Abstract Background DNA methylation in the form of 5-methylcytosine (5mC) is the most abundant base modification in animals. However, 5mC levels vary widely across taxa. While vertebrate genomes are hypermethylated, in most invertebrates, 5mC concentrates on constantly and highly transcribed genes (gene body methylation; GbM) and, in some species, on transposable elements (TEs), a pattern known as “mosaic”. Yet, the role and developmental dynamics of 5mC and how these explain interspecies differences in DNA methylation patterns remain poorly understood, especially in Spiralia, a large clade of invertebrates comprising nearly half of the animal phyla. Results Here, we generate base-resolution methylomes for three species with distinct genomic features and phylogenetic positions in Annelida, a major spiralian phylum. All possible 5mC patterns occur in annelids, from typical invertebrate intermediate levels in a mosaic distribution to hypermethylation and methylation loss. GbM is common to annelids with 5mC, and methylation differences across species are explained by taxon-specific transcriptional dynamics or the presence of intronic TEs. Notably, the link between GbM and transcription decays during development, alongside a gradual and global, age-dependent demethylation in adult stages. Additionally, reducing 5mC levels with cytidine analogs during early development impairs normal embryogenesis and reactivates TEs in the annelid Owenia fusiformis. Conclusions Our study indicates that global epigenetic erosion during development and aging is an ancestral feature of bilateral animals. However, the tight link between transcription and gene body methylation is likely more important in early embryonic stages, and 5mC-mediated TE silencing probably emerged convergently across animal lineages.

BackgroundVenous thromboembolism (VTE) is a common complication after major orthopedic surgery. The venous foot pump (VFP) is an effective mechanical preventive measure against VTE in patients. However, the differences in effectiveness based on varying usage times of VFP remain unclear.ObjectiveTo explore the effectiveness of VFP with different usage times in preventing VTE in patients undergoing major orthopedic surgery.MethodsNine databases (PubMed, Web of Science, CINAHL, Embase, Cochrane Library, CBM, VIP, CNKI, and Wanfang) were searched to identify randomized controlled trials (RCTs) evaluating VFP interventions for VTE prevention in major orthopedic surgery patients. The risk of bias in each study was assessed using the Cochrane Collaboration tool. Meta-analysis was conducted using RevMan 5.3.ResultsA total of 36 RCTs involving 3,791 patients undergoing major orthopedic surgery were included. ABSTRACTObjectives Diabetic gastrointestinal dysfunction (DGD) is a serious complication of diabetic mellitus (DM), affecting the enteric nervous system (ENS), particular enteric glial cells (EGCs). This study aimed to elucidate the effects and underlying molecular mechanisms of hyperglycemic stress on EGCs in in vitro and in vivo models of DM.Methods In in vitro studies, enteric glial cell line CRL-2690 was exposed to hyperglycemia stress, and cell viability, cell apoptosis and oxidative damage were assessed. In in vivo studies, STZ-induced diabetic mice were constructed, and cell apoptosis and oxidative damage of EGCs in the duodenum of DM mice were assessed.Results The results showed that hyperglycemic stress markedly induced oxidative damage of EGCs in in vitro and in vivo models of DM. This damage was found to be dependent on the activation of redoxosomes, which involved the phosphorylation of SRC and Vav2, the up-regulation of active RAC1-GTP, and the activation of NADPH oxidase (NOX). Moreover, inhibitors of redoxosomes, such as the RAC1 inhibitor NSC23766 and the NOX inhibitor VAS2870, effectively mitigated the hyperglycemic stress-induced oxidative damage of EGCs. Additionally, inhibition of p66SHC, a downstream target of redoxosomes, attenuated oxidative damage of EGCs under hyperglycemic stress.Discussion Our findings suggest that the redoxosomes/p66SHC signaling is involved in the oxidative damage of EGCs during the pathological process of DGD. This signaling cascade may represent a potential therapeutic target for the treatment of DGD.

Abstract Metal halide perovskites (MHPs) are considered as promising candidates in the application of nonvolatile high‐density, low‐cost resistive switching (RS) memories and artificial synapses, resulting from their excellent electronic and optoelectronic properties including large light absorption coefficient, fast ion migration, long carrier diffusion length, low trap density, high defect tolerance. Among MHPs, 2D halide perovskites have exotic layered structure and great environment stability as compared with 3D counterparts. Herein, recent advances of 2D MHPs for the RS memories and artificial synapses realms are comprehensively summarized and discussed, as well as the layered structure properties and the related physical mechanisms are presented. Furthermore, the current issues and developing roadmap for the next‐generation 2D MHPs RS memories and artificial synapse are elucidated.

The design and preparation of high-performance separators for lithium-ion batteries (LIBs) have far-reaching practical significance in enhancing the overall performance of LIBs. Electrospun nanofiber separators (ENSs) have the characteristics of large specific surface area, high porosity, small pore size and good affinity with the electrolyte, making them become ideal candidates for LIB separators. In this work, polyacrylonitrile (PAN)/polyurethane (PU) (PAU) ENSs loaded with boehmite (BM) particles (BM/PAU ENSs) were mass-produced using spherical section free surface electrospinning (SSFSE), and used as LIB separators. Their morphology, structures and performances were tested and characterized. The results showed that all BM/PAU ENSs maintained excellent thermal dimensional stability in the range of 140–180 °C, and had good electrolyte wettability and high porosity. The composite BM/PAU-2 ENS with the best performance had a porosity of 52.5%, an electrolyte uptake rate of 822.1%, and an ionic conductivity of 1.97 mS/cm. Additionally, the battery assembled with BM/PAU-2 separator also demonstrated best electrochemical performance, cycling performance, and rate capability, with a capacity retention rate of 94.4% after 80 cycles at 0.5 C, making it a promising high-performance separator for LIBs.

Abstract Optoelectronic logic gates have emerged as one of the key candidates for the creation of next generation logic devices. However, current optoelectronic logic gates can provide only one or two logic gates, severely limiting their applications. Here we report a self‐powered and mechanically flexible device based on a BaTiO3 ferroelectric film to produce multi‐modal logic gates. By exploiting the photo‐induced photovoltaic and pyroelectric effects of a Schottky junction which is created between BaTiO3 and LaNiO3, the device is able to provide five different optoelectronic logic gates, which can be operated using input lasers of different wavelength (405 or 785 nm). The mode of operation of the logic gate can be switched by controlling the wavelength and intensity of the input laser, where the switching process is both lossless and reversible. A logic gate array was designed to conduct the five logic operations, with 100% accuracy, thereby providing application potential for the Internet of Things, big data, and secure solutions for data processing and transmission.

Objectives To investigate how core competency and self-efficacy of newly graduated nurses affect their experience of transition shock, and to determine the relationship between these factors.Design A cross-sectional study.Methods 262 newly graduated nurses participated in a cross-sectional study by using demographic data, the transition shock scale, the competency inventory for registered nurses scale and the self-efficacy scale.Results Among newly graduated nurses, the score of transition shock was 77.641±24.140, the score of core competency was 125 (109.5, 163.5) and the score of self-efficacy was 2.5 (2,3), all of which were at a moderate level. Abstract In this study, a method for assimilating FY4A advanced geostationary radiance imager (AGRI) cirrus‐effected radiances (CER) is investigated, and the impact of this method on water vapor analysis and rainstorm forecasting is examined through observing system simulation experiments and actual case experiments. The high proportion of inverted humidity profiles in the cirrus‐effected pixels is the main reason for the negative effect of assimilation in the mid‐to‐lower troposphere. To address this, relevant constraint conditions are incorporated into the cost function. The statistical results reveal that the addition of a CER assimilation improves the analysis increment of water vapor, with pattern correlation coefficients of 0.33, 0.35, and 0.20 at 200, 300, and 400 hPa, respectively, which are greater than those of a clear‐sky radiance assimilation (0.28, 0.33, and 0.17, respectively). Moreover, the inclusion of a CER assimilation greatly improves data utilization, and has a neutral to positive effect on precipitation forecasting.

