Peat moss (Sphagnum) is a non-vascular higher plant with unique xylem-like hyaline (H) cells that are accompanied by photosynthetic chlorophyllous cells. These cellular structures play crucial roles in water storage and carbon sequestration. However, it is largely unknown how peat moss develops the H cells. This study systematically explored the Sphagnum Developmental Cell Atlas and Lineage and classified leaf cell development into two lineages with six stages (S0-S5) based on changes in key cellular traits, including the formation of spiral secondary cell walls (S4) and the presence of water pores (S5). Cell lineage-specific subcellular remodeling was transcriptionally regulated during leaf development, and vacuole-mediated clearance of organelles and cell death led to mature dead H cells. Interestingly, expression of land plant conserved Vascular-related NAC Domain (VND) genes correlated with H cell formation. Overall, these results suggest that the origination of xylem-like H cells is related to VND, likely through the neofunctionalization of vacuole-mediated cell death to attempt xylem formation in peat moss, suggesting potential uncoupling of xylem and phloem cell origins. This study positions peat moss as a potential model organism for studying integrative evolutionary cell biology. High‐latitude vegetation experience different temperatures than the ambient air temperature. While lacking a regional plant temperature product, we drove the dynamic ecosystem model, LPJ‐GUESS, with widely used ERA5‐land surface temperature (Tsurf, at radiative equilibrium) and air temperature to understand ecosystem process responses to these two temperatures. We show that tundra plants' growth is stimulated by warmer Tsurf in the summer, but in the boreal forests, colder Tsurf in the non‐summer months constrains leaf development and enzyme activity the following growing season. Tsurf drives higher productivity of tundra plant individuals but leads to less productive individuals in the boreal forest, although with compensatory changes (almost 68%) in vegetation structure. We demonstrate the importance of forcing temperature in simulating high‐latitude ecosystem processes and call for a community effort to measure plant temperatures across canopy heights and seasons to reduce uncertainties in estimating high‐latitude plant responses and feedback to climate. Plain Language Summary: The temperature experienced by plants often differs from the surrounding air temperature. This plant temperature plays a large role in how plants grow and function. By using two common observation‐based temperature data sets (air and surface temperatures) to drive an ecosystem model, we found that warmer summer surface temperatures stimulate tundra plant growth, but colder temperatures in other seasons than summer can limit growth in the boreal forest. Leaf ontogeny of tropical evergreen tree species lasts several months with changes in size, shape, colouration and internal tissue distribution of leaves. Leaf initiation in Saraca asoca generally occurs once in a year during February–April, followed by very limited leafing thereafter. We measured the rate of photosynthesis, chlorophyll a fluorescence, energy quenching and PSII functions during the leaf ontogeny process. Observations were taken up to 35 days after opening of lamina (DAOL). Significant increase in the synthesis and accumulation of photosynthetic pigments but negative net photosynthesis was noticed during initial days of the ontogeny. The leaf moved from heterotrophy to autotrophy with gradual improvement of PSII functions. The ratio of intercellular CO2 (C i) and ambient CO2 (C a) showed significant change at ≥11 DAOL. Increase in the age of the leaf (between 5 and 28 DAOL) caused decrease in O-J rise and corresponding increase in J-I and I-P rise as well as of fluorescence maximum (F M) of the OJIP curve. The improvement of the electron transport components of the donor side of PSII was seen with increase in the functional oxygen evolving complex. The functional improvements of the donor and acceptor side of PSII during leaf ontogeny are discussed. The leaf ontogeny in Saraca asoca usually occurs once in a year and involves variation in PSII function. The developing leaves metabolically switch from heterotrophy to autotrophy. During leaf development, the carotenoids are accumulated earlier than chlorophylls. Nodular clusters are a morphogenetic pathway used in the in vitro propagation of Bromeliaceae due to their high responsiveness in the regeneration of microshoots. Additionally, endogenous polyamines, especially putrescine, may contribute to the induction of these nodular clusters. However, the potential effects of this regulator on the regeneration of microshoots when applied to explants are unclear, especially in traditional pineapple cultivars such as "Turiaçu". Therefore, our objective here was to induce and characterize the morphoanatomy of nodular clusters in combinations of 6-benzylaminopurine (BAP) and putrescine (PUT) in "Turiaçu" pineapple. Leaf segments induced in culture medium supplemented with 4 μM naphthaleneacetic acid (NAA), 8 μM BAP, and 100 μM PUT resulted in an induction rate of 56 %, with the formation of microshoots via nodular clusters, increased length, and the number of leaves per microshoot. Morphoanatomical studies revealed that in the medium with 4 μM NAA, 8 μM BAP, and 100 μM PUT, the clusters presented a voluminous size, microshoots with leaves, and the development of new leaves along the base of the explant, and the formation of meristemoids with small cells arranged in a spiral. The results indicate that auxins and cytokinins combined with exogenous putrescine are efficient for inducing nodule clusters. Tomato (Solanum lycopersicum L.), the second most important vegetable crop globally, faces a significant threat from various viral diseases. A newly emerging disease, characterised by distinctive shoestring symptoms on leaves and the development of unripe, small, and hard fruit, poses a serious challenge to tomato cultivation in India. An initial survey in an experimental field revealed more than 50% of the plants displayed symptoms of the shoestring disease, resulting in substantial reductions in fruit yield and quality. Transmission electron microscopy (TEM) and molecular analyses identified an isolate of the tomato mottle mosaic virus (ToMMV) in the affected plants. When the partially purified virus was mechanically inoculated into tomato cv. Pusa Ruby plants, it reproduced the characteristic shoestring symptoms, confirming its causal relationship with the disease. Notably, the present shoestring isolate of ToMMV (ToMMV-Tss) was found to induce similar shoestring symptoms in most of the major commercial tomato varieties when inoculated under controlled experimental conditions in the glasshouse, indicating its aggressive nature. Host range studies demonstrated that the ToMMV-Tss can infect several solanaceous species, while cucurbitaceous hosts remained unaffected. Moreover, the virus was found to be seed-transmissible, with a small percentage of seedlings from infected plants displaying symptoms. These findings underscore the significant impact of ToMMV on tomato production in India and emphasise the need for reliable diagnostic tools and effective management strategies to curb the spread and mitigate the impact of this virus on commercial tomato cultivation. Leaves are the most significant parts of forage crops such as alfalfa. Senescence is the terminal stage of leaf development and is controlled by an integrated myriad of endogenous signals and environmental stimuli. WRKY transcription factors (TFs) play essential roles in regulating leaf senescence; however, only a few studies on the analysis and identification of the WRKY TF family in Medicago Sativa have been reported. In this study, we identified 198 WRKY family members from the alfalfa (M. sativa L.) cultivar 'XinjiangDaye' using phylogenetic analysis and categorized them into three subfamilies, Groups I, II, and III, based on their structural characteristics. Group II members were further divided into five subclasses. In addition, several hormone- and stress-related cis-acting elements were identified in the promoter regions of MsWRKYs. Furthermore, 14 aging-related MsWRKYs genes from a previous transcriptome in our laboratory were selected for RT-qPCR validation of their expression patterns, and subsequently cloned for overexpression examination. Finally, MsWRKY5, MsWRKY66, MsWRKY92, and MsWRKY141 were confirmed to cause leaf yellowing in Nicotiana benthaminana using a transient expression system. SUMMARY Leaves, as primary photosynthetic organs essential for high crop yield and quality, have attracted significant attention. The functions of DNA topoisomerase 1α (TOP1α) in various biological processes, including leaf development, in Brassica napus remain unknown. Here, four paralogs of BnaTOP1α, namely BnaA01.TOP1α, BnaA02.TOP1α, BnaC01.TOP1α and BnaC02.TOP1α, were identified and cloned in the B. napus inbred line ‘K407’. Expression pattern analysis revealed that BnaA02.TOP1α and BnaC02.TOP1α, but not BnaA01.TOP1α and BnaC01.TOP1α, were persistently