Background: Drug promotional literatures (DPLs) are one of the well-known promotional activities of pharmaceutical industries which are sometimes inaccurate as well as of poor educational value. Angiotensin receptor blockers (ARBs) are one of the most commonly used antihypertensives. Therefore, this study was done to estimate the accuracy of DPLs on ARBs as per the World Health Organization (WHO) criteria. Aims and Objectives: The aims of this study were to estimate the accuracy of DPLs on ARBs as per the WHO and to estimate the DPLs for types of claims and appropriateness of claims. Materials and Methods: A cross-sectional observational study was carried out for 1 month. All the required information of selected DPLs on ARBs were recorded in a pro forma and were evaluated according to the WHO criteria. Results: In this study, a total of 20 (twenty) DPLs were evaluated only on ARBs. It was observed that none of the DPLs fulfilled all the WHO criteria. In this study, some DPLs made multiple claims, as much as five per DPL. Claims were, further, analyzed and divided into appropriate and inappropriate. We have observed that 65.96% claims were appropriate and 34.04% claims were inappropriate. Conclusion: This type of study can contribute to make prescribing practices rational as promotional activities influence the prescribing behavior of the health-care provider.

Rhizoctonia solani is a soil-borne fungal plant pathogen causing damping-off of economically important crops. The pathogen is generally controlled with fungicides which can be toxic to the environment. Plants are abundant and renewable sources of natural antimicrobial chemicals and their phytochemicals are receiving increasing attention as broad-based “green pesticides". 28 plant extracts consisting of essential oils and vegetable oils, and six chemicals derived from plant essential oils were evaluated for antifungal activity against R. solani using in vitro poisoned food technique and hyphal extension bioassay. Several plant extracts including allylanisole, allyl isothiocyanate, anethole, anise oil, and lemon eucalyptus oil were found to be potent inhibitors of R. solani growth in both the in vitro assays. Selected plant chemicals showing strong in vitro antagonistic potentials were tested for control of cucumber seedling damping-off caused by R. solani. In the greenhouse trials, allyl isothiocynate, geraniol, lemongrass, clove, origanum, pennyroyal, and peppermint oils as soil amendments at 0.3% (v/w) suppressed damping-off by >70%, which compared well with a commercial fungicide Banrot® 40WP demonstrating their potentials as safer disease management alternatives. Foodborne pathogens such as Escherichia coli O157:H7, Salmonella spp., Listeria monocytogenes are responsible for frequent occurrences of illness and mortality in humans. Several economically important plant diseases are caused by pathogenic bacteria. Economically important plant diseases and post-harvest losses and decay are also incited by plant pathogens such as Erwinia, Pectobacterium, Botrytis, and Pseudomonas spp. This paper discusses the potential of predatory bacteria for biocontrol of foodborne and plant pathogens with emphasis on the deltaproteobacteria group. Deltaproteobacteria (Bdellovibrio and Bacteriovorax sp. as well as Daptobacter, and Myxobacteria) are Gramnegative predatory microorganisms which prey mainly on Gram-negative bacteria and occur in diverse soil, marine and fresh water ecologies. The predation of Gram-negative bacteria by Bdellovibrio and Bdellovibrio-like organisms (BALOs) may enhance their potential application for biocontrol of foodborne and plant pathogens. The difficulty in controlling foodborne and plant pathogenic bacteria is due to the paucity of available bactericidal chemicals and regulations limiting the use of antibiotics in pathogen control. In this review, the types and mechanisms of predation by predatory bacteria (PB) are discussed with respect to potential utilization as biocontrol agents. Their diversity and relationships is highlighted by genomic research. Potential for biocontrol of foodborne and plant pathogens by PB is discussed in terms of attributes and limitations. Although research in animal systems (Salmonella/chicken) indicates significant biocontrol potential when PB are applied as probiotics, their application to foodborne and plant pathogens are limited. With increased research in metagenomics on PB effects on animal cells /tissues, a better understanding of regulation of cellular metabolism and their association with growth of PB and prey degradation will ultimately enhance the utility of PB as biocontrol agents of foodborne and plant pathogens. Rhizoctonia blight is a common and serious disease of many turfgrass species. The most widespread causal agent, Thanatephorus cucumeris (anamorph: R. solani), consists of several genetically different subpopulations. In addition, Waitea circinata varieties zeae, oryzae and circinata (anamorph: Rhizoctonia spp.) also can cause the disease. Accurate identification of the causal pathogen is important for effective management of the disease. It is challenging to distinguish the specific causal pathogen based on disease symptoms or macroscopic and microscopic morphology. Traditional methods such as anastomosis reactions with tester isolates are time consuming and sometimes difficult to interpret. In the present study universally primed PCR (UP-PCR) fingerprinting was used to assess genetic diversity of Rhizoctonia spp. infecting turfgrasses. Eighty-four Rhizoctonia isolates were sampled from diseased turfgrass leaves from seven distinct geographic areas in Virginia and Maryland. Rhizoctonia isolates were characterized by ribosomal DNA internal transcribed spacer (rDNA-ITS) region and UP-PCR. The isolates formed seven clusters based on ITS sequences analysis and unweighted pair group method with arithmetic mean (UPGMA) clustering of UP-PCR markers, which corresponded well with anastomosis groups (AGs) of the isolates. Isolates of R. solani AG 1-IB (n = 18), AG 2-2IIIB (n = 30) and AG 5 (n = 1) clustered separately. Waitea circinata var. zeae (n = 9) and var. circinata (n = 4) grouped separately. A cluster of six isolates of Waitea (UWC) did not fall into any known Waitea variety. The binucleate Rhizoctonia-like fungi (BNR) (n = 16) clustered into two groups. Rhizoctonia solani AG 2-2IIIB was the most dominant pathogen in this study, followed by AG 1-IB. There was no relationship between the geographic origin of the isolates and clustering of isolates based on the genetic associations. To our knowledge this is the first time UP-PCR was used to characterize Rhizoctonia, Waitea and Ceratobasidium isolates to their infra-species level. [ABSTRACT FROM AUTHOR]

