Background: Acute lymphoblastic leukemia with MLL/AF4 rearrangement remains a major hurdle to improving outcomes. Gene network and circRNAs have been found to participate in tumorigenesis, while their roles in leukemia still need to be explored. Recent patents have shown that circRNAs exhibit the markers for the children ALL, although the target and related mechanism remain to be elucidated. Objective: This study aims to explore the possible targets and mechanisms of ALL with MLLAF4 rearrangement. Methods: We first generated a gene network focusing on MLL-AF4 rearrangement. Cell viability was determined with Cell Counting Kit-8 assay. The cell apoptosis was tested by the Annexin V/PI assay. The RNA-protein complexes were analyzed by qRT-PCR, and the pathway proteins were analyzed by western blot. Results: This gene network was associated with biological processes, such as nucleic acid metabolism and immunity, indicating its key role in inflammation. We found that circ_0008012 was upregulated in MLL/AF4 ALL cells and regulated cell proliferation and apoptosis. Further computed simulation and RIP showed that IKKβ was the strongest protein in the NF-κB pathway binding with circ_0008012. As a result, possible regulation of circ_0008012 is suggested by binding IKKβ in the IKKα:IKKβ:IKKγ compound, which then phosphorylates IκB and activates NF- κB:p65:p300 compound in cell nucleus, thereby leading to leukemia. Conclusion: We identified a gene network for MLL/AF4 ALL. Moreover, circ_0008012 may be a therapeutic target for this subtype of ALL.

The thermal decomposition of AgNO3 is known to produce metallic Ag, but single-atomic dispersion is hard to achieve instead of the aggregation state of nanoparticles. Herein, we develop an efficient approach to thermally generate and stabilize single Ag atoms via the coordination effect. Two desired Co-Ag phosphonates [AgI2CoIII2(notpH3)2(NO3)]X [X = NO3− (1) or ClO4− (2)] were synthesized by solid-phase grinding method or solution crystallization. Helical architectures with controllable helical sense bias have recently attracted considerable interest for mimicking biological helices and developing novel chiral materials. Coordination polymers (CPs), composed of metal ion nodes and organic linkers, are intriguing systems showing tunable structures and functions. However, CPs with helical morphologies have rarely been explored so far. Particularly, chirality inversion through external stimulus has not been achieved in helical CPs. In this work, we carried out an in-depth investigation on the self-assembly of 1D gadolinium(III) phosphonate CPs using GdX3 (X=Cl, Br, I) and Gd(RSO3 ) (R=CH3 , C6 H5 , CF3 ) as metal sources and R-(1-phenylethylamino)methyl phosphonic acid (R-pempH2 ) as ligand. Superhelices were formed by precise control of the interchain interactions through different intercalated anions. Furthermore, the twisting direction of superhelices could be controlled by synergistic effect of anions and pH. This study may provide a new route to fabricate helical nanostructures of CPs with a desirable chiral sense and help understand the inner mechanism of the self-assembly process of macroscopic helical structures of molecular systems.

Photosensitive lanthanide-based single-molecule magnets (Ln-SMM) are very attractive for their potential applications in information storage, switching, and sensors. However, the light-driven structural transformation in Ln-SMMs hardly changes the coordination number of the lanthanide ion. Herein we report for the first time that X-ray (λ = 0.71073 Å) irradiation can break the coordination bond of Dy-OH2 in the three-dimensional (3D) metal-organic framework Dy2(amp2H2)3(H2O)6∙4H2O (MDAF-5), where the {Dy2(OPO)2} dimers are crosslinked by dianthracene-phosphonate ligands. The structural transformation proceeds in a single-crystal-to-single-crystal (SC-SC) fashion, forming the new phase Dy2(amp2H2)3(H2O)4∙4H2O (MDAF-5X). The phase transition is accompanied by a significant change in magnetic properties due to the alteration in coordination geometry of the DyIII ion from a distorted pentagonal bipyramid in MDAF-5 to a distorted octahedron in MDAF-5-X.

