Dams constructed along waterways interrupt the dispersion and migration of aquatic organisms, affecting mainly the abundance of migratory fish species. Translocation mechanisms have been constructed at dams aiming to minimize their impact on fish species migration behavior. There is little information available about the effect of the construction of dams on the genetic structure of the Neotropical migratory fish fauna. Therefore, RAPD molecular markers and microsatellites were utilized to evaluate the diversity and genetic structure of the migratory species Leporinus elongatus (piapara) in the Canoas Complex - Paranapanema River - Brazil. Ten groups were sampled in the fish ladders of the hydroelectric dam Canoas I and Canoas II during the reproductive period in three consecutive years. Both markers showed a high level of genetic diversity within these groups. The microsatellite markers demonstrated a loss of heterozygosity and a considerable level of inbreeding in the species. The genetic differentiation found among the groups with both markers utilized is within a range from low to moderate. The data obtained with the parameter of genetic diversity among the groups led to the conclusion that the groups of L. elongatus of the Canoas Complex are structured as a single population composed of sub-populations with low genetic diversity among them. The data on genetic diversity and population structure of L. elongatus are of great importance for the development of the species management and conservation programs in the Canoas Complex, which can also be utilized in aquaculture programs. As barragens construídas ao longo de sistemas hídricos interrompem a dispersão e a migração dos organismos aquáticos, afetando principalmente a abundância das espécies de peixes migradores. Mecanismos para transposição foram construídos em barragens visando minimizar esses impactos. Poucas são as informações disponíveis sobre o efeito da construção de barragens na estrutura genética populacional da fauna neotropical de peixes migradores. Nesse contexto, marcadores moleculares RAPD e microssatélites foram utilizados para avaliar a diversidade e a estrutura genética da espécie migradora Leporinus elongatus (piapara) no Complexo Canoas - rio Paranapanema - Brasil. Dez grupos foram amostrados nas escadas para transposição de peixes das UHEs Canoas I e Canoas II durante o período reprodutivo em três anos consecutivos. Ambos os marcadores evidenciaram uma alta diversidade genética para esses grupos. Os marcadores microssatélites mostraram uma perda de heterozigosidade e uma considerável taxa de endocruzamento para a espécie. A diferenciação genética encontrada entre os grupos, com ambos os marcadores utilizados, pode ser considerada de moderada a baixa. Os dados obtidos com os parâmetros de diversidade genética entre os grupos permitiram concluir que os grupos de L. elongatus do Complexo Canoas estão estruturados como uma única população composta por sub-populações com baixa diversidade genética entre elas. Os dados obtidos sobre a diversidade genética e estrutura populacional de L. elongatus são de grande importância para o desenvolvimento de programas de manejo e conservação da espécie no Complexo Canoas, podendo também ser utilizados em programas de aqüicultura.

Despite the great anthropogenic interference on urban streams, information is still scarce about the genetic variability and structure of native fish populations inhabiting such streams. In the present study, random amplified polymorphic DNA (RAPD) markers were used to analyze genetic variability and structure of populations assigned to the Neotropical fish species Astyanax scabripinnis from an urban stream located in Londrina, Parana State, southern Brazil. Thirty individuals of this species were collected from three sites throughout the upper Cambe stream. A total of 10 primers amplified 159 loci, of which 128 (80.5%) were polymorphic. Each of the three populations showed very similar proportions of polymorphic loci, which ranged from 63.5 to 64.8%. Unbiased genetic distances varied from 0.0612 to 0.0646. Thetap-test values indicated moderate to high genetic differentiation among individuals from different localities. The number of migrants varied from 1.34 to 1.46, suggesting a low gene flow between populations. The genetic similarity among all individuals studied ranged from 0.424 to 0.848. The results suggest that populations of A. scabripinnis in Cambe stream are undergoing genetic differentiation.

Random amplified polymorphic DNA (RAPD) markers were used to analyze the genetic structure of Eufriesea violacea populations in three fragments (85.47, 832.58 and 2800 ha) of Atlantic rainforest located in the north of the Brazilian state of Paraná. A total of twelve primers produced 206 loci, of which 129 were polymorphic (95% criterion). The proportions of polymorphic loci in each population ranged from 57.28% to 59.2%, revealing very similar levels of genetic variability in the groups of bees from each fragment. Unbiased genetic distances between groups ranged from 0.0171 to 0.0284, the smallest genetic distance occurring between bees from the two larger fragments. These results suggest that the E. violacea populations from the three fragments have maintained themselves genetically similar to native populations of this species originally present in northern Paraná.

