Folate metabolism is involved in the development and progression of various cancers. We investigated the association of single nucleotide polymorphisms (SNP) in folate-metabolising genes and their interactions with serum folate concentrations with overall survival (OS) and liver cancer-specific survival (LCSS) of newly diagnosed hepatocellular carcinoma (HCC) patients. We detected the genotypes of six SNP in three genes related to folate metabolism: methylenetetrahydrofolate reductase (MTHFR), 5-methyltetrahydrofolate-homocysteine methyltransferase reductase (MTRR) and 5-methyltetrahydrofolate-homocysteine methyltransferase (MTR). Cox proportional hazard models were used to calculate multivariable-adjusted hazard ratios (HR) and 95 % CI. This analysis included 970 HCC patients with genotypes of six SNP, and 864 of them had serum folate measurements. During a median follow-up of 722 d, 393 deaths occurred, with 360 attributed to HCC. In the fully-adjusted models, the MTRR rs1801394 polymorphism was significantly associated with OS in additive (per G allele: HR = 0·84, 95 % CI: 0·71, 0·99), co-dominant (AG v. AA: HR = 0·77; 95 % CI: 0·62, 0·96) and dominant (AG + GG v. AA: HR = 0·78; 95 % CI: 0·63, 0·96) models. Carrying increasing numbers of protective alleles was linked to better LCSS (HR
10–12 v .2–6 = 0·70; 95 % CI: 0·49, 1·00) and OS (HR10–12 v .2–6 = 0·67; 95 % CI: 0·47, 0·95). Furthermore, we observed significant interactions on both multiplicative and additive scales between serum folate levels and MTRR rs1801394 polymorphism. Carrying the variant G allele of the MTRR rs1801394 is associated with better HCC prognosis and may enhance the favourable association between higher serum folate levels and improved survival among HCC patients. An unprecedented dyotropic rearrangement of β‐lactams has been developed, which provides an enabling tool for the synthesis of structurally diverse γ‐butyrolactams. Unlike the well‐established dyotropic rearrangements of β‐lactones, the present reaction probably proceeds through a dual‐activation mode, and thus displays unusual reactivity and chemoselectivity. The combined computational and experimental results suggest that the dyotropic rearrangement of β‐lactams may proceed through different mechanisms depending on the nature of migrating groups (H, alkyl, or aryl). Hinging on a chemoselective H‐migration dyotropic rearrangement of β‐lactams, we have completed the divergent synthesis of tricyclic marine alkaloids (−)‐lepadiformine A, (+)‐cylindricine C, and (−)‐fasicularin within 11–12 longest linear steps. Background Little is known about the role of vitamin D receptor polymorphisms and their interaction with vitamin D status in hepatocellular carcinoma (HCC) prognosis. Methods We evaluated the association of TaqI , BsmI , Cdx-2, and ApaI polymorphisms, individually and in combination, with liver cancer-specific (LCSS) and overall survival (OS) among 967 patients with newly diagnosed HCC. Subsequently, we examined whether these polymorphisms modified the association between serum bioavailable 25-hydroxyvitamin D (25OHD) concentrations and survival. Cox proportional hazard models were used to calculate hazard ratios (HRs) and 95% confidence intervals (CIs). Results During a median follow-up of 1017 days, 393 deaths occurred, with 360 attributed to HCC. Having TaqI G allele (HR
per allele = 1.30, 95% CI = 1.08 to 1.57) or BsmI T allele (HRper allele = 1.41, 95% CI = 1.01 to 1.99) was associated with worse LCSS. Carrying increasing numbers of protective alleles was associated with superior LCSS (HR6-8 vs 0-3 = 0.52, 95% CI = 0.34 to 0.80). The inverse association of bioavailable 25OHD with LCSS was statistically significant only in patients with TaqI AA (HRQuartile 4 vs Quartile 1 = 0.63, 95% CI = 0.44 to 0.92), BsmI CC (HRQuartile 4 vs Quartile 1 = 0.62, 95% CI = 0.44 to 0.88), and 6 to 8 protective alleles (HRQuartile 4 vs Quartile 1 = 0.45, 95% CI = 0.23 to 0.87). Similar associations were observed for OS. 15. TYK2 correlates with immune infiltration: A prognostic marker for head and neck squamous cell carcinoma

