Animal studies have shown that capsaicin plays a positive role in weight management. However, the results in human research are controversial. Therefore, the present systematic review and meta-analysis aimed to evaluate the effect of capsaicin on weight loss in adults. We searched PubMed, Embase, China Biomedical Literature Database (CBM), Cochrane library and clinical registration centre, identifying all randomised controlled trials (RCT) published in English and Chinese to 3 May 2022. A random-effect model was used to calculate the weighted mean difference (WMD) and 95 % CI. Heterogeneity between studies was assessed by the Cochran Q statistic and I-squared tests (I 2 ). Statistical analyses were performed using STATA version 15.1. P-values < 0·05 were considered as statistically significant. From 2377 retrieved studies, fifteen studies were finally included in the meta-analyses. Fifteen RCT with 762 individuals were included in our meta-analysis. Compared with the control group, the supplementation of capsaicin resulted in significant reduction on BMI (WMD: −0·25 kg/m2, 95 % CI = –0·35, –0·15 kg/m2, P < 0·05), body weight (BW) (WMD: −0·51 kg, 95 % CI = –0·86, –0·15 kg, P < 0·05) and waist circumference (WC) (WMD: −1·12 cm, 95 % CI = –2·00, –0·24 cm, P < 0·05). We found no detrimental effect of capsaicin on waist-to-hip ratio (WMD: −0·05, 95 % CI = –0·17, 0·06, P > 0·05). The current meta-analysis suggests that capsaicin supplementation may have rather modest effects in reducing BMI, BW and WC for overweight or obese individuals.

Here, the MO (coumarin pyridine derivatives) was devised by engineering lipophilic coumarin and cationic pyridine salt, which not only lastingly anchored onto the plasma membrane in dark due to appropriate amphipathicity and electrostatic interactions but also in situ reflected the membrane damage and heterogeneity with secretion of extracellular vesicles (EVs) under reactive oxygen species regulation and was investigated by two-photon fluorescence lifetime imaging microscopy. Background Non-communicable diseases (NCD) have become the leading cause of death in the world at present. Among them, the most important are cardiovascular diseases (CVD), cancer, chronic respiratory diseases and diabetes. This study aimed to analyze the time trends of mortality rates caused by the four major non-communicable diseases (NCD4) in China between 2009 and 2020, and to provide the evidence basis for policy implementation, information system, and health resource management of chronic NCD in China. Methods Data on resident death was collected from the National Mortality Surveillance data set and used to analyze the crude mortality rates, standardized mortality rates and changing trends of NCD4 among different genders, urban and rural areas, and geographical regions in China from 2009 to 2020. The Joinpoint Regression Models were fitted by the weighted least squares method. The average annual percent change (AAPC) and its 95% confidence interval (CI) were calculated for the entire time period. Results From 2009 to 2020, the standardized mortality from four major NCD combined in China decreased from 534.51 to 395.84%, with the AAPC value at -2.8% (95% CI [-3.7% to -1.8%]). The standardized mortality from CVD, cancer, and chronic respiratory diseases decreased, but the standardized mortality of diabetes increased, with AAPC values at -2.2%, -1.8%, -8.0% and 1.9% respectively. Conclusions From 2009 to 2020, the mortality rate of chronic NCD is on the decline, but chronic NCD have become the leading cause of death of residents. Close attention needs to be paid on NCD which affecting the health of the labor force population in China. The prevention and treatment of diabetes, male and west region NCD should be enhanced.

Observational studies suggest birth weight and childhood obesity are closely associated with age at menarche. However, the relationships between them are currently inconsistent and it remains elusive whether such associations are causal. Therefore, the aim of the study was to investigate whether there existed causal relationships between birth weight, childhood obesity and age at menarche.A two-sample Mendelian randomization (MR) study. The standard inverse variance weighted MR analyses were adopted to evaluate the causal effects of birth weight (n = 143,677), childhood body mass index (BMI) (n = 39,620) on age at menarche (n = 182,416) with summary statistics from large-scale genome-wide association studies (GWASs). Meanwhile, we validated our MR results with some sensitivity analyses including maximum likelihood, weighted-median and MR pleiotropy residual sum and outlier methods.The present study showed that each one standard deviation (1-SD) lower birth weight was predicted to result in a 0.1479 years earlier of age at menarche (β = .1479, 95% confidence interval [CI] = 0.0422-0.2535; p = 0.0061). We also found that genetically predicted 1-SD increase in childhood BMI was causally associated with early age at menarche (β = -.3966, 95% CI = -0.5294 to -0.2639; p = 4.73E-09).Our MR study suggests the causal effect of lower birth weight and higher childhood BMI on the increased risk of earlier menarche. It may be the opportune time to carry out weight control intervention in prenatal and early childhood development periods to prevent early menarche onset, thus decreasing the future adverse consequences.

