Abstract: A computational method in conjunction with indentation measurements is presented to determine the yield stress and the tangential modulus of different polymeric materials. A novel approach is developed to select representative points at which finite element calculations are performed. Yield stress and tangential modulus can be obtained by minimizing the deviation between the calculated and the experimental data. Hardness values are also derived using finite element calculations and compared to those obtained from the fitting to the loading curve of depth-sensing experiments. © 1998 Elsevier Science B.V. Abstract: International audience; Following the success of the Plateau de Bure high-z Blue Sequence Survey (PHIBSS), we present the PHIBSS2 legacy program, a survey of the molecular gas properties of star-forming galaxies on and around the star-formation main sequence (MS) at different redshifts using IRAM's NOrthern Extended Millimeter Array (NOEMA). This survey significantly extends the existing sample of star-forming galaxies with CO molecular gas measurements, probing the peak epoch of star formation (z = 1 − 1.6) as well as its building-up (z = 2 − 3) and winding-down (z = 0.5 − 0.8) phases. The targets are drawn from the well-studied GOODS, COSMOS, and AEGIS cosmological deep fields and uniformly sample the MS in the stellar mass (M)-star formation rate (SFR) plane with log(M /M) = 10 − 11.8 and SFR = 3.5 − 500 M yr −1 without morphological selection, thus providing a statistically meaningful census of star-forming galaxies at different epochs. We describe the survey strategy and sample selection before focusing on the results obtained at redshift z = 0.5 − 0.8, where we report 60 CO(2-1) detections out of 61 targets. We determine molecular gas masses between 2 × 10 9 and 5 × 10 10 M and separately obtain disc sizes and bulge-to-total (B/T) luminosity ratios from HST I-band images. The median molecular gas-to-stellar mass ratio µ gas = 0.28 ± 0.04, gas fraction f gas = 0.22 ± 0.02, and depletion time t depl = 0.84 ± 0.07 Gyr as well as their dependence with stellar mass and offset from the MS follow published scaling relations for a much larger sample of galaxies spanning a significantly wider range of redshifts, the cosmic evolution of the SFR being mainly driven by that of the molecular gas fraction. The galaxy-averaged molecular Kennicutt-Schmidt (KS) relation between molecular gas and SFR surface densities is strikingly linear, pointing towards similar star formation timescales within galaxies at any given epoch. In terms of morphology, the molecular gas content, the SFR, the disc stellar mass, and the disc molecular gas fraction do not seem to correlate with B/T and the stellar surface density, which suggests an ongoing supply of fresh molecular gas to compensate for the build-up of the bulge. Our measurements do not yield any significant variation of the depletion time with B/T and hence no strong evidence for morphological quenching within the scatter of the MS.