Tyrosine kinase 2 (TYK2) is a member of the Janus kinase (JAK) family and is involved in immune and inflammatory signaling. TYK2 is overexpressed in several types of cancers and promotes the invasion and proliferation of cancer cells. Nevertheless, the roles of TYK2 in the prognosis and immune infiltration of head and neck squamous cell carcinoma (HNSCC) remain to be elucidated. In this study, the expression of TYK2 in HNSCC was evaluated based on the data retrieved from multiple databases and quantitative real-time polymerase chain reaction (qRT-PCR) analysis. The prognostic potential of TYK2 in patients with HNSCC was analyzed by Kaplan-Meier curves and Cox regression analysis. A TYK2-related risk assessment model was subsequently constructed by Least Absolute Shrinkage and Selection Operator (LASSO) regression analysis and stepwise multivariate Cox regression analysis. The association between the expression of TYK2 and the tumor immune microenvironment, immune checkpoints, and drug sensitivity was explored various packages in R. Cell function assays were finally used for exploring the effects of TYK2 on the growth and metastasis of HNSCC tumors. The expression of TYK2 was significantly upregulated in HNSCC and was found to be closely correlated with HPV status, gender, clinical grade, and TP53 mutation status. Survival analysis suggested that TYK2 is associated with better survival outcomes and acts as an independent prognostic indicator of HNSCC. The model constructed herein also performed well in terms of predicting patient prognosis. The expression of TYK2 was positively associated with the population of tumor-infiltrating immune cells, expression of immune checkpoint genes, and antitumor drug susceptibility. Functionally, TYK2 knockdown significantly promoted the proliferation, migration, and invasion of HNSCC cell lines in vitro. The findings demonstrated that TYK2 could serve as a suppressor of tumor growth and holds significant promise as a novel biomarker for assessing the prognosis of patients with HNSCC and aid in immunotherapy against HNSCC.

Types of ncRNAs include microRNAs (miRNAs), circular RNAs, and long ncRNAs. miRNAs are about 21 nucleotides long and form a broad class of post-transcriptional regulators of gene expression that affect numerous developmental and physiological processes in eukaryotes. They usually act as negative regulators of mRNA expression through complementary binding sequences in the 3â-UTR of the target mRNA, leading to translation inhibition and target degradation. In recent years, the importance of ncRNA in oral lichen planus (OLP), particularly miRNA, has attracted extensive attention. However, the biological functions of miRNAs and their mechanisms in OLP are still unclear. In this review, we discuss the role and function of miRNAs in OLP, and we also describe their potential functional roles as biomarkers for the diagnosis of OLP. Background: Head and neck squamous cell carcinoma (HNSCC) is one of the most common and highly heterogeneous malignancies worldwide. Increasing studies have proven that hypoxia and related long non‐coding RNA (lncRNA) are involved in the occurrence and prognosis of HNSCC. The goal of this work is to construct a risk assessment model using hypoxia‐related lncRNAs (hrlncRNAs) for HNSCC prognosis prediction and personalized treatment. Methods: Transcriptome expression matrix, clinical follow‐up data, and somatic mutation data of HNSCC patients were obtained from The Cancer Genome Atlas (TCGA). We used co‐expression analysis to identify hrlncRNAs, then screened for differentially expressed lncRNAs (DEhrlncRNAs), and paired these DEhrlncRNAs. The risk model was established through univariate, least absolute shrinkage and selection operator (LASSO), and stepwise multivariate Cox regression. Finally, we assessed the model from multiple perspectives of tumor mutation burden (TMB), tumor immune infiltration, chemotherapeutic sensitivity, immune checkpoint inhibitor (ICI), and functional enrichment. Results: The risk assessment model included 14 hrlncRNA pairs. The risk score was observed to be a reliable prognostic factor. The high‐risk patients had an unfavorable prognosis and significant differences from the low‐risk group in TMB and tumor immune infiltration. In the high‐risk patients, the common immune checkpoints were down‐regulated, including CTLA4 and PDCD1, and the sensibility to paclitaxel and docetaxel was higher. The functional enrichment analysis suggested that the low‐risk group was accompanied by activated immune function. Conclusions: The risk assessment model of 14‐hrlncRNA‐pairs demonstrated a promising prognostic prediction for HNSCC patients and can guide personalized clinical treatment. [ABSTRACT FROM AUTHOR]

