1. Four HD 209458 b transits through CRIRES+: Detection of H$_2$O and non-detections of C$_2$H$_2$, CH$_4$, and HCN

Abstract

HD 209458 b is one of the most studied exoplanets to date. Despite this, atmospheric characterisation studies yielded inconsistent species detections and abundances. Values reported for the C/O ratio range from 0.1 to 1.0. Of particular interest is the simultaneous detection of H2O and HCN reported by some studies using high-resolution ground-based observations, which would require the atmospheric C/O ratio to be fine-tuned to a narrow interval around 1. HCN has however not been detected from recent space-based observations. We aim to provide an independent study of HD 209458 b's atmosphere with high-resolution observations, in order to infer the presence of several species, including H2O and HCN. We observed four primary transits of HD 209458 b at a high resolution (R=92000) with CRIRES+ in the near infrared (band H, 1.4--1.8 um). After reducing the data with pycrires, we prepared the data using the SysRem algorithm and performed a cross-correlation (CCF) analysis of the transmission spectra. We also compared the results with those obtained from simulated datasets constructed by combining the Exo-REM self-consistent model with the petitRADTRANS package. Combining the four transits, we detect H2O with a signal-to-noise CCF metric of 8.7. This corresponds to a signal emitted at $K_p=151.3^{+31.1}_{-23.4}$ km/s and blueshifted by $-6^{+1}_{-2}$ km/s, consistent with what is expected for HD 209458 b. We do not detect any other species among C2H2, CH4, CO, CO2, H2S, HCN, and NH3. Comparing this with our simulated datasets, this result is consistent with a C/O ratio of 0.1 and an opaque cloud top pressure of 50 Pa, at a 3 times solar metallicity. This would also be consistent with recent JWST observations. However, none of the simulated results obtained with a bulk C/O ratio of 0.8, a value suggested by previous studies using GIANO-B and CRIRES, are consistent with our observations., Comment: 15 pages, 13 figures, 5 tables, accepted for publication in Astronomy & Astrophysics

Abstract

Young brown dwarfs exhibit atmospheric characteristics similar to those of super-Jupiters, providing a unique opportunity to study planetary atmospheres. The ESO SupJup Survey, utilizing CRIRES$^+$ on the Very Large Telescope, aims to assess the role of $^{12}$C/$^{13}$C as a formation tracer. We present observations of three young brown dwarfs: 2MASS J12003792-7845082, TWA 28, and 2MASS J08561384-1342242, with the goal of constraining their chemical compositions, thermal profiles, surface gravities, spin rotations, and $^{12}$C/$^{13}$C. Atmospheric retrievals of CRIRES$^+$ K-band spectra were conducted using the radiative transfer code petitRADTRANS coupled with the Bayesian inference algorithm MultiNest, resulting in a detailed characterization of the atmospheres of these objects. We report the volume mixing ratios of main molecular and atomic species, including the novel detection of hydrogen fluoride (HF) in a brown dwarf's atmosphere, and determine $^{12}$C/$^{13}$C values of $81^{+28}_{-19}$ and $79^{+20}_{-14}$ in the atmospheres of TWA 28 and J0856, respectively, with strong significance ($>3\sigma$). Tentative evidence ($\sim 2\sigma$) of $^{13}$C in J1200 was found, with $^{12}$C/$^{13}$C = $114^{+69}_{-33}$, along with $^{18}$O detected at moderate significance in J0856 (3.3$\sigma$) and TWA 28 (2.1$\sigma$). The retrieved thermal profiles indicate hot atmospheres (2300-2600 K) with low surface gravities and slow spins, consistent with young objects. The consistent carbon isotope ratios among the three objects, showing no significant deviation from the local ISM, suggest a fragmentation-based formation mechanism similar to star formation. The tentative detection of $^{18}$O in two objects highlights the potential of high-resolution spectroscopy to probe additional isotope ratios, such as $^{16}$O/$^{18}$O, in the atmospheres of brown dwarfs and super-Jupiters., Comment: Published in A&A Volume 689, September 2024

