Abstract Previous studies on the correlation between serum selenium and hypertension have yielded inconsistent results. Our previous analysis of participants from the National Health and Nutrition Examination Survey (NHANES) 2011–2018 indicated that elevated serum selenium concentrations were associated with an increased risk of metabolic abnormalities in obese individuals, with the primary effect being on blood pressure in males. The aim of this study was to further elucidate the relationship between serum selenium and the risk of hypertension in obese males. In this study, we examined the correlation between serum selenium concentrations and hypertension in 2,585 male participants with a body mass index (BMI) ≥ 30 kg/m2 aged between 20 and 80 years from the 2011–2018 NHANES database. The associations between serum selenium levels and hypertension were evaluated through weighted generalized linear regression analyses. To examine the saturation threshold effect between serum selenium and hypertension, a generalized additive model (GAM) and a two-piecewise linear regression model were employed. Furthermore, the saturation threshold effect was evaluated separately in subgroups stratified by BMI and age. The weighted prevalence of hypertension (51.84%) was slightly higher than that of nonhypertension (48.16%) in the participants included in this study. After rigorous adjustment for sociodemographic, physical, and laboratory test covariates, the weighted odds ratio (OR) of hypertension increased by 103% for every 1 standard deviation (SD) increase (approximately 24.41 µg) in the serum selenium concentration in participants assigned to the highest serum selenium group (weighted OR = 2.03; 95% CI = 1.24–3.32; P = 0.013). A calculation was subsequently performed to determine the saturation threshold effect of selenium on hypertension among participants in the medium and highest selenium concentration subgroups. The findings indicated that participants with serum selenium concentrations exceeding the saturation threshold (2.56 µM) demonstrated an elevated risk of developing hypertension (weighted OR = 9.58; 95% CI = 2.74–33.46; P = 0.000) in comparison to those with serum selenium concentrations below the threshold. Subgroup analyses demonstrated that serum selenium concentrations exceeding the saturation threshold were associated with an increased risk of hypertension in participants with a BMI ≤ 35 kg/m2 (weighted OR = 9.11; 95% CI = 1.43–58.24; P = 0.030) or those aged less than 55 years or younger (weighted OR = 8.37; 95% CI = 1.71–40.94; P = 0.014). For obese adult males who require additional selenium supplementation to enhancing their overall health and well-being, it is strongly recommended that the serum selenium concentrations be monitored throughout the course of supplementation to ensure that they remain within the relatively safe range (approximately less than 215.75 µg/L).

Abstract In order to solve the problems of rutting and early fatigue cracks in emulsified asphalt cold recycled pavement, and the shortage of natural stone resources and new environmental hazards caused by the use of traditional limestone powder filler. In this study, coal gangue powder was added to prepare Emulsified Asphalt Mastic (EAM) to improve the rheological properties and fatigue performance. A series of tests, including frequency scanning, temperature scanning, Multiple Stress Creep Recovery (MSCR), Linear Amplitude Scanning (LAS), and Fourier Transform Infrared spectroscopy (FTIR) were conducted. Through rheology theory, viscoelastic damage theory, the impact of three fillers: Coal Gangue Powder (CGP), Limestone Powder (LP) and Portland Cement (PC), as well as four kinds of Powder-Binder ratios (P/B) (0.6, 0.9, 1.2, 1.5) on the high temperature rheology and medium temperature fatigue performance of EAM was analyzed. The findings reveal that the filler content has a great influence on the proportion of viscoelastic components of EAM in the low frequency domain. The complex shear modulus (G*) and phase angle (δ) of EAM were suitable for Christensen-Anderson-Marasteanu model (CAM) model in the wide frequency domain. Increasing the filler content and Rotating Thin Film Oven Test (RTFOT) aging can improve the high-temperature stability and stress sensitivity of EAM, but it reduced its fatigue resistance. The high temperature performance of EAM containing PC and CGP exhibit superior high-temperature performance compared to LP fillers. Conversely, LP mastic demonstrate superior anti-fatigue performance under actual strain levels in both thin and thick asphalt pavement layers, surpassing the performance of PC mastic and CGP mastic. Therefore, coal gangue powder can be used as a new type of green filler to replace limestone powder. An appropriate amount of incorporation can improve the high temperature stability of the mortar and does not have a huge impact on the fatigue performance. The most suitable range of P/B is identified as 0.6–1.0 when utilizing LP as the filler, 1.2–1.5 when utilizing PC or CGP as the filler.

Abstract Anthrax is an acute infectious disease caused by Bacillus anthracis, which can infect various animals and humans. Cutaneous anthrax primarily presents as infiltrative, edematous erythema, surface vesicles, hemorrhagic vesicles, and necrotic eschar; some patients may also experience systemic symptoms such as fever and leukocytosis. With economic development and improvements in public health conditions, naturally occurring cases of cutaneous anthrax have significantly decreased, leading to limited reports on the pathological manifestations of this disease. This article reports a case of a patient with cutaneous anthrax diagnosed and treated in our hospital, aiming to explore the laboratory examinations, imaging, pathological features, and clinical treatment methods of the disease. The goal is to enhance understanding of anthrax and increase vigilance in clinical practice to avoid misdiagnosis and missed diagnosis. The described diagnostic and therapeutic processes are accurate and reliable, with no modifications or exaggerations. It is important to note that the patient’s treatment may have been influenced by local social, economic, health, and epidemiological conditions, which introduces certain limitations. We hope that our colleagues will approach this study with an objective mindset for learning and reference.

Abstract Bipolar disorder (BD) is a neuropsychiatric disorder characterized by severe disturbance and fluctuation in mood. Dynamic functional connectivity (dFC) has the potential to more accurately capture the evolving processes of emotion and cognition in BD. Nevertheless, prior investigations of dFC typically centered on larger time scales, limiting the sensitivity to transient changes. This study employed hidden Markov model (HMM) analysis to delve deeper into the moment-to-moment temporal patterns of brain activity in BD. We utilized resting-state functional magnetic resonance imaging (rs-fMRI) data from 43 BD patients and 51 controls to evaluate the altered dynamic spatiotemporal architecture of the whole-brain network and identify unique activation patterns in BD. Additionally, we investigated the relationship between altered brain dynamics and structural disruption through the ridge regression (RR) algorithm. The results demonstrated that BD spent less time in a hyperconnected state with higher network efficiency and lower segregation. Conversely, BD spent more time in anticorrelated states featuring overall negative correlations, particularly among pairs of default mode network (DMN) and sensorimotor network (SMN), DMN and insular-opercular ventral attention networks (ION), subcortical network (SCN) and SMN, as well as SCN and ION. Interestingly, the hypoactivation of the cognitive control network in BD may be associated with the structural disruption primarily situated in the frontal and parietal lobes. This study investigated the dynamic mechanisms of brain network dysfunction in BD and offered fresh perspectives for exploring the physiological foundation of altered brain dynamics.

