The objective of this article is to address the challenges of physical sample retrieval from all locations along patients' gastrointestinal (GI) tracts for studying of interactions between GI microbial communities (microbiomes) and host immune systems. We propose a novel tetherless, blindly controlled, magnetically actuated capsule for noninvasive sampling of GI microbiomes and liquid digesta. The mesoscale capsule, a soft mobile robot platform, is made from two permanent magnets encapsulated in a soft elastomer and actuated remotely by an external magnet. The capsule is controlled blindly using an external magnet to orient and activate the capsule. Following oral administration, capsules transit the GI tract and are activated at specified locations. The capsule is recovered via routine stool passage and collected samples are processed for downstream analyses (e.g., 16S rRNA surveys, metagenomics, metatranscriptomics and/or metabolomics). The localization of the capsule is estimated using the traveling time and the location of collected microbiomes will be confirmed by downstream analyses. A mathematical model of capsule activation is used to predict capsule activation and guide design optimization. We demonstrate the ability of the capsule to collect samples, as well as maintain sample integrity in vitro, ex vivo , and with twelve capsules in in vivo swine models. Eleven capsules successfully collected digesta (in the range of 18 to 61 mg). This sampling apparatus offers a technological advance for the robust sampling of GI tract contents.

Eighty-eight gilts [initial body weight (BW) 49.8 ± 0.8 kg] were recruited to determine the effects of moderate energy and protein restriction during the development period on changes in BW and backfat depth (BF) and subsequent lactation performance. Gilts were randomly assigned to one of four feeding programs: 1) standard commercial diet fed ad libitum (CON), 2) standard commercial diet fed 10% or 3) 20% below ad libitum, or 4) a high–fiber diet fed ad libitum [2.5 times more fiber (neutral detergent fiber) than the commercial diet to dilute net energy and crude protein by approximately 20% and 13%, respectively; FIB]. The gilts were housed individually and received the feeding programs between 90 and 190 (breeding) d of age and standard gestation and lactation diets thereafter. Litters were standardized to 12 ± 1 pigs within 48 h of farrowing; weaning occurred at 20.0 ± 0.4 d of age. Gilts that received the 20% restricted program had lower overall average daily feed intake (ADFI) during the development period (2.64 ± 0.04 kg; P < 0.05) versus all other feeding programs and gilts that received the 10% restricted program had lower ADFI than FIB and CON, which were not different (2.96 vs. 3.44 and 3.47 ± 0.04 kg for 10%, FIB and CON, respectively; P < 0.05). Plasma free fatty acid (FFA) concentrations on day 180 of age were lower for gilts that received the 10% and 20% restricted programs compared to gilts that received the FIB and CON programs, which were not different (97 and 86 vs. 220 and 149 ± 29 µEq/L, respectively; P < 0.05). Plasma concentrations of glucose, urea, prolactin, and IGF-1 were not different among feeding programs on day 180 of age. At breeding, gilts that received the FIB and 10% programs had lower BW and BF versus CON (145.7 and 144.8 vs. 155.2 ± 0.9 kg and 14.4 and 14.8 vs. 16.5 ± 0.2 mm for BW and BF, respectively; P < 0.05) but greater BW than gilts that received the 20% restricted program (137.9 kg; P < 0.05). The BW and BF of gilts did not differ at the end of gestation or at weaning. The ADFI of sows during lactation and offspring birth weight and growth rate during lactation and the 5-wk nursery period were not influenced by gilt development feeding program. Therefore, a high-fiber feeding program could be used in group-housing gilt-development scenarios, where feed is offered ad libitum, to control BW and BF prior to breeding without influencing milk production in the subsequent lactation period.

The physiochemical properties and digestible, metabolizable, and predicted net energy contents in high-protein dried distillers’ grain (HiPro) were determined to assess the nutritive value for growing pigs. Twelve Yorkshire × Landrace barrows (initial body weight 25 ± 0.5 kg) were used in a partially replicated Latin square design over three periods (n = 7 or 8) and assigned to one of five experimental diets. In each period, pigs were adapted to diets for 7 d, followed by 5 d of total urine collection and fecal grab sampling. The experimental diets included a corn- and soybean-meal-based diet (CON) or diets containing dried distillers’ grains with solubles (DDGS) or HiPro to partially replace corn and soybean meal, without or with (i.e., DDGS+ and Hipro+) a multi-carbohydrase enzyme blend (0.05% inclusion). The HiPro ingredient contained half as much starch (2.6% vs. 5.2%; DM-basis), 20% more protein (32.5% vs. 27.1%), and had 14% greater water binding capacity versus DDGS. Standardized ileal digestibility (SID, %) of crude protein (CP) and amino acid (AA) and the metabolic availability (MA) of Met using the indicator amino acid oxidation (IAAO) method, in partially defatted black soldier fly larvae (PD-BSFL) meal were determined in growing pigs in 2 experiments. The Met SID value was then compared numerically with the Met MA to understand how different SID is compared with its MA value. In Exp. 1, 6 ileal-cannulated barrows (initial body weight [BW] = 18.03 ± 0.34 kg) were used in a 2-period switch back design and fed either a nitrogen-free diet (NFD) or test diet, with PD-BSFL meal as the sole source of AA, over two 11-d experimental periods, at a feeding level of 2.8 × estimated maintenance digestible energy requirement. Barrows were adapted for 9-d to the diet, followed by continuous 8-h ileal digesta collection on day 10 and 11. Digesta were pooled per pig within period. The SID of CP and Met of PD-BSFL meal were 76.1 ± 6.2% and 90.4 ± 3.9%, respectively. In Exp. 2, 7 barrows (initial BW = 18.77 ± 0.69 kg) were used in a 7 × 7 Latin square design with L-[1-13C]-Phe as the indicator AA. Each pig was randomly assigned to 1 of 7 dietary treatments over seven 3-d experimental periods. Two diet types were studied including reference (crystalline AA) and PD-BSFL test diets, each supplying graded intakes of Met at 55, 65, and 75% of the estimated SID requirement (NRC, 2012). The MA of Met was determined by comparing the IAAO response between the reference and PD-BSFL test diet using the slope-ratio method. Linear regression determined a negative slope of the best fit line for both the reference and test diets (P < 0.05). The MA of Met in PD-BSFL meal was 53.3%, which is as expected lower than the SID value. While it is generally appreciated that MA will be less than SID, the use of SID is more practical. In cases where SID cannot explain physiological outcomes of feeding a novel ingredient, IAAO may provide additional insight into whether MA should be explored.

Six ileal-cannulated barrows (28.0 ± 1.3 kg initial BW) were used in a replicated 3 × 3 Latin square design with one additional period (n = 7 or 6) to determine standardized ileal digestible (SID) AA and digestible energy of two modified soy protein concentrates [MSPC1 and MSPC2] and soybean meal (SBM). Pigs were fed one of three cornstarch-based diets with either MSPC1 or MSPC2 or SBM as the sole source of AA at a rate of 2.8 times the estimated maintenance energy requirement. In each period, pigs were adapted to diets for 7 d followed by 2 d of fecal collection and subsequently, 2 d of continuous ileal digesta collection for 8 h. The SID of AA was calculated using basal endogenous losses from a previous study for pigs fed a nitrogen-free diet. The digestible energy of the ingredients was calculated according to the difference method using a nitrogen-free diet that contained the same cornstarch:sucrose:oil ratio as the three test diets. The total Lys content was 33% and 38% greater for MSPC1 vs. MSPC2 and SBM, respectively. The SID of crude protein was greater for MSPC1 (96.9%) than for SBM (91.3%; P < 0.05), whereas an intermediate value was observed for MSPC2 (94.3% ± 1.2%). The SID of Ile (93.8%), Leu (93.6%), Lys (93.9%), Phe (96.7%), and Val (93.2%) were not different between MSPC1 and MSPC2 but greater than for SBM (88.8% ± 1.3%, 87.8% ± 1.2%, 84.5% ± 1.7%, 92.9% ± 1.0%, 86.5% ± 1.7% for Ile, Leu, Lys, Phe, and Val, respectively; P < 0.05). The SID of His and Thr was greater for MSPC1 than MSPC2 and SBM (P < 0.05), which were not different. The SID of Met was greater for MSPC1 and SBM vs. MSPC2 (P < 0.05). The SID of Arg was greater for MSPC1 than MSPC2 and SBM (P < 0.05), and greater for MSPC2 than SBM (P < 0.05). The digestible energy was greater for MSPC1 (4,677 kcal/kg) than MSPC2 and SBM (average; 3,896 ± 239 kcal/kg; P < 0.05), which were not different. Therefore, the MSPC1 was a better source of SID Lys and digestible energy than either MSPC2 or SBM and could be used as a highly digestible protein ingredient in swine rations.