and highly expressed in B. napus true leaves. Preliminary analysis in Arabidopsis thaliana revealed that BnaA02.TOP1α and BnaC02.TOP1α paralogs, but not BnaA01.TOP1α and BnaC01.TOP1α, performed biological functions. Targeted mutations of four BnaTOP1α paralogs in B. napus using the CRISPR‐Cas9 system revealed that BnaA02.TOP1α and BnaC02.TOP1α served as functional paralogs and redundantly promoted true leaf number and size, thereby promoting true leaf biomass accumulation. Moreover, BnaA02.TOP1α modulated the levels of endogenous gibberellins, cytokinins and auxins by indirectly regulating several genes related to their metabolism processes. BnaA02.TOP1α directly activated BnaA03.CCS52A2 and BnaC09.AN3 by facilitating the recruitment of RNA polymerase II and modulating H3K27me3, H3K36me2 and H3K36me3 levels at these loci and indirectly activated the BnaA08.PARL1 expression, thereby positively controlling the true leaf size in B. napus. Additionally, BnaA02.TOP1α indirectly activated the BnaA07.PIN1 expression to positively regulate the true leaf number. ABSTRACT Plant organs achieve their specific size and shape through the coordination of cell division and cell expansion, processes that are profoundly influenced by environmental cues. Cytokinesis during cell division depends on the position of the cytokinetic wall, but how this process responses to environment fluctuations remains underexplored. Here, we investigated a regulatory module involving C2H2‐type zinc finger protein (C2H2‐ZFP) in leaf morphology during drought stress. A total of 123 C2H2‐ZFP members were identified through a comparative genome survey in Populus alba × P. glandulosa ‘84K’. Among them, PagSUPa, an orthologous gene of Arabidopsis SUPERMAN, was selected due to its responsiveness to drought stress and was further confirmed to play a role in leaf development. Phenotypic characterization and cellular analysis revealed that PagSUPa fine‐tunes the duration of cell proliferation in the adaxial epidermis, thereby influencing leaf morphology by modulating leaf adaxial‐abaxial polarity. Additionally, we found that PagSUPa directly suppresses the expression of PHRAGMOPLAST ORIENTING KINESIN1 (PagPOK1) and PagPOK2, genes encoding proteins involved in phragmoplast orientation and position, which results in impaired cytokinesis and cell wall organization. Barley development from seedling to flowering involves both external and internal changes, the latter requiring microscopic observation. Internal changes allow for the classification of preflowering development into three phases: vegetative, early reproductive, and late reproductive. Genetic and environmental factors influence the duration of these phases, impacting grain yield. Photoperiod-sensitivity genes PPD-H1 play amajor role in flowering time, affecting adaptation; however, the effect might also be direct (beyond affecting phenology). In this paper, we aimed to assess how PPD-H1 alleles affect barley development, including the progression of growth phases, leaf emergence, tillering dynamics, and spikelet development. Two experiments (field and controlled conditions) were conducted with a factorial combination of (i) four near-isogenic lines (NILs) for PPD-H1 alleles (ppd-H1 or Ppd-H1) under two contrasting PHYC genetic backgrounds (PhyC-l and PhyC-e) and (ii) two photoperiod conditions (short and long days). As expected, longer photoperiods led to a shorter growth cycle. All subphases of time to flowering, final leaf number, and phyllochron were affected by photoperiod. The effects of PPD-H1 on flowering time depended on the PHYC genetic backgrounds and photoperiod conditions. PPD-H1 effects on flowering time were associated with leaf number and phyllochron; the interplay between leaf number and phyllochron affected mainly the late reproductive phase. We also found that although PPD-H1 did not affect the phyllochron of the first six leaves, the phyllochron of leaves appearing later, when grown under a short photoperiod, was consistently increased in lines carrying the ppd-H1 allele. The integration of nanoparticles into plant cryopreservation protocols holds great promise for improving the survival rates and recovery potential of explants. This study aimed to verify the effect of nanoparticles on the ex-vitro performance of cryopreservation-derived plants. Lamprocapnos spectabilis (L.) Fukuhara (bleeding heart) 'Gold Heart' and 'Valentine' cultivars were used as the plant material. The encapsulation-vitrification cryopreservation protocol of shoot tips included the preculture, encapsulation, dehydration, storage in liquid nitrogen, rewarming, and recovery steps. Gold (AuNPs), silver (AgNPs), or zinc oxide (ZnONPs) nanoparticles were added at varying concentrations, either into the preculture medium or the protective bead matrix during encapsulation. After the in vitro recovery, the plants were transferred to the glasshouse and subjected to detailed biometrical, biochemical and cytogenetic analyses. Nanoparticles had no evident effect on the acclimatization efficiency (80–100% survival) and leaf number in L. spectabilis 'Gold Heart'. Nonetheless, shoots developed from alginate beads supplemented with 5 ppm AuNPs were twice as long as the control, while the leaves of plants grown on the preculture medium with ZnONPs contained significantly more chlorophyll and had higher Leaf Soil-Plant Analysis Development (SPAD) values. Moreover, several NPs treatments stimulated the development of leaves, including their surface area, length, and perimeter. Higher ZnONPs levels enhanced also the replication process, resulting in higher nuclear DNA content. As for L. spectabilis 'Valentine', alginate augmentation with 5 ppm AgNPs or 5 ppm ZnONPs stimulated the elongation of shoots. There was also a tendency suggesting a positive influence of 5 ppm AgNPs in the alginate bead matrix on foliar growth. The effect of nanoparticles on the content of flavonoids, anthocyanins, and stress markers in the plants varied depending on the treatment and cultivar, but also on the organ studied (leaf or stem). Overall, L. spectabilis 'Gold Heart' was more stress-tolerant and genetically stable than 'Valentine' judging by the activity of Photosystem II (PSII) and flow cytometric analyses, respectively. The complex effects of nanoparticles on survival, biometric parameters, physiological responses, and cytogenetic events underscore the intricate interplay between nanoparticles and plant systems. SUMMARY Trichomes, which originate from the epidermal cell of aerial organs, provide plants with defense and secretion functions. Although numerous genes have been implicated in trichome development, the molecular mechanisms underlying trichome cell formation in plants remain incompletely understood. Here, we using genome‐wide association study (GWAS) across 1037 diverse accessions in upland cotton (Gossypium hirsutum) to identify three loci associated with leaf pubescence (hair) amount, located on chromosome A06 (LPA1), A08 (LPA2) and A11 (LPA3), respectively. GhHD1, a previously characterized candidate gene, was identified on LPA1 and encodes an HD‐Zip transcription factor. For LPA2 and LPA3, we identified two candidate genes, GhGIR1 and GhGIR2, both encoding proteins with WD40 and RING domains that act as inhibitors of leaf hair formation. Expression analysis revealed that GhHD1 was predominantly expressed in hairy accessions, whereas GhGIR1 and GhGIR2 were expressed in hairless accessions. Silencing GhHD1 or overexpressing GhGIR1 in hairy accessions induced in a hairless phenotype, whereas silencing GhGIR2 in hairless accessions resulted in a hairy phenotype. We also demonstrated that GhHD1 interact with both GhGIR1 and GhGIR2, and GhGIR1 can interact with GhGIR2. Further investigation indicated that GhHD1 functions as a transcriptional activator, binding to the promoters of the GhGIR1 and GhGIR2 to active their expression, whereas GhGIR1 and GhGIR2 can suppress the transcriptional activation of GhHD1. Seagrass seed‐based restoration has been developed in several places worldwide, but disturbance at the vulnerable initial stages of seedling recruitment has proved to be a major bottleneck to successful restoration. A detailed investigation of seagrass seedling survival and growth at the earliest stages of seedling development is an important contribution to developing techniques to improve seedling establishment and survival. Here, we investigated the early seedling development of Posidonia australis and P. sinuosa as well as quantifying the variation in seedling survival and development under different seed‐based restoration methods. Early seedling development was documented in indoor aquaria during the first month after fruit dehiscence. In the second ex situ experiment, we determined the effects of three different restoration methods (surface sowing, seedling planting, and planting inside a hessian bag) on seedling survival and development over the first 2 months of life. In the first experiment, a primary root with the first root hairs developed after 7 days. After 1 month, roots were 20–60 mm in length, firmly establishing the seedling into the sediment. Compared to surface sowing, seedling