1. Agrimol B alleviates cisplatin-induced acute kidney injury by activating the Sirt1/Nrf2 signaling pathway in mice

Abstract

Cisplatin (CDDP) is a widely used chemotherapeutic agent that has remarkable antineoplastic effects. However, CDDP can cause severe acute kidney injury (AKI), which limits its clinical application. Agrimol B is the main active ingredient found in Agrimonia pilosa Ledeb and has a variety of pharmacological activities. The effect of agrimol B on CDDP-induced renal toxicity has not been determined. To investigate whether agrimol B has a protective effect against CDDP-induced AKI, we first identify Sirtuin 1 (Sirt1) as a critical target protein of agrimol B in regulating AKI through network pharmacology analysis. Subsequently, the AKI mouse model is induced by administering a single dose of CDDP via intraperitoneal injection. By detecting the serum urea nitrogen and creatinine levels, as well as the histopathological changes, we confirm that agrimol B effectively reduces CDDP-induced AKI. In addition, treatment with agrimol B counteracts the increase in renal malondialdehyde level and the decrease in superoxide dismutase (SOD), catalase and glutathione levels induced by CDDP. Moreover, western blot results reveal that agrimol B upregulates the expressions of Sirt1, SOD2, nuclear factor erythroid2-related factor 2, and downstream molecules, including heme oxygenase 1 and NAD(P)H quinone dehydrogenase 1. However, administration of the Sirt1 inhibitor EX527 abolishes the effects of agrimol B. Finally, we establish a tumor-bearing mouse model and find that agrimol B has a synergistic antitumor effect with CDDP. Overall, agrimol B attenuates CDDP-induced AKI by activating the Sirt1/Nrf2 signaling pathway to counteract oxidative stress, suggesting that this compound is a potential therapeutic agent for the treatment of CDDP-induced AKI.

Abstract

We report the discovery of a peculiar X-ray transient, EP240408a, by Einstein Probe (EP) and follow-up studies made with EP, Swift, NICER, GROND, ATCA and other ground-based multi-wavelength telescopes. The new transient was first detected with Wide-field X-ray Telescope (WXT) on board EP on April 8th, 2024, manifested in an intense yet brief X-ray flare lasting for 12 seconds. The flare reached a peak flux of 3.9x10^(-9) erg/cm2/s in 0.5-4 keV, about 300 times brighter than the underlying X-ray emission detected throughout the observation. Rapid and more precise follow-up observations by EP/FXT, Swift and NICER confirmed the finding of this new transient. Its X-ray spectrum is non-thermal in 0.5-10 keV, with a power-law photon index varying within 1.8-2.5. The X-ray light curve shows a plateau lasting for about 4 days, followed by a steep decay till becoming undetectable about 10 days after the initial detection. Based on its temporal property and constraints from previous EP observations, an unusual timescale in the range of 7-23 days is found for EP240408a, which is intermediate between the commonly found fast and long-term transients. No counterparts have been found in optical and near-infrared, with the earliest observation at 17 hours after the initial X-ray detection, suggestive of intrinsically weak emission in these bands. We demonstrate that the remarkable properties of EP240408a are inconsistent with any of the transient types known so far, by comparison with, in particular, jetted tidal disruption events, gamma-ray bursts, X-ray binaries and fast blue optical transients. The nature of EP240408a thus remains an enigma. We suggest that EP240408a may represent a new type of transients with intermediate timescales of the order of about 10 days. Abstract

In this paper we construct a new family of sets via Diophantine approximation, in which the classical examples are endpoints. Our first application is on their Hausdorff dimension. We show a recent result of Ren and Wang, known sharp on orthogonal projections in the plane, is also sharp on $A+cB$, $c\in C$, thus completely settle this ABC sum-product problem. Higher dimensional examples are also discussed. In addition to Hausdorff dimension, we also consider Fourier dimension. In particular, now for every $0\leq t\leq s\leq 1$ we have an explicit construction in $\mathbb{R}$ of Hausdorff dimension $s$ and Fourier dimension $t$, together with a measure $\mu$ that captures both dimensions. In the end we provide a perspective of Knapp's example and treat our Diophantine approximation as its analog in $\mathbb{R}$, that naturally leads to the sharpness of Mockenhaupt-Mitsis-Bak-Seeger Fourier restriction theorem. These are alternatives of recent examples due to Fraser-Hambrook-Ryou. Abstract

Spectral computed tomography based on a photon-counting detector (PCD) attracts more and more attentions since it has the capability to provide more accurate identification and quantitative analysis for biomedical materials. The limited number of photons within narrow energy bins leads to imaging results of low signal-noise ratio. The existing supervised deep reconstruction networks for CT reconstruction are difficult to address these challenges because it is usually impossible to acquire noise-free clinical images with clear structures as references. In this paper, we propose an iterative deep reconstruction network to synergize unsupervised method and data priors into a unified framework, named as Spectral2Spectral. Our Spectral2Spectral employs an unsupervised deep training strategy to obtain high-quality images from noisy data in an end-to-end fashion. The structural similarity prior within image-spectral domain is refined as a regularization term to further constrain the network training. The weights of neural network are automatically updated to capture image features and structures within the iterative process. Three large-scale preclinical datasets experiments demonstrate that the Spectral2spectral reconstructs better image quality than other the state-of-the-art methods., Comment: Accepted by IEEE TCI

