gives examples of their utilization and discusses the possibilities of quantitative analysis of data generated with other RET-based techniques.

An ultra-sensitive digital imaging system was employed to visualize oxidative stress in intact L. minor plants exposed to Cd, Cu, menadione, AAPH, and ascorbate in real time. The increase of ROS production was assessed by measuring the rate of fluorescence intensity increases of the test medium supplemented with a fluorescing probe (dichlorofluorescein diacetate). The addition of 100 μM CdCl₂ or 100 μM CuSO₄ to the growth medium resulted in a significant increase of medium fluorescence. Additionally, CuSO₄ caused a significantly higher fluorescence intensity than CdCl₂ did. A strong positive correlation (R² = 0.99) between menadione concentration and fluorescence intensity was observed. The positive correlation between AAPH concentration and fluorescence intensity was not as strong as in the case of menadione (R² = 0.81). Menadione induced a stronger oxidative stress than similar concentration of AAPH. The addition of 100 μM ascorbate to L. minor treated with 50 μM menadione significantly reduced the fluorescence intensity increase. A linear trend of the fluorescence increase was observed in all treatments, indicating that chemical-induced oxidative stress is a gradual process and that the applied concentrations of the chemicals caused a constant increased production of ROS with different intensities, depending on the treatment. This is the combined result of a gradual diminishing of antioxidant reserves and accumulating oxidative damage. The applicability of the delayed fluorescence (DF) for the purpose of distinguishing the cells growing in different nutrient conditions was researched on the marine unicellular algae Dunaliella tertiolecta Butcher (Chlorophyta). The DF intensity (DFI), as a measure of living algal biomass, was compared with other biomass measures--the cell concentration, chlorophyll a and fluorescence. The photosynthetic activity index (PhAI), a non-dimensional physiological index of photosynthesis calculated from a combination of DFI and F(0) was introduced. The nitrogen deprivation was indicated by more than 50% drop of PhAI. DF decay kinetics was measured with two different illuminations (<600 nm and >650 nm). The measured curves were divided and the resulting peak utilized for the differentiation among nutrient conditions. DF decay kinetics of D. tertiolecta differed among the cells growing in various nutrient conditions, indicating changes in the photosynthesis physiology.

Prorocentrum minimum is a potentially harmful and widely distributed marine dinoflagellate. Several P. minimum strains have already been studied, showing phylogenetical relations of strains isolated from the same geographical regions. Similarity among the strains was further examined on the basis of their physiology. Pigment composition and concentration, as well as delayed fluorescence (DF) decay kinetics and intensity, were measured in four P. minimum strains isolated from the Baltic and Adriatic Seas. The strains were grown at two salinities characteristic of the Baltic (8 PSU) and North Adriatic Seas (32 PSU). Strain differences in DF decay kinetics and growth did not always follow their genetic relations. While two strains showed similarities to the previously described strains from the Baltic and Adriatic Seas in DF parameters, the other two strains seemed to be specific. The differences among strains isolated from the same sea could stem from adaptations to conditions in the specific habitats. Cluster analysis based on the ratio of individual carotenoid pigments concentrations to the chlorophyll a concentration or to total carotenoids were not conclusive in showing relations among the strains. Among the measured pigments, only peridinin concentration depended on salinity in all strains.

Goal, Scope and Background: Elevated concentrations of copper in the environment result in accumulation of the metal in plants and cause an increase in reactive oxidative species (ROS). The first response to elevated amounts of ROS is increased levels of enzymatic and non-enzymatic antioxidants that reduce oxidative stress. The aim of our study was to evaluate the early stages of antioxidative responses to the low copper concentrations usually present in moderately polluted environments. In addition, some other parameters were examined to evaluate the effect of copper on plants., Methods: Duckweed (Lemna minor L.) was exposed to different concentrations of copper sulphate for up to 24 hours. Glutathione concentration and enzymatic activities of catalase, guaiacol peroxidase and glutathione reductase were measured spectrophotometrically. Additionally, delayed and prompt chlorophyll fluorescence was measured by luminometry and fluorometry, respectively. The accumulation of copper in plants exposed for 24 hours to various concentrations of copper sulphate was measured by flame atomic absorption spectrophotometry., Results: The treatment of plants with copper sulphate resulted in an immediate decrease of the glutathione pool, which was replenished after 24 hours at CuSO4 concentrations lower than 2 microM. Higher CuSO4 concentrations caused a decrease of reduced glutathione. The responses of the antioxidant enzymes glutathione reductase, guaiacol peroxidase and catalase to CuSO4 differed during the first six hours of exposure, but their enzyme activities all increased after 24 hours of exposure. All these enzymes displayed biphasic activity curves with maximum values between 0.5 microM and 1 microM CuSO4. The response of guaiacol peroxidase was the most pronounced and statistically significantly specific and that of catalase the least. Delayed chlorophyll fluorescence decreased after exposure to 1 microM CuSO4, but no significant effect on maximum quantum yield of photosystem II (Fv/Fm) was observed. L. minor accumulated relatively high concentrations of copper. The accumulation rate was higher at lower concentrations of copper in the test medium (up to 2 microM CuSO4) than at concentrations above 2 microM CuSO4., Discussion: One of the most pronounced antioxidative responses to copper exposure was modified levels of oxidized and reduced forms of glutathione. The decrease of the glutathione pool is most probably coupled with induced production of phytochelatins. Antioxidative enzymes showed the biphasic enzyme activity characteristic of stress response. Guaiacol peroxidase exhibited the greatest significant increase of activity, even at higher CuSO4 concentrations at which the activity of catalase and glutathione reductase dropped. The intensity of delayed chlorophyll fluorescence decreased, indicating reduced photosynthesis of plants under stress. All the measured parameters showed that plants respond to even low copper concentrations very soon after exposure. The accumulation rate of copper in duckweed tissues indicates that L. minor is an accumulator species., Conclusions: The synchronized and prompt inducibility of antioxidants indicates their involvement in a general plant defence strategy for coping with metal-induced oxidative stress. Glutathione concentration and guaiacol peroxidase activity were found to be the most sensitive of the early indicators of exposure to copper concentrations present in polluted water bodies., Recommendation and Perspectives: The experimental design of the present study allowed us to compare the sensitivity of various methods and parameters for detecting plant responses to heavy metal-induced oxidative stress. The level of glutathione and the enzyme activities of guaiacol peroxidase and glutathione reductase could be used as a rapidly determined early warning system in toxicity studies.

Using bioluminescence resonance energy transfer (BRET) we studied opsin oligomerization in heterologous expression systems and quantitatively assessed its oligomerization state. BRET2 saturation and competition experiments were performed with live COS-7 cells expressing Rluc-and GFP2-tagged receptor constructs. BRET2 saturation curves obtained were hyperbolic, and the calculated oligomerization state (N = 1 for dimers) suggested that opsin (N = 1.34 +/- 0.25) forms higher oligomers. Very high BRET2 values obtained for the opsin homo-dimer pair indicated a large energy transfer efficiency (E) and for cases where E >> 0.1 a modified saturation curve was proposed. The existence of homo-dimer complexes was additionally supported by competition assay results and was also observed in HEK-293 cells. Furthermore, evidence was provided for homo-and hetero-dimerization of family A (beta2-adrenergic) and B (gastric inhibitory polypeptide, GIP) receptors. In summary, these experiments demonstrate homo-and hetero-dimerization for opsin, beta 2-adrenergic, and GIP receptors.