In lambs, weaning imposes stress that can contribute to impaired rumen epithelial barrier functionality and immunological dysregulation. In this study, the effects of a yeast co-culture consisting of Saccharomyces cerevisiae and Kluyveromyces marxianus (NM) on rumen health in lambs was evaluated, with a focus on parameters including growth performance, ruminal fermentation, and epithelial barrier integrity, ruminal metabolic function, and the composition of the ruminal bacteria. In total, 24 lambs were grouped into four groups of six lambs including a control (C) group fed a basal diet, and N, M, and NM groups in which lambs were fed the basal diet respectively supplemented with S. cerevisiae yeast cultures (30 g/d per head), K. marxianus yeast cultures (30 g/d per head), and co-cultures of both yeasts (30 g/d per head), the experiment lasted for 42 d. Subsequent analyses revealed that relative to the C group, the average daily gain (ADG) of lambs in the NM group was significantly greater and exhibited significant increases in a range of mRNA relative expression including monocarboxylate transporter 1 (MCT1), (Na+)/hydrogen (H+) exchanger 1 (NHE1), (Na+)/hydrogen (H+) exchanger 3 (NHE3), proton-coupled amino acid transporter 1 (PAT1), vacuolar H+-ATPase (vH+ ATPase), claudin-1, occludin in the rumen epithelium (P

Delivery and weaning are major stressful events in sows and piglets, adversely affecting production and growth performance and causing economic losses to swine farms. Probiotics as safe antibiotic alternatives have great potential for use across all stages of livestock farming. Here, 18 pregnant sows from clinical farms randomly were divided into two groups: one fed a basal diet (CON group) and the other fed a basal diet plus a probiotic mixture CBB-mix (containing 1×1012 CFU/g of Lactobacillus johnsonii [CJ21], 1×109 CFU/g of Bacillus subtilis [BS15], and 1×109 CFU/g of Bacillus licheniformis [BL21]), for 20 days before delivery. The effects of maternal CBB-mix supplementation on sow colostrum metabolome and offspring piglets' clinical performance, immune status, and gut microbiota were investigated. Additionally, 177 piglets were randomly divided into 4 groups, including CC group (piglets and sows fed a basal diet, n = 40 from 5 litters), CP group (piglets fed the basal diet plus CBB-mix and sows fed the basal diet, n = 38 from 4 litters), PC group (piglets fed the basal diet and sows fed the basal diet plus CBB-mix, n = 50 from 4 litters), and PP group (both piglets and sows fed the basal diet plus CBB-mix, n = 49 from 5 litters). Context Liuwei Dihuang pill (LWDH) has been used to treat postmenopausal osteoporosis (PMOP).Objective To explore the effects and mechanisms of action of LWDH in PMOP.Materials and methods Forty-eight female Sprague-Dawley rats were divided into four groups: sham-operated (SHAM), ovariectomized (OVX), LWDH high dose (LWDH-H, 1.6 g/kg/d) and LWDH low dose (LWDH-L, 0.8 g/kg/d); the doses were administered after ovariectomy via gavage for eight weeks. After eight weeks, the bone microarchitecture was evaluated. The effect of LWDH on the differentiation of bone marrow mesenchymal stem cells (BMSCs) was assessed via osteogenesis- and lipogenesis-induced BMSC differentiation. The senescence-related biological indices were also detected using senescence staining, cell cycle analysis, quantitative real-time polymerase chain reaction and western blotting. Finally, the expression levels of autophagy-related proteins and Yes-associated protein (YAP) were evaluated.Results LWDH-L and LWDH-H significantly modified OVX-induced bone loss. LWDH promoted osteogenesis and inhibited adipogenesis in OVX-BMSCs. Additionally, LWDH decreased the positive ratio of senescence OVX-BMSCs and improved cell viability, cell cycle, and the mRNA and protein levels of p53 and p21. LWDH upregulated the expression of autophagy-related proteins, LC3, Beclin1 and YAP, in OVX-BMSCs and downregulated the expression of p62.Discussion and conclusions LWDH improves osteoporosis by delaying the BMSC senescence through the YAP-autophagy axis.

Abstract Non-Hermitian physical systems have attracted considerable attention in recent years for their unique properties around exceptional points (EPs), where the eigenvalues and eigenstates of the system coalesce. Phase transitions near exceptional points can lead to various interesting phenomena, such as unidirectional wave transmission. However, most of those studies are in the classical regime and whether these properties can be maintained in the quantum regime is still a subject of ongoing studies. Using a non-Hermitian on-chip superconducting quantum circuit, here we observe a phase transition and the corresponding exceptional point between the two phases. Furthermore, we demonstrate that unidirectional microwave transmission can be achieved even in the few-photon regime within the broken symmetry phase. This result holds some potential applications, such as on-chip few-photon microwave isolators. Our study reveals the possibility of exploring the fundamental physics and practical quantum devices with non-Hermitian systems based on superconducting quantum circuits.

EPSPS is a key gene in the shikimic acid synthesis pathway that has been widely used in breeding crops with herbicide resistance. However, its role in regulating cell elongation is poorly understood. Through the overexpression of EPSPS genes, we generated lines resistant to glyphosate that exhibit an unexpected dwarf phenotype. A representative line, DHR1, exhibits a stable dwarf phenotype throughout its entire growth period. Except for plant height, the other agronomic traits of DHR1 are similar to its transgenic explants ZM24. Paraffin section observations showed that DHR1 internodes are shortened due to reduced elongation and division of the internode cells. Exogenous hormones confirmed that DHR1 is not a classical brassinolide (BR)- or gibberellin (GA)-related dwarfing mutant. Hybridization analysis and fine mapping confirmed that the EPSPS gene is the causal gene for dwarfism, and the phenotype can be inherited in different genotypes. Transcriptome and metabolome analyses showed that genes associated with the phenylpropanoid synthesis pathway are enriched in DHR1 compared with ZM24. Flavonoid metabolites are enriched in DHR1, whereas lignin metabolites are reduced. The enhancement of flavonoids likely results in differential expression of auxin signal pathway genes and alters the auxin response, subsequently affecting cell elongation. This study provides a new strategy for generating dwarfs and will accelerate advancements in light simplification in the cultivation and mechanized harvesting of cotton.

Abstract Episodic memory is essential for forming and retaining personal experiences, representing a fundamental aspect of human cognition. Traditional studies of episodic memory have typically used static analysis methods, viewing the brain as an unchanging entity and overlooking its dynamic properties over time. In this study, we utilized dynamic functional connectivity analysis on fMRI data from healthy adults performing an episodic memory task. We quantified integration and recruitment metrics and examined their correlation with memory performance using Pearson correlation. During encoding, integration across the entire brain, especially within the frontoparietal subnetwork, was significantly correlated with memory performance. During retrieval, recruitment becomes significantly associated with memory performance in visual subnetwork, somatomotor subnetwork, and ventral attention subnetwork. At the nodal level, a significant negative correlation was observed between memory scores and integration of the anterior cingulate gyrus, precentral gyrus, and inferior frontal gyrus within the frontoparietal network during encoding task. During retrieval task, a significant negative correlation was found between memory scores and recruitment in the left progranular cortex and right transverse gyral ventral, whereas positive correlations were seen in the right posterior inferior temporal, left middle temporal, right frontal operculum, and left operculum nodes. Moreover, the dynamic reconfiguration of the functional network was predictive of predict memory performance, as demonstrated by a significant correlation between actual and predicted memory scores. These findings advance our understanding network mechanisms underlying memory processes and developing intervention approaches for memory-related disorders as they shed light on critical factors involved in cognitive processes and provide a deeper understanding of the underlying mechanisms driving cognitive function.

Abstract Background Airway pressure release ventilation (APRV) has become increasingly popular for the management of acute respiratory distress syndrome (ARDS); however, its clinical impact remains a topic of debate. Furthermore, there is a gap between the guidelines and the actual clinical practices in mechanical ventilation management for ARDS. This survey aimed to explore the utilization of APRV and mechanical ventilation strategies for ARDS in Chinese intensive care unit (ICU) clinicians. Methods A comprehensive 34‐item survey was distributed online platforms amongst ICU clinicians across mainland China from June to August 2019. Results A total of 420 valid responses were collected, with 57.4% (241) originating from academic hospitals and 42.6% (179) from non‐academic hospitals. Of the respondents, 98.6% (414) recognized the significance of low tidal volume ventilation for ARDS prognosis, 85.2% adhered to a tidal volume below 8 mL/kg predicted body weight, and most (46.4%) selected the initial positive end‐expiratory pressure within the range of 5–10 cmH2O based on experience. Among the respondents, 62.1% (261) reported familiarity with APRV and 41.9% (176) had implemented APRV. Of those who had utilized APRV, 93.2% (164) believed in its effectiveness for ARDS patients and 69.3% (122) advocated for early application of APRV. Substantial variations were noted regarding APRV initiation settings and the preservation of spontaneous breathing during APRV. Academic hospitals exhibited higher usage rates of lung recruitment, neuromuscular blockade, prone ventilation, and acquaintance with and utilization of APRV compared to non‐academic hospitals (all p values ≤ 0.001). Conclusions Our findings highlight opportunities for improvement in mechanical ventilation management for ARDS, particularly in non‐academic hospitals. Additionally, a significant proportion of clinicians demonstrated limited knowledge of APRV, and there was a lack of consensus on its application. Further training and larger‐scale clinical trials are required to validate the efficacy and utilization of APRV in managing ARDS.

