Obesity represents the single greatest ongoing roadblock to improving cardiovascular health. Prolonged obesity is associated with fundamental changes in the integrative control of energy balance, including the development of selective leptin resistance, which is thought to contribute to obesity-associated hypertension, and adaptation of resting metabolic rate (RMR) when excess weight is reduced. Leptin and the melanocortin system within the hypothalamus contribute to the control of both energy balance and blood pressure. While the development of drugs to stimulate RMR and thereby reverse obesity through activation of the melanocortin system has been pursued, most of the resulting compounds simultaneously cause hypertension. Evidence supports the concept that although feeding behaviors, RMR, and blood pressure are controlled through mechanisms that utilize similar molecular mediators, these mechanisms exist in anatomically dissociable networks. New evidence supports a major change in molecular signaling within AgRP (Agouti-related peptide) neurons of the arcuate nucleus of the hypothalamus during prolonged obesity and the existence of multiple distinct subtypes of AgRP neurons that individually contribute to control of feeding, RMR, or blood pressure. Finally, ongoing work by our laboratory and others support a unique role for AT 1 (angiotensin II type 1 receptor) within one specific subtype of AgRP neuron for the control of RMR. We propose that understanding the unique biology of the AT 1 -expressing, RMR-controlling subtype of AgRP neurons will help to resolve the selective dysfunctions in RMR control that develop during prolonged obesity and potentially point toward novel druggable antiobesity targets that will not simultaneously cause hypertension.

Resting metabolic rate (RMR) adaptation contributes to the maintenance of human obesity, yet mechanisms controlling RMR in lean vs obese states remain poorly defined. Leptin contributes to RMR control in part through suppression of inhibitory Agouti-related peptide (AgRP) neurotransmission in the hypothalamic arcuate nucleus (ARC). Within AgRP neurons, leptin activates the extracellular signal-regulated kinase (ERK) cascade, which is thought to interrupt Agrp transcription through interference with the Krüppel-like factor 4 (KLF4) transcription factor, and our published data support altered ARC ERK signaling during obesity. We therefore hypothesized a role for ERK/KLF4 signaling within AgRP neurons for the suppression of AgRP transcription and thereby stimulation of RMR in the lean state, and changes in this pathway during obesity. In the GT1-7 cell model of AgRP neurons, leptin increased the phosphorylated ERK (pERK)/ERK ratio (n=4, 1 uM, 15 min +89% pAgrp mRNA (n=4, 1 uM, 1 hr -43.0% pKlf4 reduced Agrp mRNA (n=4, -21.6% pAgrp -Cre x Klf4 flox ) exhibited increased RMR (n=13 KO +13.48% vs controls n=22 pAgrp and RMR in the lean state. Ten weeks of high fat feeding strongly increases endogenous plasma leptin in C57BL/6J mice, and this treatment increased the ARC pERK/ERK ratio (n=5 +54.4% pAgrp mRNA was not suppressed (n=4 +1.6-fold, p=ns). Therefore, these data also support the concept that loss of ERK-mediated suppression of KLF4 within the AgRP neuron during obesity may contribute to the pathogenesis of RMR adaptation.

Local paracrine versions of the renin-angiotensin (ANG) system within the brain have been implicated in aspects of cardiovascular and metabolic control, including fluid and electrolyte homeostasis, blood pressure, and energy expenditure, and these effects are often mediated via the ANG type 1 (AT1) receptor. Previous radiographic studies of human brain support ligand binding to AT1 within the hypothalamic arcuate nucleus (ARC), but its localization to individual cell types remains unresolved. Studies of rodent brain demonstrate expression of the rodent AT1 homolog AT1A within the murine ARC almost exclusively within a subset of cells that also express Agouti-related peptide (AgRP) and neuropeptide-Y (NPY). Rodent studies also demonstrate a major role for AT1A within AgRP/NPY neurons in integrative control of energy expenditure, autonomic function, and possibly blood pressure. Thus, we hypothesized that AgRP/NPY neurons of the human ARC would express AT1. ARC sections were obtained after autopsy from a 74 year old male subject diagnosed with Amyotrophic Lateral Sclerosis, body mass index of 34, with self-reported occasional use of over the counter fluticasone propionate, fexofenadine, and topical hydrocortisone. Formalin-fixed, paraffin-embedded ARC was sectioned at 4 microns. RNAscope in situ hybridization methods were used to localize mRNA for AT1 (ACDBio catalog 602841-C2) and AgRP (557451), and nuclei were visualized using DAPI. A similar 4 micron section, obtained within 12 microns of the first section, was analyzed as a negative control (AT1 control: 310043-C2, AgRP control: 310043). As expected, robust expression of AgRP was observed within the ARC (51/106 cells), but not in regions lateral to the ARC (4/114 cells). AT1 expression was observed in cells both within (49/106 cells) and outside the ARC (26/114 cells). Within the ARC, AT1 expression was detected in approximately 40% of AgRP neurons (double-positive n=21, AgRP-only n=30, AT1-only n=28, total cells n=106 in a 95,000 μm 2 region). Only a very small number of signal artifacts (AgRP 0.13; AT1 0.01 % total area) were detected in the negative control. These findings indicate that within the human ARC, and similar to rodents, AT1 is expressed within a unique subset of AgRP/NPY neurons.

The target sits at the core of each facility for the production of radioactive ion beams for nuclear physics research. The materials which constitutes it must withstand extremes conditions of temperature and pressure, being bombarded by intense beams of light particles for several days. This is particularly important in high-power facilities such as SPES (Selective Production of Exotic Species) in which the targets are subjected to strong thermal gradients. Accurate choice and design of materials are therefore crucial. On recent years, several studies have been carried out in the framework of the SPES target research and development. An overview of recently exploited production techniques and a selection of synthesized materials are presented.

Prolonged obesity is associated with blunted feeding and thermogenic autonomic responses to leptin, but cardiovascular responses to leptin are maintained. This state of selective leptin resistance is, therefore, proposed to contribute to the pathogenesis and maintenance of obesity-associated hypertension. Cells of the arcuate nucleus of the hypothalamus detect leptin, and although the cellular and molecular mechanisms remain unclear, altered arcuate nucleus biology is hypothesized to contribute to selective leptin resistance. Male C57BL/6J mice were fed a high-fat diet (HFD) or chow from 8 to 18 weeks of age, as this paradigm models selective leptin resistance. Nuclei were then isolated from arcuate nucleus for single-nucleus RNA sequencing. HFD caused expected gains in adiposity and circulating leptin. Twenty-three unique cell-type clusters were identified, and Ingenuity Pathway Analysis was used to explore changes in gene expression patterns due to chronic HFD within each cluster. Notably, gene expression signatures related to leptin signaling exhibited suppression predominantly in neurons identified as the Agouti-related peptide ( Agrp ) subtype. Ingenuity Pathway Analysis results were also consistent with alterations in CREB (cAMP response element-binding protein) signaling in Agrp neurons after HFD, and reduced phosphorylated CREB was confirmed in arcuate nucleus after prolonged HFD by capillary electrophoresis-based Western blotting. These findings support the concept that prolonged HFD-induced obesity is associated with selective changes in Agrp neuron biology, possibly secondary to altered CREB signaling.