Controllable synthesis of uniformly distributed nanowires of coordination polymers with inherent physical functions is highly desirable but challenging. In particular, the combination of chirality and magnetism into nanowires has potential applications in multifunctional materials and spintronic devices. Herein, we report four pairs of enantiopure coordination polymers with formulae S-, R-Dy(cyampH)3 ⋅ CH3 COOH ⋅ 2H2 O (S-1, R-1), S-, R-Dy(cyampH)3 ⋅ 3H2 O (S-2, R-2), S-, R-Dy(cyampH)2 (C2 H5 COO) ⋅ 3H2 O (S-3, R-3) and S-, R-Dy(cyampH)3 ⋅ 0.5C2 H5 COOH ⋅ 2H2 O (S-4, R-4) [cyampH2 =S-, R-(1-cyclohexylethyl)aminomethylphosphonic acids], which were obtained depending on the pH of the reaction mixtures and the specific carboxylic acid used as pH regulator. Interestingly, compounds 3 were obtained as superlong nanowires, showing 1D neutral chain structure which contains both phosphonate and propionate anion ligands. While compounds 1, 2 and 4 appeared as block-like crystals, superhelices and nanorods, respectively, and exhibited similar neutral chain structures containing only phosphonate ligand. Slow magnetization relaxation characteristic of single-molecule magnet (SMM) behavior was observed for compounds S-1 and S-3. Theoretical calculations were performed to rationalize the magneto-structural relationships.

Dinuclear ytterbium and erbium based bifunction complexes Ln2L2(depma2)Cl2 (1-Ln, Ln = Yb and Er, H2L = N1,N3-bis(salicylideneimino)diethylenetriamine, depma2 = dimerized 9-diethyl-phosphonomethylanthracene) are reported. They undergo thermo-induced consecutive phase transitions, first the dissociation of depma2 ligand forming LnL(depma)Cl (2-Ln) and then the release of chloroethane forming LnL(epma) (3-Ln, epma = 9-ethylphosphonomethylanthrancene). The structural transformations are accompanied with synergetic switch of the luminescence in visible and NIR regions and also magnetic dynamics.

A scheme for achieving atom localization in the noise spectrum in phase quadrature of resonance fluorescence from a three-level A atom is suggested. We add a microwave field to the lower energy states in the three-level A atom localization scheme, and form a three-level loop atom system. When spontaneous emission photons are detected, by changing the relative phase between the three nonresonant fields, the atom is located in one of two half wavelengths, so the probability of atomic localization is 50%. We also see that the intensity and the detuning of the microwave field affect intensively localization probability. Compared with the scheme based on electromagnetically induced transparency, the detection probability of atoms in the sub wavelength range is only 25%, and the resonance condition is useless.

The tritrophic interactions among plant virus, host plants, and insect vectors directly influence the natural ecosystem, which, in turn, has tremendous practical implications in the sustainable pest control strategies. Cucumber mosaic virus (CMV), transmitted by aphids in a non-persistent manner, causes severe damage in diverse crops worldwide. There is a wealth of information on the initial round of interactions within this tritrophic system. However, knowledge on the subsequent round of interactions is very limited. In this research, we focused on their interactions among Nicotiana tabacum cv. Samsum plants, CMV, and green peach aphids, Myzus persicae specifically after the initial round of aphid feeding on CMV-infected plants. Our results show that initial aphid feeding on CMV-infected plants reduces the fitness of the subsequent aphids. The reproduction capacity, longevity, and survival rate of M. persicae are reduced on CMV-infected plants, previously foraged by M. persicae. Furthermore, the initial aphid feeding on CMV-infected plants induce gene expression involved in the salicylic acid (SA) signaling pathway and suppresses the expression of downstream genes associated with jasmonic acid (JA) signaling pathway. Besides, plant chlorophyll content and nitrogen source are reduced on those CMV-infected plants, previously foraged by aphids. The negative impacts on the fitness and performance of the subsequent aphids may have significant implications in virus transmission, distribution, and epidemiology.