BackgroundThere is currently a lack of studies examining the long-term therapeutic effectiveness of the third-generation anti-sezure medication, perampanel (PER), for focal-onset seizures (FOS), particularly in Chinese patients with sleep-related epilepsy (SRE). Additionally, the appropriate dosage, plasma concentration, and the relationship between dose and plasma concentration of PER in Chinese patients are still uncertain.MethodsA prospective, single-center, 24-month observational study was conducted in patients diagnosed with FOS, with a focus on patients with SRE. Changes in seizure frequency from baseline, adverse events, and retention rates were analyzed at 12 and 24 months following the start of the treatment. Tolerability was evaluated based on adverse events and discontinuation profiles. PER plasma concentrations were used to assess dose-concentration-response relationships.ResultsA total of 175 patients were included (median age: 25 years; range: 4–72 years; 53. 1% males and 46.9% females), with the SRE population accounting for 49. 1% (n = 86). The patients diagnosed with SRE showed considerably higher response rates than those who did not have this diagnosis (p = 0.025, odds ratio = 3.8). Additionally, the SRE group adhered better to PER treatment (r = 0.0009). Patients with a shorter duration of epilepsy (median: 3 years; range:2–7 years) demonstrated a more favorable therapeutic response to PER (p = 0.032). Throughout the administration of maintenance doses, among the entire FOS population, the concentration of PER (C0) ranged between 101.5 and 917.4 ng/mL (median, 232.0 ng/mL), and the mean plasma concentration of PER in the responders was 292.8 ng/mL. We revealed a linear relationship between PER dose and plasma concentration, regardless of whether PER was used as monotherapy or add-on therapy. The retention rates were 77.7% and 65. 1% at 12 and 24 months, respectively. Drug-related adverse events occurred in 45.0% of the patients and were mostly manageable.ConclusionPER effectively reduced seizure frequency in Chinese patients with FOS, particularly in those with SRE, over a 24-month period. The treatment was well-tolerated and had a clear linear dose-plasma concentration relationship.

Timber is the most widely used material for furniture in view of its characteristics of light mass, high strength, easy processing, coloring, and decorative appearance. However, the flammability of wood has been frequently associated with increased fire intensity and the rapid spread of fire in buildings. In this paper, the combustion performance of six kinds of common furniture timber was investigated based on thermogravimetric analysis at 25–500 °C, cone calorimetry with 50 kW/m2 thermal radiation intensity, and flame spread experiments with 3 kW/m2 thermal radiation intensity. The ignition, weight loss, thermogenesis, smoke, and flame spread characteristics of these timbers were obtained. Subsequently, a comprehensive index system including thermal stability, heat release ability, smoke production capacity, and flame spreading speed was constructed to evaluate the combustion performance of the selected timbers. In addition, a grey correlation method relying on the game theory to assign weight was proposed for the quantitative analysis of the relevant evaluation indexes. As a result, the combustion performance of the six kinds of timber, which was defined as a specific value from poor to good, was as follows: pine (0.8696) > Chinese fir (0.8568) > Oriented Strandboard (OSB) (0.8425) > density board (0.8122) > plywood (0.8087) > elm (0.7909). Timber with poor combustion performance contributes to the reduction in fire risk in buildings. Our suggestions are of great significance for selecting furniture timber from the perspective of the prevention and control of building fires.

IntroductionBelowground bud banks play integral roles in vegetation regeneration and ecological succession of plant communities; however, human-caused changes in land use severely threaten their resilience and regrowth. Although vegetation attributes and soil properties mediate such anthropogenic effects, their influence on bud bank size and composition and its regulatory mechanisms under land use change have not been explored.MethodsWe conducted a field investigation to examine impacts of land use change on bud bank size and composition, vegetation attributes, and soil properties in wetlands (WL), farmlands (FL), and alpine meadow (AM) ecosystems in Zhejiang Province, China.ResultsOverall, 63 soil samples in close proximity to the vegetation quadrats were excavated using a shovel, and samples of the excavated soil were placed in plastic bags for onward laboratory soil analysis. The total bud density (1514.727 ± 296.666) and tiller bud density (1229.090 ± 279.002) in wetland ecosystems were significantly higher than in farmland and alpine meadow ecosystems [i.e., total (149.333 ± 21.490 and 573.647 ± 91.518) and tiller bud density (24.666 ± 8.504 and 204.235 ± 50.550), respectively]. While vegetation attributes critically affected bud banks in WL ecosystems, soil properties strongly influenced bud banks in farmland and alpine meadow ecosystems. In wetland ecosystems, total and tiller buds were predominantly dependent on soil properties, but vegetation density played a significant role in farmlands and alpine meadow ecosystems. Root sprouting and rhizome buds significantly correlated with total C in the top 0 – 10 cm layer of farmland and alpine meadow ecosystems, respectively, and depended mainly on soil properties.DiscussionOur results demonstrate that land use change alters bud bank size and composition; however, such responses differed among bud types in wetland, farmland, and alpine meadow ecosystems.

Abstract Introduction This study aimed to investigate the potential application of plasma signal peptide‐complement C1r/C1s, Uegf and Bmp1‐epidermal growth factor domain‐containing protein 1 (SCUBE‐1) as a biomarker in the diagnosis of pulmonary embolism (PE). Methods This cross‐sectional study enrolled 177 patients who underwent PE diagnostic test and 87 healthy controls. The results of CT pulmonary angiogram (CTPA) were used as reference standards for PE diagnosis. The levels of SCUBE‐1 and D‐dimer in participants' plasma were detected with enzyme‐linked immunosorbent assay and compared among patients with confirmed PE, suspicious PE and healthy controls. The diagnostic values were analysed using receiver operating characteristic (ROC) curve analysis. In addition, differences in plasma SCUBE‐1 levels were compared among patients with different risk stratifications. Results The plasma SCUBE‐1 concentration levels in patients with CTPA confirmed PE (14.28 ± 7.74 ng/ml) was significantly higher than those in the suspicious patients (11.11 ± 4.48 ng/ml) and in healthy control (4.40 ± 3.23 ng/ml) (P 0.05). Conclusion We believe that SCUBE‐1 could be a potential coagulation‐related marker for the diagnosis of PE.

Abstract Phosphatidylinositol 3-kinase alpha (PI3Kα) inhibitors are currently evaluated for the therapy of esophageal squamous cell carcinoma (ESCC). It is of great importance to identify potential biomarkers to predict or monitor the efficacy of PI3Kα inhibitors in an aim to improve the clinical responsive rate in ESCC. Here, ESCC PDXs with CCND1 amplification were found to be more sensitive to CYH33, a novel PI3Kα-selective inhibitor currently in clinical trials for the treatment of advanced solid tumors including ESCC. Elevated level of cyclin D1, p21 and Rb was found in CYH33-sensitive ESCC cells compared to those in resistant cells. CYH33 significantly arrested sensitive cells but not resistant cells at G1 phase, which was associated with accumulation of p21 and suppression of Rb phosphorylation by CDK4/6 and CDK2. Hypo-phosphorylation of Rb attenuated the transcriptional activation of SKP2 by E2F1, which in turn hindered SKP2-mediated degradation of p21 and reinforced accumulation of p21. Moreover, CDK4/6 inhibitors sensitized resistant ESCC cells and PDXs to CYH33. These findings provided mechanistic rationale to evaluate PI3Kα inhibitors in ESCC patients harboring amplified CCND1 and the combined regimen with CDK4/6 inhibitors in ESCC with proficient Rb.