We use dust masses ($M_{dust}$) derived from far-infrared data and molecular gas masses ($M_{mol}$) based on CO luminosity, to calibrate proxies based on a combination of the galaxy Balmer decrement, disk inclination and gas metallicity. We use such proxies to estimate $M_{dust}$ and $M_{mol}$ in the local SDSS sample of star-forming galaxies (SFGs). We study the distribution of $M_{dust}$ and $M_{mol}$ along and across the Main Sequence (MS) of SFGs. We find that $M_{dust}$ and $M_{mol}$ increase rapidly along the MS with increasing stellar mass ($M_*$), and more marginally across the MS with increasing SFR (or distance from the relation). The dependence on $M_*$ is sub-linear for both $M_{dust}$ and $M_{mol}$. Thus, the fraction of dust ($f_{dust}$) and molecular gas mass ($f_{mol}$) decreases monotonically towards large $M_*$. The star formation efficiency (SFE, the inverse of the molecular gas depletion time) depends strongly on the distance from the MS and it is constant along the MS. As nearly all galaxies in the sample are central galaxies, we estimate the dependence of $f_{dust}$ and $f_{gas}$ on the host halo mass and find a tight anti-correlation. As the region where the MS is bending is numerically dominated by massive halos, we conclude that the bending of the MS is due to lower availability of molecular gas mass in massive halos rather than a lower efficiency in forming stars., Accepted for publication in MNRAS, 12 pages, 9 figures

We report the serendipitous discovery of a bright galaxy (Gal-A) observed as part of the ALMA Large Program to INvestigate [CII] at Early times (ALPINE). While this galaxy is detected both in line and continuum emission in ALMA Band 7, it is completely dark in UV/optical filters and only presents a marginal detection in the UltraVISTA Ks band. We discuss the nature of the observed ALMA line, i.e. whether the emission comes from [CII] at z~4.6, or from high-J CO transitions at z~2.2. In the first case we find a [CII]-to-FIR luminosity ratio of log(L_[CII]/L_FIR)=-2.5, consistent with the average value for local star-forming galaxies (SFGs); in the second case, instead, the source would lie outside of the empirical relations between L_CO and L_FIR found in the literature. At both redshifts, we derive the star-formation rate (SFR) from the ALMA continuum, and the stellar mass (M*) by using stellar population synthesis models as input for LePHARE spectral energy distribution (SED) fitting. Exploiting our results, we believe that Gal-A is a Main-Sequence (MS), dusty SFG at z=4.6 (i.e. [CII] emitter) with log(SFR/[M/yr])~1.4 and log(M*/M)~9.7. This work underlines the crucial role of the ALPINE survey in making a census of this class of objects, in order to unveil their contribution to the global star-formation rate density (SFRD) of the Universe at the end of the Reionisation epoch., Comment: 13 pages, 6 figures. Angiotensin II (ANG II) Agtr1a receptor (AT1A) is expressed in cells of the arcuate nucleus of the hypothalamus that express the leptin receptor ( Lepr) and agouti-related peptide ( Agrp). Agtr1a expression in these cells is required to stimulate resting energy expenditure in response to leptin and high-fat diets (HFDs), but the mechanism activating AT1Asignaling by leptin remains unclear. To probe the role of local paracrine/autocrine ANG II generation and signaling in this mechanism, we bred mice harboring a conditional allele for angiotensinogen ( Agt, encoding AGT) with mice expressing Cre-recombinase via the Lepr or Agrp promoters to cause cell-specific deletions of Agt ( AgtLepr-KOand AgtAgrp-KOmice, respectively). AgtLepr-KOmice were phenotypically normal, arguing against a paracrine/autocrine AGT signaling mechanism for metabolic control. In contrast, AgtAgrp-KOmice exhibited reduced preweaning survival, and surviving adults exhibited altered renal structure and steroid flux, paralleling previous reports of animals with whole body Agt deficiency or Agt disruption in albumin ( Alb)-expressing cells (thought to cause liver-specific disruption). Surprisingly, adult AgtAgrp-KOmice exhibited normal circulating AGT protein and hepatic Agt mRNA expression but reduced Agt mRNA expression in adrenal glands. Reanalysis of RNA-sequencing data sets describing transcriptomes of normal adrenal glands suggests that Agrp and Alb are both expressed in this tissue, and fluorescent reporter gene expression confirms Cre activity in adrenal gland of both Agrp-Cre and Alb-Cre mice. These findings lead to the iconoclastic conclusion that extrahepatic (i.e., adrenal) expression of Agt is critically required for normal renal development and survival.

The brain renin-angiotensin system (RAS) is implicated in control of blood pressure (BP), fluid intake, and energy expenditure (EE). Angiotensin II (ANG II) within the arcuate nucleus of the hypothalamus contributes to control of resting metabolic rate (RMR) and thereby EE through its actions on Agouti-related peptide (AgRP) neurons, which also contribute to EE control by leptin. First, we determined that although leptin stimulates EE in control littermates, mice with transgenic activation of the brain RAS (sRA) exhibit increased EE and leptin has no additive effect to exaggerate EE in these mice. These findings led us to hypothesize that leptin and ANG II in the brain stimulate EE through a shared mechanism. Because AgRP signaling to the melanocortin MC4R receptor contributes to the metabolic effects of leptin, we performed a series of studies examining RMR, fluid intake, and BP responses to ANG II in mice rendered deficient for expression of MC4R via a transcriptional block ( Mc4r-TB). These mice were resistant to stimulation of RMR in response to activation of the endogenous brain RAS via chronic deoxycorticosterone acetate (DOCA)-salt treatment, whereas fluid and electrolyte effects remained intact. These mice were also resistant to stimulation of RMR via acute intracerebroventricular (ICV) injection of ANG II, whereas BP responses to ICV ANG II remained intact. Collectively, these data demonstrate that the effects of ANG II within the brain to control RMR and EE are dependent on MC4R signaling, whereas fluid homeostasis and BP responses are independent of MC4R signaling.