Two hundred and forty weaned pigs [initial body weight (BW) 7.2 ± 0.07 kg] were allocated to three diets (eight pens per treatment, 10 pigs per pen) to determine the effects of a temporary lysine (Lys) restriction on subsequent growth, body composition, as well as carcass and loin quality at slaughter. For a 3 wk restriction period, pigs were fed diets that were 110% (control), 20% (Lys20), or 40% (Lys40) below estimated Lys requirements. Thereafter, all pigs were fed a common grower diet containing 120% of the estimated Lys requirement for 6 wk (recovery period) and commercial diets until slaughter at ∼125 kg BW. Fifty-six litters from first-parity sows standardized to 12 piglets were used to determine the effects of creep feed composition and form and the provision of low- or high-complexity nursery diets on the evolution of small intestinal histomorphology and jejunal mucosa-specific enzyme activities postweaning. At 5 d of age, litters (initial bodyweight [BW] 2.31 ± 0.61 kg) were assigned to one of four creep feeding regimens (n = 14): 1) commercial creep feed (COM), 2) liquid milk replacer (LMR), 3) pelleted milk replacer (PMR), or 4) no creep feed (NO). At weaning (21 d of age), six pigs per litter were provided a HIGH- (contained highly digestible animal proteins) or LOW- (contained corn and soybean meal as main protein sources) complexity nursery diet (n = 7). At 21, 28, and 59 d of age, two pigs per pen (one castrated male and one female) were euthanized, and ileal and jejunal segments for histomorphological measurements and jejunal mucosal scrapings were collected to determine specific mucosa enzyme activities. At weaning, pigs provided COM had a greater ileal absorptive capacity (M) than LMR or NO, which were not different (14.1 vs. 10.4 and 10.5 ± 0.9 μm2; P0.05); PMR was intermediate. On days 28 and 59, M was not different among pigs regardless of creep feed treatments. Pigs fed LOW had reduced jejunal villus height (VH; P0.001) and M (P0.001) on day 28 vs. day 21. The VH and M were not different for pigs fed HIGH or LOW by the end of the nursery period. For all dietary treatments except COM-HIGH and COM-LOW, jejunal mucosal maltase-specific activity was not different between days 21 and 28 of age but greater on day 59 (P0.05). For pigs that received COM-HIGH, maltase-specific activity was not different between days 21 and 28 but greater on day 59 than day 28 (P0.05). For pigs that received COM-LOW, maltase-specific activity was not different between days 21, 28, and 59. Regardless of creep or nursery treatment, sucrase-specific activity was the greatest on day 59, followed by days 21 and 28 (P0.001), and lactase-specific activity was greater on day 21 than on days 28 and 59 (P0.001), which were not different. Therefore, pigs that provided LOW diet had greater villus atrophy and reduced M during the first week after weaning vs. pigs that provided HIGH, regardless of creep feeding regimen, but were able to recover by the end of the nursery period.After weaning, the piglet digestive tract must adapt in order to effectively break down and absorb nutrients derived from plant-based ingredients, contributing to the postweaning growth lag. Feeding strategies applied both preweaning and postweaning with the intent to assist in intestinal adaptation have been developed; however, little work has been done examining the interaction between creep and nursery feeding strategies on intestinal histomorphology and jejunal mucosa-specific enzyme activities after weaning. In the current study, piglets that provided creep feed with higher starch content had greater jejunal mucosa maltase-specific enzyme activity and ileal absorptive capacity at weaning. However, these parameters were still negatively affected by weaning, regardless of nursery feeding strategy. Additionally, nursery diets with greater inclusion of soybean meal to replace animal protein sources (low complexity) exacerbated reductions in jejunal absorptive capacity. Conversely, by the end of the nursery period, there were no differences in intestinal histomorphology and jejunal mucosa-specific enzyme activities, regardless of creep or nursery feeding program. Therefore, exposing piglets to starch during creep feeding and reducing the inclusion of soybean meal in nursery diets improved intestinal morphology and mucosa disaccharidase activity directly following weaning, but benefits did not extend to the end of the nursery period.

Therefore, the MSPC1 was a better source of SID AA and digestible energy than either MSPC2 or SBM and could be used as a high-quality protein ingredient in swine rations.

Three hundred twenty newly weaned pigs (21 days of age; 6.7 ± 0.3 kg BW) were used to determine the effects of supplementing low-complexity (LC) deoxynivalenol- (DON) contaminated nursery diets with a feed additive or fish oil on growth performance and immune response to an Escherichia coli lipopolysaccharide (LPS) challenge. Pens were randomly assigned to 1 of 5 dietary treatments (n = 8 pens per treatment): positive control (PC; contained multiple animal protein sources), or 1 of 4 LC diets (contained only plant-based protein sources) without (NC; negative control) or with ~ 3.5 ppm DON contamination, without (DON-) or with a feed additive containing a blend of immune-modulating components (DON+; 2 mg/kg, as-fed) or fish oil (DONω3; 2.5%, as-fed). Dietary treatments were fed during phases I and II (7 and 15 days, respectively) and a common phase III diet was fed for 20 days. On day 22, two pigs per pen were injected IM with 30 μg/kg BW LPS and 1 pig per pen with 1 mL saline. Rectal temperatures were recorded at 0, 1, 2, 3 h after injection. At 3 h, blood was collected for plasma cytokine analysis and small intestinal histomorphology was assessed. In phase I, pigs fed PC and NC did not differ for ADG, ADFI and G:F, but these outcomes were greater than for pigs fed DON+ and DONω (P < 0.05). In phase II, pigs fed NC had greater ADG and PC had greater ADFI but lower G:F than pigs fed DON- and DONω3 (P < 0.05). At the end of phase II, pigs fed DONω3 tended to have lower BW than PC and NC (P = 0.084 and 0.079, respectively). In phase III and overall, there were no differences among dietary treatments for ADG, ADFI, G:F, or final BW. The LPS injection increased rectal temperature and reduced jejunal and ileal villus height (versus saline; P < 0.05). Plasma interferon-γ concentration was only increased by LPS for pigs fed PC, NC, and DON+ compared to the saline-injected counterparts (P < 0.05). Regardless of LPS injection, jejunal villus height was greater for pigs fed DON+ than DONω3 (P < 0.05) and ileal villus height was greater for pigs fed DON+ and PC than DONω3 (P < 0.05). Therefore, nursery diet complexity did not affect growth performance or immune response to LPS. Regardless of DON contamination and feed additive inclusion in phases I and II, pigs were able to achieve nursery exit BW not different from those fed PC. The feed additive offered marginal benefits for small intestinal villus height and immune response for pigs fed DON-contaminated LC nursery diets.

A total of 70 gestating gilts (166±13 kg BW) were used to determine whole-body N retention when standardized ileal digestible (SID) methionine (Met) was fed at one of seven levels (n=10), ranging between 50 and 150% of estimated requirements during late gestation. The experimental diets provided excess Cys and 2.61, 3.39, 4.44, 5.22, 6.00, 7.05, and 7.83 g SID Met/d, respectively, with a feed allowance of 2.61 kg/d. Diets were fed to individual gilts for a 7-d adaptation period followed by a N balance period between gestation days 109 and 112. Contrast statements were used to determine linear and quadratic effects of the dietary inclusion level of Met. The average N intake was 70.8 g/d among treatments. Urinary N excretion decreased with increasing SID Met content, with no further change after 3.39 g/d SID Met (quadratic; P < 0.0001). Fecal N excretion decreased with increasing SID Met content, with no further change after 5.22 g/d SID Met (linear and quadratic; P < 0.0001 and P < 0.05, respectively). Whole-body N retention increased as SID Met content increased between 2.61 and 4.44 g/d SID Met, no differences were observed between 4.44 and 7.05 g/d SID Met, with intermediate N retention for gilts fed 7.83 g/d SID Met (linear and quadratic; P < 0.01 and P < 0.0001). Whole-body N retention efficiency (% of intake) was greatest (60.74%) at 7.05 g/d SID Met. Whole-body N retention (g/d) was optimized at 4.04 and 4.58 g/d SID Met based on linear and quadratic broken-line linear models, respectively, for gilts during late gestation. Therefore, for whole-body protein retention, the average SID Met requirements (4.31 g/d) appear to be less versus those recommended by the NRC in late gestation (5.22 g SID Met/day), which do not take into consideration the non-protein partitioning of dietary Met.