Objectives: To describe the epidemiological, clinical, and household transmission characteristics of pediatric COVID-19 cases in Shanghai, China. Methods: Pediatric patients with COVID-19 hospitalized in Shanghai from March-May 2022 were enrolled in this retrospective, multicenter cohort study. The symptoms and the risk factors associated with disease severity were analyzed. Results: In total, 2620 cases (age range, 24 days-17 years) were enrolled in this study. Of these, 1011 (38.6%) were asymptomatic, whereas 1415 (54.0%), 190 (7.3%), and 4 (0.2%) patients developed mild, moderate, and severe illnesses, respectively. Household infection rate was negatively correlated with household vaccination coverage. Children aged 0-3 years, those who are unvaccinated, those with underlying diseases, and overweight/obese children had a higher risk of developing moderate to severe disease than children aged 12-17 years, those who were vaccinated, those without any underlying disease, and those with normal weight, respectively (all P

Abstract Background Intraperitoneal metastasis is one of the major causes of the high mortality rate of ovarian cancer. Bufalin (BU) is an effective component of the traditional Chinese medicine Chansu that exerts antitumor effects, including metastasis inhibition. In our previous studies, we found that BU inhibited the migration and invasion of ovarian cancer cells. However, the application of BU is limited due to its insolubility, toxicity and imprecise targeting. The aim of this study was to use vitamin E succinate (VES)-grafted chitosan oligosaccharide (CSO)/arginine-glycine-aspartic acid peptide (RGD)-conjugated d-alpha-tocopheryl polyethylene glycol 1000 succinate (TPGS) mixed micelles (VeC/T-RGD MMs) to deliver BU to ovarian cancer cells to inhibit intraperitoneal metastasis. Moreover, the toxicity of BU was reduced by coating it with the mixed micelles to increase its biocompatibility for practical applications. Results The BU-loaded VeC/T-RGD MMs (BU@MMs) had an average diameter of 161 ± 1.4 nm, a zeta potential of 4.49 ± 1.54 mV and a loading efficiency of 2.54%. The results showed that these micelles inhibited cell proliferation, induced apoptosis, and reduced the migration and invasion of A2780 and SKOV3 cells. Further studies indicated that BU@MMs enhanced the levels of e-cadherin and decreased the expression levels of N-cadherin, vimentin and Snail in vitro. In addition, the mixed micelles effectively enhanced the anticancer effect and inhibited intraperitoneal metastasis in intraperitoneal metastatic models. The BU@MMs exhibited fewer toxic side effects than BU, indicating better biocompatibility and biosafety for in vivo applications. Conclusions Our studies show that BU@MMs are a potential multifunctional nano-drug delivery system that can effectively inhibit the intraperitoneal metastasis of ovarian cancer.

BackgroundMultiple endocrine neoplasia type 1 (MEN1) is an inherited endocrine syndrome caused by the mutation in the tumor suppressor gene MEN1. The recurrence rate of primary hyperparathyroidism (PHPT) in patients with MEN1 after parathyroidectomy remains high, and the management of recurrent hyperparathyroidism is still challenging.Case presentationWe reported a 44-year-old woman with MEN1 combined with PHPT who was diagnosed through genetic screening of the patient and her family members. After parathyroidectomy to remove one parathyroid gland, the patient suffered from persistent high levels of serum calcium and parathyroid hormone, which returned to normal at up to 8 months after ultrasound-guided microwave ablation (MWA) for bilateral parathyroid glands, suggesting an acceptable short-term prognosis.ConclusionUltrasound-guided MWA for parathyroid nodules may be an effective therapeutic strategy for recurrent PHPT in MEN1 patients.

Objectives: To evaluate excess deaths of gastrointestinal, liver, and pancreatic diseases in the United States during the COVID-19 pandemic.Methods: We retrieved weekly death counts from National Vital Statistics System and fitted them with a quasi-Poisson regression model. Cause-specific excess deaths were calculated by the difference between observed and expected deaths with adjustment for temporal trend and seasonality. Demographic disparities and temporal-spatial patterns were evaluated for different diseases.Results: From March 2020 to September 2022, the increased mortality (measured by excess risks) for Clostridium difficile colitis, gastrointestinal hemorrhage, and acute pancreatitis were 35.9%; 24.8%; and 20.6% higher than the expected. For alcoholic liver disease, fibrosis/cirrhosis, and hepatic failure, the excess risks were 1.4–2.8 times higher among younger inhabitants than older inhabitants. The excess deaths of selected diseases were persistently observed across multiple epidemic waves with fluctuating trends for gastrointestinal hemorrhage and fibrosis/cirrhosis and an increasing trend for C. difficile colitis.Conclusion: The persistently observed excess deaths of digestive diseases highlights the importance for healthcare authorities to develop sustainable strategies in response to the long-term circulating of SARS-CoV-2 in the community.

With the increasing application of electrospun nanofibers, the batch preparation of high-performance functional nanofibers containing nanoparticles has become a research hotspot. As the distribution uniformity of nanoparticles in functional nanofibers has a great impact on their performance, an electrospinning device with multiple air inlets, which has a copper porous spinneret, is proposed to obtain functional nanofibers with higher yield and more uniform distribution of nanoparticles. The mechanism of batch preparation of functional nanofibers containing ZnO nanoparticles by the device was studied through experiments and theoretical analysis. The experimental data are in good agreement with the theoretical analysis results, which showed that under the appropriate voltage (50 kV) and air flow (50 m3/h), the device could keep ZnO nanoparticles contained in the spinning solution evenly dispersed during the spinning process, thus obtaining functional nanofibers with more uniform distribution of ZnO nanoparticles, whose quality and yield were higher than those prepared by other high-yield electrospinning devices.

Nanofluids have recently been utilized in experimental studies to enhance the performance of flat plate solar collectors (FPSC). The reported results for the nanofluids’ effect on this solar collector are ambiguous and sometimes contradictory. Furthermore, there is no reliable model to analyze the impact of nanofluids’ properties on the FPSC thermal performance. Therefore, this research develops a straightforward approach to predict the thermal efficiency of nanofluid-based FPSC. Pearson’s analysis confirmed that the three-quarters root of the FPSC’s thermal efficiency is the best transformation for simulating the considered problem. The machine learning models are then applied to relate the transformed thermal efficiency to the absorbed energy, energy loss, reduced temperature, the tilt angle of a flat plate, and nanoparticles’ size. Prediction performance of artificial neural networks (ANN), least-squares support vector regression (LS-SVR), adaptive neuro-fuzzy inference system (ANFIS), and available correlations have been compared to distinguish the highest accurate tool for the considered task. The results demonstrate that the LS-SVR has higher accuracy than other correlations for numerically analyzing the thermal efficiency of the FPSC. This highest accurate paradigm predicts 545 experimental datasets with the absolute average relative deviation (AARD) of 2.77%, mean squared errors (MSE) of 0.00039, and coefficient of determination (R2) of 0.99311.

Objective Ecological momentary assessment (EMA) refers to the repeated sampling of information about an individual’s symptoms and behaviours, enabling the capture of ecologically meaningful real-time information in a timely manner. Compliance with EMA is critical in determining the validity of an assessment. Background: Hemodialysis patients have a high risk of severe/critical COVID-19 and related high mortality, but nirmatrelvir/ritonavir is not recommended for hemodialysis patients with COVID-19 infection because of lack of evidence of safety.Objectives: Our study aims to evaluate the minimum plasma concentration (Cmin) of nirmatrelvir and its safety of different doses of nirmatrelvir/ritonavir in hemodialysis patients with mild COVID-19.Method: This was a prospective, two step, nonrandomized, open-label study. Participants were treated with nirmatrelvir 150 mg or 300 mg once a day (another 75 mg or 150 mg supplied after hemodialysis) and ritonavir 100 mg twice daily for 5 days, respectively. The primary outcome was the safety of nirmatrelvir/ritonavir, including the Cmin of nirmatrelvir and the number of adverse events (AE). The secondary outcome was the time of viral elimination in hemodialysis patients.Results: Adverse events were happened in 3 and 7 participants in the step 1 and step 2 group, respectively (p = 0.025). Among them, 2 and 6 participants were identified as drug-related adverse events (p = 0.054). No SAE or liver function damage happened. The Cmin of nirmatrelvir in step 1 and step 2 group were 5,294.65 ± 2,370.59 ng/mL and 7,675.67 ± 2,745.22 ng/mL (p = 0.125). The Cmin of the control group was 2,274.10 ± 1,347.25 ng/mL (p = 0.001 compared to step 2 and p = 0.059 compared to step 1). Compared to hemodialysis patients without nirmatrelvir/ritonavir, there were no statistical differences in overall viral elimination time (p = 0.232).Conclusion: In our study, two doses of nirmatrelvir/ritonavir appeared to be excessive for hemodialysis patients. Although all of the patients tolerated 5-day administration, nearly half of the patients experienced drug-related adverse events. In addition, the medication group did not show a significant advantage in the time of viral elimination.

