Abstract

Fasting glucose and insulin are intermediate traits for type 2 diabetes. Here we explore the role of coding variation on these traits by analysis of variants on the HumanExome BeadChip in 60,564 non-diabetic individuals and in 16,491 T2D cases and 81,877 controls. We identify a novel association of a low-frequency nonsynonymous SNV in GLP1R (A316T; rs10305492; MAF=1.4%) with lower FG (β=-0.09±0.01 mmol l(-1), P=3.4 × 10(-12)), T2D risk (OR[95%CI]=0.86[0.76-0.96], P=0.010), early insulin secretion (β=-0.07±0.035 pmolinsulin mmolglucose(-1), P=0.048), but higher 2-h glucose (β=0.16±0.05 mmol l(-1), P=4.3 × 10(-4)). We identify a gene-based association with FG at G6PC2 (pSKAT=6.8 × 10(-6)) driven by four rare protein-coding SNVs (H177Y, Y207S, R283X and S324P). We identify rs651007 (MAF=20%) in the first intron of ABO at the putative promoter of an antisense lncRNA, associating with higher FG (β=0.02±0.004 mmol l(-1), P=1.3 × 10(-8)). Our approach identifies novel coding variant associations and extends the allelic spectrum of variation underlying diabetes-related quantitative traits and T2D susceptibility.

Abstract

High-quality draft genome sequences were determined for 10 Exiguobacterium strains in order to provide insight into their evolutionary strategies for speciation and environmental adaptation. The selected genomes include psychrotrophic and thermophilic species from a range of habitats, which will allow for a comparison of metabolic pathways and stress response genes.

25. Resistance to Opportunities of Plastic Recycling

Abstract

Plastics present a vast and pressing issue in modern society. Currently recycling efforts fall dangerously short of dealing with even a small percent of the millions of tons of plastic waste produced yearly across the globe. This article investigates resistance toward plastic recycling in three areas from both a contemporary and a historical context, highlighting the magnitude of the problem and the insufficient nature of current solutions. The three primary areas covered are the plastics problem from (1) a design perspective, (2) a material science perspective, and (3) a systems perspective. Solutions are proposed that emphasize a synergistic collaboration across disciplines and research modes. Ultimately, the conclusions point to a need for stronger engagement at the level of people (both consumers and decision makers) and reintegrating reused and recycled plastics into everyday life to build a solid foundation for success.

Abstract

An insertion in the promoter of the operon that encodes the molecular chaperone GroE was isolated as an antimutator for stationary-phase or adaptive mutation. The groE operon consists of two genes, groES and groEL; point mutations in either gene conferred the same phenotype, reducing [Lac.sup.+] adaptive mutation 10- to 20-fold. groE mutant strains had 1/10 the amount of error-prone DNA polymerase IV (Pol IV). In rec[G.sup.+] strains, the reduction in Pol IV was sufficient to account for their low rate of adaptive mutation, but in recG mutant strains, a deficiency of GroE had some additional effect on adaptive mutation. Pol IV is induced as part of the SOS response, but the effect of GroE on Pol IV was independent of LexA. We were unable to show that GroE interacts directly with Poi IV, suggesting that GroE may act indirectly. Together with previous results, these findings indicate that Pol IV is a component of several cellular stress responses.

Abstract

Researchers were able to measure the amount of ammonia-oxidizing bacteria and nitrite-oxidizing bacteria in wastewater treatment plants by using two sets of PCR primers. One was designed for a region of the amoA gene and one for 16S rDNA. Nitrospira sp. was more numerous than Nitrosomonas oligotropha.

Abstract

Tuberculosis screening at single-room-occupancy hotels may be an effective way of tracking tuberculosis patients. In New York, many residents of such facilities are AIDS patients, a group at high risk for tuberculosis. Researchers investigated tuberculosis screening strategies at 32 New York single-room-occupancy hotels. The Tuberculosis Registry was consulted to identify known cases. A group of 93 residents, of which 73 were AIDS patients, were interviewed for symptoms and given a tuberculin skin test and a chest X ray. Sputum specimens were cultured for residents with suspicious symptoms or signs. Sixteen cases of tuberculosis were identified, all in AIDS patients. All were known cases, but eight had been lost to medical follow-up. Five cases were not listed in the tuberculosis registry. Taking a history and cross-checking the registry was the most cost-effective and accurate screening technique. Skin tests and chest X-rays added to the cost without adding to the accuracy of identification.

Abstract

The role of the extra DNA fragments, which is found in the plasmids pSS60 and pSS70, in the degradation of 4-chlorobenzoic acid (4CBA) was elucidated. 4CBA is an intermediate in the biodegradation of 4-chlotobiphenyl. The study showed that Alcaligenes sp. strain ALP83 can degrade 4CBA to 4-hydroxybenzoate. Results showed that this activity was correlated with a 10-kb fragment on plasmid pSS70.

Abstract The genome of a fecal pollution indicator phage, Bacteroides fragilis ATCC 51477-B1, was sequenced and consisted of 44,929 bases with a G+C content of 38.7%. Forty-six putative open reading frames were identified and genes were organized into functional clusters for host specificity, lysis, replication and regulation, and packaging and structural proteins.