A massive market penetration of electric vehicles (EVs) associated with nonnegligible energy consumption and environmental issues has imposed a big challenge on evaluating electrical power distribution and related transportation facilities improvement in response to the large-scale EV charging service need. Strategical deployment of EV charging stations including location and determination of number of slow charging stations and fast charging stations has become an emerging concern and one of the most pressing needs in planning. This paper conducts a comprehensive survey of EV charging demand and distribution models with consideration of realistic driver behaviors impacts. This is currently a shortage in academic literature, but indeed has drawn practical attention in the strategic planning process. To address the need, this paper presents an in-depth literature review of relevant studies that have identified different types of EV charging facilities, needs or concerns that are considered into EV charging demand and distribution modeling, alongside critical impacting factor identification, mathematical relationships of the contributing factors and EV charging demand and distribution modeling. Key findings from the current literature are summarized with strategies for optimized plan of charging station deployments (i.e., location and related number of charging station), in an attempt to provide a valuable reference for interested readers.

IntroductionIn lambs, the function of the rumen is incompletely developed at weaning, and the inclusion of yeast cultures in the diet can profoundly influence the morphological and functional development of the rumen.MethodsIn this study, the effects of Saccharomyces cerevisiae and Kluyveromyces marxianus (NM) yeast co-cultures on ruminal histomorphology were assessed, and corresponding transcriptomic changes within the rumen epithelium were identified. In total, 24 lambs were grouped into four groups of six lambs including a control (C) group fed a basal diet, and N, M, and NM groups in which lambs were fed the basal diet, respectively, supplemented with Saccharomyces cerevisiae yeast cultures (30 g/d per head), Kluyveromyces marxianus yeast cultures (30 g/d per head), and co-cultures of both yeasts (30 g/d per head), the experiment lasted for 42 d.ResultsIn morphological analyses, lambs from the NM group presented with significant increases in papilla length, papilla width, and epithelial thickness in the rumen relative to lambs in the C group (p < 0.05). Transcriptomic analyses revealed 202 genes that were differentially expressed between samples from the C and NM groups, with the largest proportion of these genes being associated with the oxidative phosphorylation pathway. In a weighted gene coexpression network analysis, a positive correlation was observed between the MEgreen and MEpurple modules and rumen morphology. Of these modules, the MEgreen module was found to be more closely linked to fatty acid metabolism and oxidative phosphorylation, whereas the MEpurple module was linked to oxidative phosphorylation and fatty acid degradation. Ultimately, these results suggest that dietary supplementation with NM has driven the degradation of fatty acids, the induction of oxidative phosphorylation, the acceleration of lipid metabolism, the production of ATP to sustain ruminal growth, and the maintenance of intracellular NADH/NAD+ homeostasis on weaned lambs and is superior to single yeast fermentation.DiscussionThese results thus offer a theoretical foundation for further studies examining the mechanisms through which NM cultures can influence ruminal development in lambs.

BackgroundDespite a consensus on the negative implications of problematic internet use (PIU) for mental health, there is a notable gap in research concerning older demographics, particularly older teachers. This study aimed to investigate the prevalence of PIU and its impact on the mental wellbeing of this population.MethodsThree sub-studies were conducted: Study 1 (2020) investigated how fear induced by COVID-19 influenced PIU and psychological distress among 3,929 older teachers. Study 2 (2021) examined the effects of PIU on psychological needs thwarting (the frustration of basic psychological needs) and psychological distress, involving 3,502 participants. Study 3 (2022) explored the impact of PIU on occupational burnout and psychological distress, with 1,276 participants. The Generalized Linear Model in Jamovi 2.3.23 was used to evaluate the three mediation models.ResultsThe three sub-studies revealed a high prevalence of PIU among older teachers (27.4% in Study 1, 27.4% in Study 2, and 24.5% in Study 3). High levels of PIU were associated with elevated psychological distress during the 3 years of the COVID-19 pandemic. In Study 1, fear of COVID-19 indirectly influenced psychological distress through PIU as a mediator. In Studies 2 and 3, respectively, psychological needs thwarting and occupational burnout mediated the relationship between PIU and psychological distress.ConclusionThis study confirmed the high prevalence of PIU among older school teachers and the detrimental effect of PIU on psychological needs thwarting, occupational burnout, and psychological distress. Given these findings, education authorities and school management should take proactive steps to mitigate PIU and ensure the health and wellbeing of older teachers.

During the late laying period, the intestinal barrier of laying hens is susceptible to damage, resulting in enteric infections and even systemic inflammatory responses, posing a major challenge for the poultry industry. Therefore, it is crucial to investigate methods for addressing intestinal inflammation in late laying hens. In order to maximize the production potential of egg laying chickens, farmers usually use various feed additives to prevent damage to the intestinal barrier. Composite yeast cultures have shown advantages in broiler applications. This study aims to assess the impact of composite yeast culture (CYC) on the intestinal barrier function, inflammatory cytokines, and microbial community structure of Hy-Line Brown laying hens. A total of 160 healthy Hy-Line Brown hens, aged 58 weeks and of similar weight, were randomly assigned to two groups, with four replicates per group and 20 hens in each replicate. The control group was fed a basal diet (Con), while the experimental group was provided with a diet supplemented with 40 g/kg of composite yeast culture (CYC). The test period was 25 days. Continuous emergence of new variants of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), enhanced transmissibility, significant immune escape, and waning immunity call for booster vaccination. We evaluated the safety, immunogenicity, and efficacy of heterologous booster with a SARS-CoV-2 mRNA vaccine SYS6006 versus an active control vaccine in a randomized, open-label, active-controlled phase 3 trial in healthy adults aged 18 years or more who had received two or three doses of SARS-CoV-2 inactivated vaccine in China. The trial started in December 2022 and lasted for 6 months. The participants were randomized (overall ratio: 3:1) to receive one dose of SYS6006 (N = 2999) or an ancestral receptor binding region-based, alum-adjuvanted recombinant protein SARS-CoV-2 vaccine (N = 1000), including 520 participants in an immunogenicity subgroup. SYS6006 boosting showed good safety profiles with most AEs being grade 1 or 2, and induced robust wild-type and Omicron BA.5 neutralizing antibody response on Days 14 and 28, demonstrating immunogenicity superiority versus the control vaccine and meeting the primary objective. The relative vaccine efficacy against COVID-19 of any severity was 51.6% (95% CI, 35.5–63.7) for any variant, 66.8% (48.6–78.5) for BA.5, and 37.7% (2.4–60.3) for XBB, from Day 7 through Month 6. In the vaccinated and infected hybrid immune participants, the relative vaccine efficacy was 68.4% (31.1–85.5) against COVID-19 of any severity caused by a second infection. All COVID-19 cases were mild. SYS6006 heterologous boosting demonstrated good safety, superior immunogenicity and high efficacy against BA.5-associated COVID-19, and protected against XBB-associated COVID-19, particularly in the hybrid immune population.Trial registration: Chinese Clinical Trial Registry: ChiCTR2200066941