The goal of this project was to determine if standardized ileal digestible (SID) lysine provided at 40% above estimated requirements, with the concomitant increase in protein intake, from days 90 to 110 of gestation would stimulate mammary development in gilts. From day 90 of gestation, Yorkshire × Landrace gilts were fed 2.65 kg of either a conventional diet (CTL, control, n = 19) providing 18.6 g/d of SID Lys or a diet providing 26.0 g/d of SID Lys via additional soybean meal (HILYS, n = 19). Both diets were isoenergetic. Jugular blood samples obtained on days 90 and 110 of gestation were used to measure concentrations of insulin-like growth factor-1 (IGF-1), metabolites, and amino acids (AA). Gilts were necropsied on day 110 ± 1 of gestation to obtain mammary glands for compositional analyses, immunohistochemistry, and analysis of mRNA abundance for AA transporters and markers of cell proliferation and differentiation. The HILYS gilts gained more body weight (P0.01) during the experimental period compared with CTL gilts, and had greater fetal weights (1.29 vs. 1.21 ± 0.03 kg, P0.05). There was no difference in circulating IGF-1, glucose, or albumin (P0.10) between HILYS and CTL gilts on day 110 of gestation, whereas concentrations of urea and free fatty acids were greater (P0.01), and those of Trp and Ala were lower (P0.05), in HILYS than CTL gilts. The provision of lysine at 40% above estimated requirements increased total mammary parenchymal mass by 44%, as well as total parenchymal fat, protein, DNA, and RNA (P0.01). The mRNA abundance of ACACA was greater (P0.05) in HILYS than CTL gilts, while only the AA transporter SLC6A14 tended (P0.10) to be greater. Results demonstrate that providing dietary Lys above current National Research Council recommendations in late gestation increases mammary development in gilts. Results also indicate that Lys may have been limiting for protein retention. These data suggest that the use of a two-phase feeding strategy during gestation, whereby dietary Lys is increased from day 90, could benefit potential sow milk yield in the subsequent lactation.Results indicate that the current National Research Council recommendations for dietary lysine during late pregnancy in pigs, the period when most mammary gland development takes place, are underestimated. From days 90 to 110 of gestation, gilts were fed 2.65 kg of either a conventional diet providing 18.6 g/d of standardized ileal digestible (SID) lysine, or a diet providing 26.0 g/d of SID lysine via the inclusion of additional soybean meal. Diets were isoenergetic. Feeding 26.0 g/d of SID lysine increased the mass of mammary parenchymal tissue (where milk is synthesized) by 44%. Findings suggest that a greater mammary uptake of lysine in supplemented sows supported enhanced accretion of mammary parenchyma. Such information is most pertinent in the actual context where milk yield of hyperprolific sows must be maximized to sustain optimal growth of all their piglets. Furthermore, these data indicate that the use of a two-phase feeding strategy during gestation, whereby dietary lysine is increased from day 90, could benefit potential sow milk yield in the subsequent lactation.

Growth performance and physiological responses of nursery piglets when fed enzymatically treated yeast (HY40) and pharmacological ZnO alone or in combination were investigated. A total of 144 pigs (21 d old, BW 7.32 ± 0.55 kg) were placed in 36 pens (4 pigs/pen). Pigs were randomly assigned to one of four dietary treatments (n = 9): 1) control corn-wheat-soybean meal diet (control), 2) control + HY40 (HY40), 3) control + (ZnO) and 4) control + HY40 + ZnO (HY40+ZnO). Inclusion of HY40 and ZnO was 0.5% and 3,000 ppm in phase I (days 0 to 14), respectively, and halved in phase II (days 15 to 42). All diets contained 0.2% TiO2 for determination of apparent total tract digestibility (ATTD) of components. Body weight and feed disappearance were recorded weekly. One pig per pen was killed for organ weights, blood, and tissue samples on day 14. Except for phase II, when HY40 + ZnO pigs had greater average daily feed intake (P = 0.004) than all other treatments, there were no (P0.05) interactions between HY40 and ZnO on growth performance. Pigs fed HY40 or ZnO containing diets were heavier (P0.05) than pigs fed without by the end of the study. On day 14, pigs fed additives exhibited higher (P ≤ 0.009) ATTD of dry matter (DM) and gross energy (GE) than control pigs. On day 28, pigs fed control, HY40, and HY40 + ZnO had greater (P ≤ 0.022) ATTD of DM, crude protein, and GE than piglets fed ZnO only. Pigs fed HY40 + ZnO had lower ileal digesta Escherichia coli concentration (P0.05) than HY40 and control pigs. Ileal digesta of pigs fed ZnO diets had higher lactobacillus to E. coli ratio (1.44 vs. 1.20; P = 0.001), exhibited higher concentrations of acetic (P = 0.01) and butyric acid (P = 0.01) but lower lactic (P = 0.02) and total short chain fatty acids (P = 0.033) than pigs fed non-ZnO diets. Greater (P0.05) mRNA expression of nutrient transporters, tight junction proteins, and fecal excretion of zinc (Zn) was observed in ZnO pigs relative to non-ZnO pigs. Pigs fed HY40 diets had greater (P = 0.002) villus height to crypt depth ratio (VH:CD) than non-HY40 pigs. The concentration of plasma IgA was higher (P = 0.04) in HY40 + ZnO pigs relative to other pigs, whereas HY40 pigs showed higher (P0.001) jejunal IgA than non-HY40 pigs. Although the mode of action of HY40 and ZnO differed, the present study indicated that HY40 improved growth performance and jejunal function and immunity, making HY40 an effective alternative to pharmacological ZnO in nursery pigs feeding programs.Abrupt weaning of pig results in nutritional, environmental, and psychological stresses leading to reduced feed intake, little or no weight gain and in some instances diarrhea, morbidity, and mortality. The post-weaning growth performance lag is often managed by use of antimicrobial growth promoters (AGP), pharmacological levels of trace elements such as zinc oxide (ZnO), specialty feed ingredients, and additives. However, many jurisdictions have already banned, and others are considering banning/restricting use of AGP to abate burgeoning levels of antimicrobial resistance in pathogens of animal and human. The European Union confirmed banning pharmacological ZnO effective June 2022 because of environmental pollution. In this context, identifying alternative nutritional strategies for managing newly weaned pigs is of global concern. Yeast derivatives have been proposed to be beneficial in transitioning piglets. However, there is little available information on comparative efficacy of yeast derivatives and pharmacological ZnO. The current study demonstrated that enzymatically treated yeast (HY40) had positive effects on growth, intestinal histomorphology, nutrient digestibility, and immune indices in nursery pigs. Through differing modes of action, overall growth performance of HY40 piglets was comparable to that of pharmacological ZnO piglets.

A total of 601 pigs from 65 litters were used to determine the effects of closely meeting estimated daily Lys and energy requirements for sows during gestation for three consecutive parities on offspring postweaning growth performance and carcass and loin quality at slaughter. Sows were assigned a control (static diet composition; CON) or precision (individual daily blend of two diets to meet estimated Lys and energy requirements; PRE) feeding program between days 7 and 110 of gestation for three consecutive pregnancy cycles, starting with primiparous sows (parity 1: 12 CON and 12 PRE sows; parity 2: 8 CON and 13 PRE sows; parity 3: 8 CON and 12 PRE sows). At weaning (20 ± 2 d of age), up to 10 pigs per litter were randomly selected and placed in a pen (1 litter per pen). All pens received ad libitum access to commercial diets in six phases (four-phase nursery, grower, and finisher, respectively). Four pigs per pen were slaughtered at ~125 kg BW for evaluation of carcass characteristics and loin quality. The ADG and ADFI of offspring were not influenced by maternal feeding program in any parity during nursery phases I through III. During nursery phase IV, ADG and ADFI were greater for litters from PRE- vs. CON-fed sows (0.70 vs. 0.66 ± 0.03 and 1.15 vs. 1.08 ± 0.06 kg/d for ADG and ADFI, respectively; P < 0.05). The BW for litters from PRE- vs. CON-fed sows tended to be greater by day 66 of age (end of nursery period; 29.7 vs. 28.7 ± 1.1 kg; P = 0.076). Within the grower phase, litters from PRE-fed sows had a greater ADG in parity 2 but lower ADG in parity 3 vs. litters from CON-fed sows (0.99 vs. 0.94 and 0.93 vs. 1.01 ± 0.03 kg/d for parities 2 and 3, respectively; P < 0.05). No differences were observed for ADG or ADFI in the finisher phase or G:F in any phase for any parity. Loin eye area was smaller (52.2 vs. 55.0 ± 1.8 cm2; P < 0.05) for offspring from PRE- vs. CON-fed sows. In parity 2, carcass lean yield tended to be less for offspring from PRE- vs. CON-fed sows (58.6 vs. 59.6 ± 0.4%; P = 0.051). Minimal differences were observed for subjective and objective evaluations of loin quality. Closely meeting the estimated daily energy and Lys requirements for sows throughout gestation for three consecutive pregnancy cycles improved offspring growth performance (ADG and ADFI) in the final nursery stage, but generally did not affect growth performance in grower/finisher periods or carcass and loin quality at ~125 kg BW.