Abstract Background The development of plant gene transfer systems has allowed for the introgression of alien genes into plant genomes for novel disease control strategies, thus providing a mechanism for broadening the genetic resources available to plant breeders. Using the tools of plant genetic engineering, a broad-spectrum antimicrobial gene was tested for resistance against head blight caused by Fusarium graminearum Schwabe, a devastating disease of wheat (Triticum aestivum L.) and barley (Hordeum vulgare L.) that reduces both grain yield and quality. Results A construct containing a bovine lactoferrin cDNA was used to transform wheat using an Agrobacterium-mediated DNA transfer system to express this antimicrobial protein in transgenic wheat. Transformants were analyzed by Northern and Western blots to determine lactoferrin gene expression levels and were inoculated with the head blight disease fungus F. graminearum. Transgenic wheat showed a significant reduction of disease incidence caused by F. graminearum compared to control wheat plants. The level of resistance in the highly susceptible wheat cultivar Bobwhite was significantly higher in transgenic plants compared to control Bobwhite and two untransformed commercial wheat cultivars, susceptible Wheaton and tolerant ND 2710. Quantification of the expressed lactoferrin protein by ELISA in transgenic wheat indicated a positive correlation between the lactoferrin gene expression levels and the levels of disease resistance. Conclusions Introgression of the lactoferrin gene into elite commercial wheat, barley and other susceptible cereals may enhance resistance to F. graminearum.

Cercospora leaf spot (CLS) is a destructive disease limiting sugar beet production and is managed using resistant cultivars, crop rotation, and timely applications of effective fungicides. Since 2016, its causal agent, Cercospora beticola , has been reported to be resistant to quinone outside inhibitors (QoIs) and to have reduced sensitive to demethylation inhibitors (DMIs) in sugar beet growing areas in North Dakota and Minnesota. Isolates of C. beticola resistant to QoIs, DMIs, and both QoIs and DMIs were collected from fields in Foxhome, Minnesota, in 2017. Fitness of these resistant isolates was compared with that of QoI- and DMI-sensitive isolates in laboratory and greenhouse studies. In the lab, mycelial growth, spore production, and spore germination were measured. The results showed that resistant isolates had significantly less mycelial growth and spore production than sensitive isolates, while no significant difference in spore germination was detected. In the greenhouse, six leaf-stage sugar beets were inoculated with a spore suspension made from each resistant group and incubated in separate humidity chambers. CLS disease severity was evaluated visually at 7, 14, and 21 days after inoculation (DAI), and the areas under disease progress curve (AUDPC) were calculated. Resistant isolates had significantly smaller AUDPC but still caused as high disease severity as the sensitive ones at 21 DAI. Although QoI- and/or DMI-resistant isolates had a relatively slower disease development, they still caused high disease severity and need to be factored in disease management practices., Competing Interests: The author(s) declare no conflict of interest.