In this study, Continuous Discontinuous Element Method (CDEM) was employed to analyze the failure of concrete slab under drop-hammer impact at the mesoscopic level. Initially, simulation models of the impact system were established, and concrete slab models, including three-dimensional random aggregates, were generated. Mesoscopic simulations of concrete failure were then conducted by considering different variables, such as impact energy, slab size and the number of hammerheads. Corresponding drop hammer tests were used to validate the simulation results. Furthermore, an energy conversion formula for the dynamic failure of concrete slabs was derived, and the energy conversion process within the impact system was analyzed. The study also revealed the dynamic failure mechanisms of concrete slabs under various impact conditions. The research demonstrates that the simulation results are generally consistent with the impact tests. CDEM effectively investigates the impact characteristic and the energy conversion process of concrete slab failure. Under varying impact energies, slab sizes and hammerhead numbers, distinct differences in the failure characteristics of concrete-slab structure were observed. Increased impact energy accelerates the failure of concrete slabs, whereas larger concrete slab sizes inhibit crack propagation. The number of impact heads determines the initial crack position and its propagation path within the concrete slab. Moreover, the energy conversion process of the impact system significantly influences the failure behavior of the concrete slab. The effect of slab size on the energy utilization rate is greater than those of hammerhead number and impact speed.

In order to investigate the rheological behavior and the influence of solid waste coal gangue powder on the high-temperature performance of cold recycled Emulsified Asphalt Mastics (EAM), Dynamic Shear Rheometer (DSR) and Multiple Stress Creep Recovery (MSCR) tests were conducted using three different fillers and four powder-to-binder ratios of EAM. Evaluation indexes such as rutting factor (G*/sinδ), improved rutting factor (G*/1−1/sinδ∙tanδ), Unrecoverable Creep Compliance (Jnr), Cumulative Strain (CS) and Creep Recovery Ratio (R) were used to analyze the influence of filler type and dosage on high temperature of EAM, and the effectiveness of these indexes to evaluate high temperature is considered. Moreover, atomic force microscopy experiments were carried out to study on interaction mechanism between the fillers and emulsified asphalt. Finally, grey relational analysis method was used for quantitative assessment of these indexes for EAM. The results indicate that all three fillers effectively improve the high-temperature performance, with a larger improvement observed as the filler dosage increases, among which the tunneling coal gangue powder is the most significant. Under a stress level of 0.1 KPa, the coal gangue filler mastic appears the best deformation recovery ability, whereas under a stress level of 3.2 KPa, the cement filler mastics appears the best deformation recovery ability. The additional fillers significantly influence the nano-scale surface morphology of the residues, and the tunneling coal gangue powder appears a stronger adsorption capacity for free polar components compared to the cement and limestone mineral powder. It is suggested that the Jnr and CS can be used as evaluation indexes of high temperature evaluation index of EAM, instead of G*/1−1/sinδ∙tanδ or G*/sinδ or R.

Yin deficiency syndrome is a theoretical concept of traditional Chinese medicine (TCM) that claims an imbalance between Yin and Yang serves as a potential etiological factor disrupting physiological homeostasis. Its diagnosis in TCM is hindered by the intricate and diverse etiology, resulting in the absence of quantification and standardization. Hence, this study developed a hydrocortisone (HC) induced Yin deficiency syndrome model to investigate the intricate network underlying TCM and elucidate its intervention mechanism. In the findings, the model was characterized by weight loss, elevated drinking water, yellow urination, dry stools, variations in inflammatory factors, and higher levels of oxidative stress. Based on metabolomics, 56 metabolites showed different expressions; among them, 19 were upregulated, and 37 were downregulated. Bioinformatics analysis revealed that identified metabolites are mainly involved in glycerol phospholipid metabolism, pyrimidine metabolism, amino acid biosynthesis, arginine and proline metabolism, and arachidonic acid metabolism pathways. Finally, the metabolic network association confirmed the diagnostic accuracy of the Yin deficiency syndrome as established by HC. We propose for the first time an animal model of Yin deficiency syndrome induced by hydrocortisone in TCM clinical state. This research presents experimental evidence for establishing TCM symptoms and serves as a fundamental basis for the scientific awareness of their etiology.

ABSTRACT Hawk tea (Litsea coreana var. sinensis), derived from the tender shoots or leaves, rich in flavonoids can promote healthcare for humans. The primary flavonoid are kaempferol‐3‐O‐β‐D‐glucoside, kaempferol‐3‐O‐β‐D‐galactoside, quercetin‐3‐O‐β‐D‐glucoside, and quercetin‐3‐O‐β‐D‐galactoside. The existence of an association between leaf phenotype and flavonoid content, along with the underlying mechanisms of flavonoid biosynthesis, remains incompletely understood. In this study, 109 samples were analyzed to determine the correlation and genetic variability in leaf phenotype and flavonoid content. Furthermore, a transcriptome‐wide association study identified candidate loci implicated in the biosynthesis of four key flavonoids. The study revealed that genetic variability in leaf traits and flavonoid concentrations is predominantly attributed to interpopulation differences. Flavonoid accumulation was significantly correlated with tree DBH, indicative of age‐related traits. Transcriptome‐wide association analysis identified 84 significant SNPs associated with flavonoid content, with only 13 located within gene regions. The majority of these genes are implicated in metabolic processes and secondary metabolite biosynthesis. Notably, structural genes within these regions are directly involved in pathways known to regulate flavonoid metabolism, exerting a pivotal influence on flavonoid biosynthesis. These results revealed the physiological basis for the regulation of flavonoid content, as well as the molecular mechanisms for the biosynthesis of flavonoids in hawk tea. It also lays theoretical groundwork for subsequent explorations into the genetic determinants influencing flavonoid accumulation of hawk tea.

Abstract Background Chronic obstructive pulmonary disease (COPD) remains a leading cause of morbidity and mortality worldwide, characterized by persistent respiratory symptoms and airflow limitation. The involvement of C–C motif chemokine ligand 2 (CCL2) in COPD pathogenesis, particularly in macrophage regulation and activation, is poorly understood despite its recognized role in chronic inflammation. Our study aims to elucidate the regulatory role and molecular mechanisms of CCL2 in the pathogenesis of COPD, providing new insights for therapeutic strategies. Methods This study focused on the CCL2-CCR2 signaling pathway, exploring its role in COPD pathogenesis using both Ccl2 knockout (KO) mice and pharmacological inhibitors. To dissect the underlying mechanisms, we employed various in vitro and in vivo methods to analyze the secretion patterns and pathogenic effects of CCL2 and its downstream molecular signaling through the CCL2-CCR2 axis. Results Elevated Ccl2 expression was confirmed in the lungs of COPD mice and was associated with enhanced recruitment and activation of macrophages. Deletion of Ccl2 in knockout mice, as well as treatment with a Ccr2 inhibitor, resulted in protection against CS- and LPS-induced alveolar injury and airway remodeling. Mechanistically, CCL2 was predominantly secreted by bronchial epithelial cells in a process dependent on STAT1 phosphorylation and acted through the CCR2 receptor on macrophages. This interaction activated the PI3K-AKT signaling pathway, which was pivotal for macrophage activation and the secretion of inflammatory cytokines, further influencing the progression of COPD. Conclusions The study highlighted the crucial role of CCL2 in mediating inflammatory responses and remodeling in COPD. It enhanced our understanding of COPD's molecular mechanisms, particularly how CCL2's interaction with the CCR2 activates critical signaling pathways. Targeting the CCL2-CCR2 axis emerged as a promising strategy to alleviate COPD pathology. Graphical Abstract

This systematic review aims to offer a comprehensive analysis of recent empirical studies on technology-supported scaffoldings (TSS) within the education sector. A total of 31 TSS studies published between 2017 and 2022 were selected from the reputable databases. The analysis focuses on trends, research methods, intervention design, and student learning outcomes in TSS studies. The findings reveal that TSS applications encompass a diverse range of participants, subjects, pedagogical approaches, teaching strategies, and technological tools, impacting cognitive and affective domains. However, recent TSS research demonstrates a bias towards upper primary and junior secondary students, with a focus on science and language subjects. While established e-learning tools are commonly integrated into TSS designs, there is limited exploration of metacognitive and motivational scaffoldings. This study highlights the importance of dynamic scheduling and adjustment of TSS to cater to individual student needs and capabilities, laying the groundwork for future research to develop highly customized TSS that effectively support student learning.

