Background and Objectives Insufficient ethnoracial diversity is a pervasive challenge in Alzheimer’s disease (AD) research. The Recruitment Innovations for Diversity Enhancement (RIDE) is grounded in the premise that culturally informed narratives of research participation can inspire individuals from a given culture-sharing group to consider research enrollment. This study examines factors associated with interest in AD research among Black or African American adults following exposure to RIDE narrative campaign materials. Research Design and Methods A community-based sample of 500 Black or African American adults viewed RIDE narrative materials online and completed a survey of perceptions about research, AD risk, and likelihood of enrolling in AD research. Logistic regression examined predictors and mediators of self-reported likelihood of participating in AD research. Results Most (72%) participants reported interest in being contacted for AD research opportunities. After controlling for key variables, prior experience with clinical research and trust in medical researchers emerged as independent predictors of likelihood of enrolling in AD research. Perceived burden of AD research partially mediated the effects of prior research experience and trust on likelihood of enrollment. Perceived benefits of AD research also played a mediating role, accounting for over one third of the effect of trust on likelihood of enrollment. Discussion and Implications This study advances the field’s understanding of how narrative may function to enhance diversity in AD research. Findings suggest that participant narratives should address experiences regarding the burdens and potential benefits of AD research participation as these factors may influence decisions leading to subsequent research enrollment.

This study aimed to examine the complex relations between two known predictors of bystander decisions in bullying incidents—empathy and family contextual factors—and bystander help-seeking from two preferred choices (i.e., adults and peers). In particular, we examined the mediating role of cognitive and affective empathy on the relation between parental attachment and bystander help-seeking in 826 adolescents in the 6th and 9th grade (49.9% female) using four bullying/acts of aggression scenarios. Results indicated that affective, but not cognitive, empathy served as a partial mediator for the relationship in boys, suggesting that both empathy and the family context can play a role in bystander decisions to intervene. On the other hand, empathy did not serve as a mediator for girls, suggesting that girls do not need the added push of empathy in order to intervene in bullying situations. Results suggest emphasizing home-school collaboration for boys, in particular, in bullying prevention programs in order to maximize efforts of bystanders. © 2022, The Author(s), under exclusive licence to Springer Science+Business Media, LLC, part of Springer Nature.

Artificial levees are a major human modification of river corridors, but we still do not have a clear understanding of how artificial levees affect floodplain extent at regional and larger scales. We estimated changes in river-floodplain connectivity due to artificial levees in the contiguous United States (CONUS) using a combination of artificial levee databases, delineations of floodplain areas, and deletion of artificial levees from topography. Our results indicate that artificial levees do not only decrease floodplain extent but also alter locations of floodplain connectivity. Anthropogenically connected and disconnected locations are similar in land cover and are predominantly, in decreasing order of extent, cultivated, wetland, forested, and developed land cover types, with more than 30% of the entire floodplain area in the CONUS cultivated or developed. This study indicates that artificial levees cause complex changes in river-floodplain connectivity and can increase flooded areas in some rivers.

Cobalamin-dependent radical S-adenosylmethionine (SAM) methylases catalyze key steps in the biosynthesis of numerous biomolecules, including protein cofactors, antibiotics, herbicides, and other natural products, but have remained a relatively understudied subclass of radical SAM enzymes due to their inherent insolubility upon overproduction in Escherichia coli. These enzymes contain two cofactors: a [4Fe-4S] cluster that is ligated by three cysteine residues, and a cobalamin cofactor typically bound by residues in the N-terminal portion of the enzyme. Recent advances in the expression and purification of these enzymes in their active states and with both cofactors present has allowed for more detailed biochemical studies as well as structure determination by X-ray crystallography. Herein, we use KsTsrM and TokK to highlight methods for the structural characterization of cobalamin-dependent radical SAM (RS) enzymes and describe recent advances in in the overproduction and purification of these enzymes.

