Abstract Background Circular RNAs (circRNAs), one of the major contents of exosomes, have been shown to participate in the occurrence and progression of cancers. The role and the diagnostic potential of exosome-transported circRNAs in non-small-cell lung cancer (NSCLC) remain largely unknown. Methods The NSCLC-associated exosomal circ_0061407 and circ_0008103 were screened by circRNA microarray. The role of circ_0061407 and circ_0008103 in NSCLC was examined in vitro and in vivo. The encapsulation of the two circRNAs into exosomes and the transport to recipient cells were observed by confocal microscopy. The effects of exosome-transported circ_0061407 and circ_0008103 on recipient cells were investigated using a co-culture device. Bioinformatics analyses were performed to predict the mechanisms by which circ_0061407 and circ_0008103 affected NSCLC. The quantitative polymerase chain reaction was used to quantify the exosome-containing circ_0061407 and circ_0008103 in the serum samples of healthy, pneumonia, benign lung tumours, and NSCLC. The diagnostic efficacy was evaluated using receiver operating characteristic curves. Results The levels of circ_0061407 and circ_0008103 within exosomes were down-regulated in the serum of patients with NSCLC. The up-regulation of circ_0061407 and circ_0008103 inhibited the proliferation, migration/invasion, cloning formation of NSCLC cells in vitro and inhibited lung tumour growth in vivo. Circ_0061407 and circ_0008103 were observed to be packaged in exosomes and transported to recipient cells, where they inhibited the proliferation, migration/invasion, and cloning formation abilities of the recipient cells. Moreover, circ_0061407 and circ_0008103 might be involved in the progression of NSCLC by interacting with microRNAs and proteins. Additionally, lower serum exosomal circ_0061407 and circ_0008103 levels were associated with advanced pathological staging and distant metastasis. Conclusions This study identified two novel exosome-transported circRNAs (circ_0061407 and circ_0008103) associated with NSCLC. These findings may provide additional insights into the development of NSCLC and potential diagnostic biomarkers for NSCLC.

The dynamic changes of amniotic fluid during pregnancy are crucial for the development, protection, defense and nutrition of the fetus. Amniotic fluid contains a variety of trace biomarkers, such as microRNA, free DNA, reactive oxygen species, interleukin and metabolomics, etc., which are of great significance for fetal health, diagnosis and drug control of various diseases. This article reviews the latest advances in the detection of microbial markers in amniotic fluid, and introduces the methods of amniotic fluid collection and attention. In addition, the biological approach to amniotic fluid testing is discussed. Finally, the challenges of amniotic fluid sample testing are discussed, with a view to promoting the clinical application of amniotic fluid in the field of assisted reproduction.

Abstract Background Extracellular vesicles (EVs) participate in cancer development via cell-to-cell communication. Long non-coding RNAs (lncRNAs), one component of EVs, can play an essential role in non-small-cell lung cancer (NSCLC) through EV-mediated delivery. Methods The NSCLC-associated lncRNA AL139294.1 in EVs was identified via lncRNA microarray analysis. The role of AL139294.1 in NSCLC was examined in vitro and in vivo. Confocal microscopy was used to observe the encapsulation of AL139294.1 into EVs and its transport to recipient cells. A co-culture device was used to examine the effects of transported AL139294.1 on the oncogenic behaviour of recipient cells. Dual-luciferase reporter assay was performed to verify the direct interaction of miR-204-5p with AL139294.1 and bromodomain-containing protein 4 (BRD4). AL139294.1 and miR-204-5p in EVs were quantified using quantitative polymerase chain reaction. Receiver operating characteristic analyses were conducted to evaluate the diagnostic efficiency. Results The lncRNA AL139294.1 in EVs promoted NSCLC progression in vitro and in vivo. After AL139294.1 was encapsulated into EVs and transported to recipient cells, it promoted the cells’ proliferation, migration, and invasion abilities by competitively binding with miR-204-5p to regulate BRD4, leading to the activation of the Wnt and NF-κB2 pathways. Additionally, the expression of serum lncRNA AL139294.1 in EVs was increased, whereas miR-204-5p in EVs was decreased in NSCLC. High levels of lncRNA AL139294.1 and low levels of miR-204-5p in EVs were associated with advanced pathological staging, lymph node metastasis, and distant metastasis, underscoring their promising utility for distinguishing between more and less severe manifestations of the disease. Conclusions This study reveals a novel lncRNA in EVs associated with NSCLC, namely, AL139294.1, providing valuable insights into the development of NSCLC and introducing potential diagnostic biomarkers for NSCLC.