Fifty-six litters from first-parity sows standardized to 12 piglets were used to determine the effects of creep feed composition and form on pre- and post-weaning pig growth performance and the utilization of low-complexity nursery diets. At 5 days of age, litters (initial body weight [BW] 2.31 ± 0.61 kg) were assigned to one of four creep feeding regimens (n = 14): 1) pelleted commercial creep feed (COM), 2) liquid milk replacer (LMR), 3) pelleted milk replacer (PMR), or 4) no creep feed (NO); creep feeds contained 1.0% brilliant blue as a fecal marker. Individual piglet BW and fecal swabs were collected every 3 ± 1 days during the creep-feeding period. The latter was to identify piglets that regularly consumed creep feed via the visual appearance of blue dye in the feces. At weaning (21 ± 2 days of age), six pigs per litter with median BW that consumed creep feed were placed on either a HIGH− (contained highly digestible animal proteins) or LOW− (contained corn and soybean meal as the main protein sources) complexity nursery diet (n = 7) in a three-phase feeding program over 39 days. On day 8, two pigs per pen were sacrificed to collect organ weights and digesta. The LMR disappeared at the greatest rate (average 37.7 g/pig/d; dry matter-basis) versus COM and PMR (10.6 and 10.3 ± 1.5 g/pig/d, respectively; P < 0.001). Litters that received LMR had the greatest proportion of pigs with blue fecal swabs throughout the creep feeding period (85.0 vs. 54.9 and 63.0% ± 0.4% for COM and PMR, respectively; P < 0.05) and LMR piglets had greater BW at weaning versus all other treatments (6.32 vs. 6.02, 5.92, and 5.67 ± 0.14 kg, for LMR, COM, NO, and PMR, respectively; P < 0.001). Overall, pigs given LOW (vs. HIGH) diets in the nursery period had reduced average daily gain (25.1 vs. 27.7 ± 0.4 g/kg BW; P < 0.001), gain:feed (0.75 vs. 0.81 ± 0.02; P < 0.001), and exit BW (21.2 vs. 24.4 ± 0.6 kg; P < 0.001); no carryover effects of creep feeding program were observed. Creep feed regimen had limited effects on nutrient digestibility of nursery diets but the apparent ileal digestibility of organic matter tended to be less at 28 days of age for pigs that received the LOW nursery diet (64.2 vs. 68.8% ± 2.5%; P = 0.076). Providing supplemental nutrition during the suckling period via LMR improved piglet BW at weaning, which did not correspond to improved post-weaning growth performance, regardless of nursery diet complexity.

This experiment evaluated how maternal metabolizable protein level and rumen-protected methionine supplementation during late gestation impacts expression of genes associated with fetal programming in beef steer offspring. In a 3 x 2 factorial arrangement, 138 Angus crossbred cows were randomly assigned one of three metabolizable protein (MP) treatments (fed to meet 90, 100, or 110% of MP requirements), with(without) 9 g/d of rumen-protected methionine (MET; Smartamine®M, Adisseo Inc.). The isocaloric diets were fed for the last 8 weeks of gestation. Steer progeny (n = 51) were assigned to one of seven pens by weight and received a common corn-based grower diet (58% corn silage, 26% alfalfa haylage, 15% soybean meal) for 47 days, followed by a finisher diet (78% high moisture corn, 12% alfalfa haylage, 8% soybean meal) for 115 ±31.5 days until slaughter. At slaughter, samples of Sternomandibularis muscle and liver were collected and snap-frozen in liquid nitrogen for RNA isolation and analysis of gene expression for growth, myogenic, and adipogenic genes. Data were analyzed using PROC GLIMMIX in SAS, with maternal MP level and MET supplementation as fixed effects, and pen as a random effect. Exceeding maternal MP requirements increased expression of myogenic regulatory factor, myogenin (MYOG; 90% MP: 0.72, 100% MP: 0.94, and 110% MP: 1.00; SEM = 0.083, respectively; P = 0.02) in muscle from steer progeny at slaughter. Methionine supplementation decreased expression of pyruvate kinase (PKM; MET: 0.81, No MET: 1.16; SEM = 0.092; P = 0.02) in muscle, which is associated with lean tissue growth. Maternal nutritional treatment did not influence hepatic gene expression (P ≥ 0.06). These data suggest that maternal nutrition may impact progeny muscle development, while maintaining metabolic function of the liver.

OBJECTIVES The purpose of the present study was to determine if protecting parenteral nutrition solutions from ambient light and supplementing with N-acetylcysteine (NAC) improves mesenteric blood flow, gut morphology, and oxidative status of parenterally fed neonates. METHODS Neonatal Yucatan miniature piglets (n = 23, 7-11 days old) were surgically fitted with central venous catheters and an ultrasonic blood flow probe around the superior mesenteric artery. Piglets were fed continuously for 7 days either light-protected (LP) or light-exposed (LE) complete parenteral nutrition that was enriched with either NAC or alanine (ALA). RESULTS There were no differences in body weight or overall gut morphology among groups after 7 days. The objective of the current study was to determine the effects of precisely meeting estimated daily energy and Lys requirements for gestating sows over three consecutive pregnancies on sow reproductive and lactation performance. A total of 105 sows (initial reproductive cycle 1.4 ± 0.5) were randomly assigned to a precision (PF; n = 50) or control (CON; n = 55) feeding program between days 2 and 9 of gestation and housed in group-pens equipped with electronic sow feeders capable of blending two diets. The PF sows received unique daily blends of two isocaloric diets (2518 kcal/kg NE; 0.80% and 0.20% standardized ileal digestible [SID] Lys, respectively), whereas CON sows received a static blend throughout gestation to achieve 0.56% SID Lys. After weaning, sows were re-bred and entered the same feeding program as in the previous pregnancy for two subsequent pregnancy cycles (PF: n = 36; CON: n = 37; average reproductive cycle: 2.4 ± 0.5; PF: n = 25; CON: n = 24; average reproductive cycle: 3.5 ± 0.5). Sows on the PF program received 97%, 105%, and 118% (average over three pregnancy cycles) of dietary energy and 67%, 79%, and 106% of SID Lys intakes compared to CON between days 5 and 37, 38 and 72, and 73 and 108 of gestation, respectively. Estimated N (26.1%) retention did not differ between gestation feeding programs in any pregnancy, but excess N excretion was less (1617 vs. 1750 ± 54 g/sow; P < 0.01) for PF vs. CON sows. Regardless of pregnancy cycle, sows that received the PF program had greater ADG between days 38 and 72 (614 vs. 518 ± 63 g/d; P < 0.05) and between days 73 and 108 (719 vs. 618 ± 94 g/d; P = 0.063) of gestation, and greater loin depth gain between days 63 and 110 of gestation (0.7 vs. −1.1 ± 1.6 mm; P < 0.05), but BW (235.1 kg) and backfat (17.8 mm) and loin (70.5 mm) depths on day 110 of gestation did not differ. The number of piglets born alive, stillborn, and mummified, and litter birth weight (16.5 kg) did not differ in any pregnancy cycle, nor did piglet ADG during lactation (250 g/d) and piglet BW (6.7 kg) at weaning. Sows that received the PF program during gestation had lower ADFI during lactation (5.7 vs. 6.2 ± 0.2 kg; P < 0.01). Therefore, using feeding programs that precisely match estimated daily energy and Lys requirements for gestating sows provides the opportunity to reduce N losses to the environment and reduce lactation feed usage, without negatively affecting sow reproductive and lactation performance.

Three hundred twenty newly weaned pigs (6.7±0.3 kg BW) were used to determine the effect of low complexity diets contaminated with deoxynivalenol (DON) and supplemented with NutraMixTM or fish oil on nursery pig growth performance. Pigs were randomly divided into 40 pens and assigned to 1 of 5 dietary treatments (n = 8): [1] high-complexity diet containing animal proteins (HC) or one of four low complexity diets with protein supplied only by corn and soybean meal with [2] no DON contamination (LC), or [3] DON contamination of 3 ppm without supplements (DON-), [4] with NutraMixTM supplementation (2 g/kg; DONNM), or [5] with fish oil supplementation (2.5%, as-fed; DONω3). Diets were fed over two phases (7 and 15 days, respectively) and a common phase III diet was fed to all pigs for 20 days. In phase I, ADG, ADFI, and G:F were not different between pigs fed the HC and LC diets, but were lower for pigs fed DONNM and DONω3(P < 0.05). In phase II, pigs fed the DON- and DONω3 diets had lower ADG than LC (375 vs. 410 g/d; P < 0.05) and lower ADFI than HC (452 vs. 519 g/d; P < 0.05), while pigs fed DON- and DONω3 had greater G:F than those fed HC (0.83 vs. 0.78; P < 0.05). The BW at the end of phase II were not different between HC and LC (13.0 kg), but tended to be less for DONω3 (12.6 kg; P = 0.084 and 0.079, respectively). In phase III and over the entire nursery period, there were no treatment effects on ADG, ADFI, G:F, or final BW (26.0±0.7 kg). Feeding low complexity diets contaminated with 3 ppm DON initially reduced growth performance, but pigs were still able to achieve BW not different from HC pigs at the end of the nursery period, regardless of supplementation.