Pimelodus maculatus populations from the Tietê and Paranapanema rivers were sampled and had their genetic structure analyzed by using RAPD markers, with the aim of contributing to future conservation studies. The proportion of polymorphic loci was greater than 50% in the populations of both rivers. Genetic diversity data showed that, in spite of its nine hydroelectric plants, the Tietê river population was genetically homogeneous, whereas the Paranapanema river population was structured. This might be due to the presence of high waterfalls distributed all along its course. These data may serve as indicators for future conservation studies on the Tietê and Paranapanema rivers.

A similaridade genética de seis espécies, pertencentes à família Pimelodidae (Osteichthyes, Siluriformes), coletadas em quatro localidades na bacia do rio Tibagi, foi analisada utilizando-se a técnica de RAPD. A análise comparativa de RAPD foi realizada para observar o grau de similaridade genética entre seis espécies de Pimelodidae. O dendrograma obtido através da análise comparativa dos padrões de RAPD mostrou claramente que os indivíduos estudados de cada uma das espécies se agrupam entre si, ou seja, há uma separação definida entre as seis espécies. Dois agrupamentos foram obtidos, um formado pelas espécies P. maculatus, P. cf. absconditus, I. labrosus e P. pirinampu (Subfamília Pimelodinae), e outro formado pelas espécies P. aff. avanhandavae e Pimelodella aff. meeki (subfamília Heptapterinae). A similaridade genética entre esses agrupamentos foi de 0.084.The genetic similarity of six species from the Pimelodidae family (Osteichthyes, Siluriformes), collected at four sites in the Tibagi River basin, was analyzed using RAPD. RAPD comparative analysis was carried out to observe the degree of genetic similarity among the six species of Pimelodidae. The dendrogram obtained by analysis of the comparative RAPD profiles clearly showed that the individuals investigated from each species clustered together, that is, there was a defined separation among the six species. Two clusters also separated distinctly, one formed by the P. maculatus, P. cf. absconditus, I. labrosus and P. pirinampu (Pimelodinae subfamily) species and the other by the P. aff. avanhandavae and Pimelodella aff. meeki (Heptapterinae subfamily) species. The genetic similarity among these groupings was 0.084.

A técnica de Polimorfismo de DNA Amplificado ao Acaso (RAPD) foi utilizada na análise de oito espécies de peixe da família Anostomidae, com a finalidade de quantificar a variabilidade genética e estimar a similaridade genética dentro e entre essas espécies. Os resultados foram comparados com dados obtidos anteriormente com isoenzimas, para a maioria das espécies analisadas. A proporção de locos polimórficos obtida, com dez primers, está acima de 40 %, exceto para Leporinus amblyhynchus. Esse é um fato importante desde que a existência de variabilidade genética é uma condição importante para a sobrevida das espécies no meio ambiente Com dendrograma obtido, através de análise comparativa, foi possível discriminar as duas espécies de Schizodon daquelas do gênero Leporinus, no entanto não foi possível separar L. obtusidens e L. elongatus. A análise de RAPD indica que essas duas espécies são genéticamante muito similares, conclusão também sustentada por dados de estudos de isoenzimasRandom Amplified Polymorphic DNA (RAPD) techniques were applied to eight Anostomidae species to analyze the genetic variability and genetic similarity within and between species. This is especially important since genetic variability is a population feature both for short term fitness of the individual and for the long term survival of populations. Through subsequent analysis of dendrograms it was possible to discriminate the two Schizodon species from those of the genus Leporinus, even though it was impossible to differentiate L. obtusidens and L. elongates. Genetic variability in three species of Pimelodidae catfish (Iheringichthys labrosus, Pimelodus maculatus and Pimelodus cf. absconditus), collected at four sites in the Tibagi river-basin: Sertaneja, Londrina (lower Tibagi), Sapopema (middle Tibagi) and Tibagi (upper Tibagi) was analysed using RAPD and isoenzymatic techniques. Iheringichthys labrosus had the greatest genetic variability showing a proportion of mean polymorphic loci (P¯m) of 53.7%. The dendrogram showed that the individuals from the locations were clustered together, supporting the hypothesis that I. labrosus has a sedentary or localized geographical distribution. Pimelodus maculatus is the only species studied that is considered to be highly migratory. The P¯m obtained for this species was 30.2%. Their genetic dendrogram showed that the individuals were not clustered, based on the place of collection. P. cf. absconditus had a P¯m value of 47.4%, and the external morphology of this species is similar to that of I. labrosus, which hindered differentiation of individuals from the two species. RAPD and isoenzyme analysis, however, provided differential patterns for P. cf. absconditus from I. labrosus.

