Abstract The successful accomplishment of the first telomere-to-telomere human genome assembly, T2T-CHM13, marked a milestone in achieving completeness of the human reference genome. The upcoming era of genome study will focus on fully phased diploid genome assembly, with an emphasis on genetic differences between individual haplotypes. Most existing sequencing approaches only achieved localized haplotype phasing and relied on additional pedigree information for further whole-chromosome scale phasing. The short-read-based Strand-seq method is able to directly phase single nucleotide polymorphisms (SNPs) at whole-chromosome scale but falls short when it comes to phasing structural variations (SVs). To shed light on this issue, we developed a Nanopore sequencing platform-based Strand-seq approach, which we named NanoStrand-seq. This method allowed for de novo SNP calling with high precision (99.52%) and acheived a superior phasing accuracy (0.02% Hamming error rate) at whole-chromosome scale, a level of performance comparable to Strand-seq for haplotype phasing of the GM12878 genome. Importantly, we demonstrated that NanoStrand-seq can efficiently resolve the MHC locus, a highly polymorphic genomic region. Moreover, NanoStrand-seq enabled independent direct calling and phasing of deletions and insertions at whole-chromosome level; when applied to long genomic regions of SNP homozygosity, it outperformed the strategy that combined Strand-seq with bulk long-read sequencing. Finally, we showed that, like Strand-seq, NanoStrand-seq was also applicable to primary cultured cells. Together, here we provided a novel methodology that enabled interrogation of a full spectrum of haplotype-resolved SNPs and SVs at whole-chromosome scale, with broad applications for species with diploid or even potentially polypoid genomes.

Abstract Orthopedic conditions have emerged as global health concerns, impacting approximately 1.7 billion individuals worldwide. However, the limited understanding of the underlying pathological processes at the cellular and molecular level has hindered the development of comprehensive treatment options for these disorders. The advent of single-cell RNA sequencing (scRNA-seq) technology has revolutionized biomedical research by enabling detailed examination of cellular and molecular diversity. Nevertheless, investigating mechanisms at the single-cell level in highly mineralized skeletal tissue poses technical challenges. In this comprehensive review, we present a streamlined approach to obtaining high-quality single cells from skeletal tissue and provide an overview of existing scRNA-seq technologies employed in skeletal studies along with practical bioinformatic analysis pipelines. By utilizing these methodologies, crucial insights into the developmental dynamics, maintenance of homeostasis, and pathological processes involved in spine, joint, bone, muscle, and tendon disorders have been uncovered. Specifically focusing on the joint diseases of degenerative disc disease, osteoarthritis, and rheumatoid arthritis using scRNA-seq has provided novel insights and a more nuanced comprehension. These findings have paved the way for discovering novel therapeutic targets that offer potential benefits to patients suffering from diverse skeletal disorders.

Abstract Background The effects of female chromosomal polymorphisms (FCPs) on various aspects of reproductive health have been investigated, yet the findings are frequently inconsistent. This study aims to clarify the role of FCPs on the outcomes of in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI). Methods This retrospective cohort study comprised 951 couples with FCPs and 10,788 couples with normal karyotypes who underwent IVF/ICSI treatment at Peking University Third Hospital between 2015 and 2021. The exposure was FCPs. The embryological outcomes and clinical outcomes were compared. Results The FCPs, as a whole, compromised the oocyte maturation rate (76.0% vs. 78.8%, P = 0.008), while they did not adversely affect other IVF/ICSI outcomes. Further detailed analyses showed that every type of FCPs contributed to the lower oocyte maturation rate, particularly the rare FCPs (69.0% vs. 78.8%, P = 0.008). The female qh + was associated with a higher normal fertilization rate (63.0% vs. 59.2%, adjusted P = 0.022), a higher clinical pregnancy rate (37.0% vs. 30.7%, adjusted P = 0.048), and a higher live birth rate (27.0% vs.19.0%, adjusted P = 0.003) in couples undergoing IVF. Conversely, in couples undergoing ICSI, female qh + was found to be related to a lower normal fertilization rate (58.8% vs. 63.8%, P = 0.032), a comparable clinical pregnancy rate (25.7% vs. 30.9%, P = 0.289), and a comparable live birth rate (19.8% vs. 19.2%, P = 0.880) compared to the control group. Abstract Fluorescent‐patterned materials are widely used in information storage and encryption. However, preparing a patterned fluorescent display on a matrix currently requires a time‐consuming (hours or even days) and complex multi‐step process. Herein, a rapid and mild technique developed for the in‐situ controllable synthesis of fluorescent nitrogen‐doped carbon dots (NCDs) on eco‐friendly transparent wood films (TEMPO‐oxidized carboxyl wood film [TOWF]) within a few minutes was developed. A wood skeleton was employed as the carbon precursor for NCD synthesis as well as the matrix for the uniform and controlled distribution of NCDs. Moreover, the in‐situ synthesis mechanism for preparing NCDs in TOWF was proposed. The resulting fluorescent wood films have excellent tensile strength (310.00 ± 15.57 MPa), high transmittance (76.2%), high haze (95.0%), UV‐blocking properties in the full ultraviolet (UV) range, and fluorescent performance that can be modified by changing the heating parameters. Fluorescent patterning was simply achieved by regulating the in‐situ NCD synthesis regions, and the fluorescent patterns were formed within 10 s. These fluorescent‐patterned wood films can effectively store and encrypt information, and they can interact with external information through a transparent matrix. This work provides a green and efficient strategy for fabricating fluorescent information storage and encryption materials.

Osteochondral defect (OCD) is a common but challenging condition in orthopaedics that imposes huge socioeconomic burdens in our aging society. It is imperative to accelerate the R&D of regenerative scaffolds using osteochondral tissue engineering concepts. Yet, all innovative implant-based treatments require animal testing models to verify their feasibility, biosafety, and efficacy before proceeding to human trials. Rabbit models offer a more clinically relevant platform for studying OCD repair than smaller rodents, while being more cost-effective than large animal models. The core-decompression drilling technique to produce full-thickness distal medial femoral condyle defects in rabbits can mimic one of the trauma-relevant OCD models. This model is commonly used to evaluate the implant’s biosafety and efficacy of osteochondral dual-lineage regeneration. In this article, we initially indicate the methodology and describe a minimally-invasive surgical protocol in a step-wise manner to generate a standard and reproducible rabbit OCD for scaffold implantation. Besides, we provide a detailed procedure for sample collection, processing, and evaluation by a series of subsequent standardized biochemical, radiological, biomechanical, and histological assessments. In conclusion, the well-established, easy-handling, reproducible, and reliable rabbit OCD model will play a pivotal role in translational research of osteochondral tissue engineering.