One hundred and forty-four newly weaned pigs (6.74 ± 0.23 kg initial BW; 21 d of age) were used to determine the effect of partially replacing animal protein sources with black solider fly larvae meal (BSFLM) in nursery diets on growth performance, gut morphology, and immune response. After weaning, pigs were placed in 24 pens (six pigs per pen) and pens were randomly assigned to one of four dietary treatments (study d 0; n = 6), which were fed over three phases (phases I, II, and III were fed for 7, 14, and 21 d, respectively). Two nursery diets were formulated with 25% (LowFF) and 50% (HighFF) of the animal protein sources replaced by full fat BSFLM. Conventional nursery diets including animal protein sources without (CON−) and with antibiotics (220 mg Aureomycin per kg of complete feed; CON+) served as controls. On d 8, two pigs per pen were sacrificed to collect organ weights and for intestinal histomorphological measurements. On d 9 and d 23, two pigs per pen were vaccinated with the novel antigen ovalbumin (OVA). Blood samples were collected on d 9, d 23, and d 38 to assess concentrations of plasma haptoglobin and OVA-specific immunoglobulins G (IgG) and IgG1. On d 38, the same two pigs per pen underwent a dermal hypersensitivity test and skin-fold thickness was measured at 0, 6, 24, and 48 h postintradermal injection with OVA. Pigs fed the CON- had greater ADFI and lower G:F in phase II vs. those fed CON+ and HighFF diets (P < 0.05 and P < 0.05); intermediate ADFI was observed for pigs fed the LowFF diet. Overall in the nursery period, ADG (496 ± 13 g), ADFI (743 ± 23 g), G:F, and final BW (27.61 ± 0.66 kg) were not different among dietary treatments. There were no differences in organ weights, jejunal or ileal villus heights, or crypt depths among dietary treatments. There were no differences in OVA-specific IgG, IgG1, or plasma haptoglobin among dietary treatments at any of the blood sampling times. Although not different, pigs fed the LowFF, HighFF, and CON+ diets had respectively 2.0×, 1.7×, and 1.4× greater dermal hypersensitivity response to OVA versus those fed CON−. Both inclusion levels of BSFLM in nursery diets supported growth performance, gut morphology, and indices of immune function not different from the CON+, which suggest that full fat BSFLM can replace at least 50% of animal protein sources in nursery diets of pigs without any deleterious effects on pig growth.

Comparative efficacy of deoxynivalenol (DON) detoxifying feed additives (FA) was evaluated in growth performance (exp. 1) and apparent total tract digestibility (ATTD; exp. 2) nursery pig studies. Six corn–soybean meal-based diets were used: 1) positive control (PC, formulated with 0.05) to that of pigs fed PC diet but greater (P = 0.01) than for pigs fed NC without or with other FA. Pigs fed NC and NC + FA2 had lower (P = 0.026) average daily feed intake (ADFI) than pigs fed PC and NC + FA3. Pigs fed NC + FA4 had greater (P = 0.003) G:F than pigs fed the other diets. Diets had no effect (P > 0.05) on ADG, ADFI, and G: F after first week, plasma concentration of urea and creatinine or liver and spleen weight. Pigs fed NC diets had greater (P = 0.01) jejunal mRNA expression of superoxide dismutase 1 relative to pigs fed PC or NC plus FA. Jejunal histomorphology and mRNA expression of nutrient transporters, inflammatory cytokines, and tight junction proteins and ceca digesta concentration of short-chain fatty acids were not affected (P > 0.05) by the diet. In exp. 2, 24 barrows (BW 10.2 ± 0.3 kg) were individually placed in metabolism crates and allocated to four diets: PC, NC, NC + FA3, and NC + FA4 (n = 6) containing TiO2 as digestibility marker. Pigs were adjusted to diets for 5 d, followed by a 2-d grab fecal sample collection. Pigs fed PC and NC + FA4 diets had higher ATTD of dry matter, gross energy, and crude protein than NC fed pigs. The FA3 was intermediate in digestibility response. In conclusion, FA containing sequestering component plus fungal extract or SMB in DON-contaminated feed resulted in commensurate nursery pig performance to PC. The tested FA mitigated intestinal oxidative stress through decreased expression of genes for superoxide dismutase.

New ingredients need to be characterized both chemically and in vivo, then compared to commonly-used protein ingredients before they can be considered for use in feeds. The objective of the present study was to determine the standardized ileal digestibility (SID, %) of amino acids (AA) of partially defatted black soldier fly larvae meal (BSFLM) (49.7% CP; 14.4% crude fat as fed). Subsequently, these results were compared to the SID AA of common protein ingredients in swine feed, fish meal (FM) and soybean meal (SBM) (NRC 2012), and to published full-fat (FF) BSFLM SID values. Six ileal-cannulated barrows (18.03 ± 0.67 kg BW) were used in a 2x2 Latin square design and fed either a nitrogen-free diet (NFD) or BSFLM-containing diet over two 11-d experimental periods. In each period, barrows were adapted for 7 days to the diet and ileal digesta collection was conducted on day 10 and 11 for 8h per day. Basal endogenous AA losses (EAAL) were calculated from pigs fed the NFD and as expected, the ileal AA content were significantly higher for pigs fed the BSFLM diet than NFD (P < 0.05). SID for AA were then calculated based on the obtained EAAL values following Stein et al., 2007. For essential AA, the SID of BSFLM were all above 82%, the highest being Arg (93.76%), followed by Met (91.32%), and Phe (89.80%). The SID of essential AA were on average 5.0% higher in BSFLM than FM, but 2.7% lower than SBM. When compared to the FF BSFLM, SID of essential AA of BSFLM were 1–5% higher, except for Phe and Thr. These results suggest that partially defatted BSFLM is a promising alternative protein ingredient in growing pig diets, which has a more digestible AA profile than FM, only slightly lower than SBM, and comparable to the FF BSFLM.

The objective of this study was to assess if rumen-protected methionine supplementation during gestation would impact male offspring performance and carcass quality in an applied setting. Sixty-seven gestating cows were randomly assigned to control (CON), or methionine (MET) treatments. Cows had ad libitum access to hay from a round bale feeder and were fed once daily in a bunk for approximately eight weeks prior to calving either: 0.75 kg/head/d of supplement pellet supplying 12 g rumen-protected MET/cow/d, or identical pellet with no added MET. The 34 steer progeny (MET n = 18; CON n = 16) were transported to a feedlot, assigned to one of seven pens by weight and fed a corn-based grower diet (58% corn silage, 26% alfalfa haylage, 15% soybean meal) for 47 days, followed by a finisher diet (78% high moisture corn, 12% alfalfa haylage, 8% soybean meal) for 115±31.5 days until slaughter. Body weights were recorded biweekly. Organ weights were recorded at slaughter. Carcass quality, meat quality, and rib composition were recorded 24 to 48 hours after slaughter. Data were analyzed using PROC GLIMMIX in SAS, with treatment as the fixed effect, and pen as the random effect. Supplementation of MET during pregnancy increased weaning weight (MET: 311, CON: 291 ±14.8 kg; P ≤ 0.001), final weight (ME: 668, CON: 631 ±16.8 kg; P = 0.01), and individual dry matter intake (MET: 13, CON: 12 ±0.5 kg/d; P = 0.04) during the finishing phase. There was no effect of maternal methionine supplementation (P ≥ 0.05) for average daily gain. Hot carcass weight was greater for MET steers (MET 372 vs. CON 353 ±9.5 kg; P = 0.03), while organ weights and carcass quality were not affected (P ≥ 0.05) by MET supplementation. This study showed that supplementing methionine during late gestation in an industry applied setting improved some offspring performance measures but did not result in enhanced carcass quality.

Sixty-three gilts were recruited at 90 days of age to evaluate the effects of a high-fiber gilt development feeding program on body weight and composition at breeding. Gilts (initial BW 50.9 ± 0.9 kg) were housed individually and randomly attributed to one of four feeding programs: [1] commercial diet fed ad libitum (CON), [2] commercial diet fed 10%, or [3] 20% below ad libitum, and [4] a high-fiber diet fed ad libitum (25% more fiber than the commercial diet and energy density reduced by 5%; FIB). Gilts received the feeding program between 90 days of age and breeding at ~190 days of age. Body weight and feed disappearance were determined weekly. Backfat depth was determined at 90, 145 (puberty), 160, and 190 (breeding) days of age. Over the entire experimental period, CON and FIB gilts had greater ADFI (mean of 3.50 ±0.07 kg) compared to 10% (2.95 ± 0.06 kg) and 20% gilts (2.70 ± 0.07kg; P < 0.05). The FIB feeding program reduced total energy and lysine intakes to amounts similar to 10%; both intakes were less than CON but greater than 20% gilts (P < 0.05). The ADG of FIB was less between days 145 (puberty) and 160 of age compared to CON gilts (0.86 vs. 1.09 ± 0.07 kg; P < 0.05). At breeding, FIB and 10% weighed less (146.5 ± 1.6 kg) than CON (152.7 ± 1.6 kg) and more than 20% gilts (138.7 ± 1.5 kg; P < 0.05). The FIB had less backfat than CON at breeding (14.9 vs. 16.7 ± 0.5 mm; P < 0.05), but did not differ from 10% or 20% gilts. In conclusion, the FIB feeding program limited energy intake, growth, and body fatness of gilts at breeding, even though gilts were offered feed ad libitum. Therefore, high-fiber feeding programs could be a practical means to control growth rates of developing gilts in commercial scenarios.