In the current literature, information is scarce on which part of the adult insect body is suitable for isolation of genomic DNA for genetic analysis based on DNA-markers. In this study, we evaluated RAPD profiles generated from total genomic DNA isolated from distinct body parts (head, legs, thorax + wings and abdomen) of 12 males of Euglossa pleosticta Dressler. From the total of bands analyzed, 9.0% did not show reproducibility. Percent variations of bands in each body segment were: 1.1% (head); 0.4% (legs); 0.8% (thorax/wings) and 6.7% (abdomen). The much higher variation (chi2one sample = 10.27; df = 1; P < 0.01) in the RAPD profiles obtained by using DNA isolated from abdomen of the euglossine males suggests that this body part of adult insects should be avoided in DNA extraction procedures. Conversely, the low variation among the RAPD profiles obtained from amplifications of genomic DNA extracted from head, legs and thorax/wings indicates that all these body parts of male bees are equally useful and secure for using in isolation and amplification procedures of total genomic DNA. Na literatura atual, são escassas as informações sobre qual a parte do corpo do inseto adulto é mais adequada para a extração de DNA genômico para estudos de análises genéticas baseadas em marcadores de DNA. Neste estudo, foram analisados os perfis de RAPD produzidos a partir da amplificação do DNA genômico extraído de partes distintas do corpo (cabeça, pernas, tórax + asas e abdome) de 12 machos de Euglossa pleosticta Dressler. Do total de bandas analisadas, 9,0% não mostraram repetibilidade. As porcentagens de variação de bandas em relação às diferentes partes do corpo das abelhas foram: 1,1% (cabeça); 0,4% (pernas); 0,8% (tórax/asas) e 6,7% (abdome). A maior variação observada (chi2para uma amostra = 10,27; gl = 1; P < 0,01), nos perfis eletroforéticos de RAPD, produzidos nas amplificações do DNA extraído do abdome de machos euglossíneos sugere que essa parte do corpo de insetos adultos deve ser evitada em procedimentos de extração de DNA. De modo diverso, a baixa variação entre os perfis de RAPD obtidos a partir das amplificações de DNA genômico extraído da cabeça, pernas e tórax/asas das abelhas indica que todas essas partes do corpo de machos euglossíneos são igualmente úteis e confiáveis para serem utilizadas para a extração e a amplificação do DNA genômico.

The genetic RAPD (random amplified polymorphic DNA) markers have been used successfully in taxonomical studies of several groups of organisms. In the present study these molecular markers were used to analyze the genetic similarity among eighteen males of Euglossa truncata Rebêlo & Moure exhibiting variations in two morphological characters (colour of the antennal scape and metaepisternal hairs) which are frequently used to identify this species of euglossine bee. The twelve primers used in the RAPD analysis amplified 127 loci, of which 40 (31.5%) were polymorphic, showing some variation among the individuals. The coefficients of genetic similarity among the individuals ranged from 0.79 to 0.95, indicating a rather high genetic similarity among the 18 male bees studied. No RAPD band was specific to any morphological character analyzed. The results indicate that all bees analyzed belong to the same species. The high genetic similarity among the eighteen euglossine males studied indicates that the variations observed in the morphological characters are not in disagreement with the identification of this species of Euglossina and these characters can vary among males of E. truncata. Os marcadores genéticos RAPD (DNA polimórfico amplificado ao acaso) têm sido empregados com sucesso em estudos taxonômicos de diversos grupos de organismos. No presente trabalho esses marcadores moleculares foram utilizados para analisar a similaridade genética entre dezoito machos de Euglossa truncata Rebêlo & Moure apresentando variações em dois caracteres morfológicos (coloração do escapo antenal e da pilosidade dos tufos metaepisternais), os quais normalmente são utilizados na identificação dessa espécie de abelha Euglossina. Os 12 primers utilizados nas análises produziram 127 locos de RAPD, dos quais 40 (31,5%) foram polimórficos, revelando alguma variação genética entre os indivíduos analisados. Os coeficientes de similaridade genética entre os indivíduos variaram de 0,79 a 0,95, indicando alta similaridade entre os 18 machos analisados. Nenhuma banda de RAPD mostrou-se específica para os caracteres morfológicos considerados. Os resultados obtidos indicam que todos os machos analisados pertencem à mesma espécie euglossina. A alta similaridade genética entre os dezoito machos eulossíneos indica que as variações observadas nos caracteres morfológicos destes não estão em discordância com a identificação dessa espécie de Euglossina, e que tais caracteres podem variar entre os machos de E. truncata.

