In recent years, water (R718) as a kind of natural refrigerant--which is environmentally-friendly, safe and cheap--has been reconsidered by scholars. The systems of using water as the refrigerant, such as water vapor compression refrigeration and heat pump systems run at sub-atmospheric pressure. So, the research on water boiling heat transfer at sub-atmospheric pressure has been an important issue. There are many research papers on the evaporation of water, but there is a lack of data on the characteristics at sub-atmospheric pressures, especially lower than 3 kPa (the saturation temperature is 24 °C). In this paper, the experimental research on water boiling heat transfer on a horizontal copper rod surface at 1.8-3.3 kPa is presented. Antiplatelet aggregation agents have demonstrated clinical benefits in the treatment of ischemic stroke. In our study, a series of novel nitric oxide (NO)-donating ligustrazine derivatives were designed and synthesized as antiplatelet aggregation agents. They were evaluated for the inhibitory effect on 5′-diphosphate (ADP)-induced and arachidonic acid (AA)-induced platelet aggregation in vitro. The results showed that compound 15d displayed the best activity in both ADP-induced and AA-induced assays, and compound 14a also showed quite better activity than ligustrazine. The preliminary structure-activity relationships of these novel NO-donating ligustrazine derivatives were discussed. Moreover, these compounds were docked with the thromboxane A2 receptor to study the structure-activity relationships. The potential for off-target mutations is a critical concern for the therapeutic application of CRISPR-Cas9 gene editing. Current detection methodologies, such as GUIDE-seq, exhibit limitations in oligonucleotide integration efficiency and sensitivity, which could hinder their utility in clinical settings. To address these issues, we introduce OliTag-seq, an in-cellulo assay specifically engineered to enhance the detection of off-target events. OliTag-seq employs a stable oligonucleotide for precise break tagging and an innovative triple-priming amplification strategy, significantly improving the scope and accuracy of off-target site identification. This method surpasses traditional assays by providing comprehensive coverage across various sgRNAs and genomic targets. Our research particularly highlights the superior sensitivity of induced pluripotent stem cells (iPSCs) in detecting off-target mutations, advocating for using patient-derived iPSCs for refined off-target analysis in therapeutic gene editing. Furthermore, we provide evidence that prolonged Cas9 expression and transient HDAC inhibitor treatments enhance the assay's ability to uncover off-target events. OliTag-seq merges the high sensitivity typical of in vitro assays with the practical application of cellular contexts. This approach significantly improves the safety and efficacy profiles of CRISPR-Cas9 interventions in research and clinical environments, positioning it as an essential tool for the precise assessment and refinement of genome editing applications. Background: The gut microbiota is crucial in the process of radiation enteritis (RE). Asrecommended by Chinese expert consensus, three formulas such as Baitouweng decoction, Gegen Qinlian decoction, and Shaoyao decoction, are used to prevent the same type of RE. Methods: Herein, we aimed to investigate the common pharmaceutical ingredients andmolecular mechanisms of the three formulas on RE from the perspective of gut microbiotathrough multi-bioinformatics analysis and mouse experiment. Results: Interestingly, theimportant active ingredient screened was quercetin in all three herbal formulas, andthedominant targets that participated in RE treatment by targeting gut microbiota regulationwere IL10, STAT1, IL1B, and IL1A. The concentration of quercetin was measured inGegenQinlian decoction > Shaoyao decoction > Baitouweng decoction (P <0.001) usinghighperformance liquid chromatography. Moreover, a total of 40 SPF C57/BL6J (male, 12–14weeks old) mice were assigned to perform the validation experiment with various quercetinintervention doses. Quercetin significantly protected against inflammatory responseinradiation induced-mice. The microbial alterations in the gut caused by quercetinintervention in radiation-induced mice were also explained. We further confirmedthealterations in the relative mRNA expression of the three formulas for the treatment of RE, which have four similar gut microbiota-regulated targets (IL10, STAT1, IL1B, andIL1A). Conclusion: These findings show that quercetin regulated the gut microbiota for REtherapy, a connotation that works on the principle of “different treatments for the samedisease” for RE. Background: Natural killer (NK) cells hold great promise in treating