4. Evaluating impacts of environmental stress and bioactive chemicals on the North Carolina blue crab population: An individual‐based model.

Abstract

Objective: Recent estimates of the North Carolina blue crab Callinectes sapidus stock found that the stock is overfished and overfishing is occurring. Threats outlined in the 2018 stock assessment include climate change and estrogenic endocrine‐disrupting chemicals (EEDCs). The objective of this study was to use an individual‐based modeling approach to simulate the long‐term effects of climate change and EEDCs on the North Carolina blue crab stock. Methods: To do this, we built an individual‐based model that simulated the life history of individual blue crabs, including but not limited to growth, reproduction, and mortality. We then tested our population of blue crabs against 30 different combinations of temperature and EEDC scenarios over 50 years to determine the long‐term effects on the population. Result: Our simulations suggested that the North Carolina blue crab population may be relatively resilient to climate change‐related temperature shifts but that there may be significant impacts at the population level as summer temperatures become more extreme. Endocrine‐disrupting chemical effects resulted in an alternative stable state of lower catch or the total extinction of the population. Conclusion: These results suggest that management strategy changes may be necessary as temperatures become more extreme in the region. In addition, more research is necessary to fully understand the effects of EEDCs on blue crabs and other crustaceans at the individual and population level. Impact statementThis study used a novel approach to connect fine‐scale blue crab ecology to broadscale population ecology. This model can be a useful predictive tool that allows for assessment of the effects of future threats or regulation changes and can be modified to simulate other important crustacean species. Abstract

Modeling population dynamics and establishing a comprehensive population assessment for fishery species that are difficult to age have been challenging. Determination of age for such species is still an unresolved issue or is at best uncertain. Catch-survey analysis does not require full age information but can still provide a comprehensive population assessment. It was extended to incorporate multiple surveys and multiple sources of uncertainties within the statistical catch-at-age framework in the applications to crustaceans. Here, we further generalize and extend the multiple survey catch-survey analysis into a hierarchical Bayesian two-stage model by applying the hierarchical Bayesian approach. The hierarchical Bayesian approach can sufficiently incorporate uncertainty and expert opinions in parameter estimation. We developed a series of models with different assumptions for natural mortality and catchability, including nonstationary (i.e., time-varying) assumptions. We evaluated model robustness to these assumptions and compared population dynamics estimates and population status determination. We demonstrated the application of the hierarchical Bayesian two-stage model using the North Carolina blue crab (Callinectes sapidus) example. In this example, estimation of population size and fishing mortality and determination of population status were robust to the natural mortality and catchability assumptions. The North Carolina blue crab population is less likely to have nonstationary catchability or nonstationary natural mortality. Its natural mortality is more likely to vary by stage than by sex or over time. La modelisation de la dynamique des populations et la realisation devaluations exhaustives des populations pour des especes de poissons commerciaux pour lesquelles la determination de l'age est difficile constituent un defi. La determination de l'age pour de telles especes demeure une question non resolue ou au mieux incertaine. Si l'analyse de releves de prises ne necessite pas d'information exhaustive sur l'age, elle peut neanmoins fournir une evaluation exhaustive de la population. Elle a ete elargie afin d'y incorporer des releves multiples et des sources d'incertitude multiples dans le cadre statistique des prises par age dans les applications aux crustaces. Nous generalisons et elargissons encore davantage l'analyse de releves de prises a releves multiples dans un modele bayesien hierarchique a deux etapes en appliquant l'approche bayesienne hierarchique. Cette derniere permet une integration suffisante de l'incertitude et des opinions d'experts dans l'estimation de parametres. Nous developpons une serie de modeles reposant sur differentes hypotheses concernant la mortalite naturelle et la capturabilite, y compris des hypotheses non stationnaires (c.-a-d. variables dans le temps). Nous evaluons la robustesse des modeles par rapport a ces hypotheses et en comparons les estimations de la dynamique de la population et la determination du statut de la population. Nous faisons la demonstration du modele bayesien hierarchique a deux etapes en utilisant l'exemple du crabe bleu (Callinectes sapidus) en Caroline du Nord. Dans cet exemple, l'estimation de la taille de la population et de la mortalite par peche et la determination du statut de la population sont robustes pour les differentes hypotheses concernant la mortalite naturelle et la capturabilite. 15. Sarcinasp. as a presumptive cause of fatal acute gastric dilation and gastric emphysema in rhesus macaques

Abstract

A 4-y-old female and 3-y-old male rhesus macaque (Macaca mulatta), both housed in the same facility, died unexpectedly within 2 wk. Postmortem examination revealed severe gastric dilation in both macaques and gastric emphysema in the female macaque. Histologically, bacteria consistent with Sarcinasp. were present in both macaques within the lungs and lumen of the trachea, esophagus, and gastrointestinal (GI) tract without associated inflammation. Additionally, in the female macaque, the bacteria were found in the gastric mucosa and associated with emphysematous spaces in the gastric wall without associated inflammation. PCR and Sanger sequencing of amplicons were subsequently performed on GI contents and non-alimentary tissues from the 2 affected monkeys and on comparative samples from unaffected rhesus monkeys in the same facility and an adjacent primate facility. The cases were compared using the 2-tailed Fisher exact test (p-value at 95% confidence). PCR identified Sarcinain GI contents of both affected and unaffected monkeys (p= 0.6084) and in non-alimentary tissues of affected monkeys only (p= 0.0083). These results suggest that the presence of Sarcinasp. in non-alimentary tissues is associated with gastric distension, gas accumulation, and unexpected death in nonhuman primates.