Sows were assigned a control (static diet composition; CON) or precision (specialized daily blend of two diets to meet estimated nutrient and energy requirements; PF) feeding regimen between d7 and 110 of gestation (n=12). At weaning (21 days of age), six pigs per litter were randomly selected and placed in a pen (1 pen per litter). All pens received ad libitum access to commercial diets in 6 phases (4-phase nursery, grower, and finisher, respectively). Individual body weights and per pen feed intakes were recorded weekly (nursery and finishing phases) or biweekly (growing phase). Four pigs per pen were slaughtered at ~125kg BW to measure carcass and loin quality. During nursery phase-3, ADG (0.60 vs. 0.53±0.02 kg/d;P< 0.05) and G:F (0.88 vs. 0.79±0.02;P< 0.05) were greater for pigs from PF versus CON sows. The ADFI in nursery phase-4 tended to be greater for pigs from PF versus CON sows (1.9 vs. 1.2±0.06 kg/d; P=0.05). By day 66 of age (end of nursery phase), pigs from PF sows had greater BW than those from CON (31.8 vs. 30.0±0.5 kg; P< 0.05); no further differences were observed for growth performance. After slaughter, there were no differences in carcass quality (e.g., hot carcass weight and lean yield) but the loin color for pigs from PF sows tended to be darker (L*; 47.4 vs. 46.5±0.1; P=0.06) and less red (a*; 6.5 vs. 6.8±0.3P=0.09) than CON. Closely meeting daily requirements for first parity sows during gestation improved offspring growth in the nursery phase, while carcass and loin quality at ~125 kg BW were generally not affected.

To evaluate if maternal supplementation of protein and rumen-protected methionine during late gestation affects steer progeny performance and carcass quality, 138 Angus X cows were used in a 3 x 2 factorial arrangement of treatments. Cows were randomly assigned one of three protein treatments (feeding to meet 90%, 100%, or 110% of metabolizable protein (MP) requirements), with(without) 9 g/d of rumen-protected methionine (MET) and fed these isocaloric diets for ~8 weeks before parturition. Steer progeny (n = 56) were assigned to one of seven pens by weight and received a common corn-based grower diet (58% corn silage, 26% alfalfa haylage, 15% soybean meal) for 47 days, followed by a finisher diet (78% high moisture corn, 12% alfalfa haylage, 8% soybean meal) for 115±31.5 days until slaughter. Body weights were recorded biweekly. Monthly ultrasound images of back and rump fats were captured. Organ weights were recorded at the time of slaughter, with carcass and meat quality recorded 24 to 48 hours later. Data were analyzed using PROC GLIMMIX in SAS, with maternal treatment as the fixed effect, and pen as the random effect. Feeding to meet 90% MP requirements resulted in heavier calf weaning weights compared to 100% and 110% (262, 251, and 245 ±13.9 kg, respectively; P = 0.01). Maternal nutritional treatment did not affect average daily gain, dry matter intake, feed conversion during finishing or organ weights. Increasing dietary MP content from 90%, 100% to 110% reduced grade fat thickness (15.5, 14.7, and 11.8 ±1.11 mm, respectively; P = 0.04) and yield grades (2.7, 2.9, and 2.3 ±0.17, respectively; P = 0.04), but increased lean meat yield (49.5, 49.4, and 50.6 ±0.3%, respectively; P = 0.04). These data suggest that maternal MP content may alter steer progeny carcass traits while maintaining animal performance during the finishing period.

A total of 144 newly weaned pigs (21 days of age) were used to determine the effect of deoxynivalenol (DON)-contaminated corn, with or without NutraMix™ supplementation, on growth performance and indices of immune system functionality during the nursery period. Pigs were randomly allocated to 24 pens of six pigs and assigned to 1 of 4 dietary treatments according to a 2X2 factorial design (n=6); treatments were fed for the 43±1-day nursery period in three phases (phases I, II, and III fed for 8, 14, and 21±1 days, respectively). The factors were clean or DON-contaminated corn and with (2 g/kg in complete diet) or without NutraMix™ supplementation. The DON diets were formulated to ensure a step-up in DON concentration for each phase (3, 4, and 5 ppm in phases I, II, and III, respectively; analyzed concentrations were 3, 3 and 4 ppm). Individual pig body weights and per pen feed intake were recorded weekly. Two pigs per pen were vaccinated against ovalbumin and candida albicans to measure humoral and cell-mediated immune responses by the cutaneous hypersensitivity response test. The ADG (600 vs 693±28 g) and ADFI (979 vs 1110±51 g) in phase III and final BW (25.2 vs 27.1±0.9 kg) were reduced for pigs fed contaminated corn (P< 0.005, P=0.088, and P< 0.05, respectively). NutraMix™ supplementation tended (P=0.086) to improve ADFI (181 vs 142±14 g) in the first week after weaning. All pigs responded to both ovalbumin and candida albicans during the cutaneous hypersensitivity response (P< 0.05), but DON nor NutraMix™ influenced the response. The DON-contaminated diets had the greatest impact on pig growth performance during phase III and when DON concentration was 4 ppm, with no modulation of the cutaneous hypersensitivity response. NutraMix™ supplementation during the early weaning period may be a means to stimulate nursery pig feed intake.

This study was conducted to test the hypothesis that supplemental dietary Arg to late-pregnant and lactating sows increases serum prolactin concentrations and mRNA abundance of SLC7A1, SLC7A2, and SLC6A14 in mammary parenchymal tissue. From day 108 of gestation and until day 21 of lactation, sows were fed a diet either supplemented with 0.10 g of l-Arg/kg body weight (BW) per day (n = 10, ARG) or 0.34 g of l-Glu/kg BW per day (n = 10, control). Litters were standardized to 10 piglets on day 1 of lactation and piglets were weighed on days 1, 7, 14, and 21 of lactation. Sow BW was recorded on day 108 of gestation and days 1, 10, and 21 of lactation. Lactation sow feed intake was recorded daily. Mammary parenchymal tissue was biopsied on day 5 of lactation to measure mRNA abundance SLC7A1, SLC7A2, and SLC6A14. On days 4 and 18 of lactation, blood samples were collected from sows at 2, 4, and 6 hr postfeeding to measure serum prolactin concentrations. Milk samples were collected on days 4, 10, and 18 of lactation to measure fat, lactose, urea N, and true protein concentrations. Sow BW, backfat, and feed intake over all sampling days did not differ between treatments. Piglet BW on d 1 tended to be greater for the ARG treatment than the control treatment (P = 0.12). Sow milk yield and composition (fat, protein, lactose, and urea N) and mammary mRNA abundance of candidate genes did not differ between the ARG and the control group. Compared to controls, serum prolactin concentrations tended to be greater (P = 0.08) in ARG sows on day 4 of lactation, and did not differ on day 18. Current findings show a potential beneficial effect of dietary supplementation with Arg to late-pregnant multiparous sows on BW of their piglets on day 1. Dietary Arg supplementation at a rate of 0.10 g/kg BW during late pregnancy and lactation tended to increase serum prolactin concentrations with no increase in mammary transcript abundance of SLC7A1, SLC7A2, and SLC6A14 in early lactation.