In this study, the RAPD technique was used to analyze the genetic structure of populations of the fish Astyanax altiparanae (Characidae, Tetragonopterinae) living in the lower, middle and upper Paranapanema River, Brazil. The aim was to assess this structure regarding fish handling and conservation programs. The genetic variability (P) was found to be 42.64%, 75% and 75% in the low, middle and upper reaches, respectively. The dendrogram of genetic similarity, obtained by comparative analysis of the sets of samples from the three sites, showed the formation of three clusters. All of the genetic parameters used indicate that the population in the lower Paranapanema is genetically different from those in the middle and upper sections. The theta P test shows that the low Paranapanema is highly differentiated from the middle (0.2813) and upper (0.2912) Paranapanema, while the differentiation between the last two is moderate (0.0895). The data obtained in the present work suggest that recolonization and conservation studies should not be focused on the species A. altiparanae as such, but on the conservation units, because they are the genetically differentiated populations.

Genetic variability was studied in five fish species (Anostomidae): Schizodon intermedius and S. nasutus and Leporinus friderici, L. elongatus and L. obtusidens, collected at one location on the Tibagi River (Paraná, Brazil). The protein data from seven systems coded collectively for 19 loci in the liver, muscle and heart. Nine of these loci were polymorphic. The estimated proportion of polymorphism loci () varied from 16.7% in S. intermedius to 36.9% in L. friderici; the mean heterozygosity observed (o) was 0.027 ± 0.015 and 0.109 ± 0.042, respectively. The estimated value of the genetic identity among L. friderici and S. intermedius (0.749) and S. nasutus (0.787) suggested that these are "congeneric" species. Morphological characteristics indicate that these species belong to distinct genera, while isoenzymatic data show that they are very similar at the genetic/biochemical level. A variabilidade genética de 5 espécies da família Anostomidae pertencentes aos gêneros Schizodon (S. intermedius e S. nasutus) e Leporinus (L. friderici, L. elongatus e L. obtusidens) coletadas em uma localidade do rio Tibagi (Paraná, Brasil) foi analisada comparativamente utilizando dados protéicos de 7 sistemas que codificam l9 locos no fígado, músculo e coração. Dos locos identificados, 9 são polimórficos, com valores estimados de proporção de locos polimórficos () que variaram de 16.7% em S. intermedius a 36.85% em L. friderici, e a heterozigosidade média observada (o) foi de 0.027 ± 0.015 e 0.109 ± 0.042, nessas mesmas espécies. O valor estimado de identidade genética (I) entre L. friderici e S. intermedius (0.749) e S. nasutus (0.787) sugere que estas são espécies congenéricas. As características morfológicas determinam que estas espécies pertencem a gêneros distintos, no entanto os dados de identidade e distância genética obtidos demonstram que essas três espécies, no nível genético-bioquímico, têm uma maior similaridade.

Genetic variability of three Pimelodidae species, Pimelodus maculatus, Iheringichthys labrosus, and Pinirampus pirinampu, collected at one site in the Tibagi River, was comparatively analyzed using protein data for six systems which code 15 loci in liver, muscle, and heart. The proportion of polymorphic loci () for P. maculatus, I. labrosus, and P. pirinampu was 13.33, 20, and 6.67%, respectively, and mean heterozigosity was 6, 8.3, and 4.3%. The genetic identity value (I) was 0.32 between P. maculatus and I. labrosus, 0.37 between P. maculatus and P. pirinampu, and 0.58 between I. labrosus and P. pirinampu. This value suggests that these two latter species are congeneric. However, morphological characteristics place these species in distinct genera.A variabilidade genética de 3 espécies da família Pimelodidae, Pimelodus maculatus, Iheringichthys labrosus e Pinirampus pirinampu, coletadas em um ponto do rio Tibagi, foi analisada comparativamente utilizando dados protéicos de 6 sistemas que codificam 15 locos em fígado, músculo e coração. A proporção de locos polimórficos () para P. maculatus, I. labrosus e P. pirinampu foi de 13,33, 20 e 6,67%, respectivamente, e a heterozigosidade média foi de 6, 8,3 e 4,3%. A identidade genética (I) foi de 0,32 entre P. maculatus e I. labrosus, 0,37 entre P. maculatus e P. pirinampu e 0,58 entre I. labrosus e P. pirinampu. O valor de I (0,58) encontrado entre I. labrosus e P. pirinampu sugere que estas são espécies congenéricas. No entanto, as características morfológicas colocam estas espécies em gêneros distintos. Os resultados obtidos nesse estudo podem ser úteis para um melhor conhecimento de espécies de Pimelodidae. Elas também reforçam a necessidade da preservação do rio Tibagi (Paraná - Brasil) através de uma análise cuidadosa no caso de construção de hidroelétricas.

