Retrogressive thaw slumps (RTSs) are spectacular landforms that occur due to the thawing of ice-rich permafrost or melting of massive ground ice, often in hillslope terrain. RTSs occur in the Arctic, the subarctic, and high mountain (Qinghai–Tibet Plateau) permafrost regions and are observed to expand in size and number due to climate warming. As the observation of RTSs is receiving more and more attention due to their important role in permafrost thaw; impacts on topography; mobilization of sediment, carbon, nutrients, and contaminants; and their effects on downstream hydrology and water quality, the thematic breadth of studies increases and scientists from different scientific backgrounds and perspectives contribute to new RTS research. At this point, a wide range of terminologies originating from different scientific schools is used, and we identified the need to provide an overview of variable characteristics of RTSs to clarify terminologies and ease the understanding of the literature related to RTS processes, dynamics, and feedbacks. We review the theoretical geomorphological background of RTS formation and landform characteristics to provide an up-to-date understanding of the current views on terminology and underlying processes. The presented overview can be used not only by the international permafrost community but also by scientists working on ecological, hydrological, and biogeochemical consequences of RTS occurrence and by remote-sensing specialists developing automated methods for mapping RTS dynamics. The review will foster a better understanding of the nature and diversity of RTS phenomena and provide a useful base for experts in the field but also ease the introduction to the topic of RTSs for scientists who are new to it.

Pattern-triggered immunity (PTI) is a critical defense mechanism employed by plants against pathogen attacks. This study explores the role of PTI induced by the Xyn11/eix fungal elicitor in two commercially valuable Rosaceae species, Prunus persica (peach) and Prunus avium (sweet cherry). Our findings demonstrate that Xyn11/eix triggers two specific defense responses: the increase in ethylene production and the induction of cell death. Furthermore, Xyn11/eix-mediated PTI significantly reduces the susceptibility to Botrytis cinerea infection in both species. The study reveals changes in gene expression patterns after Xyn11/eix treatment. Notably, ACO1 and SARDEF1 genes, involved in ethylene and salycilic acid biosynthesis, respectively, are upregulated in P. persica, but not in P. avium at the time point analyzed. This result suggests a potential role for the ethylene and salicylic acid signaling in Xyn11/mix-mediated PTI in P. persica. Additionally, the research identified functional orthologues of LeEIX2, the receptor for Xyn11/eix in Solanum lycopersicum, within both Prunes genomes. Altogether, these results suggest a remarkable functional convergence between Rosaceae and Solanaceae plants in the Xyn11/eix mediated defense responses although not at the transcriptional level, and opens new avenues for developing novel disease control strategies for stone fruits.

Land cover heterogeneity information considering soil wetness across the entire Arctic tundra is of interest for a wide range of applications targeting climate change impacts and ecological research questions. Patterns are potentially linked to permafrost degradation and affect carbon fluxes. First, a land cover unit retrieval scheme which provides unprecedented detail by fusion of satellite data using Sentinel-1 (synthetic aperture radar) and Sentinel-2 (multispectral) was adapted. Patterns of lakes, wetlands, general soil moisture conditions and vegetation physiognomy are interpreted at 10 m nominal resolution. Units with similar patterns were identified with a k-means approach and documented through statistics derived from comprehensive in situ records for soils and vegetation (more than 3500 samples). The result goes beyond the capability of existing land cover maps which have deficiencies in spatial resolution, thematic content and accuracy, although landscape heterogeneity related to moisture gradients cannot be fully resolved at 10 m. Wetness gradients were assessed, and measures for landscape heterogeneity were derived north of the treeline. About 40 % of the area north of the treeline falls into three units of dry types with limited shrub growth. Wetter regions have higher land cover diversity than drier regions. An area of 66 % of the analysed Arctic landscape is highly heterogeneous with respect to wetness at a 1 km scale (representative scale of frequently used regional land cover and permafrost modelling products). Wetland areas cover 9 % and moist tundra types 32 %, which is of relevance for methane flux upscaling.

