Sheath blight (ShB), caused by Rhizoctonia solani, is one of the major threats to rice (Oryza sativa L.) production. However, it is not clear how the risk of rice ShB will respond to elevated CO2 and temperature under future climate change. Here, we conducted, field experiments of inoculated R. solani under combinations of two CO2 levels (ambient and enriched up to 590 μmol mol-1) and two temperature levels (ambient and increased by 2.0°C) in temperature by free-air CO2 enrichment (T-FACE) system for two cultivars (a susceptible cultivar, Lemont and a resistant cultivar, YSBR1). Results indicate that for the inoculation of plants with R. solani, the vertical length of ShB lesions for cv. Lemont was significantly longer than that for cv. YSBR1 under four CO2 and temperature treatments. The vertical length of ShB lesions was significantly increased by elevated temperature, but not by elevated CO2, for both cultivars. The vertical length of ShB lesions under the combination of elevated CO2 and elevated temperature was increased by 21–38% for cv. Lemont and by -1–6% for cv. YSBR1. A significant increase in MDA level was related to a significant increase in the vertical length of ShB lesions under the combination of elevated CO2 and elevated temperature. Elevated CO2 could not compensate for the negative effect of elevated temperature on yield of both cultivars under future climate change. Rice yield and biomass were further decreased by 2.0–2.5% and 2.9–4.2% by an increase in the severity of ShB under the combination of elevated CO2 and elevated temperature. Thus, reasonable agronomic management practices are required to improve both resistance to ShB disease and grain yield for rice under future climate change.

In view of the problem that the measurement accuracy of screen gaps is affected by the R region of the transition rounded corner of the large-size flat back cover, a mobile vision system with dual linear motor synchronous drive is designed to realize precise positioning of monocular vision in 16 local image acquisition positions set in the R region. Full-size measurement of the R region is achieved using local information fusion to accurately measure the screen gap. First, to solve the problem of edge breaking and the blurring of local images, a region extraction and segmentation method based on block statistics is proposed to quickly obtain accurate outer edges. Then, an adaptive Hough line detection method based on dichotomy is proposed to accurately locate the outer edges. The experimental results show that the accuracy of region extraction and segmentation is up to 99.68% within 60 ms; the angle error and position error of line detection are less than 0.01° and 0.2 pixels, respectively. With optimal anti-interference performance, the proposed algorithm realizes the adaptive and accurate positioning of the outer edge within 200 ms; the measurement time and the measurement error of the measurement system are less than 6.0 s and 0.03 mm, respectively.

Welding joint is a key component in steel structure engineering, which is often subjected to complex stress and becomes the weak link of building structure, and different fracture modes may appear under different stress. In this study, the ductile fracture of Q345 steel welded joint was investigated by selecting the base material and heat-affected zone (HAZ) material. Based on the J–C (Johnson–Cook) fracture model of stress triaxiality factor, a series of round bar specimens were designed for the tensile test, and the variation of minimum diameter and gauge distance was tracked in real time by digital image correlation (DIC) technology, and the cloud map of strain distribution in the whole loading process was obtained. The constitutive model and fracture criterion model of Q345 steel welded joint base material and HAZ material are established through experimental measurement. The results show that the maximum triaxial stress is at the center root of the minimum diameter of the notched round bar tensile specimen, the sudden change of the notched edge will lead to stress concentration, the stress change of the notch section is the most significant, extending from near the notch, the maximum strain is at the root of the notch, and the fracture position appears at the minimum section of the notch. According to the true stress-strain curve of a standard tensile specimen and the least square method, the values of parameters A, B, and N of the J–C (Johnson–Cook) constitutive model of base material and HAZ material are 379.80, 452.89, and 0.37 and 392.90, 540.61, and 0.49, respectively. The fracture strains of the base metal and HAZ material under different triaxial stresses were measured experimentally, and the J–C fracture model was fitted. The values of failure criterion parameters D1, D2, and D 3 were 1.025, −0.008, and 3.617 and 0.678, −2.689E-5, and 7.683, respectively. This provides experimental parameters for mechanical properties and fracture models of welded joints in structural engineering, refined finite element models considering material failure for structural numerical analysis, and basic data for evaluation and design of building structures, which have good social and economic benefits.