In this study, random amplified polymorphic DNA (RAPD) markers were applied to analyze the genetic diversity of samples of the Neotropical catfish Hypostomus ancistroides, collected from four sites (S1, S2, S3 and S4) along an urban stream in Southern Brazil. The 11 primers used in RAPD analysis amplified 147 loci, 76 (51.7%) of which were polymorphic. The proportions of polymorphic loci observed in the four samples were: 29.93% (S1), 31.97% (S2), 23.81% (S3) and 38.77% (S4). The average heterozygosity within sampling localities ranged from 0.1230 to 0.1526 and unbiased genetic distances ranged from 0.0253 to 0.0445. AMOVA partitioned 90.85% of the total variation within samples and 9.15% among samples. Excepting for the sample pair S1-S2 (phiST = 0.02784; p > 0.05), all others pairwise phiST values were significantly greater than zero, indicating moderate genetic differentiation among catfish samples from four localities. The relative low levels of genetic variation detected in all groups studied could be related to different factors, including the sedentary habit of these fish, which can be eroding the genetic variation of H. ancistroides from each locality.

Um aumento de salmonelose humana causada pelo sorovar Enteritidis relacionado com o consumo de carne de aves, ovos e derivados foi observado em todo o mundo nos últimos anos. A implantação do Sistema de Análise de Perigos e Pontos Críticos de Controle (APPCC), embora tenha diminuído o número de aves contaminadas, não foi capaz de eliminar o problema. A salmonelose é a doença alimentar de origem bacteriana mais prevalente no Paraná e o sorovar Enteritidis foi responsável por aproximadamente 80% dos surtos ocorridos nesse Estado, entre 1999 e 2006. Como S. Enteritidis não provoca sintomas clínicos graves nas aves, o monitoramento da sua presença em alimentos é muito importante para o controle da doença humana. Os métodos convencionais de isolamento de Salmonella em alimentos, água ou amostras ambientais são muito demorados. Devido à rapidez, especificidade e sensibilidade, métodos moleculares, como a reação em cadeia da polimerase (PCR), têm sido padronizados para detecção e identificação de Salmonella em alimentos. O objetivo deste trabalho foi padronizar um ensaio PCR multiplex para a detecção de Salmonella spp. e identificação simultânea de Salmonella Enteritidis em carcaças de frango. A PCR foi padronizada utilizando-se os pares de iniciadores INV A, específico para Salmonella spp. e IE1, específico para o sorovar Enteritidis, resultando em fragmentos amplificados de DNA alvo específicos de 796 e 316 pares de base, respectivamente. A especificidade do ensaio PCR padronizado, testada com culturas puras de diferentes sorovares de Salmonella e outras espécies bacterianas, foi de 100%. Após 24 h de enriquecimento não seletivo foi possível detectar aproximadamente 1 unidade formadora de colônia (UFC) de Salmonella por 10 g de pele de frango artificialmente contaminada. Das 66 amostras de carcaça de frango analisadas, adquiridas no comércio de Londrina, nove (13,0%) estavam contaminadas com Salmonella spp. A presença dessa bactéria foi confirmada pelo método tradicional de cultura e identificados os sorovares Schwarzengrund e Montevideo, respectivamente, em seis e em três amostras. Através da PCR foram detectados fragmentos de DNA específicos que confirmaram a presença do gênero Salmonella, porém não do sorovar Enteritidis. A PCR multiplex padronizada apresentou sensibilidade e especificidade para detectar o gênero Salmonella e, simultanemante identificar o sorovar Enteritidis, em carne de frango, após 24 horas de análise. An increase in human salmonellosis caused by serovar Enteritidis related to consumption of poultry products was observed worldwide in the last years. Safety measures have been adopted to control this serovar. The implementation of Hazard Analysis and Critical Control Points (HACCP) System was not able to eliminate the problem although it has considerably reduced the number of infected chickens. Salmonellosis is the most frequent bacterial foodborne disease in Parana State, Brazil and serovar Enteritidis caused approximately 80% of outbreaks occurred between 1999 and 2006. S. Enteritidis causes subclinical infection in chickens. Thus, the constant checking of Salmonella contamination in the food chain is essential to control human disease. Salmonella detection in food, water or environmental samples by traditional methods is labor-intensive and time-consuming. Molecular methods, such as polymerase chain reaction (PCR), are rapid, specific and sensitive and several methods have been developed to detect and identify Salmonella in foods. The objective of this study was to develop a multiplex PCR for detection of Salmonella spp. and Salmonella Enteritidis in chicken carcasses. A PCR was developed using Salmonella genus-specific primers INVA and serovar Enteritidis-specific primers IE1, which produce specific DNA fragments of 796 and 316 base-pairs, respectively. The standardized PCR was tested with different isolates of Salmonella and other bacterial species and the specificity was 100%. Salmonella was detected from chicken skin artificially contaminated with approximately 1 colonyforming unit (CFU) of Salmonella per 10 g, after 24 h of non-selective enrichment. Salmonella spp. was detected in nine (13%) of 66 analyzed chicken carcasses purchased from local retail market. The presence of Salmonella was confirmed by traditional culture method and serovars Schwarzengrund and Montevideo were identified in six and three carcass samples, respectively. Specific DNA fragments of the PCR confirmed the presence of genus Salmonella, but not serovar Enteritidis. The standardized multiplex PCR was found to be sensitive and specific on the detection of genus Salmonella and simultaneous identification of serovar Enteritidis in chicken meat after 24 hours of analysis.

