The Al/Ni magnetic core–shell pigments were successfully synthesized in this study using the galvanic displacement method. A dense nickel shell was formed on the surface of a flake Al pigment (Al:Ni2+ = 1:0.5), resulting in a decrease in its lightness by 9 units while only slightly increasing the average infrared emissivity by 0.1 compared with the uncoated flake Al pigments. And it also exhibited soft magnetic characteristics, with a saturated magnetization value of 4.5 emu/g. Moreover, the impact of Al/Ni core-shell pigments on the infrared-radar stealth performance of the target was investigated based on the structure of “low-emissivity coating (LEC) + radar absorbing coating (RAC)”. The results showed that the emissivity of Al/Ni composite coatings maintained a low infrared emissivity (ε ≤ 0.23). The introduction of Ni shell improved the impedance matching between the Al/Ni coating and free space, allowing greater penetration of microwaves into the LEC and improving radar stealth performance of materials. The Al/Ni composite coatings (Al:Ni2+ = 1:0.5, the thickness of RAC:1.2 mm, and LEC: 60 μm) exhibited an extended effective absorption bandwidth (Reflection loss (RL) ≤ −10 dB) from 2.6 to 4.25 GHz and an enhancement of the average RL from −6.43 to −7.1 dB in the range of 2–18 GHz compared to the RAC. Consequently, the Al/Ni magnetic core–shell pigments can be used as novel low-emissivity pigments for infrared-radar compatible stealth research.

Abstract The non-coordination between the socio-economic systems and ecosystems of a region is a crucial obstacle to sustainable development. To reveal the relationships between complex urban systems and achieve the goal of sustainable and coordinated urban development, we constructed a coupling coordination degree model (CCDM) and coupling angle model (CAM) and analyzed the degree of coupling coordination and evolution process among the population, water resources, economy, and ecology (PWEE) system of the Tuha region for 2005–2020. The results indicated that: (1) During 2005–2020, the comprehensive development index (CDI) of the population, water resources and economy subsystems was 0.21–0.65, with the three subsystems portraying an overall increase; the average values of the RSEI at five-year intervals were 0.29, 0.28, 0.28, and 0.26, indicating a downward trend in the environmental quality. (2) The coupling coordination effect of the PWEE system portrayed a low level; the coupling coordination degree (CCD) values were 0.28–0.58, portraying a fluctuating upward trend. The level of CCD increased from low disorder to marginal coordination. (3) The PWEE system’s scissor difference reflects large evolutionary characteristics. The ecological support capacity was not observed until the late stage. We conclude that the PWEE composite system of the region is in a stage of disordered development. These findings significantly bolster the theoretical underpinnings of sustainable development studies, offering essential scientific theories and methodological frameworks for crafting sustainable development policies tailored to urban systems in the Tuha region.

Abstract In order to enhance the hyperspectral camouflage efficacy of stealth coatings against a natural vegetative backdrop, LiCl, known for its significant hygroscopic properties, was incorporated into green Mg–Al layered double hydroxide (Mg–Al LDHs) material. Micron-sized composite microspheres were subsequently synthesized via the spray-drying granulation technique. The structure, morphology, and chemical composition of these microspheres were thoroughly characterized by X-ray diffraction, scanning electron microscopy, laser particle size analysis, nitrogen adsorption–desorption isotherms, and Fourier-transform infrared spectroscopy. The effect of LiCl content on the moisture absorption capacity and near-infrared reflectance spectra of the microspheres was systematically evaluated. We found that incorporating an optimal amount of LiCl into the internal pores of the Mg–Al LDHs microspheres did not compromise their smooth surface morphology and uniform particulate distribution. Notably, when the LiCl content was 10%, the maximum saturation moisture uptake ratio of the coating increased to 0.75 g/g. This hygroscopicity significantly enhanced the absorption and scattering of near-infrared radiation by the coating while concurrently improving its ability to modulate the shape and reflectance of both the visible and near-infrared spectral curves. Spectral congruence between the synthetic coating and natural green foliage was quantified at 97.41%. Moreover, this performance was maintained over 10 cycles of programmed drying and re-humidification, and the coating consistently demonstrated stable hygroscopic properties and sustained over 95% spectral congruence. These optimized artificial coatings were found to effectively confuse hyperspectral classification algorithms, thus blending seamlessly into a natural foliage backdrop. This study provides a new method for regulating VIS and NIR spectral (visible–near infrared spectrum) features, which will be critical for applications in advanced hyperspectral camouflage materials.

