Abstract

Mucin-domain glycoproteins are densely O-glycosylated and play critical roles in a host of biological functions. In particular, the T cell immunoglobulin and mucin-domain containing family of proteins (TIM-1, -3, -4) decorate immune cells and act as key regulators in cellular immunity. However, their dense O-glycosylation remains enigmatic, primarily due to the challenges associated with studying mucin domains. Here, we demonstrate that the mucinase SmE has a unique ability to cleave at residues bearing very complex glycans. SmE enables improved mass spectrometric analysis of several mucins, including the entire TIM family. With this information in-hand, we perform molecular dynamics (MD) simulations of TIM-3 and -4 to understand how glycosylation affects structural features of these proteins. Finally, we use these models to investigate the functional relevance of glycosylation for TIM-3 function and ligand binding. Overall, we present a powerful workflow to better understand the detailed molecular structures and functions of the mucinome.

5. A study of daytime convective vortices and turbulence in the martian Planetary Boundary Layer based on half-a-year of InSight atmospheric measurements and Large-Eddy Simulations

Abstract

Studying the atmospheric Planetary Boundary Layer (PBL) is crucial to understand the climate of a planet. The meteorological measurements by the instruments onboard InSight at a latitude of 4.5$^{\circ}$N make a uniquely rich dataset to study the active turbulent dynamics of the daytime PBL on Mars. Here we use the high-sensitivity continuous pressure, wind, temperature measurements in the first 400 sols of InSight operations (from northern late winter to midsummer) to analyze wind gusts, convective cells and vortices in Mars' daytime PBL. We compare InSight measurements to turbulence-resolving Large-Eddy Simulations (LES). The daytime PBL turbulence at the InSight landing site is very active, with clearly identified signatures of convective cells and a vast population of 6000 recorded vortex encounters, adequately represented by a power-law with a 3.4 exponent. While the daily variability of vortex encounters at InSight can be explained by the statistical nature of turbulence, the seasonal variability is positively correlated with ambient wind speed, which is supported by LES. However, wind gustiness is positively correlated to surface temperature rather than ambient wind speed and sensible heat flux, confirming the radiative control of the daytime martian PBL; and fewer convective vortices are forming in LES when the background wind is doubled. Thus, the long-term seasonal variability of vortex encounters at the InSight landing site is mainly controlled by the advection of convective vortices by ambient wind speed. Abstract

Effective treatment of pediatric solid tumors has been hampered by the predominance of currently "undruggable" driver transcription factors. Improving outcomes while decreasing the toxicity of treatment necessitates the development of novel agents that can directly inhibit or degrade these elusive targets. MYCN in pediatric neural-derived tumors, including neuroblastoma and medulloblastoma, is a paradigmatic example of this problem. Attempts to directly and specifically target MYCN have failed due to its similarity to MYC, the unstructured nature of MYC family proteins in their monomeric form, the lack of an understanding of MYCN-interacting proteins and ability to test their relevance in vivo, the inability to obtain structural information on MYCN protein complexes, and the challenges of using traditional small molecules to inhibit protein-protein or protein-DNA interactions. However, there is now promise for directly targeting MYCN based on scientific and technological advances on all of these fronts. Here, we discuss prior challenges and the reasons for renewed optimism in directly targeting this "undruggable" transcription factor, which we hope will lead to improved outcomes for patients with pediatric cancer and create a framework for targeting driver oncoproteins regulating gene transcription.

Abstract

Northern line-of-sight extinction within Gale Crater during the 2018 global dust storm was monitored daily using MSL's Navcam. Additional observations with Mastcam (north) and Navcam (all directions) were obtained at a lower cadence. Using feature identification and geo-referencing, extinction was estimated in all possible directions. Peak extinction of $>1.1$ km$^{-1}$ was measured between sols 2086 and 2090, an order of magnitude higher than previous maxima. Northern and western directions show an initial decrease, followed by a secondary peak in extinction, not seen in column opacity measurements. Due to foreground topography, eastern direction results are provided only as limits, and southern results were indeterminable. Mastcam red and green filter results agree well, but blue filter results show higher extinctions, likely due to low signal-to-noise. Abstract