From 17 to 22 July 2021, the “21·7” extreme rainfall event (“21·7” ERE) hit Henan Province, breaking the record for mainland China with a maximum hourly rainfall of 201.9 mm at the Zhengzhou station. The long-lived (20 h) mesoscale Huang-Huai vortex (HHV) was an important system that directly affected the major rainfall stage, including the extreme hourly rainfall. This study investigates the formation and development mechanism of the HHV, as well as its association with the simulation of extreme hourly rainfall through numerical simulations. The simulated rainfall and radar composite reflectivity were in good agreement with the observations, thus effectively reproducing the generation and developmental process of the HHV. The analysis results showed that the HHV initially formed at 850 hPa on 19 July at 1800 UTC and eventually developed to 550 hPa. The positive feedback formed by the horizontal convergence and vertical vorticity transport was the main mechanism leading to the generation and deepening of the HHV. The stretching effect (STR) term played an absolutely dominant role in the increase in the vorticity tendency, and it primarily originated from the coupling effect of boundary layer jets (BLJs) and synoptic-weather-system-related low-level jets (SLLJs). The accurate simulation of the HHV allowed the early rainfall to reasonably reproduce the surface cold pool near the Zhengzhou station, and then the cooperation of the SLLJs, the BLJs, and the cold pool made the simulated extreme hourly rainfall exactly close to the Zhengzhou station, but with a weaker intensity, due to the fact that the HHV moved northeastward after its formation, resulting in a narrow range of southerly flow in southern Henan, which is not conducive to convective triggering in the southerly flow.

As an important element of lithium-ion batteries (LIBs), the separator plays a critical role in the safety and comprehensive performance of the battery. Electrospun nanofiber separators have a high porosity and good electrolyte affinity, which are favorable to the transference of lithium ions. In this paper, the batch preparation of polyacrylonitrile (PAN)-based nanofiber separators are obtained via spherical section free surface electrospinning (SSFSE). Introducing an appropriate amount of polyester polyurethane (PU) can effectively enhance the mechanical property of PAN nanofiber separators and help the separators resist the external force extrusion. The results show that when PAN:PU = 8:2, the porosity and electrolyte uptake rate of the composite nanofiber separator (PAN-2) are 62.9% and 643.3%, respectively, exhibiting a high ionic conductivity (1.90 mS/cm). Additionally, the coin battery assembled with PAN-2 as a separator (LiFePO4/PAN-2/lithium metal) shows good cycling performance and good rate performance, with a capacity retention rate of 93.9% after 100 cycles at 0.5 C, indicating that the battery with PAN-2 has a good application potential in advanced energy storage.

Undesired maturation and reproduction are major challenges in fish aquaculture regarding genetic introgression, precocious maturation, and reproduction-related mortality, which can have profound ecological or economic impacts. Farming reproductively sterile fish can effectively mitigate these challenges. In this paper, we transferred and applied a novel immersion-based, non-transgenic gene silencing technology to sterilize coho salmon for the first time. Unfertilized eggs were bath immersion-treated with csdnd-MO-Vivo in different immersion media. Eyed rates of treated groups ranged from 0.9 to 63.5%. Sterile fish lacking germ cells, and those with arrested germ cells/atretic oocytes, were obtained at 14 and 20 months of age, albeit at a low percentage (2.3 to 10.0% based on females). Gonadal histology and vasa/nanos3 gene expression profile were provided for comparing fertile and sterile gonads, as well as retarded ovaries. Future directions and strategies for optimizing the technology and improving sterility induction were also proposed. The successful production of sterile coho salmon achieved in this study demonstrates the proof of principle for this new sterilization technology. As we continue to expand upon these findings and refine the technology, achieving coho salmon sterile population farming would facilitate the future transfer and application to other commercially important aquaculture fish.

Facing the era of information explosion and the advent of artificial intelligence, there is a growing demand for information technologies with huge storage capacity and efficient computer processing. However, traditional silicon-based storage and computing technology will reach their limits and cannot meet the post-Moore information storage requirements of ultrasmall size, ultrahigh density, flexibility, biocompatibility, and recyclability. As a response to these concerns, polymer-based resistive memory materials have emerged as promising candidates for next-generation information storage and neuromorphic computing applications, with the advantages of easy molecular design, volatile and non-volatile storage, flexibility, and facile fabrication. Herein, we first summarize the memory device structures, memory effects, and memory mechanisms of polymers. Then, the recent advances in polymer resistive switching materials, including single-component polymers, polymer mixtures, 2D covalent polymers, and biomacromolecules for resistive memory devices, are highlighted. Finally, the challenges and future prospects of polymer memory materials and devices are discussed. Advances in polymer-based memristors will open new avenues in the design and integration of high-performance switching devices and facilitate their application in future information technology.

Abstract Background Arsenic trioxide (ATO) is highly effective in acute promyelocytic leukemia (APL) patients, but it fails to show satisfactory efficacy in other acute myeloid leukemia (AML) patients with non-APL subtypes. Different from the APL cells, most non-APL AML cells express low levels of the ATO transporter Aquaporin-9 (AQP9) protein, making them less sensitive to ATO treatment. Recently, we found that granulocyte colony stimulating factor (G-CSF) can upregulate the expression of AQP9. We hypothesized that the pretreatment with G-CSF may enhance the antitumor effect of ATO in non-APL AML cells. In addition, we aimed to elucidate the underlying mechanisms by which G-CSF upregulates the expression of AQP9. Methods Non-APL AML cell lines including THP-1 and HL-60 were pretreated with or without G-CSF (100 ng/ml) for 24 h, followed by the treatment with ATO (2 μM) for 48 h. Cell morphology was observed under the microscope after Wright-Giemsa staining. Flow cytometry was performed to evaluate the cell apoptosis levels. The intracellular concentrations of ATO were determined by atomic fluorescence spectrometry. The mRNA and protein expression were respectively measured by quantitative reverse transcription PCR (RT-qPCR) and western blotting. Target genes were knocked down by transfection with small interfering RNA (siRNA), or overexpressed by transfection with overexpression plasmids. The cell line derived xenograft mouse model was established to confirm the results of the in vitro experiments. Results Compared with using ATO alone, the combination of G-CSF with ATO induced the cell apoptosis more dramatically. G-CSF upregulated the expression of AQP9 and enhanced the intracellular concentrations of ATO in AML cells. When AQP9 was overexpressed, it markedly enhanced the cytotoxic activity of ATO. On the other hand, when AQP9 was knocked down, it profoundly attenuated the combinational effect. Moreover, we found that the upregulation of AQP9 by G-CSF depends on the transcription factor CCAAT enhancer binding protein beta (CEBPB). We also demonstrated that the combination of G-CSF and ATO significantly inhibited tumor growth in the xenograft mouse model. Conclusions The combination of G-CSF and ATO may be a potential therapeutic strategy for AML patients.

