Purpose:Innate immunity is an indispensable arm of tumor immune surveillance, and the liver is an organ with a predominance of innate immunity, where mucosal-associated invariant T (MAIT) cells are enriched. However, little is known about the phenotype, functions, and immunomodulatory role of MAIT cells in hepatocellular carcinoma (HCC).Experimental Design: The distribution, phenotype, and function of MAIT cells in patients with HCC were evaluated by both flow cytometry (FCM) and in vitro bioassays. Transcriptomic analysis of MAIT cells was also performed. Prognostic significance of tumor-infiltrating MAIT cells was validated in four independent cohorts of patients with HCC.Results:Despite their fewer densities in HCC tumor than normal liver, MAIT cells were significantly enriched in the HCC microenvironment compared with other mucosa-associated organs. Tumor-derived MAIT cells displayed a typical CCR7−CD45RA−CD45RO+CD95+ effector memory phenotype with lower costimulatory and effector capabilities. Tumor-educated MAIT cells significantly upregulated inhibitory molecules like PD-1, CTLA-4, TIM-3, secreted significantly less IFNγ and IL17, and produced minimal granzyme B and perforin while shifting to produce tumor-promoting cytokines like IL8. Transcriptome sequencing confirmed that tumor-derived MAIT cells were reprogrammed toward a tumor-promoting direction by downregulating genes enriched in pathways of cytokine secretion and cytolysis effector function like NFKB1 and STAT5B and by upregulating genes like IL8, CXCL12, and HAVCR2 (TIM-3). High infiltration of MAIT cells in HCC significantly correlated with an unfavorable clinical outcome, revealed by FCM, qRT-PCR, and multiplex IHC analyses, respectively.Conclusions:HCC-infiltrating MAIT cells were functionally impaired and even reprogrammed to shift away from antitumor immunity and toward a tumor-promoting direction.See related commentary by Carbone, p. 3199

As China is implementing the policy ofquot;Announcement on Ending the Pilot Work of Chinese medicine formula gra-nulesquot;, the standard of Chinese medicine formula granules has gradually become the focus of industry development. Up to now, 196 national drug standards for Chinese medicine formula granules have been published by China, which guaranteed the production quality of Chinese medicine formula granules. However, there are still several challenges such as the rational application of national drug standards and the enrichment and improvement of varieties. The basic content of the issued national drug standards for Chinese medicine formula granules was analyzed and compared with the quality standard provisions of the corresponding decoction pieces in the Chinese Pharmacopoeia(2020 edition) in this paper. This paper discussed the main characteristics of paste-forming rate of each medicinal raw materials,quot;quantity-qualityquot; transformation, equivalent ratio, and so on, and clarified the characteristics of the national standard for Chinese medicine formula granules. This paper provided references for achieving the unified quality control and meeting the overall quality requirements of Chinese medicine formula granules.

To investigate the incidence of Runt-related transcription factor 1 (RUNX1) gene and its associated gene mutations in patients with acute myeloid leukemia (AML), and analyze its clinical characteristics and prognosis.The genomic DNA-PCR method was used to detect the exon of RUNX1 gene, and the gene mutations were analyzed by genetic sequencing. NPM1, DNMT3A, FLT3-ITD, IDH1/2, K/N-RAS, CEPBA, TET2, and WT1 co-mutations were also detected. Patients were followed up to determine efficacy and prognosis.Among 171 patients, the RUNX1 gene mutation was detected in 17 cases, and the mutation rate was 9.9%. The type of RUNX1 gene mutation was 9 missense mutations, 4 frameshift mutations, and 4 nonsense mutations. The peripheral blood leukocyte count of the patients in mutation group was 3 (1-101) ×10AML patients with RUNX1 gene mutation shows unique clinical and biological characteristics, RUNX1 mutation can be regarded as a molecular marker of poor prognosis in AML patients.伴RUNX1基因突变急性髓系白血病患者临床特征的分析.探讨急性髓系白血病（AML）患者中RUNX1基因及其伴随基因突变的发生率，并分析其临床特征及预后。.采用基因组DNA-PCR方法扩增RUNX1基因外显子，应用基因测序分析其基因突变，同时检测NPM1、DNMT3A、FLT3-ITD、IDH1/2、K/N-RAS、CEPBA、TET2、WT1伴随突变的情况，随访患者并判定疗效和预后。.171例AML患者中，17例检测存在RUNX1基因突变，突变率为9.9%。RUNX1基因突变类型为错义突变9例，框移突变4例，无义突变4例，突变组患者初诊时外周血白细胞数[3（1-101）×10伴RUNX1基因突变的AML患者具有独特的临床及生物学特点，RUNX1基因突变可以作为AML患者预后不良的分子学标志。.