The [CII] 158$~\mu$m emission line represents so far one of the most profitable tools for the investigation of the high-redshift galaxies in the early Universe. Being one of the brightest cooling lines in the rest-frame far-infrared regime of star-forming galaxies, it has been successfully exploited as a tracer of star-formation rate (SFR) in local sources. The picture is more complex at higher redshifts, where its usability in this context is still under investigation. Recent results from the ALMA Large Program to INvestigate [CII] at Early times (ALPINE) survey suggest that there is no (or weak) evolution of the L$\mathrm{_{[CII]}}$-SFR relation up to $z\sim6$ but their reliability is hampered by the presence of a large population of [CII] non-detected galaxies. In this work, we characterize the population of [CII] non-detections in ALPINE. By stacking their ALMA spectra, we obtain a signal detected at $\sim5.1\sigma$, resulting in a [CII] luminosity of $\mathrm{log(L_\mathrm{[CII]}}/\mathrm{L_{\odot}})$ $\sim7.8$. When combining this value with those from the [CII] detections, we find a $\mathrm{L_{[CII]}}$-SFR relation with a slope $b=1.14\pm0.11$, in agreement within the uncertainties both with the linear relation found in the local Universe, and with the previous findings from ALPINE at $z\sim5$. This suggests that the [CII] line can be considered a good tracer of star formation up to the distant Universe. Finally, we show that the galaxies of our sample that most deviate from the observed L$_\mathrm{[CII]}$-SFR relation could suffer from a less precise redshift estimation, perhaps artificially reducing their [CII] luminosity. We use a sample of 706 galaxies, selected as [O ii]λ3727 ([O ii]) emitters in the Survey for High-z Absorption Red and Dead Sources (SHARDS) on the CANDELS/GOODS-N field, to study the differential attenuation of the nebular emission with respect to the stellar continuum. The sample includes only galaxies with a counterpart in the infrared and log10(M∗/M⊙) > 9, over the redshift interval 0.3 ≲ z ≲ 1.5. Our methodology consists in the comparison of the star formation rates inferred from [O ii] and Hα emission lines with a robust quantification of the total star-forming activity (SFR TOT) that is independently estimated based on both infrared and ultraviolet (UV) luminosities. We obtain $f\, =\, E(B-V)_{\mathrm{stellar}}$/E(B - V)nebular = 0.69$^{0.71}_{0.69}$ and 0.55$^{0.56}_{0.53}$ for [O ii] and Hα, respectively. Our resulting f-factors display a significant positive correlation with the UV attenuation and shallower or not-significant trends with the stellar mass, the SFRTOT, the distance to the main sequence, and the redshift. Adaptation of resting metabolic rate (RMR) contributes to the maintenance of human obesity, yet the control of RMR - and how this system changes during obesity - remains poorly defined. Leptin normally suppresses Agouti-related peptide (AgRP) expression in the arcuate nucleus of the hypothalamus (ARC) to disinhibit melanocortin-mediated stimulation of RMR. Leptin stimulates ERK signaling in the ARC (a known suppressor of AgRP), and the inhibition of ARC ERK attenuates thermogenic sympathetic nervous activity responses to leptin. Diet-induced obesity (DIO) increases leptin, but paradoxically exaggerates ARC AgRP expression and causes RMR adaptation. Thus, we hypothesize that leptin increases ERK signaling in AgRP cells, to reduce AgRP and thereby ultimately increase RMR, but that this signaling mechanism becomes dysfunctional during DIO. The Isotope Separation On-Line (ISOL) technique is today established as one of the primary methods to produce high-intensity and high-quality radioactive beams. This technique produces, for a given amount of the desired isotope, many orders of magnitude of other radioactive species. Due to the activation generated by interactions of the primary beam, intense neutron fields, and deposition of the produced radioactive ions inside beam line elements, an ISOL facility in operation becomes an intense radioactive source. Therefore, the biological hazard imposes severe radiological safety challenges to the design, operation, maintenance, and final decommissioning of such facilities. A challenging component is the ion source complex, where the ion extraction electrode provides the extraction of radioactive ions from the ion source and the first acceleration to the extracted beam. The radioactive contamination of this sub-component is studied, by means of the FLUKA code, in the case of the Selective Production of Exotic Species facility, which is in the advanced construction phase at Istituto Nazionale di Fisica Nucleare, Laboratori Nazionali di Legnaro, Padua, Italy. The developed model includes isotope production by the interactions of a 40 MeV energy proton beam on a 238UCx target, selection of radioactive isotopes that are able to stick on the electrode tip, time evolution of the deposited isotopes during the operation and cooling periods before maintenance interventions, and evaluation of the ambient dose equivalent rate generated by the contamination of the electrode tip. Based on these results, the possibility of manual interventions for maintenance and emergency vs the use of remote handling systems is discussed.

The Selective Production of Exotic Species project is under construction at Laboratori Nazionali di Legnaro-INFN. The aim of the collaboration is to produce highly pure Radioactive Ion Beams (RIBs) from fission fragments of a uranium carbide (UC x) target activated by a cyclotron proton beam. In order to select a specific atomic species, the main tool to be applied is the resonant laser ionization technique. We have just completed the installation of a dedicated all solid state laser system whose elements are tunable to transitions of all the elements/isotopes of interest for the project. The new laser system is based on three Titanium:sapphire laser sources, independently pumped by three Nd:YLF pump lasers, and it can be coupled to two high harmonic generation (second harmonic generation, third harmonic generation, and fourth harmonic generation) setups. The power, wavelength, and position of the laser beams are continuously monitored and stabilized by using automated active systems to improve the beam production stability of RIBs. This paper presents the main features of the laser system and examples of application of a laser ion source, including a first demonstration of photoionization of stable silver, one of the most requested elements for RIB application.

