Abstract

Simple Summary: This research focuses on finding more efficient and cost-effective methods for the development of therapeutic agents to treat malignant tumors. The study explores the design of special molecules called aptamers, which can bind to cancer cells and help in diagnosis and treatment. However, creating these molecules can be expensive and technically challenging. Using computer modeling, a way was found to simplify these molecules, reducing unnecessary parts while keeping their effectiveness intact. This method not only cuts down on production costs but also improves how quickly and precisely these molecules can work within the body. This approach promises to benefit researchers in developing targeted cancer therapies, even when detailed information about the cancer targets is lacking. Background: Oncological diseases are a major focus in medicine, with millions diagnosed each year, leading researchers to seek new diagnostic and treatment methods. One promising avenue is the development of targeted therapies and rapid diagnostic tests using recognition molecules. The pharmaceutical industry is increasingly exploring nucleic acid-based therapeutics. However, producing long oligonucleotides, especially aptamers, poses significant production challenges. Objectives: This study aims to demonstrate the efficacy of using molecular modeling, supported by experimental procedures, for altering aptamer nucleotide sequences while maintaining their binding capabilities. The focus is on reducing production costs and enhancing binding dynamics by removing nonfunctional regions and minimizing nonspecific binding. Methods: A molecular modeling approach was employed to elucidate the structure of a DNA aptamer, Gli-55, facilitating the truncation of nonessential regions in the Gli-55 aptamer, which selectively binds to glioblastoma (GBM). This process aimed to produce a truncated aptamer, Gli-35, capable of forming similar structural elements to the original sequence with reduced nonspecific binding. The efficiency of the truncation was proved by flow cytometry, fluorescence polarization (FP), and confocal microscopy. Results: The molecular design indicated that the new truncated Gli-35 aptamer retained the structural integrity of Gli-55. In vitro studies showed that Gli-35 had a binding affinity comparable to the initial long aptamer while the selectivity increased. Gli-35 internalized inside the cell faster than Gli-55 and crossed the blood–brain barrier (BBB), as demonstrated in an in vitro model. Conclusions: The success of this truncation approach suggests its potential applicability in scenarios where molecular target information is limited. The study highlights a strategic and resource-efficient methodology for aptamer development. By employing molecular modeling and truncation, researchers can reduce production costs and avoid trial and error in sequence selection. Abstract

Colloidal solutions of nanoparticles (NPs) are prepared by nanosecond pulsed laser ablation (upon exposure to Nd:YAG laser radiation at a wavelength of 1064 nm with a pulse duration of 7 ns) of a copper target in water and ethyl alcohol. Their composition, structure, and optical properties are investigated. It is shown that monovalent Cu2O NPs are mainly formed in water, whereas metal Cu NPs are mainly formed in ethanol. The nonlinear transmission of colloids at wavelengths of 532 and 1064 nm is investigated. It is established that they effectively limit the nanosecond laser pulse power at a wavelength of 532 nm.

Abstract

NaSrR(BO3)2(R = Ho–Lu, Y, Sc) compounds were obtained for the first time. Their structures exhibit disordered positions of Sr2+and Na+atoms while RO6polyhedra are connected through the BO3groups. Large distances between R atoms and high transparency in the range of 250–900 nm make them promising for phosphor applications. A pathway to obtain single crystals was shown by growing NaSrY(BO3)2and NaSrYb(BO3)2by the top seeded solution growth method with Na2O-B2O3-NaF flux.

Abstract

A new SmSc(BO3)2 compound was discovered from the study of SmBO3–ScBO3 by solid-state synthesis. The obtained compound was grown from a stoichiometric melt by a spontaneous crystallization method, and crystallizes in the space group R3¯ with the cell parameters of a = 4.8951(6) Å and c = 16.3012(2) Å. In addition, phase equilibria in this system in the 900–1300 °C temperature range were studied by X-ray diffraction and thermal analyses, as well as special diffusion experiments. Abstract

Colloidal solutions of copper (I) oxide, Cu2O, were obtained by pulsed laser ablation of metallic copper target in distilled water using fundamental harmonic of Nd:YAG laser (1064 nm, 7 ns, 20 Hz). Nanocolloids obtained were applied to cotton fabric. Nanoparticles composition and structure were studied by UV-Vis and Raman spectroscopy, transmission and scanning electron microscopy, and XRD. Antibacterial activity of Cu2O nanoparticles on cotton fabric to E.coli bacteria was demonstrated in comparison with zinc oxide nanoparticles.

Abstract

Dispersions of cerium oxide nanoparticles in water, ethanol, and water-ethanol solution were synthesized for the first time using laser ablation of metallic target. The fundamental harmonic of nanosecond Nd:YAG laser was used. Nanocrystalline powders of cerium oxide were obtained from the dispersions. The average size of the crystallites was 17-19 nm. Phase composition of nanoparticles was confirmed by X-ray diffraction and Raman spectroscopy. It was found that carbon present on the surface of CeO2 particles. The materials obtained may be used as catalyst carriers for CO oxidation, and as active components of sunscreen cosmetic products.

Abstract

We selected DNA aptamers to the epithelial cell adhesion molecule (EpCAM) expressed on primary lung cancer cells isolated from the tumors of patients with non-small cell lung cancer using competitive displacement of aptamers from EpCAM by a corresponding antibody. The resulting aptamers clones showed good nanomolar affinity to EpCAM-positive lung cancer cells. Confocal microscopy imaging and spectral profiling of lung cancer tissues confirmed the same protein target for the aptamers and anti-EpCAM antibodies. Furthermore, the resulted aptamers were successfully applied for isolation and detection of circulating tumor cells in clinical samples of peripheral blood of lung cancer patients.
