Abstract

The interaction of ultraintense laser pulses with solids is largely affected by the plasma gradient at the vacuum-solid interface, which modifies the absorption and ultimately, controls the energy distribution function of heated electrons. A micrometer scale-length plasma has been predicted to yield a significant enhancement of the energy and weight of the fast electron population and to play a major role in laser-driven proton acceleration with thin foils. We report on recent experimental results on proton acceleration from laser interaction with foil targets at ultra-relativistic intensities. We show a three-fold increase of the proton cut-off energy when a micrometer scale-length pre-plasma is introduced by irradiation with a low energy femtosecond pre-pulse. Our realistic numerical simulations agree with the observed gain of the proton cut-off energy and confirm the role of stochastic heating of fast electrons in the enhancement of the accelerating sheath field.

Abstract

In this paper we report the measurement of laser-driven proton acceleration obtained by irradiating nanotube array targets with ultrashort laser pulses at an intensity in excess of 1020 W cm-2. The energetic spectra of forward accelerated protons show a larger flux and a higher proton cutoff energy if compared to flat foils of comparable thickness. Particle-In-Cell 2D simulations reveal that packed nanotube targets favour a better laser-plasma coupling and produce an efficient generation of fast electrons moving through the target. Due to their sub-wavelength size, the propagation of e.m. field into the tubes is made possible by the excitation of Surface Plasmon Polaritons, travelling down to the end of the target and assuring a continuous electron acceleration. The higher amount and energy of these electrons result in turn in a stronger electric sheath field on the rear surface of the target and in a more efficient acceleration of the protons via the target normal sheath acceleration mechanism.

Abstract

X-ray free electron laser (XFEL) sources coupled to high-power laser systems offer an avenue to study the structural dynamics of materials at extreme pressures and temperatures. The recent commissioning of the DiPOLE 100-X laser on the high energy density (HED) instrument at the European XFEL represents the state-of-the-art in combining x-ray diffraction with laser compression, allowing for compressed materials to be probed in unprecedented detail. Here, we report quantitative structural measurements of molten Sn compressed to 85(5) GPa and ∼ 3500 K. The capabilities of the HED instrument enable liquid density measurements with an uncertainty of ∼ 1 % at conditions which are extremely challenging to reach via static compression methods. We discuss best practices for conducting liquid diffraction dynamic compression experiments and the necessary intensity corrections which allow for accurate quantitative analysis. We also provide a polyimide ablation pressure vs input laser energy for the DiPOLE 100-X drive laser which will serve future users of the HED instrument.

Abstract

The construction of a novel Laser driven Light Ions Acceleration Line(L3IA) is progressing rapidly towards the operation, following the recent upgrade of the ILIL-PW laser facility. The Line was designed following the pilot experimental activity carried out earlier at the same facility to define design parameters and to identify main components including target control and diagnostic equipment, also in combination with the numerical simulations for the optimization of laser and target parameters. A preliminary set of data was acquired following the successful commissioning of the laser system >100 TW upgrade. Data include output from a range of different ion detectors and optical diagnostics installed for qualification of the laser-target interaction. Abstract

Recently a new injection scheme for Laser Wake Field Acceleration, employing a single 100-TW-class laser system, has been proposed. In the Resonant Multi-Pulse Ionization injection (ReMPI) a resonant train of pulses drives a large amplitude plasma wave that traps electrons extracted from the plasma by further ionization of a high-Z dopant (Argon in the present paper). While the pulses of the driver train have intensity below the threshold for the dopant's ionization, the properly delayed and frequency doubled (or more) ionization pulse possesses an electric field large enough to extract electrons, though its normalized amplitude is well below unity. In this paper we will report on numerical simulations results aimed at the generation of GeV-scale bunches with normalized emittance and {\it rms} energy below $80\, nm \times rad $ and $0.5\, \%$, respectively. Abstract

The construction of a novel user facility employing laser-driven plasma acceleration with superior beam quality will require an industrial grade, high repetition rate petawatt laser driver which is beyond existing technology. However, with the ongoing fast development of chirped pulse amplification and high average power laser technology, options can be identified depending on the envisioned laser-plasma acceleration scheme and on the time scale for construction. Abstract