A conservação da diversidade genética é de fundamental importância para sobrevivência de uma espécie a longo tempo, pois a manutenção de um pool gênico com variabilidade suficiente faz-se necessário para a adaptação da espécie a alterações ambientais. Nesse contexto, marcadores moleculares RAPD e microssatélites, foram utilizados para avaliar a diversidade e estrutura genética da espécie migradora Leporinus friderici no Complexo Canoas – rio Paranapanema – Brasil. Sete grupos foram amostrados nas escadas para transposição de peixes das UHEs Canoas I e Canoas II durante o período reprodutivo em três anos consecutivos. Ambos os marcadores evidenciaram uma alta diversidade genética para esses grupos. Os marcadores microssatélites mostraram uma perda de heterozigosidade e uma baixa taxa de endocruzamento para a espécie. A diferenciação genética encontrada entre os grupos, com ambos os marcadores utilizados, pode ser considerada de baixa a moderada. Os dados obtidos com os parâmetros de diversidade genética (distância e identidade genética, FST e AMOVA) permitiram concluir que os grupos de L. friderici do Complexo Canoas estão estruturados como uma única população. The dams built along systems of water interrupt the dispersion and the migration of the aquatic organisms affecting mainly the abundance of the species of fish migraters. Mechanisms for transposition were built in dams seeking to minimize those impacts. Little are the available information on the effect of the construction of dams in the population genetic structure of the fauna Neotropical of fish migraters. The conservation of the genetic diversity is of fundamental importance for survival of a species at long time because the maintenance of a genic pool with enough variability is made necessary for the adaptation of the species to environmental alterations. In that context, molecular markers RAPD and microsatellites, were used to evaluate the diversity and structure genetics of the migraters species Leporinus friderici in the “Complexo Canoas” – Paranapanema river - Brazil. Seven groups were collected in the ladder for transposition of fish of HPS Canoas I and Canoas II during the reproductive period in three consecutive years. Both markers evidenced a high genetic diversity for those groups. The markers microsatellites showed a loss of heterozygosity and a considerable inbreeding tax for the species. The genetic differentiation found among the groups with both used markers can be considered low until moderate. The data obtained with the parameters of genetic diversity (distance and genetic identity, FST and AMOVA) allowed to conclude that the groups of L. friderici of the “Complexo Canoas” are structured as a single population. The data obtained on the genetic diversity and population structure of L. friderici are of great importance for the development of handling programs and conservation of the species in the “Complexo Canoas” and could also be used in aquaculture programs.