Rabies is a fatal encephalomyelitis mainly transmitted to humans and other animals by rabid dog bites. Hence, vaccination programs are being instituted for the control of rabies in dogs. Though stray dogs have been vaccinated for years under various programs initiated for control of the disease, the effectiveness of these programs can be ascertained only by assessing the immunity of these dogs. With this in view, a study was conducted to assess the effectiveness of the ongoing mass dog vaccination (MDV) program by the Bengaluru City Municipal Corporation, Bengaluru, India. Whole blood and serum samples (n = 260) from vaccinated stray dogs in 26 wards of 8 corporation zones were tested by rapid fluorescent focus inhibition test (RFFIT) as well as an in-house quantitative indirect enzyme-linked immunosorbent assay (iELISA) for a humoral response and by interferon-gamma (IFN-γ) ELISA for a cellular response. As determined by the cut-off value of 0.5 IU/mL of serum, 71% and 87% of the samples from vaccinated dogs revealed adequate levels of antibodies presumed to confer protection by RFFIT and iELISA, respectively. The sensitivity and specificity of the iELISA were 100% and 63.3%, respectively. The IFN-γ ELISA revealed adequate cellular response in 50% of the samples. The quantitative iELISA was found to be useful in large-scale seromonitoring of MDV programs to aid in the elimination of dog-mediated rabies.

Rabies is a progressively fatal viral disease affecting a wide variety of warm-blooded animals and human beings. With cattle being major part of Indian livestock population, rabies can result in significant financial losses. Immunization of livestock vulnerable to exposure is the best way to control rabies. The present study was undertaken to investigate the efficacy of a rabies pre-exposure prophylactic vaccine administered through different routes and to sequentially monitor the levels of rabies virus-neutralizing antibody (RVNA) titers in cattle. Thirty cattle were divided into five groups of six animals each. Group I and III animals were immunized with 1 mL and 0.2 mL of rabies vaccine through intramuscular (IM) and intradermal (ID) routes, respectively, on day 0, with a booster dose on day 21; Group II and IV animals were immunized with 1 mL and 0.2 mL of rabies vaccine, respectively, without the booster dose; unvaccinated animals served as a control (Group V). Serum samples were collected on days 0, 14, 28, and 90 to estimate RVNA titers using the rapid fluorescent focus inhibition test (RFFIT). The titers were above an adequate level (≥0.5 IU/mL) on day 14 and maintained up to 90 days in all animals administered the rabies vaccine through the IM and ID route with or without a booster dose. The study indicated that both routes of vaccination are safe and effective in providing protection against rabies. Hence, both routes can be considered for pre-exposure prophylaxis. However, the ID route proved to be more economical due to its dose-sparing effect.

Chilli is an important commercial crop grown in tropical and subtropical climates. The whitefly-transmitted chilli leaf curl virus (ChiLCV) is a serious threat to chilli cultivation. Vector migration rate and host-vector contact rate, the major drivers involved in the epidemic process, have been pinpointed to link management. The complete interception of migrant vectors immediately after transplantation has been noted to increase the survival time (to remain infection free) of the plants (80%) and thereby delay the epidemic process. The survival time under interception (30 days) has been noted to be nine weeks ( p < 0.05), as compared to five weeks, which received a shorter period of interception (14-21 days). Non-significant differences in hazard ratios between 21- and 30-day interceptions helped optimize the cover period to 26 days. Vector feeding rate, estimated as a component of contact rate, is noted to increase until the sixth week with host density and decline subsequently due to plant succulence factor. Correspondence between the peak time of virus transmission or inoculation rate (at 8 weeks) and contact rate (at 6 weeks) suggests that host succulence is of critical importance in host-vector interactions. Infection proportion estimates in inoculated plants at different leaf stages have supported the view that virus transmission potential with plant age decreases, presumably due to modification in contact rate. The hypothesis that migrant vectors and contact rate dynamics are the primary drivers of the epidemic has been proved and translated into rules to guide management strategies.