Background: Exogenous HMGB1 plays a vital role in tumor recurrence, and HMGB1 is ubiquitous in the tumor microenvironment. However, the mechanism of action is still unclear. We investigated the role of exogenous HMGB1 in tumor proliferation and metastasis using human SW1990 and PANC-1 cells after radiotherapy and explored the possible molecular mechanism.Materials and Methods: Residual PANC-1 cells and SW1990 cells were isolated after radiotherapy. The supernatant after radiotherapy was collected. The relative expression of HMGB1 was evaluated by Enzyme Linked Immunosorbent Assay (ELISA). Electron microscope (EMS) was used to collect the images of pancreatic cancer cells pre and post radiotherapy treatment. The proliferation of pancreatic cancer cells which were treated with different radiation doses was measured by Carboxy Fluorescein Succinimidyl Ester (CFSE). The migration rates of pancreatic cancer cells were measured by wound healing assays. Subsequently, the expression of related proteins was detected by Western Blot. In vivo, the subcutaneous pancreatic tumor models of nude mice were established, and therapeutic capabilities were tested.Results: HMGB1 was detected in the supernatant of pancreatic cancer cells after radiotherapy. The results of CFSE showed that exogenous HMGB1 promotes the proliferation and metastasis of pancreatic cancer cells. The western blot results showed activation of p-GSK 3β and up-regulation of N-CA, Bcl-2, and Ki67 in response to HMGB1 stimulation, while E-CA expression was down-regulated in pancreatic cancer cells in response to HMGB1 stimulation. In vivo, ethyl pyruvate (EP, HMGB1 inhibitor) inhibits the growth of tumors and HMGB1 promotes the proliferation of tumors after radiation.Conclusion: Radiotherapy induces HMGB1 release into the extracellular space. Exogenous HMGB1 promotes the proliferation and metastasis of PANC-1 cells and SW1990 cells by activation of p-GSK 3β which is mediated by Wnt pathway.

Background: High-mobility group box 1 (HMGB1), a common extracellular damage associated molecular pattern molecule, is overexpressed in several solid tumors including pancreatic carcinoma. We previously observed that radiotherapy induced dying cells secrete HMGB1 and accelerate pancreatic carcinoma progression through an unclear mechanism. Methods: Using the Millicell system as an in vitro co-culture model, we performed quantitative reverse transcriptase-polymerase chain reaction, western blot and sphere forming ability analyses to access the effect of dying-cell-derived HMGB1 on CD133+ cancer cell stemness in vitro and in vivo. Interactions between HMGB1 and Toll-like receptor 2(TLR2)/TLR4 were studied by co- immunoprecipitation. Western blot and short-hairpin RNA-based knockdown assays were conducted to detect HMGB1 and TLR2/TLR4 signaling activity. Findings: Radiation-associated, dying-cell-derived HMGB1 maintained stemness and contributed to CD133+ cancer stem cell self-renewal in vitro and in vivo. In overexpressing and silencing experiments, we demonstrated that the process was activated by TLR2 receptor, whereas TLR4 antagonized HMGB1-TLR2 signaling. Wnt/β-catenin signaling supported the HMGB1-TLR2 mediated stemness of CD133+ cancer cells. Interpretation: Our results show how irradiation-induced cell death might enhance the stemness of resident cancer cells, and indicate HMGB1-TLR2 signaling as a potential therapeutic target for preventing pancreatic cancer recurrence. The progressive collapse of a concrete-filled steel tubular (CFST) frame structure is studied subjected to impact loading of vehicle by the finite-element software ABAQUS, in the direct simulation method (DS) and alternate path method (AP), respectively. Firstly, a total of 14 reference specimens including 8 hollow steel tubes and 6 CFST specimens were numerically simulated under transverse impact loading for verification of finite-element models, which were compared with the existing test results, confirming the overall similarity between them. Secondly, a finite-element analysis (FEA) model is established to predict the impact behaviour of a five-storey and three-span composite frame which was composed of CFST columns and steel beams under impact vehicle loading. The failure mode, internal force-time curve, displacement-time curve, and mechanical performance of the CFST frame were obtained through analyzing. Finally, it is concluded that the result by the DS method is closer to the actual condition and the collapse process of the structure under impact load can be relatively accurately described; however, the AP method is not.