Abstract The two-dimensional electron gas (2DEG) in BaSnO $$_3$$ 3 -based heterostructure (HS) has received tremendous attention in the electronic applications because of its excellent electron migration characteristic. We modeled the n-type (LaO) $$^+$$ + /(SnO $$_2$$ 2 ) $$^0$$ 0 interface by depositing LaGaO $$_3$$ 3 film on the BaSnO $$_3$$ 3 substrate and explored strain effects on the critical thickness for forming 2DEG and electrical properties of LaGaO $$_3$$ 3 /BaSnO $$_3$$ 3 HS system using first-principles electronic structure calculations. The results indicate that to form 2DEG in the unstrained LaGaO $$_3$$ 3 /BaSnO $$_3$$ 3 HS system, a minimum thickness of approximately 4 unit cells of LaGaO $$_3$$ 3 film is necessary. An increased film thickness of LaGaO $$_3$$ 3 is required to form the 2DEG for -3%-biaxially-strained HS system and the critical thickness is 3 unit cells for 3%-baxially-strained HS system, which is caused by the strain-induced change of the electrostatic potential in LaGaO $$_3$$ 3 film. In addition, the biaxial strain plays an important role in tailoring the electrical properties of 2DEG in LaGaO $$_3$$ 3 /BaSnO $$_3$$ 3 HS syestem. The interfacial charge carrier density, electron mobility and electrical conductivity can be optimized when a moderate tensile strain is applied on the BaSnO $$_3$$ 3 substrate in the ab-plane.

Megalobrama amblycephala (Wuchang bream) is a widely farmed native species with the lowest hypoxic tolerance among the major aquaculture species in China. It is often threatened by hypoxic environments in aquaculture due to high culture densities and weather changes. Identification of the key genes and elucidation of the mechanisms involved in hypoxic tolerance provide the basis for genetic breeding of hypoxia-tolerant Megalobrama amblycephala strains. Here, we report that fih is evolutionarily conserved across species. CRISPR/Cas9-generated fih heterozygotes in Megalobrama amblycephala facilitate hypoxic tolerance and exhibit increased expression of hypoxia-responsive genes, including pdh3, glut1 and vegfa. Mechanistically, coimmunoprecipitation experiments indicated that overexpressed Megalobrama amblycephala fih (Ma-fih) interacts with the transcription factor hypoxia inducible factor (Ma-hif-α). Dual-luciferase assay showed that overexpression of Ma-fih inhibited the transcriptional activity of Ma-hif-α. In addition, we found that overexpression of Ma-fih suppressed hypoxia-induced glucose uptake in EPC cells. This study not only highlights the critical function of Ma-fih in regulating the hypoxia signaling pathway, but also provides a candidate gene for breeding hypoxia-tolerant Megalobrama amblycephala strains.