A new method for the generation of a train of pulses from a single high-energy, ultra short pulse is presented, suited for Resonant Multi-Pulse Ionization injection. The method is based on different transverse portion of the pulse being delayed by a "mask" sectioned in concentric zones with different thicknesses, in order to deliver multiple laser pulses. The mask is placed right before the last focusing parabola. A hole in the middle of the mask lets part of the original pulse to pass through to drive electron injection. In this paper a full numerical modelling of this scheme is presented. In particular we discuss the spatial and temporal profile of the pulses emerging from the mask and how they are related to the radius and thickness of each section.

Abstract

On the wake of the results obtained so far at the SPARC\_LAB test-facility at the Laboratori Nazionali di Frascati (Italy), we are currently investigating the possibility to design and build a new multi-disciplinary user-facility, equipped with a soft X-ray Free Electron Laser (FEL) driven by a $\sim$1 GeV high brightness linac based on plasma accelerator modules. This design study is performed in synergy with the EuPRAXIA design study. In this paper we report about the recent progresses in the on going design study of the new facility.

Abstract

The interaction of laser pulses with thin grating targets, having a periodic groove at the irradiated surface, has been experimentally investigated. Ultrahigh contrast ($\sim 10^{12}$) pulses allowed to demonstrate an enhanced laser-target coupling for the first time in the relativistic regime of ultra-high intensity $>10^{19} \mbox{W/cm}^{2}$. A maximum increase by a factor of 2.5 of the cut-off energy of protons produced by Target Normal Sheath Acceleration has been observed with respect to plane targets, around the incidence angle expected for resonant excitation of surface waves. Abstract

We report spatially and temporally resolved measurements of magnetic fields generated by petawatt laser-solid interactions with high spatial resolution, using optical polarimetry. The polarimetric measurements map the megagauss magnetic field profiles generated by the fast electron currents at the target rear. The magnetic fields at the rear of a 50 $\mu$m thick aluminum target exhibit distinct and unambiguous signatures of electron beam filamentation. Abstract

This report describes the scientific aims and potentials as well as the preliminary technical design of IRIDE, an innovative tool for multi-disciplinary investigations in a wide field of scientific, technological and industrial applications. IRIDE will be a high intensity 'particle factory', based on a combination of a high duty cycle radio-frequency superconducting electron linac and of high energy lasers. Conceived to provide unique research possibilities for particle physics, for condensed matter physics, chemistry and material science, for structural biology and industrial applications, IRIDE will open completely new research possibilities and advance our knowledge in many branches of science and technology. IRIDE will contribute to open new avenues of discoveries and to address most important riddles: What does matter consist of? What is the structure of proteins that have a fundamental role in life processes? What can we learn from protein structure to improve the treatment of diseases and to design more efficient drugs? But also how does an electronic chip behave under the effect of radiations? How can the heat flow in a large heat exchanger be optimized? The scientific potential of IRIDE is far reaching and justifies the construction of such a large facility in Italy in synergy with the national research institutes and companies and in the framework of the European and international research. It will impact also on R&D work for ILC, FEL, and will be complementarity to other large scale accelerator projects. Abstract

We report on an experimental study on the interaction of a high-contrast 40 fs duration 2.5 TW laser pulse with an argon cluster target. A high-charge, homogeneous, large divergence electron beam with moderate kinetic energy (~2 MeV) is observed in the forward direction. The results show, that an electron beam with a charge as high as 10 nC can be obtained using a table-top laser system. The accelerated electron beam is suitable for a variety of applications such as radiography of thin samples with a spatial resolution better than 100 micron., Comment: 4 pages, 5 figures