SUMMARY: This study estimates causality of physical activity (PA) on bone mineral density (BMD) by conducting multivariable Mendelian randomization (MR). The findings suggest that habitual vigorous PA increases lumbar spine BMD, and higher overall acceleration average would improve forearm BMD. The results could promote PA intervention targeting individuals with optimized type. INTRODUCTION: Evidence from epidemiologic studies showed type, frequency, and duration of PA influenced BMD. However, these observational studies may be confounded by many factors, resulting in spurious associations. We aimed to conduct multivariable MR to estimate the causal effect of self-reported and device-measured PA on osteoporosis. METHODS: Three self-reported and two device-measured PA-related traits were selected as exposures. Outcomes were BMD at different skeletal sites: femoral neck BMD (FN BMD), lumbar spine BMD (LS BMD), and forearm BMD (FA BMD). Exposure datasets were obtained from UK Biobank with total 377,234 subjects. Outcome datasets were obtained from GEFOS consortium with 53,236 subjects. Standard MR analysis and multivariable MR were conducted to assess the total and direct causal effect of PA on BMD. RESULTS: For self-reported PA, inverse-normalized moderate-to-vigorous had a direct causal effect on FN BMD independently (β = − 1.116 (95% confidence interval, 95%CI: − 2.210, − 0.023), P = 0.045); vigorous PA showed a direct effect (β = 3.592 (95%CI: 0.310, 6.874), P = 0.032) on LS BMD independently. While overall acceleration average and fraction of accelerations both had a direct causal effect on FA BMD independently. CONCLUSIONS: Habitual vigorous PA could increase LS BMD. Individuals with higher overall acceleration average would have a higher FA BMD.

Recently, phototherapy has attracted much attention due to its negligible invasiveness, insignificant toxicity and excellent applicability. The construction of a newly proposed nanosystem with synergistic photothermal and photodynamic tumor-eliminating properties requires a delicate structure design. In this work, a novel therapeutic nanoplatform (denoted as BCS-Ce6) based on defective cobalt hydroxide nanosheets was developed, which realized hypoxia-relieved photothermal-enhanced photodynamic therapy against cancer. Defective cobalt hydroxide exhibited high photothermal conversion efficacy at the near-infrared region (49.49% at 808 nm) as well as enhanced catalase-like activity to produce oxygen and greatly boost the singlet oxygen generation by a photosensitizer, Ce6, realizing efficacious dual-modal phototherapy. In vivo and in vitro experiments revealed that BCS-Ce6 can almost completely extinguish implanted tumors in a mouse model and present satisfactory biocompatibility during the treatment. This work sets a new angle of preparing photothermal agents and constructing comprehensive therapeutic nanosystems with the ability to modulate the hypoxic tumor microenvironment for efficient cancer therapy.

Phototheranostics, the integration of phototherapy and multimodal imaging into one nanoagent, is highly significant for precise cancer treatment, during which how the photo-energy is converted plays a critical role. The Forster resonance energy transfer (FRET) mechanism has opened up a new avenue for diversifying energy conversion, demonstrating great promise for enhanced phototheranostics. Herein, a theranostic nanoagent (HCY–TPA), based on single-molecule FRET, was designed, where the harvested photo-energy was not only transformed into near-infrared fluorescence but also acoustic energy for deep-seated tumor imaging, as well as enhanced thermal and type-I ROS generation for efficient treatment, realizing optimized phototheranostics. Furthermore, the dual cation endowed it with superior mitochondrial targeting ability for precise treatment. In vivo mouse model experiments demonstrated that the proliferation of tumors was effectively inhibited, compared to that by single HCY or TPA, which ultimately demonstrated a synergistic effect in multimodal imaging guiding synergistic cancer treatment.