Abstract

It has been proposed that the distinct formation and evolution of exoplanets and brown dwarfs may affect the chemical and isotopic content of their atmospheres. Recent work has indeed shown differences in the $^{12}$C/$^{13}$C isotope ratio, provisionally attributed to the top-down formation of brown dwarfs and the core accretion pathway of super-Jupiters. The ESO SupJup Survey aims to disentangle the formation pathways of isolated brown dwarfs and planetary-mass companions using chemical and isotopic tracers. The survey uses high-resolution spectroscopy with the recently upgraded VLT/CRIRES$^+$ spectrograph, covering a total of 49 targets. Here, we present the first results: an atmospheric characterisation of DENIS J0255-4700, an isolated brown dwarf near the L-T transition. We analyse its K-band spectrum using a retrieval framework where the radiative transfer code petitRADTRANS is coupled to PyMultiNest. Gaussian Processes are employed to model inter-pixel correlations and we adopt an updated parameterisation of the PT-profile. Abundances of CO, H$_2$O, CH$_4$, and NH$_3$ are retrieved for this fast-rotating L-dwarf. The ExoMol H$_2$O line list provides a significantly better fit than that of HITEMP. A free-chemistry retrieval is strongly favoured over equilibrium chemistry, caused by an under-abundance of CH$_4$. The free-chemistry retrieval constrains a super-solar C/O-ratio of $\sim0.68$ and a solar metallicity. We find tentative evidence ($\sim3\sigma$) for the presence of $^{13}$CO, with a constraint on the isotope ratio of $\mathrm{^{12}C/^{13}C}=184^{+61}_{-40}$ and a lower limit of $\gtrsim97$, suggesting a depletion of $^{13}$C compared to the interstellar medium ($\sim68$). High-resolution, high signal-to-noise K-band spectra provide an excellent means to constrain the chemistry and isotopic content of sub-stellar objects, as is the main objective of the ESO SupJup Survey., Comment: Submitted to A&A

Abstract

SPHERE+ is a proposed upgrade of the SPHERE instrument at the VLT, which is intended to boost the current performances of detection and characterization for exoplanets and disks. SPHERE+ will also serve as a demonstrator for the future planet finder (PCS) of the European ELT. The main science drivers for SPHERE+ are 1/ to access the bulk of the young giant planet population down to the snow line ($3-10$ au), to bridge the gap with complementary techniques (radial velocity, astrometry); 2/ to observe fainter and redder targets in the youngest ($1-10$\,Myr) associations compared to those observed with SPHERE to directly study the formation of giant planets in their birth environment; 3/ to improve the level of characterization of exoplanetary atmospheres by increasing the spectral resolution in order to break degeneracies in giant planet atmosphere models. Achieving these objectives requires to increase the bandwidth of the xAO system (from $\sim$1 to 3\,kHz) as well as the sensitivity in the infrared (2 to 3\,mag). These features will be brought by a second stage AO system optimized in the infrared with a pyramid wavefront sensor. As a new science instrument, a medium resolution integral field spectrograph will provide a spectral resolution from 1000 to 5000 in the J and H bands. 5. Searching for the origin of the Ehrenreich effect in ultra-hot Jupiters: Evidence for strong C/O gradients in the atmosphere of WASP-76b?

Abstract

Extreme temperature contrasts between the day and nightside of ultra-hot Jupiters result in significantly asymmetric atmospheres, with a large expansion occurring over a small range of longitude around the terminator. Over the course of a transit, WASP-76b rotates by about 30 degree, changing the observable part of the atmosphere and invoking variations in the appearance of its constituents. As recently reported, this results in time-variable effects in the neutral iron signal, which are amplified by its possible condensation on the nightside. Here, we study the presence of molecular signals during a transit of WASP-76b observed with the CARMENES spectrograph and compare the contributions from this planet's morning and evening terminators. The results are somewhat puzzling, with formal detections of water vapor (5.5$\sigma$) and hydrogen cyanide (5.2$\sigma$) but at significantly different positions in the K$_p$-V$_{sys}$ diagram, with a blueshift of -14.3 $\pm$ 2.6 km/s and a redshift of $+$20.8 $^{+7.8}_{-3.9}$ km/s respectively, and a higher K$_p$ than expected. The H$_2$O signal also appears stronger later on in the transit, in contrast to that of HCN, which seems stronger early on. We tentatively explain this by silicate clouds forming and raining out on the nightside, partially removing oxygen from the upper atmosphere. For C/O values between 0.7 and 1, this leads to the formation of HCN at the morning limb. At the evening terminator, with the sequestered oxygen being returned to the gas phase due to evaporation, these C/O values lead to formation of H$_2$O instead of HCN. If confirmed, these results indicate that individual molecules trace different parts of the atmosphere, as well as nightside condensation, allowing spatial characterization. Abstract