In this paper we present an attempt to estimate the redshift evolution of the molecular to neutral gas mass ratio within galaxies (at fixed stellar mass). For a sample of five nearby grand design spirals located on the Main Sequence (MS) of star forming galaxies, we exploit maps at 500 pc resolution of stellar mass and star formation rate ($M_{\star}$ and SFR). For the same cells, we also have estimates of the neutral ($M_{\rm HI}$) and molecular ($M_{\rm H_2}$) gas masses. To compute the redshift evolution we exploit two relations: {\it i)} one between the molecular-to-neutral mass ratio and the total gas mass ($M_{\rm gas}$), whose scatter shows a strong dependence with the distance from the spatially resolved MS, and {\it ii)} the one between $\log(M_{\rm{H_2}}/M_{\star})$ and $\log(M_{\rm{HI}}/M_{\star})$. For both methods, we find that $M_{\rm H_2}$/$M_{\rm HI}$ within the optical radius slightly decreases with redshift, contrary to common expectations of galaxies becoming progressively more dominated by molecular hydrogen at high redshifts. Galaxy mergers are thought to be one of the main mechanisms of the mass assembly of galaxies. Recently, many works have suggested a possible increase in the fraction of major mergers in the early Universe, reviving the debate on which processes (e.g., cold accretion, star formation, mergers) most contribute to the mass build-up of galaxies through cosmic time. To estimate the importance of major mergers in this context, we make use of the new data collected by the ALMA Large Program to INvestigate [CII] at Early times (ALPINE), which observed the [CII] 158 $��$m emission line from a sample of 75 main-sequence star-forming galaxies at 4.4 < z < 5.9. We used, for the first time, the morpho-kinematic information provided by the [CII] emission to obtain the fraction of major mergers ($f_{MM}$) at z~5. By adopting different prescriptions for the merger timescales ($T_{MM}$), we converted this fraction into the merger rate per galaxy ($R_{MM}$) and per volume ($��_{MM}$). We then combined our results with those at lower redshifts from the literature, computing the cosmic evolution of the merger fraction. This is described by a rapid increase from z~0 to higher redshifts, a peak at z~3, and a slow decrease towards earlier epochs. Depending on the timescale prescription used, this fraction translates into a merger rate ranging between ~0.1 and ~4.0 Gyr$^{-1}$ at z~5. Finally, we compare the specific star formation and star-formation rate density with the analogous quantities from major mergers. Our new ALPINE data reveal the presence of a significant merging activity in the early Universe. However, whether this population of mergers can provide a relevant contribution to the galaxy mass assembly at these redshifts and through the cosmic epochs is strongly dependent on the assumption of the merger timescale., Accepted for publication in section 4. Extragalactic astronomy of Astronomy and Astrophysics

While the kinematics of galaxies up to z~3 have been characterized in detail, only a handful of galaxies at high redshift (z>4) have been examined in such a way. The Atacama Large Millimeter/submillimeter Array (ALMA) Large Program to INvestigate [CII] at Early times (ALPINE) survey observed a statistically significant sample of 118 star-forming main sequence galaxies at z=4.4-5.9 in [CII]158um emission, increasing the number of such observations by nearly 10x. A preliminary qualitative classification of these sources revealed a diversity of kinematic types (i.e., rotators, mergers, and dispersion-dominated systems). In this work, we supplement the initial classification by applying quantitative analyses to the ALPINE data: a tilted ring model (TRM) fitting code (3DBarolo), a morphological classification (Gini-M20), and a set of disk identification criteria. Of the 75 [CII]-detected ALPINE galaxies, 29 are detected at sufficient significance and spatial resolution to allow for TRM fitting and the derivation of morphological and kinematic parameters. These 29 sources constitute a high-mass subset of the ALPINE sample (M_*>10^9.5Msol). We robustly classify 14 of these sources (six rotators, five mergers, and three dispersion-dominated systems); the remaining sources showing complex behaviour. By exploring the G-M20 of z>4 rest-frame FIR and [CII] data for the first time, we find that our 1"~6kpc resolution data alone are insufficient to separate galaxy types. We compare the rotation curves and dynamical mass profiles of the six ALPINE rotators to the two previously detected z~4-6 unlensed main sequence rotators, finding high rotational velocities (~50-250km/s) and a diversity of rotation curve shapes., ERC STFC

Future surveys focusing on understanding the nature of dark energy (e.g., Euclid and WFIRST) will cover large fractions of the extragalactic sky in near-IR slitless spectroscopy. These surveys will detect a large number of galaxies that will have only one emission line in the covered spectral range. In order to maximize the scientific return of these missions, it is imperative that single emission lines are correctly identified. Using a supervised machine-learning approach, we classified a sample of single emission lines extracted from the WFC3 IR Spectroscopic Parallel survey, one of the closest existing analogs to future slitless surveys. Our automatic software integrates a spectral energy distribution (SED)-fitting strategy with additional independent sources of information. We calibrated it and tested it on a "gold" sample of securely identified objects with multiple lines detected. The algorithm correctly classifies real emission lines with an accuracy of 82.6%, whereas the accuracy of the SED-fitting technique alone is low (∼50%) due to the limited amount of photometric data available (≤6 bands). While not specifically designed for the Euclid and WFIRST surveys, the algorithm represents an important precursor of similar algorithms to be used in these future missions.

Perception of weight loss requirements before bariatric surgery varies among patients, physicians, and health insurance payers. Current clinical guidelines do not require preoperative weight loss because of a lack of scientific support regarding its benefits.To examine the association of preoperative body mass index (BMI) and weight loss with 30-day mortality after bariatric surgery.This cohort study used data from 480 075 patients who underwent bariatric surgery from 2015 to 2017 in the Metabolic and Bariatric Surgery Accreditation and Quality Improvement Program, which covers more than 90% of all bariatric surgery programs in the United States and Canada. Clinical and demographic data were collected at all participating institutions using a standardized protocol. Data analysis was performed from December 2018 to November 2019.Preoperative BMI and weight loss.30-day mortality after bariatric surgery.Of the 480 075 patients (mean [SD] age 45.1 [12.0] years; 383 265 [79.8%] women), 511 deaths (0.1%) occurred within 30 days of bariatric surgery. Compared with patients with a preoperative BMI of 35.0 to 39.9, the multivariable-adjusted odds ratios for 30-day mortality for patients with preoperative BMI of 40.0 to 44.9, 45.0 to 49.9, 50.0 to 54.9, and 55.0 and greater were 1.37 (95% CI, 1.02-1.83), 2.19 (95% CI, 1.64-2.92), 2.61 (95% CI, 1.90-3.58), and 5.03 (95% CI, 3.78-6.68), respectively (P for trend .001). Moreover, compared with no preoperative weight loss, the multivariable-adjusted odds ratios for 30-day mortality for patients with weight loss of more than 0% to less than 5.0%, 5.0% to 9.9%, and 10.0% and greater were 0.76 (95% CI, 0.60-0.96), 0.69 (95% CI, 0.53-0.90), and 0.58 (95% CI, 0.41-0.82), respectively (P for trend = .003).In this study, even moderate weight loss (ie,0% to5%) before bariatric surgery was associated with a lower risk of 30-day mortality. These findings may help inform future updates of clinical guidelines regarding bariatric surgery.