Numerous studies have shown that urban rivers are becoming a source of methane emissions, which poses a challenge to developing a “net zero carbon city.” Most studies at this stage focus on the influence of single urban elements on river methane emissions, such as land use, water facilities, etc. It is necessary to consider the impact of urban development on river methane emissions in an integrated manner. This study aggregated relevant data sets for seven municipal districts in Shanghai during 2011–2020, collected through literature and statistical yearbooks. We constructed an “urban river methane emission” system based on the Driver-Pressure-State-Response model and used partial least squares-path modeling to verify the rationality of the system’s influence pathways. The results showed (goodness-of-fitness = 0.4446) that driver (population density, urbanization rate), pressure (annual water supply, total yearly household waste), and state (deteriorating water quality environment) all increased the methane emission potential of urban rivers (total effect = 0.1917; 0.3932; 0.1394). Response (sewage treatment rate, environmental investment) would mitigate river methane emissions (total effect = -0.2230). An “urban river methane emission potential” assessment model was then developed. Partial least squares-path modeling and generalized linear mixed model were used to calculate the “urban river methane emission potential” index in seven municipal districts of Shanghai over ten years, respectively. And the results of the two methods were similar. Abstract Exploration of exotic transport behavior for quantum materials is of great interest and importance for revealing exotic orders to bring new physics. In this work, we report the observation of unconventional prominent planar Hall effect (PHE) and planar anisotropic magnetoresistivity (PAMR) in strange kagome metal KV3Sb5. The PHE and PAMR, which are driven by an in-plane magnetic field and display sharp difference from other Hall effects driven by an out-of-plane magnetic field or magnetization, exhibit exotic higher-order oscillations in sharp contrast to those following empirical rule only allowing two-fold symmetrical oscillations. These higher-order oscillations exhibit strong field and temperature dependence and vanish around charge density wave (CDW) transition. The unique transport properties suggest a significant interplay of the lattice, magnetic and electronic structure in KV3Sb5. This interplay can couple the hidden anisotropy and transport electrons leading to the unconventional PHE and PAMR in contrast to other materials.

Oocyte senescence alters the shape and function, thereby weakening the fertilization potential. Nicotinamide mononucleotide (NMN) reverses age-related dysfunctions in various organs. Studies had shown long-term administration of NMN reduced the physiological decline associated in aged mice and reversed the aging of the ovaries. However, the protective effect of NMN on aged porcine oocytes is still unclear. In this study, we investigated the effects of NMN on aging porcine oocytes and subsequent embryonic development. We established a model of senescence of porcine oocytes after ovulation by extending the culture time in vitro. NMN supplementation significantly reduced reactive oxygen species (ROS) levels in senescence oocytes and increased the mRNA levels of antioxidant genes SOD1 and Cat. The mitochondrial membrane potential of aged oocytes treated with NMN was increased compared with that of untreated oocytes. In addition, the mRNA level of apoptosis-related gene Bax was significantly decreased in senescence oocytes treated with NMN, while the mRNA level of anti-apoptosis-related gene BCL-2 was significantly increased. Furthermore, NMN supplementation enhanced the subsequent development ability of senescent oocytes during in vitro aging. Compared with untreated senescent oocytes, the blastocyst formation rate and pluripotent genes of senescent oocytes treated with NMN were significantly increased. Taken together, these results suggest that NMN is beneficial for delaying the aging process in porcine oocytes.

Respiratory syncytial virus (RSV) infection can cause life-threatening pneumonia and bronchiolitis, posing a significant threat to human health worldwide, especially to children and the elderly. Currently, there is no specific treatment for RSV infection. The most effective measures for preventing RSV infection are vaccines and prophylactic medications. However, not all population groups are eligible for the approved vaccines or antibody-based preventive medications. Therefore, there is an urgent need to develop novel vaccines and prophylactic drugs available for people of all ages. High-throughput assays that evaluate the efficacy of viral entry inhibitors or vaccine-induced neutralizing antibodies in blocking RSV entry are crucial for evaluating vaccine and prophylactic drug candidates. We developed an efficient entry assay using a lentiviral pseudovirus carrying the fusion (F) protein of type A or B RSV. In addition, the essential parameters were systematically optimized, including the number of transfected plasmids, storage conditions of the pseudovirus, cell types, cell numbers, virus inoculum, and time point of detection. Furthermore, the convalescent sera exhibited comparable inhibitory activity in this assay as in the authentic RSV virus neutralization assay. We established a robust pseudovirus-based entry assay for RSV, which holds excellent promise for studying entry mechanisms, evaluating viral entry inhibitors, and assessing vaccine-elicited neutralizing antibodies against RSV.