Abstract: We present deep high resolution (0.03", 200pc) ALMA Band 7 observations covering the dust continuum and [CII] $\lambda157.7\mu$m emission in four $z\sim4.4-4.8$ sub-millimeter galaxies (SMGs) selected from the ALESS and AS2UDS surveys. The data show that the rest-frame 160$\mu$m (observed 345 GHz) dust emission is consistent with smooth morphologies on kpc scales for three of the sources. One source, UDS47.0, displays apparent substructure but this is also consistent with a smooth morphology, as indicated by simulations showing that smooth exponential disks can appear clumpy when observed at high angular resolution (0.03") and depth of these observations ($\sigma_{345\text{GHz}} \sim27-47\mu$Jy beam$^{-1}$). The four SMGs are bright [CII] emitters, and we extract [CII] spectra from the high resolution data, and recover $\sim20-100$% of the [CII] flux and $\sim40-80$% of the dust continuum emission, compared to the previous lower resolution observations. When tapered to 0.2" resolution our maps recover $\sim80-100$% of the continuum emission, indicating that $\sim60$% of the emission is resolved out on $\sim200$pc scales. We find that the [CII] emission in high-redshift galaxies is more spatially extended than the rest-frame 160$\mu$m dust continuum by a factor of $1.6\pm0.4$. By considering the $L_{\text{[CII]}}$/$L_{\text{FIR}}$ ratio as a function of the star-formation rate surface density ($\Sigma_{\text{SFR}}$) we revisit the [CII] deficit, and suggest that the decline in the $L_{\text{[CII]}}$/$L_{\text{FIR}}$ ratio as a function of $\Sigma_{\text{SFR}}$ is consistent with local processes. Abstract: We present the "SINS/zC-SINF AO survey" of 35 star-forming galaxies, the largest sample with deep adaptive optics-assisted (AO) near-infrared integral field spectroscopy at z~2. The observations, taken with SINFONI at the Very Large Telescope, resolve the Ha and [NII] line emission and kinematics on scales of ~1.5 kpc. In stellar mass, star formation rate, rest-optical colors and size, the AO sample is representative of its parent seeing-limited sample and probes the massive (M* ~ 2x10^9 - 3x10^11 Msun), actively star-forming (SFR ~ 10-600 Msun/yr) part of the z~2 galaxy population over a wide range in colors ((U-V)_rest ~ 0.15-1.5 mag) and half-light radii (R_e,H ~ 1-8.5 kpc). The sample overlaps largely with the "main sequence" of star-forming galaxies in the same redshift range to a similar K_AB = 23 magnitude limit; it has ~0.3 dex higher median specific SFR, ~0.1 mag bluer median (U-V)_rest color, and ~10% larger median rest-optical size. We describe the observations, data reduction, and extraction of basic flux and kinematic properties. With typically 3-4 times higher resolution and 4-5 times longer integrations (up to 23hr) than the seeing-limited datasets of the same objects, the AO data reveal much more detail in morphology and kinematics. The now complete AO observations confirm the majority of kinematically-classified disks and the typically elevated disk velocity dispersions previously reported based on subsets of the data. We derive typically flat or slightly negative radial [NII]/Ha gradients, with no significant trend with global galaxy properties, kinematic nature, or the presence of an AGN. Azimuthal variations in [NII]/Ha are seen in several sources and are associated with ionized gas outflows, and possible more metal-poor star-forming clumps or small companions. Abstract: We present Karl G. Jansky Very Large Array observations of the CO J=1-0 transition in a sample of four $z\sim2$ main sequence galaxies. These galaxies are in the blue sequence of star-forming galaxies at their redshift, and are part of the IRAM Plateau de Bure HIgh-$z$ Blue Sequence Survey (PHIBSS) which imaged them in CO J=3-2. Two galaxies are imaged here at high signal-to-noise, allowing determinations of their disk sizes, line profiles, molecular surface densities, and excitation. Using these and published measurements, we show that the CO and optical disks have similar sizes in main-sequence galaxies, and in the galaxy where we can compare CO J=1-0 and J=3-2 sizes we find these are also very similar. Assuming a Galactic CO-to-H$_2$ conversion, we measure surface densities of $\Sigma_{mol}\sim1200$ M$_\odot$pc$^{-2}$ in projection and estimate $\Sigma_{mol}\sim500-900$ M$_\odot$pc$^{-2}$ deprojected. Finally, our data yields velocity-integrated Rayleigh-Jeans brightness temperature line ratios $r_{31}$ that are approximately unity. In addition to the similar disk sizes, the very similar line profiles in J=1-0 and J=3-2 indicate that both transitions sample the same kinematics, implying that their emission is coextensive. We conclude that in these two main sequence galaxies there is no evidence for significant excitation gradients or a large molecular reservoir that is diffuse or cold and not involved in active star-formation. We suggest that $r_{31}$ in very actively star-forming galaxies is likely an indicator of how well mixed the star formation activity and the molecular reservoir are., Comment: To appear in ApJ

Abstract: International audience; The micro-convection caused by the ponderomotive forces of the self-magnetic field in a magnetic fluid is studied here both numerically and experimentally. The theoretical approach based on the general Brinkman model substantially improves the description with respect to the previously proposed Darcy model. The predictions of both models are here compared to finely controlled experiments. The Brinkman model, in contrast to the Darcy model, allows us to describe the formation of mushrooms on the plumes of the micro-convective flow and the width of the fingers. In the Brinkman approach, excellent quantitative agreement is also obtained for the finger velocity dynamics and the velocity maximal values as a function of the magnetic Rayleigh number. The diffusion coefficient of particles of the water-based magnetic colloid determined by the threshold field strength value of the micro-convection is significantly larger than the diffusion coefficient of individual particles. This result is confirmed by independent measurements of the diffusion coefficient at the smearing of the diffusion front.