Five new species of the genus Bifurcia are described: B. kangding sp. nov. (♂♀), B. labahe sp. nov. (♂), B. luding sp. nov. (♂♀), B. shuangqiao sp. nov. (♂♀), and B. xiaojin sp. nov. (♂♀) from western Sichuan, China. A distribution map of the species and illustrations of genital characters are provided in this paper.

Abstract Bacterial ClpB is an ATP-dependent disaggregate that belongs to the Hsp100/Clp family and facilitates bacterial survival under hostile environmental conditions. Streptococcus agalactiae, which is regarded as the major bacterial pathogen of farmed Nile tilapia (Oreochromis niloticus), is known to cause high mortality and large economic losses. Here, we report a ClpB homologue of S. agalactiae and explore its functionality. S. agalactiae with a clpB deletion mutant (∆clpB) exhibited defective tolerance against heat and acidic stress, without affecting growth or morphology under optimal conditions. Moreover, the ΔclpB mutant exhibited reduced intracellular survival in RAW264.7 cells, diminished adherence to the brain cells of tilapia, increased sensitivity to leukocytes from the head kidney of tilapia and whole blood killing, and reduced mortality and bacterial loads in a tilapia infection assay. Furthermore, the reduced virulence of the ∆clpB mutant was investigated by transcriptome analysis, which revealed that deletion of clpB altered the expression levels of multiple genes that contribute to the stress response as well as certain metabolic pathways. Collectively, our findings demonstrated that ClpB, a molecular chaperone, plays critical roles in heat and acid stress resistance and virulence in S. agalactiae. This finding provides an enhanced understanding of the functionality of this ClpB homologue in gram-positive bacteria and the survival strategy of S. agalactiae against immune clearance during infection.

Abstract We propose a new “disappearing positron track” channel at Belle II to search for dark matter, in which a positron that is produced at the primary interaction vertex scatters with the electromagnetic calorimeter to produce dark matter particles. Such scatterings can occur via either annihilation with atomic electrons, or the bremsstrahlung process with target nuclei. The main backgrounds are due to photons and neutrons that are produced in the same scatterings and then escape detection. We require a large missing energy and further veto certain activities in the KLM detector to suppress such backgrounds. To illustrate the sensitivity of the new channel, we consider a new physics model where dark matter interacts with the standard model via a dark photon, which decays predominantly to dark matter; we find that our proposed channel can probe some currently unexplored parameter space, surpassing both the mono-photon channel at Belle II and the NA64 constraints.

Cognitive dysfunction has become the second leading cause of death among the diabetic patients. In pre-diabetic stage, blood-brain barrier (BBB) injury occurs and induced the microvascular complications of diabetes, especially, diabetes-associated cognitive dysfunction (DACD). Endothelial cells are the major component of BBB, on which the increased expression of CD40 could mediate BBB dysfunction in diabetics. Since platelets play an important role in regulating endothelial cell barrier function and over 95 % of the circulating soluble CD40 ligand (sCD40L) is derived from activated platelets, we speculated that the release of CD40L from activated platelets induced by diabetes was the key mechanism that aggravated BBB injury and leaded to DACD. We performed inhibition of platelet activation on diabetic and non-diabetic mice, with or without cilostazol treatment, and then compared cognitive function, platelet activation, BBB structure and permeability. In vitro, mouse brain microvascular endothelial cell line (b.End3) were exposed to CD40L for 24 h at 5.5 mM or 30 mM glucose media after silencing CD40 and HIF1α or not to investigate the effects of CD40 on BBB disruption and the underlying molecular pathways. Inhibition of platelet activation improved cognitive behaviors in diabetic mice, accompanied with reduced BBB permeability, increased tight junction proteins, balanced Aβ transporters, as well as attenuated Aβ deposition and hippocampal neurons damage. In vitro, CD40L increased HIF1α, diminished tight junction proteins and dysregulated Aβ transporters in b.End3 cells, which could be restored by CD40 siRNA and HIF1α siRNA. Hence, inhibition of platelet activation ameliorates DACD via alleviating BBB injury, which involving the regulation of CD40L-CD40-HIF1α signaling pathway. Our study may demonstrate a potential therapeutic target for the treatment of DACD.

Summary: Background: Complete and unbiased reporting of clinical trial results is essential for evaluating medical advances, yet publication bias and reporting discrepancies in research on the clinical application of artificial intelligence (AI) remain unknown. Methods: We conducted a comprehensive search of research publications and clinical trial registries focused on the application of AI in healthcare. Our search included publications in Dimensions.ai and pre-registered records from ClinicalTrials.gov and the EU Clinical Trials Registry before 31 December 2023. We linked registered trials to their corresponding publications, analysed the registration, reporting and different dissemination patterns of results, identified discrepancies between clinical trial registries and published literature, and assessed the use of these results in secondary research. Findings: We identified 28,248 publications related to the use of AI in clinical settings and found 1863 publications that included a clinical trial registration ID. The clinical trial registry search identified 3710 trials evaluating the use of AI in clinical settings, of which 1106 trials are completed, yet only 101 trials have published results. By linking the trials to their corresponding publications, we found that 26 trials had results available from both registries and publications. There were more results in trial registries than in articles, but researchers showed a clear preference for rapid dissemination of results through peer-reviewed articles (37.6% published within one year) over trial registries (15.8%). Discrepancies and omissions of results were common, and no complete agreement was observed between the two sources. Selective reporting of publications occurred in 53.6% of cases, and the underestimation of the incidence of adverse events is alarming. Interpretation: This research uncovers concerns with the registration and reporting of AI clinical trial results. While trial registries and publications serve distinct yet complementary roles in disseminating research findings, discrepancies between them may undermine the reliability of the evidence. We emphasise adherence to guidelines that promote transparency and standardisation of reporting, especially for investigator-initiated trials (IITs). Achilles tendon is a motor organ that is prone to tissue adhesion during its repair process after rupture. Therefore, developing motion-responsive and anti-adhesive biomaterials is an important need for the repair of Achilles tendon rupture. Here, we report an injectable lubricative hydrogel (ILH) based on hydration lubrication mechanism, which is also motion-responsive based on sol-gel reversible transmission. The lubrication performance is achieved by zwitterionic polymers as we previously proved, and the sol-gel reversible transmission is enabled by dynamic disulfide bonds. Firstly, ILH was proved to be successfully prepared and lubricated as well as sol-gel reversible via FTIR characterization, rheological measurement and tribological tests. Then, in vitro cell experiments and coagulation tests demonstrated the optimal cytocompatibility and hemocompatibility of ILH. To evaluate the potential of ILH's biofunction in vivo, SD rats' Achilles tendon rupture & repair model was established. The animal experiments' results showed that ILH significantly prevented tendon adhesion and thus promote tendon healing by inhibiting TGFβ1-Smad2/3 pathway. We believe this work will open a new horizon for tendon adhesion-free repair.