Growth performance, liver and spleen weight, plasma, and ceca digesta metabolites and incidences of diarrhea were investigated in growing pigs fed spent biomass of Pichia kudriavzevii. Ninety six barrows (~25 kg, 4 pigs/pen) were fed 1 of 4 experimental diets (n = 6) for 7 weeks. The diets were control, corn-, and soybean meal-based diet or control plus 2.5%, 3.75%, or 5.0% P. kudriavzevii. Diets were formulated to be isocaloric and iso nitrogenous. Feed intake and body weight (BW) were recorded weekly for calculation of average daily gain (ADG), average daily feed intake (ADFI), and gain to feed ratio (G:F). Fecal scores were taken 3 d/wk to assess incidence and severity of diarrhea. One pig/pen close to pen average was bled for plasma metabolites on days 7 and 49 and subsequently euthanized for spleen and liver weight, ileal and cecum digesta samples for concentration of short-chain fatty acids (SCFA). The concentration of crude protein, crude fat, and non-fiber carbohydrates in P. kudriavzevii biomass was 36.4%, 9.6%, and 50.8% DM, respectively. Inclusion of P. kudriavzevii tended (P = 0.06) to linearly reduce ADG from days 8 through 49 resulting in a trend (P = 0.06) for linear reduction in the final BW. The final BW was 79.0, 79.2, 76.8, and 75.5 kg for the 0%, 2.5%, 3.75%, and 5.0% P. kudriavzevii, respectively. Diets had no effect (P > 0.10) on ADFI, G:F, spleen, and liver weight throughout the entire experiment. On day 7, there was cubic (P = 0.03) decrease and quadratic (P = 0.02) increase in plasma concentration of creatinine and urea N, respectively. However, there were no (P > 0.10) diet effects on plasma metabolites on day 49. There was a tendency (P = 0.08) for linear increase in cecum digesta concentration of acetic acid. There were no diet effects (P > 0.10) on fecal score in the first 4 wk of feeding. In conclusion, feeding P. kudriavzevii yeast tended to depress growth and stimulate cecum fermentation at higher dose and had no detrimental effects on organ weights or plasma metabolites in growing pigs.

Two experiments were conducted to determine standardized ileal digestibility (SID) of amino acids (AA; Exp. 1) and net energy (Exp. 2) in two black soldier fly larvae meal (BSFLM) samples [full fat (FF; 42.5% crude protein (CP), as-fed) and defatted (DF; 40.8% CP; as-fed)] for growing pigs. Two cornstarch-based diets were formulated with FF and DF BSFLM as the sole sources of AA. A nitrogen-free diet was also used, and the corn starch:sucrose:oil ratio was kept constant among diets to calculate digestible energy (DE) by difference method. In each experiment, pigs were fed 2.8 × estimated maintenance energy requirement. In Exp. 1, eight ileal-cannulated barrows (25.1 ± 0.41 kg initial body weight) were used in a replicated 2 × 2 Latin square design (n = 8). In each period, pigs were adapted to diets for 5 d followed by 2 d of continuous ileal digesta collection for 8 h. The SID of AA were calculated using basal endogenous losses for pigs fed a nitrogen-free diet. In Exp. 2, eight barrows [23.4 ± 0.54 kg initial body weight (BW)] were used in a partially replicated Latin square design (n = 8). In each period, pigs were adapted to diets for 7 d, followed by 5 d of total urine collection and fecal grab sampling. The SID of CP (80.6 ± 1.1%) and Lys (88.0 ± 1.4%) were not different between FF and DF BSFLM. The SID of Arg, Val, Ala, and Pro tended to be less, and the SID of Met tended to be greater for the FF versus the DF BSFLM (P = 0.034, 0.090, 0.053, 0.065, 0.074, respectively). Digestible energy (4,927 vs. 3,941 ± 75 kcal/kg), metabolizable energy (4,569 vs. 3,396 ± 102 kcal/kg), and predicted net energy (3,477 vs. 2,640 ± 30 kcal/kg, using equations from Noblet; 3,479 vs. 2,287 ± 28 kcal/kg, using equations from Blok, respectively) were greater for the FF versus the DF BSFLM (P < 0.05). The apparent total tract digestibility of neutral detergent fiber and acid detergent fiber were greater for the FF versus the DF BSFLM (P ≤ 0.05). Both FF and DF BSFLM had high SID for most AA; however, FF BSFLM was a better source of net energy for growing pigs. Therefore, both FF and DF BSFLM could be used as protein alternatives in growing pig diets.

Offspring adrenal function may be negatively affected in utero by maternal stressors such as microbial infection. Maternal supplementation with immunomodulatory compounds such as omega-3 polyunsaturated fatty acids (n-3 PUFA) may help minimize the adverse effects of maternal stress on fetal hypothalamic-pituitary-adrenal development and improve offspring health. Presently, n-3 PUFA sources are primarily fish-based, but n-3 PUFA microalgae (AL) may be an alternative. Previously, it was determined that maternal AL or fish oil (FO) supplementation to sows, in addition to maternal stress induced by Escherichia coli lipopolysaccharide (LPS) challenge appeared to have a greater influence on the stress response of male offspring compared to females. To further elaborate on these findings, this study assessed the effects of maternal AL or FO supplementation combined with a maternal LPS challenge on adrenal gene expression in male offspring fed a nursery diet containing low-quality protein sources. Forty-eight sows were fed gestation diets starting on gestation day (gd) 75 containing either 3.12% AL, 3.1% FO, or a control diet containing 1.89% corn oil. On gd 112, half the sows in each treatment were administered 10 ​μg/kg LPS i.m. Piglets were weaned at 21 days of age onto a common low-quality plant-based protein diet, and one week after weaning, four piglets per sow were administered 40 ​μg/kg LPS i.m. Two hours later, the piglets were euthanized to obtain adrenal tissue, and total RNA was extracted to carry out transcriptome analysis using the Affymetrix GeneChip WT Plus assay and subsequent validation by real-time PCR. Analysis revealed that adrenal steroidogenesis, fatty acid metabolism and immune function were significantly influenced by maternal diet and stress. Increased expression of immune-related genes including lymphocyte antigen 96, TLR-2 and NF-κB suggests that maternal AL supplementation may increase offspring sensitivity to inflammation after weaning. Decreased expression of lymphocyte antigen 96 in male offspring from sows receiving maternal LPS challenge also suggests a possible role of maternal stress in diminishing the offspring immune response to immune stress challenge. Increased expression of the genes encoding the 11BHSD2 enzyme in offspring from sows fed FO may also reduce the magnitude of the stress response. These data provide insight to the immune and metabolic mechanisms that may be influenced by maternal diet and stress., Highlights • Expression of adrenal steroidogenesis genes were influenced by maternal treatment. • Expression of lipid metabolism genes and immune function genes were enriched. • Maternal algae supplementation may increase offspring sensitivity to inflammation. • Maternal stress may reduce the offspring immune response to immune challenges. • Maternal fish oil supplementation may reduce the offspring stress response.

Thirty five barrows (initial body weight [BW]: 15.1 ± 1.0 kg) were used to determine the effect of partially replacing Gly + Ser with Thr in reduced crude protein (CP) diets on growth performance, protein deposition in carcass and viscera, and skin collagen abundance during the late nursery phase to 25 kg BW. Pigs were individually fed one of five iso-nitrogenous diets (n = 7) for 21 d. The basal diet met estimated essential amino acids (AA) requirements by using all essential AA plus Gly and Ser in free form (CON; 12.1% CP; as-fed, analyzed contents). The remaining four diets were formulated by reducing total Gly and Ser concentrations to 60% or 20% of the CON diet. The N removed with Gly and Ser was replaced with either crystalline Thr or Glu. Total analyzed Thr made up either 1.59% (T1; 12.5% CP) or 2.34% (T2; 12.2% CP) of the Thr-supplemented diets, and total analyzed Glu made up either 3.47% (G1; 12.7% CP) or 4.64% (G2; 12.9% CP) of the Glu-supplemented diets. Pigs were slaughtered on day 21 to determine body composition and skin collagen abundance via bright field microscopy. Overall, average daily gain (ADG) and G:F and final carcass weights were greater for pigs fed diets supplemented with Glu (G1 + G2) vs. those fed diets supplemented with Thr (T1 + T2; P < 0.05, P = 0.060, and P = 0.050 for ADG, G:F, and final carcass weight, respectively); intermediate values were observed for CON. Nitrogen retention in carcass plus viscera and the AA profile of deposited protein in the carcass were not influenced by dietary treatment. Pigs fed the T2 and G2 diets had greater retention of Thr (vs. CON and G2) and Glu (vs. CON and T2) in the viscera protein, respectively (P < 0.05). The apparent utilization efficiency of standardized ileal digestible Thr for protein deposition in carcass plus viscera was less for pigs fed T2 (15.1%) vs. those fed CON (56.7%) or G2 (58.6% ± 2.9%) diets (P < 0.001). Only pigs fed T1 had skin collagen abundance not different from CON; pigs fed G1, G2, and T2 had reduced skin collagen abundance compared with CON and T1 (P < 0.01). Using Glu as an N source when Gly and Ser were reduced to 60% and 20% of CON in reduced CP diets maintained ADG for pigs between 15 and 25 kg BW, whereas supplying Thr as a N source reduced ADG and carcass weight. When dietary Gly and Ser were supplied at 60% of CON, only Thr supplementation rescued skin collagen abundance. Therefore, supplemental Thr at excess levels is not sufficient to replace N from Gly and Ser in reduced CP diets fed to late nursery pigs, despite supporting skin collagen abundance as a secondary indicator of Gly status.

