To explore the effect of room temperature storage on the flavor quality and biochemical composition of flower and fruit scented black tea, this study compared the sensory flavor quality of flower and fruit scented black tea produced in the years 2019, 2020, 2021 and 2022 and stored at room temperature, and it detected volatile and non-volatile compounds in the tea by headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) and ultra-performance liquid chromatography coupled to orbitrap high resolution mass spectrometry (UPLC-Orbitrap-MS). The results showed that the sensory quality of flower and fruit scented black tea was significantly affected by storage at room temperature, and the tea lost its floral and fruity aroma characteristics after three years of storage at room temperature; its quality characteristic was mature sour taste. The tea samples were divided into four groups of age by principal component analysis (PCA) and hierarchical cluster analysis (HCA). Totally 15 volatile differential compounds such as linalool, cis-β-ocimene, and hexanoic acid (VIP > 1 and P 2 and P < 0.05) were selected by partial least squares discriminant analysis (PLS-DA) and one-way analysis of variance (ANOVA). The content of volatile compounds such as linalool, hexanal, cis-β-ocimene, and 2-pentylfuran decreased with increasing storage time, while the contents of hexanoic acid, dihydroactinidolide 1-ethyl-2-formyl-1H-pyrrole, and β-ionone increased. The contents of non-volatile compounds such as most amino acids, nucleotides, sugars, tea polyphenols tended to decrease with increasing storage time, while the contents of organic acids and lipids showed an increasing trend. The results obtained from this study can provide a scientific basis for elucidating the quality changes of flower and fruit scented black tea during storage and help guide its rational storage and scientific consumption.

Transforming growth factor (TGF)-β1 and mesenchymal stromal cells (MSCs) are two effective immunosuppressive agents for organ transplantation technology. This study aims to explore the molecular mechanism of TGF-β1-overexpressed MSCs on T cell immunosuppression. To achieve that, BM-MSCs were isolated from canine bone marrow, and their osteogenic differentiation and surface markers were detected. The TGF-β1 gene was transferred into lentivirus and modified MSCs (TGF-β1/MSCs) by lentivirus transfection. Furthermore, TGF-β1/MSCs were co-cultured with T cells to investigate their effect on differentiation and immune regulation. Results showed that TGF-β1/MSCs significantly downregulated the proportion of CD4+ CD8+ T cells in lymphocytes and significantly upregulated the proportion of CD4+ CD25+ T cells. Moreover, TGF-β1/MSCs significantly upregulated the expression of IL-10 in CD4+ T cells and downregulated the expression of IL-17A, IL-21, and IL-22. Meanwhile, interferon-γ (IFN-γ) neutralizing antibody blocked the effects of TGF-β1/MSCs on the differentiation inhibition of Th17. Overall, our results confirm the strong immunosuppressive effect of TGF-β1/MSCs in vitro and demonstrate that IFN-γ mediates the immunosuppressive effect of TGF-β1/MSC.

BACKGROUND: Intelligent monitoring of fixation quality is a prerequisite for automated green tea processing. To meet the requirements of intelligent monitoring of fixation quality in large‐scale production, fast and non‐destructive detection means are urgently needed. Here, smartphone‐coupled micro near‐infrared spectroscopy and a self‐built computer vision system were used to perform rapid detection of the fixation quality in green tea processing lines. RESULTS: Spectral and image information from green tea samples with different fixation degrees were collected at‐line by two intelligent monitoring sensors. Competitive adaptive reweighted sampling and correlation analysis were employed to select feature variables from spectral and color information as the target data for modeling, respectively. The developed least squares support vector machine (LS‐SVM) model by spectral information and the LS‐SVM model by image information achieved the best discriminations of sample fixation degree, with both prediction set accuracies of 100%. Compared to the spectral information, the image information‐based support vector regression model performed better in moisture prediction, with a correlation coefficient of prediction of 0.9884 and residual predictive deviation of 6.46. CONCLUSION: The present study provided a rapid and low‐cost means of monitoring fixation quality, and also provided theoretical support and technical guidance for the automation of the green tea fixation process. © 2022 Society of Chemical Industry. Bone marrow mesenchymal stem cells (BMSCs) have been identified as a potential therapeutic approach to immune-related diseases. Here, we show that BMSC-derived exosomes promote FOXP3 expression and induce the conversion of CD4+ T cells into CD4+CD25+FOXP3+ Treg cells, which is significant for immunosuppressive activity. We found that miR-181a-5p is upregulated in BMSC-derived exosomes and can be transferred to CD4+ T cells. In CD4+ cells, miR-181a directly targets SIRT1 and suppresses its expression. Moreover, downregulated SIRT1 enhances FOXP3 via protein acetylation. In conclusion, our data demonstrated that BMSC-derived exosomal miR-181a is critical in the maintenance of immune tolerance. Furthermore, our results reveal that BMSC-derived exosomal miR-181a induces the production of CD4+CD25+FOXP3+ Treg cells via SIRT1/acetylation/FOXP3. [ABSTRACT FROM AUTHOR]

Objective: To Analysis on psychological pressure of college football players. Method: Theoretical research and interview method. Process: Firstly, the source of psychological stress of college football players is analyzed in the competition. Secondly, a survey of sources of psychological stress and coping strategies are carried out for 21 football players from Sichuan University. Thirdly, the results of psychological stress sources and coping strategies are obtained. Finally, he method of psychological training for soccer players is studied. Result& Analysis: After the investigation, we found that the psychological pressure of college football players is a variety of sources in the competition. Among them, the game's pressure is the most important, accounting for 50%. Most athletes can take positive coping strategies for psychological stress. But there are still close to 40% of athletes to take a negative coping strategies. Result: Colleges and universities should take active and effective psychological training for football players. Background: Numerous deregulated long non-coding RNAs (lncRNAs) accompany the initiation and progression of carcinogenesis. The present study aimed to explore the prognostic significance of lncRNA-SNHG3 on intrahepatic cholangiocarcinoma (ICC) patients., Methods: LncRNA microarray assays were used to evaluate lncRNA expression profiling in three pairs of ICC tissues and adjacent non-tumorous tissues. RT-qPCR was performed to further validate the accuracy of the microarray results., Results: lncRNA microarray and RT-qPCR assays revealed that SNHG3 expression levels were significantly increased in ICC tissues compared to the adjacent non-tumor tissues. A high SNHG3 expression level was significantly correlated with shorter OS in ICC patients. A multivariate regression analysis discovered that SNHG3 could serve as an independent prognostic factor for predicting the OS of ICC patients., Conclusion: We found SNHG3 to be an independent risk factor for predicting the prognosis of ICC.