Photodynamic therapy (PDT), as an emerging treatment modality, which takes advantage of reactive oxygen species (ROS) generated upon light illumination to ablate tumours, has suffered from a limited treatment depth, strong oxygen dependence and short ROS lifespan. Herein, we developed a highly efficient NIR-I light (808 nm laser) initiated theranostic system based on a fluorescent photosensitizer (EBD-1) with cancer cell membrane targeting ability, which can realize large penetration depth in tissue, generate superoxide radicals (O2−˙) to relieve the oxygen-dependence, confine the ROS oxidation at the cell membrane, and self-report the cell viability during the PDT process. In vivo experiments demonstrated that EBD-1 under 808 nm light successfully accomplished remarkable cancer ablation. Cell viability is greatly affected by external stimulus eliciting correlated dynamical physiological processes for cells to choose survival or death. A few fluorescent probes have been designed to detect whether the cell is in survival state or apoptotic state, but monitoring the regulation process of the cell undergoing survival to death remains a long-standing challenge. Herein, we highlight the in situ monitor of mitochondria regulating the cell viability by the RNA-specific fluorescent photosensitizer L. At normal conditions, L anchored mitochondria and interacted with mito-RNA to light up the mitochondria with red fluorescence. With external light stimulus, L generated reactive oxide species (ROS) and cause damage to mitochondria, which activated mitochondrial autophagy to prevent death, during which the red fluorescence of L witnessed dynamical distribution in accordance with the evolution of vacuole structures containing damaged mitochondria into autophagosomes. However, with ROS continuously increasing, the mitochondrial apoptosis was eventually commenced and L with red fluorescent was gradually accumulated in the nucleoli, indicating the programmed cell death. This work demonstrated how the delicate balance between survival and death are regulated by mitochondria.

Iron centers featuring thiolates in their metal coordination sphere (as ligands or substrates) are well-known to activate dioxygen. Both heme and non-heme centers that contain iron-thiolate bonds are found in nature. Investigating the ability of iron-thiolate model complexes to activate O2 is expected to improve the understanding of the key factors that direct reactivity to either iron or sulfur. We report here the structural and redox properties of a thiolate-based dinuclear Fe complex, [FeII 2(LS)2] (LS2- = 2,2′-(2,2′-bipyridine-6,6′-iyl)bis(1,1-diphenylethanethiolate)), and its reactivity with dioxygen, in comparison with its previously reported protonated counterpart, [FeII 2(LS)(LSH)]+. When reaction with O2 occurs in the absence of protons or in the presence of 1 equiv of proton (i.e., from [FeII 2(LS)(LSH)]+), unsupported μ-oxo or μ-hydroxo FeIII dinuclear complexes ([FeIII 2(LS)2O] and [FeIII 2(LS)2(OH)]+, respectively) are generated. [FeIII 2(LS)2O], reported previously but isolated here for the first time from O2 activation, is characterized by single crystal X-ray diffraction and Mössbauer, resonance Raman, and NMR spectroscopies. The addition of protons leads to the release of water and the generation of a mixture of two Fe-based "oxygen-free" species. Density functional theory calculations provide insight into the formation of the μ-oxo or μ-hydroxo FeIII dimers, suggesting that a dinuclear μ-peroxo FeIII intermediate is key to reactivity, and the structure of which changes as a function of protonation state. Compared to previously reported Mn-thiolate analogues, the evolution of the peroxo intermediates to the final products is different and involves a comproportionation vs a dismutation process for the Mn and Fe derivate, respectively.

Considering the multiple biological barriers before the entry of photosensitizers (PSs) into cytoplasm, it is of paramount importance to track PSs to elucidate their behaviors and distributions to guide the photodynamic therapy (PDT). Also, the developed PSs suffer from strong oxygen dependency. However, reports on such ideal theranostic platforms are rare. Herein, we developed a theranostic platform (CMTP-2) based on the coumarin-based D-π-A system, which, for the first time, can reveal the holistic intracellular delivery pathway and near-infrared (NIR)-activated mitophagy to guide synergistic type-I PDT and photothermal therapy. The dynamic endo-lysosomal escape of CMTP-2 was monitored, as well as its changeable distributions in endosomes, lysosomes, and mitochondria, demonstrating the preferential accumulation in mitochondria at the end. Upon NIR-I irradiation, CMTP-2 generated toxic radicals and heat, triggering the execution of mitophagy and apoptosis. In vivo experiments on mice indicated that CMTP-2 under 808 nm irradiation realized complete cancer ablation, showing great potential for advancements in synergistic phototherapy.