Abstract

We discuss a new compact gamma-ray source aiming at high spectral density, up to two orders of magnitude higher than currently available bremsstrahlung sources, and conceptually similar to Compton Sources based on conventional linear accelerators. This new source exploits electron bunches from laser-driven electron acceleration in the so-called self-injection scheme and uses a counter-propagating laser pulse to obtain X and gamma-ray emission via Thomson/Compton scattering. The proposed experimental configuration inherently provides a unique test-bed for studies of fundamental open issues of electrodynamics. A novel X-ray diagnostic of laser-fusion plasmas is described, allowing 2D monochromatic images of hot, dense plasmas to be obtained in any X-ray photon energy range, over a large domain, on a single-shot basis. The device (named Energy-encoded Pinhole Camera - EPiC) is based upon the use of an array of many pinholes coupled to a large area CCD camera operating in the single-photon mode. The available X-ray spectral domain is only limited by the Quantum Efficiency of scientific-grade X-ray CCD cameras, thus extending from a few keV up to a few tens of keV. Spectral 2D images of the emitting plasma can be obtained at any X-ray photon energy provided that a sufficient number of photons had been collected at the desired energy. Results from recent ICF related experiments will be reported in order to detail the new diagnostic.

Abstract

The advance in laser plasma acceleration techniques pushes the regime of the resulting accelerated particles to higher energies and intensities. In particular the upcoming experiments with the FLAME laser at LNF will enter the GeV regime with almost 1pC of electrons. From the current status of understanding of the acceleration mechanism, relatively large angular and energy spreads are expected. There is therefore the need to develop a device capable to measure the energy of electrons over three orders of magnitude (few MeV to few GeV) under still unknown angular divergences. Within the PlasmonX experiment at LNF a spectrometer is being constructed to perform these measurements. It is made of an electro-magnet and a screen made of scintillating fibers for the measurement of the trajectories of the particles. The large range of operation, the huge number of particles and the need to focus the divergence present unprecedented challenges in the design and construction of such a device. The spectrum of relativistic electron bunches with large energy dispersion is hardly obtainable with conventional magnetic spectrometers. We present a novel spectroscopic concept, based on the analysis of the photons generated by Thomson Scattering of a probe laser pulse inpinging with arbitrary incidence angle onto the electron bunch. The feasibility of a single-pulse spectrometer, using an energy-calibrated CCD device as detector, is investigated. Abstract

Background The goal of this study was to investigate the prevalence and factors associated with persistent viral shedding (PVS) in hospitalized patients with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection. Methods This was a prospective observational study including all consecutive adults hospitalized with SARS-CoV-2 infection. When the first nasopharyngeal swab was positive for SARS-CoV-2 RNA (day 0), additional samples were obtained on days + 3, + 5, + 7 and then once every 7 days until virus detection was negative. PVS was defined as the duration of shedding of at least 21 days after diagnosis. The primary endpoint of this study was the prevalence of PVS. Results Data were obtained regarding 121 consecutive hospitalized patients with SARS-CoV-2 infection (median age 66 years, male sex 65.3%). Overall, the prevalence of PVS was 38% (46/121 patients). According to univariate analysis, factors associated with PVS were immunosuppression (6.7% vs 21.7%, p = 0.02), increased interleukin-6 (IL-6) levels (≥ 35 ng/ml) at the time of diagnosis (43.4% vs 67.3%, p = 0.02), time from onset of symptoms to diagnosis (median days 7.0 vs 3.5, p = 0.001), intensive care unit admission (22.7% vs 43.5%, p = 0.02), and need for invasive mechanical ventilation (20.0% vs 41.3%, p = 0.01). The multivariate analysis indicated that immunosuppression, increased IL-6 levels at the time of diagnosis, time from onset of symptoms to diagnosis, and need for mechanical ventilation were independent factors associated with PVS. Conclusions PVS was detected in up to 38% of hospitalized patients with SARS-CoV-2 infection and was strongly associated with immunosuppression, increased IL-6 levels, and the need for mechanical ventilation. Abstract