Stripe rust, caused by Puccinia striiformis f. sp. tritici , is an important wheat disease in China, seriously threatening wheat production. Understanding the winter survival of the fungus is a key for predicting the spring epidemics of the disease, which determines the crop loss. Estimation of P. striiformis f. sp. tritici winter survival requires processing a large number of samples for sensitive detection of the pathogen in wheat leaf tissue using real-time quantitative reverse transcription PCR (qRT-PCR). A bottleneck for the analysis is the acquisition of a good yield of high quality RNA suitable for qRT-PCR to distinguish dead and alive fungal hyphae inside leaves. Although several methods have been described in the literatures and commercial kits are available for RNA extraction, these methods are mostly too complicated, expensive and inefficient. Thus, we modified three previously reported RNA extraction methods with common and low-cost reagents (LiCl, SDS and NaCl) to solve the problems and selected the best to obtain high quality and quantity RNA for use in qRT-PCR. In the three improved methods, the NaCl method was proven to be the best for extracting RNA from urediniospores of and wheat leaves infected by P. striiformis f. sp. tritici , although the modified LiCl and SDS methods also increased yield of RNA compared to the previous methods. The improved NaCl method has the following advantages: 1) Complete transfer of urediniospores of P. striiformis f. sp. tritici from the mortar and pestle can ensure the initial amount of RNA for the qRT-PCR analysis; 2) the use of low-cost NaCl to replace more expensive Trizol can reduce the cost; 3) the yield and quality of RNA can be increased; 4) the improved method is more suitable for a large number and high quantity of samples from fields. Using the improved NaCl method, the amount of RNA was increased three times from urediniospores of P. striiformis f. sp. tritici compared from the extraction kit. Approximately, 10.11 μg total RNA of high quality was obtained from 100 mg of infected leaves, which was 8.8, 6.5, 3.4 and 2.1 folds of the amounts obtained from the previous LiCl, SDS, NaCl and traditional Trizol methods, respectively. The method could be used to study the overwintering rates of P. striiformis f. sp. tritici over a large region of wheat production for predicting epidemic levels by determining pathogen survival levels after winter. The method can also be used in any studies which need a large number of high quality RNA samples.

The ultra-thin 304 stainless steel sheet will be used in flexible displays for substrate material. In chemical mechanical polishing of ultra-thin stainless steel, the pH value of polishing slurry has an important influence on the material removal rate (MRR). In this paper, the influences of pH regulator in slurry on MRR had been studied in CMP of ultra-thin 304 stainless steel based on alumina (Al2O3) abrasive in acidic polishing slurry. The research results show that, in the same conditions, different pH value had a different chemical action mechanism and a different MRR. And also, different oxidant with pH regulator had a different chemical action mechanism and a different MRR. The research results can provide the reference for studying the slurry in CMP of ultra-thin stainless steel.

Chemical mechanical polishing (CMP) had been considered as the most practical and effective method of achieving an ultra-smooth and non-damage surface in manufacturing SiC crystal substrate. CMP slurry was one of the key factors of CMP technology. In this paper, through investigating the changes of several core factors to evaluate the performance of CMP, such as the material removal rate (MRR), surface roughness Ra, 3D surface profiler, etc., the influence of various slurry and its content on the polishing efficiency and surface finish quality had been studied. The research results showed that different oxidant had different chemical action mechanism, also affecting the stability of CMP slurry and surface quality of specimen; adding suitable an oxidant to slurry could effectively improve the CMP performance.