Summary: Background: Hospitals in China are classified into tiers (1, 2 or 3), with the largest (tier 3) having more equipment and specialist staff. Differential health insurance cost-sharing by hospital tier (lower deductibles and higher reimbursement rates in lower tiers) was introduced to reduce overcrowding in higher tier hospitals, promote use of lower tier hospitals, and limit escalating healthcare costs. However, little is known about the effects of differential cost-sharing in health insurance schemes on choice of hospital tiers. Methods: In a 9-year follow-up of a prospective study of 0.5 M adults from 10 areas in China, we examined the associations between differential health insurance cost-sharing and choice of hospital tiers for patients with a first hospitalisation for stroke or ischaemic heart disease (IHD) in 2009–2017. Analyses were performed separately in urban areas (stroke: n = 20,302; IHD: n = 19,283) and rural areas (stroke: n = 21,130; IHD: n = 17,890), using conditional logit models and adjusting for individual socioeconomic and health characteristics. Findings: About 64–68% of stroke and IHD cases in urban areas and 27–29% in rural areas chose tier 3 hospitals. In urban areas, higher reimbursement rates in each tier and lower tier 3 deductibles were associated with a greater likelihood of choosing their respective hospital tiers. In rural areas, the effects of cost-sharing were modest, suggesting a greater contribution of other factors. Higher socioeconomic status and greater disease severity were associated with a greater likelihood of seeking care in higher tier hospitals in urban and rural areas. Interpretation: Patient choice of hospital tiers for treatment of stroke and IHD in China was influenced by differential cost-sharing in urban areas, but not in rural areas. Further strategies are required to incentivise appropriate health seeking behaviour and promote more efficient hospital use. Abstract Knowledge tracking is to analyze the mastery of students' knowledge through the learning track. This is very important for online education, since it can determine a learner’s current knowledge level by analyzing the learning history and then make recommendations for future learning. In the past, the commonly used model for knowledge tracking is the convolutional neural network, but it has long-term sequence dependencies. With the invention of Transformer, it has excellent performance in long-sequence modeling by virtue of the attention mechanism, and is gradually introduced into the field of knowledge tracking. However, through our research, some knowledge tracking data sets have a large number of continuous and repetitive training, which will cause Transformer model to ignore the potential connections between some knowledge points. To overcome this problem, we introduce a convolutional attention mechanism to help the model perceive contextual information better. In addition, we simulate the forgetting phenomenon of students during the learning process by calculating the forgetting factor, and fuse it with the weight matrix generated by the model to improve the accuracy of the model. As a result, a Transformer-based Convolutional Forgetting Knowledge Tracking (TCFKT) model is presented in this paper. According to the experimental results conducted on the real world ASSITments2012, ASSISTments2017, KDD a, STATIC datasets, the TCFKT model outperforms other knowledge tracking models.

Abstract Regulating electron transport rate and ion concentrations in the local microenvironment of active site can overcome the slow kinetics and unfavorable thermodynamics of CO2 electroreduction. However, simultaneous optimization of both kinetics and thermodynamics is hindered by synthetic constraints and poor mechanistic understanding. Here we leverage laser-assisted manufacturing for synthesizing CuxO bipyramids with controlled tip angles and abundant nanograins, and elucidate the mechanism of the relationship between electron transport/ion concentrations and electrocatalytic performance. Potassium/OH− adsorption tests and finite element simulations corroborate the contributions from strong electric field at the sharp tip. In situ Fourier transform infrared spectrometry and differential electrochemical mass spectrometry unveil the dynamic evolution of critical *CO/*OCCOH intermediates and product profiles, complemented with theoretical calculations that elucidate the thermodynamic contributions from improved coupling at the Cu+/Cu2+ interfaces. Through modulating the electron transport and ion concentrations, we achieve high Faradaic efficiency of 81% at ~900 mA cm−2 for C2+ products via CO2RR. Similar enhancement is also observed for nitrate reduction reaction (NITRR), achieving 81.83 mg h−1 ammonia yield rate per milligram catalyst. Coupling the CO2RR and NITRR systems demonstrates the potential for valorizing flue gases and nitrate wastes, which suggests a practical approach for carbon-nitrogen cycling.

Lakes on the Tibetan Plateau (TP) serve as both indicators of and safeguards against climate change, playing a crucial role in the aquatic ecosystems of the TP. While considerable attention has been devoted to studying the thermal and dynamic processes of TP lakes, research focusing on their ecological variations has been limited. In this study, we selected Namco, a representative lake on the TP, to investigate its water ecological processes using the AQUATOX lake ecological model. Long-term ecological variations spanning from 1980 to 2020 were analyzed based on lake observations. Our results revealed a consistent increase in water nutrients, particularly total nitrogen (WTN), and total phosphorus (WTP), over the study period. Additionally, the concentrations of chlorophyll-a (Chl-a) and water gross and net primary production (WGPP and WNPP) exhibited a significant upward trend. Despite the persistent state of poor nutrition in the lake, the ecological conditions improved. Multiple linear regression analysis indicated that the concentrations of WGPP, WNPP, and Chl-a were more sensitive to the local climate and hydrology compared to WTN and WTP. A continuously warming climate would heat up the lake water body, further enhancing primary production and improving water quality in the future. This study provides insights for lake limnological and ecological research and can be used to inform water management strategies in high-altitude alpine regions.

Ultra-low Earth orbit (LEO) satellites are widely used in the military, remote sensing, scientific research, and other fields. The ultra-LEO satellite faces the harsh aerothermal environment, and the complex variable attitude task requires the radiator of the satellite to not only meet the heat dissipation requirements of the load but also to resist aerothermal flux. In this study, the aerothermal flux of 160–110 km was calculated, and the loop heat pipe (LHP) coupled with a thermo-electric cooler (TEC) and multi-layer insulation (MLI) were applied to ultra-LEO satellites to determine the variable switching and fast response of heat dissipation and heat protection. An aerothermal flux simulation test platform was built. After the assessment of the ultra-LEO aerothermal flux test, even when the head temperature was as high as 350 °C and the side radiator temperature was as high as 160 °C, the temperature of the internal heat source could be controlled within 22.5 °C through the efficient work of the thermal variable switch system. The study confirms the accuracy and feasibility of the system, which provides an important reference for the subsequent actual on-orbit mission.

In the domain of space rendezvous and docking, visual navigation plays a crucial role. However, practical applications frequently encounter issues with poor image quality. Factors such as lighting uncertainties, spacecraft motion, uneven illumination, and excessively dark environments collectively pose significant challenges, rendering recognition and measurement tasks during visual navigation nearly infeasible. The existing image enhancement methods, while visually appealing, compromise the authenticity of the original images. In the specific context of visual navigation, space image enhancement’s primary aim is the faithful restoration of the spacecraft’s mechanical structure with high-quality outcomes. To address these issues, our study introduces, for the first time, a dedicated unsupervised framework named SpaceLight for enhancing on-orbit navigation images. The framework integrates a spacecraft semantic parsing network, utilizing it to generate attention maps that pinpoint structural elements of spacecraft in poorly illuminated regions for subsequent enhancement. To more effectively recover fine structural details within these dark areas, we propose the definition of a global structure loss and the incorporation of a pre-enhancement module. The proposed SpaceLight framework adeptly restores structural details in extremely dark areas while distinguishing spacecraft structures from the deep-space background, demonstrating practical viability when applied to visual navigation. This paper is grounded in space on-orbit servicing engineering projects, aiming to address visual navigation practical issues. It pioneers the utilization of authentic on-orbit navigation images in the research, resulting in highly promising and unprecedented outcomes. Comprehensive experiments demonstrate SpaceLight’s superiority over state-of-the-art low-light enhancement algorithms, facilitating enhanced on-orbit navigation image quality. This advancement offers robust support for subsequent visual navigation.

The hydrogeochemical research of groundwater in the Bieletan area, China’s largest potash producing zone, used a variety of methods, including multivariate analysis, saturation index, and hydrogeochemical modeling. Water samples were collected and analyzed for physicochemical parameters, along with soluble ions from soil cores. The results showed that total dissolved solids (TDS) of groundwater exceeded 300 g/L, with the main hydrochemical characteristics being Cl-Mg type and Cl-Na type. Groundwater is recharged by lake water and canal water, with evaporation being the main factor affecting water chemistry. Hydrogeochemical modeling analyzed the processes occurring from these two different recharge sources: mineral precipitation mainly occurred with lake water recharge, while mineral dissolution mainly occurred with canal water recharge. Regarding potash dissolution, canal water and lake water recharge resulted in 8.860 mmol/L of polyhalite dissolution and 0.278 mmol/L of carnallite dissolution, respectively. This study highlights the complex hydrogeochemical processes controlling groundwater in the potash-rich Bieletan area, providing insights for water resource management and potash mining.