We derive water vapor column abundances and aerosol properties from Mars Science Laboratory (MSL) ChemCam passive mode observations of scattered sky light. Each ChemCam passive sky observation acquires spectra at two different elevation angles. We fit these spectra with a discrete-ordinates multiple scattering radiative transfer model, using the correlated-k approximation for gas absorption bands. The retrieval proceeds by first fitting the continuum of the ratio of the two elevation angles to solve for aerosol properties, and then fitting the continuum-removed ratio to solve for gas abundances. The final step of the retrieval makes use of the observed CO2 absorptions and the known CO2 abundance to correct the retrieved water vapor abundance for the effects of the vertical distribution of scattering aerosols and to derive an aerosol scale height parameter. The ChemCam-retrieved water abundances show, with only a few exceptions, the same seasonal behavior and the same timing of seasonal minima and maxima as the TES, CRISM, and REMS-H data sets that we compare them to. However ChemCam-retrieved water abundances are generally lower than zonal and regional scale from-orbit water vapor data, while at the same time being significantly larger than pre-dawn REMS-H abundances. Pending further analysis of REMS-H volume mixing ratio uncertainties, the differences between ChemCam and REMS-H pre-dawn mixing ratios appear to be much too large to be explained by large scale circulations and thus they tend to support the hypothesis of substantial diurnal interactions of water vapor with the surface. Abstract

In the active HER receptor dimers, kinases play distinct roles; one is the catalytically active kinase and the other is its allosteric activator. This specialization enables signaling by the catalytically inactive HER3, which functions exclusively as an allosteric activator upon heterodimerization with other HER receptors. It is unclear whether the allosteric activation mechanism evolved before HER receptors functionally specialized. We determined the crystal structure of the kinase domain of the only EGF receptor in Caenorhabditis elegans, LET-23. Our structure of a non-human EGFR kinase reveals autoinhibitory features conserved in the human counterpart. Strikingly, mutations within the putative allosteric dimer interface abrogate activity of the isolated LET-23 kinase and of the full-length receptor despite these regions being only partially conserved with human EGFR. Our results indicate that ancestral EGFRs have built-in features that poise them for allosteric activation that could facilitate emergence of the catalytically dead, yet functional, orthologs.

Abstract

The Mars Science Laboratory Mast camera and Descent Imager investigations were designed, built, and operated by Malin Space Science Systems of San Diego, CA. They share common electronics and focal plane designs but have different optics. There are two Mastcams of dissimilar focal length. The Mastcam-34 has an f/8, 34 mm focal length lens, and the M-100 an f/10, 100 mm focal length lens. The M-34 field of view is about 20° × 15° with an instantaneous field of view (IFOV) of 218 μrad; the M-100 field of view (FOV) is 6.8° × 5.1° with an IFOV of 74 μrad. The M-34 can focus from 0.5 m to infinity, and the M-100 from ~1.6 m to infinity. All three cameras can acquire color images through a Bayer color filter array, and the Mastcams can also acquire images through seven science filters. Images are ≤1600 pixels wide by 1200 pixels tall. The Mastcams, mounted on the ~2 m tall Remote Sensing Mast, have a 360° azimuth and ~180° elevation field of regard. Mars Descent Imager is fixed-mounted to the bottom left front side of the rover at ~66 cm above the surface. Its fixed focus lens is in focus from ~2 m to infinity, but out of focus at 66 cm. The f/3 lens has a FOV of ~70° by 52° across and along the direction of motion, with an IFOV of 0.76 mrad. All cameras can acquire video at 4 frames/second for full frames or 720p HD at 6 fps. Images can be processed using lossy Joint Photographic Experts Group and predictive lossless compression.