planting and hessian bag restoration treatments did not significantly affect survival for P. australis but increased seedling mortality in P. sinuosa, although growth was greater (shoot and root lengths were approximately 50 and 40% longer, respectively). These aquarium‐scale experiments suggest that planting methods would enhance seedling establishment at larger scales in some species, promoting restoration of damaged seagrass habitats. Amaranthus palmeri S. Watson is a problematic weed that has been encountered almost all over the world because of its stronger adaptability, which causes huge economic losses and local plant diversity decline. As an invasive plant, A. palmeri has been found throughout China. We thus make a code system to describe the phenological growth process of A. palmeri according to the extended BBCH scale to better understand the phenological growth of this weed in China. The growth process of A. palmeri was divided into vegetative growth phases and reproductive growth phases. The vegetative growth phases are composed of seed germination (0), leaf development (1), formation of side shoots (2) and main stem elongation (3). Likewise, the reproductive growth phases include inflorescence emergence (5), blooming (6), fruit development (7), seed maturity (8) and senescence (9). Based on the extended BBCH scale, the first digit stands for the primary growth stages, and the third digit represents the secondary growth stages. In particular, the second digit defines the growth progress: the number 0, means the vegetative growth phases, and the number 1 stands for the reproductive growth phases. Owing to the fact that its seed can germinate multiple times within a year, a general illustration of the phenological development stages of A. palmeri germinating in different months was provided here to describe the growth dynamic characteristic at different times. Background Vulvovaginal candidiasis (VVC) is a condition that impacts women from various ethnic backgrounds and social strata. The precise epidemiology of the disease is difficult to determine due to inconsistent data; however, available information suggests that 70-75% of women will experience VVC at least once in their lifetime. Common symptoms include itching, vaginal discomfort, pain during intercourse, external pain during urination, and unusual discharge. Methods The current study investigates the anticandidal properties of neem (Azadirachta indica) leaf extracts. It compares the anticandidal effects of aqueous and methanolic leaf extracts, as well as green-synthesized silver nanoparticles (AgNPs/SNPs) derived from methanolic neem leaf extract, against Candida albicans isolated from clinical samples of patients with vaginal infection. Results The results showed that AgNPs exhibited a stronger inhibitory effect than both the aqueous and methanol extracts. The methanol extract demonstrated anticandidal activity against C. albicans, while the aqueous extracts had no effect. The anticandidal effect of AgNPs was evaluated using the agar well diffusion method. A topical formulation was created by incorporating the optimized AgNPs into an ointment base, and the in vitro anticandidal assay of this ointment was conducted using the same agar well diffusion method. The inhibitory effectiveness of the AgNPs-infused ointment was found to be comparable to that of a commercial product against Candida albicans. Summary: C4 photosynthesis is a complex trait requiring multiple developmental and metabolic alterations. Despite this complexity, it has independently evolved over 60 times. However, our understanding of the transition to C4 is complicated by the fact that variation in photosynthetic type is usually segregated between species that diverged a long time ago.Here, we perform a genome‐wide association study (GWAS) using the grass Alloteropsis semialata, the only known species to have C3, intermediate, and C4 accessions that recently diverged. We aimed to identify genomic regions associated with the strength of the C4 cycle (measured using δ13C), and the development of C4 leaf anatomy.Genomic regions correlated with δ13C include regulators of C4 decarboxylation enzymes (RIPK), nonphotochemical quenching (SOQ1), and the development of Kranz anatomy (SCARECROW‐LIKE). Regions associated with the development of C4 leaf anatomy in the intermediate individuals contain additional leaf anatomy regulators, including those responsible for vein patterning (GSL8) and meristem determinacy (GIF1).The parallel recruitment of paralogous leaf anatomy regulators between A. semialata and other C4 lineages implies the co‐option of these genes is context‐dependent, which likely has implications for the engineering of the C4 trait into C3 species. GhPEL48_Dt, a Pectate lyase (PEL, EC4.2.2.2), is a crucial enzyme involved in cell-wall modification and pectin degradation. Studies have shown that the GhPEL48_Dt also plays a significant role in cotton-fiber development; however, the specific function and regulatory mechanism of GhPEL48_Dt in cotton-fiber development are still not fully understood. Here, we found that the histone deacetylase inhibitor-Trichostatin A significantly reduces the transcript levels of GhPEL48_Dt and its enzyme activity. Further, silencing of GhPEL48_Dt significantly inhibits the initiation and elongation of cotton fibers by promoting pectin degradation, and the heterologous expression of GhPEL48_Dt promotes the development of trichomes and root hairs in Arabidopsis, which suggests that GhPEL48_Dt plays a positive and conserved role in single cell i.e., fiber, root hair, and leaf trichome development. Collectively, this paper provides a comprehensive analysis of the fundamental characteristics and functions of GhPEL48_Dt in fiber development, including the regulatory role of histone acetylation on GhPEL48_Dt, which contributes to the understanding of pectin degradation pathways and establishes a theoretical foundation for elucidating its regulatory mechanism. Leaves play a crucial role in photosynthesis and respiration, ultimately affecting the final grain yield of crops, including wheat (Triticum aestivum L.); however, the molecular mechanisms underlying wheat leaf development remain largely unknown. Here, we isolated a narrow-leaf gene, TaWAK2-A, through a map-based cloning strategy. TaWAK2-A encodes a wall-associated kinase (WAK), for which a single Ala-to-Val amino acid substitution reduces the protein stability, leading to a narrow-leaf phenotype in wheat. Further investigation suggests that TaWAK2 directly interacts with and phosphorylates TaNAL1, a trypsin-like serine/cysteine protease. The phosphorylated TaNAL1 is then involved in the degradation of the zinc finger transcription factor TaDST, which acts as a repressor of leaf expansion by activating the expression of the cytokinin oxidase gene TaCKX9 and triggering in vivo cytokinin degradation. The components of the mediator kinase module are highly conserved across all eukaryotic lineages, and cyclin‐dependent kinase 8 (CDK8) is essential for correct cell proliferation and differentiation in diverse eukaryotic systems. We show that CDK8 couples leaf development with the establishment of correct stomata patterning for prevailing CO2 conditions. In Arabidopsis, the basic helix‐loop‐helix (bHLH) transcription factor SPEECHLESS (SPCH) controls cellular entry into the stomatal cell lineage, and CDK8 interacts with and phosphorylates SPCH, controlling SPCH protein levels and thereby also expression of the SPCH target genes encoding key regulators of cell fate and asymmetric cell divisions. The lack of the CDK8‐mediated control of SPCH results in an increased number of meristemoid and guard mother cells, and increased stomata index in the cdk8 mutants. Increasing atmospheric CO2 concentrations trigger a developmental programme controlling cell entry into stomatal lineage by limiting the asymmetric divisions. In cdk8, the number of meristemoids and guard mother cells remains the same under ambient and high CO2 concentrations, as the accumulated levels of SPCH caused by the lack of CDK8 appear to override the negative regulation of increased CO2. Background: Type II diabetes mellitus is posing a severe health threat throughout the globe due to its associated pathophysiological risks and high mortality rate. Carbohydrate catabolic enzymes, including α-amylase, α-glucosidase and aldose reductase, play an important role in the development of diabetes. The natural or synthetic inhibitors of these enzymes are crucial in reducing diabetes and its related complications. Tinosporacordifolia is a plant of great significance in Ayurveda due to its unique biological activities, including anti-diabetic properties. The present study aims to identify the active constituents of T. cordifolia leaves and evaluate the in vitro inhibitory potential of its ethanol extract constituents against α-amylase, α-glucosidase and aldose reductase activities. Results: The ethanolic leaf extract of T. cordifolia inhibited the activities of α-amylase, α-glucosidase and aldose reductase in a dose-dependent manner. It was on par with the standard inhibitors acarbose and quercetin. At 5 mg/ml, the