Affected by the pressure and constraints of available resources, plant growth and development, as well as plant life history strategies, usually vary with environmental conditions. Plant buds play a crucial role in the life history of woody plants. Nitraria tangutorum is a common dominant woody species in desertified areas of northern China and its growth is critical to the desert ecosystem. Revealing the allometry of N. tangutorum aboveground bud fates and the linkage between bud traits and plant nutrient contents and stoichiometric ratios can be useful in understanding plant adaptation strategy. We applied seven nitrogen and phosphorus fertilizer addition treatments to natural N. tangutorum ramets in Ulan Buh Desert in three consecutive years. We surveyed three types of aboveground buds (dormant buds, vegetative buds, and reproductive buds) in each N. tangutorum ramet, then measured the plant carbon (C), nitrogen (N), and phosphorus (P) contents and ratios during three consecutive years. We specified that reserve growth potential (RGP), vegetative growth intensity (VGI) and sexual reproduction effort (SRE) are the three indices of bud dynamic pattern. The results showed that the bud dynamic pattern of N. tangutorum ramets differed significantly among different fertilizer addition treatments and sampling years. The allometry of RGP, VGI, and SRE was obvious, showing size dependence. The allometric growth relationship fluctuated among the sampling years. The linkage between bud traits and plant stoichiometric characteristics of N. tangutorum ramets showed close correlation with plant P content, C:P and N:P ratios, no significant correlation with plant C content, N content and C:N ratio. These results contribute to an improved understanding of the adaptive strategies of woody plants growing in desert ecosystems and provide insights for adoption of effective measures to restore and conserve plant communities in arid and semi-arid regions.

ObjectivesThis study aimed to explore the reasons and influencing factors for non-return to work (non-RTW) within 1 year among young and middle-aged patients with stroke and to assess their health-related quality of life (HRQoL) at 1 year across different reasons.MethodsThe study was conducted as a telephone-based cross-sectional survey. Seven hundred eighty-nine young and middle-aged patients with stroke aged between 18 and 54 years for men and 18 and 49 years for women in the electronic medical system were included. Data collection included demographic characteristics, socioeconomic status, behavioral habits, history of chronic diseases, work status, reasons for non-RTW, and HRQoL.ResultsOf 789 patients, 435 (55.1%) (mean [SD] age, 47.7 [7.8] years) did not return to work within 1 year after stroke. Among the patients who did not RTW, 58.9% were unable to work, 9.7% retired early, 11.03% became full-time homemakers or were unemployed, and 20.5% were reluctant to work. The disordered multiclass logistic regression model showed that the factors influencing the reasons for non-RTW included age, gender, education, income, health insurance, diabetes comorbidity, ability to perform activities of daily living, and mobility of the right upper extremity. Furthermore, patients who were unable to work had significantly lower HRQoL compared to those who had RTW, followed by those who retired early.ConclusionsMore than half did not RTW within 1 year in our study. The results will help inform future research to identify interventions to promote RTW and improve HRQoL for young and middle-aged patients with stroke.

Abstract Climate change and disturbance can alter invasion success of clonal plants by differentially affecting the clonal traits influencing their establishment as young plants. Clonal traits related to the vegetative reproduction of native Pascopyrum smithii and non-native Bromus inermis grass seedlings were evaluated under altered precipitation frequencies and a single grazing event. Pascopyrum smithii maintained similar vegetative reproduction under three simulated precipitation frequencies whereas B. inermis vegetative reproduction declined as precipitation became more intermittent. Vegetative reproduction of the non-native B. inermis was greater than the native P. smithii under all simulated precipitation frequencies except the most intermittent scenario. A single grazing event did not affect either species’ response to intra-annual precipitation variability but did slightly reduce their clonal growth and increase their bud dormancy. In young plants, clonal traits of the invasive grass favored its superior expansion and population growth compared to the native grass except under the most severe climate change scenario. Grassland restoration using native P. smithii seeds would be successful in most years due to its resilient clonal growth in a changing climate. Clonal infrastructure development in young plants is critical to clonal plant establishment and persistence in a changing climate and under disturbed conditions.

Electrospun nanofiber membranes (NFMs) have high porosity and a large specific surface area, which provide a suitable environment for the complex and dynamic wound healing process and a large number of sites for carrying wound healing factors. Further, the design of the nanofiber structure can imitate the structure of the human dermis, similar to the natural extracellular matrix, which better promotes the hemostasis, anti-inflammatory and healing of wounds. Therefore, it has been widely studied in the field of wound dressing. This review article overviews the development of electrospinning technology and the application of electrospun nanofibers in wound dressings. It begins with an introduction to the history, working principles, and transformation of electrospinning, with a focus on the selection of electrospun nanofiber materials, incorporation of functional therapeutic factors, and structural design of nanofibers and nanofiber membranes. Moreover, the wide application of electrospun NFMs containing therapeutic factors in wound healing is classified based on their special functions, such as hemostasis, antibacterial and cell proliferation promotion. This article also highlights the structural design of electrospun nanofibers in wound dressing, including porous structures, bead structures, core-shell structures, ordered structures, and multilayer nanofiber membrane structures. Finally, their advantages and limitations are discussed, and the challenges faced in their application for wound dressings are analyzed to promote further research in this field.

With the strengthening of the public awareness of food safety and environmental protection, functional food packaging materials have received widespread attention. Nanofibers are considered as promising packaging materials due to their unique one-dimensional structure (high aspect ratio, large specific surface area) and functional advantages. Electrospinning, as a commonly used simple and efficient method for preparing nanofibers, can obtain nanofibers with different structures such as aligned, core-shell, and porous structures by modifying the devices and adjusting the process parameters. The selection of raw materials and structural design of nanofibers can endow food packaging with different functions, including antimicrobial activity, antioxidation, ultraviolet protection, and response to pH. This paper aims to provide a comprehensive review of the application of electrospun nanofibers in functional food packaging. Advances in electrospinning technology and electrospun materials used for food packaging are introduced. Moreover, the progress and development prospects of electrospun nanofibers in functional food packaging are highlighted. Meanwhile, the application of functional packaging based on nanofibers in different foods is discussed in detail.

BackgroundThe triple combination of hepatic arterial infusion chemotherapy (HAIC) with fluorouracil, leucovorin, and oxaliplatin (FOLFOX) plus tyrosine kinase inhibitor (TKI) and immune checkpoint inhibitors (ICIs) is expected to have a synergistic anticancer effect in HCC. We conducted this meta-analysis to evaluate the efficacy and safety of the triple combination treatment in advanced HCC patients.MethodsPubMed, Embase, Cochrane Library, Web of Science databases were systematically searched for relevant studies from the inception of each database to May 10, 2023. All articles focusing the triple combination treatment of HAIC-FO plus TKI and ICIs for advanced HCC were eligible. The meta-analysis was conducted following the PRISMA guidelines. The risk of bias was assessed using the Joanna Briggs Institute (JBI) for case series and Newcastle-Ottawa Scale (NOS) for cohort studies. The primary outcomes were overall survival (OS), progression-free survival (PFS), objective response rate (ORR) and disease control rate (DCR). The secondary results were adverse events. Further meta-analysis of control studies demonstrated the superiority of the triple combination modality to TKI plus ICIs, and TKI alone.ResultsNine articles (four cohort studies and five one-arm studies) involving 777 advanced HCC patients were included in this meta-analysis. In terms of survival analysis, the pooled median PFS was 11 months (95% CI: 10.1-12.0 months) with low heterogeneity (I2 = 0%, p = 0.97). With regard to tumor response, the pooled ORR and DCR was 61.6% (I2=0%, p = 0.71) and 87.9% (I2 = 13%, p = 0.33) with low heterogeneity, respectively. As compared with TKIs plus ICIs, and TKIs alone, the triple combination thrapy was associated with improved median OS (HR=0.51, 95%CI 0.41-0.62) with low heterogeneity across studies (I2 = 0%, p = 0.47), median PFS (HR=0.51, 95%CI 0.41-0.64) with low heterogeneity across studies (I2 = 0%, p = 0.41), ORR (RR = 0.56, 95% CI: 0.42-0.74) with high heterogeneity across studies (I2 = 69%, p = 0.02), and DCR (RR = 0.38, 95%CI 0.27-0.54) with low heterogeneity across studies (I2 = 14%, p = 0.32). The most common 3/4 AEs were elevated ALT and AST, thrombocytopenia, hypertension, nausea and vomiting in this meta-analysis.ConclusionsThe triple combination therapy of HAIC-FO plus TKI and ICIs showed promising efficacy and safety in patients with advanced HCC.RegistrationThe protocol was registered with PROSPERO (ID:CRD42023424281).

