6. PAT-CNN: Automatic Segmentation and Quantification of Pericardial Adipose Tissue from T2-Weighted Cardiac Magnetic Resonance Images

Abstract

Background: Increased pericardial adipose tissue (PAT) is associated with many types of cardiovascular disease (CVD). Although cardiac magnetic resonance images (CMRI) are often acquired in patients with CVD, there are currently no tools to automatically identify and quantify PAT from CMRI. The aim of this study was to create a neural network to segment PAT from T2-weighted CMRI and explore the correlations between PAT volumes (PATV) and CVD outcomes and mortality. Methods: We trained and tested a deep learning model, PAT-CNN, to segment PAT on T2-weighted cardiac MR images. Using the segmentations from PAT-CNN, we automatically calculated PATV on images from 391 patients. We analysed correlations between PATV and CVD diagnosis and 1-year mortality post-imaging. Results: PAT-CNN was able to accurately segment PAT with Dice score/ Hausdorff distances of 0.74 +- 0.03/27.1 +- 10.9~mm, similar to the values obtained when comparing the segmentations of two independent human observers ($0.76 +- 0.06/21.2 +- 10.3~mm$). Regression models showed that, independently of sex and body-mass index, PATV is significantly positively correlated with a diagnosis of CVD and with 1-year all cause mortality (p-value < 0.01). Conclusions: PAT-CNN can segment PAT from T2-weighted CMR images automatically and accurately. Abstract

Assessing the structure and function of the right ventricle (RV) is important in the diagnosis of several cardiac pathologies. However, it remains more challenging to segment the RV than the left ventricle (LV). In this paper, we focus on segmenting the RV in both short (SA) and long-axis (LA) cardiac MR images simultaneously. For this task, we propose a new multi-input/output architecture, hybrid 2D/3D geometric spatial TransformEr Multi-Pass fEature pyRAmid (Tempera). Our feature pyramid extends current designs by allowing not only a multi-scale feature output but multi-scale SA and LA input images as well. Tempera transfers learned features between SA and LA images via layer weight sharing and incorporates a geometric target transformer to map the predicted SA segmentation to LA space. Our model achieves an average Dice score of 0.836 and 0.798 for the SA and LA, respectively, and 26.31 mm and 31.19 mm Hausdorff distances. This opens up the potential for the incorporation of RV segmentation models into clinical workflows.

Abstract

Simple Summary: In this study, the expression, biological function and potential molecular mechanisms of a novel long non-coding RNA 495810 were detected in hepatocellular carcinoma cells. The results demonstrate a significant up-regulation of long non-coding RNA 495810 in hepatocellular carcinoma, and hepatocellular carcinoma patients with higher long non-coding RNA 495810 shows poorer overall survival and disease-free survival than those with lower long non-coding RNA 495810 level. Moreover, long non-coding RNA 495810 overexpression promotes the proliferation and migration, and blocks apoptosis and cell cycle arrest of hepatocellular carcinoma cells; while knock it down get the opposite results. Mechanistically, long non-coding RNA 495810 directly binds and up-regulates fatty acid binding protein 5 expression. In further, it was found that fatty acid binding protein 5 is up-regulated in hepatocellular carcinoma and associates with poor prognosis. More importantly, fatty acid binding protein 5 knockdown reverses the enhanced abilities of proliferation and metastasis induced by long non-coding RNA 495810 overexpression. These results reveal a novel mechanism of hepatocellular carcinoma metastasis guided by long non-coding RNA, providing a potential target for the treatment of liver cancer. Hepatocellular carcinoma (HCC) is one of the malignant tumors with high morbidity and mortality. Long non-coding RNAs (lncRNAs) are frequently dysregulated in human cancers and play an important role in the initiation and progression of HCC. Here, we investigated the expression of a new reported lncRNA495810 in our previous study by analyzing the publicly available datasets and using RT-qPCR assay. The cell proliferation experiment, cell cycle and apoptosis assay, wound healing assay, cell migration assay were used to explore the biological function of lncRNA495810 in HCC. The western blot, RNA pull down and RNA immunoprecipitation (RIP) detection were used to investigate the potential molecular mechanisms of lncRNA495810. The results demonstrated that lncRNA495810 was significantly upregulated in hepatocellular carcinoma and associated with poor prognosis of hepatocellular carcinoma patients. Moreover, it proved that lncRNA495810 promotes the proliferation and metastasis of hepatoma cells by directly binding and upregulating the expression of fatty acid-binding protein 5. These results reveal the oncogenic roles of lncRNA495810 in HCC and provide a potential therapeutic target for HCC. [ABSTRACT FROM AUTHOR]

Abstract

Hepatocellular carcinoma (HCC) is one of the most lethal cancers worldwide, and therapeutic agents for this malignancy are lacking. MicroRNAs play critical roles in carcinogenesis and present tremendous therapeutic potential. Here, we report that microRNA-206 is a robust tumor suppressor that plays important roles in the development of HCC by regulating cell-cycle progression and the cMet signaling pathway. MicroRNA-206 was underexpressed in livers of two HCC mouse models, human individuals bearing HCC, and human HCC cell lines. Combining bioinformatic prediction and molecular and cellular approaches, we identified cMET (Met proto-oncogene), cyclin D1 (CCND1), and cyclin-dependent kinase 6 (CDK6) as functional targets of microRNA-206. By inhibiting expression of cMET, CCND1, and CDK6, microRNA-206 delayed cell-cycle progression, induced apoptosis, and impaired proliferation of three distinct human HCC cell lines. Systemic administration of microRNA-206 completely prevented HCC development in both cMyc and V-Akt murine thymoma viral oncogene homolog 1/neuroblastoma RAS viral oncogene homolog (AKT/Ras) HCC mice, whereas 100% of control mice died from lethal tumor burdens. Conversely, reintroduction of cMet or Cdk6 into livers of cMyc and AKT/Ras HCC mice recovered growth of HCC inhibited by microRNA-206. These results strongly suggested that cMet and Cdk6 were two functional targets that mediated the inhibitory effect of microRNA-206 on the development of HCC. MicroRNA-206 overexpression demonstrated a profound therapeutic effect on HCC in xenograft and cMyc HCC mice.ConclusionIn summary, this study defines a potentially critical role of microRNA-206 in preventing the growth of HCC and suggests its use as a potential therapeutic strategy for this malignancy.