Background: Promoting cholesterol reverse transport (RCT) has been proven to be a promising hyperlipidemia therapy since it is more effective for the treatment of atherosclerosis (AS) caused by hyperlipidemia. Liver X receptor (LXR) agonists can accelerate RCT, but most of them trigger undesirable liver steatosis due to the activation of liver LXRα.Aim: We aim to figure out whether isochlorogenic acid C (ICAC) facilitates RCT without causing hepatic steatosis.Methods:In vitro study, we established foam macrophages and macrophages with loaded NBD-cholesterol models to investigate the competence of RCT promoting ICAC. RT-qPCR and Western blot were used to verify ICAC’s regulation of RCT and NF-κB inflammatory pathways. In this in vivo study, male 6-week-old C57BL/6 mice were fed a high-fat diet (HFD) to investigate ICAC’s anti-hyperlipidemic effect and its functions in regulating RCT. The anti-hyperlipidemic effect of ICAC was evaluated by blood and liver lipid levels, liver hematoxylin, oil red o staining, and liver coefficient. Finally, mRNA levels of genes involved in RCT and inflammation pathways in the liver and intestine were detected by RT-qPCR.Results: ICAC prevented macrophages from foaming by up-regulating the LXRα mediated RCT pathway and down-regulating expression of the cholesterol absorption genes LDLR and CD36, as well as suppressing iNOS, COX2, and IL-1β inflammatory factors. In HFD-fed mice, ICAC significantly lowered the lipid level both in the serum and the liver. Mechanistic studies showed that ICAC strengthened the RCT pathway in the liver and intestine but didn’t affect liver LXRα. Furthermore, ICAC impeded both adipogenesis and the inflammatory response in the liver.Conclusion: ICAC accelerated RCT without affecting liver LXRα, thus resulting in a lipid-lowering effect without increasing liver adipogenesis. Our results indicated that ICAC could be a new RCT promoter for hyperlipidemia treatment without causing liver steatosis.

Paleolatitude evolution could provide a general paleo-location framework for explaining the paleoclimate change and tectonic deformation in geological time. Strengthening the paleolatitude study of the Tuotuohe Basin is important for understanding the history and mechanism of the tectonic uplift process in the north-central Tibetan Plateau. In this study, we introduced the magnetostratigraphy for the Tuotuohe-D (TTH-D) section in the Tuotuohe Basin, central-northern Tibetan Plateau, in order to constrain the chronology and to reconstruct the paleolatitude of the basin during the deposition of the Tuotuohe Formation. The results indicated that the Tuotuohe Formation in the TTH-D section was deposited between 38.5 and ~36.7 Ma. Combining this age with the results from the Tuotuohe section indicates that the age of the Tuotuohe Formation spans the interval from >38.5 Ma to ~33 Ma. Additionally, other paleomagnetic data of the Tuotuohe Formation from the Tuotuohe section, combined with the data from this study, indicate that the paleolatitude of the Tuotuohe Basin during the late Eocene was 25.9 ± 4.2°. That means that the Tuotuohe Basin was located in a subtropical anticyclonic zone and that the paleoenvironment during the late Eocene might be controlled by subtropical high pressure. Additionally, paleomagnetic results from the Qiangtang terrane and the bordering regions are combined with the results of our study, which suggest that the paleolatitude of the Tuotuohe Basin at ~26 Ma coincides well with the Eurasian apparent polar wander path for that interval, and that the N-S India–Asia convergence was reduced or ceased at ~26 Ma in the Tuotuohe Basin.