BACKGROUND Increases in cardiac troponin (cTn) in coronavirus disease 2019 (COVID-19) have been associated with worse prognosis. Nonetheless, data about the significance of cTn in elderly subjects with COVID-19 are lacking. METHODS From a registry of consecutive patients with COVID-19 admitted to a hub hospital in Italy from 25/02/2020 to 03/07/2020, we selected those ≥ 60 year-old and with cTnI measured within three days from the molecular diagnosis of SARS-CoV-2 infection. When available, a second cTnI value within 48 h was also extracted. The relationship between increased cTnI and all-cause in-hospital mortality was evaluated by a Cox regression model and restricted cubic spline functions with three knots. RESULTS Of 343 included patients (median age: 75.0 (68.0−83.0) years, 34.7% men), 88 (25.7%) had cTnI above the upper-reference limit (0.046 µg/L). Patients with increased cTnI had more comorbidities, greater impaired respiratory exchange and higher inflammatory markers on admission than those with normal cTnI. Furthermore, they died more (73.9%vs. 37.3%, P < 0.001) over 15 (6−25) days of hospitalization. The association of elevated cTnI with mortality was confirmed by the adjusted Cox regression model (HR = 1.61, 95%CI: 1.06−2.52, P = 0.039) and was linear until 0.3 µg/L, with a subsequent plateau. Of 191 (55.7%) patients with a second cTnI measurement, 49 (25.7%) had an increasing trend, which was not associated with mortality (univariate HR = 1.39, 95%CI: 0.87−2.22, P = 0.265). CONCLUSIONS In elderly COVID-19 patients, an initial increase in cTn is common and predicts a higher risk of death. Serial cTn testing may not confer additional prognostic information.

Abstract

Background: Chronic Obstructive Pulmonary Disease (COPD) encompasses various phenotypes that severely limit the applicability of precision respiratory medicine. The present investigation is aimed to assess the circadian rhythm of symptoms in pre-defined clinical COPD phenotypes and its association with health-related quality of life (HR-QoL), the quality of sleep and the level of depression/anxiety in each clinical phenotype. Methods: The STORICO (NCT03105999) Italian observational prospective cohort study enrolled COPD subjects. A clinical diagnosis of either chronic bronchitis (CB), emphysema (EM) or mixed COPD-asthma (MCA) phenotype was made by clinicians at enrollment. Baseline early-morning, day-time and nocturnal symptoms (gathered via the Night-time, Morning and Day-time Symptoms of COPD questionnaire), HR-QoL (via the St. George's Respiratory Questionnaire), anxiety and depression levels (via the Hospital Anxiety and Depression Scale), quality of sleep (via COPD and Asthma Sleep Impact Scale), physical activity (via the International Physical Activity Questionnaire) as well as lung function were recorded. Results: 606 COPD subjects (age 71.4 ± 8.2 years, male 75.1%) were studied. 57.9, 35.5 5.3 and 1.3% of the sample belonged to the CB, EM, MCA and EM + CB phenotypes respectively. The vast majority of subjects reported early-morning and day-time symptoms (79.5 and 79.2% in the CB and 75.8 and 77.7% in the EM groups); the proportion suffering from night-time symptoms was higher in the CB than in the EM group (53.6% vs. 39.5%, p = 0.0016). In both CB and EM, indiscriminately, the presence of symptoms during the 24-h day was associated with poorer HR-QoL, worse quality of sleep and higher levels of anxiety/depression. Conclusions: The findings highlight the primary classificatory role of nocturnal symptoms in COPD. Trial registration: Trial registration number: NCT03105999, date of registration: 10th April 2017.

Abstract

The Horizon 2020 Project EuPRAXIA (European Plasma Research Accelerator with eXcellence In Applications) is producing a conceptual design report for a highly compact and cost-effective European facility with multi-GeV electron beams accelerated using plasmas. EuPRAXIA will be set up as a distributed Open Innovation platform with two construction sites, one with a focus on beam-driven plasma acceleration (PWFA) and another site with a focus on laser-driven plasma acceleration (LWFA). User areas at both sites will provide access to FEL pilot experiments, positron generation and acceleration, compact radiation sources, and test beams for HEP detector development. Support centres in four different countries will complement the pan-European implementation of this infrastructure., Proceedings of the 10th Int. Abstract