We show the improvement to cosmological constraints from galaxy cluster surveys with the addition of CMB-cluster lensing data. We explore the cosmological implications of adding mass information from the 3.1$\sigma$ detection of gravitational lensing of the cosmic microwave background (CMB) by galaxy clusters to the Sunyaev-Zel'dovich (SZ) selected galaxy cluster sample from the 2500 deg$^2$ SPT-SZ survey and targeted optical and X-ray followup data. In the $\Lambda$CDM model, the combination of the cluster sample with the Planck power spectrum measurements prefers $\sigma_8 \left(\Omega_m/0.3 \right)^{0.5} = 0.831 \pm 0.020$. Adding the cluster data reduces the uncertainty on this quantity by a factor of 1.4, which is unchanged whether or not the 3.1$\sigma$ CMB-cluster lensing measurement is included. We then forecast the impact of CMB-cluster lensing measurements with future cluster catalogs. Adding CMB-cluster lensing measurements to the SZ cluster catalog of the on-going SPT-3G survey is expected to improve the expected constraint on the dark energy equation of state $w$ by a factor of 1.3 to $\sigma(w) = 0.19$. We find the largest improvements from CMB-cluster lensing measurements to be for $\sigma_8$, where adding CMB-cluster lensing data to the cluster number counts reduces the expected uncertainty on $\sigma_8$ by factors of 2.4 and 3.6 for SPT-3G and CMB-S4 respectively., Comment: Comments are welcome

Citrullinemia type I (CTLN-I) results from the absence or deficiency of argininosuccinate synthetase (AS), a 46 kDa enzyme that acts in the cytosol of hepatocytes to convert aspartic acid and citrulline into argininosuccinic acid. AS is an essential component of the urea cycle, and its absence or deficiency results in the harmful accumulation of ammonia in blood and cerebrospinal fluid. No disease-modifying treatment of CTLN-I exists. Here we report that the cell-permeant miniature protein (CPMP) ZF5.3 (ZF) can deliver AS to the cytosol of cells in culture and the livers of healthy mice. The fusion protein ZF-AS is catalytically active in vitro, stabilized in plasma, and traffics successfully to the cytosol of cultured Saos-2 and SK-HEP-1 cells, achieving cytosolic concentrations greater than 100 nM. This value is 3–10-fold higher than the concentration of endogenous AS (11 ± 1 to 44 ± 5 nM). When injected into healthy C57BL/6 mice, ZF-AS reaches the mouse liver to establish concentrations almost 200 nM above baseline. These studies demonstrate that ZF5.3 can deliver a complex enzyme to the cytosol at therapeutically relevant concentrations and support its application as an improved delivery vehicle for therapeutic proteins that function in the cytosol, including enzyme replacement therapies., The miniature protein ZF5.3 delivers argininosuccinate synthetase to the cytosol of hepatocytes and the livers of healthy mice, establishing its potential for cytosolic enzyme replacement therapies.

Social influence is described as a communications theory which involves three psychological processes: compliance, identification and internalization. The theory applies supplemental data from social psychology frameworks consisting of normative conformity and informative conformity research. Individuals can be influenced to demonstrate social behaviors and many of us can be influenced to change our behaviors depending on our psychological or social needs at that time. Propaganda is another social influence that affects crisis communications. These topics will be discussed to better understand social influence and social behaviors while also analyzing legitimate authority interpositions during the coronavirus global crisis. Further, this research task will examine observational research data, compare antecedent events that prompted these discussions, to support a theoretic approach that consists of organizing ontological perspectives to adduce an analysis of understanding intervals of interspersed social influences resulting in the propagation of public policy, particularly, police reform.

Despite the recognition of floodplain importance in the scientific community, floodplains are not afforded the same legal protection as river channels. In the United States alone, flood-related economic losses were much higher in the second half of the 20th century than the first half despite the expenditure of billions of dollars on flood defenses. Partially to blame are the low appraisal and understanding of human impacts to floodplain functions. Here, we explore the impacts of levees on floodplain functions and analyze case studies of floodplain restoration through levee removal. Floodplain functions include (1) fluxes of water, solutes, and particulate materials; (2) enhanced spatial heterogeneity of hydrology and biogeochemistry; (3) enhanced habitat abundance and diversity; (4) enhanced biomass and biodiversity; and (5) hazard mitigation. Case studies of floodplain restoration involving artificial levee adjustment are heavily concentrated in North America, Europe, and Japan, and those case studies assess floodplain functions within 30 years of restoration. In the United States, restoration through levee removal comprises less than 1% of artificial levee length and 1-2% of disconnected floodplains. In Europe, restoration effectiveness was severely limited by upstream flow regulation. Most case studies were impacted by stressors outside the study site and took place in lowland alluvial rivers. Reconfiguration was successful at achieving limited aims while reconnection set floodplains on a trajectory to more fully restore floodplain functions. Case studies illustrated the tension between restoration scale and study resolution in time and space as well as the role of site-specific characteristics in determining restoration outcomes. Numerous knowledge gaps surrounding the integrative relationships between floodplain functions must be addressed in future studies. The ubiquity of flow regulation demands that future floodplain restoration occur in a whole-of-basin manner. Monitoring of restoration must take place for longer periods of time and include multiple functions.