As barragens construídas ao longo de sistemas hídricos interrompem a dispersão e a migração dos organismos aquáticos, afetando principalmente a abundância das espécies de peixes migradores. Mecanismos para transposição foram construídos em barragens visando minimizar esses impactos. Poucas são as informações disponíveis sobre o efeito da construção de barragens na estrutura genética populacional da fauna Neotropical de peixes migradores. A conservação da diversidade genética é de fundamental importância para sobrevivência de uma espécie a longo tempo, pois a manutenção de um pool gênico com variabilidade suficiente faz-se necessário para a adaptação da espécie a alterações ambientais. Nesse contexto, marcadores moleculares RAPD e microssatélites, foram utilizados para avaliar a diversidade e estrutura genética da espécie migradora Leporinus elongatus no Complexo Canoas ? rio Paranapanema ? Brasil. Dez grupos foram amostrados nas escadas para transposição de peixes das UHEs Canoas I e Canoas II durante o período reprodutivo em três anos consecutivos. Ambos os marcadores evidenciaram uma alta diversidade genética para esses grupos. Os marcadores microssatélites mostraram uma perda de heterozigosidade e uma considerável taxa de endocruzamento para a espécie. A diferenciação genética encontrada entre os grupos, com ambos os marcadores utilizados, pode ser considerada de baixa a moderada. Gymnotus inaequilabiatus (Valenciennes, 1847) is distributed across the Paraná-Paraguai basin and in some coasts drainages of Uruguai and in southeast of Brazil. It inhabits either areas with grasses or floating macrophytes in small rivers and along the banks of larger dark water rivers. In Brazil, the common names of this species are; morenita, tuvira and sarapó, and it isn't a migratory species. From December 2005 until March 2006, 63 specimens were collected at upper Paraná River floodplain and 30 at Paraguay River. Ten enzyme systems (AAT: Aspartate aminotransferase, EST: Esterase, ADH: Alcohol dehydrogenase, GDH: Glucose dehydrogenase, SOD: superoxide dismutase, LDH: lactate dehydrogenase, G3PDH: glyceraldehyde 3-phosphate dehydrogenase, MDH: Malate dehydrogenase, SORB: Sorbitol dehydrogenase, IDH: Isocitrate dehydrogenase) were analyzed by starch gel electrophoresis technique. Seventeen loci were detected for each population. The population of upper Paraná River floodplain stream showed the biggest genetic variability either by proportion of polymorphic loci (76.47%), or by average number of alleles by locus (2,12 ± 0,86) and expected heterozygosity (0,2415 ± 0,2072). Some alleles were detected only in the upper Paraná River floodplain population: Adh-1(b), Adh-(c), Est-1(a), Est-2(a), Gdh-1(a), Gdh-1(c), Idh-1(b), Ldh-B(a), Mdh-A(c), Sod-1(b), Sod-2(b) e Sorb-1(b, c, d). The allele frequencies in the two populations are significantly different at 11 of the 17 loci. The Nei's genetic identity and genetic distance between the populations were I = 0,7889 and D = 0,2246, respectively. The data indicate these populations of the G. inaequilabiatus are diverging genetically and could be two distinct species in the future. A management program should inhibit the gene flux between them. Habita regiões com gramíneas e áreas com macrófitas flutuantes em pequenos rios e ao longo de margens de rios maiores de águas escuras. É popularmente conhecido como; morenita, tuvira ou sarapó, e não é uma espécie migradora. De dezembro de 2005 a março de 2006 foram coletados 63 espécimes na planície de inundação do alto rio Paraná e 30 no rio Paraguai. Foram analisados 10 sistemas enzimáticos (AAT: Aspartato amino Transferase, EST: Esterase, ADH: Alcool desidrogenase, GDH: Glicose desidrogenase, SOD: Superóxido dismutase, LDH: Lactato desidrogenase, G3PDH: Glicerol-3-fosfato desidrogenase, MDH: Malato desidrogenase, SORB: Sorbitol desidrogenase, IDH: Isocitrato desidrogenase) por meio da técnica de eletroforese em gel de amido. 17 loci foram detectados em cada população. A população da planície de inundação do alto rio Paraná foi a que apresentou a maior variabilidade genética, tanto pela freqüência de loci polimórficos (76,47%), quanto pelo número médio de alelos por locus (2,12 ± 0,86) e heterozigosidade esperada (0,2415 ± 0,2072). Foram detectados 14 alelos exclusivos para a população da planície de inundação do alto rio Paraná: Adh-1(b), Adh-(c), Est-1(a), Est-2(a), Gdh-1(a), Gdh-1(c), Idh-1(b), Ldh-B(a), Mdh-A(c), Sod-1(b), Sod-2(b) e Sorb-1(b, c, d). As duas populações diferem significativamente quanto às freqüências alélicas em 11 dos 17 loci. A identidade e a distância genética de Nei entre as populações da planície de inundação do alto rio Paraná e do rio Paraguai foram I = 0,7889 e D = 0,2246, respectivamente. Os dados indicam que as populações de G. inaequilabiatus da planície de inundação do alto rio Paraná e do rio Paraguai estão em processo de divergência genética e podem futuramente se tornar espécies distintas. Um projeto de manejo desta espécie deveria evitar o fluxo gênico entre elas. 19 f