Abstract Inhibin subunit beta A (INHBA) is a member of the transforming growth factor-beta (TGF-β) superfamily that plays a fundamental role in various cancers. However, a systematic analysis of the exact role of INHBA in patients with gastric cancer (GC) has not yet been conducted. We evaluated the expression levels of INHBA and the correlation between INHBA and GC prognosis in GC. The relationship between INHBA expression, immune infiltration levels, and type markers of immune cells in GC was also explored. In addition, we studied INHBA mutations, promoter methylation, and functional enrichment analysis. Besides, high expression levels of INHBA in GC were significantly related to unfavorable prognosis. INHBA was negatively correlated with B cell infiltration, but positively correlated with macrophage and most anticancer immunity steps. INHBA expression was positively correlated with the type markers of CD8+ T cells, neutrophils, macrophages, and dendritic cells. INHBA has a weak significant methylation level change between tumor and normal tissues and mainly enriched in cancer-related signaling pathways. The present study implies that INHBA may serve as a potential biomarker for predicting the prognosis of patients with GC. INHBA is a promising predictor of immunotherapy response, with higher levels of INHBA indicating greater sensitivity.

Ni625/WC composite coatings added with different amounts of Y 2O 3were prepared on the surface of 304 stainless steels by laser cladding. This study focused on the microstructure characteristics, microhardness, and corrosion performances of Ni625/WC composite coatings. The results showed that Y 2O 3 can effectively improve the corrosion resistance of the composite coatings. The microstructure from the bottom to the surface of composite coatings consists of plane crystal, cellular crystal, columnar crystal and equiaxed crystal. The Y 2O 3content of optimum composite coating was 1.0%. Its microhardness was three times that of matrix material. In addition, the corrosion current density of the composite coating was only 2% of Ni625/WC coating, which was attributed to the good properties of Y 2O 3 and appropriate Y 2O 3 refined microstructure.

Abstract Incorporating knowledge graphs into recommendation systems has attracted wide attention in various fields recently. A Knowledge graph contains abundant information with multi-type relations among multi-type nodes. The heterogeneous structure reveals not only the connectivity but also the complementarity between the nodes within a KG, which helps to capture the signal of potential interest of the user. However, existing research works have limited abilities in dealing with the heterogeneous nature of knowledge graphs, resulting in suboptimal recommendation results. In this paper, we propose a new recommendation method based on iterative heterogeneous graph learning on knowledge graphs (HGKR). By treating a knowledge graph as a heterogeneous graph, HGKR achieves more fine-grained modeling of knowledge graphs for recommendation. Specifically, we incorporate the graph neural networks into the message passing and aggregating of entities within a knowledge graph both at the graph and the semantic level. Furthermore, we designed a knowledge–perceiving item filter based on an attention mechanism to capture the user’s potential interest in their historical preferences for the enhancement of recommendation. Extensive experiments conducted on two datasets in the context of two recommendations reveal the excellence of our proposed method, which outperforms other benchmark models.

Abstract Face attribute classification (FAC) has received considerable attention due to its excellent application value in bio‐metric verification and face retrieval. Current FAC methods suffer two typical challenges: complex inter‐attribute correlations and imbalanced learning. Aims at the challenges, presents an end‐to‐end FAC framework with integrated use of multiple strategies, which consists of a convolutional neural network (CNN) and a graph convolutional network (GCN). The GCN is used to model the semantic correlations among attributes and capture inter‐dependency among them. The correlation information learnt via the GCN is used to guide the learning of the inter‐dependent classification features of the FAC network. An adaptive thresholding strategy and a boosting scheme are adopted to alleviate the effect of the class‐imbalance. To deal with the task imbalance problem, a new dynamic weighting scheme is proposed to update the weight of each attribute classification task in the training process. We apply four evaluation metrics to evaluate the proposed method. Experimental results show all the proposed strategies are effective, and our approach outperforms state‐of‐the‐art FAC methods on two challenging datasets CelebA and LFWA.

Mandibular defect becomes a prevalent maxillofacial disease resulting in mandibular dysfunctions and huge psychological burdens to the patients. Considering the routine presence of oral contaminations and aesthetic restoration of facial structures, the current clinical treatments are however limited, incapable to reconstruct the structural integrity and regeneration, spurring the need for cost-effective mandibular tissue engineering. Hydrogel systems possess great merit for mandibular reconstruction with precise involvement of cells and bioactive factors. In this review, current clinical treatments and distinct mode(s) of mandible formation and pathological resorption are summarized, followed by a review of hydrogel-related mandibular tissue engineering, and an update on the advanced fabrication of hydrogels with improved mechanical property, antibacterial ability, injectable form, and 3D bioprinted hydrogel constructs. The exploration of advanced hydrogel systems will lay down a solid foundation for a bright future with more biocompatible, effective, and personalized treatment in mandibular reconstruction.

Sleep disorders affect mental and physical health. Infertile women undergoing assisted reproductive technology (ART) treatment are prone to sleep disorders. Sleep condition, its influencing factors, and the association between sleep condition and ART treatment outcomes before treatment have not been explored within a population with a large sample size. Therefore, we investigated the sleep characteristics of 1002 Chinese infertile women before ovulation induction and investigated the influencing factors (negative and positive psychological factors, demographics, and fertility characteristics). We also examined whether sleep conditions before treatment predicted reproductive outcomes. We found that 24.1% of participants reported poor sleep quality. Women with primary infertility reported poorer sleep than women with secondary infertility. Negative psychological factors, including depression, anxiety, and perceived stress were associated with poor sleep, whereas positive affect was linked with good sleep. Adverse sleep characteristics, including poor subjective sleep quality, sleep disturbances, and poor sleep efficiency, decreased the quantity and quality of oocytes retrieved, fertilization rates, and clinical pregnancy rates. This study indicates that before ART treatment, a large number of females with infertility suffer from sleep problems, which are affected by psychological factors and infertility type, and unhealthy sleep characteristics may impair treatment outcomes. Our findings highlight the importance of screening and treatment for sleep disorders before the enrollment of ART treatment in infertile women.

IntroductionIntercropping and straw mulching are sustainable agricultural practices that can positively affect crop growth and development, especially together.MethodsA split-plot experimental design was used to investigate the effects of intercropping and straw mulching on crop growth, crop yield, nitrogen uptake, and photosynthetic characteristics. The main plot focused on three planting patterns: soybean monoculture (S), maize monoculture (M), and maize/soybean intercropping (I). The subplot structure consisted of four levels of straw mulching (0, 4.8, 7.2, 9.6 t ha-1).ResultsInteraction and variance analyses showed that straw mulching, intercropping, and their interaction had significant effects on plant height, stem diameter, leaf area index, chlorophyll content, nitrogen uptake, photosynthetic characteristics, and crop yield. Based on two-year averages for maize and soybean, the net photosynthetic rate (Pn) was up to 51.6% higher, stomatal conductance (Sc) was up to 44.0% higher, transpiration rate (Tr) was up to 46.6% higher, and intercellular carbon dioxide concentration (Ci) was up to 25.7% lower relative to no mulching. The maximum increases of Pn, Sc, and Tr of intercropped maize were 15.48%, 17.28%, and 23.94%, respectively, and the maximum Ci was 17.75% lower than that of monoculture maize. The maximum increase of Pn, Sc, and Tr of monoculture soybean was 24.58%, 16.90%, and 17.91%, respectively, and the maximum Ci was 13.85% lower than that of intercropped soybean. The nitrogen uptake of maize and soybean in the mulching treatment was 24.3% higher than that in the non-mulching treatment; the nitrogen uptake of intercropped maize was 34.2% higher than that of monoculture maize, and the nitrogen uptake of monoculture soybean was 15.0% higher than that of intercropped soybean. The yield of maize and soybean in the mulching treatment was 66.6% higher than that in the non-mulching treatment, the maize yield under intercropping was 15.4% higher than that under monoculture, and the yield of monoculture soybean was 9.03% higher than that of intercropped soybean.DiscussionThe growth index and photosynthesis of crops are important parts of yield formation. The results of this study confirmed that straw mulching, intercropping, and their interaction can ultimately increase crop yield by improving crop growth, nitrogen uptake, and photosynthesis. This result can be used as the theoretical basis for the combined application of these measures in agriculture.