Ultrathin nanosheets of luminescent metal-organic frameworks or coordination polymers have been widely used for sensing ions, solvents and biomolecules but, as far as we are aware, not yet used for temperature sensing. Herein we report two luminescent uranyl phosphonates based on 2-(phosphonomethyl)benzoic acid (2-pmbH3), namely (UO2)(2-pmbH2)2 (1) and (H3O)[(UO2)2(2-pmb)(2-pmbH)] (2). The former has a supramolecular layer structure, composed of chains of corner-sharing {UO6} octahedra and {PO3C} tetrahedra which are connected by hydrogen bonds between phosphonate and carboxylic groups. Compound 2 possesses a unique 2D anionic framework structure, where the inorganic uranyl phosphonate chains made up of {UO7} and {PO3C} polyhedra are cross-linked by 2-pmb3- ligands. The carboxylic groups of 2-pmbH2- ligands are pendant on the two sides of the layers and form hydrogen bonds between the layers. Both compounds can be exfoliated in acetone via a top-down freeze-thaw method, resulting in nanosheets of two-layer thickness. Interestingly, the photoluminescence (PL) of 1 and 2 is highly temperature sensitive. Variable temperature PL studies revealed that compounds 1 and 2 can be used as thermometers in the temperature ranges 120-300 K and 100-280 K, respectively. By doping the nanosheets into polymer matrix, 1-ns@PMMA and 2-ns@PMMA were prepared. The PL intensity of 1-ns@PMMA is insensitive to temperature, unlike that of the bulk sample. While 2-ns@PMMA exhibits similar temperature-dependent luminescence behaviour to its bulk counterpart, thereby enabling its potential application as a thermometer in the temperature range 100-280 K.

Chiral transcription from the molecular level to the macroscopic level by self-organization has been a topic of considerable interest for mimicking biological systems. Homochiral coordination polymers (CPs) are intriguing systems that can be applied in the construction of artificial helical architectures, but they have scarcely been explored to date. Herein, we propose a new strategy for the generation of superhelices of 1D CPs by introducing flexible cyclohexyl groups on the side chains to simultaneously induce interchain van der Waals interactions and chain misalignment due to conformer interconversion. Superhelices of S- or R-Tb(cyampH)3·3H2O (S-1H, R-1H) [cyampH2 = S- or R-(1-cyclohexylethyl)aminomethylphosphonic acid] were obtained successfully, the formation of which was found to follow a new type of “chain-twist-growth” mechanism that had not been described previously. The design strategy used in this work may open a new and general route to the hierarchical assembly and synthesis of helical CP materials., Superhelices of terbium phosphonates formed following a new type of “chain-twist-growth” mechanism involving flexible cyclohexyl groups.

Metal–organic frameworks showing stimuli-responsive magneto-optical properties are of intensive current interest for potential applications in spintronics and molecular devices. By incorporating a thermo-responsive dianthracene component into a lanthanide phosphonate framework, we successfully obtained compounds Ln2(amp2H2)2(C2O4)(H2O)2·xH2O (MDAF-4Ln, Ln = Dy, x = 10; Gd, x = 9; Er, x = 7; and amp2H4 = pre-photodimerized 9-anthrylmethylphosphonic acid). These compounds are isostructural showing three-dimensional open framework structures composed of Ln-chains and dianthracence linkages. Within the chain, the dimers of {Ln2O2(OPO)2} are connected by oxalate anions. The MOFs generate channels along the a-axis which are hydrophilic lined with protonated phosphonate moieties, and hence exhibit high affinity towards adsorption of water and ammonia. Thermogravimetric analysis and differential scanning calorimetry (DSC) measurements revealed that MDAF-4Ln experienced two consecutive phase transitions: first, the loss of all lattice and coordinated water molecules and then, the dissociation of amp2H22−. Detailed studies on MDAF-4Dy demonstrated that the thermo-induced structural transformations were accompanied by significant changes in its single-molecule magnetic behavior and luminescence properties.