The aim of the current study was to prepare the efficacious amorphous solid dispersion of poorly water-soluble compound, Dolutegravir. After theoretical and experimental determination of drug-polymer miscibility, polyvinyl caprolactam-polyvinyl acetate-polyethylne glycol graft copolymer was chosen as a polymer. The solid dispersions of Dolutegravir were prepared by quench cooling and solvent evaporation method. Though quench cooling successfully stabilized the drug into amorphous form, solvent evaporation technique failed to render the drug completely amorphous. Owing to the negative Gibbs free energy at room temperature, the prepared dispersions were found stable at room temperature for 60 days. To resolve the overlapping contribution of micellar solubilization and amorphicity in improving the dissolution characteristics of Dolutegravir, the in vitro dissolution studies were performed in USP phosphate buffer as well as bio-relevant media. The dissolution advantage between prepared dispersions and pure drug in USP phosphate buffer was found bridged in the bio-relevant media. For this, the micellar solubilization driven dissolution of Dolutegravir in the presence of bile and lecithin micelles was thought as a contributing factor. Nevertheless, the dissolution advantage of dispersions prepared by quench cooling method was found endured in FeSSIF, which was thought to be due to its amorphicity leading to molecular level dissolution. Subsequently, the dissolution advantage was translated into the improved flux. Further, in vivo oral bioavailability was investigated for the dispersion prepared by quench cooling by using crystalline Dolutegravir as a control. The overall exposure of Dolutegravir was improved by 1.7 fold (AUC), while the maximum plasma concentration (Cmax) demonstrated 2 fold increase after comparing with crystalline Dolutegravir., (Copyright © 2019 Elsevier B.V. This study investigated the dissolution behavior of BCS class II ionizable weak base Saquinavir and its mesylate salt in the multi-compartment transfer setup employing different composition of dissolution media. The dissolution behavior of Saquinavir was studied by using a two-compartment transfer model representing the transfer of drug from the stomach (donor compartment) to the upper intestine (acceptor compartment). Various buffers like phosphate, bicarbonate, FaSSIF, and FeSSIF were employed. The dissolution was also studied in the concomitant presence of the additional solute, i.e., Quercetin. Further, the dissolution profiles of Saquinavir and its mesylate salt were simulated by GastroPlus TM , and the simulated dissolution profiles were compared against the experimental ones. The formation of in situ HCl salt and water-soluble amorphous phosphate aggregates was confirmed in the donor and acceptor compartments of the transfer setup, respectively. As the consequence of the lower solubility product of HCl salt of Saquinavir, the solubility advantage of mesylate salt was vanished leading to the lower than the predicted dissolution in the acceptor compartment. However, the formation of water-soluble aggregates in the presence of the phosphate salts was observed leading to the higher than the predicted dissolution of the free base in the transfer setup. Interestingly, the formation of such water-soluble aggregates was found to be hindered in the concomitant presence of an ionic solute resulting in the lower dissolution rates. The in situ generation of salts and aggregates in the transfer model lead to the inconsistent prediction of dissolution profiles by GastroPlus TM .

Eight fungal isolates (ELRF 1 to 8) were recovered from necrotic roots of Easter lilies, Lilium longiflorum cv. Nellie White, grown in a field in the U.S. Pacific Northwest. The eight fungal isolates were identified by sequencing and molecular phylogenetic analyses based on their ITS rDNA region. Five isolates were identified as Fusarium oxysporum, two as F. tricinctum, and one as Rhizoctonia sp. AG-I. This constitutes the first report of Rhizoctonia sp. AG-I infecting lilies worldwide and the first report of F. tricinctum infecting lilies in the United States. To study and validate their pathogenicity, pure cultures of each isolate were used to infect the roots of Easter lily plants growing in vitro. In addition, Easter lily plants growing in vitro were infected either with or without Pratylenchus penetrans, the root lesion nematode, prior to placing a culture plug of fungus 1 cm from a lily root. Pratylenchus penetrans is a nematode species commonly found in the sampled fields. The presence of both nematode and Rhizoctonia sp. AG-I isolate ELRF 3 in infected lilies was evaluated by molecular analyses, confirming the infection of roots 3 days after inoculation, prior to development of disease symptoms. Necrosis and root rot developed more rapidly with all eight fungal isolates when there had been prior infection with P. penetrans, the major nematode parasitizing Easter lily roots in the field in Oregon.