Context. High-resolution spectrographs fed by adaptive optics (AO) provide a unique opportunity to characterize directly imaged exoplanets. Observations with such instruments allow us to probe the atmospheric composition, spin rotation, and radial velocity of the planet, thereby helping to reveal information on its formation and migration history. The recent upgrade of the Cryogenic High-Resolution Infrared Echelle Spectrograph (CRIRES+) at the VLT makes it a highly suitable instrument for characterizing directly imaged exoplanets. Aims. In this work, we report on observations of β Pictoris b with CRIRES+ and use them to constrain the planets atmospheric properties and update the estimation of its spin rotation. Methods. The data were reduced using the open-source pycrires package. We subsequently forward-modeled the stellar, planetary, and systematic contribution to the data to detect molecules in the planet's atmosphere. We also used atmospheric retrievals to provide new constraints on its atmosphere. Results. We confidently detected water and carbon monoxide in the atmosphere of β Pictoris b and retrieved a slightly sub-solar carbon-to-oxygen ratio, which is in agreement with previous results. The interpretation is hampered by our limited knowledge of the C/O ratio of the host star. We also obtained a much improved constraint on its spin rotation of 19.9 ± 1.0 kms−1, which gives a rotation period of 8.7 ± 0.8 h, assuming no obliquity. We find that there is a degeneracy between the metallicity and clouds, but this has minimal impact on the retrieved C/O, υ sin i, and radial velocity. Our results show that CRIRES+ is performing well and stands as a highly useful instrument for characterizing directly imaged planets. [ABSTRACT FROM AUTHOR]

Abstract

International audience; Objectives: Our aim was to assess if switching from a protease inhibitors (PI)-based regimen to a PI-free one is associated with an increased risk of Kaposi Sarcoma (KS) relapse among patients living with HIV (PLHIV) with history of KS and controlled HIV replication.Methods: In a retrospective analysis of the prospectively collected Dat'AIDS database we selected patients who both had a past KS history and a HIV-1 viral load below 200 copies/mL while being PI-treated. We searched for KS relapses while persistent virological success was maintained for at least 6 months, whether patients kept taking the PI, or switched to PI-free regimen.Results: Among the 216 patients with past KS event and a history of HIV-1 infection efficiently treated by a PI-based regimen, 148 patients (68.5%) later switched to a PI-sparing regimen. Their baseline characteristics were not different from non-switching patients. We described 7 cases of relapse (3.2% of the 216 patients). Five cases of relapse occurred in switching patients (3.4%). The remaining two relapses occurred in PI-treated patients (2.9%). At KS relapse, CD4 cell count was 459 cells/μL (range 225-560) for switching patients, compared with 362 and 136 cells/μL for the other two patients.Conclusions: In this large cohort of PLHIV with a history of KS and ART-controlled HIV replication, KS relapses were described in 3.2% of the patients, and were not more frequent when a PI-containing ART regimen has been switched to a PI-free regimen. Our results do not support a specific effect of PI on KS.

Abstract

High-contrast imaging instruments need extreme wavefront control to directly image exoplanets. This requires highly sensitive wavefront sensors which optimally make use of the available photons to sense the wavefront. Here, we propose to numerically optimize Fourier-filtering wavefront sensors using automatic differentiation. First, we optimize the sensitivity of the wavefront sensor for different apertures and wavefront distributions. We find sensors that are more sensitive than currently used sensors and close to the theoretical limit, under the assumption of monochromatic light. Subsequently, we directly minimize the residual wavefront error by jointly optimizing the sensing and reconstruction. This is done by connecting differentiable models of the wavefront sensor and reconstructor and alternatingly improving them using a gradient-based optimizer. We also allow for nonlinearities in the wavefront reconstruction using Convolutional Neural Networks, which extends the design space of the wavefront sensor. Our results show that optimization can lead to wavefront sensors that have improved performance over currently used wavefront sensors. Abstract

Objectives Even in an ‘optimal’ health system, patients’ characteristics may have an impact on their care. We investigated whether age, gender and place of birth have an impact in the HIV care continuum in France, a country with a universal free healthcare system. Methods We estimated differences in the 5 year restricted mean percentage of person-time spent (i) in care, (ii) receiving ART and (iii) on ART and virally suppressed among 2432 (30.2%) women, 3925 MSM (48.7%) and 1709 men who have sex with women (MSW; 21.2%) entering care in the Dat’AIDS French prospective cohort between 1 January 2013 and 31 December 2017. Trial registration: Clinicaltrials.gov reference NCT02898987. Results Men and women spent 85.6% and 82.8% of person-time on ART and 69.9% and 65% suppressed, respectively. MSM, MSW and women spent 86.9%, 82.6% and 82.8% of person-time on ART and 72.5%, 63.7% and 65% suppressed, respectively. Patients born in France (47%) and patients born abroad spent 87.9% and 81.9% of person-time on ART and 74.6% and 62.9% suppressed, respectively. Young men born abroad were found to spend the smallest person-time with non-detectable viral load (53% for MSW and 58.1% for MSM). Conclusions Despite free access to care and universal ART in France, disparities remain in the HIV continuum care across age, country of birth and way of HIV acquisition. Clinical and public health interventions targeting specific patients’ conditions are needed.