IFN regulatory factor 3 (IRF3) is the transcription factor crucial for the production of type I IFN in viral defence and inflammatory responses. The activity of IRF3 is strictly modulated by post-translational modifications (PTMs) to effectively protect the host from infection while avoiding excessive immunopathology. Here, we report that zebrafish myosin-regulated light chain interacting protein b (mylipb) inhibits virus-induced type I IFN production via two synergistic mechanisms: induction of autophagic degradation of irf3 and reduction of irf3 phosphorylation. In vivo, mylipb-null zebrafish exhibit reduced lethality and viral mRNA levels compared to controls. At the cellular level, overexpression of mylipb significantly reduces cellular antiviral capacity, and promotes viral proliferation. Mechanistically, mylipb associates with irf3 and targets Lys 352 to increase K6-linked polyubiquitination, dependent on its E3 ubiquitin ligase activity, leading to autophagic degradation of irf3. Meanwhile, mylipb acts as a decoy substrate for the phosphokinase tbk1 to attenuate irf3 phosphorylation and cellular antiviral responses independent of its enzymatic activity. These findings support a critical role for zebrafish mylipb in the limitation of antiviral innate immunity through two synergistic mechanisms targeting irf3.

In recent years, increasing urbanization has profoundly impacted the quality of regional habitats, presenting a severe risk to the ability of a region to develop in a high-quality manner. Therefore, the scientific assessment of the features of habitat quality (HQ) evolution over time and space and the prediction of future trends in changes in the HQ are of great significance for the formulation of effective ecological protection policies. Based on five periods of land use and land cover (LULC) data from 2000 to 2020, InVEST model was used to estimate both geographical and chronological trends in the HQ in the Tuha region, China. Spatial autocorrelation analysis methods were used to assess HQ and spatial aggregation of habitat degradation, and ecological zoning was delineated in conjunction with the Human Footprint Index (HFI). Based on the results of ecological zoning, the study predicted changes in habitat quality (HQ) in 2040 under three scenarios: natural development (ND), ecological preservation (EP), and urban development (UD) by applying the Patch-Generating Land Use Simulation (PLUS) model. The results demonstrated that (1) from 2000 to 2020, the habitat quality in the Tuha region exhibited a downward trend, with the proportion of low HQ increasing from 83.63% to 84.24%. Spatially, high habitat quality (HQ) is mainly concentrated in the Tianshan Mountains. From 2000 to 2020, the Moran index for habitat quality (HQ) decreased from 0.967 to 0.959, while the Moran index for habitat degradation declined from 0.805 to 0.780. The habitat quality (HQ) and degradation exhibit significant spatial aggregation, and the degree of degradation has increased incrementally. (2) From 2000 to 2020, human activities in the Tuha area increased continuously and were mainly concentrated in Tuha district and counties. The proportion of high Human Footprint Index (HFI) increased from 0.66% to 1.32%, while the proportion of medium HFI increased from 3.13% to 7.46%. (3) The expansion of urbanized land has exacerbated habitat degradation. The proportion of high HQ in the EP scenario is higher than that in ND and UD scenario. The results show that the ecological protection scenario is more conducive to the sustainable development of habitat quality in the Tuha region. The results can provide a scientific basis for ecological management and protection in the Tuha area.