We observe a signal for the doubly charmed baryon Ξ c c + in the decay mode Ξ c c + → p D + K − to complement the previous reported decay Ξ c c + → Λ c + K − π + in data from SELEX, the charm hadroproduction experiment at Fermilab. In this new decay mode we observe an excess of 5.62 events over a combinatoric background estimated by event mixing to be 1.38 ± 0.13 events. The mixed background has Gaussian statistics, giving a signal significance of 4.8σ. The Poisson probability that a background fluctuation can produce the apparent signal is less than 6.4 × 10 −4 . The observed mass of this state is 3518 ± 3 MeV / c 2 , consistent with the published result. Averaging the two results gives a mass of 3518.7 ± 1.7 MeV / c 2 . The observation of this new weak decay mode confirms the previous SELEX suggestion that this state is a double charm baryon. The relative branching ratio for these two modes is 0.36 ± 0.21 .

We present data from Fermilab experiment E781 (SELEX) on the hadroproduction asymmetry for anti-Lambda_c compared to Lambda_c+ as a function of xF and pt2 distributions for Lambda_c+. These data were measured in the same apparatus using incident pi-, sigma- beams at 600 GeV/c and proton beam at 540 GeV/c. The asymmetry is studied as a function of xF. In the forward hemisphere with xF >= 0.2 both baryon beams exhibit very strong preference for producing charm baryons rather than charm antibaryons, while the pion beam asymmetry is much smaller. In this energy regime the results show that beam fragments play a major role in the kinematics of Lambda_c formation, as suggested by the leading quark picture.

We report new precision measurements of the lifetimes of the {Lambda}{sup +}{sub c} and D{sup 0} from SELEX, the charm hadroproduction experiment at Fermilab. Based upon 1630 {Lambda}{sup +}{sub c} and 10210 D{sup 0} decays we observe lifetimes of {tau}[{Lambda}{sup +}{sub c}]=198.1{+-} 7.0{+-}5.6 fs and {tau}[D{sup 0}]=407.9{+-}6.0{+-}4 .3 fs .

Total cross sections for Sigma- and pi- on beryllium, carbon, polyethylene and copper as well as total cross sections for protons on beryllium and carbon have been measured in a broad momentum range around 600GeV/c. These measurements were performed with a transmission technique adapted to the SELEX hyperon-beam experiment at Fermilab. We report on results obtained for hadron-nucleus cross sections and on results for sigma_tot(Sigma- N) and sigma_tot(pi- N), which were deduced from nuclear cross sections.

We utilize the angle and momentum resolution of our apparatus to study the polarization of 375 GeV/c Sigma(+) hyperons produced by 800 GeV/c protons incident on a Cu target. By examining in detail two of our high statistics data samples, we find evidence for structure in the p(t) dependence of Sigma(+) polarization and are able to extract the x(F) dependence of the Sigma(+) polarization and compare it with x(F) behavior in the Lambda(0) and Xi(-) systems.

We have measured the branching ratio for the hyperon radiative decay ${\mathrm{\ensuremath{\Sigma}}}^{+}$\ensuremath{\rightarrow}p\ensuremath{\gamma} using the Fermilab polarized charged hyperon beam. This measurement and our previously published result on the asymmetry parameter in the same decay are part of Fermilab experiment E761. We find B(${\mathrm{\ensuremath{\Sigma}}}^{+}$\ensuremath{\rightarrow}p\ensuremath{\gamma})/B(${\mathrm{\ensuremath{\Sigma}}}^{+}$\ensuremath{\rightarrow}p${\mathrm{\ensuremath{\pi}}}^{0}$) to be [2.32\ifmmode\pm\else\textpm\fi{}0.11(stat)\ifmmode\pm\else\textpm\fi{}0.10(syst)]\ifmmode\times\else\texttimes\fi{}${10}^{\mathrm{\ensuremath{-}}3}$ with a sample of 31 901 events. The higher statistics and careful attention to systematic uncertainties make these significant improvements over previous measurements. We describe how our measurements were performed and briefly review the theoretical implications of these results.