Abstract

Philadelphia chromosome-like acute lymphoblastic leukemia (Ph-like ALL) is a high-risk ALL subtype with high rates of relapse and poor patient outcome. Activating mutations affecting components of the JAK-STAT signaling pathway occur in the majority of Ph-like ALL cases. The use of JAK inhibitors represents a potential treatment option for Ph-like ALL, although we and others have shown that CRLF2-rearranged Ph-like ALL responds poorly to single-agent JAK inhibitors in the preclinical setting. Therefore, the aim of this study was to identify effective combination treatments against CRLF2-rearranged Ph-like ALL, and to elucidate the underlying mechanisms of synergy. We carried out a series of high-throughput combination drug screenings and found that ruxolitinib exerted synergy with standard-of-care drugs used in the treatment of ALL. In addition, we investigated the molecular effects of ruxolitinib on Ph-like ALL by combining mass spectrometry phosphoproteomics with gene expression analysis. Based on these findings, we conducted preclinical in vivo drug testing and demonstrated that ruxolitinib enhanced the in vivo efficacy of an induction-type regimen consisting of vincristine, dexamethasone, and l-asparaginase in 2/3 CRLF2-rearranged Ph-like ALL xenografts. Overall, our findings support evaluating the addition of ruxolitinib to conventional induction regimens for the treatment of CRLF2-rearranged Ph-like ALL.

Abstract

Current and future high-contrast imaging instruments require extreme Adaptive Optics (XAO) systems to reach contrasts necessary to directly image exoplanets. Telescope vibrations and the temporal error induced by the latency of the control loop limit the performance of these systems. Optimization of the (predictive) control algorithm is crucial in reducing these effects. We describe how model-free Reinforcement Learning can be used to optimize a Recurrent Neural Network controller for closed-loop adaptive optics control. We verify our proposed approach for tip-tilt control in simulations and a lab setup. The results show that this algorithm can effectively learn to suppress a combination of tip-tilt vibrations. Furthermore, we report decreased residuals for power-law input turbulence compared to an optimal gain integrator. Finally, we demonstrate that the controller can learn to identify the parameters of a varying vibration without requiring online updating of the control law. We conclude that Reinforcement Learning is a promising approach towards data-driven predictive control; future research will apply this approach to the control of high-order deformable mirrors, 15 pages, 6 figures, submitted to SPIE Astronomical Telescopes + Instrumentation (2020)

Abstract

Background The arrival of highly effective, well-tolerated, direct-acting antiviral agents (DAA) led to a dramatic decrease in hepatitis C virus (HCV) prevalence. Human immunodeficiency virus (HIV)-HCV–coinfected patients are deemed a priority population for HCV elimination, while a rise in recently acquired HCV infections in men who have sex with men (MSM) has been described. We describe the variations in HIV-HCV epidemiology in the French Dat’AIDS cohort. Methods This was a retrospective analysis of a prospective cohort of persons living with HIV (PLWH) from 2012 to 2018. We determined HCV prevalence, HCV incidence, proportion of viremic patients, treatment uptake, and mortality rate in the full cohort and by HIV risk factors. Results From 2012 to 2018, 50 861 PLWH with a known HCV status were followed up. During the period, HCV prevalence decreased from 15.4% to 13.5%. HCV prevalence among new HIV cases increased from 1.9% to 3.5% in MSM but remained stable in other groups. Recently acquired HCV incidence increased from 0.36/100 person-years to 1.25/100 person-years in MSM. The proportion of viremic patients decreased from 67.0% to 8.9%. MSM became the first group of viremic patients in 2018 (37.9%). Recently acquired hepatitis represented 59.2% of viremic MSM in 2018. DAA treatment uptake increased from 11.4% to 61.5%. More treatments were initiated in MSM in 2018 (41.2%) than in intravenous drug users (35.6%). In MSM, treatment at the acute phase represented 30.0% of treatments in 2018. Conclusions A major shift in HCV epidemiology was observed in PLWH in France from 2012 to 2018, leading to a unique situation in which the major group of HCV transmission in 2018 was MSM. Abstract