diverse hematopoietic and solid tumors. Despite their availability from peripheral blood and cord blood, stem cell-derived NK cells offer an 'off-the-shelf' solution. Hematopoietic stem and progenitor cells (HSPCs) derived from cord blood pose no risk to the newborn or mother and are virtually ideal sources for NK cell differentiation. Methods: We developed a modified protocol to differentiate HSPCs to NK cells under serum-free conditions using defined factors. The HSPC-derived NK (HSC-NK) cells could be expanded in a K562 feeder cell-dependent manner. Furthermore, using lentivirus transduction, chimeric antigen receptor (CAR)-modified HSPCs could be differentiated into NK cells, leading to the establishment of CAR-NK cells. Results: The efficiency of NK cell differentiation from HSPCs was increased through the simple modulation of osmotic pressure by the addition of sodium chloride or glucose. Furthermore, the hyperosmosis-primed HSC-NK cells exhibited enhanced proliferation capacity and maintained normal functional characteristics, including transcriptome and antitumor efficacy. The optimized protocol yielded approximately 1.8 million NK cells from a single CD34-positive cell within a 28-day cycle, which signifies more than a ten-fold increase in efficiency relative to the conventional methods. This optimized protocol was also suitable for generating CAR-NK cells with high yields compared to standard conditions. Conclusions: The results of this study establish high osmotic pressure as a simple yet powerful adjustment that significantly enhances the efficiency and functionality of HSC-NK cells, including CAR-NK cells. A certain amount of cyanide is present in wastewater of various industrial processes, such as wet extraction of gold, coal processing, electroplating and other industries. In this work, an experimental study regarding transport of cyanide through a dispersion supported liquid membrane was performed. A model was established to describe the reaction and transport of CN(I) in the supported liquid membrane and the mass transfer kinetics equations were deduced. Through mass transfer kinetic equation it was derived that, when the carrier concentration was under certain conditions, there was a linear relationship between the reciprocal of the permeability coefficient of CN(I) (1/Pc) and n-th power of the concentration of H+ (cnH+), and the parameters Δa(δa/da) and Δo(δ0/d0) could be obtained from the slope and intercept of the straight line. Then the diffusion coefficient do and the diffusion layer thickness δo of the phase interface between the feed phase and membrane phase could be calculated. Factors affecting migration of CN(I) were analyzed, and the stable removal rate of CN(I) was more than 90% with carrier concentration (%TOA) of 2%, feed phase pH of 4, initial CN(I) concentration of 30 mg/L, stirring time of 1 hour, volume ratio of membrane solution to NaOH solution of 2:1, strip phase concentration of 2 mol/L. The results showed that the overall mass transfer rate increased first and then decreased with an increase of TOA concentration, organic-to-strip volume ratio, and strip concentration. The threshold and frequency of action potentials, fast inactivating transient K+ channels (IA) are important regulators of membrane excitability in sensory neurons because of its vital role in the control of the spike onset. In this study, pain behavior tests, QT-RTPCR, immunohistochemical staining, and patch-clamp recording, were used to investigate the role of P2Y2 receptors in pain behaviour. Results In control rats: 1) UTP, an agonist of P2Y2/P2Y4 receptors, caused a significant decrease in the mean threshold intensities for evoking action potentials and a striking increase in the mean number of spikes evoked by TG neurons. 2) UTP significantly inhibited IA and the expression of Kv1.4, Kv3.4 and Kv4.2 subunits in TG neurons, which could be reversed by the P2 receptor antagonist suramin and the ERK antagonist U0126. In ION-CCI (chronic constriction injury of infraorbital nerve) rats: 1) mRNA levels of Kv1.4, Kv3.4 and Kv4.2 subunits were significantly decreased, while the protein level of phosphorylated ERK was significantly increased. 