Detecting changes in two remote sensing images of the same region but at different times is of great significance in applications, such as land management and urban planning, which also prompts the continuous development and progress of change detection (CD) technology. The current deep learning-based methods make full use of the excellent feature learning ability of deep convolution to show excellent detection performance. However, advances in remote sensing technology also mean that detected objects have higher resolution and more complex content, which is more challenging for CD techniques. Strengthening the model's ability to learn the context of the detected remote sensing image can effectively improve the model's ability to distinguish between changing features and nonchanging features, thereby achieving higher-precision detection results. In order to explore and utilize the contextual information of different levels of features as much as possible, we design a dual-attention cross fusion module in our method to realize the cross-learning of contextual information of different scales during the decoding process. It will be able to complementarily fuse feature content of different granularities. We also propose an Atrous Pyramid Difference Module (APDM) to efficiently capture the difference information of two refined features by exploiting receptive fields of different sizes. In addition, in order to further improve the context modeling ability of the model, we introduce a context transformer block (Cot). Different from other transformer-based self-attention methods, Cot dynamically guides the learning of the attention matrix by the contextual information of the input keys. Our method achieves F1-scores of 91.08%/91.93%/79.80% on the LEVIR-CD/WHU-CD/DSIFN-CD datasets, respectively. Extensive qualitative and quantitative experiments on these datasets validate the effectiveness of our method.

Abstract Quantifying carbon dioxide (CO2) emissions from China’s wood and bamboo processing industry is associated with China’s emissions reduction targets, as well as mitigating global climate change. This study employed the Intergovernmental Panel on Climate Change Tier-2 methodology to investigate spatio-temporal evolution characteristics of carbon dioxide emission from the wood and bamboo processing industry in China from 2000 to 2019. The results showed that energy consumption reached a maximum value of 312,900.35 TJ in 2012. Energy consumption has been gradually transformed from raw coal to electricity and other clean energy. Energy intensity dropped from 1.39 TJ per million yuan of corrected production value in 2000 to 0.15 TJ per million yuan of corrected production value in 2019. Accordingly, CO2 emissions reached their peak value of 31,148.1 thousand tons of CO2 in 2012. Raw coal and electricity had profound impacts on CO2 emissions. The CO2 emission intensity declined from 140.04 tons CO2 per million yuan of corrected production value in 2000 to 19.62 tons CO2 per million yuan of corrected production value in 2019. We conclude that China’s wood and bamboo processing sector is a green, low-carbon industry. The spatial distribution pattern of CO2 emissions is highly consistent with the industrial spatial layout. Furthermore, several mitigation paths were put forward.

In order to enhance the degree of binding reaction of TiO2 in titanium-containing ceramic glazes and prevent the reaction of its transformation into rutile to eliminate the yellowing phenomenon of the glaze surface, an apatite–TiO2 composite opacifier (ATO) was prepared through the mechanical grinding of hydroxyapatite and anatase TiO2. The properties, opacification mechanism, and yellowing inhibition of the prepared ceramic glazes were studied. The results show that the ATO is characterized by a uniform coating of TiO2 on the surface of the apatite and the formation of close chemical bonding between the apatite and TiO2. The ceramic glaze surface when using an ATO has a white appearance and excellent opacification performance. When an ATO was used, the L*, a*, and b* values of the glaze were 89.99, −0.85, and 3.37, respectively, which were comparable to those of a ZrSiO4 glaze (L*, a*, and b* were 88.24, −0.02, and 2.29, respectively). The opacification of the glaze was slightly lower than that of the TiO2 glaze (L* value was 92.13), but the appearance changed from yellow to the white of the TiO2 glaze (b* value was 9.18). The ceramic glaze layer when using an ATO mainly consists of titanite, glass phase, and a small amount of quartz, and the opacification mechanism is the crystallization of the generated titanite. ATOs can play an active role in solving the critical problem that arises when TiO2 replaces ZrSiO4 as an opacifier.

Abstract The knowledge of osteoarthritis (OA) has nowadays been extended from a focalized cartilage disorder to a multifactorial disease. Although recent investigations have reported that infrapatellar fat pad (IPFP) can trigger inflammation in the knee joint, the mechanisms behind the role of IPFP on knee OA progression remain to be defined. Here, dysregulated osteopontin (OPN) and integrin β3 signaling are found in the OA specimens of both human and mice. It is further demonstrated that IPFP‐derived OPN participates in OA progression, including activated matrix metallopeptidase 9 in chondrocyte hypertrophy and integrin β3 in IPFP fibrosis. Motivated by these findings, an injectable nanogel is fabricated to provide sustained release of siRNA Cd61 (RGD−Nanogel/siRNA Cd61) that targets integrins. The RGD−Nanogel possesses excellent biocompatibility and desired targeting abilities both in vitro and in vivo. Local injection of RGD−Nanogel/siRNA Cd61 robustly alleviates the cartilage degeneration, suppresses the advancement of tidemark, and reduces the subchondral trabecular bone mass in OA mice. Taken together, this study provides an avenue for developing RGD−Nanogel/siRNA Cd61 therapy to mitigate OA progression via blocking OPN‐integrin β3 signaling in IPFP.

Green ceramsite concrete is an environmentally friendly sound-absorbing material for high-speed railway sound barriers. However, given its low sound absorption, this material is not widely used. Therefore, this study proposes a triple-hole structure to improve the sound absorption of green ceramsite concrete. The triple-hole structure comprised micro-sized holes in the ceramsite, medium-sized holes between the ceramsite, and surface macro-sized holes. The surface macro-sized holes were obtained by changing the mold shape, and the other holes were formed by formula control. The influence of the surface macro-sized holes on the acoustic performance of the triple-hole structure was analyzed using an impedance tube test. Results showed that the sound absorption coefficient increased when the hole diameter decreased or the surface porosity increased. The optimal surface macro-sized holes increased the average sound absorption coefficient by 83 %. The surface macro-sized holes improved the acoustic performance by absorbing the reflection of incident sound and not by resonance effect. This effect was attenuated when the relative surface area was increased by more than 100 %. Compared with the ANN method, the finite element simulation method cannot accurately reflect the acoustic effects of the triple hole structure. Back propagation neural network was used to predict the effect and can realize an accuracy of 95.5 %. Thus, this study proposes a reference for the design of sound-absorbing concrete based on frequency.