Background Chronic Obstructive Pulmonary Disease (COPD) encompasses various phenotypes that severely limit the applicability of precision respiratory medicine. The present investigation is aimed to assess the circadian rhythm of symptoms in pre-defined clinical COPD phenotypes and its association with health-related quality of life (HR-QoL), the quality of sleep and the level of depression/anxiety in each clinical phenotype. Methods The STORICO (NCT03105999) Italian observational prospective cohort study enrolled COPD subjects. A clinical diagnosis of either chronic bronchitis (CB), emphysema (EM) or mixed COPD-asthma (MCA) phenotype was made by clinicians at enrollment. Baseline early-morning, day-time and nocturnal symptoms (gathered via the Night-time, Morning and Day-time Symptoms of COPD questionnaire), HR-QoL (via the St. George’s Respiratory Questionnaire), anxiety and depression levels (via the Hospital Anxiety and Depression Scale), quality of sleep (via COPD and Asthma Sleep Impact Scale), physical activity (via the International Physical Activity Questionnaire) as well as lung function were recorded. Results 606 COPD subjects (age 71.4 ± 8.2 years, male 75.1%) were studied. 57.9, 35.5 5.3 and 1.3% of the sample belonged to the CB, EM, MCA and EM + CB phenotypes respectively. The vast majority of subjects reported early-morning and day-time symptoms (79.5 and 79.2% in the CB and 75.8 and 77.7% in the EM groups); the proportion suffering from night-time symptoms was higher in the CB than in the EM group (53.6% vs. 39.5%, p = 0.0016). In both CB and EM, indiscriminately, the presence of symptoms during the 24-h day was associated with poorer HR-QoL, worse quality of sleep and higher levels of anxiety/depression. Conclusions The findings highlight the primary classificatory role of nocturnal symptoms in COPD. Trial registration Trial registration number: NCT03105999, date of registration: 10th April 2017.

Abstract

The SARS-CoV-2 infection causes severe respiratory involvement (COVID-19) in 5–20% of patients through initial immune derangement, followed by intense cytokine production and vascular leakage. Evidence of immune involvement point to the participation of T, B, and NK cells in the lack of control of virus replication leading to COVID-19. NK cells contribute to early phases of virus control and to the regulation of adaptive responses. The precise mechanism of NK cell dysregulation is poorly understood, with little information on tissue margination or turnover. We investigated these aspects by multiparameter flow cytometry in a cohort of 28 patients hospitalized with early COVID-19. Relevant decreases in CD56brightCD16+/- NK subsets were detected, with a shift of circulating NK cells toward more mature CD56dimCD16+KIR+NKG2A+ and “memory” KIR+CD57+CD85j+ cells with increased inhibitory NKG2A and KIR molecules. Impaired cytotoxicity and IFN-γ production were associated with conserved expression of natural cytotoxicity receptors and perforin. Moreover, intense NK cell activation with increased HLA-DR and CD69 expression was associated with the circulation of CD69+CD103+ CXCR6+ tissue-resident NK cells and of CD34+DNAM-1brightCXCR4+ inflammatory precursors to mature functional NK cells. Severe disease trajectories were directly associated with the proportion of CD34+DNAM-1brightCXCR4+ precursors and inversely associated with the proportion of NKG2D+ and of CD103+ NK cells. Intense NK cell activation and trafficking to and from tissues occurs early in COVID-19, and is associated with subsequent disease progression, providing an insight into the mechanism of clinical deterioration. Strategies to positively manipulate tissue-resident NK cell responses may provide advantages to future therapeutic and vaccine approaches., Author summary This is a detailed study of activating and inhibitory receptors in NK cells of COVID-19 patients when first admitted to the hospital for respiratory insufficiency. NK cells are known to be the first line of defense against invading viruses, and regulate downstream B and T cell responses, including antibody production. We observed intense NK cell activation with decreased functional activity, as well as intense circulation of putative tissue resident CD69+CD103+CXCR6+ NK cells, with a related surge in inflammatory CD34+ precursors from the bone marrow. The findings suggest that there is unprecedented trafficking of NK cells from peripheral tissues, their increased death with recruitment of emergency precursors from the bone marrow, and a relationship with the subsequent course of the disease of the patients. This in turn suggests possible areas of treatment and prevention.