The timing and intensity of deformation of the Tanggula Range and Tuotuohe Basin are in debatable but strengthening the research on them is necessary for better understanding the geodynamic models of the Tibet and the tectonic-climate connections during the Cenozoic. Here we present the anisotropy of magnetic susceptibility (AMS) records from the foreland Tuotuohe Basin to understand the tectonic uplift history of the Tanggula Range for the interval of ∼37–19.7 Ma. Rock magnetic analyses indicate that the main magnetic carrier is paramagnetic and the minor is hematite. Thus, the AMS in the Tuotuohe Section (TS) can be used to track the tectonic strain related to tectonic processes. The distribution direction of the maximum principle susceptibility (Kmax) is approximately NW–SE, which intersects at about 10°with the strike of the strata and is nearly parallel to the Tanggula thrust fault system, whereas the minimum principle susceptibility (Kmin) is oriented in the NE–SW direction with a girdle distribution. This is consistent with the sedimentary fabric imprinted by the NE–SW shortening direction, representing the earliest deformation stage. AMS data in the TS can be divided into two phases at 30 Ma. From ∼37 to 30 Ma, the AMS parameters indicate an active tectonic strain and the Tanggula Range uplift induced by the India–Asia convergence, whereas between 30 and 19.7 Ma, the strain is weak compared with the earlier phase. Finally, the weak strain environment between 30 and 19.7 Ma indicated by the AMS indexes of the TS and the paleoelevation evidence from the Tuotuohe Basin all together suggest that an additional 1000–2000 m of surface uplift of the basin may have begun after 19.7 Ma.

In recent decades, the increasing incidence of depression has contributed to an increase in the use of serotonergic drugs, such as antidepressants, which predisposes humans to serotonin syndrome. Serotonin syndrome is caused by elevated serotonin levels in the central and peripheral nervous systems. It has been well documented that the development of offspring can be affected by maternal exposure to environmental challenges, such as stress, diseases, or an unhealthy diet during pregnancy. Serotonin, also called 5-hydroxytryptamine (5-HT), is widely expressed in the female reproductive system and plays an important role in the development of follicles and embryos. However, whether the suffering of the mother from serotonin syndrome before pregnancy affects fetal development is still uncertain. In the present study, to explore the effect of maternal prepregnancy 5-HT exposure on the fetus, intraperitoneal injection of 5-HT was used to change maternal prepregnancy 5-HT levels. It was found that maternal prepregnancy 5-HT exposure significantly reduced the body weight and liver weight and the levels of estrogen and progesterone in female mice. Although there was no significant difference in the cleavage rate and blastocyst rate between the 5-HT and control groups, maternal prepregnancy 5-HT exposure increased the percentage of embryo resorption, decreased placental weight, and led to placental inflammation at E13.5. Notably, 5-HT exposure caused weight loss in the offspring at 2 weeks. These results suggested that maternal prepregnancy 5-HT exposure could affect the development of the offspring, which was partly caused by reduced hormonal secretion and placental inflammation.

RGBD salient object detection, based on the convolutional neural network, has achieved rapid development in recent years. However, existing models often focus on detecting salient object edges, instead of objects. Importantly, detecting objects can more intuitively display the complete information of the detection target. To take care of this issue, we propose a RGBD salient object detection method, based on specific object imaging, which can quickly capture and process important information on object features, and effectively screen out the salient objects in the scene. The screened target objects include not only the edge of the object, but also the complete feature information of the object, which realizes the detection and imaging of the salient objects. We conduct experiments on benchmark datasets and validate with two common metrics, and the results show that our method reduces the error by 0.003 and 0.201 (MAE) on D3Net and JLDCF, respectively. In addition, our method can still achieve a very good detection and imaging performance in the case of the greatly reduced training data.

Reported herein is the atomic layer deposition (ALD) of novel ternary ZnCoxOy films possessing p-type semiconducting behavior. The preparation comprises of optimized ZnO and Co3O4 deposition in sub-cycles using the commercially available precursors cyclopentadienylcobalt dicarbonyl (CpCo(CO)2), diethylzinc (DEZ) and ozone (O3). A systematic exploration of the film’s microstructure, crystallinity, optical properties and electrical properties was conducted and revealed an association with Zn/Co stoichiometry. The noteworthy results include the following: (1) by adjusting the sub-cycle of ZnO/ Co3O4 to 1/10, a spinel structured ZnCoxOy film was grown at 150 °C, with it exhibiting a smooth surface, good crystallinity and high purity; (2) the material transmittance and bandgap decreased as the Co element concentration increased; (3) the ZnCoxOy film is more stable than its p-type analog Co3O4 film; and (4) upon p-n diode fabrication, the ZnCoxOy film demonstrated good rectification behaviors as well as very low and stable reverse leakage in forward and reverse-biased voltages, respectively. Its application in thin film transistors and flexible or transparent semiconductor devices is highly suggested.