In Ghana, the management of hypertension in primary health care is a cost-effective way of addressing premature deaths from vascular disorders that include hypertension. There is little or no evidence of large-scale studies on the prevalence, risk, and knowledge/awareness of hypertension in students aged 12-22 years in Ghana. In a cross-sectional study, blood pressure, anthropometric indices, and knowledge/awareness assessment of students at second-cycle schools were recorded from 2018 to 2020 in three regions of Ghana. Multistage cluster sampling was used in selecting regions and the schools. Prevalence of prehypertension and hypertension was categorized by the Joint National Committee 7, where appropriate, chi-square, scatter plots, and correlations were used in showing associations. A total of 3165 students comprising 1776 (56.1%) females and 1389 (43.9%) males participated in this study within three regions of Ghana. Background. Hypertension (HTN) is the second main source of outpatient morbidity in Ghana, and the understanding of a disease is necessary for its prevention and management. Language and communication are contributing factors to HTN in Ghana. No studies have been conducted to assess knowledge/awareness of HTN (in the context of its understanding) among students in Ghana. Following a local name for HTN in Ghana, researchers interviewed students through a focus group to assess their understanding/perception (meaning, cause, and prevention) of the disease. Available literature has concerned itself with clients’ knowledge of their condition (diagnosis) rather than their comprehension of the true nature of what HTN is. The objective of this study is to assess the knowledge/awareness of HTN in the context of its understanding of the meaning, perception, causes, and prevention of hypertension among students of Ghana’s Senior High School (Second Cycle). Semistructured interviews with the use of the theme lists were employed. Background Alzheimer's disease (AD) is a progressive brain disorder characterized by cognitive skills deterioration that affects many elderly individuals. The identified genetic loci for AD failed to explain the large variability in AD and very few causal factors have been identified so far. Results mvMR showed that increasing years of schooling (OR=0.674, 95%CI: 0.571-0.796, P=3.337E-06) and genetically elevated HDL cholesterol (OR ranging from 0.697 to 0.830, P=6.940E-10) were inversely associated with AD risk, genetically predicted total cholesterol (OR=1.300, 1.196 to 1.412; P=6.223E-10) and LDL cholesterol (OR=1.193, 1.097 to 1.296, P=3.564E-05) were associated with increasing AD risk. Genetically predicted FG was suggestively associated with increased AD risk. Furthermore, MR-BMA analysis also confirmed FG and years of schooling as two of the top five causal risk factors for AD. Conclusions Our findings might provide us novel insights for treatment and intervention into the causal risk factors for AD or AD-related complex diseases. Methods By using extension methods of Mendelian randomization (MR)--multivariable MR (mvMR) and MR based on Bayesian model averaging (MR-BMA), we intend to estimate the potential causal relationship between nine risk factors and AD outcome and try to prioritize the most causal risk factors for AD.

To address this issue, the structure of the previously described bioinspired [NiFe]-hydrogenase complex [(LNiFII)-Ni-N2S2-F-II Cp(CO)](+) ((LNiFeCp)-Fe-II-Cp-II, with L-N2S2 = 2,2'-(2,2'-bipyridine-6,6'-diy1)bis(1,1'-diphenylethanethiolate) and Cp = cyclopentadienyl) has been fine-tuned by modifying exclusively the Fe site. In [(LNiFeCp)-Ni-N2S2-Fe-II-Cp-II* (CO)](+) ((LNiFeCp)-Fe-II-Cp-II*, with Cp* = pentamethylcyclopentadienyl), the Cp- ligand has been replaced by Cp*- to change both the redox and structural properties of the overall complex as a consequence of the steric hindrance of Cp*-. The (LNiFeCp)-Fe-II-Cp-II* complex acts as an efficient electrocatalyst to produce H-2. Density functional theory (DFT) calculations support a CEEC cycle, following an initial reduction. The initial protonation leads to the cleavage of one thiolate-iron bond and the next reduction to the generation of a bridging Fe-based hydride moiety. Interestingly, the second protonation step generates a species containing a terminal Ni-based thiol and a bridging hydride. In the presence of CO, the electrocatalytic activity of (LNiFeCp)-Fe-II-Cp-II* for H-2 production is markedly inhibited (about 90% of loss), while only a partial inhibition (about 30% of loss) is observed in the case of (LNIFeCp)-I-II-Cp-II. DFT calculations rationalized this effect by predicting that interactions of the one- and two-electron-reduced species for (LNiFeCp)-Fe-II-Cp-II* with CO are thermodynamically more favorable in comparison to those for (LNiFeCp)-Fe-II-Cp-II.

