3. Getting under the skin-whitening cultures : discourses, rhetoric and representations across text types and media in Taiwan in the early 21st century

Abstract

This thesis concerns the ways in which the importance of practicing skin whitening is promoted and represented in newspaper advertorial articles and visual advertisements across different media in contemporary Taiwan, approximately between 2008 and 2012. Theoretically, it is inspired by structuralists such as Foucault in the discursive formation of discourse and knowledge legitimation, Bourdieu in classification of cultural consumptions, Barthes in the process of mythologisation, sociocultural theory in everyday practice and identity construction, and linguistic theories both from CDA and semiotics. Methodologically, I undertake qualitatively textual and visual analyses of articles from the Liberty Times newspaper, and graphic and video advertisements from product leaflets, television and the Internet. In terms of analytical approaches, CDA, semiotics and other critical visual analysing methods are applied to interpret and articulate dominant discourses, rhetorical tropes and representations of idealised female images with whitened skin. Through the exploratory investigation of the data, common themes such as scientific endorsed and legitimated discourses, an extended metaphor of fighting and the ability of whitening ingredients to penetrate/invade the skin are found in both textual and visual materials. In terms of results, the research echoes Foucault’s idea of interdiscursivity and suggests that these discourses are transferable and represented across both textual and visual media. Finally, a critical assessment is made on the extraction, appropriation and distribution of knowledge by cosmetics producers and the media to consumers; the hegemonic power of the mutual co-alignment between cosmeceutical industry, mass media, retailing channels, legislation, and advertising and research agencies intersected by ‘medical’ and ‘scientific’ qualities; and the implications behind the phenomenon of promoting skin whitening.

4. Fighting whom? A critical approach to marketing communication of skin whitening product in Taiwan.

Abstract

This paper investigates the discursive formation of non-multimodal and multimodal text for promoting skin whitening products from 2006 to 2020. Text types include advertorial articles, product naming, graphic adverts, and commercial films. It is found that the fighting metaphor treating melanin or blackness as a target to expel, suppress, or eliminate is predominant in both written and visual texts. Throughout the analysis, the results suggest that the skin-whitening discourses intersect and promote the unequal power relations of the macro-structures of gender, class, and race. Following the findings, concerns about the application of fighting metaphor and rhetoric of skin whitening products in globalized society are discussed. [ABSTRACT FROM AUTHOR]

Abstract

Background: Maize is an important crop that has a complex genome. A better understanding of maize chromatin architecture provides great opportunities for crop improvement, because chromatin accessibility influences gene expression, thereby affecting agricultural traits. The newly developed method for chromatin profiling, Assay for Transposase Accessible Chromatin with high-throughput sequencing (ATAC-seq), has been developed to investigate chromatin accessibility. Result: We adapt this method by testing parameters of several key steps and generate the first ATAC-seq protocol for maize. We demonstrate that purification of maize nuclei to eliminate organelles can be achieved without the need for cell sorting, and that only a standard bench-top centrifuge is required for sample preparation. Finally, our sequence analyses confirm that our protocol of ATAC-seq can be successfully used to assess the chromatin landscape in maize. Conclusion: The ATAC-seq provides a useful technique to study the chromatin accessibility. Given the parameters tested in our study, it can be a simple and practical method for maize and may be a foundation for similar studies in other crop species.

Abstract

2,5,7,8-Tetramethyl-2-(2'-carboxyethyl)-6-hydroxychroman (alpha-CEHC), the water-soluble metabolite of alpha-tocopherol (alpha-TOH) with a shortened side chain but an intact hydroxychroman structure, has been identified in human urine and are thought to be produced in significant amount at excess intake of alpha-TOH. In previous studies, CEHCs in biological specimens were measured by HPLC, GC-MS or LC-MS, preceded by a hydrolysis procedure using either enzyme or methanolic HCl. In an attempt to analyze alpha-CEHC in rat urine accordingly, we observed that enzyme hydrolysis was relatively inefficient in releasing alpha-CEHC compared to high concentrations of HCl. The HCl releasable alpha-CEHC conjugate was isolated and chemically identified as 6-O-sulfated alpha-CEHC (alpha-CEHC sulfate). Using the synthetic alpha-CEHC sulfate standard, it was found that sulfatase could not hydrolyze to a significant extent. On the other hand, pretreatment with HCl at 60 degrees C in the presence of ascorbate, followed by a one-step ether extraction, not only hydrolyzed the sulfate conjugate completely but also extracted alpha-CEHC with high recovery. The inclusion of ascorbate minimized the conversion of alpha-CEHC to alpha-tocopheronolactone in the HCl pretreatment. A complete procedure for the quantitative analysis of alpha-CEHC including HCl hydrolysis, ether extraction and reverse phase isocratic HPLC-ECD was thus established. In conclusion, alpha-CEHC sulfate was isolated and identified as the HCl-releasable conjugate of alpha-CEHC in rat urine. A rapid and sensitive method with high reproducibility for the determination of free, conjugated and total alpha-CEHC is then established.