Ultra-thin stainless steel sheet is likely to be the ideal substrate materials used for flexible display. In order to obtain ultra-thin stainless steel substrate with high accuracy, low surface roughness and without damage, the ultra-precision lapping and chemical mechanical polishing technology must be used. In this paper, the lapping paste for stainless steel has been designed using the orthogonal experiment method. According to the range analysis method, an optimal lapping paste had been obtained. Tests lapping the 304 stainless steel sheet with the optimal lapping paste have been done. The test results show that the MRR is about 240nm/min and the surface rough Ra is about 128 nm. This research results can provide theory support for ultra precision machining the stainless steel.

In this study, according to physic-chemical characteristic of the SiC crystals, we analyzed and researched the role of pH modifier in SiC crystal substrate chemical mechanical polishing and the surface after polishing. We used different polish agents which was made up with inorganic and organic bases to experiment, and then analyzed the results. The parameters of the polishing specimen, such as the removal rate of material, surface scanning and the roughness, were analyzed and used to determine the different pH modifiers played different roles in the stability of polish agents and surface quality of the specimen.

Flexible displays will become the mainstream display of next generation, so the manufacturing technology of the flexible display substrate is one of very important technologies. In this paper, according to orthogonal design, the composition selection and optimization of chemical mechanical polishing (CMP) slurry based on alumina abrasive had been done in CMP ultra-thin stainless steel surface by a lot of tests. The CMP slurry based on alumina abrasive for ultra-thin stainless steel surface had been obtained. According to the test results, the material removal rate (MRR) is about 177 nm/min and the surface roughness (Ra) is about 0.018μm.

Chemical mechanical polishing (CMP) is the most effective technology to achieve ultra-smooth without damage surface in ultra-precision machining of stainless steel substrate. In this paper, according to the slurry ingredients obtained by former research, the influences of the CMP process parameters, such as the rotational velocity of the platen and the carrier, the polishing pressure and the abrasive size on the material removal rate (MRR), have been studied in CMP stainless steel substrate based on the alumina (Al2O3) abrasive. The research results show that the material removal rate increases with the increase of the abrasive size, the rotational velocity of the platen and the polishing pressure significantly and the surface roughness increases with the increase of the abrasive size. This study results will provide the reference for optimizing the process parameters and researching the material removal mechanism in CMP stainless steel sheet.

There is increasing evidence that free radicals play an important role in neuronal damages induced by diabetes mellitus or cerebral ischemia insults. Antioxidants with free radical scavenging activities have been shown to be beneficial and neuroprotective for these pathological conditions. Here, we report free radical scavenging activity and neuroprotective potential of D138, one copper(II)/zinc(II) Schiff-base complex derived from N,N'-2(2-hydroxynaphthylmethylidene)-1,3-propanediamine. The data from three in vitro assays, 2,2-diphenyl-1-picrylhydrazyl assay, nitro blue tetrazolium assay and hydroxyl radical scavenging assay, indicated that D138 presented a potent free radical scavenging activity. The neuroprotective and antioxidative effects of D138 were further evaluated in vivo using bilateral common carotid artery occlusion (BCCAO) mouse model and streptozotocin (STZ) diabetic mouse model. Our results indicated that treatment of D138 significantly ameliorated the hippocampal neuronal damage and the oxidative stress levels in these animal models. Moreover, D138 also reversed the behavioral deficiencies induced by BCCAO or STZ, as assessed by Y-maze test and fear conditioning test. In conclusion, all these findings support that D138 exerts free radical scavenging and neuroprotective activities and has the potentials to be a potent therapeutic candidate for brain oxidative damage induced by cerebral ischemia or diabetes mellitus.

The durability of coastal area bridge structures is more prominent, first, the understanding of the durability of the concrete is insufficient before, and the corresponding measures has not taken; Second, accelerating deterioration of the reinforced concrete is due to the environment and climate factors. Experience has shown that both home and abroad: Starting from the design, take preventive protection measures is effective means to improve the durability of reinforced concrete bridge,then it can get twice the result with half the effort. Based on the Tangshan Coastal Highway construction, considering of the leve of the engineering environmental and so on, on the basis of the service life of the bridge design, the durability of concrete structure would be studied.