Immune system stimulation (ISS) adversely affects protein and AA metabolism and reduces productivity in pigs. Leucine (Leu) has a regulatory role in skeletal muscle protein turnover, which may be affected by ISS. The objective of this study was to evaluate the effects of ISS and dietary Leu supplementation on the protein fractional synthesis rate (FSR) of various tissues in pigs. Yorkshire barrows were surgically fitted with jugular vein catheters and assigned to one of three dietary treatments: (i) CON, 1.36% standardized ileal digestible (SID) Leu; (ii) LEU-M, 2.04% SID Leu; and (iii) LEU-H, 2.72% SID Leu. The diets were formulated to contain all essential AA 10% above estimated requirements for maximum whole-body protein deposition for this BW range. At the start of the 36-h challenge period (initial BW = 14.5 ± 0.8 kg), ISS was induced in pigs with lipopolysaccharide (ISS+; n = 7, 8, and 7 for CON, LEU-M, and LEU-H pigs, respectively); a subset of CON pigs was injected with sterile saline (ISS-; n = 6). During challenge period, pigs were fed every 4 h and feed intake of ISS- pigs was kept equal to ISS+ pigs. At the end of the challenge period, FSR of liver, plasma, gastrocnemius, and LD proteins were determined with a flooding dose of l-[ring-2H5]phenylalanine (40 mol%). All essential AA, most nonessential AA, and plasma urea-N peaked at 12 h and declined to baseline levels at 36 h after ISS was induced in ISS+ pigs (P < 0.05), whereas plasma AA and urea-N concentrations were constant in ISS- pigs. At 36 h, dietary Leu supplementation resulted in a linear decline in plasma isoleucine, valine, glutamine, and urea nitrogen concentrations (P < 0.05), whereas plasma Leu concentration was unaffected. Liver protein FSR was increased in ISS+ pigs (P < 0.05), whereas plasma and skeletal muscle protein FSR was not affected by ISS. Dietary Leu supplementation tended to diminish liver protein FSR (linear reduction; P = 0.052) and increase gastrocnemius protein FSR (linear increase; P = 0.085) in ISS+ pigs. Leucine supplementation above estimated requirements may support repartitioning of AA from visceral to peripheral protein deposition during ISS.

Physical and chemical body composition of gilts and parity 3 sows were used to determine current prediction equation accuracy and propose alternative prediction equations that incorporate additional variables. Longissimus dorsi muscle depth and parity can be combined with body weight and backfat to improve gilt and sow body composition prediction.

A total of 240 newly weaned pigs (5.25 ± 0.15 kg BW) were used to determine the dietary omega-6-to-omega-3 (ω-6:ω-3) fatty acid ratio that optimized growth performance and immune responses when fed corn and soybean meal (SBM)-based diets with low protein quality. Pigs were randomly assigned to 1 of 5 dietary treatments (n = 6 pens per treatment; day 0 of study): [1] positive control (High; included animal proteins and 5% corn oil), [2] negative control (Low0; corn- and SBM-based and 5% corn oil), or 1 of 3 Low diets with increasing supplementation of fish oil to replace corn oil: [3] 1.25% (Low1.25), [4] 2.5% (Low2.5), [5] 5% (Low5) to achieve 5:1, 3:1, and 1:1 ω-6:ω-3 ratios, respectively. Pigs were fed dietary treatments in 2 phases for 7 and 14 d, respectively, followed by a common phase III diet for 21 d. On day 6 and 20, 12 pigs per treatment were immune sensitized with 0.5 mg ovalbumin (OVA) and 0.5 mg Quil A adjuvant in 1 mL saline. The dermal hypersensitivity response (DHR) was evaluated on day 40 in these same pigs, using intradermal injection of OVA; changes in skin-fold thickness were measured. On day 21, 4 pigs per pen were immune challenged with LPS (30 µg Escherichia coli LPS per kg BW) or saline (n = 12); rectal temperature was monitored over 3 h. During phase I only, ADG, ADFI, and G:F were greater for pigs fed the High diet vs. those fed the Low diet (P < 0.05), and increased with increasing fish oil supplementation up to 2.5% (Low2.5), but decreased for pigs fed the Low5 diet (quadratic; P < 0.05, P = 0.086, and P < 0.05 for ADG, ADFI, and G:F, respectively). On day 21, LPS increased rectal temperature (vs. saline at 1-, 2-, and 3-h post-challenge; P < 0.001); fish oil supplementation reduced rectal temperature 2-h post-challenge in the Low-fed pigs (linear; P < 0.05). On day 22, serum haptoglobin was greatest for pigs fed Low0 and decreased with increasing fish oil supplementation (linear; P < 0.05). Immunization with OVA induced a serum anti-OVA IgG response, which was reduced on day 34 among pigs fed Low diets with increasing fish oil supplementation (linear; P = 0.050). On day 40, and 6 h after intradermal injection of OVA, the DHR was least for pigs fed the Low2.5 diet (P < 0.05). Inclusion of 2.5% fish oil (3:1, ω-6:ω-3) optimized growth performance during the early nursery phase when pigs were most sensitive to diets with low protein quality; the ideal ω-6-to-ω-3 fatty acid ratio may differ when using immune responses as the major outcome.

Twenty-eight lactating Yorkshire and Yorkshire × Landrace primiparous sows were used to test the hypothesis that feeding a diet with reduced CP concentration and supplemented with crystalline AA (CAA) does not decrease milk protein yield and litter growth but improves apparent N utilization for milk protein production. Sows were assigned to 1 of 2 dietary treatments: 1) control (CON; 16.2% CP; analyzed content) or 2) low CP with CAA to meet estimated requirements of limiting AA (LCP; 12.7% CP) over a 17-d lactation period. A N balance was conducted for each sow between days 13 and 17 of lactation. On day 17, a 12-h primed continuous infusion of l-[ring-2H5]-Phe was conducted on 12 sows (n = 6) with serial blood and milk sampling to determine plasma AA concentrations and Phe enrichment, and milk casein synthesis, respectively. Thereafter, sows were sacrificed and tissues were collected to determine tissue protein fractional synthesis rates (FSR). Litter growth rate and milk composition did not differ. Sows fed the LCP diet had reduced N intake (122.7 vs. 153.2 g/d; P < 0.001) and maternal N retention (13.5 vs. 24.6 g/d; P < 0.05) and greater apparent efficiency of using dietary N intake for milk production (85.1% vs. 67.5%; P < 0.001). On day 17 of lactation, all plasma essential AA concentrations exhibited a quartic relationship over time relative to consumption of a meal, where peaks occurred at approximately 1- and 4-h postprandial (P < 0.05). Protein FSR in liver, LM, gastrocnemius muscle, mammary gland, and in milk caseins did not differ between treatments. Feeding primiparous sows with a diet containing 12.7% CP and supplemented with CAA to meet the limiting AA requirements did not reduce milk protein yield or piglet growth rate and increased the apparent utilization of dietary N, Arg, Leu, Phe+Tyr, and Trp for milk protein production. The improved apparent utilization of N and AA appears to be related exclusively to a reduction in N and AA intake.

Fifty-six litters standardized to 12 piglets from first-parity sows were used to determine the effects of creep and nursery diet compositions on pre- and post-weaning pig growth performance. At three days of age, litters (initial BW 2.31±0.61kg) were assigned to one of four creep feeding regimens (n=14): [1] commercial creep feed (COM), [2] liquid milk replacer (LMR), [3] pelleted milk replacer (PMR), or [4] no creep feed (NO); creep feeds contained 1.0% brilliant blue as a fecal marker. Fecal swabs were collected every 3±1 days to identify piglets that regularly consumed creep feed. At weaning (18±1 days of age), six pigs per litter that consumed creep feed were placed on either a HIGH- (contained highly digestible animal proteins) or LOW- (contained corn and soybean meal as the main protein sources) quality nursery diet (n = 7) in a three-phase feeding program over 38 days. The LMR disappeared at the greatest rate (37.7 g/pig/d; DM-basis) versus COM and PMR (10.8±1.5 g/pig/d; P < 0.001). Litters that received LMR had the greatest proportion of pigs with blue fecal swabs between study days 4 and 15 (85.0 vs 59.0±0.4%; P < 0.05) and LMR piglets had greater BW at weaning versus all other treatments (6.32, 6.02, 5.92, 5.67±0.14 kg, for LMR, COM, NO, and PMR, respectively; P < 0.001). Over the entire nursery period, pigs that received LOW diets had reduced ADG (399 vs 485±42 g; P < 0.001), ADFI (520 vs 595±37 g; P< 0.001), G:F (0.77 vs 0.82±0.03; P < 0.01), and BW at the end of the nursery period (21.2 vs 24.4±1.6 kg; P < 0.001), with no carryover effects of creep feeding regimen. Providing supplemental nutrition during the suckling period via LMR improved pig body weight at weaning, but did not improve post-weaning growth performance, regardless of nursery diet quality.