Phages and phage-encoded lytic enzymes are promising antimicrobial agents. In this study, we report the isolation and identification of bacteriophage KP2025 from Klebsiella pneumoniae. Bioinformatics analysis of KP2025 revealed a putative endolysin, LysKP213, containing a T4-like_lys domain. Purified LysKP213 was found to be highly thermostable, retaining approximately 44.4% of its lytic activity after 20 h of incubation at 95°C, and approximately 57.5% residual activity after 30 min at 121°C. Furthermore, when administered in combination with polymyxin B or fused at the N-terminus with the antimicrobial peptide cecropin A (CecA), LysKP213 exhibited increased antibacterial activity against Gram-negative pathogens, including K. pneumoniae, Pseudomonas aeruginosa, Acinetobacter baumannii, and Escherichia coli, both in vitro and in vivo. These results indicated that LysKP213 is a highly thermostable endolysin that, when combined with or fused with an outer membrane permeabilizer, has enhanced antibacterial activity and is a candidate agent for the control of infections by Gram-negative pathogens.

PurposeThis article explored the causal relationship between immune cells and diabetic retinopathy (DR) using single nucleotide polymorphisms (SNPs) as an instrumental variable and Mendelian randomization (MR).MethodsStatistical data were collected from a publicly available genome-wide association study (GWAS), and SNPs that were significantly associated with immune cells were used as instrumental variables (IVs). Inverse variance weighted (IVW) and MR−Egger regression were used for MR analysis. BackgroundThe adenosine–adenosine receptor pathway plays important roles in the immune system and inflammation. Four adenosine receptors (i.e., A1R, A2AR, A2BR, and A3R) have been identified. However, the roles of these receptors were different in the disease progress and even play opposite roles in the same disease. This study aims to investigate the roles of A1R/A2AR/A2BR/A3R activation in liver fibrosis.MethodsIntraperitoneal injection of CCl4 into C57BL/6 mice was used to induce liver fibrosis in the models. Adenosine receptor agonists CCPA, CGS21680, BAY 60-6583, and namodenoson were used for A1R/A2AR/A2BR/A3R activation, respectively. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels were used to evaluate the liver function. Hematoxylin and eosin (H&E) staining was used to investigate the pathological damage. Masson staining and Sirius Red staining were performed to evaluate the degree of collagen deposition. CCK8 and scratch assays were used to investigate the proliferation and migration ability of hepatic stellate cells (HSCs).ResultsBy using liver fibrosis mouse models, we observed that the A1R and A2AR agonists aggravated liver fibrosis, characterized by increasing ALT and AST levels, more serious liver pathological damage, and collagen deposition. However, the A2BR and A3R agonists alleviated liver fibrosis. Moreover, the A1R and A2AR agonist treatment promotes the proliferation and migration of HSC line LX2, while A2BR and A3R agonist treatment inhibited LX2 proliferation and migration. Consistently, A1R and A2AR agonist treatment elevated the expression of α-SMA and Col1α1 in LX2, whereas A2BR and A3R agonist treatment inhibited the expression of α-SMA and Col1α1 in LX2 cells. Additionally, 5′-N-ethyl-carboxamidoadenosine (NECA), a metabolically stable adenosine analog, alleviated liver fibrosis and inhibited LX2 cell activity, proliferation, and migration.ConclusionThis study demonstrated the different roles of A1R/A2AR/A2BR/A3R during liver fibrosis development via regulating the HSC activity and proliferation.

BackgroundObesity represents a significant risk factor for the development of metabolic abnormalities. However, it is not inevitable that all individuals with obesity will develop these disorders. Selenium has been demonstrated to play a role in maintaining metabolic homeostasis in vivo, with the ability to regulate relevant signaling pathways involved in glucose and lipid metabolism processes. Previous studies have indicated that selenium concentrations in obese individuals are higher than those reported in the general population. These findings the question of whether altered selenium concentrations may act as important triggers for accelerating metabolic imbalances in the obese population. The aim of this study was to examine the potential correlation between serum selenium concentrations and the risk of developing metabolic abnormalities in individuals with obesity.MethodsThe present study included 6,125 participants from the 2011-2018 National Health and Nutrition Examination Survey (NHANES) who were aged between 20 and 80 years, with a body mass index (BMI) of 30 kg/m2 or greater, and met the inclusion and exclusion criteria. Weighted generalized linear regression analyses were conducted to evaluate the associations between serum selenium concentrations and the conversion of metabolically healthy obesity (MHO) to metabolically unhealthy obesity (MUO). A generalized additive model (GAM) and a two-piecewise linear regression model were employed to investigate the saturation threshold effect between selenium and MUO. The correlation between different selenium concentration intervals and metabolic diseases was evaluated by categorizing selenium concentrations according to the saturation threshold. Furthermore, this study investigated the correlation between serum selenium and lipid concentrations in obese females and between serum selenium and blood pressure in obese males.ResultsThe weighted prevalence of MUO in the study population was 48.35%. After rigorous adjustment for sociodemographic, physical, and laboratory test covariates, the weighted odds ratio (OR) of MUO increased by 44% for every 1 µM increase (approximately 78.74 µg) in the serum selenium concentration (weighted OR=1.44; 95% CI=1.09 - 1.91; P=0.018). Second, GAM analysis and saturation threshold analyses revealed an inverted U-shaped relationship between serum selenium and metabolic abnormalities in males, with a corresponding inflection point (K) of 2.82 µM. When the serum selenium concentration was below the K-value, the effects of serum selenium were mainly on blood pressure, especially diastolic blood pressure (DBP) (weighted β: 3.34; 95% CI= 0.25 - 6.44; P=0.038). Conversely, the correlation between the serum selenium concentrations and metabolic homeostasis imbalance in females was linear. When the selenium concentration exceeded 2.12 µM, the increase in selenium content was accompanied by increases in total cholesterol (TC, weighted β=0.54, 95% CI=0.32 - 0.76; P=0.000) and triglyceride (TG, weighted β=0.51, 95% CI=0.27 - 0.75; P=0.000) concentrations.ConclusionsThe findings of our study indicate that selenium supplementation strategies for individuals with obesity should be tailored to the sex of the individual. In females, serum selenium concentration above the saturation threshold primarily facilitates the transition from MHO to MUO by influencing alterations in serum lipid metabolism. Maintaining selenium concentrations below the threshold levels is highly important for preventing the conversion of MHO to MUO. In males, serum selenium concentrations above the threshold were found to be effective in preventing an elevation in blood pressure, particularly in improving systolic blood pressure (SBP). Nevertheless, serum selenium concentrations below the threshold are linked to an increased risk of hypertension in obese individuals, particularly those with elevated diastolic blood pressure (DBP). Further research is needed to elucidate the optimal serum selenium concentration that exerts deleterious effects on blood pressure.

Abstract Litsea coreana Levl. var. sinensis (Allen) Yang et P. H. Huang is a popular ethnic herb and beverage plant known for its high flavonoid content, which has been linked to a variety of pharmacological benefits and crucial health-promoting impacts in humans. The progress in understanding the molecular mechanisms of flavonoid accumulation in this plant has been hindered due to the deficiency of genomic and transcriptomic resources. We utilized a combination of Illumina and Oxford Nanopore Technology (ONT) sequencing to generate a de novo hybrid transcriptome assembly. In total, 126,977 unigenes were characterized, out of which 107,977 were successfully annotated in seven public databases. Within the annotated unigenes, 3,781 were categorized into 58 transcription factor families. Furthermore, we investigated the presence of four valuable flavonoids—quercetin-3-O-β-D-galactoside, quercetin-3-O-β-D-glucoside, kaempferol-3-O-β-D-galactoside, and kaempferol-3-O-β-D-glucoside in 98 samples, using high-performance liquid chromatography. A weighted gene co-expression network analysis identified two co-expression modules, MEpink and MEturquoise, that showed strong positive correlation with flavonoid content. Within these modules, four transcription factor genes (R2R3-MYB, NAC, WD40, and ARF) and four key enzyme-encoding genes (CHI, F3H, PAL, and C4H) emerged as potential hub genes. Among them, the R2R3-MYB (LcsMYB123) as a homologous gene to AtMYB123/TT2, was speculated to play a significant role in flavonol biosynthesis based on phylogenetic analysis. Our findings provided a theoretical foundation for further research into the molecular mechanisms of flavonoid biosynthesis. Additionally, The hybrid transcriptome sequences will serve as a valuable molecular resource for the transcriptional annotation of L. coreana var. sinensis, which will contribute to the improvement of high-flavonoid materials.