Background Most plant viruses rely on vectors for their transmission and spread. One of the outstanding biological questions concerning the vector-pathogen-symbiont multi-trophic interactions is the potential involvement of vector symbionts in the virus transmission process. Here, we used a multi-factorial system containing a non-persistent plant virus, cucumber mosaic virus (CMV), its primary vector, green peach aphid, Myzus persicae, and the obligate endosymbiont, Buchnera aphidicola to explore this uncharted territory. Results Based on our preliminary research, we hypothesized that aphid endosymbiont B. aphidicola can facilitate CMV transmission by modulating plant volatile profiles. Gene expression analyses demonstrated that CMV infection reduced B. aphidicola abundance in M. persicae, in which lower abundance of B. aphidicola was associated with a preference shift in aphids from infected to healthy plants. Volatile profile analyses confirmed that feeding by aphids with lower B. aphidicola titers reduced the production of attractants, while increased the emission of deterrents. As a result, M. persicae changed their feeding preference from infected to healthy plants. Conclusions We conclude that CMV infection reduces the B. aphidicola abundance in M. persicae. When viruliferous aphids feed on host plants, dynamic changes in obligate symbionts lead to a shift in plant volatiles from attraction to avoidance, thereby switching insect vector’s feeding preference from infected to healthy plants.

Background: The poor prognosis of acute myeloid leukemia (AML) is partly due to the immunosuppressive leukemia environment. A therapeutic approach to induce pyroptosis is promising to work on this issue. Pyroptosis, a form of inflammatory cell death, is capable to release pro-immune factors and enhance the infiltration and cytotoxicity of natural killer (NK) cells. Chemical drugs have been verified to enhance anti-tumor immunity by inducing pyroptosis. Arsenic trioxide (ATO), a clinical drug in leukemia, is reported to be effective to kill AML cells, but it’s still unclear about its mechanism in AML. Our research is aimed at exploring the ATO effect on pyroptosis induction and the anti-tumor immunity of NK cells in AML.Methods: Pyroptotic phenomenon was detected by confocal microscopy, Annexin-V / propidium iodide staining, LDH release assay, and western blotting; The release of cytokines was detected by SDS-PAGE, proteomic analysis, ELISA, etc.; AML xenograft models were established to explore the ATO effect in vivo and the anti-tumor effect of ATO treatment was identified by tumor-volume measurement, HE staining and KI67 staining; The activity of NK cells was evaluated by immunofluorescence, flow cytometric analysis, etc.; The differential expressed proteins were identified by quantitative proteomic analysis and western blotting.Results: We first discovered that ATO induced pyroptosis in AML cells and activated caspase-3/gasdermin E (GSDME) pathway to induce pyroptosis, and enhances the release of LDH, HMGB1, and IL-1β. Moreover, we revealed that ATO treatment promoted the proliferation, tumor-infiltration, and degranulation of NK cells in AML and its leukemia microenvironment. The further proteomic clues indicate the potential mechanisms of the ATO anti-leukemia effect in vivo.Conclusions: These findings indicate that ATO treatment induces pyroptosis via caspase-3/GSDME pathway, enhances the release of pro-inflammatory cytokines, and activates NK cells in AML. Our data identify what is to our knowledge the first clinical drug that induces pyroptosis in AML, revealing the potential anti-leukemic and pro-immune properties of ATO treatment in the leukemia microenvironment of AML, which may be beneficial in reducing the recurrence problems associated with the leukemia microenvironment.

The prognosis of patients with acute myeloid leukemia (AML) caused by the FLT3-ITD mutation is poor. Arsenic trioxide (ATO) and all-trans retinoic acid (ATRA) can down-regulate FLT3-ITD level and selectively kill leukemia cells carrying the FLT3-ITD mutation. However, the mechanisms of action of these two compounds are unknown. Here, we found that ATO could bind FLT3-ITD at Lys91 and Asp225, whereas ATRA could bind FLT3-ITD at Lys5 and Gln6. Both compounds could not bind wild-type FLT3. Further studies revealed that ATO/ATRA may suppress the Expression of FLT3-ITD by promoting the UBE2L6-mediated ubiquitination pathway and decreasing the expression of C-MYC. However, further studies are needed to define the mechanisms of these compounds on AML. Our research provides an experimental basis for the use of ATO/ATRA in FLT3-ITD AML in clinical practice.