BackgroundTransforming growth factor (TGF)-ß1 is a pleiotropic cytokine that can promote tumor growth and suppress antitumor immune responses. Latent TGF-ß1 associates with glycoprotein-A repetition predominant (GARP) on the surface of regulatory T cells prior to its activation and release. Livmoniplimab is a monoclonal antibody (mAb) that binds the GARP:TGF-ß1 complex to inhibit activation and release of TGF-ß1. It is in clinical development in combination with budigalimab, an anti-programmed cell death protein 1 Fc-modified mAb. The first-in-human, phase 1, dose-escalation results are presented herein (ClinicalTrials.gov: NCT03821935).MethodsThe dose-escalation phase enrolled adult patients with advanced solid tumors. Patients received escalating doses of livmoniplimab ranging from 3mg to 1500mg, once every 2 weeks (Q2W), as monotherapy or in combination with a 500mg fixed dose of budigalimab Q4W. The primary objective of the dose escalation was to determine the recommended phase 2 dose. Secondary objectives were to assess safety and pharmacokinetics (PK), and exploratory objectives included evaluating preliminary efficacy.ResultsFifty-seven patients enrolled in the dose escalation: 23 in monotherapy cohorts and 34 in combination therapy cohorts. Dose-limiting toxicities were limited, no maximum tolerated dose was reached, and the maximum administered dose of 1500mg was selected for dose expansion. The most common adverse events reported in monotherapy-treated patients were fatigue, anemia, and nausea, and those in combination therapy-treated patients were pruritus, fatigue, nausea, and anemia. Livmoniplimab exhibited dose-proportional PK, and peripheral blood biomarker data demonstrated saturation of the GARP:TGF-ß1 complex on platelets at livmoniplimab doses within the linear PK range. No objective tumor responses were observed in the monotherapy dose escalation. However, the objective response rate was 15% in the combination dose escalation, with a median response duration of 8.4 months.ConclusionLivmoniplimab was well-tolerated as monotherapy and in combination with budigalimab in the dose-escalation phase. Encouraging preliminary efficacy was demonstrated in the combination dose escalation in heavily pretreated patients, supporting further development of this novel drug combination in patients with advanced solid tumors.

RNA-dependent RNA polymerases (RDRs) regulate important aspects of plant development and resistance to pathogens. The role of RDRs in virus resistance has been demonstrated using siRNA signal amplification and through the methylation of viral genomes. Cucumber (Cucumis sativus) has four RDR1 genes that are differentially induced during virus infection: CsRDR1a, CsRDR1b, and duplicated CsRDR1c1/c2. The mode of action of CsRDR1s during viral infection is unknown. Transient expression of the cucumber mosaic virus (CMV)-2b protein (the viral suppressor of RNA silencing) in cucumber protoplasts induced the expression of CsRDR1c, but not of CsRDR1a/1b. Results from the yeast two-hybrid system showed that CsRDR1 proteins interacted with CMV-2b and this was confirmed by bimolecular fluorescence complementation assays. In protoplasts, CsRDR1s localized in the cytoplasm as punctate spots. Colocalization experiments revealed that CsRDR1s and CMV-2b were uniformly dispersed throughout the cytoplasm, suggesting that CsRDR1s are redistributed as a result of interactions. Transient overexpression of individual CsRDR1a/1b genes in protoplasts reduced CMV accumulation, indicating their antiviral role. However, overexpression of CsRDR1c in protoplasts resulted in relatively higher accumulation of CMV and CMVΔ2b. In single cells, CsRDR1c enhances viral replication, leading to CMV accumulation and blocking secondary siRNA amplification of CsRDR1c by CMV-2b protein. This suggests that CMV-2b acts as both a transcription factor that induces CsRDR1c (controlling virus accumulation) and a suppressor of CsRDR1c activity.