The Lactobacillus strains with excellent fermentation performance play a crucial role in the preparation of starter cultures, which has a significant impact on the quality of fermented dairy products. Therefore, to select the Lactobacillus strains with high fermentation performance is essential for the preparation of starter cultures. In this study, four Lactobacillus strains with high stability of milk-clotting after the 10th passage were derived from three samples of Aksu handmade yogurt (a sort of Xinjiang specialty). By means of 16S rRNA sequencing and morphophysiological identification, q8-1 and q28-2 were identified as Lactobacillus casei, R22-2 was identified as Lactobacillus paracasei, and q4-1 was identified as Pediococcus acidilactici. Meanwhile, some other bacteria with the stability of milk-clotting that had been previously selected in the lab including K5-2 (Pediococcus pentosaceus), 7-1 (Lactobacillus bulgaricus) and A-2 (Streptococcus thermophilus) were juxtaposed with these four Lactobacillus strains for performance assessment. In this assessment, a measurement was performed to determine their single-strain growth ability, acid production performance, post-acidification performance, viable cell count (VCC) during storage in diary products, sensory analysis results, texture properties and aroma-producing ability. Results showed that, the Lactobacillus bulgaricus 7-1 had the best fermentation performance with coagulation time and fermentation end times of 8 and 9.5 hours, respectively. It had a high acid yield but a weak capacity for post-acidification. After 1 day of post-ripening, the acidity was 97.6 °T. After 20 days of storage at 4 ℃, the acidity did not vary by more than 20 °T, and the number of viable bacteria remained above 1×106 CFU/mL. Good texture characterized the fermented milk. The firmness, consistency, cohesiveness and index of viscosity were (60.26±0.11) g, (1209.37±0.62) g·s, (−11.81±0.04) g, (−5.37±0.04) g·s, respectively. In the fermented milk, the concentrations of acetaldehyde and diacetyl were 14.48 and 1.23 mg/L, respectively. Good fermentation performance was demonstrated by Streptococcus thermophilus A-2, and Pediococcus pentosaceus K5-2, which also met the fundamental parameters of starter culture. Results showed that Lactobacillus bulgaricus 7-1, Streptococcus thermophilus A-2, and Pediococcus pentosaceus K5-2 had good yoghurt fermentation abilities and could be employed as yoghurt starter cultures.

Abstract The increasing development of biomedicine and bioelectronics has highlighted the requirement for smart materials that can respond to changes in physical and chemical properties under external environments, such as magnetic fields, electric fields, and temperature. Accordingly, hydrogels have been widely evaluated as promising candidates for smart materials owing to their intriguing structures comprising a cross‐linked network of polymer chains with interstitial spaces filled with solvent water. This feature endows hydrogels with soft and wet characteristics, which not only induce high tissue affinity but also allow the introduction of environmentally responsive nanoparticles to release specific smart properties. Herein, we reviewed novel smart hydrogels that can be applied in biomedicine and bioelectronics, and highlighted and discussed existing challenges in current technologies and research.

Due to the complexity of the protein secretory pathway, strategy suitable for the production of a certain recombination protein cannot be generalized. Here, the authors construct a proteome-constrained genome-scale protein secretory model for yeast and show its application in the production of different misfolded or recombinant proteins.

Intestinal microbiota is a unique ecosystem, known as the ''second genome'' of human beings. With the widespread application of next generation sequencing (NGS), especially 16 S rRNA and shotgun sequencing, numerous studies have shown that dysregulation of intestinal microbiota is associated with many central nervous system diseases. Ischemic stroke (IS) is a cerebrovascular disease with high morbidity and mortality. Brain damage in IS affects intestinal function, and intestinal dysfunction further aggravates brain damage, forming a vicious circle of mutual interference in pathology. The microbiota-gut-brain axis study based on the intestinal microbiota has opened up broader ideas for exploring its pathogenesis and risk factors, and also provided more possibilities for the selection of therapeutic targets for this type of drug. This review discussed the application of NGS technology in the study of intestinal microbiota and the research progress of microbiota-gut-brain axis in recent years, and systematically sorts out the literature on the relationship between ischemic stroke and intestinal microbiota. It starts with the characteristics of microbiota-gut-brain axis’ bidirectional regulation, respectively discusses the high risk factors of IS under intestinal microbiota imbalance and the physiological and pathological changes of intestinal microbiota after IS, and summarizes the related targets, in order to provide reliable reference for the treatment of IS from intestinal microbiota. In addition, natural botanical active ingredients have achieved good results in the treatment of IS based on regulating the homeostasis of gut microbiota, providing new evidence for studying the potential targets and therapies of IS based on the microbiota-gut-brain axis.