noted % inhibition values of extract were 69.27 ± 0.17, 67.8 ± 0.26 and 62.55 ± 0.24, respectively, for α-amylase, α-glucosidase and aldose reductase. Using GC-MS analysis, neophytadiene, γ-sitosterol, phytol, phytyl palmitate, and phytyl acetate were identified as prominent constituents of the ethanolic extract. Based on molecular docking and ADME analysis, γ-sitosterol was found as the major reactive phytoconstituent, which showed the highest inhibitory potential against α-amylase, α-glucosidase and aldose reductase activities. Conclusions: The present study identified γ-sitosterol as triplet inhibitor of α-amylase, α-glucosidase and aldose reductase and affirmed the ethno-medicinal significance of T. cordifolia leaves in the development of new anti-diabetic leads. Societal Impact Statement Summary Grapevine leaves are emblematic of the strong visual associations people make with plants. Leaf shape is immediately recognizable at a glance, and therefore, this is used to distinguish grape varieties. In an era of computationally enabled machine learning‐derived representations of reality, we can revisit how we view and use the shapes and forms that plants display to understand our relationship with them. Using computational approaches combined with time‐honored methods, we can predict theoretical leaves that are possible, enabling us to understand the genetics, development, and environmental responses of plants in new ways. Grapevine leaves are a model morphometric system. Sampling over 10,000 leaves using dozens of landmarks, the genetic, developmental, and environmental basis of leaf shape has been studied and a morphospace for the genus Vitis predicted. Yet, these representations of leaf shape fail to capture the exquisite features of leaves at high resolution. We measure the shapes of 139 grapevine leaves using 1672 pseudo‐landmarks derived from 90 homologous landmarks with Procrustean approaches. From hand traces of the vasculature and blade, we have derived a method to automatically detect landmarks and place pseudo‐landmarks that results in a high‐resolution representation of grapevine leaf shape. Using polynomial models, we create continuous representations of leaf development in 10 Vitis spp. We visualize a high‐resolution morphospace in which genetic and developmental sources of leaf shape variance are orthogonal to each other. Using classifiers, Vitis vinifera, Vitis spp., rootstock and dissected leaf varieties as well as developmental stages are accurately predicted. Theoretical eigenleaf representations sampled from across the morphospace that we call synthetic leaves can be classified using models. By predicting a high‐resolution morphospace and delimiting the boundaries of leaf shapes that can plausibly be produced within the genus Vitis, we can sample synthetic leaves with realistic qualities. From an ampelographic perspective, larger numbers of leaves sampled at lower resolution can be projected onto this high‐resolution space, or, synthetic leaves can be used to increase the robustness and accuracy of machine learning classifiers. Foliar development involves successive phases of cell proliferation and expansion that determine the final leaf size, and is characterized by an early burst of reactive oxygen species generated in the photosynthetic electron transport chain (PETC). Introduction of the alternative PETC acceptor flavodoxin in tobacco chloroplasts led to a reduction in leaf size associated to lower cell expansion, without affecting cell number per leaf. Proteomic analysis showed that the biogenesis of the PETC proceeded stepwise in wild-type leaves, with accumulation of light-harvesting proteins preceding that of electron transport components, which might explain the increased energy and electron transfer to oxygen and reactive oxygen species build-up at this stage. Flavodoxin expression did not affect biogenesis of the PETC but prevented hydroperoxide formation through its function as electron sink. Mature leaves from flavodoxin-expressing plants were shown to contain higher levels of transcripts encoding components of the proteasome, a key negative modulator of organ size. Proteome profiling revealed that this differential accumulation was initiated during expansion and led to increased proteasomal activity, whereas a proteasome inhibitor reverted the flavodoxin-dependent size phenotype. Cells expressing plastid-targeted flavodoxin displayed lower endoreduplication, also associated to decreased organ size. These results provide novel insights into the regulation of leaf growth by chloroplast-generated redox signals, and highlight the potential of alternative electron shuttles to investigate the link(s) between photosynthesis and plant development. To investigate the dynamics of stomata, transpiration, and photosynthesis under varying light intensities and CO2 conditions during leaf development, the light response and CO2 response of stomatal conductance (gsw), transpiration rate (Tr), and net photosynthetic rate (Pn) were observed for rice leaves at different days after leaf emergence (DAE). The results showed that (1) as photosynthetically active radiation (PAR) increased, leaf gsw, Tr, and Pn initially increased rapidly and linearly, followed by a more gradual rise to maximum values, and then either stabilized or showed a declining trend. The maximum gsw, Tr, and Pn were smaller and occurred earlier for old leaves than for young leaves. The gsw, Tr, and Pn all exhibited a linear decreasing trend with increasing DAE, and the rate of decrease slowed down with the reduction in PAR; (2) as the CO2 concentration (Ca) increased, gsw and Tr decreased gradually to a stable minimum value, while Pn increased linearly and slowly up to the maximum and then kept stable or decreased. The gsw, Tr, and Pn values initially kept high and then decreased with the increase of DAE. These results contribute to understanding the dynamics in gsw, Tr, and Pn during rice leaf growth and their response to varied light and CO2 concentration conditions and provide mechanistic support to estimate dynamic evapotranspiration and net ecosystem productivity at field-scale and a larger scale in paddy field ecosystems through the upscaling of leaf-level stomatal conductance, transpiration, and photosynthesis. The leaf is a determinate organ with a final size under genetic control. Numerous factors that regulate the final leaf size have been identified in Arabidopsis thaliana ; although most of these factors play their roles during the growth of leaf primordia, much less is known about leaf initiation and its effects on the final leaf size. In this study, we characterized oligocellula6-D (oli6-D), a semidominant mutant of A. thaliana with smaller leaves than the wild type (WT) due to its reduced leaf cell numbers. A time-course analysis showed that oli6-D had approximately 50% fewer leaf cells even immediately after leaf initiation; this difference was maintained throughout leaf development. Next-generation sequencing showed that oli6-D had chromosomal duplications involving 2-kb and 3-Mb regions of chromosomes 2 and 4, respectively. Several duplicated genes examined had approximately 2-fold higher expression levels, and at least one gene acquired a new intron/exon structure due to a chromosome fusion event. oli6-D showed reduced auxin responses in leaf primordia, primary roots and embryos, as well as reduced apical dominance and partial auxin-resistant root growth. CRISPR-associated protein-9-mediated genome editing enabled the removal of a 3-Mb duplicated segment, the largest targeted deletion in plants thus far. As a result, oli6-D restored the WT leaf phenotypes, demonstrating that oli6-D is a gain-of-function mutant. Our results suggest a new regulatory point of leaf size determination that functions at a very early stage of leaf development and is negatively regulated by one or more genes located in the duplicated chromosomal segments. The enzyme C-14 sterol reductase is involved in biosynthesis of brassinosteroids (BR) and sterols, as well as plant development. OsFK1, a member of the sterol biosynthesis pathway located in the endoplasmic reticulum (ER), encodes C-14 sterol reductase. However, there is little research on the function of C-14 sterol reductase in rice. Compared with the wild type, an osfk1 mutant showed dwarf phenotype and premature aging in the second leaf during the trefoil stage, and abnormal development of leaf veins during the tillering stage. The osfk1 mutant showed signs of aberrant PCD, as evidenced by TUNEL staining. This suggested that high ROS buildup caused DNA damage and ROS-mediated cell death in the mutant. The osfk1 mutant also showed decreased chlorophyll content and aberrant chloroplast structure. Sequencing of the osfk1 mutant allele revealed a non-synonymous G to A mutation in the final intron, leading to early termination. Here, we identified the OsFK1 allele, cloned it by Mutmap sequencing, and verified it by complementation. HPLC-MS/MS assays demonstrated that the osfk1 mutation caused lower phytosterol levels. Oak gall wasps have evolved strategies to manipulate the developmental pathways of their host to induce gall formation. This