International audience; Background - In late 2013, France was one of the first countries to recommend initiation of combination antiretroviral therapy (cART) irrespective of CD4 cell count. Methods - To assess the impact of achieving the second and third Joint United Nations Programme on HIV/AIDS 90-90-90 targets (ie, 90% of diagnosed people on sustained cART, and, of those, 90% virologically controlled) on human immunodeficiency virus (HIV) incidence, we conducted a longitudinal study to describe the epidemiology of primary HIV infection (PHI) and/or recent HIV infection (patients with CD4 cell count ≥500/mm3 at HIV diagnosis; (PRHI) between 2007 and 2017 in a large French multicenter cohort. To identify changes in trends in PHI and PRHI, we used single breakpoint linear segmented regression analysis. Results - During the study period, 61 822 patients were followed in the Dat'AIDS cohort; 2027 (10.0%) had PHI and 7314 (36.1%) had PRHI. The second and third targets were reached in 2014 and 2013, respectively. The median delay between HIV diagnosis and cART initiation decreased from 9.07 (interquartile range [IQR], 1.39-33.47) months in 2007 to 0.77 (IQR, 0.37-1.60) months in 2017. A decrease in PHI (-35.1%) and PRHI (-25.4%) was observed starting in 2013. The breakpoints for PHI and PRHI were 2012.6 (95% confidence interval [CI], 2010.8-2014.4) and 2013.1 (95% CI, 2011.3-2014.8), respectively. Conclusions - Our findings show that the achievements of 2 public health targets in France and the early initiation of cART were accompanied by a reduction of about one-third in PHI and PRHI between 2013 and 2017. Abstract

International audience; Background - Although antiretroviral therapy (ART) has reduced the risk of Kaposi sarcoma (KS), KS cases still occur in HIV-infected people. Objective - To describe all KS cases observed between 2010 and 2015 in a country with high ART coverage. Methods - Retrospective study using longitudinal data from 44 642 patients in the French Dat'AIDS multicenter cohort. Patients' characteristics were described at KS diagnosis according to ART exposure and to HIV-plasma viral load (HIV-pVL) (≤50 or >50) copies/mL. Results - Among the 209 KS cases diagnosed during the study period, 33.2% occurred in ART naïve patients, 17.3% in ART-experienced patients and 49.5% in patients on ART, of whom 23% for more than 6 months. Among these patients, 24 (11.5%) had HIV-pVL ≤50 cp/mL, and 16 (66%) were treated with a boosted-PI-based regimen. The distribution of KS localization did not differ by ART status nor by year of diagnosis. Limitations - Data on human herpesvirus 8, treatment modalities for KS and response rate were not collected. Conclusion - Half of KS cases observed in the study period occurred in patients not on ART, reflecting the persistence of late HIV diagnosis. Factors associated with KS in patients on ART with HIV-pVL ≤50 cp/mL remain to be explored.

Abstract

Background We assessed prevalence of multimorbidity (MM) according to year of human immunodeficiency virus (HIV) diagnosis in elderly people living with HIV (PLWH). Methods This was a cross-sectional study of MM in PLWH aged ≥70 years from the Dat’AIDS French multicenter cohort. MM was defined as at least 3 coexistent morbidities of high blood pressure, diabetes mellitus, osteoporosis, non-AIDS cancer, chronic renal failure, cardiovascular and cerebrovascular disease, obesity, undernutrition, or hypercholesterolemia. Logistic regression models evaluated the association between MM and calendar periods of HIV diagnosis (1983–1996, 1997–2006, and 2007–2018). The secondary analysis evaluated MM as a continuous outcome, and a sensitivity analysis excluded PLWH with nadir CD4 count Results Between January 2017 and September 2018, 2476 PLWH were included. Median age was 73 years, 75% were men, median CD4 count was 578 cells/μL, and 94% had controlled viremia. MM prevalence was 71%. HBP and hypercholesterolemia were the most prevalent comorbidities. After adjustment for age, gender, smoking status, hepatitis C and hepatitis B virus coinfection, group of exposure, nadir CD4 count, CD4:CD8 ratio, and last CD4 level, calendar period of diagnosis was not associated with MM (P = .169). MM was associated with older age, CD4/CD8 ratio Conclusions MM prevalence was high and increased with age, low CD4/CD8 ratio, and nadir CD4 count