The identification of an emission line is unambiguous when multiple spectral features are clearly visible in the same spectrum. However, in many cases, only one line is detected, making it difficult to correctly determine the redshift. We developed a freely available unsupervised machine-learning algorithm based on unbiased topology (UMLAUT) that can be used in a very wide variety of contexts, including the identification of single emission lines. To this purpose, the algorithm combines different sources of information, such as the apparent magnitude, size and color of the emitting source, and the equivalent width and wavelength of the detected line. In each specific case, the algorithm automatically identifies the most relevant ones (i.e., those able to minimize the dispersion associated with the output parameter). The outputs can be easily integrated into different algorithms, allowing us to combine supervised and unsupervised techniques and increasing the overall accuracy. We tested our software on WISP (WFC3 IR Spectroscopic Parallel) survey data. WISP represents one of the closest existing analogs to the near-IR spectroscopic surveys that are going to be performed by the future Euclid and Roman missions. These missions will investigate the large-scale structure of the universe by surveying a large portion of the extragalactic sky in near-IR slitless spectroscopy, detecting a relevant fraction of single emission lines. In our tests, UMLAUT correctly identifies real lines in 83.2% of the cases. The accuracy is slightly higher (84.4%) when combining our unsupervised approach with a supervised approach we previously developed.

The Lya line in the UV and the [CII] line in the FIR are widely used tools to identify galaxies and to obtain insights into ISM properties in the early Universe. By combining data obtained with ALMA in band 7 at ~ 320 GHz as part of the ALMA Large Program to INvestigate [CII] at Early Times (ALPINE) with spectroscopic data from DEIMOS at Keck, VIMOS and FORS2 at the VLT, we assembled a unique sample of 53 main-sequence star-forming galaxies at 4.4 < z < 6 in which we detect both the Lya line and the [CII]. We used [CII], observed with ALMA, as a tracer of the systemic velocity of the galaxies, and we find that 90% of the selected objects have Lya-[CII] velocity offsets in the range 0 < Dv_Lya-[CII] < 400 km/s, in line with the few measurements available so far in the early Universe, and significantly smaller than those observed at lower z. We observe ISM-[CII] offsets in the range -500 < Dv_ISM-[CII] < 0 km/s, in line with values at all redshifts. We find significant anticorrelations between Dv_Lya-[CII] and the Lya rest-frame equivalent width EW0(Lya) (or equivalently, the Lya escape fraction f_esc(Lya)). According to available models for the radiative transfer of Lya photons, the escape of Lya photons would be favored in galaxies with high outflow velocities, in agreement with our observations. The uniform shell model would also predict that the Lya escape in galaxies with slow outflows (0 < v_out < 300 km/s) is mainly determined by the neutral hydrogen column density (NHI), while the alternative model by Steidel+10 would favor a combination of NHI and covering fraction as driver of the Lya escape. We suggest that the observed increase in Lya escape that is observed between z~2 and z~6 is not due to a higher incidence of fast outflows at high redshift, but rather to a decrease in average NHI along the line of sight, or alternatively, a decrease in HI covering fraction. The ALMA-ALPINE [CII] survey is aimed at characterizing the properties of a sample of normal star-forming galaxies (SFGs). The ALMA Large Program to INvestigate (ALPINE) features 118 galaxies observed in the [CII]-158$\mu$m line and far infrared (FIR) continuum emission during the period of rapid mass assembly, right after the end of the HI reionization, at redshifts of 45). Based on a visual inspection of the [CII] data cubes together with the large wealth of ancillary data, we find a surprisingly wide range of galaxy types, including 40% that are mergers, 20% extended and dispersion-dominated, 13% compact, and 11% rotating discs, with the remaining 16% too faint to be classified. This diversity indicates that a wide array of physical processes must be at work at this epoch, first and foremost, those of galaxy mergers. We present ALMA observations of a merging system atz ∼ 4.57, observed as a part of the ALMA Large Program to INvestigate [CII] at Early times (ALPINE) survey. Combining ALMA [CII]158 μm and far-infrared continuum data with multi-wavelength ancillary data, we find that the system is composed of two massive (M⋆ ≳ 1010 M⊙) star-forming galaxies experiencing a major merger (stellar mass ratiormass ≳ 0.9) at close spatial (∼13 kpc; projected) and velocity (Δv < 300 km s−1) separations, and two additional faint narrow [CII]-emitting satellites. The overall system belongs to a larger scale protocluster environment and is coincident to one of its overdensity peaks. Additionally, ALMA reveals the presence of [CII] emission arising from a circumgalactic gas structure, extending up to a diameter-scale of ∼30 kpc. Our morpho-spectral decomposition analysis shows that about 50% of the total flux resides between the individual galaxy components, in a metal-enriched gaseous envelope characterised by a disturbed morphology and complex kinematics. Similarly to observations of shock-excited [CII] emitted from tidal tails in local groups, our results can be interpreted as a possible signature of interstellar gas stripped by strong gravitational interactions, with a possible contribution from material ejected by galactic outflows and emission triggered by star formation in small faint satellites. Our findings suggest that mergers could be an efficient mechanism of gas mixing in the circumgalactic medium around high-zgalaxies, and thus play a key role in the galaxy baryon cycle at early epochs.

In the second work of this series, we analyse the connection between the availability of gas and the position of a region with respect to the spatially resolved main sequence (MS) relation. Following the procedure presented in Paper I we obtain 500pc scales estimates of stellar mass and star formation rate surface densities ($\Sigma_{\star}$ and $\Sigma_{\rm{SFR}}$). Our sample consists of five face-on, grand design spiral galaxies located on the MS. Thanks to HI 21cm and $^{12}$CO(2-1) maps, we connect the gas surface densities and gas fractions to the observed star formation properties of each region. We find that the spatially resolved MS ($\sigma=0.23$ dex) is the combination of two relations: the Kennicutt-Schmidt law ($\sigma=0.19$ dex) and the molecular gas MS (MGMS, $\sigma=0.22$ dex); $\Sigma_{\star}$, $\Sigma_{\rm{SFR}}$ and the surface density of the molecular gas, $\Sigma_{\rm{H_2}}$, define a 3D relation as proposed by \citet{2019ApJ...884L..33L}. We find that $\Sigma_{\rm{H_2}}$ steadily increases along the MS relation, varies little towards higher $\Sigma_{\rm{SFR}}$ at fixed stellar surface densities (not enough to sustain the change in SFR), and it is almost constant perpendicular to the relation. The surface density of neutral gas ($\Sigma_{\rm{HI}}$) is constant along the MS, and increases in its upper envelop. $\Sigma_{\rm{SFR}}$ can be expressed as a function of $\Sigma_{\star}$ and $\Sigma_{\rm{HI}}$, following the Equation: $\log\Sigma_{\rm{SFR}}$ = 0.97$\log\Sigma_{\star}$ + 1.99$\log\Sigma_{\rm{HI}}$ - 11.11. Finally, we show that f$_{\rm{gas}}$ increases significantly towards the starburst region in the $\log\Sigma_{\star}$ - $\log\Sigma_{\rm{SFR}}$ plane, accompanied by a slight increase in SFE., Comment: Accepted for publications on MNRAS