provides shelter and nutrients for the developing larva. Galls are entirely host tissue; however, the initiation, development, and physical appearance are controlled by the inducer. The underlying molecular mechanisms of gall formation, by which one or a small number of cells are reprogrammed and commit to a novel developmental path, are poorly understood. In this study, we sought a deeper insight into the molecular underpinnings of this process.Oak gall wasps have two generations each year, one sexual, and one asexual. Galls formed by these two generations exhibit a markedly different appearance. We sequenced transcriptomes of both the asexual and sexual generations of Neuroterus quercusbaccarum and Neuroterus numismalis. We then deployed Nanopore sequencing to generate long‐read sequences to test the hypothesis that gall wasps introduce DNA insertions to determine gall development.We detected potential genome rearrangements but did not uncover any non‐host DNA insertions. Transcriptome analysis revealed that transcriptomes of the sexual generations of distinct species of wasp are more similar than inter‐generational comparisons from the same species of wasp.Our results highlight the intricate interplay between the host leaves and gall development, suggesting that season and requirements of the gall structure play a larger role than species in controlling gall development and structure. Phosphorus (P) and potassium (K) stress significantly affect the growth, physiological characteristics, and nutrient uptake of rice plants. This study investigated the photosynthetic nitrogen (N) metabolism, N uptake, and N utilization of plants under varied P and K supplies. Two local conventional high-yield rice varieties (Shennong 265 and Liaojing 294) were used. These varieties were subjected to the following hydroponic experimental treatments: HPHK (normal P and K concentrations), HPLK (normal P and 1/20 normal K concentration), LPHK (normal K and 1/20 normal P concentration), and LPLK (1/20 normal P and K concentrations). The results showed that the mesophyll cells had a relatively complete nuclear and chloroplast structures, and the antioxidant enzymes of the plants were significantly reduced under the HPHK treatment. Compared to the LP treatments (LPHK and LPLK), the HPHK treatment was found to have the following potential effects: effectively optimize plant configuration; promote leaf development (Pn, E, Ci, and Tr, chlorophyll, and leaf area index); significantly increase the N-metabolism-related enzyme activity of leaves and roots and the accumulation of N in the plant in the main growth stages; and significantly increase the rice yield and N-related efficiency. In conclusion, the HPHK treatment was found to be beneficial in improving the plant configuration, promoting photosynthetic N metabolism, and increasing grain yield and N-related utilization efficiency. Lobed leaves are vital in the high-density cultivation and breeding of wax gourd (Benincasa hispida). Thus, determining the molecular mechanisms underlying the development of lobed leaves is important. To this end, this study aimed to resequence 105 recombinant inbred lines, constructed using the parental lines, GX-7 and my-1, to elucidate the molecular mechanisms underlying leaf development in wax gourd. Genes associated with lobed leaves in wax gourds were first evaluated via quantitative trait loci (QTL) mapping. Next, the F2 population was expanded to 2000 plants for fine mapping and candidate gene analysis. Thus, the candidate area was reduced to 1.129 Mb, located between markers InDel980 and InDel853. Functional analyses of candidate genes were performed using gene functional annotation, coding sequence analyses, and expression analyses. Among 48 genes in the candidate region, only Bch04G012650 (termed BhDDL4.1) showed differences in expression between the two parents. Using sequence differences of previously screened candidate genes, an InDel marker (InDel623) was developed in BhDDL4.1 for molecular marker–assisted breeding of wax gourd; the accuracy rate was 74.03%. Our results indicate that BhDDL4.1 may play a key role in the regulation of the lobed leaf trait; thereby, we provide a theoretical basis for further exploration of the molecular mechanisms underlying the lobed leaf trait in wax gourd. Leaf is an important organ of photosynthesis in plants, and leaf color has an important influence on photosynthesis. However, the molecular mechanisms of leaf color regulation are still unclear. In this study, we identified a rice (Oryza sativa) temperature sensitive albino trans-green (tag) mutant, which showed albino leaves at the seedling stage, and gradually turned green. Map-based cloning showed that TAG is a Novel allele gene of OsBT3, encoding an adenine nucleotide transport protein. Compared with the allele mutants of sla and wsl2, tag displayed some new phenotypes, such as it was a low-temperature-sensitive albino trans-green mutant with serious DNA damage in the albino leaves, and the PSI photosynthetic efficiency and photosynthesis of it were enhanced significantly in mature stage. Besides, we also found TAG was also localized in chloroplasts and played an important role on chloroplast development. Moreover, we newly discovered that TAG is highly expressed in L3U, which is induced by low temperature, and in situ hybridization results showed TAG expressed in the shoot apical meristem, leaf primordium and vascular bundle. Considering that the presence of a leafy head is a key morphological characteristic that determines the yield and quality of Chinese cabbage, identifying the major genes regulating the formation of a leafy head is crucial for variety improvement. A heading-related gene, BrKS, was previously predicted from a non-heading mutant, nhm1, derived from a heading variety, which encodes a key enzyme involved in gibberellin biosynthesis. Here, another mutant, nhm2, was identified from the same EMS-mutagenized population, and the phenotype of nhm2 was consistent with that of nhm1. We crossed mutants nhm1 and nhm2, and their F1 plants exhibited the mutant phenotype, which indicated that their mutant genes were allelic. A single non-synonymous mutation in the fourth exon of BrKS in mutant nhm1 and another single non-synonymous mutation in the tenth exon of BrKS in mutant nhm2. The same gene mutation in mutants nhm1 and nhm2 produced a similar non-heading phenotype, which confirmed the role of BrKS in the leafy head formation of Chinese cabbage. RNA-Seq analysis indicated that a transcription factor gene, BrERF1A, which is associated with leaf development, significantly down-regulated expression in mutant nhm1, and after the mutant was treated with GA3, the expression level of BrERF1A was recovered, which indicated that BrKS might be involved in leafy head formation through regulating the expression level of BrERF1A. This study examines the growth patterns, leaf morpho-anatomical traits and physiological characteristics of three ornamental taxa of Alocasia: A. melo, A reginae and Alocasia × morfontanensis 'Bambino'. Using optical and scanning electron microscopy, the research obtained anatomical details of the leaves. Alocasia × morfontanensis 'Bambino' exhibited the highest leaf growth rate among the taxa studied, showing significant leaf number increases from the 8th to the 12th week after planting. Alocasia melo produced 2–3 new basal shoots after 20 weeks, whereas the other two taxa did not produce any. Alocasia melo displayed the greatest relative water content and leaf thickness, while A. reginae showed the highest chlorophyll stability. This study also revealed variations in the adaxial and abaxial leaf color, stomatal width, epidermal thickness, and elemental composition among the taxa involved, with A. melo containing rhodium and high levels of calcium on their leaf surface. These findings contribute to the understanding of the Alocasia leaf morpho-anatomy and growth, which is useful for optimizing Alocasia cultivation and production across diverse ecosystems. Sesame, a prized oil crop, is acknowledged for its distinctive aroma, health benefits, and copious antioxidants, including sesamin and sesamol. Despite the importance of features like wrinkled leaf and pollen sterility in sesame breeding, scant research into the former impedes progress in cultivation and production. This study endeavors to illuminate the etiology of wrinkled leaves in sesame by probing agronomic and biochemical factors like hydration, nutrient makeup, antioxidant enzymes, and hormones. The investigation found substantial disparities between mutant wrinkled leaves and normal ones; the mutants exhibited heightened water content (86.60% vs 84.99%), augmented dry leaf weight, greater stomata density per unit area (174.58 vs 146.32 cm–2), and a significantly amplified carotenoid content by 27.5%. The study deduced that deficient gibberellic acid (GA3) production in mutant leaves during the seedling stage (< 0.005 ng g−1) restricts leaf elongation, causing wrinkles. Moreover, abnormal auxin (IAA) production during the seedling stage (< 8.57 ng g−1) and deficient jasmonic acid (JA) in both central and flat leaves (< 12.0 ng g−1) were