The Community Pedestrian and Bicycle Safety Training Program (CPBST) is a joint project of the University of California Berkeley’s Safe Transportation Research and Education Center (SafeTREC) and California Walks (Cal Walks). Founded in 2009, the purpose of the CPBST is to: 1. Educate local residents and safety advocates on how to improve pedestrian and bicycle safety;2.Empower community partners to advocate for safety improvements in their neighborhoods; and3. Strengthen collaborations with local officials and agency staff to make California neighborhoods safer and more pleansant for walking and bicycling.SafeTREC and Cal Walks work with a Planning Committee, a group of local safety stakeholders, to convene a workshop, recruit participants, and tailor the curriculum to address the community’s needs and priorities. During the workshop, participants conduct a walking and biking assessment of priority areas in the community, learn about Safe System strategies to address walking and biking concerns, and develop an action plan with short-, mid-, and long-term recommendations.

SUMMARY: Photoperiod insensitivity (auto‐flowering) in drug‐type Cannabis sativa circumvents the need for short day (SD) flowering requirements making outdoor cultivation in high latitudes possible. However, the benefits of photoperiod insensitivity are counterbalanced by low cannabinoid content and poor flower quality in auto‐flowering genotypes. Despite recent studies in cannabis flowering, a mechanistic understanding of photoperiod insensitivity is still lacking. We used a combination of genome‐wide association study and genetic fine‐mapping to identify the genetic cause of auto‐flowering in cannabis. We then used gene expression analyses and transient transformation assays to characterize flowering time control. Herein, we identify a splice site mutation within circadian clock gene PSEUDO‐RESPONSE REGULATOR 37 (CsPRR37) in auto‐flowering cannabis. We show that CsPRR37 represses FT expression and its circadian oscillations transition to a less repressive state during SD as compared to long days (LD). We identify several key circadian clock genes whose expression is altered in auto‐flowering cannabis, particularly under non‐inductive LD. Research into the pervasiveness of this mutation and others affecting flowering time will help elucidate cannabis domestication history and advance cannabis breeding toward a more sustainable outdoor cultivation system. Significance Statement: Auto‐flowering (photoperiod insensitive) cannabis can be cultivated outdoors even in northern latitudes, a key domestication trait that allows to harvest before the fall and rain season starts. Eukaryotes and prokaryotes possess fatty acid synthase (FAS) biosynthetic pathways that comprise iterative chain elongation, reduction, and dehydration reactions. The bacterial FASII pathway differs significantly from human FAS pathways and is a long-standing target for antibiotic development against Gram-negative bacteria due to differences from the human FAS, and several existing antibacterial agents are known to inhibit FASII enzymes. N-Acetylcysteamine (NAC) fatty acid thioesters have been used as mimics of the natural acyl carrier protein pathway intermediates to assay FASII enzymes, and we now report an assay of FabV from Pseudomonas aeruginosa using (E)-2-decenoyl-NAC. In addition, we have converted an existing UV absorbance assay for FabA, the bifunctional dehydration/epimerization enzyme and key target in the FASII pathway, into a high-throughput enzyme coupled fluorescence assay that has been employed to screen a library of diverse small molecules. With this approach, N-(4-chlorobenzyl)-3-(2-furyl)-1H-1,2,4-triazol-5-amine (N42FTA) was found to competitively inhibit (pIC50=5.7±0.2) the processing of 3-hydroxydecanoyl-NAC by P. aeruginosa FabA. N42FTA was shown to be potent in blocking crosslinking of Escherichia coli acyl carrier protein and FabA, a direct mimic of the biological process. The co-complex structure of N42FTA with P. aeruginosa FabA protein rationalises affinity and suggests future design opportunities. Employing NAC fatty acid mimics to develop further high-throughput assays for individual enzymes in the FASII pathway should aid in the discovery of new antimicrobials.

Sixty participants from all over the world gathered in Banff, Alberta, in Aug 1991 to discuss systems tracts expression, sequence stratigraphy in terrestrial systems, parasequences and key surfaces. The NUNA Conference saw geologists agreeing on otherwise confusing aspects of high-resolution sequence stratigraphy and allowed them to advance proposals to manage this problem.
