The spread of antibiotic resistance is turning many of the currently used antibiotics less effective against common infections. To address this public health challenge, it is critical to enhance our understanding of the mechanisms of action of these compounds. Aminoglycoside drugs bind the bacterial ribosome, and decades of results from in vitro biochemical and structural approaches suggest that these drugs disrupt protein synthesis by inhibiting the ribosome's translocation on the messenger RNA, as well as by inducing miscoding errors. So far, however, we have sparse information about the dynamic effects of these compounds on protein synthesis inside the cell. In the present study, we measured the effect of the aminoglycosides apramycin, gentamicin, and paromomycin on ongoing protein synthesis directly in live Escherichia coli cells by tracking the binding of dye-labeled transfer RNAs to ribosomes. Our results suggest that the drugs slow down translation elongation two- to fourfold in general, and the number of elongation cycles per initiation event seems to decrease to the same extent. Hence, our results imply that none of the drugs used in this study cause severe inhibition of translocation.

The chromatin modifier PRDM2/RIZ1 is inactivated by mutation in several forms of cancer and is a putative tumor suppressor gene. Frameshift mutations in the C-terminal region of PRDM2, affecting (A)8 or (A)9 repeats within exon 8, are found in one third of colorectal cancers with microsatellite instability, but the contribution of these mutations to colorectal tumorigenesis is unknown. To model somatic mutations in microsatellite unstable tumors, we devised a general approach to perform genome editing while stabilizing the mutated nucleotide repeat. We then engineered isogenic cell systems where the PRDM2 c.4467delA mutation in human HCT116 colorectal cancer cells was corrected to wild-type by genome editing. Restored PRDM2 increased global histone 3 lysine 9 dimethylation and reduced migration, anchorage-independent growth and tumor growth in vivo. Gene set enrichment analysis revealed regulation of several hallmark cancer pathways, particularly of epithelial-to-mesenchymal transition (EMT), with VIM being the most significantly regulated gene. These observations provide direct evidence that PRDM2 c.4467delA is a driver mutation in colorectal cancer and confirms PRDM2 as a cancer gene, pointing to regulation of EMT as a central aspect of its tumor suppressive action.

With a multitude of images available on the Internet, how do we find what we are looking for? This project tries to determine how much the precision and recall of search queries is improved by using a word taxonomy on traditional Text-Based Image Search and Content-Based Image Search. By applying a word taxonomy to different data sources, a strong keyword filter and a keyword extender were implemented and tested. The results show that depending on the implementation, the precision or the recall can be increased. By using a similar approach on real life implementations, it is possible to force images with higher precisions to the front while keeping a high recall value, thus increasing the experienced relevance of image search. Med den mängd bilder som nu finns tillgänglig på Internet, hur kan vi fortfarande hitta det vi letar efter? Denna uppsats försöker avgöra hur mycket bildprecision och bildåterkallning kan öka med hjälp av appliceringen av en ordtaxonomi på traditionell Text-Based Image Search och Content-Based Image Search. Genom att applicera en ordtaxonomi på olika datakällor kan ett starkt ordfilter samt en modul som förlänger ordlistor skapas och testas. Resultaten pekar på att beroende på implementationen så kan antingen precisionen eller återkallningen förbättras. Genom att använda en liknande metod i ett verkligt scenario är det därför möjligt att flytta bilder med hög precision längre fram i resultatlistan och samtidigt behålla hög återkallning, och därmed öka den upplevda relevansen i bildsök.

Detta arbetes syfte är att undersöka hur fritidshemmet kan komplettera skolans undervisning i SO och hur möjligheter och hinder med ett sådant arbete kan se ut. För att undersöka detta har tre intervjuer med lärare i fritidshem genomförts. Deras utsagor har sedan analyserats utifrån teorier om formellt och informellt lärande samt de klassiska lärandeteorierna pragmatism och sociokulturellt lärande. Resultaten visar på att fritidshemmet kan komplettera skolans undervisning i SO på tre olika sätt: Genom tränande av sociala färdigheter, genom att väcka intresse för samhällsvetenskapliga ämnen samt genom överföring av kunskaper. De visar också att fritidshemmet inte löper risk att bli för skolifierat när verksamheten syftar till att komplettera skolans undervisning, sålänge man bygger verksamheten på valfrihet och elevgruppens intressen och behov. Man kan också genom resultaten konstatera att saker som personalresurser och tidsbrist kan utgöra hinder för att komplettera skolans undervisning i SO.