This research task examines crisis communication strategies of police organizations acting as spokespersons, subsequent negative public perceptions of police organizations, while attempting to understand how external and some internal variables, particularly, human processes of emotions and behaviors of police officers, victims of violence and minority groups, might contribute to negative crisis outcomes, as described in the regenerative crisis model, as the crisis becomes more prevalent and persuasive through media framing.

The use of protein sequence to inform enzymology in terms of structure, mechanism, and function has burgeoned over the past two decades. Referred to as genomic enzymology, the utilization of bioinformatic tools such as sequence similarity networks and phylogenetic analyses has allowed the identification of new substrates and metabolites, novel pathways, and unexpected reaction mechanisms. The holistic examination of superfamilies can yield insight into the origins and paths of evolution of enzymes and the range of their substrates and mechanisms. Herein, we highlight advances in the use of genomic enzymology to address problems which the in-depth analyses of a single enzyme alone could not enable.

We perform the first simultaneous Bayesian parameter inference and optimal reconstruction of the gravitational lensing of the cosmic microwave background (CMB), using 100 deg2 of polarization observations from the SPTpol receiver on the South Pole Telescope. These data reach noise levels as low as 5.8 μK arcmin in polarization, which are low enough that the typically used quadratic estimator (QE) technique for analyzing CMB lensing is significantly suboptimal. Conversely, the Bayesian procedure extracts all lensing information from the data and is optimal at any noise level. We infer the amplitude of the gravitational lensing potential to be A ϕ = 0.949 ± 0.122 using the Bayesian pipeline, consistent with our QE pipeline result, but with 17% smaller error bars. The Bayesian analysis also provides a simple way to account for systematic uncertainties, performing a similar job as frequentist “bias hardening” or linear bias correction, and reducing the systematic uncertainty on A ϕ due to polarization calibration from almost half of the statistical error to effectively zero. Finally, we jointly constrain A ϕ along with A L, the amplitude of lensing-like effects on the CMB power spectra, demonstrating that the Bayesian method can be used to easily infer parameters both from an optimal lensing reconstruction and from the delensed CMB, while exactly accounting for the correlation between the two. These results demonstrate the feasibility of the Bayesian approach on real data, and pave the way for future analysis of deep CMB polarization measurements with SPT-3G, Simons Observatory, and CMB-S4, where improvements relative to the QE can reach 1.5 times tighter constraints on A ϕ and seven times lower effective lensing reconstruction noise.

The lack of watershed-scale estimates of floodplain carbon stocks limits recognition of the important role of floodplains and river corridor restoration in efforts to enhance carbon sequestration. We use the South Platte River watershed of Colorado, USA as a case study to illustrate spatial patterns of, and controls on, floodplain carbon stocks at the watershed scale. This case study illustrates the disproportionate importance of floodplains for soil carbon stocks relative to adjacent uplands and provides an example of how spatially explicit data can be used to prioritize floodplain restoration with regard to carbon sequestration. We use the hydrogeomorphic floodplain tool GFPLAIN to delineate the extent of 100-year floodplains in the South Platte River watershed. We distinguish elevation bands for the steppe, montane, subalpine, and alpine zones. We also differentiate bead (floodplain width/channel width ≥ 5) and string (floodplain width/channel width5) reaches within the montane and subalpine zones. Drawing on prior, field-based measurements of organic carbon stock in downed, dead wood and soil in these floodplain types, we estimate total floodplain organic carbon stock based on median values of stock in different floodplain types and the spatial extent of these floodplain types. This estimate includes organic carbon stocks in lake and reservoir sediments in the watershed. Soil constitutes the greatest reservoir of floodplain carbon. The total estimated area of floodplain is 2916 km