Abstract Yeasts are known to have versatile metabolic traits, while how these metabolic traits have evolved has not been elucidated systematically. We performed integrative evolution analysis to investigate how genomic evolution determines trait generation by reconstructing genome‐scale metabolic models (GEMs) for 332 yeasts. These GEMs could comprehensively characterize trait diversity and predict enzyme functionality, thereby signifying that sequence‐level evolution has shaped reaction networks towards new metabolic functions. Strikingly, using GEMs, we can mechanistically map different evolutionary events, e.g. horizontal gene transfer and gene duplication, onto relevant subpathways to explain metabolic plasticity. This demonstrates that gene family expansion and enzyme promiscuity are prominent mechanisms for metabolic trait gains, while GEM simulations reveal that additional factors, such as gene loss from distant pathways, contribute to trait losses. Furthermore, our analysis could pinpoint to specific genes and pathways that have been under positive selection and relevant for the formulation of complex metabolic traits, i.e. thermotolerance and the Crabtree effect. Our findings illustrate how multidimensional evolution in both metabolic network structure and individual enzymes drives phenotypic variations.

Long-term ozone (O3) exposure may lead to non-communicable diseases and increase mortality risk. However, cohort-based studies are relatively rare, and inconsistent exposure metrics impair the credibility of epidemiological evidence synthetization. To provide more accurate meta-estimations, this study updates existing systematic reviews by including recent studies and summarizing the quantitative associations between O3 exposure and cause-specific mortality risks, based on unified exposure metrics. Cross-metric conversion factors were estimated linearly by decadal observations during 1990–2019. The Hunter-Schmidt random-effects estimator was applied to pool the relative risks. A total of 25 studies involving 226,453,067 participants (14 unique cohorts covering 99,855,611 participants) were included in the systematic review. After linearly unifying the inconsistent O3 exposure metrics , the pooled relative risks associated with every 10 nmol mol−1 (ppbV) incremental O3 exposure, by mean of the warm-season daily maximum 8-h average metric, were as follows: 1.014 with 95% confidence interval (CI) ranging 1.009–1.019 for all-cause mortality; 1.025 (95% CI: 1.010–1.040) for respiratory mortality; 1.056 (95% CI: 1.029–1.084) for COPD mortality; 1.019 (95% CI: 1.004–1.035) for cardiovascular mortality; and 1.074 (95% CI: 1.054–1.093) for congestive heart failure mortality. Insignificant mortality risk associations were found for ischemic heart disease, cerebrovascular diseases, and lung cancer. Adjustment for exposure metrics laid a solid foundation for multi-study meta-analysis, and widening coverage of surface O3 observations is expected to strengthen the cross-metric conversion in the future. Ever-growing numbers of epidemiological studies supported the evidence for considerable cardiopulmonary hazards and all-cause mortality risks from long-term O3 exposure. However, evidence of long-term O3 exposure-associated health effects was still scarce, so more relevant studies are needed to cover more populations with regional diversity.

Abstract Background Foxp3+CD4+ regulatory T cells (Treg) constitutes a key event in autoimmune diseases. STAT5b is the critical link between the IL-2/15 and FOXP3, the master regulator of Treg cells. Methods The CD3+T cell and Foxp3+CD4+ regulatory T cells were overexpressioned or knockdown MKL-1 and STAT5a and tested for Treg cell development and function. Direct interaction of MKL-1 and STAT5a were analyzed by coimmunoprecipitation assays, Luciferase assay, Immunofluoresence Staining and Yeast two-hybrid screening. The effect of MKL-1 and STAT5a on the Treg genes expression was analyzed by qPCR and western blotting and Flow cytometry. Results However, the molecular mechanisms mediating STAT5b-dependent Treg genes expression and Treg cell phenotype and function in autoimmune diseases are not well defined. Here, we report that the MKL-1 is a coactivator for the major Treg genes transcription factor STAT5b, which is required for human Treg cell phenotype and function. The N terminus of STAT5b, which contains a basic coiled-coil protein–protein interaction domain, binds the C-terminal activation domain of MKL-1 and enhances MKL-1 mediated transcriptional activation of Treg-specific, CArG containing promoters, including the Treg-specific genes Foxp3. Suppression of endogenous STAT5b expression by specific small interfering RNA attenuates MKL-1 transcriptional activation in cultured human cells. The STAT5b–MKL-1 interaction identifies a role of Treg-specific gene regulation and regulated mouse Treg cell development and function and suggests a possible mechanism for the protective effects of autoimmune disease Idiopathic Thrombocytopenic Purpura (ITP). Conclusions Our studies demonstrate for the first time that MKL-1 is a coactivator for STAT5b, the regulator of Treg cell development and function. Video abstract