associated with wrinkling. Furthermore, excessive late-stage GA3 production (> 0.041 ng g−1) led to persistent wrinkling and postponed maturity. This research, the first of its kind, elucidates the biochemical origins of sesame's wrinkled leaf formation, offering insights into the mechanisms of this trait and potentially facilitating the creation of new sterile lines and endorsement of sesame as a fresh-leaf vegetable. Taraxacum kok-saghyz (TKS) is a natural rubber (NR)-producing plant and a model plant for studying the biosynthesis of NR. Analyzing and studying the biosynthetic mechanism of NR is an important way to cultivate high-yield rubber TKS varieties. JAZ proteins, which belong to the Jasmonate ZIM domain family, function as negative regulators in the jasmonic acid (JA) signal transduction pathway. MYC2 is typically regarded as a regulatory factor for the target genes of JAZ proteins; JAZ proteins indirectly influence the gene expression regulated by MYC2 by modulating its activity. Theoretically, JAZ is expected to participate in growth, development, and responses to environmental cues related to rubber and biomass accumulation in TKS, all of which rely on the interaction between JAZ and MYC2. In this study, we identified 11 TkJAZs through homology searching of the TKS genomes and bioinformatics analyses. Subcellular localization, Y2H, and BiFC analysis demonstrate that TkJAZs and TkMYC2 are localized in the nucleus, with all TkJAZs and TkMYC2 showing nuclear colocalization interactions. Overexpression of TkMYC2 in TKS inhibited leaf development, promoted root growth, and simultaneously increased NR production. RNA-seq and qRT-PCR analysis revealed that the TkSRPP/REF genes exhibit varying degrees of upregulation compared to the wild type, upregulating the TkREF1 gene by 3.7-fold, suggesting that TkMYC2 regulates the synthesis of NR by modulating the TkSRPP/REF genes. Plant anatomy is patterned early during leaf development which suggests studying the spatial–temporal transcriptomes of primordia will help identify critical regulative and functional genes. We successfully isolated the leaf primordia tissues from the C3 grass rice and the C4 grass foxtail millet by laser capture microdissection (LCM) and studied the gene expression throughout leaf developmental stages. Our data analysis uncovered the conserved expression patterns of certain gene clusters both in rice and foxtail millet during leaf development. We revealed genes and transcription factors involved in vein formation, stomatal development, and suberin accumulation. We identified 79 candidate genes associated with functional regulation of C4 anatomy formation. Screening phenotype of the candidate genes revealed that knock-out of a putative polar auxin transport related gene NAL1 resulted significantly reduced veinal space in rice leaf. Our present work provides a foundation for future analyses of genes with novel functions in grasses and their role in leaf development, in particular the role in leaves with a contrasting C3 vs. C4 biosynthetic pathway.

Abstract Background The plant-specific YABBY transcription factor family plays important roles in plant growth and development, particularly leaf growth, floral organ formation, and secondary metabolite synthesis. Results Here, we identified a total of 13 OfYABBY genes from the Osmanthus fragrans genome. These 13 OfYABBY genes were divided into five subfamilies through phylogenetic analysis, and genes in the same subfamily showed similar gene structures and conserved protein motifs. Gene duplication promoted the expansion of the OfYABBY family in O. fragrans. Tissue-specific expression analysis showed that the OfYABBY family was mainly expressed in O. fragrans leaves and floral organs. To better understand the role of OfYABBY genes in plant growth and development, OfYABBY12 was selected for heterologous stable overexpression in tobacco, and OfYABBY12-overexpressing tobacco leaves released significantly fewer volatile organic compounds than wild-type tobacco leaves. Overexpression of OfYABBY12 led to the downregulation of NtCCD1/4 and decreased β-ionone biosynthesis. Correspondingly, a dual-luciferase assay showed that OfYABBY12 negatively regulated the expression of OfCCD4, which promotes β-ionone synthesis. Furthermore, tobacco leaves overexpressing OfYABBY12 were curled and wrinkled and had significantly reduced leaf thickness and leaf inclusions and significantly extended flower pistils (styles). Conclusion Overall, the results suggest that the OfYABBY gene family may influence the biosynthesis of the floral scent (especially β-ionone) in O. fragrans and may regulate leaf morphogenesis and lateral organs.

Heterophylly is a phenomenon whereby an individual plant dramatically changes leaf shape in response to the surroundings. Hygrophila difformis (Acanthaceae; water wisteria), has recently emerged as a model plant to study heterophylly because of its striking leaf shape variation in response to various environmental factors. When submerged, H. difformis often develops complex leaves, but on land it develops simple leaves. Leaf complexity is also influenced by other factors, such as light density, humidity, and temperature. Here, we sequenced and assembled the H. difformis chromosome‐level genome (scaffold N50: 60.43 Mb, genome size: 871.92 Mb), which revealed 36 099 predicted protein‐coding genes distributed over 15 pseudochromosomes. H. difformis diverged from its relatives during the Oligocene climate‐change period and expanded gene families related to its amphibious habit. Genes related to environmental stimuli, leaf development, and other pathways were differentially expressed in submerged and terrestrial conditions, possibly modulating morphological and physiological acclimation to changing environments. We also found that auxin plays a role in H. difformis heterophylly. Finally, we discovered candidate genes that respond to different environmental conditions and elucidated the role of LATE MERISTEM IDENTITY 1 (LMI1) in heterophylly. Plants must cope with ever-changing temperature conditions in their environment. In many plant species, suboptimal high and low temperatures can induce adaptive mechanisms that allow optimal performance. Thermomorphogenesis is the acclimation to high ambient temperature, whereas cold acclimation refers to the acquisition of cold tolerance following a period of low temperatures. The molecular mechanisms underlying thermomorphogenesis and cold acclimation are increasingly well understood but neither signalling components that have an apparent role in acclimation to both cold and warmth, nor factors determining dose-responsiveness, are currently well defined. This can be explained in part by practical limitations, as applying temperature gradients requires the use of multiple growth conditions simultaneously, usually unavailable in research laboratories. Here we demonstrate that commercially available thermal gradient tables can be used to grow and assess plants over a defined and adjustable steep temperature gradient within one experiment. We describe technical and thermodynamic aspects and provide considerations for plant growth and treatment. We show that plants display the expected morphological, physiological, developmental and molecular responses that are typically associated with high temperature and cold acclimation. This includes temperature dose-response effects on seed germination, hypocotyl elongation, leaf development, hyponasty, rosette growth, temperature marker gene expression, stomatal conductance, chlorophyll content, ion leakage and hydrogen peroxide levels. In conclusion, thermal gradient table systems enable standardized and predictable environments to study plant responses to varying temperature regimes and can be swiftly implemented in research on temperature signalling and response. Introduction: The research on plant leaf morphology is of great significance for understanding the development and evolution of plant organ morphology. As a relict plant, the G. biloba leaf morphology typically exhibits bifoliate and peltate forms. However, throughout its long evolutionary history, Ginkgo leaves have undergone diverse changes. Methods: This study focuses on the distinct "trumpet" leaves and normal fan-shaped leaves of G. biloba for analysis of their phenotypes, photosynthetic activity, anatomical observations, as well as transcriptomic and metabolomic analyses. Results: The results showed that trumpet-shaped G. biloba leaves have fewer cells, significant morphological differences between dorsal and abaxial epidermal cells, leading to a significantly lower net photosynthetic rate. Additionally, this study found that endogenous plant hormones such as GA, auxin, and JA as well as metabolites such as flavonoids and phenolic acids play roles in the formation of trumpet-shaped G. biloba leaves. Moreover, the experiments revealed the regulatory mechanisms of various key biological processes and gene expressions in the trumpet-shaped leaves of G. biloba. Discussion: Differences in the dorsal and abdominal cells of G. biloba leaves can cause the leaf to curl, thus reducing the overall photosynthetic efficiency