2) When blocking P2Y2 receptors by suramin or injection of P2Y2R antisense oligodeoxynucleotides both led to a time- and dose-dependent reverse of allodynia in ION-CCI rats. 3) Injection of P2Y2 receptor antisense oligodeoxynucleotides induced a pronounced decrease in phosphorylated ERK expression and a significant increase in Kv1.4, Kv3.4 and Kv4.2 subunit expression in trigeminal ganglia. Conclusions Our data suggest that inhibition of P2Y2 receptors leads to down-regulation of ERK-mediated phosphorylation and increase of the expression of IA-related Kv channels in trigeminal ganglion neurons, which might contribute to the clinical treatment of trigeminal neuropathic pain. Background: The Southeast Asian deletion (--SEA) is the most commonly observed mutation among diverse α-thalassemia alleles in Southeast Asia and South China. It is generally argued that mutation --SEA, like other variants causing hemoglobin disorders, is associated with protection against malaria that is endemic in these regions. However, little evidence has been provided to support this claim. Results: We first examined the genetic imprint of recent positive selection on the --SEA allele and flanking sequences in the human α-globin cluster, covering a genomic region spanning ∼410 kb, by genotyping 28 SNPs in a Chinese population consisting of 76 --SEA heterozygotes and 138 normal individuals. The pattern of linkage disequilibrium (LD) and the long-range haplotype test revealed a signature of positive selection. The network of inferred haplotypes suggested a single origin of the --SEA allele. Conclusions: Thus, our data support the hypothesis that the --SEA allele has been subjected to recent balancing selection, triggered by malaria. [ABSTRACT FROM AUTHOR]

Impacts of hot water extraction (HWE), pressurized water extraction (PWE), high-speed shearing homogenization extraction, microwave assisted extraction (MAE), ultrasound assisted extraction (UAE), ultrasound assisted enzymatic extraction, and ultrasound-microwave assisted extraction (UMAE) on physicochemical characteristics and bioactivities of polysaccharides from loquat (Eriobotrya japonica) leaves (LLPs) were investigated. Results showed that the degrees of esterification, contents of phenolics and uronic acids, constituent monosaccharides, apparent viscosities, and molecular weights of LLPs varied by different extraction methods. Bioactivities of LLPs were also significantly affected by different extraction methods. The high molecular weight and high degree of esterification of LLP-W and LLP-P extracted by HWE and PWE, respectively, might contribute to their strong binding capacities. The strong antioxidant activities and inhibitory effects on α-amylase and α-glucosidase were found in LLP-M and LLP-U extracted by MAE and UAE, respectively, which might be attributed to their contents of uronic acids, contents of total phenolics, and molecular weights. The low molecular weights and viscosities of LLP-U and LLP-UM extracted by UMAE might contribute to their strong prebiotic effects. In the present study, freeze drying, hot-air drying, vacuum drying, and microwave drying at the microwave powers of 400, 600, and 800 W, respectively, were utilized to dry loquat leaves for evaluating the effects of different drying techniques on the physicochemical structures and bioactivities of polysaccharides extracted from loquat leaves (LLPs). Results demonstrated that the physicochemical structures and bioactivities of LLPs significantly affected by different drying techniques. The degrees of esterification, molar ratios of constituent monosaccharides, contents of uronic acids, apparent viscosities, and molecular weights of LLPs were varied by different drying techniques. Additionally, LLPs, particularly LLP-M4 which extracted from loquat leaves prepared by microwave drying at the power of 400 W, exerted remarkable in vitro binding capacities, strong inhibitory effects on α-amylase and α-glucosidase, and obvious antioxidant activities. In order to well understand the physicochemical characteristics and biological activities of polysaccharides (OPPs) from different cultivars of okra collected in China, the chemical characteristics, rheological properties, antioxidant activities, in vitro binding properties, and in vitro inhibitory effects on α-glucosidase of polysaccharides from five representative okra cultivars, including 'Lvjian', 'Kalong8', 'Shuiguo', 'Taiwanwufu', and 'Kalong3', were investigated and compared. Results showed that the constituent monosaccharides of OPPs were similar, which composed of rhamnose, galacturonic acid, galactose, and arabinose. However, their weight-average molecular weights varied from 2.76 × 103 to 4.20 × 103 kDa, and from 0.11 × 103 to 0.90 × 103 kDa, respectively. The uronic acids and degrees of esterification of OPPs ranged from 39.32% to 61.68%, and from 21.66% to 30.02%, respectively. OPPs exhibited typical shear-thinning behavior and viscoelastic properties. Furthermore, OPPs exhibited remarkable antioxidant activities, in vitro binding capacities, and inhibitory effects on α-glucosidase, which might be attributed to their relatively high content of uronic acids, high degrees of esterification, and high molecular weights. The sol-gel transition of methylcellulose (MC) solutions in the presence of ortho-methoxycinnamic acid (OMCA) or cetyltrimethylammonium bromide (CTAB) and