Probiotics exert beneficial effects on the host. This study aimed to investigate whether maternally ingested Lacticaseibacillus rhamnosus Probio-M9 during pregnancy could access and colonize the infant gut. This study recruited one pregnant woman, who ingested Probio-M9 daily from 35 weeks of gestation to delivery. Feces of the mother-infant pair were regularly collected from one month before delivery to 6 months of infant’s age for metagenomic sequencing. Probio-M9 genomes were mappable to all infant fecal samples, suggesting the ingested probiotics could be vertically transmitted from mother to infant. Infant- or mother-specific differential metabolic pathways were found between the maternal and infant’s gut microbiome, implicating apparent differences in the intestinal metagenomic potential/function between the mother and the infant. In conclusion, maternal ingestion of Probio-M9 during the final weeks of gestation could deliver to the infant gut. The findings provided novel insights into shaping infant’s gut microbiota.

Osteosarcoma is usually resistant to immunotherapy and, thus primarily relies on surgical resection and high-dosage chemotherapy. Unfortunately, less invasive or toxic therapies such as photothermal therapy (PTT) and chemodynamic therapy (CDT) generally failed to show satisfactory outcomes. Adequate multimodal therapies with proper safety profiles may provide better solutions for osteosarcoma. Herein, a simple nanocomposite that synergistically combines CDT, PTT, and chemotherapy for osteosarcoma treatment was fabricated. In this composite, small 2D NiFe-LDH flakes were processed into 3D hollow nanospheres via template methods to encapsulate 5-Fluorouracil (5-FU) with high loading capacity. The nanospheres were then adsorbed onto larger 2D Ti3C2 MXene monolayers and finally shielded by bovine serum albumin (BSA) to form 5-FU@NiFe-LDH/Ti3C2/BSA nanoplatforms (5NiTiB). A mixed nitrate (NO3−) and ammonium (NH4+) supply can promote root growth in maize (Zea mays), however, the changes in root morphology and the related physiological mechanism under different N forms are still unclear. Here, maize seedlings were grown hydroponically with three N supplied in three different forms (NO3− only, 75/25 NO3−/NH4+ and NH4+ only). Compared with sole NO3− or NH4+, the mixed N supply increased the total root length of maize but did not affect the number of axial roots. The main reason was the increased total lateral root length, while the average lateral root (LR) length in each axle was only slightly increased. In addition, the average LR density of 2nd whorl crown root under mixed N was also increased. Compared with sole nitrate, mixed N could improve the N metabolism of roots (such as the N influx rate, nitrate reductase (NR) and glutamine synthase (GS)enzyme activities and total amino content of the roots). Experiments with exogenously added NR and GS inhibitors suggested that the increase in the average LR length under mixed N was related to the process of N assimilation, and whether the NR mediated NO synthesis participates in this process needs further exploration. Meanwhile, an investigation of the changes in root-shoot ratio and carbon (C) concentration showed that C transportation from shoots to roots may not be the key factor in mediating lateral root elongation, and the changes in the sugar concentration in roots further proved this conclusion. Furthermore, the synthesis and transportation of auxin in axial roots may play a key role in lateral root elongation, in which the expression of ZmPIN1B and ZmPIN9 may be involved in this pathway. This study preliminarily clarified the changes in root morphology and explored the possible physiological mechanism under a mixed N supply in maize, which may provide some theoretical basis for the cultivation of crop varieties with high N efficiency.

Four morphologically similar species of the Pholcus yichengicus species group, occurring in geographic proximity of China’s Qinling Mountains, were recognised, based on morphology and four methods of molecular species delimitation. They comprise two new species, namely Pholcus ankang sp. nov. and P. baoji sp. nov. and two previously described species: P. ovatus Yao & Li, 2012 and P. taibaiensis Wang & Zhu, 1992. Their DNA barcodes were obtained to estimate p-distances and K2P distances. In addition, an identification key for the four closely-related species is presented.

We report 18 spider species of the genus Pholcus Walckenaer, 1805 from a survey in the Qinling Mountains of central China. They belong to four species groups and include seven species new to science: Pholcus jiaozuo Yang & Yao, sp. nov. (♂♀) in the taishan species group; P. luonan Yang & Yao, sp. nov. (♂♀), P. luoyang Yang & Yao, sp. nov. (♂♀), P. lushan Yang & Yao, sp. nov. (♂♀), P. shangluo Yang & Yao, sp. nov. (♂♀), P. weinan Yang & Yao, sp. nov. (♂♀) and P. yuncheng Yang & Yao, sp. nov. (♂♀) in the yichengicus species group. Detailed diagnoses, descriptions, photomicroscopy images and DNA barcodes of all new species are provided. Our study will make a significant contribution to understanding species diversity and zoogeography of the region.

Abstract Background Previous studies have observed a link between immunophenotypes and lung cancer, both of which are closely associated with genetic factors. However, the causal relationship between them remains unclear. Methods Bidirectional Mendelian randomization (MR) was performed on publicly available genome-wide association study (GWAS) summary statistics to analyze the causal relationships between 731 immunophenotypes and lung cancer. Sensitivity analyses were conducted to verify the robustness, heterogeneity, and potential horizontal pleiotropy of our findings. Results Following Bonferroni adjustment, CD14− CD16+ monocyte (OR = 0.930, 95%CI 0.900–0.960, P = 8.648 × 10− 6, P Bonferroni = 0.006) and CD27 on CD24+ CD27+ B cells (OR = 1.036, 95%CI 1.020–1.053, P = 1.595 × 10 − 5, P Bonferroni = 0.012) were identified as having a causal role in lung cancer via the inverse variance weighted (IVW) method. At a more relaxed threshold, CD27 on IgD+ CD24+ B cell (OR = 1.035, 95%CI 1.017–1.053, P = 8.666 × 10− 5, P Bonferroni = 0.063) and CD27 on switched memory B cell (OR = 1.037, 95%CI 1.018–1.056, P = 1.154 × 10− 4, P Bonferroni = 0.084) were further identified. No statistically significant effects of lung cancer on immunophenotypes were found. Conclusions The elevated level of CD14− CD16+ monocytes was a protective factor against lung cancer. Conversely, CD27 on CD24+ CD27+ B cell was a risk factor. CD27 on class-switched memory B cells and IgD+ CD24+ B cells were potential risk factors for lung cancer. This research enhanced our comprehension of the interplay between immune responses and lung cancer risk. Additionally, these findings offer valuable perspectives for the development of immunologically oriented therapeutic strategies.