We analyse the spatially resolved relation between stellar mass (M$_{\star}$) and star formation rate (SFR) in disk galaxies (i.e. the Main Sequence, MS). The studied sample includes eight nearby face-on grand-design spirals, e.g. the descendant of high-redshift, rotationally-supported star-forming galaxies. We exploit photometric information over 23 bands, from the UV to the far-IR, from the publicly available DustPedia database to build spatially resolved maps of stellar mass and star formation rates on sub-galactic scales of 0.5-1.5 kpc, by performing a spectral energy distribution fitting procedure that accounts for both the observed and the obscured star formation processes, over a wide range of internal galaxy environments (bulges, spiral arms, outskirts). With more than 30 thousands physical cells, we have derived a definition of the local spatially resolved MS per unit area for disks, $\log(\Sigma_{SFR})$=0.82log$(\Sigma_{*})$-8.69. This is consistent with the bulk of recent results based on optical IFU, using the H$\alpha$ line emission as a SFR tracer. Our work extends the analysis at lower sensitivities in both M$_{\star}$ and SFR surface densities, up to a factor $\sim$ 10. The self consistency of the MS relation over different spatial scales, from sub-galactic to galactic, as well as with a rescaled correlation obtained for high redshift galaxies, clearly proves its universality., Comment: 21 pages, 15 figures. Selective leptin resistance (SLR) is a pathological state caused by prolonged high fat diet (HFD) feeding in which cardiovascular responses to leptin are preserved, but metabolic effects are attenuated. Thus, SLR is hypothesized to contribute to obesity-associated hypertension, but the mechanisms causing SLR are unclear. Desensitization of leptin signaling in cells of the arcuate nucleus (ARC) of the hypothalamus has been proposed as one of potential mechanisms contributing to SLR. Tanycytes, located along the walls of the third ventricle, play critical roles in the regulation of energy balance through the transport of blood-borne metabolic hormones such as leptin and ghrelin into the cerebrospinal fluid. Previous studies have reported that 10 weeks of 45% HFD (D12451) feeding is sufficient to cause SLR in wildtype C57BL/6J mice. To examine the transcriptomic changes induced by prolonged HFD exposure in tanycytes, we performed single-nucleus RNA-sequencing on nuclei isolated from ARC of ad libitum fed male C57BL/6J mice fed chow diet (7013; CD, n=7) or HFD (n=8) from 8 to 18 weeks of age. At 18 weeks, fat mass (+10.1±1.4 vs +3.2±0.4g, pAgrp ) neurons (z-score -1.6, p=0.009) and unchanged in neurons expressing proopiomelanocortin ( Pomc ) (p=0.2). Collectively, these results lead us to hypothesize that SLR is associated with normal transport of circulating leptin into the arcuate nucleus, while the primary dysfunction that causes SLR instead occurs within selected neuronal populations such as Agrp neurons.

Autoimmune thyroid disease, characterized by the presence of specific anti-thyroid antibodies, is the most common cause of thyroid dysfunction and can lead to either hypo- or hyperthyroidism. In rare cases, thyroid function spontaneously shifts from one extreme to the other. A 32 year old man with an 8 year history of hypothyroidism was referred to our clinic for evaluation of hyperthyroidism. He had been on levothyroxine (LT4) replacement, requiring up to 300 µg qd until about 6 months before coming to our attention, when he sustained a 60 lb weight loss. TSH was suppressed and the LT4 dose was progressively decreased to 75 µg qd. On presentation, the patient was clinically euthyroid. He had no family history of thyroid disease. We use the Sloan Digital Sky Survey (SDSS) database to explore the effect of the disk inclination angle on the derived star formation rate (SFR), hence on the slope and width of the Main Sequence (MS) relation for star-forming galaxies. We find that SFRs for nearly edge-on disks are underestimated by factors ranging from $\sim$ 0.2 dex for low mass galaxies up to $\sim$ 0.4 dex for high mass galaxies. This results in a substantially flatter MS relation for high-inclination disks compared to that for less inclined ones, though the global effect over the whole sample of star-forming galaxies is relatively minor, given the small fraction of high-inclination disks. By using the deepest available mid and far infrared surveys in the CANDELS, GOODS and COSMOS fields we study the evolution of the Main Sequence (MS) of star forming galaxies (SFGs) from z~0 to` ~2.5 at stellar masses larger than 10^{10} M_{\odot}. The MS slope and scatter are consistent with a re-scaled version of the local relation and distribution, shifted at higher values of SFR according to ~(1+z)^{3.2}. The relation exhibits a bending at the high mass end and a slightly increasing scatter as a function of the stellar mass. We show that the previously reported evolution of the MS slope, in the considered mass and redshift range, is due to a selection effect. The distribution of galaxies in the MS region at fixed stellar mass is well represented by a single log-normal distribution at all redshifts and masses, with starburst galaxies (SBs) occupying the tail at high SFR., Comment: Accepted for publication by MNRAS

Based on HST and MUSE data, we probe the stellar and gas properties (i.e. kinematics, stellar mass, star formation rate) of the radio-loud brightest cluster galaxy (BCG) located at the centre of the X-ray luminous cool core cluster Abell 2667 (z = 0.2343). The bi-dimensional modelling of the BCG surface brightness profile reveals the presence of a complex system of substructures extending all around the galaxy. Clumps of different size and shape plunged into a more diffuse component constitute these substructures, whose intense 'blue' optical colour hints to the presence of a young stellar population. Our results depict the BCG as a massive (M_star ~ 1.38 x 10^11 M_sun) dispersion-supported spheroid (v_star < 150 km/s, sigma_0 ~ 216 km/s) hosting an active supermassive black hole (M_SMBH ~ 3.8 x 10^9 M_sun) whose optical features are typical of low ionisation nuclear emission line regions. Although the velocity pattern of the stars in the BCG is irregular, the stellar kinematics in the regions of the clumps show a positive velocity of ~ 100 km/s, similarly to the gas component. An analysis of the mechanism giving rise to the observed lines in the clumps through empirical diagnostic diagrams points out that the emission is composite, suggesting the contribution from both star formation and AGN. We conclude our analysis describing how scenarios of both chaotic cold accretion and merging with a gas-rich disc galaxy can efficaciously explain the phenomena the BCG is undergoing., Accepted for publication on the 10th of June 2019 (MNRAS)