A teleost’s kidney was divided into head kidney and trunk kidney. The head kidney is an important lymphatic organ, while the trunk kidney mainly performs osmotic pressure regulation and excretion functions. Previous studies have shown that the teleost’s head kidney exerts a strong immune response against pathogen invasion, while the mechanism of immune response in the trunk kidney is still rarely reported. Therefore, in this study, we established an Infectious hematopoietic necrosis virus (IHNV) immersion infection model to compare the similarities and differences of immune response mechanisms between the head kidney and trunk kidney against viral infection. The results showed that IHNV infection causes severe tissue damage and inflammatory reaction in the head and trunk kidney, triggers a series of interferon cascade reactions, and produces strong immune response. In addition, the transcriptome data showed that the head kidney and trunk kidney had similar immune response mechanisms, which showed that the NOD-like receptor signaling pathway and Toll-like receptor signaling pathway were activated. In conclusion, despite functional differentiation, the teleost’s trunk kidney still has a strong immune response, especially the interferon-stimulated genes, which have stronger immune response in the trunk kidney than in the head kidney when responding to IHNV infection. This study contributes to a more comprehensive understanding of the teleost immune system and enriches the theory of kidney immunity in teleosts.

As an emerging pollutant in the aquatic environment, microcystin-LR (MC-LR) can enter the body through multiple pathways, and then induce apoptosis and gonadal damage, affecting reproductive function. Previous studies focused on male reproductive toxicity induced by MC-LR neglecting its effects on females. The apoptotic signal-regulated kinase 1 (ASK1) is an upstream protein of P38/JNK pathway, closely associated with apoptosis and organ damage. However, the role of ASK1 in MC-LR-induced reproductive toxicity is unclear. Therefore, this study investigated the role of ASK1 in mouse ovarian injury and apoptosis induced by MC-LR. After MC-LR exposure, ASK1 expression in mouse ovarian granulosa cells was increased at the protein and mRNA levels, and decreased following pretreatment by antioxidant N-acetylcysteine, suggesting that MC-LR-induced oxidative stress has a regulatory role in ASK1 expression. Inhibition of ASK1 expression with siASK1 and NQDI-1 could effectively alleviate MC-LR-induced mitochondrial membrane potential damage and apoptosis in ovarian granulosa cells, as well as pathological damage, apoptosis and the decreased gonadal index in ovaries of C57BL/6 mice. Moreover, the P38/JNK pathway and downstream apoptosis-related proteins (P-P38, P-JNK, P-P53, Fas) and genes (MKK4, MKK3, Ddit3, Mef2c) were activated in vivo and vitro, but their activation was restrained after ASK1 inhibition. Data presented herein suggest that the ASK1-mediated P38/JNK pathway is involved in ovarian injury and apoptosis induced by MC-LR in mice. It is confirmed that ASK1 has an important role in MC-LR-induced ovarian injury, which provides new insights for preventing MCs-induced reproductive toxicity in females.