The aim of this study is to analyse the efficacy rate of folate for the treatment of hyperhomocysteinaemia (HHcy) and to explore how folate metabolism-related gene polymorphisms change its efficacy. This study also explored the effects of gene–gene and gene–environment interactions on the efficacy of folate. A prospective cohort study enrolling HHcy patients was performed. The subjects were treated with oral folate (5 mg/d) for 90 d. We analysed the efficacy rate of folate for the treatment of HHcy by measuring homocysteine (Hcy) levels after treatment. Unconditioned logistic regression was conducted to analyse the association between SNP and the efficacy of folic acid therapy for HHcy. The efficacy rate of folate therapy for HHcy was 56·41 %. Covalent organic frameworks (COFs) have appeared as a variety of promising nanomaterials for photodynamic therapy (PDT) owing to their excellent biosafety and structural diversity. However, finite aqueous dispersibility, deficient subcellular targeting ability and undesirable PDT efficiency had seriously restricted their biomedical application. Herein, we prepared two solution-dispersible nanoparticle COFs via a mild solution-phase synthesis, denoted as EB-TFP and DPP-TFP, investigating the effect of COF’s surface charges on their subcellular targeting ability and PDT efficiency. With tuning the COF’s surface charges, the positive charged COFs (EB-TFP) possessed the elevated mitochondrial targeting ability, higher O2-. generation ability and fluorescence at 510 nm. Therefore, EB-TFP successfully employed as multifunctional PSs with excellent mitochondrial targeting ability for the imaging-guided type Ⅰ PDT. This work provides a “killing two birds with one stone” strategy to improve subcellular targeting and ROS generation ability for expanding the biomedical applications via tuning the COFs’ surface charges.

This study aimed to examine the effects of different types of bullying victimization (direct, relational, and cyber) on psychological symptoms, self-harm, and suicidality (including suicidal ideation and attempts) among adolescents, and to explore whether these effects may vary by gender. The data were obtained from a cross-sectional study of adolescents (n = 11,248, 46.7% females) with a mean age of 13.83 years from grade 5 to 12 in Henan, China. A series of binary logistic regression models were conducted to estimate the associations between different types of bullying victimization and psychological symptoms, self-harm, suicidal ideation, and suicidal attempts, after adjusting for demographic covariates. All three types of bullying victimization were significantly associated with psychological symptoms, self-harm, suicidal ideation, and suicidal attempts. Adolescents who suffered from cyberbullying victimization were more likely to commit self-harm and suicidal attempts as compared to direct and relational victimization. Female adolescents who suffered from relational bullying tend to have a higher risk of suicidal attempts than male adolescents. The current study demonstrated the negative effect of bullying victimization on adolescents’ adverse psychological outcomes and gender difference need to be taken into account in developing targeted intervention strategies to address bullying victimization.

The di-iron complex [(CO)LN2S2FeIIFeII(CO)Cp]+ (with LN2S2 = 2,2′-(2,2′-bipyridine-6,6′-diyl)bis(1,1-diphenylethanethiolate and Cp = cyclopentadienyl) has been synthesized and fully characterized. In the solid state, it contains two CO ligands: one bound to the {FeCp} moiety in a semibridging manner and one terminally bound to the {FeLN2S2} moiety. This dinuclear iron complex is thus not isostructural to [LN2S2NiIIFeII(CO)Cp]+, which contains a single CO ligand terminally bound to the Fe site. However, at low concentrations in MeCN solutions, the CO ligand coordinated to the {FeLN2S2} moiety is removed and the CO ligand bound to the {FeCp} moiety becomes fully bridging between the two Fe sites. Under such conditions, the di-iron complex displays similar catalytic performances to the parent NiFe complex (a comparable overpotential, η = 730 and 690 mV, and TON = 15 and 16, respectively). Cyclic voltammetry data give direct experimental evidence for an E[ECEC] mechanism, which was also previously proposed for the NiFe complex. However, the structure of the one-electron reduced species, the entry point of the catalytic cycle, slightly differs for the two systems: in [LN2S2NiI(CO)FeIICp], this is valence localized species on the site Ni and the CO ligand bridges the two metal ions, while in [(CO)LN2S2FeFeCp], this is a type II–III mixed-valence species with the CO terminally bound to the {FeLN2S2} unit.