Abstract

Meticulous study design, exacting scientific review, scrupulous data management, rigorous human subjects' protection, and effective recruitment strategies are essential components of all clinical research studies that must be in place prior to the conduct of the study. Equally important to the success of the study and to the well-being and protection of study participants is the assurance that safe clinical environments are provided for the care of research participants. This chapter describes approaches to managing patient care and clinical research quality, as well as the application of established health care-based clinical quality methods and tools to the complex processes of clinical research. Techniques for identifying clinical risks, tools for collecting data about the quality of clinical care processes, strategies for mitigating risk and patient harm, and techniques for measuring the participants' perceptions of the research process are discussed in detail.

38. Intra-abdominal nocardiosis and scedosporiosis in a dog: case report and literature review

Abstract

A 2-y-old, intact female, mixed-breed dog was presented to the veterinary hospital with abdominal distension, anemia, and lethargy following a chronic history of nonspecific gastrointestinal signs. CBC and serum biochemistry revealed moderate nonregenerative anemia with neutrophilia, hypoalbuminemia, hyperglobulinemia, hypoglycemia, decreased urea and creatinine, and hypercholesterolemia. Abdominal radiographs and ultrasound revealed a large heterogeneous mesenteric mass and ascites. Abdominocentesis confirmed septic peritonitis with filamentous bacteria. Fine-needle aspiration of the mass yielded pyogranulomatous inflammation and hyphae. An exploratory laparotomy revealed a large cranial abdominal mass with granulomas present throughout the abdominal cavity. Due to the poor prognosis and disseminated disease, the owner elected euthanasia. Postmortem and histologic examinations detected intralesional mycetomas and bacterial colonies within the mesenteric masses. 16S ribosomal RNA gene PCR and sequencing using formalin-fixed, paraffin-embedded sections identified Nocardia yamanashiensis, Nocardioides cavernae, and Nocardioides zeicaulis. Fungal culture, PCR, and sequencing confirmed Scedosporium apiospermum. Our report highlights the importance of molecular methods in conjunction with culture and histologic findings for diagnosing coinfections caused by infrequent etiologic agents. Additionally, we provide a comprehensive literature review of Scedosporium apiospermuminfections in dogs.

Abstract

Introduction: Participants' perceptions of their research experiences provide valuable measures of ethical treatment, yet no validated instruments exist to measure these experiences. We conducted focus groups of research participants and professionals as the initial step in developing a validated instrument. Methods: Research participants enrolled in 12 focus groups, consisting of: (1) individuals with disorders undergoing interventions; (2) in natural history studies; or (3) healthy volunteers. Research professionals participated in six separate groups of: (1) institutional review board members, ethicists, and Research Subject Advocates; (2) research nurses/coordinators; or (3) investigators. Focus groups used standard methodologies. Results: Eighty-five participants and 29 professionals enrolled at eight academic centers. Altruism and personal relevance of the research were commonly identified motivators; financial compensation was less commonly mentioned. Participants were satisfied with informed consent processes but disappointed if not provided test results, or study outcomes. Positive relationships with research teams were valued highly. Research professionals were concerned about risks, undue influence, and informed consent. Conclusions: Participants join studies for varied, complex reasons, notably altruism and personal relevance. They value staff relationships, health gains, new knowledge, and compensation, and expect professionalism and good organization. Abstract

Despite intensive study in humans, responses to dying and death have been a neglected area of research in other social mammals, including nonhuman primates. Two recent reports [Anderson JR, Gillies A, Lock LC. 2010. Pan thanatology. Current Biology 20:R349-R351; Biro D, Humle T, Koops K, Souse C, Hayashi M, Matsuzawa T. 2010. Chimpanzee mothers at Bossou, Guinea carry the mummified remains of their dead infants. Current Biology 20:R351-R352] offered exciting new insights into behavior toward dying and dead conspecifics in our closest living relatives-chimpanzees. Here, we provide a comparative perspective on primate thanatology using observations from a more distant human relative-gelada monkeys ( Theropithecus gelada)-and discuss how gelada reactions to dead and dying groupmates differ from those recently reported for chimpanzees. Over a 3.75-year study period, we observed 14 female geladas at Guassa, Ethiopia carrying dead infants from 1 hr to ≥48 days after death. Dead infants were carried by their mothers, other females in their group, and even by females belonging to other groups. Like other primate populations in which extended (>10 days) infant carrying after death has been reported, geladas at Guassa experience an extreme climate for primates, creating conditions which may favor slower rates of decomposition of dead individuals. We also witnessed the events leading up to the deaths of two individuals and the responses by groupmates to these dying individuals. Our results suggest that while chimpanzee mothers are not unique among primates in carrying their dead infants for long periods, seemingly 'compassionate' caretaking behavior toward dying groupmates may be unique to chimpanzees among nonhuman primates (though it remains unknown whether such 'compassionate' behavior occurs outside captivity).