Abstract

The James Webb Space Telescope (JWST), scheduled for launch in 2018, is the successor to the Hubble Space Telescope (HST) but with a significantly larger aperture (6.5 m) and advanced instrumentation focusing on infrared science (0.6-28.0 $\mu$m ). In this paper we examine the potential for scientific investigation of Titan using JWST, primarily with three of the four instruments: NIRSpec, NIRCam and MIRI, noting that science with NIRISS will be complementary. Five core scientific themes are identified: (i) surface (ii) tropospheric clouds (iii) tropospheric gases (iv) stratospheric composition and (v) stratospheric hazes. We discuss each theme in depth, including the scientific purpose, capabilities and limitations of the instrument suite, and suggested observing schemes. We pay particular attention to saturation, which is a problem for all three instruments, but may be alleviated for NIRCam through use of selecting small sub-arrays of the detectors - sufficient to encompass Titan, but with significantly faster read-out times. We find that JWST has very significant potential for advancing Titan science, with a spectral resolution exceeding the Cassini instrument suite at near-infrared wavelengths, and a spatial resolution exceeding HST at the same wavelengths. In particular, JWST will be valuable for time-domain monitoring of Titan, given a five to ten year expected lifetime for the observatory, for example monitoring the seasonal appearance of clouds. Abstract

Cyclic absorption of solar radiation generates oscillations in atmospheric fields. These oscillations are called atmospheric or thermal tides, which are furthermore modified by topography and surface properties. This leads to a complex mix of sun‐synchronous and non‐sun‐synchronous tides that propagate around the planet eastward and westward. This study focuses on analyzing the ter‐diurnal component (period of 8 hr) from surface pressure observations by Mars Science Laboratory (MSL), InSight, Viking Lander (VL) 1, and VL2. General Circulation Model (GCM) results are used to provide a global context for interpreting the observed ter‐diurnal tide properties. MSL and InSight have a clear and similar seasonal cycle, with local amplitude peaks at around solar longitude (Ls) 60°, Ls 130°, and Ls 320°. The amplitude peak at Ls 320° is related to the annual dust storm, while the Ls 230° dust storm is not detected by either platforms. During global dust storms, MSL, VL1, and VL2 detect their highest amplitudes. The GCM predicts the weakest amplitudes at the equinoxes, while the strongest ones are predicted in summer for both hemispheres. GCM amplitudes tend to differ from the observations but match slightly better around the aphelion season. During this time, model results suggest that the two most prominent modes are the sun‐synchronous ter‐diurnal tide and an eastward propagating resonantly enhanced Kelvin wave. Simulations with and without the effect of radiative heating by water ice clouds indicate the clouds may play a significant role in forcing the ter‐diurnal tide during northern hemisphere summer season. Plain Language Summary: Atmospheric pressure is a crucial meteorological parameter, since many weather phenomena on Mars, such as the condensation and sublimation of the seasonal polar caps, global scale atmospheric tides, and local turbulent flows, are associated with changes in surface pressure. On a diurnal time scale, atmospheric tides dominate surface pressure daily variations in the Martian tropics. They are generated by the diurnally varying solar radiation and additionally modified by topography, surface properties, and atmospheric absorbers. Here, we use pressure observations from Mars Science Laboratory, InSight, Viking lander 1, and 2 to characterize the Martian atmospheric tide that has a period of 8 hr, known as the ter‐diurnal tide, and compare results with model simulations. The results show that the Mars Science Laboratory and InSight observe similar seasonal cycles for the amplitude and phase of the ter‐diurnal tide. In addition, both these platforms react similarly to atmospheric dust content. The Viking landers and Mars Science Laboratory observe their highest amplitudes during the global dust storms. We find that modeled amplitudes are the strongest during summertime for both hemispheres and typically lower than those observed throughout the year. Moreover, the ter‐diurnal tide may be significantly affected by water ice clouds. Abstract

Dust aerosol plays a fundamental role in the behavior and evolution of the Martian atmosphere. The first five Mars years of Mars Exploration Rover data provide an unprecedented record of the dust load at two sites. This record is useful for characterization of the atmosphere at the sites and as ground truth for orbital observations. Atmospheric extinction optical depths have been derived from solar images after calibration and correction for time-varying dust that has accumulated on the camera windows. The record includes local, regional, and globally extensive dust storms. Comparison with contemporaneous thermal infrared data suggests significant variation in the size of the dust aerosols, with a 1 {\mu}m effective radius during northern summer and a 2 {\mu}m effective radius at the onset of a dust lifting event. The solar longitude (LS) 20-136{\deg} period is also characterized by the presence of cirriform clouds at the Opportunity site, especially near LS=50 and 115{\deg}. In addition to water ice clouds, a water ice haze may also be present, and carbon dioxide clouds may be present early in the season.
