Multi-omics approaches use a diversity of high-throughput technologies to profile the different molecular layers of living cells. Ideally, the integration of this information should result in comprehensive systems models of cellular physiology and regulation. However, most multi-omics projects still include a limited number of molecular assays and there have been very few multi-omic studies that evaluate dynamic processes such as cellular growth, development and adaptation. Hence, we lack formal analysis methods and comprehensive multi-omics datasets that can be leveraged to develop true multi-layered models for dynamic cellular systems. Here we present the STATegra multi-omics dataset that combines measurements from up to 10 different omics technologies applied to the same biological system, namely the well-studied mouse pre-B-cell differentiation. STATegra includes high-throughput measurements of chromatin structure, gene expression, proteomics and metabolomics, and it is complemented with single-cell data. To our knowledge, the STATegra collection is the most diverse multi-omics dataset describing a dynamic biological system.

The differentiation of self-renewing progenitor cells requires not only the regulation of lineage- and developmental stage-specific genes but also the coordinated adaptation of housekeeping functions from a metabolically active, proliferative state toward quiescence. How metabolic and cell-cycle states are coordinated with the regulation of cell type-specific genes is an important question, because dissociation between differentiation, cell cycle, and metabolic states is a hallmark of cancer. Here, we use a model system to systematically identify key transcriptional regulators of Ikaros-dependent B cell-progenitor differentiation. We find that the coordinated regulation of housekeeping functions and tissue-specific gene expression requires a feedforward circuit whereby Ikaros down-regulates the expression of Myc. Our findings show how coordination between differentiation and housekeeping states can be achieved by interconnected regulators. Similar principles likely coordinate differentiation and housekeeping functions during progenitor cell differentiation in other cell lineages.

Transfer printing is a key technology in the fabrication of flexible electronics. Transfer printing method based on curvature provides a simple and effective way to transfer films onto weakly adhesive or even adhesiveless surfaces, overcoming the shortage of traditional transfer printing methods that it is difficult to print functional materials from strong interface to weak interface and can only be applicable to surfaces with certain interfacial strength. So far, the theoretical principle of the curvature‐based transfer printing method has not yet been developed. In this article, a dual‐interface competitive fracture model is established to analyze the mechanism of printing and picking up processes and quantitatively provide the critical transfer printing radius in terms of material and geometric properties of the transfer printing system. This model is verified to be both correct and widely applicable by rich experimental results, providing a new and reliable theory for the fabrication of inorganic flexible electronics. To understand the role of paddy soils in the global carbon cycle, it is necessary to analyze the characteristics of the organic carbon pool at different soil depths. It was hypothesized that soil organic carbon fractions including labile organic carbon fraction I (LOCF-I), labile organic carbon fraction II (LOCF-II), and recalcitrant organic carbon (ROC) distributed differently within the soil profile. In this study, soil was collected from 27 typical rice fields in Guangdong Province, south China. The carbon fractions of the paddy field soils were analyzed and compared over a 0–60 cm depth profile. The relationship between carbon content and the physical and chemical properties of the soils was further analyzed using correlation analysis and structural equation modeling. The results showed that soil total organic carbon concentration in paddy fields was increased by 22.1% during the last four decades. In the soil organic carbon pool of 0–60 cm profile, the proportion of 67.31 to 70.31% in ROC, 21.75 to 22.06% in LOCF-I, and 7.7 to 10.63% was recorded, respectively, indicating that ROC was the dominating fraction. Storage of soil total organic carbon and fractions all decreased with the increase in soil depth. Correlation and path analysis showed that total nitrogen was the main driving factor affecting the soil carbon fractions, whereas pH and soil bulk density indirectly affected the content of carbon fractions by influencing total nitrogen. Background: Biliary tract cancers have garnered significant attention due to their highly malignant nature. The relationship between abnormal lipid metabolism and tumor occurrence and development is a research hotspot. However, its correlation with biliary tract cancers is unclear. Methods: We enrolled 78 patients with biliary tract cancers and obtained data on clinical characteristics, pathological findings, and preoperative blood lipid indices, including total cholesterol (TC), high‐density lipoprotein cholesterol (HDL‐C), low‐density lipoprotein cholesterol (LDL‐C), triglycerides (TG), and lipoprotein (a) [Lp(a)]. Receiver operating characteristic (ROC) curves were used to determine the optimal predictive cutoff values of lipid indicators among the participants. Independent risk factors were determined using Cox regression, and survival was predicted using the Kaplan–Meier method. Statistical analyses were performed using SPSS software. Results: Univariate Cox regression analysis revealed that the body mass index (BMI), tumor location, surgical margin, N stage, and abnormally increased LDL‐C, TG, and Lp(a) levels were significantly associated with poor prognosis of biliary tract cancers (p < 0.05). Multifactor Cox regression demonstrated that only N stage (HR = 3.393, p < 0.001) and abnormally increased Lp(a) levels (HR = 2.814, p = 0.004) were significantly associated with shorter survival. N stage and Lp(a) were identified as independent prognostic risk factors for patients with biliary tract cancers. Conclusion: This study presents Lp(a) as a novel biochemical marker that can guide clinical treatment strategies for patients with biliary tract cancers. More effective treatment options and intensive postoperative testing should be considered to prolong the survival of these patients with preoperative abnormal lipid metabolism. [ABSTRACT FROM AUTHOR]
