Abstract

International audience; Urine is a highly suitable biological matrix for metabolomics studies. Total collection for 24-h periods is the gold standard as it ensures the presence of all metabolites excreted throughout the day. However, in animal studies, it presents limitations related to animal welfare and also due to alterations of the metabolome originating from the use of acid for preventing microbial growth or microbial contamination. In this study, we investigated whether spot urine collection is a practical alternative to total collection for metabolomic studies in lactating cows. For this purpose, we collected urine samples from 4 lactating Holstein cows fed 4 diets in a 4 × 4 Latin square design. Urine was collected for 24 h using a collecting device (i.e., total collection) or collected once per day 4 h after the morning feeding (i.e., spot urine collection). Dietary treatments differed by the amount of nitrogen content (high vs. low) and by the nature of the energy (starch vs. fiber). Urine metabolome was analyzed by 2 untargeted complementary methods, nuclear magnetic resonance and hydrophilicinteraction liquid chromatography (HILIC) coupled to a time-of-flight mass spectrometer, and by 1 targeted method, HILIC–tandem mass spectrometry. Although sampling technique had an effect on the abundance of metabolites detected, spot urine samples were equally capable of showing differences in urine metabolomethan samples from total collection. When considering nitrogen levels in the diet, the robustness and precision for discriminating high- and low-nitrogen dietswas equally achieved with both sampling techniques. A total of 22 discriminant metabolites associated with the N level of diets were identified from untargeted HILIC coupled to a time-of-flight mass spectrometer (n = 9) and nuclear magnetic resonance (n = 11), and 2 from targeted HILIC–tandem mass spectrometry. Alternatively, starch or fiber in the diet induced less changes in the metabolome that were not clearly discriminated independently of the sampling technique. We concluded that spot urine collection can successfully reveal differences in the urine metabolome elicited by dietary N levels and be used as a substitute of total urinary 24-h collection for metabolomic studies.

International audience; Purpose: Breast cancer is a major cause of death in occidental women. Mechanisms involved in its etiology remain misunderstood. Metabolomics is a powerful tool which may help elucidating novel biological pathways and identify new biomarkers in order to predict breast cancer well before symptoms appear. The aim of this study was to investigate whether untargeted metabolomic signatures from blood draws of healthy women could contribute to better understand and predict the long-term risk of developing breast cancer. Abstract

Contexte et objectifs : La métabolomique permet d’étudier l’ensemble des métabolites présents dans un biofluide. Cette étude basée sur une analyse non ciblée en spectrométrie de masse a pour but d’étudier si des profils métabolomiques établis à partir d’un prélèvement sanguin sur des femmes a priori en bonne santé, pourrait contribuer à prédire le risque à long terme de développer le cancer du sein et d’améliorer la compréhension de son étiologie. Méthodes : Un cas-témoin niché prospectif a été mené dans la cohorte SU.VI.MAX, incluant 211 cas de cancer du sein et 211 témoins appariés. Les profils métabolomiques ont été établis sur des échantillons de plasma prélevés à l’inclusion, avant l’apparition du cancer, grâce à une analyse non ciblée par spectrométrie de masse (LC-MS). Des modèles de régressions logistiques conditionnelles multivariées ont été calculés sur chaque ion présélectionné grâce à une ANOVA et sur les combinaisons de potentiels biomarqueurs issus d’une Analyse en Composante Principale. Une correction de test multiple de type FDR (False Discovery Rate) a été appliquée. Résultats : Plusieurs métabolites étaient associés au risque de cancer du sein. La finalisation de l’identification des métabolites discriminants est en cours. Au vu des résultats actuels, les femmes caractérisées par un niveau plasmatique plus élevé de valine (OR=1,45 [1,15-1,83] ; p= 0,009), de phénylalanine (OR=1,43 [1,14-1,78] ; p=0,009), de tryptophane (OR=1,4 [1,1-1,79] ; p=0,01), et de glutamine (OR=1,33 [1,07-1,66] ; p=0,01) et d’un niveau plus bas d’O-succinyl-L-homosérine (OR=0,7 [0,55-0,89] ; p=0,01) auraient un risque plus important de développer un cancer du sein dans la décennie qui suit. Conclusions et perspectives : Cette étude suggère plusieurs associations entre profils métabolomiques à l’inclusion et risque à long terme de développer le cancer du sein et confirme certains résultats que nous avons mis en évidence par une analyse RMN. Cette étude pourrait aider à améliorer l’identification des femmes à risque de développer un cancer du sein, bien avant l’apparition des symptômes. L’étude des biomarqueurs nutritionnels associés au risque de développer cette pathologie (actuellement en cours) pourra permettre de préciser le rôle de la nutrition (facteur modifiable) dans l’étiologie du cancer du sein et d’améliorer les stratégies préventives.