The Rotary CNC system is a novel way to position a tool in the XY-plane. The system is meant to be used in areas such as 3D-printing, laser cutting, CNC milling and other rapid prototyping equipment. Benets include lower production cost, less moving parts and a small physical footprint. This thesis has provided a "Proof of Concept" prototype of the Rotary CNC system. It has been shown that the system is a viable alternative to current CNC system designs in the eld of PCB routing.

SVT:s Uppdrag granskning sände i januari-februari 2011 två avslöjande reportage om Ikea och dess grundare Ingvar Kamprad. I den här uppsatsen görs en kvantitativ innehållsanalys av hur dagstidningarna Aftonbladet, Dagens Nyheter, Expressen, Smålandsposten och Svenska Dagbladet rapporterade kring Kamprad i 74 artiklar under efterspelet till granskningen. Forskningsfrågan är huruvida rapporteringen i ett mediedrev styrs av huvudpersonens status i samhället. Resultatet tolkas med hjälp av teorier om mediedrev och framing och jämförs med tidigare forskning om två politiska affärer: Rainer-affären 1983 och Freivalds-affären 2000. Undersökningen visar att flertalet kriterier för ett mediedrev inte stämmer in på rapporteringen kring Kamprad. Rapporteringen var inte tillräckligt omfattande, inriktningen var inte ensidig och utgången var inte oviss. Beskrivningen av Kamprad som ett nationalhelgon var relativt vanlig i de undersökta artiklarna. Däremot tycks rapporteringen inte huvudsakligen ha styrts av Kamprads status i samhället. Flera alternativa förklaringar kan finnas, däribland att affären inte var tillräckligt spektakulär, att Kamprad inte hade någon chefsposition att avgå ifrån samt att det var en tids- och resurskrävande uppgift för tidningarna att gräva fram nya uppgifter i sakfrågan.

Neural stem cell self-renewal and differentiation are tightly regulated events during CNS development, leading to cell division into new neural stem cells or the formation of neurons and glial cells. This thesis focuses on the cellular responses induced by known and novel factors in neural stem and progenitor cells (NSPCs). Platelet-derived growth factor (PDGF) signaling has previously been implicated in NSPC regulation as well as in tumor formation. In order to evaluate the differentiation process and find new regulators of NSPCs a micro-array screen was performed, evaluating transcription during normal differentiation and the effect of PDGF-AA in this process. The transcriptional profile of PDGF-AA treated NSPCs was shown to be an intermediate between the profiles of neural stem cells and their progeny. The NSPC transcriptome was also found to have similarities with that of experimental glioma. A previously non-characterized transcript, the nuclear receptor binding protein 2 (NRBP2), was identified and found to be expressed in the developing and adult mouse brain and in medulloblastoma. NRBP2 down-regulation rendered neural progenitors sensitive to induced cell death. Different PDGF ligands interact with different combinations of PDGF receptors. Therefore NSPCs were stimulated with either PDGF-AA or -BB to further evaluate cellular responses with regard to the two specific isoforms. A divergent effect between the two isoforms in long-term proliferation and cell survival was found, with PDGF-BB being the most efficient stimulator. Stem cell factor (SCF) has previously been identified as a regulator in the hematopoietic system and we showed that SCF induces a migratory response in NSPCs. In addition, SCF positively affected cell survival but had no effect on NSPC differentiation. Insights into the regulatory mechanisms involved in neural stem cell signaling are needed to develop diagnostic tools and novel treatments.

The purpose of the project is to create interest and stimulate discussions around the place and the building.In order to gain inspiration of what such a place can contain and express, a number of similar projects have been analysed, for example the Sydney Opera and the Yokohama International Port Terminal.A number of suggestions was then created and analysed, all of which are presented in chronological order in order to show the rejections and the developed designs that resulted in the final formation to proceed with.The result was finalised in a number of presentation images of the project that can be used by Cullbergs Arkitektkontor AB when presenting the project. Examensarbetet är en vision till en identitetsbyggnad i Göteborg belägen på Bananpiren.