Abstract Chromium-doped TiO2 pigments were synthesized via a solid-state reaction method and studied with X-ray diffraction, SEM, XPS, and UV-VIS-NIR reflectance spectroscopy. The incorporation of Cr3+ accelerates the transition from the anatase phase to the rutile phase and compresses the crystal lattice. Moreover, the particle morphology, energy gap, and reflectance spectrum of Cr-doped TiO2 pigments is affected by the crystal structure and doping concentration. For the rutile samples, some of the Cr3+ ions are oxidized to Cr4+ after sintering at a high temperature, which leads to a strong near-infrared absorption band due to the 3A2 → 3 T1 electric dipole-allowed transitions of Cr4+. And the decrease of the band gap causes an obvious redshift of the optical absorption edges as the doping concentration increases. Thus, the VIS and near-infrared average reflectance of the rutile Ti1 − x Cr x O2 sample decrease by 60.2 and 58%, respectively, when the Cr content increases to x = 0.0375. Meanwhile, the color changes to black brown. However, for the anatase Ti1 − x Cr x O2 pigments, only the VIS reflection spectrum is inhibited by forming some characteristic visible light absorption peaks of Cr3+. The morphology, band gap, and NIR reflectance are not significantly affected. Finally, a Cr-doped anatase TiO2 pigment with a brownish-yellow color and 90% near-infrared reflectance can be obtained.

Abstract Vascular endothelial growth factor A (VEGF-A) is a pivotal player in angiogenesis. It is capable of influencing such cellular processes as tubulogenesis and vascular smooth muscle cell (VSMC) proliferation, yet very little is known about the actual signaling events that mediate VEGF-A induced VSMC phenotypic switch. In this report, we describe the identification of an intricate VEGF-A-induced signaling cascade that involves VEGFR2, STAT3, and Myocardin. We demonstrate that VEGF-A promotes VSMC proliferation via VEGFR2/STAT3-mediated upregulating the proliferation of markers like Cyclin D1 and PCNA. Specifically, VEGF-A leads to nitrosylation of Myocardin, weakens its effect on promoting the expression of contractile markers and is unable to inhibit the activation of STAT3. These observations reinforce the importance of nitric oxide and S-nitrosylation in angiogenesis and provide a mechanistic pathway for VEGF-A-induced VSMC phenotypic switch. In addition, Myocardin, GSNOR and GSNO can create a negative feedback loop to regulate the VSMC phenotypic switch. Thus, the discovery of this interactive network of signaling pathways provides novel and unexpected therapeutic targets for angiogenesis-dependent diseases.

Microcystin-leucine arginine (MC-LR) is a cyclic heptapeptide intracellular toxin released by cyanobacteria that exhibits strong reproductive toxicity. However, little is known about its biotoxicity to the female reproductive system. The present study investigates unexplored molecular pathways by which oxidative stress acts on MC-LR-induced endoplasmic reticulum stress (ERs) and autophagy. In the present study, immortalized murine ovarian granular cells (KK-1 cells) were exposed to 8.5, 17, and 34 μg/mL (IC50) of MC-LR with or without N-acetyl-l-cysteine (NAC, 10 mM) for 24 h, and C57BL/6 mice were treated with 12.5, 25.0, and 40.0 μg/kg⋅bw of MC-LR with or without NAC (200 mg/kg⋅bw) for 14 days. The results revealed that MC-LR could induce cells apoptosis and morphologic changes in ovarian tissues, induce oxidative stress by stimulating the generation of reactive oxygen species (ROS), destroying antioxidant capacity, and subsequently trigger ERs and autophagy by inducing the hyper-expression of ATG12, ATG5, ATG16, EIF2α (phosphorylated at S51), CHOP, XBP1, GRP78, Beclin1, and PERK (Thr980). Furthermore, NAC pretreatment partly inhibited MC-LR-induced ERs and autophagy via the PERK/ATG12 and XBP1/Beclin1 pathways. These results suggest that oxidative stress mediated MC-LR-induced ERs and autophagy in KK-1 cells and C57BL/6 mice ovaries. Therefore, oxidative stress plays an important role in female toxicity induced by MC-LR.