of the leaves. However, the morphology of plant leaves is determined during the primordia leaf stage. In the early stages of leaf development, the shoot apical meristem (SAM) determines the developmental morphology of dicotyledonous plant leaves. This process involves the activity of multiple gene families and small RNAs. The establishment of leaf morphology is complexly regulated by various endogenous hormones, including the effect of auxin on cell walls. Additionally, changes in intracellular ion concentrations, such as fluctuations in Ca2+ concentration, also affect cell wall rigidity, thereby influencing leaf growth morphology. Many plants show strong heteroblastic changes in the shape and size of organs as they transition from juvenile to reproductive age. Most attention has been focused on heteroblastic development in leaves, but we wanted to understand heteroblastic changes in reproductive organ size. We therefore studied the progression of reproductive development in the model plant Arabidopsis thaliana , and found strong reductions in the size of flowers, fruit, seed, and internodes during development. These did not arise from correlative inhibition by older fruits, or from changes in inflorescence meristem size, but seemed to stem from changes in the size of floral organ primordia themselves. We hypothesized that environmental conditions might influence this heteroblastic pattern and found that the ambient temperature during organ initiation strongly influences organ size. We show that this temperature-dependent heteroblasty is dependent on FLOWERING LOCUS T (FT)-mediated signal integration, adding to the repertoire of developmental processes regulated by this pathway. Our results demonstrate that rising global temperatures will not affect just fertility, as is widely described, but also the size and seed number of fruits produced. Habitats resulting from agricultural intensification harbour a wide array of stressors for resident organisms. Environmental stress during development can leave imprints on the phenotype that are useful as indicators of its impairment. Among them, fluctuating asymmetry (FA), considered the morphological manifestation of developmental instability (DI), may reflect the increment of developmental noise and the inefficiency of buffering mechanisms. Here, we evaluated for the first time the impact of intensive agriculture on the development of a viviparous reptile, the asp viper, Vipera aspis (Linnaeus 1758), by means of FA. To this end, we first quantified unsigned asymmetry indexes (uAI) of dorsal head shape captured by geometric morphometrics (GM) and of seven pholidotic traits in 127 and 482 individuals, respectively. Using linear models, we then tested for variation in uAI according to habitat type. Furthermore, we tested the effect of climatic variables with a potential influence on development, and thus, on FA. We performed analyses at the V. aspis zinnikeri subspecies range and at the population level, including three well‐sampled natural localities (NAT) and three sites of intensive agriculture (AGR). We found significantly higher FA levels in dorsal head shape in individuals from agricultural habitats than in those from natural habitats, which suggests that asp viper development is impaired by stressors resulting from agricultural intensification. Our results also point to a lower sensitivity of traditional morphological tools to capture the signal that intensive agriculture leaves on development and a lack of capacity of climatic factors in isolation to predict variation in the degree of asymmetry. In addition to encouraging the use of high‐precision methods, we hypothesize on the overall effect of different stressors linked to intensive agriculture as the real constraint. Background: In plants, the leaf functions as a solar panel, where photosynthesis converts carbon dioxide and water into carbohydrates and oxygen. In soybean, leaf type traits, including leaf shape, leaf area, leaf width, and leaf width so on, are considered to be associated with yield. In this study, we performed morphological characterization, transcriptome analysis, and endogenous hormone analysis of a rolled and narrow leaf mutant line (rl) in soybean. Results: Compared with wild type HX3, mutant line rl showed rolled and narrower leaflet, and smaller leaf, meanwhile rl also performed narrower pod and narrower seed. Anatomical analysis of leaflet demonstrated that cell area of upper epidermis was bigger than the cell area of lower epidermis in rl, which may lead rolled and narrow leaf. Transcriptome analysis revealed that several cytokinin oxidase/dehydrogenase (CKX) genes (Glyma.06G028900, Glyma.09G225400, Glyma.13G104700, Glyma.14G099000, and Glyma.17G054500) were up-regulation dramatically, which may cause lower cytokinin level in rl. Endogenous hormone analysis verified that cytokinin content of rl was lower. Hormone treatment results indicated that 6-BA rescued rolled leaf enough, rescued partly narrow leaf. And after 6-BA treatment, the cell area was similar between upper epidermis and lower epidermis in rl. Although IAA content and ABA content were reduced in rl, but exogenous IAA and ABA didn't affect leaf type of HX3 and rl. Conclusions: Our results suggest abnormal cytokinin metabolism caused rolled and narrow leaf in rl, and provide valuable clues for further understanding the mechanisms underlying leaf development in soybean. [Objective] Using Chrysanthemum morifolium HETAI, a characteristic medicinal and edible plant in Zhaoqing, Guangdong, as the material, to explore the growth characteristics and nutritional changes of leaves and inflorescences. [Method] The growth of leaves and floral organs from Chrysanthemum morifolium was measured in the field, and corresponding colorimetric methods were used to explore the content changes of effective components. [Result] The Chrysanthemum morifolium HETAI developed from young leaves to mature leaves for about 168 days, with significant morphological changes during the leaf development process. The mature leaves were oval shaped, feathered with shallow or deep cracks, and the leaf edges were round serrated with notches. The growth cycle of flower organs was about 55 days, and it lasted for about 20 days from the initial flowering period to the full-bloom period. The tongue shaped and tubular flower clusters were fully developed; the flower diameter could reach 4.67-4.89 cm. During the development of flower organs, the content of soluble proteins generally showed a downward trend, but the content of total flavonoids and soluble sugars continued to increase, both contents maintain high levels during the development stage of 3140 days. Combined with the growth characteristics of leaves and inflorescences, the initial flowering period and early peak flowering period (31-40 days) were the suitable harvest time for this variety of chrysanthemum. ABSTRACT Domestication has shaped the population structure and agronomic traits of tea plants, yet the complexity of tea population structure and genetic variation that determines these traits remains unclear. We here investigated the resequencing data of 363 diverse tea accessions collected extensively from almost all tea distributions and found that the population structure of tea plants was divided into eight subgroups, which were basically consistent with their geographical distributions. The genetic diversity of tea plants in China decreased from southwest to east as latitude increased. Results also indicated that Camellia sinensis var. assamica (CSA) illustrated divergent selection signatures with Camellia sinensis var. sinensis (CSS). The domesticated genes of CSA were mainly involved in leaf development, flavonoid and alkaloid biosynthesis, while the domesticated genes in CSS mainly participated in amino acid metabolism, aroma compounds biosynthesis, and cold stress. Comparative population genomics further identified ~730 Mb novel sequences, generating 6,058 full‐length protein‐encoding genes, significantly expanding the gene pool of tea plants. We also discovered 217,376 large‐scale structural variations and 56,583 presence and absence variations (PAVs) across diverse tea accessions, some of which were associated with tea quality and stress resistance. Functional experiments demonstrated that two PAV genes (CSS0049975 and CSS0006599) were likely to drive trait diversification in cold tolerance between CSA and CSS tea plants. The overall findings not only revealed the genetic diversity and domestication of tea plants, but also underscored the vital role of structural variations in the diversification of tea plant traits. Plants consist of fundamental units of growth called phytomers (leaf or bract, axillary bud, node, and internode), which are repeated and modified throughout shoot development to give plants plasticity for survival and adaptation. One phytomer modification is the suppression or outgrowth of bracts, the leaves subtending the flowers. The floral meristem identity regulator LEAFY (LFY) and the organ boundary genes BLADE-ON-PETIOLE1 (BOP1) and BOP2 have been shown to suppress bract development in Arabidopsis, as mutations in these genes result in