Diabetic kidney disease (DKD) is a leading factor in end-stage renal disease. The complexity of its pathogenesis, combined with the limited treatment efficacy, necessitates deeper insights into potential causes. Studies suggest that ferroptosis-driven renal tubular damage contributes to DKD's progression, making its counteraction a potential therapeutic strategy. Quercetin, a flavonoid found in numerous fruits and vegetables, has demonstrated DKD mitigation in mouse models, though its protective mechanism remains ambiguous. In this study, we delved into quercetin’s potential anti-ferroptotic properties, employing a DKD rat model and high glucose (HG)-treated renal tubular epithelial cell models. Our findings revealed that HG prompted unusual ferroptosis activation in renal tubular epithelial cells. However, quercetin counteracted this by inhibiting ferroptosis and activating NFE2-related factor 2 (Nrf2) expression in both DKD rats and HG-treated HK-2 cells, indicating its renal protective role. Further experiments, both in vivo and in vitro, validated that quercetin stimulates Nrf2. Thus, our research underscores quercetin’s potential in DKD treatment by modulating the ferroptosis process via activating Nrf2 in a distinct DKD rat model, offering a fresh perspective on quercetin’s protective mechanisms.

Glioma grading is critical for treatment selection, and the fine classification between glioma grades II and III is still a pathological challenge. Traditional systems based on a single deep learning (DL) model can only show relatively low accuracy in distinguishing glioma grades II and III. Introducing ensemble DL models by combining DL and ensemble learning techniques, we achieved annotation-free glioma grading (grade II or III) from pathological images. We established multiple tile-level DL models using residual network ResNet-18 architecture and then used DL models as component classifiers to develop ensemble DL models to achieve patient-level glioma grading. Whole-slide images of 507 subjects with low-grade glioma (LGG) from the Cancer Genome Atlas (TCGA) were included. The 30 DL models exhibited an average area under the curve (AUC) of 0.7991 in patient-level glioma grading. Single DL models showed large variation, and the median between-model cosine similarity was 0.9524, significantly smaller than the threshold of 1.0. The ensemble model based on logistic regression (LR) methods with a 14-component DL classifier (LR-14) demonstrated a mean patient-level accuracy and AUC of 0.8011 and 0.8945, respectively. Our proposed LR-14 ensemble DL model achieved state-of-the-art performance in glioma grade II and III classifications based on unannotated pathological images.

Microorganisms play very important roles in carcinogenesis, tumor progression, and resistance upon treatment. Due to the challenge of accurately acquiring samples and quantifying low-biomass tissue microorganisms, most studies have focused on the effect of gut microorganisms on cancer treatments, especially the efficacy of immunotherapy. Although recent publications reveal the potential interactions between intratumor microorganisms and the immune microenvironment, whether and to what extent the intratumor microorganism could affect progression and treatment outcome remain controversial. This study is aiming to evaluate the associations among intratumor microorganisms, DNA methylation cancer driver genes, immune response, and clinical outcomes from a pan-cancer perspective, using 6,876 TCGA samples across 21 cancer types. We revealed that tumor microorganism dysbiosis is closely associated with the abnormal tumor methylome and/or tumor microenvironment, which might serve to enhance the proliferation ability and fitness for the therapy of tumors. These findings shed the light on a better understanding of the interactions between tumor cells and carcinogens during and after tumor formation, as well as microorganism-associated methylation alterations that could further serve as biomarkers for clinical outcome assessment.

Abstract A torque motor is a key component that directly influences the dynamic performance of two‐dimensional (2D) cartridge servo valves. Considering the effects of eddy current, a magnetic circuit model of a torque motor is proposed, and the eddy current resistance value considering the skin effect is calculated. When the eddy current effect is ignored, the natural frequency and critical damping of a torque motor are less than the calculated values under the eddy current effect, so the eddy current influence can be assumed to increase torque motor damping, as the iron loss can be equivalent to the damping energy consumption. Test rigs were set up and experiments were conducted based on a torque motor prototype. Through a finite element simulation analysis, the eddy current effect of the armature was found to be greater than that of the yokes. The magnetic leakage coefficients of the permanent magnets were calculated, and the obtained values were compared through experiments and simulations. Both the numerical simulations and experimental results showed that the eddy current model is closer to actual working conditions than the model for which the eddy current loss was ignored.

Abstract Background As a vital osmoticum, proline has an important role in enhancing the tolerance of plants to environmental stress. It is unclear whether the application of exogenous proline can improve the tolerance of Brassica juncea to cadmium (Cd). Results This study investigated the effects of different concentrations of proline (20, 40, 60, 80, and 100 mg/L) under Cd stress at different times (0 d, 2 d, and 7 d) on the growth and physiology of B. juncea. Treatment with exogenous proline not only increased the content of proline in B. juncea but also alleviated Cd-induced seedling growth inhibition via the maintenance of higher photosynthetic pigment content and cell viability and a decrease in the content of Cd. Moreover, it increased the activities of antioxidant enzymes and the glutathione/glutathione disulfide ratio to reduce the accumulation of reactive oxygen species. Compared with other concentrations, 60 mg/L of exogenous proline was the most effective at mitigating Cd toxicity in B. juncea. Conclusions Exogenous proline treatment enhanced the tolerance to Cd via a decrease in Cd accumulation and reestablishment of the redox homeostasis in B. juncea.

How to recover geometric transformations is one of the most challenging issues in image registration. To alleviate the effect of large geometric distortion in multimodal remote sensing image registration, a scale and rotate transform prediction net is proposed in this paper. First, to reduce the scale between the reference and sensed images, the image scale regression module is constructed via CNN feature extraction and FFT correlation, and the scale of sensed image can be recovered roughly. Second, the rotation estimate module is developed for predicting the rotation angles between the reference and the scale-recovered images. Finally, to obtain the accurate registration results, LoFTR is employed to match the geometric-recovered images. The proposed registration network was evaluated on GoogleEarth, HRMS, VIS-NIR and UAV datasets with contrast differences and geometric distortions. The experimental results show that the number of correct matches of our model reached 74.6%, and the RMSE of the registration results achieved 1.236, which is superior to the related methods.

In order to elucidate the importance of grafting in the compatibilization process of silane coupling agents, poplar veneer was treated with silane coupling agents and grafted under different heating conditions. The treated veneers were used composited with PE film to prepare different plywood samples. XPS and WCA were used to analyze the effect of grafting conditions on the surface properties of the silane-treated veneer. The results showed that free silanols can physically be adsorbed onto all silane-treated veneer surfaces, forming hydrogen–Si–O–Si– bonds and therefore increasing the water contact angle. Only under heating conditions could the –Si–O–Si– be converted into covalent –Si–O–C– bonds, which helped to improve the bonding strength. When silane-treated veneer was grafted at 120 °C for 90 min, the tensile shear strength of plywood reached 1.03 MPa, meeting the requirements of GB/T 9846.3-2004 for outdoor materials. Enhanced interlock between silane-modified veneer and PE film was observed under the optimal grafting condition by SEM. The better interface structure allowed improvement of thermal stability. DMA results showed that the retention rate in storage modulus at 130 °C was 60% for the grafted sample, while the retention rate for the ungrafted sample was only 31%.

Magnetic current imaging is deemed an emerging powerful technique for visualizing electrical currents in electronic devices. However, the existing magnetic-field-based Fourier Transform back-evolution method is limited by its mono-function of imaging the magnitude of current density in devices under test, and subject to background noise distortion. Here, we developed a novel vectorial current density imaging method based on the detection of the magnetic field gradient generated by current carrying conductors. A closed form solution of current density inversion was analytically derived and numerically verified. Experiments were conducted by scanning tri-axial fluxgate sensor over different shapes of electrical wires. The results show that a current density resolution of 24.15 mA/mm2, probe-to-sample separation of 2 mm, and spatial resolution of 0.69 mm were achieved over a maximum scanning area of 300 mm × 300 mm. Such a method is verified to be capable of simultaneously imaging both magnitude and directions of current density, which is a promising technique for in situ noninvasive inspection for the power electronic and semiconductor industry.