The influence of a commercial coupling agent on the dielectric properties and microstructure of ceramic-polymer nanocomposites are studied. Free-standing BaTiO _3 -P(VDF-CTFE) films were fabricated using a simple solution-casting process, and the good wettability between fillers and polymer matrix was obtained by using 3-Aminopropyltriethoxysilane as coupling agent. It is found that the coating of a small amount of coupling agent on the surface of BaTiO _3 fillers results in an enhanced dielectric constant, a higher breakdown strength and a larger energy-storage efficiency. When an excessive amount of coupling agent was used, the non-attached coupling agent molecules participated in the complex reactions and result in the aggregation of fillers and the reduction of dielectric constant. It is also found that the surface modification of fillers has a complicated influence on its dielectric behavior which leads to an increase in the dielectric loss of the nanocomposites. When an appropriate amount of coupling agent is coated on fillers, the energy storage density of the nanocomposite is improved due to the enhanced dielectric constant and higher breakdown strength. A maximal discharged energy-storage density of about 4.0 J cm ^−3 was obtained from the nanocomposite film containing 15 vol% of surface modified BaTiO _3 fillers with 1 wt% of KH550, which is about 3 times of that for the nanocomposite without coupling agent.

Currently, the climatic implications associated with the Cenozoic tectonic history and growth mechanisms of the Tibetan Plateau lack consensus and remain controversial. This is due in part to chronological uncertainties and few paleoelevation records distributed in the central to northern Tibetan Plateau, which we address here with the development of a robust chronology (using magnetostratigraphy, biostratigraphy, detrital zircons, and regional radiochronologic dating) and a paleoelevation reconstruction for the Tuotuohe Basin (central-northern Tibet). We refined the age of the Tuotuohe Formation (37-33 Ma), Yaxicuo Formation (33-23.6 Ma), and Wudaoliang Formation (23.6-19.7 Ma). We estimated early Oligocene (ca. 29 Ma) paleotemperatures of the Tuotuohe Basin from 11 °C to 16.1 °C, which correspond to paleoelevations of 2.9 km (±0.4 km) when the relative humidity is 64% and 2.5 km (±0.5 km) when this value is 75%, using various methods including ostracod assemblages, gastropods, charophytes, branched glycerol dialkyl glycerol tetraether analysis, regional empirical formulas, and climate model simulation. Paleoelevation data and existing geological evidence in the vicinity indicate that late Eocene to late Oligocene uplift was associated with upper-crustal shortening. Since the middle Miocene, uplift has been associated with convective removal of lithospheric mantle and/or lower-crustal flow beneath the Hoh Xil Basin. [ABSTRACT FROM AUTHOR]

Abstract Vascularization plays a significant role in promoting the expedited process of bone regeneration while also enhancing the stability and viability of artificial bone implants. Although titanium alloy scaffolds were designed to mimic the porous structure of human bone tissues to facilitate vascularization in bone repair, their biological inertness restricted their broader utilization. The unique attribute of Metal-organic framework (MOF) MIL-53(Fe), known as “breathing”, can facilitate the efficient adsorption of extracellular matrix proteins and thus provide the possibility for efficient interaction between scaffolds and cell adhesion molecules, which helps improve the bioactivity of the titanium alloy scaffolds. In this study, MIL-53(Fe) was synthesized in situ on the scaffold after hydrothermal treatment. The MIL-53(Fe) endowed the scaffold with superior protein absorption ability and preferable biocompatibility. The scaffolds have been shown to possess favorable osteogenesis and angiogenesis inducibility. It was indicated that MIL-53(Fe) modulated the mechanotransduction process of endothelial cells and induced increased cell stiffness by promoting the adsorption of adhesion-mediating extracellular matrix proteins to the scaffold, such as laminin, fibronectin, and perlecan et al., which contributed to the activation of the endothelial tip cell phenotype at sprouting angiogenesis. Therefore, this study effectively leveraged the intrinsic “breathing” properties of MIL-53 (Fe) to enhance the interaction between titanium alloy scaffolds and vascular endothelial cells, thereby facilitating the vascularization inducibility of the scaffold, particularly during the sprouting angiogenesis phase. This study indicates that MIL-53(Fe) coating represents a promising strategy to facilitate accelerated and sufficient vascularization and uncovers the scaffold-vessel interaction from a biomechanical perspective. Abstract Background Prostate cancer (PCa) has a high incidence in men worldwide, and almost all PCa patients progress to the androgen-independent stage which lacks effective treatment measures. PTENP1, a long non-coding RNA, has been shown to suppress tumor growth through the rescuing of PTEN expression via a competitive endogenous RNA (ceRNA) mechanism. However, PTENP1 was limited to be applied in the treatment of PCa for the reason of rapid enzymatic degradation, poor intracellular uptake, and excessively long base sequence to be synthesized. Considering the unique advantages of artificial nanomaterials in drug loading and transport, black phosphorus (BP) nanosheet was employed as a gene-drug carrier in this study. Results The sequence of PTENP1 was adopted as a template which was randomly divided into four segments with a length of about 1000 nucleotide bases to synthesize four different RNA fragments as gene drugs, and loaded onto polyethyleneimine (PEI)-modified BP nanosheets to construct BP-PEI@RNA delivery platforms. The RNAs could be effectively delivered into PC3 cells by BP-PEI nanosheets and elevating PTEN expression by competitive binding microRNAs (miRNAs) which target PTEN mRNA, ultimately exerting anti-tumor effects. Conclusions Therefore, this study demonstrated that BP-PEI@RNAs is a promising gene therapeutic platform for PCa treatment. Graphical Abstract