We investigate a sample of 152 dusty sources at 1.5 z Herschel-selected, having stellar masses M * > 1010 M ⊙ and SFR (~100–1000 M ⊙ yr−1) elevated (>4×) above the star-forming "main sequence," classifying them as starbursts (SBs). Through a multiwavelength fitting approach (including a dusty torus component), we divided the sample into active SBs (dominated by an active galactic nucleus (AGN) emission, SBs-AGN, ~23% of the sample) and purely star-forming SBs (SBs-SFR). We visually inspected their Hubble Space Telescope/ultraviolet (UV) rest frame maps: SBs-SFR are generally irregular and composite systems; ~50% of SBs-AGN are instead dominated by regular compact morphologies. We then found archival Atacama Large Millimeter/submillimeter Array continuum counterparts for 33 galaxies (12 SBs-AGN and 21 SBs-SFR). For these sources we computed dust masses, and, with standard assumptions, we also guessed total molecular gas masses. SBs turn into gas-rich systems (f gas = M gas/(M gas + M *) 20%–70%), and the gas fractions of the two SB classes are very similar (f gas = 43% ± 4% and f gas = 42% ± 2%). Our results show that SBs are consistent with a mixture of: (1) highly star-forming merging systems (dominating the SBs-SFR) and (2) primordial galaxies, rapidly growing their M * together with their black hole (mainly the more compact SBs-AGN). Feedback effects have not yet reduced their f gas. Indeed, SBs at z = 2, with relatively low bolometric AGN luminosities in the range 1044 L bol(AGN) 46 erg s−1 (compared to bright optical and X-ray quasars), are still relatively far from the epoch when the AGN feedback will quench the SFR in the host and will substantially depress the gas fractions.

By using a set of different SFR indicators, including WISE mid-infrared and Halpha emission, we study the slope of the Main Sequence (MS) of local star forming galaxies at stellar masses larger than 10^{10} M_{\odot}. The slope of the relation strongly depends on the SFR indicator used. In all cases, the local MS shows a bending at high stellar masses with respect to the slope obtained in the low mass regime. While the distribution of galaxies in the upper envelope of the MS is consistent with a log-normal distribution, the lower envelope shows an excess of galaxies, which increases as a function of the stellar mass but varies as a function of the SFR indicator used. The scatter of the best log-normal distribution increases with stellar mass from ~0.3 dex at 10^{10} M_{\odot} to ~0.45 at 10^{11} M_{\odot}. The MS high-mass end is dominated by central galaxies of group sized halos with a red bulge and a disk redder than the lower mass counterparts. We argue that the MS bending in this region is due to two processes: i) the formation of a bulge component as a consequence of the increased merger activity in groups, and ii) the cold gas starvation induced by the hot halo environment, which cuts off the gas inflow onto the disk. Prolactinomas are the most common functioning pituitary adenoma and may arise sporadically or present in the context of genetic syndromes (multiple endocrine neoplasia type 1 [MEN-1], familial isolated pituitary adenomas [FIPA]). Their prevalence and incidence have increased in recent years, due to improved awareness and diagnostic methods, and are estimated at about 50 per 105 and 2 per 105 persons, respectively. Prolactinomas are rare in children and adolescents, and little is known about their epidemiology in the elderly. Prolactinomas display clear sex differences in terms of size and age at diagnosis, with a greater proportion of macroadenomas in men, who are usually diagnosed at an older age compared to women. There may also be an effect of menopause on tumor size. Information about the natural history of prolactinomas comes from older studies conducted in women, but they seem to have limited growth potential in this population. Despite the elevated levels of estrogen, pregnancy does not appear to promote tumor growth in most patients. Prolactinomas may be more aggressive in patients with MEN-1 and FIPA, although recently published data suggest that their clinical behavior in MEN-1 may actually be similar to that of sporadic prolactinomas.

High-resolution multi-wavelength photometry is crucial to explore the spatial distribution of star formation in galaxies and understand how these evolve. To this aim, in this paper we exploit the deep, multi-wavelength Hubble Space Telescope (HST) data available in the central parts of the GOODS fields and study the distribution of star formation activity and mass in galaxies located at different positions with respect to the Main Sequence (MS) of star-forming galaxies. Our sample consists of galaxies with stellar mass $\geq 10^{9.5} M_{\odot}$ in the redshift range 0.2 $ \leq z \leq 1.2$. Exploiting 10-band photometry from the UV to the near-infrared at HST resolution, we derive spatially resolved maps of galaxies properties, such as stellar mass and star formation rate and specific star formation rate, with a resolution of $\sim 0.16$ arcsec. We find that the star formation activity is centrally enhanced in galaxies above the MS and centrally suppressed below the MS, with quiescent galaxies (1 dex below the MS) characterised by the highest suppression. The sSFR in the outer region does not show systematic trends of enhancement or suppression above or below the MS. The distribution of mass in MS galaxies indicates that bulges are growing when galaxies are still on the MS relation. Galaxies below the MS are more bulge-dominated with respect to MS counterparts at fixed stellar mass, while galaxies in the upper envelope are more extended and have S\'ersic indexes that are always smaller than or comparable to their MS counterparts. The suppression of star formation activity in the central region of galaxies below the MS hints at \textit{inside-out} quenching, as star formation is still ongoing in the outer regions., Comment: 17 pages, 12 figures, accepted for publication on A&A

Obesity is a major risk factor for development of hypertension, and evidence supports a role for the suppression of resting metabolic rate (RMR) in the development and maintenance of obesity. Angiotensin II (ANG) AT1A receptors, localized to cells expressing the leptin receptor and Agouti-related peptide (AgRP) within the arcuate nucleus of the hypothalamus (ARC), are involved in transcriptional control of AgRP and γ-aminobutyric acid (GABA) synthetic genes ( Gad65 , Gad67 , Slc32a1 ) within the ARC, as well as the integrative control of thermogenic sympathetic nerve activity (SNA) and RMR responses in vivo to various stimuli including leptin and high fat diet. These data led us to hypothesize that AT1A functions to suppress GABA signaling in AgRP neurons, ultimately to disinhibit SNA and RMR. Treatment of cultured mouse hypothalamic N43/5 cells (which express endogenous Agtr1a , AgRP and Gad67 ) with 100 nM ANG for 1 hour (n=6) resulted in reduced AgRP (0.57-fold of control, p=0.03) and Gad67 (0.72-fold of control, p=0.09) mRNA. Mice with targeted disruption of Slc32a1 (vesicular GABA transporter; VGAT) in AgRP neurons (VGAT AgRP-KO mice) were generated on a mixed background. PURPOSE OF REVIEW: Here we review the current understanding of the functional neuroanatomy of neurons expressing Agouti-related peptide (AgRP) and the angiotensin 1A receptor (AT(1A)) within the arcuate nucleus (ARC) in the control of energy balance. RECENT FINDINGS: The development and maintenance of obesity involves suppression of resting metabolic rate (RMR). RMR control is integrated via AgRP and proopiomelanocortin neurons within the ARC. Their projections to other hypothalamic and extrahypothalamic nuclei contribute to RMR control, though relatively little is known about the contributions of individual projections and the neurotransmitters involved. Recent studies highlight a role for AT(1A), localized to AgRP neurons, but the specific function of AT(1A) within these cells remains unclear. SUMMARY: AT(1A) functions within AgRP neurons to control RMR, but additional work is required to clarify its role within subpopulations of AgRP neurons projecting to distinct second-order nuclei, and the molecular mediators of its signaling within these cells.