In order to reveal the change rule of vibration acceleration during micro-end-milling straight groove, the single-factor and orthogonal experiments are employed to confirm the influence of milling parameters on vibration acceleration. On this basis, the relation equation between milling parameters and vibration accelerations is built. The result show that, the vibration acceleration along the width of groove is bigger than that of feed direction and it will increase rapidly with the improve of cutting speed; the predicted result of vibration acceleration is in good accordance with test result, therefore the relation equation can be used to predict vibration acceleration.

In this paper, according to the slurry ingredients obtained by former research, the influences of the chemical mechanical polishing (CMP) process parameters, such as the rotational velocity of the platen and the carrier, the polishing pressure and the abrasive size on the material removal rate (MRR) and surface roughness Ra have been studied in CMP SiC crystal substrate (0001) C and (0001) Si surface based on the diamond abrasive. The research results show that the material removal rate changes with the change of the abrasive size, the rotational velocity of the platen and the polishing pressure significantly, but the maximum of MRR can be obtained at a certain rotational velocity of platen, abrasive size and polishing pressure. The influence of the abrasive size, the platen velocity, the carrier velocity and the polishing pressure on surface roughness is no significant. Under the same conditions, the MRR of CMP the Si surface is larger than that of the C surface. This study results will provide the reference for optimizing the process parameters and researching the material removal mechanism in CMP SiC crystal substrate.

The bending moment changes of slab column nodes in the structure system are analyzed by applying horizontal load to concrete filled steel tubular column and girderless floor, and the experimental values and the theoretical calculation values are compared and analyzed.According to the consequence of comparison and analysis, the ratio of the test value and the theoretical value of the bending moment at the bottom of the plate of concrete filled steel tubular column ranges from 8% to 17%,and the average of the ratio is 13%, and the deformability of the node is better than the ordinary slab column node of concrete filled steel tubular column.

An ideal anodes used for the electrochemical oxidation of organic wastewater should have excellent activity, stability and high oxygen evolution potential. In this paper the CNT (Carbon Nanotubes)-PbO2 films electrodeposited on stainless steel were prepared. X-ray diffraction (XRD) patterns and SEM images indicated that CNT particles and PbO2 were able to achieve co-deposit and the composite CNT-PbO2 films were compact. The cyclic voltammograms of the CNT-PbO2 films studied in 0.5M H2SO4 at a scan rate of 100 mV/s showed that the CNT-PbO2 film has high electrochemical stability. The results of wastewater treatment indicated that the CNT-PbO2 anodes have excellent activity in ammonia wastewater treatment.

To study the effect of Cynomorium songaricum polysaccharide (CSRP) on A549 cells telomere of human non-small cell lung cancer, the mice were intragastric administrated with CSRP (0.08 g•kg⁻¹) once daily for 4 days. Then their serum was taken for preparing CSRP drug serum. A549 cells were treated by the drug serum, and the effect of drug serum with different concentrations and different treating time on the proliferation of non-small cell lung cancer A549 cells was determined by MTT test. After treating for 48 hours by the drug serum of different concentrations, the telomere length of the cells was determined by fluorescence quantitative polymerase chain reaction (qPCR); the mRNA expression of telomerase reverse transcriptase (TERT) was determined by RT-qPCR; the cells apoptosis was determined by TUNEL assay. The results demonstrated that CSRP of various concentrations could inhibit the proliferation of the lung cancer A549 cells significantly, and the inhibition effect was strongest at 48 hours with the concentration of 6.0 mL•L⁻¹. At 48 h, that CSRP of the concentrations from 1.5 to 12.0 mL•L⁻¹ could significantly shorten the telomere length of A549 cells, and the effect was strongest with the concentration of 1.5 mg•L⁻¹. CSRP of various concentrations could significantly inhibit the mRNA expression of TERT in A549 cells, and the inhibition effect was stronger when the concentration was ≥6.0 mL•L⁻¹. CSRP of various concentrations could promote A549 cells apoptosis, and the effect was stronger when the concentration was ≥6.0 mL•L⁻¹. In conclusion, CSRP has the anti-cancer effect, and the action mechanism may be associated with inhibiting TERT mRNA expression, shortening telomere length, inhibiting cells proliferation and promoting cells apoptosis.