The widespread distribution of cyanobacteria in the aquatic environment is increasing the risk of water pollution caused by cyanotoxins, which poses a serious threat to human health. However, the structural characterization, distribution and identification techniques of cyanotoxins have not been comprehensively reviewed in previous studies. This paper aims to elaborate the existing information systematically on the diversity of cyanotoxins to identify valuable research avenues. According to the chemical structure, cyanotoxins are mainly classified into cyclic peptides, alkaloids, lipopeptides, nonprotein amino acids and lipoglycans. In terms of global distribution, the amount of cyanotoxins are unbalanced in different areas. The diversity of cyanotoxins is more obviously found in many developed countries than that in undeveloped countries. Moreover, the threat of cyanotoxins has promoted the development of identification and detection technology. Many emerging methods have been developed to detect cyanotoxins in the environment. This communication provides a comprehensive review of the diversity of cyanotoxins, and the detection and identification technology was discussed. This detailed information will be a valuable resource for identifying the various types of cyanotoxins which threaten the environment of different areas. The ability to accurately identify specific cyanotoxins is an obvious and essential aspect of cyanobacterial research.

Microcystin-leucine arginine (MC-LR), a cyclic heptapeptide produced by cyanobacteria, is a strong reproductive toxin. Studies performed in rat Sertoli cells and Chinese hamster ovary cells have demonstrated typical apoptosis after MC-LR exposure. However, little is known on how to protect against the reproductive toxicity induced by MC-LR. The present study aimed to explore the possible molecular mechanism underlying the anti-apoptosis and protective effects of resveratrol (RES) on the co-culture of Sertoli–germ cells and rat testes. The results demonstrated that MC-LR treatment inhibited the proliferation of Sertoli–germ cells and induced apoptosis. Furthermore, sirtuin 1 (SIRT1) and Bcl-2 were inhibited, while p53 and Ku70 acetylation, Bax expression, and cleaved caspase-3 were upregulated by MC-LR. However, RES pretreatment ameliorated MC-LR-induced apoptosis and SIRT1 inhibition, and downregulated the MC-LR-induced increase in p53 and Ku70 acetylation, Bax expression, and caspase-3 activation. In addition, RES reversed the MC-LR-mediated reduction in Ku70 binding to Bax. The present study indicated that the administration of RES could ameliorate MC-LR-induced Sertoli–germ cell apoptosis and protect against reproductive toxicity in rats by stimulating the SIRT1/p53 pathway, suppressing p53 and Ku70 acetylation and enhancing the binding of Ku70 to Bax.

Abstract Background Previous studies indicated that type 2 diabetes mellitus (T2DM) might be associated with the risk of cancer. The aim of this study was to investigate the association between T2DM and the risk of developing common cancers in a Chinese population. Methods A population-based retrospective cohort study was carried out in the Nan-Hu district of Jiaxing city, Zhejiang province, China. The incidence of cancer cases among type 2 diabetic patients were identified through record-linkage of the Diabetic Surveillance and Registry Database with the Cancer Database from January 2002 to June 2008. The standardized incidence ratio (SIR) and 95% confidence interval (CI) were estimated for the risk of cancer among the patients with type 2 diabetes. Results The overall incidence of cancer was 1083.6 per 105 subjects in male T2DM patients and 870.2 per 105 in females. Increased risk of developing cancer was found in both male and female T2DM patients with an SIR of 1.331 (95% CI = 1.143-1.518) and 1.737 (1.478-1.997), respectively. As for cancer subtypes, both male and female T2DM patients had a significantly increased risk of pancreatic cancer with the SIRs of 2.973 (1.73-4.21) and 2.687 (1.445-3.928), respectively. Elevated risk of liver and kidney cancers was only found in male T2DM patients with SIRs of 1.538 (1.005-2.072) and 4.091 (1.418-6.764), respectively. Increased risks of developing breast cancer [2.209 (1.487-2.93)] and leukemia SIR: [4.167 (1.584- 6.749) ] were found in female patients. Conclusions These findings indicated that patients with T2DM have an increased risk of developing cancer. Additional cancer screening should be employed in the management of patients with T2DM.