We use a newly assembled large sample of 3,545 star-forming galaxies with secure spectroscopic, grism, and photometric redshifts at z=1.5-2.5 to constrain the relationship between UV slope (beta) and dust attenuation (L(IR)/L(UV)=IRX). Our sample benefits from the combination of deep Hubble WFC3/UVIS photometry from the Hubble Deep UV (HDUV) Legacy survey and existing photometric data compiled in the 3D-HST survey, and extends the range of UV luminosity and beta probed in previous UV-selected samples. IRX is measured using stacks of deep Herschel/PACS 100 and 160 micron data, and the results are compared with predictions of the IRX-beta relation for different assumptions of the stellar population model and obscuration curve. We find that z=1.5-2.5 galaxies have an IRX-beta relation that is consistent with the predictions for an SMC extinction curve if we invoke sub-solar metallicity models that are currently favored for high-redshift galaxies, while the commonly assumed starburst attenuation curve over-predicts the IRX at a given beta by a factor of ~3. The IRX of high-mass (M*>10^9.75 Msun) galaxies is a factor of >4 larger than that of low-mass galaxies, lending support for the use of stellar mass as a proxy for attenuation. The commonly observed trend of fainter galaxies having bluer beta may simply reflect bluer intrinsic UV slopes for such galaxies, rather than lower obscurations. The IRX-beta for young/low-mass galaxies implies a dust curve that is steeper than the SMC, suggesting a lower attenuation at a given beta relative to older/more massive galaxies. The lower attenuations and higher ionizing photon output implied by low metallicity stellar population models point to Lyman continuum production efficiencies, xi_ion, that may be elevated by a factor of ~2 relative to the canonical value for L* galaxies, aiding in their ability to keep the universe ionized at z~2. We present the Hubble Deep UV Legacy Survey (HDUV), a 132 orbit imaging program with the WFC3/UVIS camera onboard the Hubble Space Telescope (HST). The HDUV extends and builds on the few previous UV imaging surveys in the two GOODS/CANDELS-Deep fields to provide deep images over a total area of ~100 arcmin2 in the two filters F275W and F336W. Our release also includes all the F275W imaging data taken by the CANDELS survey, which were aligned using a novel approach and combined with the HDUV survey data. By reaching depths of 27.5-28.0 mag (5sigma, in 0.4" apertures), these are the deepest high-resolution UV data over such a large area taken to date. Such unique UV imaging enables a wide range of science by the community. Among the main goals of the HDUV survey are: (1) provide a complete sample of faint star-forming galaxies at z~1-3, (2) constrain the ionizing photon escape fraction from galaxies at z~2-3, and (3) track the build-up of bulges and the disappearance of clumpy disk galaxies through reliable internal stellar population properties at sub-kpc resolution out to z~3. The addition of the HDUV data further enhances the legacy value of the two GOODS/CANDELS-Deep fields, which now include deep 11-band HST imaging as well as very deep ancillary data from X-ray to radio, enabling unique multi-wavelength studies. We present six galaxies at z~2 that show evidence of Lyman continuum (LyC) emission based on the newly acquired UV imaging of the Hubble Deep UV legacy survey (HDUV) conducted with the WFC3/UVIS camera on the Hubble Space Telescope (HST). At the redshift of these sources, the HDUV F275W images partially probe the ionizing continuum. By exploiting the HST multi-wavelength data available in the HDUV/GOODS fields, models of the UV spectral energy distributions, and detailed Monte Carlo simulations of the intergalactic medium absorption, we estimate the absolute ionizing photon escape fractions of these galaxies to be very high -- typically >60% (>13% for all sources at 90% likelihood). Our findings are in broad agreement with previous studies that found only a small fraction of galaxies to show high escape fraction. These six galaxies comprise the largest sample yet of LyC leaking candidates at z~2 whose inferred LyC flux has been cleanly observed at HST resolution. While three of our six candidates show evidence of hosting an active galactic nucleus (AGN), two of these are heavily obscured and their LyC emission appears to originate from star-forming regions rather than the central nucleus. This suggests an AGN-aided pathway for LyC escape from these sources. Extensive multi-wavelength data in the GOODS fields, especially the near-IR grism spectra from the 3D-HST survey, enable us to study the candidates in detail and tentatively test some recently proposed indirect methods to probe LyC leakage -- namely, the [OIII]/[OII] line ratio and the H$��-$UV slope diagram. High-resolution spectroscopic followup of our candidates will help constrain such indirect methods which are our only hope of studying $f_{esc}$ at z~5-9 in the fast-approaching era of the James Webb Space Telescope., 11 pages, 6 figures, submitted to ApJ

Study Objectives Severe sleep restriction results in elevated evening cortisol levels. We examined whether this relative hypercortisolism is associated with alterations in the pituitary-adrenocortical response to evening corticotropin-releasing hormone (CRH) stimulation. Methods Eleven subjects participated in 2 sessions (2 nights of 10 hours vs. 4 hours in bed) in randomized order. Sleep was polygraphically recorded. After the second night of each session, blood was sampled at 20-minute intervals from 09:00 to 24:00 for adrenocorticotropic hormone (ACTH) and cortisol measurements, and perceived stress was assessed hourly. Ovine CRH was injected at 18:00 (1 µg/kg body weight). Results Prior to CRH injection, baseline ACTH, but not cortisol, levels were elevated after sleep restriction. Relative to the well-rested condition, sleep restriction resulted in a 27% decrease in overall ACTH response to CRH (estimated by the incremental area under the curve from 18:00 to 24:00; p = .002) while the cortisol response was decreased by 21% (p = .083). Further, the magnitude of these decreases was correlated with the individual amount of sleep loss (ACTH: rSp = -0.65, p = .032; cortisol: rSp = -0.71, p = .015). The acute post-CRH increment of cortisol was reduced (p = .002) without changes in ACTH reactivity, suggesting decreased adrenal sensitivity. The rate of decline from peak post-injection levels was reduced for cortisol (p = .032), but not for ACTH. Scores of perceived stress were unaffected by CRH injection and were low and similar under both sleep conditions. Conclusions Sleep restriction is associated with a reduction of the overall ACTH and cortisol responses to evening CRH stimulation, and a reduced reactivity and slower recovery of the cortisol response.