Concrete is one of the most widely used construction material throughout the world. But the properties of concrete under different strain rates differ from each other greatly. In order to investigate the effect of strain rate on concrete compressive mechanical properties, compressive experiments of concrete specimens (C35) are carried out on MTS, with the uniaxial strain rates ranging from 10-5/s to 10-2/s. The compressive mechanical properties of concrete under different stain rates, which include compressive strength, elastic modulus, peak strain and Poisson's ratio are studied systematically. The formulas which can describe the change laws of the compressive properties of concrete under different the strain rates are proposed. The test results show that the compresseive strength and elastic modulus of concrete would increase with the strain rate increasing. The effect of strain rate on peak strain and and Poisson's ratio is not significant. These research achievements can contribute to grasp the dynamic properties and build the dynamic constitutive models of concrete.

In order to improve and simplify the process of electroless Ni-P plating on aluminum substrate, a activated solution and process were optimized by orthogonal test, the plating morphology were analysed using Quanta 200 type scanning electron microscope. The bond between the plating and aluminum substrate were evaluated using method of water quenching and alternating bending test. The results show that good coating can be obtained on aluminum substrate under the normal condition of the acid electroless Ni-P plating solution with sulfuric acid of 120g/L and nitric acid of 10-15g/L as activated solution and activated time of 20 min. And the good bond between the plating and aluminum substrate was obtained.

According to material mechanics theory and drilling practice, the equation of lateral bending deformation (LBD) of drill was built and the inclined angle of drill tip was solved when drilling small hole. And then the drilling test was done and the character of drilling force was analyzed. Based on this, the LBD of drill was studied. The result shows that, the rise of lateral force and thrust force will increase the inclined angle of drill tip in drilling process, and lateral force is the main factor of generating LBD. By choosing suitable axial vibration parameter, axial vibration drilling can reduce the LBD of drill and improve the dimensional accuracy of hole evidently.

Using cement or lime and cement to solidify the local saline soil. The physical and mechanical properties were comparative analyzed of the curing chlorine saline soil with different ratio, through compaction test and CBR test. The best cure programuse is 6% lime and 6% cement for the saline soil.

Based on experiments of common drilling and vibration drilling on duralumin (2A12), the failure mode and failure reason of twist drill are studied theoretically and experimentally. The result shows that, compared with common drilling, the wear of clearance face and rake face decreases evidently during vibration drilling, and the wear of chisel edge and marginal point of twist drill hardly happens. Moreover the probability of drill breakage is very low during vibration drilling. However, if amplitude is bigger, the tipping of chisel edge often appears.

This paper investigates the effect of spindle clamping force on the static and dynamic characteristics of the spindle-tooling system through measuring the frequency response function of the tool end point in variable spindle clamping force. The research shows that the limited static stiffness increases accordingly and normal static stiffness changes little as the spindle champing force increases. The high spindle clamping force leads to reduce the modal damping of the spindle-tooling system while the natural frequency varies slight. The analyses of process stability indicates that the low spindle clamping force can improves the process stability because of the higher damping in some clamping force range.

Insulin resistance and vascular remodeling are prevalent and predict cardiovascular mortality in hemodialysis patients. Angiotensin II (Ang II) may be involved in both pathogenesis. In the present study, we investigated the effects of the Ang II receptor blocker losartan on insulin resistance, arterial stiffness, and carotid artery structure in hemodialysis patients. Seventy-two hemodialysis patients were randomly assigned to receive either losartan 50 mg qd (n = 36) or β-blocker bisoprolol 5 mg qd (n = 36). At the start and at month 12, ambulatory blood pressure (BP) monitoring, aortic pulse wave velocity (PWV) measurements, and carotid artery ultrasound were performed, and homeostasis model assessment index of insulin resistance (HOMA-IR) was determined. During the study period, bioimpedance method was used to evaluate volume status every 3 months. Home-monitored BPs were measured at least monthly. Ambulatory BP decreased significantly and similarly by either losartan or bisoprolol. Decreases in PWVs in losartan group at the end of month 12 were significantly greater than changes in PWV in bisoprolol group (0.9 ± 0.3 vs. 0.4 ± 0.5 m/s, P = 0.021). Common carotid artery intima-media cross-sectional area decreased significantly only in patients treated with losartan (20.3 ± 4.9 vs. 19.1 ± 5.1 mm(2) , P = 0.001), and HOMA-IR was also reduced in losartan group only (1.9 ± 1.0 vs. 1.7 ± 0.8, P = 0.003). Multiple regression analysis showed significant correlations between changes in PWV and changes in HOMA-IR. With comparable BP-lowering efficacy, losartan achieved better improvement in insulin sensitivity, arterial stiffness, and carotid artery hypertrophy in hemodialysis patients. The regression of arterial stiffness may be in part through attenuation in insulin resistance.