Glaciers are crucial water resources for arid inland rivers in Northwest China. In recent decades, glaciers are largely experiencing shrinkage under the climate-warming scenario, thereby exerting tremendous influences on regional water resources. The primary role of understudying watershed scale glacier changes under changing climatic conditions is to ensure sustainable utilization of regional water resources, to prevent and mitigate glacier-related disasters. This study maps the current (2020) distribution of glacier boundaries across the Kaidu-Kongque river basin, south slope of Tianshan Mountains, and monitors the spatial evolution of glaciers over five time periods from 2000–2020 through thresholded band ratios approach, using 25 Landsat images at 30 m resolution. In addition, this study attempts to understand the role of climate characteristics for variable response of glacier area. The results show that the total area of glaciers was 398.21 km2 in 2020. The glaciers retreated by about 1.17 km2/a (0.26%/a) from 2000 to 2020. The glaciers were reducing at a significantly rapid rate between 2000 and 2005, a slow rate from 2005 to 2015, and an accelerated rate during 2015–2020. The meteorological data shows slight increasing trends of mean annual temperature (0.02°C/a) and annual precipitation (2.07 mm/a). The correlation analysis demonstrates that the role of temperature presents more significant correlation with glacier recession than precipitation. There is a temporal hysteresis in the response of glacier change to climate change. Increasing trend of temperature in summer proves to be the driving force behind the Kaidu-Kongque basin glacier recession during the recent 20 years.© 2021 China Geology Editorial Office.

Guillain–Barré syndrome (GBS) is a potentially life-threatening post-infectious autoimmune disease characterized by rapidly progressive symmetrical weakness of the extremities. Herein, we report a case of GBS associated with drug poisoning complicated by Klebsiella pneumoniae infection. A 38-year-old woman was admitted to the intensive care unit after taking an overdose of amitriptyline and was later diagnosed with coma, Klebsiella pneumoniae infection, and septic shock. Thirteen days after admission, she was diagnosed with GBS based on acute muscle pain, flaccid paralysis, hyporeflexia, reduced amplitude of compound muscle action potential, and albuminocytologic dissociation in the cerebrospinal fluid. GBS rarely occurs after a drug overdose and septic shock, and this is the first report of a rapidly progressive GBS following amitriptyline overdose and severe Klebsiella pneumoniae infection.

Although a natural anti-inflammatory ingredient, isoliquiritigenin (ISL), plays an effective role in ulcerative colitis (UC) treatment, a series of drawbacks still limit its clinical application, including the poor solubility, instability in gastrointestinal tract, and rapid elimination rate of ISL. Zein-based NPs display the benefits on drug loading and delivery, whereas with the poor stability. In this study, an edible nano-system composed by zein/caseinate complex was fabricated for the colon-targeting delivery of ISL, to improve its colon retention and anti-UC effects. The optimized ISL loaded zein/caseinate NPs (ISL@NPs) were prepared by single-factor design by anti-solvent precipitation method, and then characterized. The improved cellular uptake of ISL@NPs on NCM460 and RAW 264.7 cells was evaluated in vitro. The colon tissue permeability and retention capacity in vivo, and the anti-UC efficacy of ISL@NPs in DSS-induce UC were implemented. As a result, ISL@NPs with the high drug loading efficiency of 9.39% ± 0.26%, the average particle diameter of 137.32 ± 2.54 nm, exhibited the pH-sensitive stability in the different simulated gastrointestinal buffer. Compared with free ISL, ISL@NPs showed significantly higher cellular uptake ability in NCM460 and RAW 264.7 cells. Based on in vivo imaging system, zein/caseinate NPs showed the prolonged colonic retention and the enhanced penetration into the colonic epithelium. Finally, the oral administration of ISL@NPs could effectively alleviate the UC-related symptoms, down-regulate the production of pro-inflammatory factors, and reduce the infiltration of macrophages and neutrophils in colon tissues. In this study, an oral colon-specific nano-system, composed with the natural compound and edible materials, was developed as the promising alternatives in the prevention and treatment of UC.

As a key region of Northeast Passage, the polynya along the Siberian coast in the East Siberian and Chukchi Seas is important to local dynamic and thermodynamic processes, sea ice production and marine ecosystem. The detailed variations of polynya and the contributions of atmospheric and oceanic factors to the polynya have not been explored quantitatively. AMSR-E satellite data from January to April during the period 2003–2011 were used to study the impacts of wind stress and ocean heat transport on variations of polynya in the East Siberian Sea and Chukchi Sea. The study region was divided into six domains. Four sets of AMSR-E data with resolutions of 6.25 km and 12.5 km were compared based on two algorithms of sea ice concentration (referred to as 6.25 km-IC and 12.5 km-IC) and sea ice thickness (referred to as 6.25 km-h and 12.5 km-h). The monthly and yearly polynya areas in the four cases and six domains had remarkable differences. The two cases of 6.25 km-h and 12.5 km-h had larger areas of polynya than the other two cases of 6.25 km-IC and 12.5 km-IC. The difference in polynya area between the 6.25 km-h and 12.5 km-h cases was much smaller than the difference between the 6.25 km-IC and 12.5 km-IC cases. The study of atmospheric and oceanic mechanisms on polynya is influenced significantly by the sensitivity of polynya areas. In general, the impact of wind stress and ocean heat transport on the polynyas had noticeable monthly and interannual variations and was dependent on the locations of the polynyas. The alongshore and offshore wind had stronger correlations with the polynya area than ocean heat transport. Although the higher resolution (6.25 km) AMSR-E data are best for the study of atmospheric and oceanic impacts on polynya area, the coarse resolution (12.5 km) AMSR-E data based on sea ice thickness can also be used. Wind direction dominated the polynya area in the East Siberian Sea and wind speed dominated the polynya area in the Chukchi Sea. The variation in ocean heat transport was influenced mainly by variation in volume transport rather than variation in water temperature.

The throughflow in the Canadian Arctic Archipelago (CAA) had a significant impact on the North Atlantic Ocean with the Arctic climate change. The findings of physical mechanisms driving the throughflow in the CAA differed and few studies about the impact of sea level pressure (SLP) on the CAA throughflow were made. A high-resolution ice-ocean coupled Arctic Ocean Finite-Volume Community Ocean Model (AO-FVCOM) was used over the period 1978–2016 to examine the interannual and seasonal variability of the outflows in the CAA and the mechanism of SLP in driving the variation of the CAA throughflow quantitively. The simulated volume transport through Davis Strait, Nares Strait, Lancaster Sound and Jones Sound showed consistent increasing trends over 1978–2016 and the larger flux in winter and spring than in summer and fall. The variation of volume transport through Nares Strait contributed more than Lancaster and Jones Sound to the variation through Davis Strait. Five process-oriented experiments were made to further explore the role of SLP in setting up and controlling the sea surface height (SSH) difference and thus the throughflow transport in the CAA. The SLP was a primary forcing to control the SSH difference and the outflow transport compared with the wind forcing. The memory of the SSH to the SLP was short and an equilibrium state could be reached if the SLP varied with time. The upstream and downstream SLP difference, however, made a slight direct contribution to driving the volume transport of the CAA throughflow. In addition to the external forcing of SLP and wind, the variability of the CAA outflow was also influenced by the variability of the inflow/outflow and SSH on boundaries connected to the Pacific and Atlantic Oceans. The choice of SLP datasets from CORE-v2, ECMWF and NCEP was sensitive to the simulated uncertainty of volume transport.

