Abstract

Recently, probiotics have been widely applied for the in situ remediation of aquatic water. Numerous studies have proved that probiotics can regulate water quality by improving the microbial community. Nitrogen cycling, induced by microorganisms, is a crucial process for maintaining the balance of the aquatic ecosystem. Nevertheless, the underlying mechanisms by which probiotics enhance water quality in aquatic systems remain poorly understood. To explore the water quality indicators and their correlation with nitrogen cycling-related functional genes, metagenomic analysis of element cycling was performed to identify nitrogen cycling-related functional genes in Coilia nasus aquatic water between the control group (C) and the groups supplemented with probiotics in feed (PF) or water (PW). The results showed that adding probiotics to the aquatic water could reduce the concentrations of ammonia nitrogen (NH4+-N), nitrite (NO2−-N), and total nitrogen (TN) in the water. Community structure analysis revealed that the relative abundance of Verrucomicrobiota was increased from 30 d to 120 d (2.61% to 6.35%) in the PW group, while the relative abundance of Cyanobacteria was decreased from 30 d to 120 d (5.66% to 1.77%). We constructed a nitrogen cycling pathway diagram for C. nasus aquaculture ponds. The nitrogen cycle functional analysis showed that adding probiotics to the water could increase the relative abundance of the amoC_B and hao (Nitrification pathways) and the nirS and nosZ (Denitrification pathways). Correlation analysis revealed that NH4+-N was significantly negatively correlated with Limnohabitans, Sediminibacterium, and Algoriphagus, while NO2−-N was significantly negatively correlated with Roseomonas and Rubrivivax. Our study demonstrated that adding probiotics to the water can promote nitrogen element conversion and migration, facilitate nitrogen cycling, benefit ecological environment protection, and remove nitrogen-containing compounds in aquaculture systems by altering the relative abundance of nitrogen cycling-related functional genes and microorganisms. Abstract

Photoacoustic spectroscopy (PAS) is a highly sensitive optical detection technique for trace gases. However, PAS is sensitive to environmental factors such as temperature and pressure during actual measurement. In this paper, we study the interference characteristics of temperature and pressure in PAS gas detection. The influence of pressure and temperature on PAS is theoretically analyzed based on the basic principle of gas photoacoustic spectroscopy. The effect of different pressure and temperature conditions on the resonance frequency and signal amplitude of PAS is studied through COMSOL software simulation and validated through experiments. Results show that changes in temperature and pressure will affect the resonance frequency and signal amplitude of PAS, thereby impacting the system's sensitivity and accuracy. To reduce the impact of temperature on the PAS sensor, a temperature compensation model was derived based on experimental analysis. Real‐time temperature detection is used to compensate for the photoacoustic signal based on temperature changes, which can improve the accuracy and stability of photoacoustic spectroscopy gas detection. For 5% CO2, the photoacoustic signal and experimental temperature data were measured for 3612 s, and after temperature compensation, the detection limit was 0.0192% at an integration time of 365 s, which is 0.0128% higher than before temperature compensation. The system's normalized noise equivalent absorption coefficient is 3.30 × 10−8 cm−1 WHz−1/2. This research improves the accuracy and reliability of photoacoustic spectroscopy gas detection technology and provides useful references for temperature correction in similar gas detection fields. Abstract

The human myxovirus resistance B (MxB) protein is an interferon-induced restriction factor that fights a wide range of viruses. We previously demonstrated that MxB binds to hepatitis C virus (HCV)-encoded non-structural protein 5A (NS5A) and inhibits HCV infection by impairing the formation of cyclophilin A (CypA)–NS5A complex. However, the molecular details about how the presence of MxB diminishes the binding of NS5A to CypA remain uncovered. In this study, through molecular dynamic simulations and biochemical assays, we characterized that MxB binds to NS5A domain I through its N-terminal and GTPase domains. Specifically, amino acids (aa.) 189–191 and aa. 330–334 within MxB, together with NS5A residues aa. 71–73, are crucial for MxB–NS5A interaction. Furthermore, we predicted the CypA:NS5A and CypA:NS5A:MxB complexes and calculated the per-residue energy decomposition for identified key residues of the CypA–NS5A interface. A 28% decrease in CypA–NS5A binding affinity was observed in the presence of MxB, suggesting a weakened CypA–NS5A association upon binding of MxB to NS5A, which may contribute to the MxB-mediated inhibitory effect on the formation of CypA–NS5A complex. This work provides information for the antiviral mechanism of MxB and may facilitate the discovery of new strategies to combat CypA-dependent viruses.