Abstract Introduction The treatment of hepatitis C has entered the pan-genotypic era, but the effectiveness is not good for the genotype 3b patients who have a large proportion in China. The guidelines for hepatitis C recommend the use of gene-specific regimens when the regional 3b prevalence rate greater than 5%. This study is to explore rationality of this proportion and the cost-effectiveness to implement pan-genotypic regimens in China. Methods A decision Markov model was developed from the health system perspective to evaluate the effectiveness and cost-effectiveness between pan-genotypic and gene-specific treatment regimens for hepatitis C patients. Additionally, we set a regional genotype 3b patient proportion of 0–100% to explore at which proportion it is necessary to perform genotype identification and typing therapy on patients. Model parameters were derived from published literature and public databases. Effectiveness was measured by cured patient numbers, newly diagnosed cases of decompensated cirrhosis, hepatocellular carcinoma, need for liver transplantation, and quality-adjusted life years (QALYs). Cost-effectiveness outcomes included costs and the incremental cost-effectiveness ratio (ICER). The 1–3 times 2022 Chinese per capita gross domestic product was used as the willingness-to-pay threshold. One-way and probabilistic sensitivity analyses were performed to assess the uncertainty of the model parameters. Results Compared with gene-specific regimens, pan-genotypic regimens resulted in an additional 0.13 QALYs and an incremental cost of $165, the ICER was $1,268/QALY. From the view of efficacy, the pan-genotypic regimens cured 5,868 more people per 100,000 patients than gene-specific regimens, avoiding 86.5% of DC cases, 64.6% of HCC cases, and 78.2% of liver transplant needs. Identifying 3b patients before treatment was definitely cost-effectiveness when their prevalence was 12% or higher. The results remained robust in sensitivity analyses. Conclusions In China, the prioritized recommendation of pan-genotypic therapeutics proves to be both cost-effective and efficacious. But, in regions where the prevalence of genotype 3b exceeds 12%, it is necessary to identify them to provision of more suitable therapies.

Abstract Background Hypoxia contributes to cancer progression through various molecular mechanisms and hepatocellular carcinoma (HCC) is one of the most hypoxic malignancies. Hypoxia-inducible gene domain protein-1a (HIGD1A) is typically induced via epigenetic regulation and promotes tumor cell survival during hypoxia. However, the role of HIGD1A in HCC remains unknown. Methods HIGD1A expression was determined in 24 pairs of human HCC samples and para-tumorous tissues. Loss-of-function experiments were conducted both in vivo and in vitro to explore the role of HIGD1A in HCC proliferation and metastasis. Results Increased HIGD1A expression was found in HCC tissues and cell lines, which was induced by hypoxia or low-glucose condition. Moreover, HIGD1A knockdown in HCC cells arrested the cell cycle at the G2/M phase and promoted hypoxia-induced cell apoptosis, resulting in great inhibition of cell proliferation, migration, and invasion, as well as tumor xenograft formation. Interestingly, these anti-tumor effects were not observed in normal hepatocyte cell line L02. Further, HIGD1A knockdown suppressed the expression of ornithine decarboxylase 1 (ODC1), a rate-limiting enzyme of polyamine metabolism under c-Myc regulation. HIGD1A was found to bind with the c-Myc promoter region, and its knockdown decreased the levels of polyamine metabolites. Consistently, the inhibitory effect on HCC phenotype by HIGD1A silencing could be reversed by overexpression of c-Myc or supplementation of polyamines. Conclusions Our results demonstrated that HIGD1A activated c-Myc–ODC1 nexus to regulate polyamine synthesis and to promote HCC survival and malignant phenotype, implying that HIGD1A might represent a novel therapeutic target for HCC.