A fragmentação dos regimes hídricos naturais, causada por interferência antrópica, é provavelmente o fator principal na redução de espécies migradoras neotropicais, uma vez que pode dificultar a estratégia reprodutiva, causar perda da variabilidade genética e redução dos estoques nativos. Em uma tentativa de minimizar o impacto do represamento para espécies migradoras de peixes, escadas de transposição são construídas ao lado das barragens hidroelétricas para permitir a ascensão dos peixes migradores à porção montante. Tendo em vista a falta de informações sobre os efeitos destas escadas de transposição sobre a diversidade genética em peixes, tem-se como objetivo deste trabalho contribuir com o conhecimento da eficiência das escadas de transposição construídas no Complexo Canoas ? rio Paranapanema (Brasil), através de análises genéticas da estrutura populacional da espécie Prochilodus lineatus utilizando as técnicas de RAPD e microssatélites. Os exemplares de P. lineatus foram divididos em grupos, conforme sua data de coleta: 4 de Canoas I e 4 de Canoas II. As análises genéticas por meio do marcador RAPD, indicaram uma variabilidade genética alta (.P ? 70%) e uma baixa diferenciação genética entre os grupos de P. lineatus de cada escada de transposição (FST = 0,0324). As análises genéticas realizadas com microssatélite de maneira comparativa entre os agrupamentos CI e CII corroboraram com as análises de RAPD exibindo um baixo valor de diferenciação genética entre os agrupamentos (FIT = 0,2592; FIS = 0,2549; FST = 0,059). Assim, os diversos parâmetros de diversidade genética utilizados, tanto nas análises com RAPD quanto com microssatélite evidenciaram a existência de uma única população de P. lineatus no Complexo Canoas proveniente do reservatório de Capivara (jusante de Canoas I) que é provavelmente mantida pelos principais afluentes deste reservatório, os rios Tibagi e das Cinzas. Diante de dados que indicam que após a transposição não há conclusão do ciclo reprodutivo, a presença das escadas de transposição de peixes no Complexo Canoas pode ser considerada inadequada e a longo prazo, comprometer a sobrevivência de P. lineatus. Assim, como medidas de manejo, torna-se indispensável a preservação desses tributários na manutenção dos estoques naturais dessa espécie, assim como o fechamento das escadas de transposição durante o período reprodutivo dessa espécie. The aim of this work was to contribute to our understanding of the effects of fish passage ladders on the genetic diversity of fish by conducting genetic study of the population structure of the species Prochilodus lineatus collected in the passage ladders of Canoas I and Canoas II - Paranapanema River (Brazil) utilizing the RAPD and microsatellite techniques. Analyses with both markers indicated a high genetic variability (`P » 70% and `Ho » 67%) and low genetic differentiation (FST =0.0324 and FST = 0.0017, for RAPD and microsatellite, respectively) among the groups of P. lineatus collected at each passage ladder. The various parameters of genetic diversity determined indicated the existence of a single population of P. lineatus in the Canoas Complex originating from the Capivara Reservoir (downstream from Canoas I), probably fed by the Tibagi and Cinzas Rivers. In view of the findings that indicated that after passage there is no completion of the reproductive cycle, the presence of the fish passage ladders in the Canoas Complex can be considered impacting, where they cause a depletion in the recruitment downstream, which in the long term can compromise the natural population of P. lineatus in the middle of the Paranapanema River.

Overfishing, pollution and construction of dams have impacted migratory fishes of Paraná River basin. Prochilodus lineatus is an economically important endemic migratory species from Platina basin. Decrease in P. lineatus landings have been observed as the need of management and conservation. This study aimed to investigate the genetic variability of this species through Random Amplified Polymorphic DNA (RAPD) and mtDNA sequencing techniques. The analyzed individuals were collected in the Paraná, Bahia, Corumbá and Miranda Rivers. A total of 1815 base pairs from 12 individuals were sequenced to seven PCRAmplified fragments representing five protein-coding mitochondrial genes. Estimates of percent nucleotide sequence divergence ranged from 0.00 to 0.95 and any geographic pattern to mtDNA haplotype distribution was observed. The RAPD study included 86 markers and 58 individuals and revealed a high level of polymorphism. Fisher?s exact test was computed to verify whether the genetic differences were significant for the total data and between geographically proximate population samples. The results suggest that Prochilodus lineatus is not genetically subdivided, although significant divergence in frequencies of some RAPD fragments was found. This information may have useful for stocking and management programs plannings of P. lineatus. Prochilodus lineatus é uma espécie economicamente importante e endêmica na Bacia Platina. Um decréscimo na quantidade desembarcada desta espécie tem sido observado assim como a necessidade de seu manejo e conservação. Este estudo teve como objetivo investigar a variabilidade genética desta espécie através das técnicas de RAPD e sequenciamento de DNA mitocondrial. Os indivíduos analisados foram coletados nos rios Paraná, Baía, Corumbá e Miranda. Um total de 1815 pares de bases de doze indivíduos foi sequenciado para sete fragmentos de genes mitocondriais codificadores de proteína. As estimativas de porcentagem de divergência na sequência de nucleotídeos variaram de 0,00 a 0,95 e nenhum padrão de distribuição geográfica dos haplótipos de DNA foi observado. O estudo com RAPD incluiu 86 fragmentos de 58 indivíduos e revelou um alto polimorfismo. O teste exato de Fisher para verificar se as divergências genéticas eram significativas foi aplicado para os dados totais e entre populações geograficamente mais próximas. Os resultados obtidos sugerem que Prochilodus lineatus não está subdividida, embora tenham sido detectadas divergências significativas nas frequências de alguns locos.