Abstract: International audience; We explore how a magnetic field breaks the symmetry of an initially spherical giant liposome filled with a magnetic colloid. The condition of rotational symmetry along the field axis leads either to a prolate or to an oblate ellipsoid. Abstract: We propose a simple micro-rheology technique to evaluate the viscoelastic properties of complex fluids. The method is based on the use of magnetic wires of a few microns in length submitted to a rotational magnetic field. In this work, the method is implemented on a surfactant wormlike micellar solution that behaves as an ideal Maxwell fluid. With increasing frequency, the wires undergo a transition between a steady and a hindered rotation regime. The study shows that the average rotational velocity and the amplitudes of the oscillations obey scaling laws with well-defined exponents. Abstract: International audience; Relaxors constitute a large class of ferroelectrics where disorder is introduced by doping with ions of different size and valence, in order to maximize their useful properties in a broad temperature range. Polarization disorder in relaxors is typically studied by dielectric and scattering techniques that do not allow direct mapping of relaxor parameters, such as correlation length or width of the relaxation time spectrum. In this paper, we introduce a novel method based on measurements of local vibrations by Piezoresponse Force Microscopy (PFM) that detects nanoscale polarization on the relaxor surface. Random polarization patterns are then analyzed via local Fast Fourier Transform (FFT) and the FFT PFM parameters, such as amplitude, correlation radius and width of the spectrum of spatial correlations, are mapped along with the conventional topography. The results are tested with transparent (Pb, La) (Zr, Ti)O3 ceramics where local disorder is due to doping with La3+. The conclusions are made about the distribution of the defects responsible for relaxor behavior and the role of the grain boundaries in the macroscopic response.

Abstract: International audience; Magnetic field-driven domain-wall propagation in the flow regime is investigated in (Ga, Mn) As ferromagnetic semiconductor layers. Square-shape magnetic domains with an unexpected orientation of their edges, at pi/8 with respect to the anisotropy axes, are found. This is shown to arise from the effect of tetragonal magnetic anisotropy on domain-wall dynamics. Using a one-dimensional model for domain-wall motion and modeling domain growth by contour dynamics the shape and orientation of domains and their field range for existence are well reproduced. These results point to the key role of the vectorial nature of the order parameter in the dynamics of ferromagnetic domains.

Both superconducting (SC) and normal state (NS) domains are analyzed using magneto-optical imaging. We measure the average width (diameter) of lamellar (circular) domains. The experimental values and predicted equilibrium values are shown to present a significantly better agreement when the screening of the magnetic flux is taken into account. On this basis, we analyze the contribution of the elongation instability of circular (NS and SC) domains on the formation of the intermediate state. Abstract: A synthesis of enantiopure thiourea organocatalyst based on (-)-(S)-3-aminoquinuclidine dihydrochloride was developed with quantitative product yield. The catalyst was tested in different reactions: asymmetric Michael addition of ketones and malonates to nitroalkenes, nitromethane 1,4-addition to trans-chalcone, and Friedel-Crafts alkylation of indoles with trans-β-nitrostyrene. The novel thiourea proved to catalyze the aforementioned reactions and expected products were obtained in mediocre yields and low enantioselectivities.