Reaction of the metalloligand IrIII(ppy-COOH)3 and the anisotropic paramagnetic CoII ion under solvothermal conditions resulted in a metal-metalloligand coordination polymer, [CoII3(μ3-O)(μ-OH2){IrIII(ppy-COO)2(ppy-COOH)}2(H2O)4]·2DMF·xH2O (I). It consists of trimeric Co3O secondary building units (SBUs) bridged by pairs of Ir to form chains of alternate orthogonal squares. The compound undergoes two single-crystal to single-crystal transformations while retaining its general structural features. A chemical transformation occurs to give [CoII3(μ3-O){IrIII(ppy-COO)2(ppy-COOH)}2(H2O)4(DMF)]·DMF·H2O (II) by soaking in acetone, where a bridging water molecule departs and the solvent DMF bonds to the vacant site of the Co center. Both I and II undergo a temperature-induced transformation to [CoII3(μ3-O){IrIII(ppy-COO)2(ppy-COOH)}2(H2O)3(DMF)]·DMF (III), where one more coordinated water molecule is lost. The major difference in the three phases is in the Co coordination spheres, which have considerable consequences on the magnetism. Compound I displays paramagnetism down to 2 K, whereas II and III show weak ferromagnetism with TC values of 14 and 17 K, respectively.

As an important class of inorganic-organic hybrid materials, metal phosphonates can exhibit versatile structures, interesting functions and high water and thermal stabilities. Despite a large number of metal phosphonates reported in the past two decades, the development of chiral metal phosphonates is still in its infancy. This review summarizes the current status in this topical field including the synthetic strategies, the crystal structures of chiral metal phosphonates reported thus far, and their physical and chemical properties. Future challenges in this promising field are also discussed.

Plant viruses can modify the performance and behavior of insect vectors to maximize their transmission rates. Whether viruses can evolve to mediate their insect vectors’ interspecific competition is poorly understood. Here, we used two vectors of tomato zonate spot virus (TZSV), one native species (Thrips palmi) and one exotic species (Frankliniella occidentalis), to investigate this in laboratory and field experiments. TZSV infection increased the population growth of its insect vectors when the thrips species were isolated from one another. When both species coexisted, F. occidentalis displaced T. palmi on both TZSV-infected and uninfected plants. However, on TZSV-infected plant, F. occidentalis completely displaced T. palmi more rapidly (within 14 days) than on uninfected plants. Our results also revealed that reproductive interference occurred between the two thrips species, whereas TZSV-infected pepper significantly decreased the reproductive interference of T. palmi to F. occidentalis. When F. occidentalis and T. palmi co-occurred in Jinning County, Yunnan Province, we found a rising trend of natural proportions of F. occidentalis in the TZSV-infected pepper fields, suggesting that TZSV-infected pepper enhanced the competitive advantage of F. occidentalis over T. palmi in the field. Together, our findings demonstrate a plant virus that plays an important role in the competitive displacement of its insect vectors.

Photochemical reactions of (PPh4)[OsVI(N)(L)(CN)3] (NO2-OsN) with piperidine and pyrrolidine afforded two osmium(v) hydrazido compounds, (PPh4)[OsV(L)(CN)3(NNC5H10)] ([PPh4]1) and (PPh4)[OsV(L)(CN)3(NNC4H8)] ([PPh4]2), respectively. Their structures consist of isolated, mononuclear distorted octahedral osmium anions that are well-separated from each other by PPh4+. Their low spin S = 1/2 and L = 1 ground state was confirmed by magnetometry and DFT calculations. Interestingly, both compounds exhibit slow magnetic relaxation under a bias dc-field. These osmium(v) complexes are potentially useful building-blocks for the construction of molecule-based architectures with interesting magnetic properties. In contrast, the structurally related (PPh4)[OsIII(L)(CN)3(NH3)] ([PPh4]3), which also has a low-spin S = 1/2 ground state but with a different electronic configuration (5d5), does not exhibit slow magnetic relaxation, due to the absence of any orbital moment (L = 0). Furthermore, the structurally different osmium(v) hydrazido compound reported by Meyer, [OsV(tpy)(Cl)2(NNC5H10)](PF6) (4[PF6]), also does not exhibit slow magnetic relaxation due possibly to a change in magnetic anisotropy from axial for [PPh4]1 and [PPh4]2 to planar.