Abstract Background Dying cells after irradiation could promote the repopulation of surviving cancer cells leading to tumor recurrence. We aim to define the role of dying cells in promoting pancreatic cancer cells metastasis following radiotherapy. Methods Using the transwell system as the in vitro co-culture model, a small number of untreated pancreatic cancer cells were seeded in the upper chamber, while a larger number of lethally treated pancreatic cancer cells were seeded in the lower chamber. A series of experiments were conducted to investigate the role of dying-cell-derived HMGB1 on the invasion of pancreatic cancer in vitro and cancer metastasis in vivo. We then designed shRNA knockdown and Western blot assays to detect signaling activity. Results We found that dying pancreatic cancer cells significantly promote the invasion of pancreatic cancer cells in vitro and cancer metastasis in vivo. HMGB1 gene knockdown attenuated the migration-stimulating effect of irradiated, dying cells on living pancreatic cancer cells. Finally, we showed that dying-cell-derived HMGB1 functions in a paracrine manner to affect cancer-cell migration dependent on acquiring an epithelial-mesenchymal transition (EMT) phenotype and PI3K/pAkt activation. This process is mediated by the receptor for TLR2. Conclusion Our study indicates that, during radiotherapy, dying pancreatic cancer cells activate paracrine signaling events that promote the mobility of surviving tumor cells. We suggest a strategy to inhibit HMGB1 for preventing pancreatic carcinoma relapse and metastasis.

Nitrogen (N) has a unique place in agricultural systems with large requirements. To achieve optimal nitrogen management that meets the needs of agricultural systems without causing potential environmental risks, it is of great significance to increase N use efficiency (NUE) in agricultural systems. A chlorophyll meter, for example, the SPAD-502, can provide a simple, nondestructive, and quick method for monitoring leaf N status and NUE. However, the SPAD-based crop leaf’s N status varies greatly due to environmental factors such as CO2 concentration ([CO2]) or temperature variations. In this study, we conducted [CO2] (ambient and enriched up to 500 μmol moL1) and temperature (ambient and increased by 1.5~2.0 °C) controlled experiments from 2015 to 2017 and in 2020 in two Free-Air CO2 Enrichment (FACE) sites. Leaf characters (SPAD readings, chlorophyll a + b, N content, etc.) of seven rice cultivars were measured in this four year experiment. Here, we provide evidence that SPAD readings are significantly linearly correlated with rice leaf chlorophyll a + b content (chl a + b) and N content, while the relationships are profoundly affected by elevated [CO2] and warming. Under elevated [CO2] treatment (E), the relationship between chl a + b content and N content remains unchanged, but SPAD readings and chl a + b content show a significant difference to those under ambient (A) treatment, which distorts the SPAD-based N monitoring. Under warming (T), and combined elevated [CO2] and warming (ET) treatments, both of the relationships between SPAD and leaf a + b content and between leaf a + b content and N content show a significant difference to those under A treatment. To deal with this issue under the background of global climate change dominated by warming and elevated [CO2] in the future, we need to increase the SPAD reading’s threshold value by at least 5% to adjust for applying N fertilizer within the rice cropping system by mid-century.