Abstract: We use the first systematic samples of CO millimeter emission in z>1 'main-sequence' star forming galaxies (SFGs) to study the metallicity dependence of the conversion factor {\alpha}CO, from CO line luminosity to molecular gas mass. The molecular gas depletion rate inferred from the ratio of the star formation rate (SFR) to CO luminosity, is ~1 Gyr-1 for near-solar metallicity galaxies with stellar masses above M_S~1e11 M_sun. In this regime the depletion rate does not vary more than a factor of two to three as a function of molecular gas surface density, or redshift between z~0 and 2. Below M_S the depletion rate increases rapidly with decreasing metallicity. We argue that this trend is not caused by starburst events, by changes in the physical parameters of the molecular clouds, or by the impact of the fundamental metallicity-SFR-stellar mass relation. A more probable explanation is that the conversion factor is metallicity dependent and that star formation can occur in 'CO-dark' gas. The trend is also expected theoretically from the effect of enhanced photodissociation of CO by ultraviolet radiation at low metallicity. From the available z~0 and z~1-3 samples we constrain the slope of the log({\alpha}CO) -log (metallicity) relation to range between -1 and -2, fairly insensitive to the assumed slope of the gas-star formation rate relation. Because of the lower metallicities near the peak of the galaxy formation activity at z~1-2 compared to z~0, we suggest that molecular gas masses estimated from CO luminosities have to be substantially corrected upward for galaxies below M_S.

Abstract: This paper provides an update of our previous scaling relations (Genzel et al.2015) between galaxy integrated molecular gas masses, stellar masses and star formation rates, in the framework of the star formation main-sequence (MS), with the main goal to test for possible systematic effects. For this purpose our new study combines three independent methods of determining molecular gas masses from CO line fluxes, far-infrared dust spectral energy distributions, and ~1mm dust photometry, in a large sample of 1444 star forming galaxies (SFGs) between z=0 and 4. The sample covers the stellar mass range log(M*/M_solar)=9.0-11.8, and star formation rates relative to that on the MS, delta_MS=SFR/SFR(MS), from 10^{-1.3} to 10^{2.2}. Our most important finding is that all data sets, despite the different techniques and analysis methods used, follow the same scaling trends, once method-to-method zero point offsets are minimized and uncertainties are properly taken into account. The molecular gas depletion time t_depl, defined as the ratio of molecular gas mass to star formation rate, scales as (1+z)^{-0.6}x(delta_MS)^{-0.44}, and is only weakly dependent on stellar mass. The ratio of molecular-to-stellar mass mu_gas depends on (1+z)^{2.5}x (delta_MS)^{0.52}x(M*)^{-0.36}, which tracks the evolution of the specific star formation rate. The redshift dependence of mu_gas requires a curvature term, as may the mass-dependences of t_depl and mu_gas. We find no or only weak correlations of t_depl and mu_gas with optical size R or surface density once one removes the above scalings, but we caution that optical sizes may not be appropriate for the high gas and dust columns at high-z., Comment: Now accepted by ApJ; Figures and tables have been updated, and an appendix has been added

Abstract: We use VLTI/GRAVITY near-infrared interferometry measurements of 8 bright, Type 1 AGN to study the size and structure of hot dust heated by the central engine. We partially resolve each source, and report Gaussian FWHM sizes in the range 0.3-0.8 milliarcseconds. In all but one object, we find no evidence for significant elongation or asymmetry (closure phases < 1 deg). The effective physical radius increases with bolometric luminosity as found from past reverberation and interferometry measurements. The measured sizes for Seyfert galaxies are systematically larger than for the two quasars in our sample when measured relative to the previously reported R ~ L^1/2 relationship explained by emission at the sublimation radius. 22. The NOAH HSQC-COSY module revisited: A theoretical and practical comparison of pulse sequences.

Abstract: NMR supersequences, as exemplified by the NOAH (NMR by Ordered Acquisition using 1 H detection) technique, are a powerful way of acquiring multiple 2D data sets in much shorter durations. This is accomplished through targeted excitation and detection of the magnetisation belonging to specific isotopologues ('magnetisation pools'). Separately, the HSQC-COSY experiment has recently seen an increase in popularity due to the high signal dispersion in the indirect dimension and the removal of ambiguity traditionally associated with HSQC-TOCSY experiments. Here, we describe how the HSQC-COSY experiment can be integrated as a 'module' within NOAH supersequences. 23. COVID-19 Lessons and Post-pandemic Recovery: A Case of Latvia.