We have measured the branching ratio for the hyperon radiative decay [Xi][sup [minus]][r arrow][Sigma][sup [minus]][gamma] from a sample of 211[plus minus]33 events obtained in the polarized 375 GeV/[ital c] charged hyperon beam at Fermilab. We find [ital B]([Xi][sup [minus]][r arrow][Sigma][sup [minus]][gamma]/[Xi][sup [minus]][r arrow][Lambda][sup 0][pi][sup [minus]])=(1.22[plus minus]0.23[plus minus]0.06)[times]10[sup [minus]4] where the quoted errors are statistical and systematic, respectively. We have also obtained an indication that the sign of the asymmetry parameter of this decay is positive.

We have measured the magnetic moments of the SIGMA+ and SIGMABAR- hyperons produced by 800-GeV protons incident on a Cu target. We determine the SIGMA+ magnetic moment to be (2.4613 +/- 0.0034 +/- 0.0040)mu(N) where the uncertainties are statistical and systematic, respectively. In this first measurement we determine the magnetic moment of the SIGMABAR- to be -(2.428 +/- 0.036 +/- 0.007)mu(N). The magnetic moments of the SIGMA+ and SIGMABAR- are consistent with each other in magnitude but opposite in sign as required by CPT invariance.

We find that the SIGMA+ polarization rises with increasing p(t) to a maximum of 16% at p(t) = 1.0 GeV/c and then decreases to 10% at p(t) = 1.8 GeV/c. We compare this SIGMA+ polarization with data at lower energies. The SIGMABAR- polarization has been measured for the first time. It has the same sign as the SIGMA+ but smaller magnitude in a similar kinematical region.

We have measured the asymmetry parameter (${\mathrm{\ensuremath{\alpha}}}_{\ensuremath{\gamma}}$) in the hyperon radiative decay ${\mathrm{\ensuremath{\Sigma}}}^{+}$\ensuremath{\rightarrow}p\ensuremath{\gamma} with a sample of 34 754\ifmmode\pm\else\textpm\fi{}212 events obtained in a polarized charged hyperon beam experiment at Fermilab. We find ${\mathrm{\ensuremath{\alpha}}}_{\ensuremath{\gamma}}$=-0.720\ifmmode\pm\else\textpm\fi{}0.086\ifmmode\pm\else\textpm\fi{}0.045, where the quoted errors are statistical and systematic, respectively.

- Abstract
We report the first observation of a charm-strange meson D(+)(sJ)(2632) at a mass of 2632.5+/-1.7 MeV/c(2) in data from SELEX, the charm hadro-production experiment E781 at Fermilab. This state is seen in two decay modes, D(+)(s)eta and D0K+. In the D(+)(s)eta decay mode we observe a peak with 101 events over a combinatoric background of 54.9 events at a mass of 2635.4+/-3.3 MeV/c(2). There is a corresponding peak of 21 events over a background of 6.9 at 2631.5+/-2.0 MeV/c(2) in the decay mode D0K+. The decay width of this state is17 MeV/c(2) at 90% confidence level. The relative branching ratio Gamma(D0K+)/Gamma(D(+)(s)eta) is 0.14+/-0.06. The mechanism that keeps this state narrow is unclear. Its decay pattern is also unusual, being dominated by the D(+)(s)eta decay mode.

We have searched for the rare hyperon radiative decay Omega- to Xi- gamma, using the Fermilab Proton Center 375 GeV/c charged hyperon beam. Our measurement of the Omega- beam fraction at 13.6 m from production is (4.8 +_ 0.4)10**(-5). No signal was found and we determine a new upper limit of (4.6 x 10**-4) at 90% CL for the Omega- to Xi- gamma branching ratio.

Pion virtual compton scattering (VCS) via the reaction pi(-)e-->pi(-)egamma was observed in the Fermilab E781 SELEX experiment. SELEX used a 600 GeV/c pi(-) beam incident on target atomic electrons, detecting the incident pi(-) and the final state pi(-), electron and gamma. Theoretical predictions based on chiral perturbation theory are incorporated into a Monte Carlo simulation of the experiment and are compared to the data. The number of reconstructed events (=9) and their distribution with respect to the kinematic variables (for the kinematic region studied) are in reasonable accord with the predictions. The corresponding pi(-) VCS experimental cross section is sigma=38.8+/-13 nb, in agreement with the theoretical expectation of sigma=34.7 nb.