in the coexistence of OMCA and CTAB was determined by the rheological measurement. It has been found that the sol-gel transition temperature of MC solutions increases linearly with the concentration of either OMCA or CTAB in solution, respectively. However, in the coexistence of OMCA and CTAB, the sol-gel transition temperature of MC solutions remains invariable, independent of the concentration of CTAB in solution. The experimental results show that OMCA has priority to adsorb on the methyl group of MC chains to form polymer-bound aggregates. In particular, these aggregates inhibit the hydrophobic interaction between CTAB and the methyl group of MC chains completely. Taking into account the fact that OMCA is almost insoluble in MC-free solutions but dissolves very well in aqueous MC solutions, we propose the formation of the core-shell architecture prompted by OMCA and the methyl group of MC chains, with the methyl group of MC chains serving as the core and the self-assembly of OMCA molecules serving as the shell. Obviously, the formation of the core-shell structure increases the solubility of OMCA, improves the stability of methyl groups of MC chains at high temperatures and inhibits the hydrophobic interaction between CTAB and the methyl group of MC chains in solution. The abnormal behavior relating to the sol-gel transition of MC solutions in the presence of OMCA or in the coexistence of OMCA and CTAB is therefore explained. Upon UV irradiation, the sol-gel transition temperature of MC solutions in the presence of OMCA, or in the coexistence of OMCA and CTAB, decreases notably. However, the dependence of the sol-gel transition temperature of MC solutions as a function of OMCA concentration, or CTAB concentration in the presence of OMCA, does not change after UV irradiation. Overexpression of the mammalian proapoptotic protein Bax induces cell death in plant and yeast cells. The Bax inihibitor-1 (BI-1) gene rescues yeast and plant from Bax-mediated lethality, Using the Arabidopsis BI-1 (AtBI-1) gene controlled by the GAL1 promoter as a cell death suppressor in yeast, Cdf1 (cell growth defect factor-1) was isolated from Arabidopsis cDNA library. Overexpression of Cdf1 caused cell death in yeast, whereas such an effect was suppressed by co-expression of AtBI-1. The Cdf1 protein fused with a green fluorescent protein was localized in the mitochondria and resulted in the loss of mitochondrial membrane potential in yeast. The Bax-resistant mutant BRM1 demonstrated tolerance against Cdf1-mediated lethality, whereas the Δatp4 strain was sensitive to Cdf1. Arabidopsis (Arabidopsis thaliana) ethylene-responsive element binding protein (AtEBP) gene was isolated as a suppressor of Bax-induced cell death by functional screening in yeast (Saccharomyces cerevisiae). To further examine the cell death suppressive action of AtEBP in plant cells, we established transgenic tobacco (Nicotiana tabacum) plants overexpressing AtEBP as well as transgenic tobacco plants ectopically expressing mouse Bax protein under a dexamethasone-inducible promoter. We prepared the crosses of the selective lines of each transgenic plant, which were evaluated in terms of cell death suppression activity. Results indicate that AtEBP suppressed Bax-induced cell death in tobacco plants, an action also associated with a lowered level of ion leakage. Furthermore, tobacco Bright Yellow-2 cells overexpressing AtEBP conferred resistance to hydrogen peroxide (H2O2) and heat treatments. AtEBP protein localized in the nucleus and functioned as an in vivo transcription activator as confirmed in transient assays and experiments using stable transgenic system. Up-regulation of defense genes was observed in transgenic Arabidopsis plants overexpressing AtEBP. The maize Hm1 gene encoding the NADPH-dependent HC-toxin reductase is capable of detoxifying HC-toxin of fungus Cochliobolus carbonum. Here, we conducted the metabolic and biochemical analysis in transgenic rice plants overexpressing an HC-toxin reductase-like gene in rice (YK1 gene). Methods employing NADPH oxidation and capillary electrophoresis mass spectrometry analysis confirmed that YK1 possessed dihydroflavonol-4-reductase activity in vitro and in vivo. The overexpression of YK1 in both suspension-cultured cells and rice plants increased NAD(H) and NADP(H) levels by causing an increase in NAD synthetase and NAD kinase activities. Activity changes in enzymes that require NAD(P) as coenzymes were also noted in rice cells ectopically expressing YK1, where the cell death caused by hydrogen peroxide and bacterial disease was down-regulated. Thus, a strategy was proposed that the combination of dihydroflavonol-4-reductase activity and the elevated level of NAD(P)H pool may confer the prevention of induced cell death in planta.