Abstract

Ultrahigh rate performance of active particles used in lithium-ion battery electrodes has been revealed by single-particle measurements, which indicates a huge potential for developing high-power batteries. However, the charging/discharging behaviors of single particles at ultrahigh C-rates can no longer be described by the traditional electrochemical kinetics in such ion-intercalation active materials. In the meantime, regular kinetic measuring methods meet a challenge due to the coupling of interface reaction and solid-state diffusion processes of active particles. Here, we decouple the reaction and diffusion kinetics via time-resolved potential measurements with an interval of 1 ms, revealing that the classical Butler-Volmer equation deviates from the actual relation between current density, overpotential, and Li+ concentration. An interface ion-intercalation model is developed which considers the excess driving force of Li+ (de)intercalation in the charge transfer reaction for ion-intercalation materials. Simulations demonstrate that the proposed model enables accurate prediction of charging/discharging at both single-particle and electrode scales for various active materials. The kinetic limitation processes from single particles to composite electrodes are systematically revealed, promoting rational designs of high-power batteries.

Abstract

Identifying overpotential components of electrochemical systems enables quantitative analysis of polarization contributions of kinetic processes under practical operating conditions. However, the inherently coupled kinetic processes lead to an enormous challenge in measuring individual overpotentials, particularly in composite electrodes of lithium-ion batteries. Herein, the full decomposition of electrode overpotential is realized by the collaboration of single-layer structured particle electrode (SLPE) constructions and time-resolved potential measurements, explicitly revealing the evolution of kinetic processes. Perfect prediction of the discharging profiles is achieved via potential measurements on SLPEs, even in extreme polarization conditions. By decoupling overpotentials in different electrode/cell structures and material systems, the dominant limiting processes of battery rate performance are uncovered, based on which the optimization of electrochemical kinetics can be conducted. Our study not only shades light on decoupling complex kinetics in electrochemical systems, but also provides vitally significant guidance for the rational design of high-performance batteries.

Abstract

Abstract Background Methamphetamine (METH) use, especially in females, has become a growing public health concern in China. In this study, we aimed to characterize the factors that contributed to drug craving in female METH users under isolated compulsory detoxification. We characterized factors contributing to craving such as duration of detoxification, history of drug use and self-reported mood state. Methods Subjects (N=113) undergoing a 1- to 3-year METH detoxification program were recruited from the Zhejiang Compulsory Detoxification Center for Women. The Questionnaire of METH-use Urge (QMU) was used to evaluate the level of craving for METH. The Abbreviate Profile of Mood States (A-POMS) was applied as an assessment for the negative mood disturbances. Results The participants were at a mean age of 25.2, primarily lowly educated and unemployed, and single. Smoking was the only route of METH administration at an average dose of 0.5 g/day, and 4 times/week. The reported craving level was positively correlated with the negative mood disturbances and the weekly dose of METH, but independent of the duration of detoxification. Furthermore, all five aspects of negative mood disturbances, including fatigue, bewilderment, anxiety, depression and hostility, were shown to positively correlate to the self-reported craving level after controlling for weekly dose of METH. Conclusions The data demonstrate a robust correlation between mood distress and craving for METH. Our results call for close evaluation of mood distress in treatment of METH users in China.

Abstract Background To date there has been limited research on both the prevalence of overdose and drug user knowledge about overdose prevention and response methods in China. In addition, there has been no effort to integrate naloxone information and distribution into pre-release services for drug users detained in isolated compulsory detoxification facilities in China. Methods The authors conducted a survey of 279 heroin users in isolated compulsory detoxification centers in Ningbo, China in an attempt to evaluate the possibility of conducting prelease peer naloxone programs in Ningbo isolated compulsory detoxification centers. Respondents' demographic background, history of heroin overdoses, and attitudes/knowledge about overdose prevention and response were collected. Results While drug users in Ningbo's compulsory detoxification centers have limited understandings of how to effectively respond to overdoses, they expressed concern about the possibility of overdose, interest in participating in overdose prevention and response programs, and a willingness to help their peers. In general, there was no significant difference in history and attitudes/knowledge of overdose between male and female participants. Conclusion Based on the findings of this research, our survey provides preliminary evidence that detained drug users have considerable interest in overdose prevention and response information and willingness to help peers. However, drug users in Ningbo isolated compulsory detoxification centers currently have limited understandings of effective ways of helping to prevent overdose deaths.
