Abstract Background Dendrobium nobile and Dendrobium chrysotoxum are important species of the genus Dendrobium and have great economic and medicinal value. However, the medicinal properties of these two plants remain poorly understood. This study aimed to investigate the medical properties of D. nobile and D. chrysotoxum by conducting a comprehensive chemical profiling of the two plants. Additionally, active compounds and predictive targets for anti-hepatoma activity in D. chrysotoxum extracts were identified using Network Pharmacology. Results Chemical profiling showed that altogether 65 phytochemicals were identified from D. nobile and D. chrysotoxum, with major classes as alkaloids, terpenoids, flavonoids, bibenzyls and phenanthrenes. About 18 compounds were identified as the important differential metabolites in D. nobile and D. chrysotoxum. Furtherly, CCK-8 results showed that the extracts of stems and leaves of D. nobile and D. chrysotoxum could inhibit the growth of Huh-7 cells, and the anti-hepatoma activity of extracts were dose-dependent. Among the extracts, the extract of D. chrysotoxum showed significant anti-hepatoma activity. In order to find the potential mechanism of anti-hepatoma activity of D. chrysotoxum, five key compounds and nine key targets were obtained through constructing and analyzing the compound-target-pathway network. The five key compounds were chrysotobibenzyl, chrysotoxin, moscatilin, gigantol and chrysotoxene. Nine key targets, including GAPDH, EGFR, ESR1, HRAS, SRC, CCND1, HIF1A, ERBB2 and MTOR, could be considered as the core targets of the anti-hepatoma activity of D. chrysotoxum. Conclusions In this study, the chemical composition difference and anti-hepatoma activity of stems and leaves of D. nobile and D. chrysotoxum were compared, and the potential anti-hepatoma mechanism of D. chrysotoxum was revealed in a multi-target and multi-pathway manner.

Simple Summary: In order to improve the comprehensive utilization of oil tea seed meal and increase the comprehensive economic benefits of the oil tea industry. In this paper, the low eutectic solvent was used as the extraction system, and the ultrasonic extraction method was combined with the enzymatic method to jointly extract the tea saponins. It was further purified with a large-pore adsorption resin to develop a set of environmentally friendly, energy-saving, and efficient tea saponin extraction and purification processes. On this basis, the tea saponins extracted from oil tea seed meal were further analyzed for antioxidant capacity and bacteriostatic activity. The results provided the scientific experimental basis and theoretical support for the efficient use of oil tea by-products. Tea saponins have high surface-active and biological activities and are widely used in chemicals, food, pharmaceuticals, and pesticides. Tea saponins are usually extracted using ethanol or water, but both methods have their disadvantages, including a negative impact on the environment, high energy consumption, and low purity. In this study, we explored an effective process for extracting tea saponins from tea meal using deep eutectic solvents combined with ultrasonic extraction and enzymatic techniques. The experimental results showed that a high extraction efficiency of 20.93 ± 0.48% could be achieved in 20 min using an ultrasonic power of 40% and a binary DES consisting of betaine and ethylene glycol (with a molar ratio of 1:3) at a material–liquid ratio of 1:35 and that the purity of the tea saponins after purification by a large-pore adsorption resin reached 95.94%, which was higher than that of commercially available standard tea saponin samples. In addition, the extracted tea saponins were evaluated for their antioxidant and bacteriostatic activities using chemical and biological methods; the results showed that the tea saponins extracted using these methods possessed antioxidant properties and displayed significant antibacterial activity. Therefore, the present study developed a method for using deep eutectic solvents as an environmentally friendly technological solution for obtaining high-purity tea saponins from tea meal oil. Objectives: The differences in chemical constituents in different parts of Tetrastigma hemsleyanum were analyzed. Methods: The contents of total phenols, total flavonoids, and polysaccharides in methanol extracts from different parts of T. hemsleyanum in Zhejiang were determined, and the chemical components of different parts of T. hemsleyanum were collected by ultra-performance liquid chromatography-tandem quadrupole-time-of-flight mass spectrometry. The differential index components of different parts of T. hemsleyanum were identified and analyzed by accurate mass-to-charge ratio of primary mass spectrometry and fragment ion of secondary mass spectrometry with reference spectrum and software database search and related literature reports. Results: A total of 20 components were identified, including 13 identified in root tubers, 7 in fibrous roots and 8 in leaves, and the structural cleavage of its compounds was analyzed. Conclusion: It provides a possibility and theoretical basis for the future use of the whole plant of T. hemsleyanum as medicine. [ABSTRACT FROM AUTHOR]