Abstract: The decision of EU and the response of the national governments to COVID-19 crisis provide the basis for returning "back to normal". A key challenge is the transition to economic recovery in the presence of the ongoing COVID-19 risk. Adequate policy mix and forward-looking actions of the public institutions are crucial to mitigate the devastating impact of the crisis and to preserve growth. Governments need to facilitate positive changes in the labor market, adjust the macroeconomic and fiscal regimes, and mitigate the post-crisis "fatigue" of societies. The turmoil of the EU economy is symmetrical, as the pandemic has affected all EU Member States, but the impact of the pandemic varies considerably from one country to another, as does their ability to absorb the economic crisis. Also, variation in the vaccination performance is partly due to different institutional characteristics across countries. Small countries are more vulnerable to external economic shocks; however, they can increase their resilience by efficient governance and social response. Extraordinary pandemic crisis can be seen as a stress test for the small and open Latvian economy, and it is worth analyzing the lessons that Latvia had learned and its future prospects. The aim of this paper is to evaluate the economic and social consequences of the ongoing crisis in Latvia, assess the effectiveness of the response of the government to the crisis, analyse people's perceptions, and to identify the future scenarios. The authors applied a special theoretical framework for the assessment of the effectiveness of institutions. Institutional analysis of crises response by the Latvian government reveals that the government managed to avoid serious functional disruptions; however, it failed to show convincing ability to learn by doing. The authors also provide a comprehensive analysis of the macroeconomic trends of the "COVID-sick" Latvian economy and conclude that future-oriented solutions relate to international competitiveness and that the key factor of competitiveness is a productivity renaissance. The pandemic crisis has fostered the state support for healthcare, which in Latvia for decades has been underfinanced. The right choice of fiscal instruments is crucial to accelerate the economic recovery and better healthcare. Research is based on the macroeconomic assessment and survey-based analysis. Abstract: The USA Defense Threat Reduction Agency provided data in 2014 on the health status, including cancer, of the 4,843 sailors on the nuclear-powered United States Ship (USS) Ronald Reagan over the 2.55-year period from May 12, 2011 to Dec 31, 2013. Also provided were data on a matched control group of 65,269 US Navy personnel. Examination of the control population relative to the US national data gives a relative risk for all malignancies of RR = 9.2 (95% CI 8.48 < 9.2 < 9.96). The result suggests a significant cancer risk associated with serving on a nuclear-powered ship, one which is not predicted by the science underlying current radiation protection legislation.

26. Mapping Disorder in Polycrystalline Relaxors: A Piezoresponse Force Microscopy Approach.

Abstract: Relaxors constitute a large class of ferroelectrics where disorder is introduced by doping with ions of different size and valence, in order to maximize their useful properties in a broad temperature range. Polarization disorder in relaxors is typically studied by dielectric and scattering techniques that do not allow direct mapping of relaxor parameters, such as correlation length or width of the relaxation time spectrum. In this paper, we introduce a novel method based on measurements of local vibrations by Piezoresponse Force Microscopy (PFM) that detects nanoscale polarization on the relaxor surface. Random polarization patterns are then analyzed via local Fast Fourier Transform (FFT) and the FFT PFM parameters, such as amplitude, correlation radius and width of the spectrum of spatial correlations, are mapped along with the conventional topography. Abstract: To investigate the structural and functional organization of the human interleukin-2 locus, the nucleotide sequence of 9339 bp of the 5' flanking region of this gene has been determined. Computer search analysis reveals five stretches with a high degree of homology between the human and mouse 5' flanking sequence, including a very distant 5' region. In this region additional binding sites for potential transcription factors were found that are identical to known regulatory sequences. The possible roles of these putative regulatory elements in the interleukin-2 gene regulation remain to be proven. The 5' end of the sequence contains almost full-length LINE element. LINE consensus open reading frames ORF1 and ORF2 in the reported sequence are interrupted by insertions, deletions or in-frame nonsense mutations. Comparative analysis of the ratio of codon changes that result in amino acid replacement to those that are silent revealed a high silent mutation frequency throughout the consensus ORF1 3' end, suggesting that this region is probably under selection for protein function.