Sea ice conditions in the Canadian Arctic Archipelago (CAA) play a key role in the navigation of the Northwest Passage (NWP). Limited by observed sea ice thickness data, the research of temporal and spatial variation of sea ice in the NWP is insufficient. Based on the observed sea ice concentration and simulated thickness data, the temporal and spatial characteristics of sea ice concentration, extent and thickness from 1979 to 2017 in the NWP of the CAA were studied. The more specific pathways of the northern and southern routes of the NWP were evaluated. Against the background of the rapid retreat of Arctic sea ice, the 39-year observed sea ice concentration and extent of the NWP exhibited a relatively large decreasing trend in summer and fall, while heavy sea ice conditions were maintained in winter and spring, with slightly increasing trend in some subregions. The sea ice thickness in most subregions of the NWP showed a decreasing trend, with exception of Lancaster Sound. The sea ice thickness was larger along the northern route than the southern routes. Abstract Background Lung adenocarcinoma (LAD) is one of the most frequently diagnosed pathological categories of human lung cancer. Nevertheless, the link between long non-coding RNA (lncRNA) LINC01116 and LAD remains poorly investigated. Methods QRT-PCR and western blot were applied for quantifying the expression of RNAs and proteins. Both functional experiments assays in vitro and xenografts model in vivo were implemented for analyzing LINC01116 function in LAD while molecular relationship among RNAs was investigated via mechanism experiments. Results LINC01116 was expressed at an abnormally high level in LAD, which was induced by transcription activator EGR1. LINC01116 depletion restrained proliferation, migration and invasion, yet facilitated apoptosis of LAD cells. MiR-744-5p could bind to LINC01116. MiR-744-5p inhibitor reversed the inhibitory effects of silencing LINC01116 on LAD malignant behaviors. In addition, cell division cycle-associated protein 4 (CDCA4) shared binding sites with miR-744-5p. Silencing LINC01116 elicited decline in CDCA4 mRNA and protein levels. Moreover, CDCA4 up-regulation could counteract the biological effects of LINC01116 knockdown on LAD cells. Conclusion Our data revealed that LINC01116 promoted malignant behaviors of LAD cells by targeting miR-744-5p/CDCA4 axis, implying the theoretical potential of LINC01116 as a novel target for LAD treatment.

Soil aggregation and aggregate-associated carbon (C) play an essential function in soil health and C sequestration. Arbuscular mycorrhizal fungi (AMF) are considered to be primary soil aggregators due to the combined effect of extraradical hyphae and glomalin-related soil proteins (GRSPs). However, the effects of diversity and network complexity of AMF community on stability of soil aggregates and their associated C under long-term climate change (CC) and land-use conversion (LUC) in relatively high-latitude regions are largely unexplored. Therefore, an 8-year soil plot (with a 30-year cropping history) transplantation experiment was conducted to simulate CC and LUC from cropland to fallow land. The results showed that Glomus, Paraglomus, and Archaeospora were the most abundant genera. The diversity of AMF community in fallow land was higher than cropland and increased with increasing of mean annual temperature (MAT) and mean annual precipitation (MAP). Fallow land enhanced the network complexity of AMF community. The abundance families (Glomeraceae and Paraglomeraceae) exhibited higher values of topological features and were more often located in central ecological positions. Long-term fallow land had a significantly higher hyphal length density, GRSP, mean weight diameter (MWD), geometric mean diameter (GMD), and C concentration of GRSP (C-GRSP) than the cropland. The soil aggregate associated soil organic carbon (SOC) was 16.8, 18.6, and 13.8% higher under fallow land compared to that under cropland at HLJ, JL, and LN study sites, respectively. The structural equation model and random forest regression revealed that AMF diversity, network complexity, and their secreted GRSP mediate the effects of CC and LUC on C-GRSP and aggregate-associated SOC. This study elucidates the climate sensitivity of C within GRSP and soil aggregates which response symmetry to LUC and highlights the potential importance of AMF in C sequestration and climate change mitigation.

In this study, we examined the effect of health on the elderly's labor supply in rural China based on the data of the Chinese Health and Nutrition Survey (CHNS) from 1997 to 2006. We used simultaneous equations to address the endogeneity problem of health and estimate the models with censored data of labor supply by the full information maximum likelihood estimation. We found that the failing health does not significantly decrease the elderly's labor supply in rural areas when using both the subjective (self-reported health status) and objective (hypertension diagnosed or not) health indicators. Our finding indicates the phenomenon of “ceaseless toil” for the elderly in rural China, i.e., the elderly almost work their whole life even if they are not physically capable. The results remain robust when using a two-stage limited information maximum likelihood estimation.

PurposeThis study aimed to investigate the effect of OCT4&SOX2 specific cytotoxic T lymphocytes (CTLs) plus programmed cell death protein-1 (PD-1) inhibitor (nivolumab) on treating breast cancer stem-like cells (BCSCs) in vitro and drug-resistance breast cancer (DRBC) mice in vivo.MethodsIn total, 160 breast cancer patients were enrolled following the immunofluorescence assay to detect tumor OCT4 and SOX2 expressions. CD154-activated B cells were co-cultured with CD8+ T cells (from breast cancer patients) in the presence of OCT4&SOX2 peptides, CMV pp65 peptides (negative control), and no peptides (normal control). MCF7-BCSCs were constructed by drug-resistance experiment and sphere-formation assay, then DRBC mice were constructed by planting MCF7-BCSCs. Subsequently, different doses of OCT4&SOX2 CTLs and PD-1 inhibitor (nivolumab) were used to treat MCF7-BCSCs and DRBC mice.ResultsOCT4 and SOX2 correlated with poor differentiation, more advanced stage, and worse prognosis in breast cancer patients. In vitro, OCT4&SOX2 CTLs with effector-target ratio (ETR) 5:1, 10:1 and 20:1 presented with increased cytotoxic activity compared to CMV pp65 CTLs with ETR 20:1 (negative control) and Control CTLs with ETR 20:1 (normal control) on killing MCF7-BCSCs. Besides, PD-1 inhibitor (nivolumab) improved the cytotoxic activity of OCT4&SOX2 CTLs against MCF7-BCSCs in a dose-dependent manner. In vivo, OCT4&SOX2 CTLs plus PD-1 inhibitor (nivolumab) decreased tumor volume and tumor weight while increased tumor apoptosis rate compared to OCT4&SOX2 CTLs alone, PD-1 inhibitor (nivolumab) alone, and control.ConclusionOCT4&SOX2 CTLs exhibit good efficiency and synergize PD-1 inhibitor (nivolumab) in treating BCSCs and DRBC.

The Huaibei grey donkey (HGD) is an endangered species and a vital native breed in Anhui Province, China. However, its complete mitogenome, phylogeny, and maternal origin remain unclear. The objectives of this study were to detect the genetic diversity of the HGD and investigate its phylogenetic relationship with other breeds to inform conservation management. The complete mitogenome of the HGD was sequenced through next-generation sequencing, and the most variable region in the mitochondrial DNA displacement-loop (D-loop) was amplified via a polymerase chain reaction (PCR). Next, we used the median-joining network (MJN) to calculate the genetic relationships among populations and the neighbor-jointing method to build a phylogenetic tree and speculate as to its origin. The results showed that the mitogenome contains 22 tRNAs, 2 rRNAs, 13 PCGs, and 1 D-loop region. Analyzing the D-loop region of the HGDs, we identified 23 polymorphic sites and 11 haplotypes. The haplotype and nucleotide diversity were 0.87000 (Hd) and 0.02115 (Pi), respectively. The MJN analysis indicated that the HGD potentially has two maternal lineages, and phylogenetic analysis indicated that the Somali lineage could be the most probable domestication center for this breed. Therefore, our mitogenome analysis highlights the high genetic diversity of the HGD, which may have originated from the Somali wild ass, as opposed to the Asian wild ass. This study will provide a useful resource for HGD conservation and breeding.