Understanding how humanity’s influence on the climate affects rainfall seasonality around the world is immensely important for agriculture production, ecology protection, and freshwater resource management. In this study, we qualitatively and quantitatively analyzed the potential influence of anthropogenic forcing on rainfall seasonality in global land monsoon (GM) regions using the Coupled Model Intercomparison Project Phase 5 (CMIP5) general circulation models. We discovered that anthropogenic forcing enhances rainfall seasonality over many parts of GM regions, and was evident in the South Asian and the most parts of the South American and the South African monsoon regions. Anthropogenic forcing partially but clearly contributed to the increasing trend of rainfall seasonality over many parts of GM regions from 1960 to 2012. Moreover, anthropogenic forcing also increased the probability of more pronounced rainfall seasonality in almost all GM regions. The results provide valuable information for agriculture, ecology, and freshwater resource management under climate warming induced by anthropogenic forcing.

Background A major workforce inadequacy and the change in skill demanded have been observed due to the digital transformation. This study aims to identify the digital skills demanded by employers, focusing on exploring the skills gaps among the graduates that impact graduate employability. Methods A cross sectional online survey was conducted among the companies registered with the Malaysian Productivity Corporation (MPC). Demand for digital skills was assessed using descriptive analysis of mean scores of the employers’ rating in digital skills at present and in future. A pair sample t-test was performed to explore the existence of skill gaps, by comparing the demand versus competencies of the graduates’ digital skills in the workplace. Results Of the 393 responses collected, 376 responses were used for final analysis. The findings show that the current top three digital skills in demand are “information and data literacy”, “problem-solving”, and “digital content creation”. Whereas the top digital skills for future demand are “problem-solving”, “safety”, and “communication and collaboration”. In addition, the most significant (digital) skill gap is found in “communication and collaboration skills” followed by “problem-solving” and “safety” skills. Conclusions These findings provide insights into the digital skills demanded by employers in IR 4.0, allowing the practitioners, education service providers and policymakers to do better planning on human capital management and training development. By focusing on identifying the current and future demand for digital skills while exploring the digital skill gaps among the graduates, this study provides insight into the employability skills required by the graduate before entering the job market.

Pancreaticobiliary endoscopy is an essential tool for diagnosing and treating pancreaticobiliary diseases. However, it does not fully meet clinical needs, which presents challenges such as significant difficulty in operation and risks of missed diagnosis or misdiagnosis. In recent years, artificial intelligence (AI) has enhanced the diagnostic and treatment efficiency and quality of pancreaticobiliary endoscopy. Diagnosis and differential diagnosis based on endoscopic ultrasound (EUS) images, pathology of EUS‐guided fine‐needle aspiration or biopsy, need for endoscopic retrograde cholangiopancreatography (ERCP) and assessment of operational difficulty, postoperative complications and prediction of patient prognosis, and real‐time procedure guidance. This review provides an overview of AI applications in pancreaticobiliary endoscopy and proposes future development directions in aspects such as data quality and algorithmic interpretability, aiming to provide new insights for the integration of AI technology with pancreaticobiliary endoscopy. Photocatalysis driven by abundant yet intermittent solar energy has considerable potential in renewable energy generation and environmental remediation. The outstanding electronic structure and physicochemical properties of graphitic carbon nitride (g-C3N4), together with unique metal-free characteristic, make them ideal candidates for advanced photocatalysts construction. This review summarizes the up-to-date advances on g-C3N4 based photocatalysts from ingenious-design strategies and diversified photocatalytic applications. Notably, the advantages, fabrication methods and limitations of each design strategy are systemically analyzed. In order to deeply comprehend the inner connection of theory--structure--performance upon g-C3N4 based photocatalysts, structure/composition designs, corresponding photocatalytic activities and reaction mechanisms are jointly discussed, associated with introducing their photocatalytic applications toward water splitting, carbon dioxide/nitrogen reduction and pollutants degradation, etc. Finally, the current challenges and future perspectives for g-C3N4 based materials for photocatalysis are briefly proposed. These design strategies and limitations are also instructive for constructing g-C3N4 based materials in other energy and environment-related applications. [ABSTRACT FROM AUTHOR]