Abstract: The Escherichia coli RNA phage Q beta coat protein-encoding gene (C) was amplified from native Q beta RNA using a reverse transcription-PCR technique. Gene C contains sequences coding for both the 133-amino acid (aa) Q beta coat protein (CP) and the 329-aa read-through protein (A1) consisting of CP and an additional 196-aa C-terminal sequence, separated from CP within the C gene by an opal (UGA) stop codon. Primers ensuring the natural environment for gene C, especially within the ribosome-binding site, and supplying C with unique restriction sites at both ends have been prepared. An amplified 1062-bp PCR fragment was positioned under the control of the strong E. coli trp promoter (Ptrp) within a pGEM-derived plasmid. The synthesis of gene C products was confirmed electrophoretically and immunologically. An immunodiffusion test with anti-Q beta phage antibodies and electron microscopy evaluation of the purified recombinant products showed that when expressed, the Q beta C gene was responsible for high-level synthesis and correct self-assembly of Q beta CP monomers into capsids indistinguishable morphologically and immunologically from Q beta phage particles, which we plan to use as surface display vectors.

Abstract: A structure-function analysis of the icosahedral RNA bacteriophage fr coat protein (CP) assembly was undertaken using linker-insertion, deletion and substitution mutagenesis. Mutations were specifically introduced into either pre-existing or artificially created restriction enzyme sites within fr CP gene expressed in Escherichia coli from a recombinant plasmid. This directs synthesis of wild type protein that undergoes self-assembly and forms capsid-like particles indistinguishable morphologically and immunologically from native phage particles. A series of fr CP variants containing sequence alterations in the regions which are (i) exposed on the external surface of capsid or (ii) located on the contacting areas between CP subunits were obtained and their assembly properties investigated. The majority of mutants demonstrated reduction of assembly ability and formed either CP dimers (mutations at residues 2, 10, 63 or 129) or both dimer and capsid structures (residue 2 or 69). The exceptions were variants demonstrating normal assembly and containing insertions at residues 2, 50 or 129 of the fr CP. A third type of assembled structure was formed by a variant with a single amino acid substitution I104T. The alpha A-helix region (residues 97-111) is particularly sensitive to mutation and any alteration in this region decreases accumulation of mutant protein in E. coli. The relative contributions of particular fr CP domains in maintenance of capsid structural integrity as well as the possible capsid assembly mechanism are discussed.

Abstract: From a panel of monoclonal antibodies (MoAbs) directed against E. coli-derived native and denatured hepatitis B virus (HBV) core antigen we have selected a set of specific MoAbs which recognize different linear antigenic determinants: MoAb C1-5--cl epitope; MoAb 14K8--less immunogenic N-terminal region; and MoAbs 13C9, 10F10 and 14E11, 14G3--the immunodominant region between amino acids 134 and 140. We have applied the polymerase chain reaction technique to clone Ig VH and VL region genes, and appropriate full-length cDNA clones were obtained and characterized by nucleotide sequence analysis. Among the six heavy chain variable region sequences examined, three VH families were represented. Two of them belong to the 7183 (MoAb C1-5) and 3609 (14B8) families respectively and four, having only two amino acid changes in the CDR2 region, to the J558 family. These four probably are derived from a single expanded B-cell clone. The light chain sequences indicate that their VL are encoded by V kappa 21, V kappa 19 and V kappa 3 germline genes. Unlike VH genes, light chain genes are closely related to known representatives of mouse kappa light chain families and are employed also by MoAbs raised against other antigens.

Abstract: Calves of two breeds--Brown Latvian and Black and White, were immunized against bovine leukemia virus with an immunogen produced from short-term cultured blood lymphocytes of BLV-positive Brown Latvian cows. The immunogen contained the viral proteins gp51 and p24 as well as some cellular proteins. As determined by bioassay in sheep, it did not contain infective bovine leukemia virus. The calves were challenged by BLV-producing lymphocytes 2 weeks after the immunization procedure. Effective protection against BLV infection was induced by the tested immunogen in all Brown Latvian calves for 6 months and in 70% of the Black and White calves.