Hepatocellular carcinoma (HCC) is the third-most lethal cancer worldwide. Understanding the molecular pathogenesis of HCC recurrence and metastasis is the key to improve patients’ prognosis. In this study, we report that protein tyrosine phosphatase receptor S (PTPRS) is significantly down-regulated in nearly 80% of HCCs, and its expression negatively correlates with aggressive pathological features, such as larger tumor size and advanced stage. In addition, PTPRS deficiency is independently associated with shorter survival and increased recurrence in patients, although 16.7% of HCCs show intratumor heterogeneous expression of PTPRS. Restoration of wild-type, but not mutant, PTPRS expression significantly inhibits HCC cell migration and invasion in vitro as well as lung metastasis in vivo, whereas knockdown of its expression significantly promotes invasion and metastasis. Notably, PTPRS-regulated HCC invasiveness is accompanied by typical changes of epithelial-mesenchymal transition (EMT). Moreover, PTPRS forms a complex with epithermal growth factor receptor (EGFR) and regulates its tyrosine residues’ phosphorylation. Ectopic expression of EGFR reverses the metastasis-inhibiting effects of PTPRS, whereas silencing of EGFR or inhibiting phosphorylation of key molecules in EGFR downstream pathways reinhibits EMT and metastasis caused by PTPRS down-regulation. Meanwhile, promoter hypermethylation of PTPRS is frequently detected in HCC samples and cell lines. Treatment with a demethylation agent, 5-aza-2′-deoxycytidine, recovers PTPRS expression in a dose-dependent manner. Conclusions: Epigenetic inactivation of PTPRS may increase phosphorylation and activity of EGFR signaling to promote EMT and metastasis in HCC. (Hepatology 2015;62:1201-1214)

This study was to investigate the influence of bone cement implantation on haemodynamics and the preventive effect of epinephrine hydrochloride on pulmonary embolism in elderly patients with cemented semihip replacement. 128 patients were retrospectively analyzed. The patients were treated with (group A, 64 cases) or without (group B, 64 cases) epinephrine hydrochloride saline. The monitoring indicators included systolic blood pressure (SBP), diastolic blood pressure (DBP), mean arterial pressure (MAP), heart rate (HR), and pulse oxygen saturation (SPO2). The indicators of the two groups were compared before and 1, 2, 3, 4, 5, 6, 7, 8, 9, and 10 minutes after bone cement implantation. Analysis of variance and SNK-q test were used for the statistical analysis. Blood pressure and SPO2of group B decreased with statistical difference (P<0.05) and HR increased without statistical significance, comparing with those of group A. In group A, SBP, DBP, MAP, HR, and SPO2after bone cement implantation did not change significantly at each time point comparing with before implantation (P>0.05). Bone cement implantation has significant influence on hemodynamics in elderly patients with hemiarthroplasty. Flushing the bone marrow cavity with saline epinephrine hydrochloride is an effective measure to reduce the incidence of bone cement pulmonary embolism.

Based on the metal cutting experiments, the forming model of feed direction burr is proposed in the paper, a physical parameter of determining the generating of typeⅠ feed direction burr — critical tool edge angle rI χ is given. And the main factors which influence rI χ are studied by the experiment, and related theory analysis is made. Furthermore, some new controlling methods and technology to restrain or decrease the feed direction burr in machining are developed.