bract outgrowth. However, much less is known about the mechanisms that promote bract outgrowth in Arabidopsis mutants such as these. Further understanding of this mechanism may provide a potential tool for modifying leaf development. Here, we showed that the MADS-box genes SUPPRESSOR OF OVEREXPRESSION OF CONSTANS1 (SOC1), FRUITFUL (FUL), and AGAMOUS-LIKE24 (AGL24) play more important roles than BOP1/2 and LFY in bract suppression, and that AINTEGUMENTA (ANT) and the partially redundant AINTEGUMENTA-LIKE6 (AIL6) are necessary for bract outgrowth in these mutant backgrounds. We also demonstrated that misexpression of AIL6 alone is sufficient for bract outgrowth. Our data reveal a mechanism for bract suppression and outgrowth and provide insight into phytomer plasticity. Leaves of flowering plants are characterized by diverse venation patterns. Patterning begins with the selection of vein-forming procambial initial cells from within the ground meristem of a developing leaf, a process which is considered to be auxin-dependent, and continues until veins are anatomically differentiated with functional xylem and phloem. At present, the mechanisms responsible for leaf venation patterning are primarily characterized in the model eudicot Arabidopsis thaliana which displays a reticulate venation network. However, evidence suggests that vein development may proceed via a different mechanism in monocot leaves where venation patterning is parallel. Here, we employed Molecular Cartography, a multiplexed in situ hybridization technique, to analyze the spatiotemporal localization of a subset of auxin-related genes and candidate regulators of vein patterning in maize leaves. We show how different combinations of auxin influx and efflux transporters are recruited during leaf and vein specification and how major and minor vein ranks develop with distinct identities. The localization of the procambial marker PIN1a and the spatial arrangement of procambial initial cells that give rise to major and minor vein ranks further suggests that vein spacing is prepatterned across the medio-lateral leaf axis prior to accumulation of the PIN1a auxin transporter. In contrast, patterning in the adaxial-abaxial axis occurs progressively, with markers of xylem and phloem gradually becoming polarized as differentiation proceeds. Leaves are the nutritive organs of rice. Leaf shape influences rice photosynthesis, subsequently impacting yield. Gibberellins, GAs, are important hormones, but the way in which GAs regulate leaf width is largely unknown. This study focuses on the d18 mutant with broader leaves due to defective GA biosynthesis. Statistical analysis indicates broader leaves in the d18 mutant compared to the wild-type group. An examination of leaf cell morphology shows a higher count of secondary vascular bundles in d18 than in the wild-type group. RNA-seq analysis demonstrates significantly higher expression of CYCB (CYCLIN B) and H4 (HISTONE H4) in d18 compared to wild type. Leaf angle (LA) is a major agronomic trait of rice plant architecture, which is determined by the development of the leaf lamina joint (LJ) and closely related to plant yield. The LJ is formed by the leaf collar belt (ligule band), leaf tongue (ligule) and leaf ear (auricle). Parenchyma cells (PCs), sclerenchyma cells (SCs), vascular bundles (VBs), and arenchyma tissue (Ac) are present in the cross sections of LJ. The genetic and molecular regulation of rice leaf angle and LJ development has been well investigated in recent years. However, the underlying complex regulatory network still requires further elucidation and comprehensive discussion. In this review, we briefly describe the cellular characteristics of four typical stages of LJ development, and systematically summarize the genes regulating rice leaf inclination based on their roles in PC proliferation and elongation, as well as SC proliferation and differentiation. We also introduce the emerging regulatory pathways of phytohormones and transcription factors (TFs), environmental cues that are involved in rice LJ development, discussing the relevant intricate signal network that is relevant to provide further insights into the genetic improvement of leaf angle traits in rice breeding. Chamaecyparis obtusa (Siebold et Zucc.) Endl. is cultivated in the temperate region of East Asia for its high-quality and profitable timber. The timber-processing industry produces a large amount of waste, such as tree bark, branches, and leaves, and finding ways to minimize such waste is economically and environmentally efficient. In the present study, the allelopathic activity and allelochemicals of the C. obtusa leaves were investigated to develop potential application methods. C. obtusa leaves were phytotoxic and associated leaf extracts significantly suppressed the growth of the weeds; Echinochloa crus-galli, Phleum pratense and Lolium multiflorum under laboratory conditions. The leaf powder applied onto the soil surface also suppressed the germination of E. crus-galli and P. pratense under greenhouse conditions. Hinokiic acid and (+)-dihydrosesamin were isolated from the leaves and structurally identified. Both compounds suppressed the growth of cress and P. pratense in a concentration dependent manner. When the leaves of C. obtua were applied as a soil additive, hinokiic acid and (+)-dihydrosesamin in the leaves potentially cause the growth inhibitory activity by leaching into the soil. These experiments showed that the leaves and the leaf extracts of C. obtusa are phytotoxic. These experiments also demonstrated that the leaves and the leaf extracts of C. obtusa have growth inhibitory potential against several plant species, and the leaves produce allelochemicals. Therefore, the leaves and leaf extracts of C. obtusa may be useful for weed suppression purposes. The leaf biomass of C. obtusa is frequently large and the timber processing industry produces significant leaf waste. The development of weed control products using waste leaves may be a solution to minimize the timber processing waste to reduce environmental impact and provide economic value. Maize is an important food and feed crop. Under limited arable land area, the cultivation of high-density-tolerance crops is a key factor in promoting yield improvement. Leaf width and stalk strength are important influences on density tolerance in maize. However, no comprehensive transcriptomic analysis has focused on maize's leaf width and stalk strength formation mechanisms. In this study, comparative transcriptomic analyses demonstrated that significant transcriptome changes occurred regarding leaf width and stalk strength of narrow-leaved and wide-leaved maize inbred lines, with a total of 5001 differentially expressed genes (DEGs) identified. Enrichment analysis showed that phenylpropanoid biosynthesis, starch and sucrose metabolism, phytohormone signaling, amino acid metabolism, and brassinosteroid biosynthesis were significantly correlated with the formation of maize leaf shape and stalk strength and that the genes in these pathways were primarily involved in cell wall formation. Weighted gene co-expression network analysis identified 2 modules significantly correlated with leaf width and stalk strength, from which 11 hub genes were further identified. The 11 leaf and stem development genes in different pathways were validated using qRT-PCR. These findings can provide a theoretical basis for the mechanism of narrow-leaf and high-strength stalk formation in high-density-tolerance maize and contribute to the proposal of a breeding strategy for yield improvement. Plant leaves contain multiple cell types which achieve distinct characteristics whilst still coordinating development within the leaf. The bundle sheath possesses larger individual cells and lower chloroplast content than the adjacent mesophyll, but how this morphology is achieved remains unknown. To identify regulatory mechanisms determining bundle sheath cell morphology we tested the effects of perturbing environmental (light) and endogenous signals (hormones) during leaf development of Oryza sativa (rice). Total chloroplast area in bundle sheath cells was found to increase with cell size as in the mesophyll but did not maintain a 'set‐point' relationship, with the longest bundle sheath cells demonstrating the lowest chloroplast content. Application of exogenous cytokinin and gibberellin significantly altered the relationship between cell size and chloroplast biosynthesis in the bundle sheath, increasing chloroplast content of the longest cells. Delayed exposure to light reduced the mean length of bundle sheath cells but increased corresponding leaf length, whereas premature light reduced final leaf length but did not affect bundle sheath cells. This suggests that the plant hormones cytokinin and gibberellin are regulators of the bundle sheath cell‐chloroplast relationship and that final bundle sheath length may potentially be affected by light‐mediated control of exit from the cell cycle. Summary statement: In rice (Oryza sativa) the timing of light exposure during leaf development changes mature bundle sheath morphology, reducing maximum cell length under extended darkness.