A series of iridium(III)–lanthanide(III) bimetallic coordination polymers were successfully obtained by hydrothermal reactions based on the Ir unit [Ir(ppy)2(Hdcbpy)], namely, [Ln{Ir(ppy)2(dcbpy)}2(NO3)(H2O)n]·solvent (Ir2Ln, n = 5, Ln = Gd and Dy; and n = 4, Ln = Er and Yb) and [Ln2(OH){Ir(ppy)2(dcbpy)}4(NO3)(H2O)4]·solvent (Ir4Ln2, Ln = Gd, Dy, Er, and Yb). Compounds Ir2Ln show chain structures in which the lanthanide ions are bridged by the Ir(ppy)2(dcbpy)− ligands through the carboxylate groups. Compounds Ir4Ln2 have double-chain structures, in which the dimers of Yb2(OH) are connected by the Ir(ppy)2(dcbpy)− ligands. Magnetic studies reveal that Ir2Dy and Ir4Dy2 exhibit slow magnetic relaxation behaviour in zero dc field, which is characteristic of single molecule magnets, but Ir2Er, Ir4Yb2, Ir4Er2 and Ir4Yb2 require a field to induce such characteristics. Moreover, since the iridium(III) moiety can serve as an “antenna” to effectively sensitize the f–f transitions of the lanthanide ions, near-infrared luminescence has been observed for the YbIII and ErIII complexes. As far as we know, this is the first observation of SMM behaviour and NIR luminescence in iridium(III)–lanthanide(III) bimetallic coordination polymers.

Metal phosphonates are an important class of metal-organic hybrid materials that exhibit versatile structures, intriguing functions and high water and thermal stability. Despite a large number of metal phosphonates achieved over the past few decades, those incorporating metalloligands are rather limited. The metalloligand approach can provide a unique opportunity in constructing homo- or mixed-metal coordination polymers with rationally immobilized functional moieties for various applications in gas storage and separation, heterogeneous catalysis, sensing and multifunctional materials. In this feature article, we shall introduce the current status of a special subclass of metal phosphonates, namely, metal-metalloligand phosphonates (MMPs), including synthetic strategies, crystal structures, and properties of those based on paddlewheel diruthenium, metallo-polyazamacrocycle, metalloporphyrin and metallo-tris-bipyridine ligands. Future challenges in this field are discussed.

Reactions of metalloligands MIII(notpH3) (M = Fe, Co and notpH6 = 1,4,7-triazacyclononane-1,4,7-triyl-tris(methylenephosphonic acid)) with Zn(OAc)2 under hydrothermal conditions resulted in new metal phosphonates Zn2Fe(notp)Cl(H2O) (1) and ZnCo(notpH)(H2O)·2H2O (2). They crystallize in polar space groups P63 (for 1) and Pca21 (for 2), respectively, and exhibit layer structures in which the inorganic layers are separated by the organic groups of the notp ligands. However, the layer topologies of the two compounds are quite different. In 1, the layer contains 6-membered rings composed of one {FeN3O3} octahedron, one {Zn1O3Cl}, one {Zn2O4} and three {PO3C} tetrahedra via corner-sharing connections, while in 2, the layer contains 10-membered rings composed of two {CoO3N3} octahedra, three {ZnO4} and five {PO3C} tetrahedra via vertex-sharing connections. Dielectric measurements on single crystals of 2 confirmed the presence of high dielectric anisotropy. Proton conductivity measurements revealed that the proton conduction is more favourable in 2 due to the presence of a continuous hydrogen bond network in this compound.

Manipulating the synergy between magnetic and optical characteristics in a predictable manner by an external stimulus is a highly desirable option to develop smart materials for advanced applications such as information processing and storage in multifunctional devices. Herein we report dinuclear complexes Ln2L2(depma2)Cl2 (1-Ln, Ln = Dy, Gd; H2L = N1,N3-bis(salicylideneimino)diethylenetriamine, depma2 = photodimerized 9-diethylphosphonomethylanthracene) where the lanthanide ions possess pseudo-D5h geometries with five equatorial positions occupied by L2− and the axial positions by depma2 and the Cl− anion. depma2 acts as a bridge between the equivalent LnIII ions and can be opened and closed upon heating and light irradiation. 1-Ln experience consecutive structural transformations, first separation of the depma2 at ca. 170 °C and second elimination of chloroethane at ca. 250 °C, accompanied by a synergetic switch of the photoluminescence and magnetic dynamics for 1-Dy.