A ceramic coating grown on the surface of aluminum wires by a micro-arc oxidation (MAO) process has high electrical breakdown strength and thermal stability. However, the aluminum wires with ceramic coating by MAO have been not used in transformer windings previously. The objective of this paper is to determine the electrical breakdown strength and thermal stability of the ceramic coating on aluminum wires by MAO for its potential usage in transformer windings. In this paper, electrical breakdown strength of aluminum windings processed by MAO was tested by a manually controlled up-down method. Thermal stability was evaluated through a Differential Scanning Calorimeter DSC822e and a thermogravimetric Analyzer TGA/SDTA851. The experimental results indicated thickness of the coating varied between 20 and 30 μm, which was measured by SEM. Electrical breakdown strength of the ceramic coating in the air was between 22.9 and 47.8 kV/mm, which was at the same level as that of epoxy resin (20 kV/mm) and NOMEX paper (18 kV/mm ~ 40 kV/mm). In the insulation oil, electrical breakdown strength of the ceramic coating was between 76.7 kV/mm and 115 kV/mm. The electrical breakdown strength of the ceramic coating in the air was recoverable because of a rapid formation of new oxide layer. Thermal properties of the ceramic coating on aluminum windings by MAO kept stable under the temperature from 25 degrees Celsius to 550 degrees Celsius, which was significantly better than those of traditional transformer winding insulation materials such as epoxy resin (25 degrees Celsius ~ 180 degrees Celsius) and NOMEX paper (25 degrees Celsius ~ 220 degrees Celsius). Therefore, the aluminum windings with ceramic coating based on MAO showed a potential as dry-type transformer windings.

Stroke is a complex disease with multiple etiologies. Numerous studies suggest an established association between obesity and stroke, which may partly arise from the shared genetic components between the two phenotypes. Despite genome-wide association studies (GWASs) have identified some loci associated with stroke and obesity individually, the estimated genetic variability explained by these loci is limited (especially for stroke) and the pleiotropic loci between them are largely unknown. In this study, we jointly applied the pleiotropy-informed conditional false discovery rate (cFDR) method and the genetic analysis incorporating pleiotropy and annotation (GPA) method on summary statistics of two large GWASs to detect the genetic overlap between stroke (n = 446,696) and obesity (n = 681,275). International audience; In the Oxygen Reduction Reaction (ORR) domain, the investigation of new homogeneous catalysts is a crucial step towards the full comprehension of the key structural and/or electronic factors that control catalytic efficiency and selectivity. Herein, we report a unique non-heme diiron complex that can act as a homogeneous ORR catalyst in acetonitrile solution. This iron(II)-thiolate dinuclear complex, [FeII2(LS)(LSH)] ([Fe2SH]+) (LS2- = 2,2'-(2,2'-bipyridine-6,6'-diyl)bis(1,1-diphenylethanethiolate)) contains a thiol group in the metal coordination sphere. [Fe2SH]+ is an efficient ORR catalyst both in the presence of a one-electron reducing agent as well as under electrochemically assisted conditions. However, its selectivity is dependent on the electron delivery pathway, in particular, the process is selective for H2O2 production under chemical conditions (up to ~95%), whereas H2O is the main product during electrocatalysis (less than ~10% H2O2). Based on computational work alongside the experimental data, a mechanistic proposal is discussed that rationalizes the selective and tunable reduction of dioxygen.

Photodynamic therapy has been generally developed and approved as a promising theranostic technique in recent years, which requires photosensitizers to bear high efficiency of reactive oxygen species production, precisely targeting ability and excellent biocompatibility. The real-time monitoring the microenvironments such as viscosity dynamic involved in mitophagy mediated by photodynamic therapy is significantly important to understand therapeutic process but barely reported. In this work, a pyridinium-functionalized triphenylamine derivative, (E)-4-(2-(4'-(diphenylamino)-[1,1′-biphenyl]-4-yl)vinyl)-1-methylpyridin-1-ium iodide (Mito-I), was exploited as photosensitizer for mitochondria-targeted photodynamic therapy and as fluorescent probe for imaging the mitochondrial viscosity dynamic during mitophagy simultaneously. The results indicated that the additional phenyl ring in Mito-I was beneficial to promote its efficiency of singlet oxygen production. The excellent capability of targeting mitochondria and singlet oxygen generation allowed Mito-I for the specifically mitochondria-targeted photodynamic therapy. Moreover, Mito-I displayed off-on fluorescence response to viscosity with high selectivity and sensitivity. The observed enhancement in fluorescence intensity of Mito-I revealed the increasingly mitochondrial viscosity during mitophagy mediated by the photodynamic therapy of Mito-I. As a result, this work presents a rare example to realize the mitochondria-targeting photodynamic therapy as well as the real-time monitoring viscosity dynamic during mitophagy, which is of great importance for the basic medical research involved in photodynamic therapy.