A traditional thermal conductivity vacuum gauge mainly detects low pressure (the degree of vacuum) by measuring the temperature change of a filament heated by the electric current. We propose a novel pyroelectric vacuum sensor that utilizes the effect of ambient thermal conductivity on the pyroelectric effect to detect vacuum through the charge density of ferroelectric materials under radiation. The functional relationship between the charge density and low pressure is derived, which is validated in a suspended (Pb,La)(Zr,Ti,Ni)O3 (PLZTN) ferroelectric ceramic-based device. The charge density of the indium tin oxide/PLZTN/Ag device under 405 nm of 60.5 mW cm−2 radiation at low pressure reaches 4.48 μC cm−2, which is increased by about 3.0 times compared with that at atmospheric pressure. The vacuum can improve the charge density without increasing the radiation energy, confirming the important role of ambient thermal conductivity on the pyroelectric effect. This research provides a demonstration for ambient thermal conductivity effectively tuning pyroelectric performance, a theoretical basis for pyroelectric vacuum sensors, and a feasible route for further optimizing the performance of pyroelectric photoelectric devices.

Due to the particularity of biomedical industry, it has become necessary for biomedical enterprises to seek innovative research and development (R&D) cooperation to maintain advanced technologies and products in multiple fields. Under such circumstance, the biomedical industry has gradually formed a certain cluster to promote the development of the industry. So far, the biomedical industry cluster has formed in China, mainly within the Yangtze River Delta, Pearl River Delta, and Beijing-Tianjin-Hebei three urban agglomerations. Within the industrial clusters, the frequency of innovation cooperation among enterprises, universities, research institutions, and other relevant organizations in the biomedical area is high, and the capacity for innovation cooperation is strong as well. This paper used the representative cross-section data of cooperative patents from the medical science and technology patent database of China National Knowledge Infrastructure (CNKI), researching the R&D cooperation within the three major urban agglomerations in China from 2008 to 2016 (Yangtze River Delta Urban Agglomeration, Pearl River Delta Urban Agglomeration, Beijing-Tianjin-Hebei Urban Agglomeration) on total 36 cities' spatial pattern characteristics of biomedical cooperation and the influencing factors. The spatial interaction model was used to study the spatial, economic, political, and R&D influencing factors of cross-city cooperation. The degree of aggregation showed that cross-city R&D cooperation mainly occurred in well-developed and central cities of urban agglomerations. Econometric results revealed that spatial, economic, political, and R&D bias factors did have a significant impact on the frequency of biomedical R&D cooperation across cities.

The list of long non-coding RNAs (lncRNAs) that participate in the function of ovarian granulosa cells (GCs) is rapidly expanding, but the mechanisms through which lncRNAs regulate GC function are not yet fully understood. Here, we recognized a minimally expressed lncRNA RP4-545C24.1 (which we named DDGC) in GCs from patients with biochemical premature ovarian insufficiency (bPOI). We further explored the role of lncRNA DDGC in GC function and its contribution to the development of bPOI. Mechanistically, silencing DDGC downregulated RAD51 by competitively binding with miR-589-5p, and this resulted in significant inhibition of DNA damage repair capacity. In addition, decreased expression of DDGC promoted ubiquitin-mediated degradation of Wilms tumor 1 (WT1) protein through interactions with heat shock protein 90 (HSP90), which led to aberrant differentiation of GCs. Moreover, DDGC was able to ameliorate the etoposide-induced DNA damage and apoptosis in vivo. Taken together, these findings provide new insights into the contribution of lncRNAs in POI pathogenesis.

Acute hyperglycemia is a powerful indicator of the severity of acute ischemic stroke (AIS); however, the relationship between these two factors is not very clear in patients with diabetes. We aimed to retrospectively evaluate data from 335 consecutive patients who experienced AIS from November 2015 to November 2016 to investigate whether a comprehensive assessment of blood glucose levels is a more valuable indicator of the severity of AIS or the presence of acute hyperglycemia in patients with diabetes. We collected demographic data, clinical manifestation information, clinical scores, and laboratory data [including fasting blood glucose and glycated hemoglobin (HbA1c) levels]. We estimated prehospital mean blood glucose concentrations using the following formula [1.59 * HbA1c (%) – 2.59] to calculate the “Acute-to-Chronic Glycemic Ratio” (AC ratio). The AC ratio differed significantly among patients grouped according to the National Institutes of Health Stroke Scale/Score (NIHSS) at admission (admission NIHSS) (p = 0.006). Univariate regression analysis revealed a correlation between the AC ratio and admission NIHSS [standardized β-coefficient (Std. B) = 0.164, p = 0.004]. The adjusted linear regression analysis revealed a correlation between both HbA1c (Std. B = 0.368, p = 0.038) and the AC ratio (Std. B = 0.262, p = 0.022) and admission NIHSS. The AC ratio (Std. B = 0.161, p = 0.012) was related to admission NIHSS in the stepwise variable selection. For an admission NIHHS > 4, the AC ratio (Std. B = 0.186, p = 0.047) was related to admission NIHSS in the stepwise variable selection. The AC ratio (Std. B = 1.163, p = 0.006 and Std. B = 0.565, p = 0.021, respectively) were related to admission NIHSS in both large-artery atherosclerosis (LAA) and small-vessel occlusion (SVO) subgroups. Thus, the AC ratio is related to admission NIHSS in patients with diabetes who experienced AIS and may be a better indicator of severity than acute blood glucose levels.

Abstract FANCI is an essential component of Fanconi anemia pathway, which is responsible for the repair of DNA interstrand cross-links (ICLs). As an evolutionarily related partner of FANCD2, FANCI functions together with FANCD2 downstream of FA core complex. Currently, growing evidences showed that the essential role of FA pathway in male fertility. However, the underlying mechanisms for FANCI in regulating spermatogenesis remain unclear. In the present study, we found that the male Fanci −/− mice were sterile and exhibited abnormal spermatogenesis, including massive germ cell apoptosis in seminiferous tubules and dramatically decreased number of sperms in epididymis. Besides, FANCI deletion impaired maintenance of undifferentiated spermatogonia. Further investigation indicated that FANCI was essential for FANCD2 foci formation and regulated H3K4 and H3K9 methylation on meiotic sex chromosomes. These findings elucidate the role and mechanism of FANCI during spermatogenesis in mice and provide new insights into the etiology and molecular basis of nonobstructive azoospermia.

The U.N. Decade on Ecosystem Restoration aims to accelerate actions to prevent, halt, and reverse the degradation of ecosystems, and re-establish ecosystem functioning and species diversity. The practice of ecological restoration has made great progress in recent decades, as has recognition of the importance of species diversity to maintaining the long-term stability and functioning of restored ecosystems. Restorations may also focus on specific species to fulfill needed functions, such as supporting dependent wildlife or mitigating extinction risk. Yet even in the most carefully planned and managed restoration, target species may fail to germinate, establish, or persist. To support the successful reintroduction of ecologically and culturally important plant species with an emphasis on temperate grasslands, we developed a tool to diagnose common causes of missing species, focusing on four major categories of filters, or factors: genetic, biotic, abiotic, and planning & land management. Through a review of the scientific literature, we propose a series of diagnostic tests to identify potential causes of failure to restore target species, and treatments that could improve future outcomes. This practical diagnostic tool is meant to strengthen collaboration between restoration practitioners and researchers on diagnosing and treating causes of missing species in order to effectively restore them.