Abstract: A possible approach to control of bovine lymphoproliferative disease caused by bovine leukaemia virus (BLV) may be the development of an "antiviral information immunity" based on the effect of anti-sense RNA (asRNA). A numbers of constructs were obtained, under control of various promotors (herpesvirus thymidine kinase, T-antigen SV40 promoter), carrying as DNA against gene X, the expression product of which is a transactivator of viral transcription from the BLV LTR promotor. As a model system for the analysis of antiviral activity of constructs developed, cloned continuous cell lines of BLV-producing FLK cells were used. The level of BLV expression in cells transfected with the constructs was determined by various parameters. Differences were detected in different clones obtained from non-transfected cells, as well as variation between transfected clones, as measured by reverse transcriptase, competitive radio-immunoassay for BLV p24, the viral particle count on agar membrane, and the tumorigenicity for nude mice. The differences in inhibition of expression of BLV genes and their products may be explained in terms of the site of integration of asDNA and the number of integrated copies.

Abstract: The data about changes in the molecular organization of the cell nucleus during aging are reviewed. Changes in DNA primary and secondary structure include deletions of some sequences, changes in base methylation pattern and the increasing number of DNA breaks. Conditions underlying poor reproducibility of the results obtained in corresponding experiments are discussed. Changes observed in the nucleosomal and supranucleosomal chromatin structure reflect either its increasing compactization or the loss the nucleosomal structure during aging. The data about the increased DNA superhelicity and topoisomerase activity in aging organisms contradict the accepted views about age-related decrease in chromatin activity. It is suggested that the gene activity in aging organisms is specifically altered rather than generally decreased. The data are presented about the ways of preventing or reverting the age-related changes in chromatin structure.

Abstract: 1. Early development of peptide hydrolysis in the digestive tract was investigated in experiments with fasted and fed ad lib. chicks during the first decade of postnatal period. 2. Pancreatic carboxypeptidase A (CPA) activity was maximal at the moment of hatch. On the second day CPA activity considerably diminished in starved and fed animal groups; further starvation (3-4 days) led to the significant increase of CPA total and specific activity, whereas the amount of enzyme in pancreas of fed chicks was rather low. 3. Aminopeptidase (AP) activity of the small intestinal surface was less sensitive to starvation. The increase of activity in all intestinal parts was observed only on the 4th day of fasting. The most sensitive to starvation were dipeptidases. Changes in their activity (2-fold increase) were detected after 24 hr of starvation. 4. The formation of specific physiological proximo-distal gradient of intestinal exopeptidase activities began only after the moment of the first feeding. 5. This gives evidence that the development of peptide hydrolysis depends not only on the age of the animal but also on the normal physiological beginning of the process of exogenous nutrition.

Abstract: A combined 1H-NMR and molecular mechanics study of [Cpp1, Sar7]AVP was performed in order to select the most probable conformations in DMSO solutions. The NMR constraints obtained were employed in the selection of starting conformations of the cyclic moiety of the analog. In particular, the diminished accessibility of the Asn5 NH proton to solvent and the close contact between Cpp1 and Cys6 C alpha H protons suggests a beta-turn conformation at the Phe3-Gln4 residues. Energy minimization was carried out both in the ECEPP/2 (rigid-valence geometry) and in the AMBER (flexible-valence geometry) force fields. Comparison of the experimental and calculated values of NMR characteristics has revealed that conformations containing type I, II, and III beta-turns at the Phe3-Gln4 residues are in reasonable agreement with the experimental data, with a dynamic equilibrium between the beta I (beta III) and beta II type structures of the cyclic part being the most probable. All of these conformations prefer the negative chirality of the disulfide bridge (theta 3 approximately -90 degrees). Five representative conformations were chosen for the acyclic tail: one with a beta I, one with a beta II'-turn at the Sar7-Arg8 residues, two extended-type conformations, and a conformation with a gamma-turn at Sar7. Because only high-energy extended conformations were in agreement with NMR data, it was concluded that the acyclic tail has considerable conformational flexibility in solution. The conformations obtained are discussed in terms of the structure-function relationship of the neurohypophyseal hormone analogs.