26. Optimisation of the downstream logistics of a division: : a case study

Abstract

The drop of oil prices has led to a significant decrease in the demand in oil-related industries, and thus to severe competition in the market. In this context, the companies are striving to reduce their costs and lead times in order to remain attractive to the customers. While companies are used to challenging the production costs and lead times, and are aware of the profits that can be drawn from such studies, logistics aspects being more difficult to assess, they are less often dealt with.This thesis aims at building a model of the costs and lead times involved in downstream logistics in order to understand the impact of downstream logistics on the global costs and lead times of a product. It also aims at providing a method to allocate the customer demand to the division’s mills in order to optimise either the downstream logistics costs or the lead times.This thesis was performed in the Oil and Gas division of an industrial French company. This division’s scope is the oil companies in Europe, Africa, the Middle East and Asia. Ten mills were considered in this study, in Germany, France, the United Kingdom, China, Nigeria, Saudi Arabia, Brazil and Indonesia.The context of the company and the concepts related to downstream logistics were investigated. Then, workgroups consisting of logisticians and the author of this thesis were organised to build a model for the costs and lead times of downstream logistics. This model was used to analyse a Sales and Operation Planning, and to optimise the allocations of the customer demand to the mills from a logistics perspective.The results show that the downstream logistics costs and lead times are very sensitive to the choice of allocations to the mills. The integration of the production is the main factor in the minimisation of the logistics costs, i.e. the downstream logistics costs are at the lowest when the manufacturing mill is the supplying mill. On the other hand, the main factor in the minimisation of the logistics lead times is the proximity between the manufacturer and the country of the customer.This thesis shows that the logistics budget of the division vary significantly depending on the choice of the supplier and the manufacturer. Besides, the optimisation of the costs and the optimisation of the lead times have opposed effects, so both optimisations cannot occur at the same time. Reduktionen i oljepriserna har lett till en betydande minskning av efterfrågan på oljerelaterade industrier och således råder en hög konkurrens på marknaden. I denna omgivning strävar företagen efter kostnad- och ledtidsreducering för att förbli attraktiva för kunderna. Medan företagen vanligtvis utmanar produktionskostnader och ledtider, samt är medvetna om de vinster som kan dras av sådana studier, är logistikaspekter svårare att bedöma då de sällan behandlas.Denna avhandling syftar till att bygga en modell av hur kostnader och ledtider involveras i nedströms logistik. Detta för att förstå effekterna av nedströmslogistik för globala kostnader och ledtider för en produkt. Modellen syftar även till att skapa en metod som fördelar kundefterfrågan på divisionens fabriker för att optimera antingen nedströmslogistikkostnaderna eller ledtiderna.Denna avhandling utfördes i en olje- och gasdivision inom ett franskt industriellt företag. Divisionsomfattningen är oljebolagen i Europa, Afrika, Mellanöstern och Asien. I denna studie har det tagits hänsyn till tio olika fabriker i Tyskland, Frankrike, Storbritannien, Kina, Nigeria, Saudiarabien, Brasilien och Indonesien.Företagsbakgrunden och nedströmslogistik-relaterade koncept undersöktes. Därefter organiserades arbetsgrupper bestående av logistiker och författaren till denna avhandling för att bygga en modell för kostnader och ledtider för nedströmslogistik. Denna modell användes sedan för att analysera försäljning och driftsplanering, samt för att optimera fördelningen av kundernas efterfrågan till fabriker utifrån logistikperspektiv.Resultaten visar att nedströmslogistikkostnader och ledtiderna är mycket känsliga för valet av fördelning till fabriker. Integrationen av produktionen är den viktigaste faktorn i minimering av logistikkostnader; nedströmslogistikkostnader är lägst när tillverkningsfabriken är den levererande fabriken. Samtidigt är den viktigaste faktorn för minimering av logistikens ledtider närheten mellan tillverkaren och landet där kunden vistas.Denna avhandling visar att divisionens logistikbudget varierar kraftigt beroende på valet av leverantör och tillverkare. Dessutom har optimering av kostnaderna och optimering av ledtider motsatta effekter, vilket resulterar i att båda optimeringarna inte kan inträffa samtidigt.