The combination of multiple electrode materials and their reasonable structural design are conducive to the preparation of composite electrodes with excellent performance. In this study, based on carbon nanofibers grown with Ni(OH)2 and NiO (CHO) prepared by electrospinning, hydrothermal growth, and low-temperature carbonization, five transition metal sulfides (MnS, CoS, FeS, CuS, and NiS) were hydrothermally grown on their surfaces, exhibiting that CHO/NiS had the optimal electrochemical properties. Subsequently, the effect of hydrothermal growth time on CHO/NiS revealed that the electrochemical performance of CHO/NiS-3h was optimal, with a specific capacitance of up to 1717 F g−1 (1 A g−1), due to its multistage core–shell structure. Moreover, the diffusion-controlled process of CHO/NiS-3h dominated its charge energy storage mechanism. Finally, the asymmetric supercapacitor assembled with CHO/NiS-3h as the positive electrode demonstrated an energy density of 27.76 Wh kg−1 at a maximum power density of 4000 W kg−1, and it still maintained a power density of 800 W kg−1 at a maximum energy density of 37.97 Wh kg−1, exhibiting the potential application of multistage core–shell composite materials in high-performance supercapacitors.

The atmospheric temperature profiles (ATPs) retrieved through the geostationary Interferometric Infrared Sounder (GIIRS) onboard the FY-4A satellite (GIIRS/FY-4A) can effectively fill the gap of the scarce conventional sounding data in the Taklimakan Desert (TD), the second largest desert in the world, with an area of 330,000 square kilometers. In this study, we take the experimental radiosonde observations (RAOB) from one RAOB station in the hinterland of TD and seven conventional radiosondes in the oasis region around the desert as the true values and analyze the bias distribution characteristics of GIIRS/FY-4A ATPs with quality control (QC) flags 0 or 1 for this region. In addition, a bias comparison is made with GIIRS/FY-4A ATPs, and the fifth generation ECMWF atmospheric reanalysis of the global climate (ERA5) ATPs. The results show that (1) Missing measurements in GIIRS/FY-4A ATPs are the most frequent in the near-surface layer, accounting for more than 80% of all the retrieved grid points. The averaged total proportion of GIIRS/FY-4A ATPs with QC marks 0 or 1 is about 33.06%. (2) The root mean square error (RMSE) of GIIRS/FY-4A ATPs is less than 3 K, smaller than that of ERA5 ATPs. The RMSE of ERA5 ATPs can exceed 10 K in the desert hinterland. The absolute mean biases of GIIRS/FY-4A ATPs and ERA5 ATPs are, respectively, smaller than 3 K and 2 K, the former being slightly larger. The correlation coefficients of GIIRS/FY-4A ATPs with ERA5 ATPs and RAOB ATPs are higher than 0.98 and 0.99, respectively, and the correlation between GIIRS/FY-4A ATPs and RAOB ATPs is inferior to the latter. (3) The overall atmospheric temperature retrieved by GIIRS/FY-4A is 0.08 K higher than the temperature of RAOB, on average, while the overall temperature from ERA5 is 0.13 K lower than that of RAOB, indicating that the temperature profile obtained by integrating GIIRS/FY-4A ATPs and ERA5 ATPs may be much closer to RAOB ATPs. (4) The probability density of the GIIRS/FY-4A ATP biases in the TD region generally follows the Gaussian distribution so that it can be effectively assimilated in the 3-D variational assimilation modules. The probability density distribution characteristics of the GIIRS/FY-4A ATP biases in the desert hinterland and oasis are not much different. However, due to the fusion analysis of the relatively rich multi-source conventional observation data from the oasis stations, the probability density of ERA5 ATPs biases at the oasis stations is nearer to Gaussian distribution than that of the GIIRS/FY-4A ATPs. In the desert hinterland, where conventional observation is not enough, the probability density distributions of the ATPs biases from ERA5 and GIIRS/FY-4A are alike. Therefore, the GIIRS FY4A can contribute to a more accurate estimation of ERA5 ATPs in the TD region.

Abstract Background Lipoprotein(a) is genetically determined and increasingly recognized as a major risk factor for arteriosclerotic cardiovascular disease. We examined whether plasma lipoprotein(a) concentrations were associated with intraplaque neovascularization (IPN) grade in patients with carotid stenosis and in terms of increasing plaque susceptibility to haemorrhage and rupture. Methods We included 85 patients diagnosed with carotid stenosis as confirmed using carotid ultrasound who were treated at Guangdong General Hospital. Baseline data, including demographics, comorbid conditions and carotid ultrasonography, were recorded. The IPN grade was determined using contrast-enhanced ultrasound through the movement of the microbubbles. Univariate and multivariate binary logistic regression analyses were used to evaluate the association between lipoprotein(a) and IPN grade, with stepwise adjustment for covariates including age, sex, comorbid conditions and statin therapy (model 1), total cholesterol, triglyceride, low-density lipoprotein cholesterol calculated by Friedwald's formula, high-density lipoprotein cholesterol, apolipoprotein A and apolipoprotein B (model 2), maximum plaque thickness and total carotid maximum plaque thickness, degree of carotid stenosis and internal carotid artery (ICA) occlusion (model 3). Results Lipoprotein(a) was a significant predictor of higher IPN grade in binary logistic regression before adjusting for other risk factors (odds ratio [OR] 1.238, 95% confidence interval [CI] (1.020, 1.503), P = 0.031). After adjusting for other risk factors, lipoprotein(a) still remained statistically significant in predicting IPN grade in all model. Abstract Aims The aims of the current study were to evaluate the association between anaemia and all‐cause mortality according to chronic kidney disease (CKD) status and to explore at what level of haemoglobin concentration would the all‐cause mortality risk increase prominently among CKD and non‐CKD patients, respectively. Methods and results This is a prospective cohort study, and 1559 patients with ischaemic heart failure (IHF) were included (mean age of 63.5 ± 11.0 years, 85.8% men) from December 2015 to June 2019. Patients were divided into the CKD (n = 481) and non‐CKD (n = 1078) groups based on the estimated glomerular filtration rate of 60 mL/min/1.73 m2. In the CKD group, the incidence rate of all‐cause mortality in anaemic and non‐anaemic patients was 15.4 per 100 person‐years and 10.8 per 100 person‐years, respectively, with an incidence rate ratio of 1.42 (95% confidence interval: 1.00–2.02; P‐value = 0.05). In the non‐CKD group, the incidence rate of all‐cause mortality in anaemic and non‐anaemic patients was 9.8 per 100 person‐years and 5.5 per 100 person‐years, respectively, with an incidence rate ratio of 1.78 (95% confidence interval: 1.20–2.59; P‐value = 0.005). Abstract Background Inherited dilated cardiomyopathy (DCM) contributes to approximately 25% of idiopathic DCM cases, and the proportion is even higher in familial DCM patients. Most studies have focused on familial DCM, whereas the genetic profile of sporadic DCM in Chinese patients remains unknown. Methods Between June 2018 and September 2019, 24 patients diagnosed with idiopathic DCM without a family history were included in the present study. All patients underwent genetic screening for 80 DCM-related genes using targeted next-generation sequencing. Results By in silico analysis, 10 of 99 detected variants were considered pathogenic or likely-pathogenic, including seven TTN truncating variants (TTNtv), one in-frame deletion in TNNT2, one missense mutation in RBM20, and one frameshift deletion variant in FLNC. Of these variants, eight are reported for the first time. Conclusions Using targeted next-generation sequencing, potential genetic causes of idiopathic DCM were identified. Sarcomere mutations remained the most common genetic cause of inherited DCM in this cohort of sporadic Chinese DCM.

