Population size has increasingly been taken as the driver of past human environmental impact worldwide, and particularly in the Arctic. However, sedimentary ancient DNA (sedaDNA), pollen and archaeological data show that over the last 12,000 years, paleoeconomy and culture determined human impacts on the terrestrial ecology of Arctic Norway. The large Mortensnes site complex (Ceavccageađgi, 70°N) has yielded the most comprehensive multiproxy record in the Arctic to date. The site saw occupation from the Pioneer period (c. 10,000 cal. years BP) with more intensive use from c. 4,200 to 2,000 cal. years BP and after 1,600 cal. years BP. Here, we combine on-site environmental archaeology with a near-site lake record of plant and animal sedaDNA. The rich animal sedaDNA data (42 taxa) and on-site faunal analyses reveal switches in human dietary composition from early-Holocene fish + marine mammals, to mixed marine + reindeer, then finally to marine + reindeer + domesticates (sheep, cattle, pigs), with highest reindeer concentrations in the last millennium. Archaeological evidence suggests these changes are not directly driven by climate or variation in population densities at the site or in the region, but rather are the result of changing socio-economic activities and culture, probably reflecting settlers’ origins. This large settlement only had discernable effects on its hinterland in the last 3,600 years (grazing) and more markedly in the last 1,000 years through reindeer keeping/herding and, possibly domestic stock. Near-site sedaDNA can be linked to and validate the faunal record from archaeological excavations, demonstrating that environmental impacts can be assessed at a landscape scale.

What drives ecosystem buildup, diversity, and stability? We assess species arrival and ecosystem changes across 16 millennia by combining regional-scale plant sedimentary ancient DNA from Fennoscandia with near-complete DNA and trait databases. We show that postglacial arrival time varies within and between plant growth forms. Further, arrival times were mainly predicted by adaptation to temperature, disturbance, and light. Major break points in ecological trait diversity were seen between 13.9 and 10.8 calibrated thousand years before the present (cal ka BP), as well as break point in functional diversity at 12.0 cal ka BP, shifting from a state of ecosystem buildup to a state where most habitat types and biotic ecosystem components were in place. Trait and functional diversity stabilized around 8 cal ka BP, after which both remained stable, although changes in climate took place and species inflow continued. During the last glacial–interglacial cycle, Arctic biotas experienced substantial climatic changes, yet the nature, extent and rate of their responses are not fully understood1–8. Here we report a large-scale environmental DNA metagenomic study of ancient plant and mammal communities, analysing 535 permafrost and lake sediment samples from across the Arctic spanning the past 50,000 years. Furthermore, we present 1,541 contemporary plant genome assemblies that were generated as reference sequences. Our study provides several insights into the long-term dynamics of the Arctic biota at the circumpolar and regional scales. Our key findings include: (1) a relatively homogeneous steppe–tundra flora dominated the Arctic during the Last Glacial Maximum, followed by regional divergence of vegetation during the Holocene epoch; (2) certain grazing animals consistently co-occurred in space and time; (3) humans appear to have been a minor factor in driving animal distributions; (4) higher effective precipitation, as well as an increase in the proportion of wetland plants, show negative effects on animal diversity; (5) the persistence of the steppe–tundra vegetation in northern Siberia enabled the late survival of several now-extinct megafauna species, including the woolly mammoth until 3.9 ± 0.2 thousand years ago (ka) and the woolly rhinoceros until 9.8 ± 0.2 ka; and (6) phylogenetic analysis of mammoth environmental DNA reveals a previously unsampled mitochondrial lineage. Our findings highlight the power of ancient environmental metagenomics analyses to advance understanding of population histories and long-term ecological dynamics.

This thesis explores the potential of sedimentary ancient DNA (sedaDNA) from lakes as a tool for studying past communities. First it sets out to determine what taxa in the environment are most likely to be detected in lake sediments. For this, vegetation surveys were carried out around eleven different lakes in Norway and the taxa detected based on plant metabarcoding of the lake sediments were compared to the recorded flora. The taxa detected in the lake sediments primarily resembled those growing either in the lake or within the immediate surrounding. Furthermore, taxa abundant in the vegetation were more likely to be detected based on DNA. Three different studies were carried out to compare plant sedaDNA results to traditional palaeoecological proxies such as pollen or macrofossils. The results indicate that overall sedaDNA is capable of detecting more taxa than the alternative methods, though differences are observed between the shotgun and metabarcode methods tested, as well as the lakes themselves. A separate study aimed to detect mammals based on sedaDNA, which proved to be more problematic. Co-amplification of off-target taxa, the presence of common lab contaminants and a general low amount of template material hindered the identification of mammals present. However, the detection of “bycatch” worm taxa demonstrates their potential for palaeoenvironmental reconstructions. Finally, the shotgun sequencing of two algal rich sediment layers allowed for both the reconstruction of the algal organelle genomes, as well as the exploration of the present within lake populations.

Pollen, botanical macrofossils, chemical components and ancient DNA were studied in samples from the rumen of a frozen Yakutian bison (Bison priscus) that lived ca. 10,500 cal a BP. The dialkyl glycerol ether lipid archaeol (2,3-di-O-phytanyl-sn-glycerol) was detected and is interpreted to have been derived from methanogenic Archaea in the rumen. This is the oldest reported occurrence of archaeol attributed to digestive tract Archaea. Remains of shrubs (Alnus, Betula, Salix) and Poaceae indicate that the animal probably lived in a landscape of predominantly dry soils, intermixed with wetlands containing herbaceous plant species, as indicated by remains of Comarum palustre, Caltha palustris, Eriophorum, Sparganium, Menyanthes trifoliata and Utricularia. All recorded taxa still occur in the present-day Yakutian tundra vegetation. We discuss the representativeness in space and time of the methods used. Both the botanical microfossil and the macrofossil records may be biased because of differences in pollen production and dispersal between species, the food choice of the bison, and the season of death of the animal. Similarities and differences are discussed to highlight pitfalls of the individual analytical techniques. We highlight the power of data integration.
