6. Silencing TLR4 using an ultrasound-targeted microbubble destruction-based shRNA system reduces ischemia-induced seizures in hyperglycemic rats

Abstract

The toll-like receptor 4 (TLR4) pathway is involved in seizures. We investigated whether ultrasound-targeted microbubble destruction (UTMD)-mediated delivery of short hairpin RNA (shRNA) targeting the TLR4 gene (shRNA-TLR4) can reduce ischemia-induced seizures in rats with hyperglycemia. A total of 100 male Wistar rats were randomly assigned to five groups: (1) Sham; (2) normal saline (NS); (3) shRNA-TLR4, where rats were injected with shRNA-TLR4; (4) shRNA-TLR4 + US, where rats were injected with shRNA-TLR4 followed by ultrasound (US) irradiation; and (5) shRNA-TLR4 + microbubbles (MBs) + US, where rats were injected with shRNA-TLR4 mixed with MBs followed by US irradiation. Western blot and immunohistochemical staining were used to measure TLR4-positive cells. Half of the rats in the NS group developed tonic-clonic seizures, and TLR4 expression in the CA3 region of the hippocampus was increased in these rats. In addition, the NS group showed an increased number of TLR4-positive cells compared with the Sham group, while there was a decreased number of TLR4-positive cells in the shRNA, shRNA + US, and shRNA + MBs + US groups. Our findings indicate that the TLR4 pathway is involved in the pathogenesis of ischemia-induced seizures in hyperglycemic rats and that UTMD technology may be a promising strategy to treat brain diseases.

Abstract

Introduction: The connection between periodontitis and mild cognitive impairment (MCI) continues to receive attention. However, whether periodontitis is a risk factor for MCI remains still uncertain. This study aims to systematically analyze the available literature regarding the relationship between periodontitis and the risk of developing MCI and whether the periodontal health of MCI patients is poorer. Methods: A literature search of PubMed, Scopus, Embase, and Web of Science databases was conducted to include all studies on the relationship between periodontitis and MCI from inception to April 2023. The studies were independently screened by 2 researchers, and those meeting the inclusion criteria were extracted and cross-checked. Pooled odds ratio (OR) or mean difference (MD) with 95% confidence intervals (CI) was calculated using either a fixed-effects or random-effects model. Results: Seven studies with a total of 3,973 participants were included. Meta-analysis results showed a statistically significant higher incidence of MCI in patients with periodontitis (OR, 1.70 (95% CI: 1.24–2.32, p < 0.001) compared to healthy participants. A subgroup meta-analysis showed that the pooled OR for the risk of MCI in patients with severe periodontitis was 2.09 (95% CI: 1.49–2.92, p < 0.001). In addition, attachment loss (MD = 0.44, 95% CI: 0.12–0.75, p < 0.001) and plaque index (MD = 0.72, 95% CI: 0.50–0.93, p < 0.001) were higher in MCI patients compared with the control group, but the pocket probing depth (MD = 0.21, 95% CI: −0.08 to 0.49, p = 0.15) was not significantly different between the two groups. Conclusions: Patients with periodontitis are at a higher risk of developing MCI, and the periodontal health of MCI patients is generally compromised. Abstract

Background: Cancer‐associated fibroblasts (CAFs) are potential targets for cancer therapy. Due to the heterogeneity of CAFs, the influence of CAF subpopulations on the progression of lung cancer is still unclear, which impedes the translational advances in targeting CAFs. Methods: We performed single‐cell RNA sequencing (scRNA‐seq) on tumour, paired tumour‐adjacent, and normal samples from 16 non‐small cell lung cancer (NSCLC) patients. CAF subpopulations were analyzed after integration with published NSCLC scRNA‐seq data. SpaTial enhanced resolution omics‐sequencing (Stereo‐seq) was applied in tumour and tumour‐adjacent samples from seven NSCLC patients to map the architecture of major cell populations in tumour microenvironment (TME). Immunohistochemistry (IHC) and multiplexed IHC (mIHC) were used to validate marker gene expression and the association of CAFs with immune infiltration in TME. Results: A subcluster of myofibroblastic CAFs, POSTN+ CAFs, were significantly enriched in advanced tumours and presented gene expression signatures related to extracellular matrix remodeling, tumour invasion pathways and immune suppression. Stereo‐seq and mIHC demonstrated that POSTN+ CAFs were in close localization with SPP1+ macrophages and were associated with the exhausted phenotype and lower infiltration of T cells. POSTN expression or the abundance of POSTN+ CAFs were associated with poor prognosis of NSCLC. Conclusions: Our study identified a myofibroblastic CAF subpopulation, POSTN+ CAFs, which might associate with SPP1+ macrophages to promote the formation of desmoplastic architecture and participate in immune suppression. Furthermore, we showed that POSTN+ CAFs associated with cancer progression and poor clinical outcomes and may provide new insights on the treatment of NSCLC. [ABSTRACT FROM AUTHOR]

42. Crustal structure beneath northern Myanmar from ambient noise tomography

Abstract

The Burmese arc consists of the Indo-Burman Ranges (IBR), an accretionary wedge in the west and the Central Myanmar Basin (CMB) in the east. It is bounded in the east by the seismically active Sagaing Fault to the Shan Plateau. Intermediate-depth seismicity below Myanmar occurs at depths up to ~150km, generally understood to be related to the subducting Burma slab. We useambient noise tomography toconstruct a regional three-dimension (3-D) shear wave velocity model, aiming at figuring out the transition from oceanic subduction to continental subduction/collisionalong the Burmese arcand how it affects the upper plate crustal deformation. Our dataset includes 30 seismic stations from a recently deployed temporary seismic array and ~50permanent stations in and around Myanmar. Ambient noise corss-correlations are calculated at periods of 5-60 s and are inverted for a 3-D velocity model from the crust to the uppermost mantle. Our model shows alow-velocity anomaly extending to >20 km depths beneath the northern part of the CMB that accurately defines the Chindwin Subbasin. The crust beneath the IBM is characterized by low velocity, which probably reflects the fluid-rich accreted sediments that has experienced metamorphism in the mid-lower crust. Underneath the Sagaing Fault an N-S trending high-velocity anomaly is clearly visible at depths of 40-70 km in the uppermost mantle extending from 20°N to 24°N. The anomaly could represent a remnant slab of the Incertus Arc collision/subduction and mark the eastern boundary of the Burma plate., The 28th IUGG General Assembly (IUGG2023) (Berlin 2023)