ObjectiveWe performed a prospective cohort study to compare the efficacy, safety, effect on mood, and quality of life between lamotrigine (LTG) and oxcarbazepine (OXC) monotherapy among Chinese adult patients with newly-diagnosed focal-onset epilepsy (FOE) with or without secondarily generalized tonic-clonic seizures.MethodsWe enrolled 106 adult patients with new-onset FOE, of whom 56 were in the OXC group and 50 in the LTG group. Their clinical characteristics were detailly recorded especially basic seizure frequency, seizure types, and drug-related adverse events. Efficacy was evaluated as seizure-free (no seizure for 6 months), effective (seizure reduction by more than 50%), and ineffective (seizure reduction by less than 50%). Both intention-to-treat and per-protocol analyses were performed. We also assessed their mood state with the Zung Self-rating Scale for anxiety (Z-SAS) and Zung Self-rating Scale for Depression (Z-SDS) and quality of life (QOL) with Quality of Life in Epilepsy (QOLIE-31) at their baseline visit, 3-month visits, and 6-month visit. Intra-group comparisons in each group and inter-group comparisons between the two groups were made. Correlation analysis and multiple regression analysis were also conducted.ResultsExcept for gender, the two groups were well matched in any other characteristics such as primary seizure frequency and seizure types. In terms of efficacy, 33 patients in the OXC group were evaluated as seizure-free and 15 as effective, while in the LTG group, 31 were seizure-free, and nine were effective. No significant differences could be observed in efficacy between the two groups (P = 0.429). Through multiple logistic regression analysis, we found that OXC monotherapy was more likely to predict a seizure-free state (OR = 1.76) than LTG, but the difference didn't reach statistical significance (P = 0.322) after correcting for other clinical variables. Both groups had adverse events such as fatigue, drowsiness, dizziness, rash, and gastrointestinal discomfort, most of which were mild and transient. In the OXC group, the scores of SAS (P = 0.067) and SDS (P = 0.004) reduced at the 6-month visit, while the score of QOLIE-31 significantly increased (P = 0.001). In the LTG group, a significant decrease in SAS and SDS scores and an increase in QOLIE-31 scores could be witnessed (All P < 0.001). The inter-group comparison showed that improvement of SAS and SDS in the LTG group was more evident than that in the OXC group, which was of statistical significance. Correlational analysis indicated that the improvement of mood and life quality scales in both groups was independent of baseline seizure frequency and treatment efficacy. Multiple linear regression analysis indicated that LTG monotherapy was the only independent factor that could predict a better SAS (P = 0.01) and SDS (P = 0.019) outcome.ConclusionsOXC and LTG are effective as monotherapy and can be considered first-line selection among adult patients with new-onset FOE. Most adverse events are mild, transient, and tolerable. The two drugs improve the mood state of patients, though LTG is superior to OXC in this respect. OXC and LTG have great power in enhancing patients' quality of life. The positive effect on the psychosocial well-being of epilepsy patients may be one of the intrinsic pharmacological properties of LTG and OXC.

With the rapid development of urbanization, fire risk factors have increased greatly, indicating a higher requirement for urban firefighting security. Fire rescue capabilities can be effectively improved by the scientific layout of fire stations, and therefore, the optimal spatial arrangement of fire stations has practical implications for urban safety. In this paper, a method for planning the locations of urban fire stations is presented, taking into account the fire risk points of interest (POIs) data, road networks and fire station planning principles. The combined model method is validated against the nearest facility point model, and the service area model is proposed for the coverage of POIs and regional areas of planned new sites. The efficacy of the model is demonstrated through an improvement in the coverage of crosspoints of the regional area and points of interest (POIs), with increases of 10.20% and 12.43%, respectively. We applied the combined model method to Fengdong New Town, Shaanxi Province, China. A total of 11 new potential sites were proposed to improve the efficiency of spatial coverage, and subsequently, the coverage rate of the POIs and regional area reached 97.66% and 84.80%, respectively. This study provides application guidelines for the decision-making of fire services and the allocation of firefighting resources.

The movement of viruses in aquatic systems is rarely studied over large geographic scales. Oceanic currents, host migration, latitude-based variation in climate, and resulting changes in host life history are all potential drivers of virus connectivity, adaptation, and genetic structure. To expand our understanding of the genetic diversity of Callinectes sapidus reovirus 1 (CsRV1) across a broad spatial and host life history range of its blue crab host (Callinectes sapidus), we obtained 22 complete and 96 partial genomic sequences for CsRV1 strains from the US Atlantic coast, Gulf of Mexico, Caribbean Sea, and the Atlantic coast of South America. Phylogenetic analyses of CsRV1 genomes revealed that virus genotypes were divided into four major genogroups consistent with their host geographic origins. However, some CsRV1 sequences from the US mid-Atlantic shared high genetic similarity with the Gulf of Mexico genotypes, suggesting potential human-mediated movement of CsRV1 between the US mid-Atlantic and Gulf coasts. This study advances our understanding of how climate, coastal geography, host life history, and human activity drive patterns of genetic structure and diversity of viruses in marine animals and contributes to the capacity to infer broadscale host population connectivity in marine ecosystems from virus population genetic data.

Abstract Background Enteral nutrition (EN) therapy is widely used in clinical practice to provide artificial nutrition to patients, while the incidence of adverse events are relatively highly. In the clinical setting, the occurrence of adverse events is associated with the nurse’s risk perception. Thus, using tool to evaluate nurse’s risk perception of enteral nutrition is necessary. Methods The draft questionnaire with 37-items was formed by comprehensive literature reviews and semi-structured in-depth interviews with 11 nurses. Two iterations of expert consultations were used to evaluate the content validity, and 4 items were deleted in this phrase. A 33-items questionnaire was used to survey 352 nurses from five tertiary hospitals in China from May to July 2019 with convenience sampling. Content validity, construct validity and known-groups validity were evaluated by content validity index (CVI), exploratory factor analysis, and the comparisons of the different EN risk perception levels of nurses at different working departments and different educational backgrounds, respectively. Reliability was tested by internal consistency, test-retest reliability, and split-half reliability. Results After the exploratory factor analysis, four items were excluded. Finally, the newly developed questionnaire included 29 items explaining 71.356% of the total variance. It consisted of three factors: Risks of operation (15 items); Risks of EN-related adverse events (11 items), and Risks of EN solution selection (3 items). The CVI of the questionnaire was 0.95 and the CVI of items ranged from 0.875–1.0. The results of known-groups validity showed that the nurses with different educational backgrounds had a statistically significant difference of EN risk perception (z = − 3.024, p = 0.002), whereas there was not significantly different between EN risk perception of nurses working in different departments (z = − 1.644, p = 0.100). The Cronbach’s α, test-retest reliability, and split-half reliability of the questionnaire were 0.967, 0.818, and 0.815, respectively. Conclusions The newly developed questionnaire for assessing nurse’s EN risk perception showed good reliability and validity. It can be used as a tool for nursing managers to assess Chinese nurses’ EN risk perception ability, so as to help to reduce the occurrence of adverse events during EN implementation.

Abstract Phosphoinositide-3 kinase alpha-specific inhibitors (PI3Kαi) displayed promising potential for the treatment of esophageal squamous cell carcinoma (ESCC) with frequent activation in PI3K signaling. However, acquired resistance is likely to develop and limit the efficacy of PI3Kαi like other targeted therapies. To identify genomic adaptation to PI3Kαi, we applied whole-genome sequencing and detected gene mutation and amplification in four lines of ESCC cells established with adapted resistance to a novel PI3Kαi CYH33. Particularly, HRAS G12S mutation was found in KYSE180C cells. Overexpression of HRASG12S in ESCC parental cells rendered resistance to CYH33. By contrast, down-regulation of HRASG12S restored the sensitivity of KYSE180C1 cells to CYH33, and combination of CYH33 and MEK162 displayed synergistic effect against KYSE180C1 cells and xenografts. Furthermore, elevated mTORC1, mitogen-activated protein kinase (MAPK), and c-Myc signaling pathways were found in resistant cells by RNA sequencing and combination of CYH33 and RAD001, MEK162, or OTX015 overcame the resistance to CYH33, which was accompanied with enhanced inhibition on S6, extracellular signal-regulated kinase 1 (ERK), or c-Myc, respectively. Overall, we characterized the adaptations to PI3Kαi in ESCC cells and identified combinatorial regimens that may circumvent resistance.