The consideration of environmental factors has long been crucial to developing theories about the spatial variability of species diversity. However, the effects of global warming on Collembola, in permafrost wetlands, are largely unknown. Understanding how Collembola are affected by climate warming is important as they directly affect the community assembly and decomposition processes of plant litter within soil ecosystems. A peatland area in a cold temperate monsoon climate zone in the Great Hing’an Mountains of Northeast China was selected as the study area. Collembola were captured using an aspirator after five years of simulated warming using open top chambers (OTCs). Sampling in different growth seasons showed different characteristics in the control (CK) and warming (OTCs) treatment. Further, the results showed that (1) warming treatment increased the species richness and abundance of Collembola in the different seasons, except in May, (2) warming increased Collembola abundance in permafrost wetlands, and the warming effect was more significant during the cold season (about eight times in April), (3) species composition differed significantly in the control and warming treatment in May and September, and (4) the Collembola species composition in permafrost wetlands was mainly determined by air humidity, indicating different responses of Collembola species to the indirect effect of warming on water availability. We found that warming was the primary factor positively affecting the abundance of Collembola. An increase of Collembola abundance and community alteration to warming could have profound cascading effects on the microbes and plants they feed on in permafrost wetlands.

Abstract Plenty of reports have probed the involvement of abnormally expressed lncRNAs in multiple cancers, including lung squamous cell carcinoma (LUSC). Through online database GEPIA, lncRNA PITPNA antisense RNA 1 (PITPNA‐AS1) was highly expressed in LUSC samples, and these tendency was further affirmed in LUSC cells. The aim of current study was to investigate the related mechanism of PITPNA‐AS1 in LUSC. Functional experiments verified that depletion of PITPNA‐AS1 hampered the proliferative and migratory abilities, but accelerated apoptosis of LUSC cells. Additionally, we observed the increased expression of HMGB3 and its positive correlation with PITPNA‐AS1 in LUSC samples. Interestingly, PITPNA‐AS1 mainly located in the cytosol of LUSC cells, and also affected mRNA stability of HMGB3. Furthermore, the repressed mRNA stability of HMGB3 by PITPNA‐AS1 via TAF15 was exposed through mechanism experiments. The mediatory function of PITPNA‐AS1 on HMGB3 was validated via rescue assays. All in all, PITPNA‐AS1 promoted the proliferation and migration of LUSC cells via stabilizing HMGB3 by TAF15. In conclusion, our study displayed a novel mechanism underlying PITPNA‐AS1 in LUSC cells.

For the evaluation of college students’ physical health, the t-test, one-way ANOVA test, fuzzy comprehensive evaluation, and other methods were used to establish the models of back generation test, ANOVA, and fuzzy comprehensive evaluation; and Matlab, Excel, VBA, and other software programming were used to derive the significant influence of body weight on college students’ physical health, no significant difference of students’ physical health in different places of birth and the comprehensive score of college students’ physical health. The results such as the comprehensive score of health evaluation were used to analyze the physical health test results of college students in Zhuzhou city colleges and universities in the context of the promulgation of the new Student Physical Health Standard and several higher school sports policies by using the fuzzy comprehensive evaluation method to comprehensively grasp the physical health condition of students and provide reference for school sports reform and students’ physical exercise. Conclusion. The current situation of physical health of college students in Zhuzhou City is less satisfactory, and most of the students’ physical test results are concentrated in the passing area, with fewer students with excellent and good proportions. The policy changes, system construction, implementation, and students’ attitudes were analyzed, and solution measures were proposed to intervene in time to turn the situation around and improve students’ physical health.

Abstract Background Sepsis represents a major worldwide healthcare burden. However, how body-mass index (BMI) is related to the long-term risk of sepsis-related mortality in low- and middle-income countries remains uncertain. Methods We examined the associations of sepsis-related mortality with both baseline BMI and waist circumference (WC) using data from China Kadoorie Biobank, a prospective cohort recruited during 2004–2008 and followed up to December 2016. After excluding participants with chronic obstructive pulmonary disease, tuberculosis, cancer, heart disease, and stroke, and omitting the first 3 years of follow-up, 440,763 participants remained for analysis. This paper investigates vibration suppression of uncertain hose and drogue systems in the presence of actuator nonlinearities. Firstly, a previously presented model of the hose and drogue systems is extended to describe how the hose and drogue systems restrain the vibration, while the accompanying unknown aerodynamic coefficients are estimated by invoking the parameter projection method. Subsequently, for the actuator nonlinearities of dead-zone and saturation, a smooth dead-zone approximate function is constructed to design the dead-zone compensation method, based upon which the proposed control scheme can handle actuator dead-zone and saturation simultaneously while improving the output efficiency of the actuator. Next, for the actuator nonlinearities of backlash and saturation, a smooth backlash inverse is constructed based upon which the presented control scheme can cope with the both actuator nonlinearities simultaneously. Finally, by utilizing backstepping method and hyperbolic tangent function, the proposed control schemes can also achieve the control objectives of vibration suppression and external disturbance attenuation. Simulation examples are included to demonstrate the validity of the proposed control schemes.

Linked open data is a relatively new topic area with great potential in a wide range of fields. In the tourism domain, many studies are using linked open data to address the problem of location-based recommendation by integrating data with other linked open datasets to enrich data and tourism content for reacting to the needs of tourists. This work aims not only to present a systematic review and mapping of the linked open data in location-based recommendation system on tourism domain, but also to provide an overview of the current research status in the area. First, we classify journal papers in this area from 2001 to 2018 by the year of publication. Second, we analyze and categorize journal papers by the different recommendation applications including problem formulations, data collections, proposed algorithms/systems, and experimental results. Third, we group the linked open data sources used in location-based recommendation system on tourism. Next, we summarize the research achievements and present the distribution of the different categories of location-based recommendation applications via linked open data. Last, we also guide the possible future research direction for the linked open data in location-based recommendations on tourism.

Traveling as a very popular leisure activity enjoyed by many people all over the world. Typically, tourists have different kinds of preferences about their itineraries, limited time budgets, unfamiliar with the wide range of Points-of-Interest (POIs) in a city, so that planning an itinerary is quite tedious, time-consuming, and challenging for them. In this paper, we propose an adaptive genetic algorithm for personalized itinerary planning for travelers to plan their itineraries better. Firstly, desired starting POIs (e.g., POIs that are close to their hotels) and destination POIs (e.g., POIs that are near train stations or airports) are considered in our approach. Secondly, we also take some general factors into account that travelers would consider in their preferences of an itinerary, which are mandatory POIs, the total number of POIs, the overall POI popularity, the overall cost, and the overall rating. Thirdly, we view this kind of recommendation task as a Multi-Objective Optimization problem, and we propose an adaptive genetic algorithm with the crossover and mutation probabilities (AGAM) for solving this problem to better find the best global solution. Fourthly, we allocate different weights to every factor which considered in our paper to generate a personalized itinerary recommendation for better meet many kinds of preferences of tourists. Finally, we compare our approach against baselines on real-world datasets which include six touristic cities, and the experimental results show that the AGAM achieves better recommendation performance in terms of the mandatory POIs, total POI visits, overall POI popularity, total travel time (including travel time and visit duration), overall cost, and overall rating.