Abstract Vaccines have proven effective in protecting populations against COVID-19, including the recombinant COVID-19 vaccine (Sf9 cells), the first approved recombinant protein vaccine in China. In this positive-controlled trial with 85 adult participants (Sf9 cells group: n = 44; CoronaVac group: n = 41), we evaluated the safety, immunogenicity, and protective effectiveness of a heterologous boost with the Sf9 cells vaccine in adults who had been vaccinated with the inactivated vaccine, and found a post-booster adverse events rate of 20.45% in the Sf9 cells group and 31.71% in the CoronaVac group (p = 0.279), within 28 days after booster injection. Neither group reported any severe adverse events. Abstract Background Multispecies biofilms located in the anatomical intricacies of the root canal system remain the greatest challenge in root canal disinfection. The efficacy of Er:YAG laser-activated irrigation techniques for treating multispecies biofilms in these hard-to-reach areas has not been proved. The objective of this laboratory study was to evaluate the effectiveness of two Er:YAG laser-activated irrigation techniques, namely, photon-induced photoacoustic streaming (PIPS) and shock wave-enhanced emission photoacoustic streaming (SWEEPS), in treating multispecies biofilms within apical artificial grooves and dentinal tubules, in comparison with conventional needle irrigation (CNI), passive ultrasonic irrigation (PUI), and sonic-powered irrigation (EDDY). Two types of multispecies root canal biofilm models were established in combination with two assessment methods using scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM) with the aim to obtain more meaningful results. Methods Ninety extracted human single-rooted premolars were chosen for two multispecies biofilm models. Each tooth was longitudinally split into two halves. In the first model, a deep narrow groove was created in the apical segment of the canal wall. After cultivating a mixed bacterial biofilm for 4 weeks, the split halves were reassembled and subjected to five irrigation techniques: CNI, PUI, EDD, PIPS, and SWEEPS. The residual biofilms inside and outside the groove in Model 1 were analyzed using SEM. For Model 2, the specimens were split longitudinally once more to evaluate the percentage of killed bacteria in the dentinal tubules across different canal sections (apical, middle, and coronal thirds) using CLSM. One-way analysis of variance and post hoc multiple comparisons were used to assess the antibiofilm efficacy of the 5 irrigation techniques. Results Robust biofilm growth was observed in all negative controls after 4 weeks. Lipid accumulation and oxidative stress, which could be improved by autophagy, are the “hits” of metabolic-associated fatty liver disease (MAFLD). Ganoderma lucidum spore powder (GLSP) has the effect of improving liver function. However, there are few reports about its effects on and mechanisms impacting MAFLD alleviation. This study investigated the effect of GLSP on hepatic lipid accumulation and oxidative stress and explored the role that autophagy played in this effect. The results showed that GLSP effectively reduced lipid accumulation and activated autophagy in the livers of mice with high-fat-diet-induced disease and palmitic acid-induced hepatocytes. GLSP reduced the lipid accumulation by reducing lipogenesis and promoting lipid oxidation in HepG2 cells. It decreased the production of ROS, increased the activity of SOD and CAT, and improved the mitochondrial membrane potential via the Keap1/Nrf2 pathway. The alleviating effects of GLSP on the lipid accumulation and oxidative stress was reversed by 3-methyladenine (3-MA), an autophagy inhibitor. GLSP activated autophagy via the AMPK pathway in HepG2 cells. In conclusion, GLSP could attenuate MAFLD by the improvement of lipid accumulation and oxidative stress via autophagy. This paper is the first to report the improvement of MAFLD through autophagy promotion. It will shed novel light on the discovery of therapeutic strategies targeting autophagy for MAFLD.

Due to the frequent occurrence of food safety problems in recent years, healthy diets are gradually receiving worldwide attention. Chemical pigments are used in smart food packaging because of their bright colors and high visibility. However, due to shortcomings such as carcinogenicity, people are gradually looking for natural pigments to be applied in the field of smart food packaging. In traditional smart food packaging, the indicator and the packaging bag substrate have different degrees of toxicity. Smart food packaging that combines natural colorants and cellulose-based hydrogels is becoming more and more popular with consumers for being natural, non-toxic, environmentally friendly, and renewable. This paper reviews the synthesis methods and characteristics of cellulose-based hydrogels, as well as the common types and characteristics of natural pigments, and discusses the application of natural colorants and cellulose-based hydrogels in food packaging, demonstrating their great potential in smart food packaging.

Ambient pH, an important environmental factor, affects the growth, pathogenicity, and mycotoxin production of pathogenic fungus. Fusarium sulphureum is one of the predominant causal agents causing fusarium rot of muskmelon. In this study, we investigated the effects of ambient pH on fusarium rot development and diacetoxyscirpenol (DAS) accumulation in muskmelon infected with F. sulphureum, then analyzed the possible mechanisms in vitro and in vivo. The results suggested that ambient pH 6 was more conducive to the growth, pathogenicity, and mycotoxin production of F. sulphureum in vitro. Ambient pH 6 was also more favorable for secretion of cell wall-degrading enzymes for the pathogen to degrade the cell wall of the host plant and up-regulated the relative expression of genes involved in DAS biosynthesis, thus aggravating fruit disease and DAS accumulation. However, when the pH of the inoculated spore suspension was too acidic or too alkaline, the opposite results were observed.

The functional connectivity (FC) graph of the brain has been widely recognized as a ``fingerprint'' that can be used to identify individuals from a group of subjects. Research has indicated that individual identification accuracy can be improved by eliminating the impact of shared information among individuals. However, current research extracts not only shared information of inter-subject but also individual-specific information from FC graphs, resulting in incomplete separation of shared information and fingerprint information among individuals, leading to lower individual identification accuracy across all functional magnetic resonance imaging (fMRI) states session pairs and poor cognitive behavior prediction performance. In this paper, we propose a method to enhance inter-subject variability combining conditional variational autoencoder (CVAE) network and sparse dictionary learning (SDL) module. By embedding fMRI state information in the encoding and decoding processes, the CVAE network can better capture and represent the common features among individuals and enhance inter-subject variability by residual. Our experimental results on Human Connectome Project (HCP) data show that the refined connectomes obtained by using CVAE with SDL can accurately distinguish an individual from the remaining participants. The success accuracies reached 99.7 % and 99.6 % in the session pair rest1-rest2 and reverse rest2-rest1, respectively. In the identification experiment involving task-task combinations carried out on the same day, the identification accuracies ranged from 94.2 % to 98.8 %. Furthermore, we showed the Frontoparietal and Default networks make the most significant contributions to individual identification and the edges that significantly contribute to individual identification are found within and between the Frontoparietal and Default networks. Additionally, high-level cognitive behaviors can also be better predicted with the obtained refined connectomes, suggesting that higher fingerprinting can be useful for resulting in higher behavioral associations. In summary, our proposed framework provides a promising approach to use functional connectivity networks for studying cognition and behavior, promoting a deeper understanding of brain functions.

To assess the energy saving and emission reduction effects of recycling technology, the quantitative analysis of energy consumption and carbon emission of recycled asphalt pavement is conducted of asphalt mixture production, transportation, paving and compaction. Firstly, the calculation model for energy consumption and carbon emissions of recycled asphalt pavement is established during the construction stage. In addition, the energy consumption and carbon emission of four recycling technologies, including the HCPR, HIR, CCPR and CIR, were compared. Finally, the FCE method is used to evaluate the actual effect of energy saving and emission reduction from the perspectives of the ecological benefit and the economic benefit. The research results indicate the most serious stage of energy consumption is the new material production stage, and the most serious stage of carbon emission is the asphalt mixture production stage of recycled asphalt pavement. Taking the HCPR technology as an example are calculated in detail. The energy consumption of the HCPR technology in the four stages of the construction stage was 17.59 MJ, 344.45 MJ, 228.51 MJ and 25.85 MJ, respectively, and the emissions in the four stages of the construction stage were 1307.89 g, 5971.28 g, 19,251.24 g and 1949.82 g, respectively. For different recycling technologies in the construction stage, the energy-saving ratios of HCPR, HIR and CCPR are 13.62%, 9.61% and 19.24% respectively when compared with the typical scheme, which have a certain energy-saving effect. However, the energy consumption of CIR reduced by 43.63%, which the energy saving effect is most significant. Moreover, the emission reduction ratios of HCPR, HIR and CCPR are 2.3%, 9.23% and 1.01% respectively when compared with the typical scheme, the emission of CIR reduced by 30.76%, so the emission reduction effect of CIR is most significant. Furthermore, the comprehensive evaluation score of the CIR technology with the most significant energy saving and emission reduction effect is 0.8021 in terms of ' ecological benefits ' and 0.8074 in terms of ' economic benefits '. In summary, it is recommended to adopt CIR technology in the engineering project, which can achieve the most significant energy-saving and emission reduction effects.