43. Inhibition of microbial growth and metabolism by excess turbulence.

Abstract: Excess turbulence caused by high-intensity stirring inhibited microbial growth and metabolism. In stirred tank bioreactors, the growth rate and lysine biosynthesis decreased in Brevibacterium flavum beyond 900 rpm, the growth rate of Trichoderma reesei on wheat straw beyond 150 rpm, and the growth rate of Saccharomyces cerevisae beyond 800 rpm. The term turbohypobiosis was introduced to describe this inhibition. Turbohypobiosis was characterized by a stress factor F(str) expressing the interaction of medium flow with microbial cells in local turbulent zones, dependent on the energy distribution of the stirring regime. Lysine synthesis was inhibited at significantly lower F(str) values than the growth of B. flavum. The main reason for the inhibition was shear effects causing decreased adenosine triphosphate (ATP) generation, lower O(2) uptake, and lower specific growth rate of bacteria.

Abstract: Mean distances between C beta atoms of redox centers and mean values of the exponentially distance dependent rate constants for intramolecular electron transfer, (k), in aqueous solution were calculated for Met5-enkephalin and its D-Ala2 analogue using the molecular mechanics and Monte Carlo techniques. The ratio of (k) values thus obtained proved similar to that determined experimentally for intramolecular electron transfer accompanying Met(S:.Br)----TyrO.radical transformation in these two peptides. This agreement indicates that the almost 2-fold difference observed between experimental k values for Met5-enkephalin and its D-Ala2 analogue can be attributed to the effect of Gly2 for D-Ala2 replacement on the average separation of the redox centers.

Abstract: Theoretical conformational analysis was carried out for several tetrapeptide analogues of beta-casomorphin and dermorphin containing a Phe residue in position 3. Sets of low-energy backbone structures of the mu-selective peptides [N-Me-Phe3, D-Pro4]-morphiceptin and Tyr-D-Orn-Phe-Asp-NH2 were obtained. These sets of structures were compared for geometrical similarity between themselves and with the low-energy conformations found for the delta-selective peptide Tyr-D-Cys-Phe-D-Pen-OH and nonactive peptide Tyr-Orn-Phe-Asp-NH2. Two pairs of geometrically similar conformations of mu-selective peptides, sharing no similarity with the conformations of peptides showing low affinity to the mu-receptor, were selected as two alternative models of probable mu-receptor-bound backbone conformations. Both models share geometrical similarity with the low-energy structures of the linear mu-selective peptide Tyr-D-Ala-Phe-Phe-NH2. Abstract: Derivative UV spectroscopic data show that the plant growth substances N6-(delta 2-isopentenyl) adenine (i6Ade) and indolylacetic acid (IAA) can bind to the yeast alcohol dehydrogenase (ADH) and affect coenzyme-enzyme binding. This is confirmed by enzyme kinetics studies. At fixed ethanol concentrations (27.8 and 111.1 mM) and varying NAD+ concentrations (0.033-2 mM), as well as at fixed levels of coenzyme (0.67 and 2 mM), and at varying concentrations of ethanol (1.4-111.1 mM), the rate of ethanol oxidation is significantly inhibited by i6Ade and IAA. The kinetics of the ADH reaction is affected by two inhibition constants (KI and K'I) which correspond to the dissociation constants of complexes EI and ESI, respectively. For i6Ade the KI = 0.52 +/- 0.06 mM and K'I = 0.74 +/- 0.07 mM, and for IAA the KI = 0.88 +/- 0.03 mM and K'I = 0.99 +/- 0.02 mM.

Abstract: [D-Arg2,Leu5]enkephalin and its analogues, shortened from the C-terminus, were studied in a rat brain membrane fraction with the aid of radioreceptor analysis. The interaction between these compounds and the peripheral opiate receptors of isolated organs (guinea pig ileum and mouse vas deferens) was also investigated. The ethyl ester of the [D-Arg2]tripeptide retains approximately 10% of the mu-receptor activity of leucine-enkephalin. This implies that for interaction with the mu-subclass of opiate receptors there is no requirement for a second aromatic ring in addition to the tyramine group in the minimal active moiety of the enkephalin molecule.
